Abstract: Magnetic structures of the noncentrosymmetric magnet Sr$_2$MnSi$_2$O$_7$ were examined through neutron diffraction for powder and single-crystalline samples, as well as magnetometry measurements. All allowed magnetic structures under the space group $P\bar{4}2_1 m$ with the magnetic wavevector $\vec{q}_{\rm m}=(0,0,1/2)$~r.l.u., were analysed via irreducible representation and magnetic space group approaches. The compound is refined to have in-plane magnetic moments within the magnetic space group $Cmc2_1.1^{\prime}_c$ (\#36.177) under zero field, which can be altered to $P2_1 2_1 2_1.1^{\prime}_c$ (\#19.28) above $\mu_0 H=0.067(5)$~T to align induced weak-ferromagnetic components within one layer on the $ab$-plane. All refined parameters are provided following the recent framework based upon the magnetic space group, which better conveys when exchanging crystallographic information for commensurate magnetic structures., Comment: 7 pages, 4 figures

Abstract: We report the first measurement of the azimuthal anisotropy of J$/\psi$ at forward rapidity ($1.2<|\eta|<2.2$) in Au$+$Au collisions at $\sqrt{s_{_{NN}}}=200$ GeV at the Relativistic Heavy Ion Collider. The data were collected by the PHENIX experiment in 2014 and 2016 with integrated luminosity of 14.5~nb$^{-1}$. The second Fourier coefficient ($v_2$) of the azimuthal distribution of $J/\psi$ is determined as a function of the transverse momentum ($p_T$) using the event-plane method. The measurements were performed for several selections of collision centrality: 0\%--50\%, 10\%--60\%, and 10\%-40\%. We find that in all cases the values of $v_2(p_T)$, which quantify the elliptic flow of J$/\psi$, are consistent with zero. The results are consistent with measurements at midrapidity, indicating no significant elliptic flow of the J$/\psi$ within the quark-gluon-plasma medium at collision energies of $\sqrt{s_{_{NN}}}=200$ GeV., Comment: 369 authors from 72 institutions, 12 pages, 7 figures, 5 tables. v1 is version submitted to Physical Review C. However, whether long-term objectively measured PA is related to glycemic control remains unclear. The aim of this study was to investigate the relationship between long-term objectively measured PA and glycemic control in T2DM patients.Research Design and Methods: This prospective cohort study recruited T2DM patients admitted to a hospital-based diabetes management and education program. The primary outcome was glycemic control by hemoglobin A1c at 6 months after discharge. We defined poor glycemic control according to the Japanese Clinical Practice Guidelines. The PA was objectively measured using a three-axis accelerometer during 6 months’ period after discharge. The representative value of PA was the average daily steps during the measurement period and was divided into quartiles. To determine the relationship between the daily steps and poor glycemic control, we performed a multivariate logistic regression analysis.Results: Ninety-four participants were enrolled in the study. Their median age was 59 years, and 38 (40.0%) of them showed poor glycemic control. Multivariate logistic regression analysis showed that the first (Q1, ≤ 6106 steps/day) and second quartiles (Q2, 6107– 8258 steps/day) had significantly elevated risks of poor glycemic control compared to Q4 (≥ 10,542 steps/day), with odds ratios of 8.55 [95% confidence intervals (CI) =1.43– 51.23] and 15.62 (95% CI 2.63– 92.87), respectively.Conclusion: We found that lesser PA was significantly associated with poor glycemic control in T2DM patients. This finding may be beneficial for clinicians while providing long-term advice to diabetic patients.Keywords: glycemic control, long-term, physical activity, type 2 diabetes

Abstract: PHENIX presents a simultaneous measurement of the production of direct $\gamma$ and $\pi^0$ in $d$$+$Au collisions at $\sqrt{s_{_{NN}}}=200$ GeV over a $p_T$ range of 7.5 to 18 GeV/$c$ for different event samples selected by event activity, i.e. charged-particle multiplicity detected at forward rapidity. Direct-photon yields are used to empirically estimate the contribution of hard-scattering processes in the different event samples. Using this estimate, the average nuclear-modification factor $R_{d\rm Au,EXP}^{\gamma^{\rm dir}}$ is $0.925{\pm}0.023({\rm stat}){\pm}0.15^{\rm (scale)}$, consistent with unity for minimum-bias (MB) $d$$+$Au events. For event classes with moderate event activity, $R_{d\rm Au,EXP}^{\gamma^{\rm dir}}$ is consistent with the MB value within 5\% uncertainty. These results confirm that the previously observed enhancement of high-$p_T$ $\pi^0$ production found in small-system collisions with low event activity is a result of a bias in interpreting event activity within the Glauber framework. In contrast, for the top 5\% of events with the highest event activity, $R_{d\rm Au,EXP}^{\gamma^{\rm dir}}$ is suppressed by 20\% relative to the MB value with a significance of $4.5\sigma$, which may be due to final-state effects., Comment: 279 authors from 69 institutions, 8 pages, 3 figures, v1 is version submitted to Physical Review Letters. Abstract: Recently, the PHENIX Collaboration has published second- and third-harmonic Fourier coefficients $v_2$ and $v_3$ for midrapidity ($|\eta|<0.35$) charged hadrons in 0\%--5\% central $p$$+$Au, $d$$+$Au, and $^3$He$+$Au collisions at $\sqrt{s_{_{NN}}}=200$ GeV utilizing three sets of two-particle correlations for two detector combinations with different pseudorapidity acceptance [Phys. Rev. C {\bf 105}, 024901 (2022)]. This paper extends these measurements of $v_2$ to all centralities in $p$$+$Au, $d$$+$Au, and $^3$He$+$Au collisions, as well as $p$$+$$p$ collisions, as a function of transverse momentum ($p_T$) and event multiplicity. The kinematic dependence of $v_2$ is quantified as the ratio $R$ of $v_2$ between the two detector combinations as a function of event multiplicity for $0.5$$<$$p_T$$<$$1$ and $2$$<$$p_T$$<$$2.5$ GeV/$c$. A multiphase-transport (AMPT) model can reproduce the observed $v_2$ in most-central to midcentral $d$$+$Au and $^3$He$+$Au collisions. However, the AMPT model systematically overestimates the measurements in $p$$+$$p$, $p$$+$Au, and peripheral $d$$+$Au and $^3$He$+$Au collisions, indicating a higher nonflow contribution in AMPT than in the experimental data. Abstract: There is strong evidence for the formation of small droplets of quark-gluon plasma in $p/d/^{3}$He+Au collisions at the Relativistic Heavy Ion Collider (RHIC) and in $p$+$p$/Pb collisions at the Large Hadron Collider. In particular, the analysis of data at RHIC for different geometries obtained by varying the projectile size and shape has proven insightful. In the present analysis, we find excellent agreement with the previously published PHENIX at RHIC results on elliptical and triangular flow with an independent analysis via the two-particle correlation method, which has quite different systematic uncertainties and an independent code base. In addition, the results are extended to other detector combinations with different kinematic (pseudorapidity) coverage. Abstract: Field-dependent magnetic structure of a layered Dirac material EuMnBi$_2$ was investigated in detail by the single crystal neutron diffraction and the resonant x-ray magnetic diffraction techniques. On the basis of the reflection conditions in the antiferromagnetic phase at zero field, the Eu moments were found to be ordered ferromagnetically within the $ab$ plane and stacked antiferromagnetically along the $c$ axis in the sequence of up-up-down-down. Upon the spin-flop transition under the magnetic field parallel to the $c$ axis, the Eu moments are reoriented from the $c$ to the $a$ or $b$ directions forming two kinds of spin-flop domains, whereas the antiferromagnetic structure of the Mn sublattice remains intact as revealed by the quantitative analysis of the change in the neutron diffraction intensities. 9. Observation of Magnetopiezoelectric Effect in Antiferromagnetic Metal EuMnBi2

Abstract: We have experimentally studied a magnetopiezoelectric effect predicted recently for magnetic metals with low crystal symmetries. In EuMnBi2 with antiferromagnetic Mn moments at 77 K, dynamic displacements emerge along the $a$ direction upon application of ac electric fields in the $c$ direction, and increase in proportion to the applied electric fields. Such displacements are not observed along the $c$ direction of EuMnBi2 or EuZnBi2 with nonmagnetic Zn ions. As temperature increases from 77 K, the displacement signals decrease and disappear at about 200 K, above which electric conduction changes from coherent to incoherent. These results demonstrate the emergence of the magnetopiezoelectric effect in a magnetic metal lacking inversion and time-reversal symmetries.

Abstract: We report spin-split Landau levels of quasi-two-dimensional Dirac fermions in a layered antiferromagnet EuMnBi$_2$, as revealed by interlayer resistivity measurements in a tilted magnetic field up to $\sim$35 T. The amplitude of Shubnikov-de Haas (SdH) oscillation in interlayer resistivity is strongly modulated by changing the tilt angle of the field, i.e., the Zeeman-to-cyclotron energy ratio. The effective $g$ factor estimated from the tilt angle, where the SdH oscillation exhibits a phase inversion, differs by approximately 50% between two antiferromagnetic phases. This observation signifies a marked impact of the magnetic order of Eu sublattice on the Dirac-like band structure. Abstract: Asymmetric nuclear collisions of $p$$+$Al, $p$$+$Au, $d$$+$Au, and $^{3}$He$+$Au at $\sqrt{s_{_{NN}}}=200$ GeV provide an excellent laboratory for understanding particle production, as well as exploring interactions among these particles after their initial creation in the collision. We present measurements of charged hadron production $dN_{\rm ch}/d\eta$ in all such collision systems over a broad pseudorapidity range and as a function of collision multiplicity. A simple wounded quark model is remarkably successful at describing the full data set. We also measure the elliptic flow $v_{2}$ over a similarly broad pseudorapidity range. Abstract: The experimental study of the collisions of heavy nuclei at relativistic energies has established the properties of the quark-gluon plasma (QGP), a state of hot, dense nuclear matter in which quarks and gluons are not bound into hadrons. In this state, matter behaves as a nearly inviscid fluid that efficiently translates initial spatial anisotropies into correlated momentum anisotropies among the produced particles, producing a common velocity field pattern known as collective flow. In recent years, comparable momentum anisotropies have been measured in small-system proton-proton ($p$$+$$p$) and proton-nucleus ($p$$+$$A$) collisions, despite expectations that the volume and lifetime of the medium produced would be too small to form a QGP. Here, we report on the observation of elliptic and triangular flow patterns of charged particles produced in proton-gold ($p$$+$Au), deuteron-gold ($d$$+$Au), and helium-gold ($^3$He$+$Au) collisions at a nucleon-nucleon center-of-mass energy $\sqrt{s_{_{NN}}}$~=~200 GeV. The unique combination of three distinct initial geometries and two flow patterns provides unprecedented model discrimination. Abstract: We present measurements of the transverse-momentum dependence of elliptic flow $v_2$ for identified pions and (anti)protons at midrapidity ($|\eta|<0.35$), in 0%--5% central $p$$+$Au and $^3$He$+$Au collisions at $\sqrt{s_{_{NN}}}=200$ GeV. When taken together with previously published measurements in $d$$+$Au collisions at $\sqrt{s_{_{NN}}}=200$ GeV, the results cover a broad range of small-collision-system multiplicities and intrinsic initial geometries. We observe a clear mass-dependent splitting of $v_2(p_{T})$ in $d$$+$Au and $^3$He$+$Au collisions, just as in large nucleus-nucleus ($A$$+$$A$) collisions, and a smaller splitting in $p$$+$Au collisions. Both hydrodynamic and transport model calculations successfully describe the data at low $p_T$ ($< 1.5$ GeV/$c$), but fail to describe various features at higher $p_T$. In all systems, the $v_2$ values follow an approximate quark-number scaling as a function of the hadron transverse kinetic energy per constituent quark($KE_T/n_q$), which was also seen previously in $A$$+$$A$ collisions., Comment: 353 authors, 12 pages, 7 figures, 3 tables. v2 is version accepted for publication in Phys. Abstract: We present a detailed measurement of charged two-pion correlation functions in 0%-30% centrality $\sqrt{s_{_{NN}}}=200$ GeV Au$+$Au collisions by the PHENIX experiment at the Relativistic Heavy Ion Collider. The data are well described by Bose-Einstein correlation functions stemming from L\'evy-stable source distributions. Using a fine transverse momentum binning, we extract the correlation strength parameter $\lambda$, the L\'evy index of stability $\alpha$ and the L\'evy length scale parameter $R$ as a function of average transverse mass of the pair $m_T$. We find that the positively and the negatively charged pion pairs yield consistent results, and their correlation functions are represented, within uncertainties, by the same L\'evy-stable source functions. The $\lambda(m_T)$ measurements indicate a decrease of the strength of the correlations at low $m_T$. The L\'evy length scale parameter $R(m_T)$ decreases with increasing $m_T$, following a hydrodynamically predicted type of scaling behavior. The values of the L\'evy index of stability $\alpha$ are found to be significantly lower than the Gaussian case of $\alpha=2$, but also significantly larger than the conjectured value that may characterize the critical point of a second-order quark-hadron phase transition., Comment: 448 authors, 25 pages, 11 figures, 4 tables, 2010 data. v3 is version accepted for publication in Phys. Abstract: We present measurements of the elliptic flow, $v_2$, as a function of transverse momentum ($p_T$), pseudorapidity ($\eta$), and centrality in $d$$+$Au collisions at $\sqrt{s_{_{NN}}}=$200, 62.4, 39, and 19.6 GeV. The beam-energy scan of $d$$+$Au collisions provides a testing ground for the onset of flow signatures in small collision systems. We measure a nonzero $v_2$ signal at all four collision energies, which, at midrapidity and low $p_T$, is consistent with predictions from viscous hydrodynamic models. Comparisons with calculations from parton transport models (based on the {\sc ampt} Monte Carlo generator) show good agreement with the data at midrapidity to forward ($d$-going) rapidities and low $p_T$. At backward (Au-going) rapidities and $p_T>1.5$ GeV/$c$, the data diverges from {\sc ampt} calculations of $v_2$ relative to the initial geometry, indicating the possible dominance of nongeometry related correlations, referred to as nonflow. We also present measurements of the charged-particle multiplicity ($dN_{\rm ch}/d\eta$) as a function of $\eta$ in central $d$$+$Au collisions at the same energies. We find that in $d$$+$Au collisions at $\sqrt{s_{_{NN}}}=$200 GeV the $v_2$ scales with $dN_{\rm ch}/d\eta$ over all $\eta$ in the PHENIX acceptance. At $\sqrt{s_{_{NN}}}=$62.4, and 39 GeV, $v_2$ scales with $dN_{\rm ch}/d\eta$ at midrapidity and forward rapidity, but falls off at backward rapidity. Abstract: Recently, multiparticle-correlation measurements of relativistic $p/d/^3$He$+$Au, $p$$+$Pb, and even $p$$+$$p$ collisions have shown surprising collective signatures. Here we present beam-energy-scan measurements of 2-, 4-, and 6-particle angular correlations in $d$$+$Au collisions at $\sqrt{s_{_{NN}}}$=200, 62.4, 39, and 19.6 GeV. We also present measurements of 2- and 4-particle angular correlations in $p$$+$Au collisions at $\sqrt{s_{_{NN}}}$=200 GeV. We find the 4-particle cumulant to be real-valued for $d$$+$Au collisions at all four energies. We also find that the 4-particle cumulant in $p$$+$Au has the opposite sign as that in $d$$+$Au, indicating that the correlations are geometrical and therefore collective in origin. Further we find that the 6-particle cumulant agrees with the 4-particle cumulant in $d$$+$Au collisions at 200 GeV, which indicates that nonflow effects are subdominant and provides strong evidence of collectivity., Comment: 317 authors, 6 pages, 4 figures. v2 is version accepted for publication by Phys. Abstract: During 2015 the Relativistic Heavy Ion Collider (RHIC) provided collisions of transversely polarized protons with Au and Al nuclei for the first time, enabling the exploration of transverse-single-spin asymmetries with heavy nuclei. Large single-spin asymmetries in very forward neutron production have been previously observed in transversely polarized $p$$+$$p$ collisions at RHIC, and the existing theoretical framework that was successful in describing the single-spin asymmetry in $p$$+$$p$ collisions predicts only a moderate atomic-mass-number ($A$) dependence. In contrast, the asymmetries observed at RHIC in $p$$+$$A$ collisions showed a surprisingly strong $A$ dependence in inclusive forward neutron production. The observed asymmetry in $p$$+$Al collisions is much smaller, while the asymmetry in $p$$+$Au collisions is a factor of three larger in absolute value and of opposite sign. Abstract: The cross section and transverse single-spin asymmetries of $\mu^{-}$ and $\mu^{+}$ from open heavy-flavor decays in polarized $p$+$p$ collisions at $\sqrt{s}=200$ GeV were measured by the PHENIX experiment during 2012 at the Relativistic Heavy Ion Collider. Because heavy-flavor production is dominated by gluon-gluon interactions at $\sqrt{s}=200$ GeV, these measurements offer a unique opportunity to obtain information on the trigluon correlation functions. Abstract: For the innovation of spintronic technologies, Dirac materials, in which the low-energy excitation is described as relativistic Dirac fermions, are one of the most promising systems, because of the fascinating magnetotransport associated with the extremely high mobility. To incorporate Dirac fermions into spintronic applications, their quantum transport phenomena are desired to be manipulated to a large extent by magnetic order in a solid. We here report a bulk half-integer quantum Hall effect in a layered antiferromagnet EuMnBi$_2$, in which field-controllable Eu magnetic order significantly suppresses the interlayer coupling between the Bi layers with Dirac fermions. In addition to the high mobility more than 10,000 cm$^2$/Vs, Landau level splittings presumably due to the lifting of spin and valley degeneracy are noticeable even in a bulk magnet. These results will pave a route to the engineering of magnetically functionalized Dirac materials., Comment: 26 pages, 4 figures, 1 table

Abstract: The fraction of $J/\psi$ mesons which come from B-meson decay, $\textrm{F}_{B{\rightarrow}J/\psi}$, is measured for J/$\psi$ rapidity \mbox{$1.2<|y|<2.2$} and $p_T>0$ in $p$+$p$ and Cu+Au collisions at $\sqrt{s_{_{NN}}}$=200 GeV with the PHENIX detector. The extracted fraction is $\textrm{F}_{B{\rightarrow}J/\psi}$ = 0.025 $\pm$ 0.006(stat) $\pm$ 0.010(syst) for $p$+$p$ collisions. For Cu+Au collisions, $\textrm{F}_{B{\rightarrow}J/\psi}$ is 0.094 $\pm$ 0.028(stat) $\pm$ 0.037(syst) in the Au-going direction ($-2.2

Abstract: We report a measurement of $e^+e^-$ pairs from semileptonic heavy-flavor decays in $p$+$p$ collisions at $\sqrt{s_{NN}}=200$~GeV. The $e^+e^-$ pair yield from $b\bar{b}$ and $c\bar{c}$ is separated by exploiting a double differential fit done simultaneously in dielectron invariant mass and $p_T$. We used three different event generators, {\sc pythia}, {\sc mc@nlo}, and {\sc powheg}, to simulate the $e^+e^-$ spectra from $c\bar{c}$ and $b\bar{b}$ production. The data can be well described by all three generators within the detector acceptance. However, when using the generators to extrapolate to $4\pi$, significant differences are observed for the total cross section. These difference are less pronounced for $b\bar{b}$ than for $c\bar{c}$. The same model dependence was observed in already published $d$+$A$ data. The $p$+$p$ data are also directly compared with $d$+$A$ data in mass and $p_T$, and within the statistical accuracy no nuclear modification is seen., Comment: 510 authors, 19 pages, 16 figures, 7 tables, 2006 data. v2 is version accepted for publication by Phys. Abstract: Magnetic structures of the noncentrosymmetric magnet Sr2MnSi2O7 were examined through neutron diffraction for powder and single‐crystalline samples, as well as magnetometry measurements. All allowed magnetic structures for space group P421m with the magnetic wavevector qm = (0, 0, ½) were refined via irreducible representation and magnetic space group analyses. The compound was refined to have in‐plane magnetic moments within the magnetic space group Cmc21.1′c (No. 36.177) under zero field, which can be altered to P212121.1′c (No. 19.28) above μ0H = 0.067 (5) T to align induced weak‐ferromagnetic components within one layer on the ab plane. All refined parameters are provided following the recent framework based upon the magnetic space group, which better conveys when exchanging crystallographic information for commensurate magnetic structures. Abstract: There is still no general consensus on how one can describe the out-of-equilibrium phenomena in matter induced by an ultrashort light pulse. We investigate the pulse-induced dynamics in a layered Dirac semimetal SrMnBi2 by pump-and-probe photoemission spectroscopy. At ~<1 ps, the electronic recovery slowed upon increasing the pump power. Such a bottleneck-type slowing is expected in a two-temperature model (TTM) scheme, although opposite trends have been observed to date in graphite and in cuprates. Subsequently, an unconventional power-law cooling took place at ~100 ps, indicating that spatial heat diffusion is still ill defined at ~100 ps. We identify that the successive dynamics before the emergence of heat diffusion is a canonical realization of a TTM scheme. Criteria for the applicability of the scheme is also provided.

Abstract: Emission of muonium ($\mu^{+}e^{-}$) atoms from silica aerogel into vacuum was observed. Characteristics of muonium emission were established from silica aerogel samples with densities in the range from 29 mg cm$^{-3}$ to 178 mg cm$^{-3}$. Spectra of muonium decay times correlated with distances from the aerogel surfaces, which are sensitive to the speed distributions, follow general features expected from a diffusion process, while small deviations from a simple room-temperature thermal diffusion model are identified. Abstract: We have applied Pade approximants to perturbative QCD calculations of event shape observables in e+e- --> hadrons. We used the exact O(alpha_s^2) prediction and the [0/1] Pade approximant to estimate the O(alpha_s^3) term for 15 observables, and in each case determined alpha_s(M_Z^2) from comparison with hadronic Z^0 decay data from the SLD experiment. Abstract: We have applied so-called `optimised' perturbation theory to resolve the renormalisation-scale (mu) ambiguity of exact O(alpha_s^2) QCD calculations of event shape observables in e+e- --> hadrons. We fitted the optimised predictions for 15 observables to hadronic Z0 decay data from the SLD experiment to determine alpha_s(M_Z^2). Comparing with results using the physical scale mu = M_Z we found no reduction in the scatter among alpha_s(M_Z^2) values from the 15 observables, implying that the O(alpha_s^2) predictions with optimised scales are numerically no closer to the exact all-orders results than those with the physical scale., Comment: 19 pages for text plus 4 pages for figures which were tar'ed, gzip'ed, uuencoded and put as one package. 40. Fabrication and thermal stability of arrays of Fe nanodots

Abstract: We have fabricated arrays of 60-nm-size magnetic Fe nanodots over a 1-cm2-size area using nanoporous alumina membranes as shadow masks. The size and size distribution of the nanodots correlate very well with that of the membrane pores. By placing an antiferromagnetic FeF2 layer underneath the Fe nanodots, an exchange anisotropy can be introduced into the Fe/FeF2 system. We have observed an increase in the magnetic hysteresis loop squareness in biased nanodots, suggesting that exchange bias may be used as a tunable source of anisotropy to stabilize the magnetization in such nanodots.

Abstract: Herein, we give an overview of several less explored structural and optical characterization techniques useful for biomaterials. New insights into the structure of natural fibers such as spider silk can be gained with minimal sample preparation. Electromagnetic radiation (EMR) over a broad range of wavelengths (from X-ray to THz) provides information of the structure of the material at correspondingly different length scales (nm-to-mm). When the sample features, such as the alignment of certain fibers, cannot be characterized optically, polarization analysis of the optical images can provide further information on feature alignment. The 3D complexity of biological samples necessitates that there be feature measurements and characterization over a large range of length scales. We discuss the issue of characterizing complex shapes by analysis of the link between the color and structure of spider scales and silk. For example, it is shown that the green-blue color of a spider scale is dominated by the chitin slab's Fabry-Pérot-type reflectivity rather than the surface nanostructure. The use of a chromaticity plot simplifies complex spectra and enables quantification of the apparent colors. All the experimental data presented herein are used to support the discussion on the structure-color link in the characterization of materials.

Abstract: The spindle pole body (SPB) is the equivalent of the centrosome in fission yeast. In vivo it nucleates microtubules (MTs) during mitosis, but, unlike animal centrosomes, does not act as a microtubule organizing center (MTOC) during interphase. We have studied the MT-nucleating activity of SPBs in vitro and have found that SPBs in permeabilized cells retain in vivo characteristics. SPBs in cells permeabilized during mitosis can nucleate MTs, and are recognized by two antibodies: anti-gamma-tubulin and MPM-2 which recognizes phosphoepitopes. SPBs in cells permeabilized during interphase cannot nucleate MTs and are only recognized by anti-gamma-tubulin. Interphase SPBs which cannot nucleate can be converted to a nucleation competent state by incubation in cytostatic factor (CSF)-arrested Xenopus egg extracts. After incubation, they are recognized by MPM-2, and can nucleate MTs. The conversion does not occur in Xenopus interphase extract, but occurs in Xenopus interphase extract driven into mitosis by preincubation with exogenous cyclin B. The conversion is ATP dependent and inhibited by protein kinase inhibitors and alkaline phosphatase. Purified, active, cdc2 kinase/cyclin B complex in itself is not effective for activation of MT nucleation, although some interphase SPBs are now stained with MPM-2. These results suggest that the ability of SPBs in vitro to nucleate MTs after exposure to CSF-arrested extracts is activated through a downstream pathway which is regulated by cdc2 kinase.

Abstract: To investigate the mechanisms of spindle elongation and chromosome separation in the fission yeast Schizosaccharomyces pombe, we have developed an in vitro assay using a temperature-sensitive mutant strain, nuc2. At the restrictive temperature, nuc2 cells are arrested at a metaphase-like stage with short spindles and condensed chromosomes. After permeabilization of spheroplasts of the arrested cells, spindle elongation was reactivated by addition of ATP and neurotubulin both at the restrictive and the permissive temperatures, but chromosome separation was not. This suggests that the nuc2 cells are impaired in function at a stage before sister chromatid disjunction. Spindle elongation required both ATP and exogenous tubulin and was inhibited by adenylyl imidodiphosphate (AMPPNP) or vanadate. The ends of yeast half-spindle microtubules pulse-labeled with biotinylated tubulin moved past each other during spindle elongation and a gap formed between the original half-spindles. These results suggest that the primary mechanochemical event responsible for spindle elongation is the sliding apart of antiparallel microtubules of the two half-spindles.
