Aim The study aimed to isolate and characterize Enterococcus species from apparently healthy waste attendants, cattle and cattle waste in Tanzania. Emphasis was given to antimicrobial resistance and in particular occurrence of vancomycin (VA)-resistant enterococci. Methods and results Faecal samples were collected from healthy cattle, cattle waste attendants and cattle house wastes, and isolation of Enterococcus species was performed using Slanetz Bartley agar. Isolates were characterized with regard to species, antimicrobial susceptibility and presence of VA resistance genes. Enterococcus faecalis was the most prevalent species from all sources of isolation (43·5%), followed by Enterococcus faecium (38·4%). Isolates of E. faecium showed a higher number of phenotypic antimicrobial resistance than isolates of E. faecalis. Fifty-eight isolates, which showed resistance or intermediate resistance to VA by disc diffusion test, were analysed for VA-resistant Enterococcus (VRE) by PCR. The vanA gene was detected in 14 isolates of E. faecium and 12 isolates of E. faecalis, while vanB was detected in three isolates. No isolates were found to carry vanC1-gene. Conclusion VRE was detected in both human and cattle samples, despite no known use of antimicrobial agents that can select for VRE in livestock in Tanzania. Enterococcus faecalis was the most commonly isolated species from cattle and humans. Significance and impact of the study The study provides information on the prevalence of VRE in human and nonhuman samples in Tanzania calling for further studies on the origin of VRE in such isolates, since no selection mechanism in Tanzania are known.

Introduction: Staphylococcus aureus is an opportunistic pathogen causing infections in humans and animals. Here we report for the first time the prevalence of nasal carriage, spa typing and antimicrobial resistance of S. aureus in a Tanzanian livestock community. Methodology: Nasal swabs were taken from 100 humans, 100 pigs and 100 dogs in Morogoro Municipal. Each swab was enriched in Mueller Hinton broth with 6.5% NaCl and subcultured on chromogenic agar for S. aureus detection. Presumptive S. aureus colonies were confirmed to the species level by nuc PCR and analysed by spa typing. Antimicrobial susceptibility patterns were determined by disc diffusion method. Results: S. aureus was isolated from 22 % of humans, 4 % of pigs and 11 % of dogs. A total of 21 spa types were identified: 13, 7 and 1 in human, dogs, and pigs, respectively. Three spa types (t314, t223 and t084) were shared between humans and dogs. A novel spa type (t10779) was identified in an isolate recovered from a colonized human. Antimicrobials tested revealed resistance to ampicillin in all isolates, moderate resistances to other antimicrobials with tetracycline resistance being the most frequent. Conclusion: S. aureus carrier frequencies in dogs and humans were within the expected range and low in pigs. The S. aureus spa types circulating in the community were generally not shared by different hosts and majority of types belonged to known clones. Besides ampicillin resistance, moderate levels of antimicrobial resistance were observed irrespective of the host species from which the strains were isolated.

In the current study, various haematological and biochemical parameters of chickens were evaluated following an oral administration of resin extract from Commiphora swynnertonii. Sixty chickens (8 months old) were randomly assigned into 5 groups (n=12): G1 served as a negative control, that is, chickens received normal saline only, while G2 to G5 chickens were given different doses of the resin extract orally for 14 consecutive days. Results revealed no signs of sickness or death in groups G1, G2 and G3 throughout the experimental period of 28 days. The body weights of chickens in G2, G3, G4 and G5 decreased significantly in a dose dependent manner from day 3 of treatment with the resin. The resin administration did not affected packed cell volume (PCV), total white blood cell (WBC) count, differential WBC count (heterophils and eosinophils). However, haemoglobin (Hb), total red blood cell (RBC) count, mean corpuscular hemoglobin (MCH) and mean corpuscular hemoglobin concentration (MCHC) decreased significantly (P < 0.05) with increasing concentration of the resin. Significant dose dependent reduction (P < 0.01) in plasma glucose and total cholesterol was observed. Levels of total protein, albumin and globulin in the resin-treated groups were not significantly affected. In the current study, liver transaminases (alanine transaminase (ALT) and aspartate aminotransferase (AST)) levels increased slightly to a maximum peak by day 21-post treatment. It is concluded that administration of the resin extract in chickens caused no undesirable effects at lower doses, but with increased doses signs of toxicity were evident. It is therefore suggested that, before the resin extract can be used for veterinary or medical purposes, safety margin must be established for each animal species in question. Urban and peri-urban livestock farming is expanding world-widely because of increased urbanization and demands for food of animal origin. Such farming practices pose a public health risk as livestock are reservoirs of several zoonotic pathogens. In an attempt to determine the fecal transmission between livestock and people, 100 household clusters keeping cattle in close proximity of humans were selected in urban and peri-urban areas of Morogoro in Tanzania. One hundred eighteen ampicillin and tetracycline resistant Escherichia coli (40 from human stool, 50 from cattle feces, 21 from soil and seven from water samples) were isolated from 44 different clusters. Pulsed-field gel electrophoresis (PFGE) of XbaI digested chromosomal DNA was used to compare the genetic relatedness of the ampicillin- and tetracycline-resistant E . coli isolates. Indistinguishable PFGE band patterns of the ampicillin- and tetracycline-resistant E. coli isolates were found in samples from 23 (52%) clusters. This suggests that transfer of fecal microorganisms between cattle, humans, water and soils within the farms and from livestock farms to the neighborhood occurred commonly. Logistic regression showed that animal housing infrastructures (Odd Ratio = 11.2, 95% CI = 1.1–119.3) were associated with E . coli showing identical PFGE types within and between clusters. There is a need to improve animal husbandry and manure management practices to reduce risks of transmission of enteropathogens between livestock and humans in urban and peri-urban farming.

Aim The aim of this study was to determine whether the practice of co-grazing with cattle and wild life constitutes a risk of transmission of antibiotic resistant bacteria to wild ungulates. Methods and Results Faecal samples were collected from buffalo (n = 35), wildebeest (n = 40), zebra (n = 40) and cattle (N = 20) from Mikumi National Park, Tanzania (MNP), where cattle is prohibited and from Ngorongoro Conservation Area (NCA) where co-grazing is practiced. The number of coliforms and enterococci resistant to selected antibiotics was determined. Wild life generally harboured higher number of resistant Escherichia coli and Enterococci than cattle, but with no general influence in wild life of co-grazing with cattle. Vancomycin-resistant Enterococci were detected in wild life samples, and E. coli resistant to cefotaxime and enrofloxacin were observed among isolates from all wild life, but not from cattle. Culture independent estimates of the number of sulII gene copies obtained by qPCR did not differ between wild life from the two sample sites, while tetW was significantly higher in samples from MPN than from NCA. Conclusions Antibiotic resistant bacteria were not more frequently found in ungulates grazing together with cattle than ungulates without this interaction. Significance and Impact of the Study This study did not indicate that transmission of antibiotic resistant bacteria is a frequent event following co-grazing of wild life and cattle.

This study provides an estimate of antimicrobial resistance in intestinal indicator bacteria from humans (n = 97) and food animals (n = 388) in Tanzania. While pathogenic types of Escherichia coli are well characterized, relatively little is known about the commensal E. coli flora. In the current study, antimicrobial resistance in commensal E. coli and distribution of ERIC-PCR genotypes among isolates of such bacteria from cattle and cattle attendants on cattle farms in Tanzania were investigated. Seventeen E. coli genomes representing different ERIC-PCR types of commensal E. coli were sequenced in order to determine their possible importance as a reservoir for both antimicrobial resistance genes and virulence factors. Both human and cattle isolates were highly resistant to tetracycline (40.8% and 33.1%), sulphamethazole-trimethoprim (49.0% and 8.8%) and ampicillin (44.9% and 21.3%). However, higher proportion of resistant E. coli and higher frequency of resistance to more than two antimicrobials was found in isolates from cattle attendants than isolates from cattle. Sixteen out of 66 ERIC-PCR genotypes were shared between the two hosts, and among these ones, seven types contained isolates from cattle and cattle attendants from the same farm, suggesting transfer of strains between hosts. Genome-wide analysis showed that the majority of the sequenced cattle isolates were assigned to phylogroups B1, while human isolates represented phylogroups A, C, D and E. In general, in silico resistome and virulence factor identification did not reveal differences between hosts or phylogroups, except for lpfA and iss found to be cattle and B1 phylogroup specific. The most frequent plasmids replicon genes found in strains from both hosts were of IncF type, which are commonly associated with carriage of antimicrobial and virulence genes. Commensal E. coli from cattle and attendants were found to share same genotypes and to carry antimicrobial resistance and virulence genes associated with both intra and extraintestinal E. coli pathotypes.

A study was conducted to evaluate the disease resistance potential in 105 chickens of six indigenous local chicken ecotypes in Tanzania by orally challenging 1-week-old chicks with 2.5 x 10(8) colony-forming units of virulent S. Gallinarum. For 14 days post infection, clinical signs, necropsy findings, antibody titres, packed cell volume, leukocyte population count, and viable bacterial cell counts in the liver and spleen were recorded. Clinical signs were recorded daily but other parameters were recorded on the day of infection, then on days 3, 6, 10 and 14 after infection. Clinical signs of fowl typhoid were evident in chickens from day 3 post infection and disappeared by day 9 post infection. Pathological lesions on sacrificed birds included enlargement of the liver and spleen with foci of necrosis on the liver, spleen and myocardium. The mean viable bacterial cell counts in the liver and spleen varied between ecotypes, although the differences were not statistically significant. There were significant differences in the leukocyte population in the peripheral blood, with one ecotype (Morogoro-medium) showing a consistent and significantly higher heterophil count compared with other ecotypes. It was concluded that there is a selectable resistance potential to S. Gallinarum among the local chicken ecotypes in Tanzania that may be attributable to non-specific host immune responses. Further studies are suggested.

The ethnoveterinary use of Aloe species extract in free-range local chickens against fowl typhoid was investigated. Five-months-old local chickens, free of antibodies against fowl typhoid were used. The chickens were randomly assorted into five groups including pretreated and infected (G1, n=21), infected and untreated (G2, n=21), infected and treated (G3, n=21), untreated and uninfected (G4, n=10) and treated uninfected (G5, n=10). Groups 1, 2 and 3 were inoculated with 5.0 x 10(8) c.f.u/ml of Salmonella gallinarum, following which the chickens were monitored for 15 days. There was a delay on the occurrence of the clinical signs and reduced severity of the disease in the Aloe treated chickens (G1 and G3). The mortality rates were 23.8% (5/21) in G1 (pretreated and infected), 42.8% (9/21) in G2 (infected and untreated) and 14.2% (3/21) in G3 (infected and treated). Antibody levels were raised among the infected and untreated group (G2) while they remained relatively low in the Aloe treated groups (G1 and G3). However, there was a sharp increase in the levels of interleukin 6 (IL-6) in the Aloe treated groups (G1 and G3) as compared to the untreated group (G2) until day 9 post-infection. The results of this work indicate that the extract of Aloe secundiflora may be used in the control of fowl typhoid in chickens. Studies to determine the active ingredients of the plant extract are in progress.

The pathogenicity of a virulent strain of Salmonella gallinarum was studied in local chickens and commercial layers. Sixty 4-month-old chickens were used. Of these, 50 chickens were orally infected with S. gallinarum, comprising 25 commercial layers and 25 local chickens. Five chickens in each group were used as uninfected negative controls. The productivity of local chickens under village management conditions was studied in six villages situated in three climatic zones within Morogoro District in Tanzania. Two villages were picked in each climatic zone (warm and wet, warm and dry, cool and wet) for the study. The data were obtained by actual measurement, qualitative observations and interview of members of the households directly responsible for the care of chickens. In addition, data sheets were given to selected farmers to record the performance of their chickens. The mean flock size for the three zones was 16.2, with a range of 2 to 58. The overall mean clutch size, egg weight and hatchability were 11.8, 44.1 g and 83.6%, respectively. The overall mean chick survival rate to 10 weeks of age was 59.7%. The mean live weights for cocks and hens were 1948 g and 1348 g, respectively. The mean growth rates to the age of 10 weeks were 4.6 g/day and 5.4 g/day, while those from 10 to 14 weeks of age were 8.4 g/day and 10.2 g/day for female and male birds, respectively. The age at first lay ranged between 6 and 8 months, and the average hen had three laying cycles per year. Most of the chickens were left to scavenge during the day and were provided with simple housing at night (95.2% of the owners). Only small amounts of supplementary feeds were occasionally given and minimal health care was provided. It was concluded that the low productivity of chickens was partly due to the prevailing poor management practices, in particular the lack of proper health care, poor nutrition and housing.

Escherichia coli strains such as Shiga toxin-producing E. coli (STEC), enteropathogenic E. coli, enterotoxigenic, attaching, and effacing E. coli, and enteroinvasive E. coli cause diarrhea in humans. Although other serotypes exist, the most commonly reported STEC in outbreaks is O157:H7. A cross-sectional study was conducted to isolate and characterize non-sorbitol-fermenting (NSF) E. coli O157:H7 from urban and periurban livestock settings of Morogoro, Tanzania. Human stool, cattle feces, and soil and water samples were collected. Observations and questionnaire interview studies were used to gather information about cattle and manure management practices in the study area. E. coli were isolated on sorbitol MacConkey agar and characterized by conventional biochemical tests. Out of 1049 samples, 143 (13.7%) yielded NSF E. coli. Serological and antimicrobial tests and molecular typing were performed to NSF E. coli isolates. These procedures detected 10 (7%) pathogenic E. coli including STEC (n=7), enteropathogenic E. coli (EPEC) (n=2), and attaching and effacing E. coli (A/EEC) (n=1) strains. The STEC strains had the ability to produce VT1 and different VT2 toxin subtypes that caused cytopathic effects on Vero cells. The prevalence of STEC in cattle was 1.6%, out of which 0.9% was serotype O157:H7 and the overall prevalence of diarrheagenic E. coli in cattle was 2.2%. The serotypes O157:H7, O142:H34, O113:H21, O+:H-, O+:H16, and O25:H4 were identified. One ESBL-producing isolate showed the MLST type ST131. To our knowledge, this is the first finding in Tanzania of this recently emerged worldwide pandemic clonal group, causing widespread antimicrobial-resistant infections, and adds knowledge of the geographical distribution of ST131. Cattle manure was indiscriminately deposited within residential areas, and there was direct contact between humans and cattle feces during manure handling. Cattle and manure management practices expose humans, animals, and the environment to pathogenic E. coli and other manure-borne pathogens. Therefore, there is a need to improve manure management practices in urban and periurban areas to prevent pathogen spread and associated human health risks.

A study was conducted to determine the variations in physical characters and immunocompetence among scavenging local chicken ecotypes in Tanzania. Eighty-four adult scavenging local chickens from four eco-climatic regions of Tanzania were studied. Measurements of adult body weight, body length, shank length and egg weight and observations of plumage colour and pattern, earlobe colour, skin colour and the shape of the comb were conducted. The antibody response to sheep red blood cells, serum haemolytic complement and the cutaneous response to phytohaemagglutinin-P were assessed. Five ecotypes were identified and named Mbeya, Morogoro-medium, Ching'wekwe, Kuchi and Singamagazi. Singamagazi and Kuchi were significantly heavier, with longer shanks and heavier eggs than the other ecotypes. The average adult body weight for males ranged from 1621 g (Mbeya) to 2915 g (Singamagazi). Average female weights ranged from 1108 g (Morogoro-medium) to 2020 g (Singamagazi). Mean egg weights ranged from 37.65 g (Ching'wekwe) to 45.61 (Singamagazi). The Kuchi had mostly rose and walnut combs, while the other ecotypes were mostly single combed. In each ecotype there were chickens with a high or low antibody response to red blood cells, but there was a significant difference between the ecotypes.

Coccidial oocysts were detected in 35% of 445 cattle in four medium-scale and 20 small-scale dairy farms in Morogoro municipality, Tanzania. The highest prevalence (56%) was observed in animals aged between 5 and 18 months, whereas lower prevalences were observed in calves (29%) aged between 12 days and 4 months and adults (30%). No coccidial oocysts were detected in calves less than 12 days old. The oocyst output was high in calves, followed by weaners; adults had the lowest oocyst output. The number of oocysts per gram of faeces was significantly higher (P < 0.001) in diarrhoeic animals than in non-diarrhoeic animals, and more so in young calves. Eimeria species infecting the animals included Eimeria bovis (68%) and Eimeria zuemii (57%), Eimeria ellipsoidalis (25%), Eimeria cylindrica (23%), Eimeria auburnensis (22%), Eimeria alabamensis (12%) and Eimeria subspherica (5%). Mixed infections involving two or three species were common. Our findings indicate that eimeriosis is common in cattle in Morogoro municipality.

Prevalences of Cryptosporidium spp. oocysts in cattle (n = 486) on five selected farms in Morogoro municipality and three species of herbivorous wildlife (n = 87) from Mikumi National Park, Morogoro, Tanzania, were determined using the modified Ziehl-Neelsen staining technique. Of 486 bovine faecal samples, 5.3% were positive for Cryptosporidium spp. The prevalence of Cryptosporidium was higher in calves less than 3 months of age compared to weaned calves and adults. Cryptosporidium spp. oocysts were detected in both diarrhoeic and non-diarrhoeic animals, but there was a significantly higher prevalence (P < 0.001) of oocyst shedding in diarrhoeic than in non-diarrhoeic animals. Of the 87 faecal specimens from the wildlife species, 36 were from the African buffaloes (Syncerus caffer), 25 from zebra (Equus zebra) and 26 from the wildebeest (Connochaetes gnou). Cryptosporidium spp. oocysts were detected in eight (22%) buffaloes, seven (28%) zebras and seven (27%) wildebeests. Confirmation of the diagnosis was performed using anti-Cryptosporidium monoclonal antibody specific for Cryptosporidium muris. Prevalences of Cryptosporidium parvum oocysts in faeces and of isotype-specific anti- C. parvum antibodies in serum of apparently healthy adult cattle on two farms were determined. On Farm 1 cryptosporidial diarrhoea had been recorded in more than 80% of calves born over the previous 5 years, whereas on Farm 2 cryptosporidiosis had never been reported. No differences were demonstrated in oocyst excretion or presence of antibodies between the two farms. C. parvum oocysts were detected in 62.4% of faecal smears collected from a total of 553 apparently healthy adult cattle. Sucrose flotation was performed on a proportion of the faecal samples. This proved a more sensitive technique, detecting oocysts in 92% of the samples tested, and highlighting the insensitivity of direct smears for detecting oocysts. More than 90% of the cattle had specific anti- C. parvum IgG, IgG 1 , IgG 2 and IgM antibodies and 58% specific anti- C. parvum IgA antibodies. Results suggest that asymptomatic adults may play an important role in the epidemiology of cryptosporidiosis in calves.

A crude resinous extract from Commiphora swynnertonii was tested against an experimental coccidial infection in local chickens. A total of 80 growing chickens were randomly assigned into five groups, which received different treatments. Chickens in G1 were not infected with coccidian oocysts and therefore served as a negative control. All chickens in G2, G3, G4 and G5 were infected through oral administration of coccidian oocysts suspension at a dosed rate of 1.5 × 10(4) Eimeria spp. oocysts per bird. Starting from day 3 post-infection (p.i), chickens in different groups were treated for 7 consecutive days as follows: G1 and G2 (positive control) received 5 ml of normal saline as placebo, G3 and G4 were given the extract at 400 and 800 mg/kg bodyweight whereas G5 received anticoccidial drug. Clinical signs, bodyweights, oocysts counts and mortality rates were observed regularly. Results showed that oral administration of the resinous extract to chickens with coccidiosis significantly reduced mortality rate from 94 to 25 % and oocysts counts from 1.03 × 10(5) to 6.55 × 10(3) oocysts/g faeces (p < 0.05). Also a body condition score chart indicated less severe clinical signs of the disease in the groups which received the extract. Mean daily body weights were slightly reduced by the administration of the extract but this effect disappeared by day 7 p.i. These findings clearly indicate that resinous extract from C. swynnertonii has significant anticoccidial effect against experimental Eimeria spp. infection in chickens. A larger field trial to validate the use of the extract in chickens naturally infected with Eimeria spp. is required.

Ethanolic extracts from resin, root bark, stem bark and leaves of Commiphora swynnertonii were tested against fungi and bacteria using agar well diffusion method. The fungi included Candida albicans and Aspergillus niger whereas the bacteria species included Staphylococcus aureus, Streptococcus pyogenes, Bacillus subtilis, Pseudomonas aeruginosa, Salmonella typhimurium and Escherichia coli . Antimicrobial activity was determined by measuring inhibition zone diameters around agar wells. Resin and root bark extracts showed significant activities against S. pyogenes, E. coli and B. subtilis compared to the other two extracts. Growth of E. coli was highly reduced by resin extract (minimum inhibition concentration = 1.9 µg/ml). The fungi, C. albicans , also showed similar sensitivity to resin and root bark extract. Growth of S. typhimurium was not reduced by all four extracts at all concentrations tested whereas that of P. aeruginosa was slightly reduced. Cytotoxicity studies using brine lethality test indicated that root bark, stem bark and resin extract had effect to brine shrimps with LC 50 of 3.5, 13.0 and 15.8 µg/ml, respectively. The current results indicate that resin and root bark extracts of C. swynnertonii have strong antimicrobial activity against most of the tested microbes and support the traditional use of the plant in treating various infectious diseases. Further studies are suggested to validate the use of this plant against the diseases caused by the tested microbes.

Studies were carried out to investigate the effect of crude extracts from resin, leaves, stem barks and root barks of Commiphora swynnertonii against Newcastle disease virus (NDV) using an in ovo assay. Nine-day-old embryonated chicken eggs were divided into seven groups (n = 6) and received various treatments. Six groups were inoculated with velogenic NDV strain; five groups out of these were treated with different concentrations of the four extracts or a diluent, dimethylsulphoxide. The uninoculated and inoculated groups were left as negative and positive controls, respectively. Embryo survival was observed daily and embryo weights were measured day 5 post-inoculation; a few eggs from selected groups were left to hatch. Allantoic fluid from treated eggs and serum from hatched chicks were collected for hemagglutination and hemagglutination inhibition (HI) tests to detect NDV in the eggs and antibodies against NDV in the hatched chicks respectively. Results showed that embryo survival and mean embryo weight were significantly higher (p < 0.001) in those groups which were treated with the crude extracts from C. swynnertonii than the positive control group. Also the extracts significantly (p < 0.001) reduced virus titres, whereas no viruses were detected in the allantoic fluids of the resin-treated group at the highest concentration of 500 μg/mL. Furthermore, the HI test results showed very low levels of antibodies against NDV in chicks hatched from resin and root bark extract-treated eggs suggesting that these plant materials were capable of destroying the NDV before stimulating the developing chick's immunity. The current findings have clearly demonstrated that crude extracts especially that of resin from C. swynnertonii have strong antiviral activity against NDV in ovo. In vivo trials are needed to validate the use of resin from the tree in controlling Newcastle disease in chickens.

Sleeping sickness is a re-emerging disease in the Serengeti ecosystem affecting both local people and tourists. Here we report the results of a survey to assess the prevalence of trypanosomiasis in both domestic and wild animals from this area. Five hundred and eighteen cattle samples were collected from 12 villages that bordered the Serengeti National Park and 220 samples from 15 different wild animal species were collected from within the park. PCR analysis, directed against the human serum resistance associated gene SRA, identified human infective Trypanosoma brucei rhodesiense parasites in both cattle and warthogs.

Two neonatal lambs were inoculated orally with purified Cryptosporidium species oocysts isolated from a farm cat. Oocysts first appeared in the faeces of the two lambs three and 10 days after infection. Two distinct sizes of oocysts were observed in the faeces of both the cat and the lambs, the smaller measuring approximately 5.0 x 4.5 microns and the larger measuring approximately 6.0 x 5.0 microns in diameter. The smaller type predominated. Histological examination of the alimentary tract of the lambs revealed endogenous stages of Cryptosporidium in the epithelial borders of the ileum. In addition, Cryptosporidium oocysts were detected in impression smears from the jejunum, ileum, caecum and colon. Suspensions of 10(3) oocysts from the faeces of the farm cat were inoculated into each of 10 newborn mice and 10(4) oocysts from the two experimentally infected lambs were inoculated into each of 20 newborn mice. Cryptosporidium oocysts were detected in gut homogenates from 19 of the 20 mice inoculated with oocysts from the lambs but in none of the mice inoculated with oocysts from the cat.

1. A total of 144 chickens purchased from peasants in Morogoro, Tanzania were slaughtered, their crops dissected and the contents analysed. The birds consisted of 48 chickens from each of three climatic zones (warm and wet, warm and dry, cool and wet). Seventy-two chickens were slaughtered in each season, that is short rainy and long rainy season. 2. Cereal grains, bran, green forages, insects/worms and kitchen food wastes were the main crop contents and their composition varied significantly with season and climate. 3. Two replicate experiments were carried out to verify the efficacy of Aloe species (Aloaceae) as used for the control of Newcastle disease (ND) in rural poultry in free-range systems among several communities in Tanzania. Four months old local chickens free of Newcastle disease antibodies were used. Following inoculation with ND virus, body weights, clinical signs, antibody levels and mortality were monitored. Results showed that there was reduced mortality rate and the severity of clinical signs during the acute phase of the infection in Aloe treated chickens compared with the non-treated ones. However, there was no significant effect of the Aloe on the antibody levels that were attributed to the recovery of the surviving chickens. The findings of this study suggest that Aloe secundiflora could be a potential candidate on the management of Newcastle disease in chickens. Further studies are in progress to identify the active ingredients of A. secundiflora against Newcastle disease virus.

Free-ranging local chicken flocks are important for the livelihood of resource-poor rural farmers in Tanzania, as they provide a critical source of animal protein and a ready source of income through the sale of chickens and eggs. An occurrence of highly pathogenic avian influenza (HPAI) in the village setting of Tanzania would result in a disastrous loss of livelihood. This paper attempts to offer an alternative method for preventing and controlling HPAI in village settings of Tanzania through community-based approaches.

Prophylactic and therapeutic efficacy of a combination of Capsicum frutescens (red pepper), Citrus limon (lemon) and Opuntia vulgaris (prickly pear) against Newcastle disease (ND) in domestic fowl were evaluated. Eighty-eight broiler chickens were divided into five groups. Birds from three groups were inoculated with velogenic ND virus strain, whereas birds from two groups were left as controls. Two groups received a mixture of the plant extract three days prior to inoculation and birds from one group were given the plant extract for two days following development of clinical signs. Blood samples were collected for haemaglutination inhibition tests (HI) for detection of ND virus antibodies. Body weights were monitored during the experiment. Three birds died from the group that was inoculated with ND virus and treated with the plant extract; two died from the group that received the plant extract as a prophylaxis and inoculated with ND virus; and one bird died from the group that was inoculated with ND virus but not given the plant extract. No death was observed in any of the birds in the control groups. Antibody titers for ND virus rose four-fold in the inoculated birds but remained low in the un-inoculated groups. Mean body weights of birds in group B declined markedly compared to the other groups. The results indicated that there was no prophylactic or therapeutic value of the plant extract against ND. The plant extract showed a negative effect on body weights in birds with ND.

We investigated the prevalence, burden and types of gastro-intestinal helminths in 424 local and cross-bred pigs kept under different management systems in two climatic zones in the Morogoro region of Tanzania. Coprological examination revealed that 53% of the pigs excreted helminth eggs in their faeces. The median eggs per gram of faeces (EPG) was 500 (range 100–22000). Local breeds in the Mgeta location with tropical highland climate showed significantly higher prevalence (P < 0.001) and median EPG values (P < 0.001) than the cross-bred animals in the semi-arid area. There was no significant difference in the prevalence (P = 0.90) of helminth infection and egg outputs (P = 0.78) in cross-bred pigs raised under the small-scale and semi-intensive management systems in the semi-arid zone. Piglets showed significantly lower prevalence of helminthosis (P < 0.001) than the weaners, growers and adults in both local and cross-bred animals. Median EPGs of growers and adult animals were significantly higher than those of piglets and weaners (P = 0.006). The prevalences of various helminth species were Oesophagostomum sp. (40%), Ascaris suum (12%), Strongyloides ransomi (9%) and Trichuris suis (5%).

Sera from 258 healthy and sick domestic and feral cats were screened for specific anti-Cryptosporidium antibodies using an indirect immunofluorescence antibody test (IFA). Sera were positive for IgG, IgM and IgA antibodies in 192 (74%), 84 (32%) and 67 (26%) samples, respectively. Antibody was not detected at dilutions of 1:10 and 1:20 or greater in any of eight specific pathogen-free kittens. IgM and IgA antibody classes were more prevalent in sick than in healthy domestic cats. The presence of IgM and/or IgA antibodies indicated early infection. However, these antibody classes were present in sera from cats either positive or negative for Cryptosporidium infection by faecal examination. Pronounced polar fluorescence was observed in the sporozoites in positive samples under fluorescence microscopy. The higher prevalence of specific anti-Cryptosporidium antibodies and the absence of Cryptosporidium oocysts in faecal samples from some IFA-positive animals suggests that detection of these antibodies in sera from cats could be helpful for the diagnosis of feline cryptosporidiosis.

Modified Ziehl-Neelsen (MZN), auramine-phenol (A-P) and fluorescein isothiocyanate-labelled (FITC-labelled) monoclonal antibody (MAb) techniques were compared for detection of Cryptosporidium parvum oocysts in cat faecal specimens inoculated with known numbers of C. parvum oocysts. Of the three techniques, the FITC-labelled MAb technique detected more oocysts than the MZN and A-P techniques (P < 0.05), but A-P was more efficient than MZN (P < 0.05). Comparison of sucrose flotation, zinc sulphate (ZnSO4) flotation and formol-ether (F-E) sedimentation techniques revealed that F-E was the most effecient of the three (P < 0.05) for concentration of C. parvum oocysts from cat faecal specimens. On average, the F-E technique recovered 37% of oocysts from the original sample, whereas the sucrose and ZnSO4 flotation techniques recovered 33% and 11%, respectively. The findings of this study suggest that MZN and A-P staining are both useful for screening C. parvum oocysts in cat faecal materials containing 106 oocysts or more, but FITC-labelled MAb should be used when the number of oocysts is low. Also, the F-E sedimentation technique is recommended for concentrating oocysts in cat faecal specimens.

A social strategy was tested for implementing Newcastle disease (ND) vaccination and biosecurity improvements among free-ranging chicken at village level in Tanzania. In addition to training the local poultry vaccinators, data recorders and poultry-keepers, the strategy involved training and empowering leaders at the district, ward and village level. The trainings covered poultry health, management, and marketing of village chickens, with an emphasis on ND vaccination and improving biosecurity against avian influenza (AI), The study sites included villages in one ward in each of three each three districts (Iringa, Mtwara-Mikindani, and Mvomero) of mainland Tanzania. Ninety-six local leaders at district level and 101 leaders at ward levels were trained. In addition, 196 farmers (households) were trained, as well as 86 vaccinators and 26 data recorders. Data recorders were also trained as poultry first aid workers. ND vaccination was conducted by the vaccinators, supervised by their local leaders with technical assistance from veterinarians. A total of 158,343 village chickens were vaccinated in three rounds of vaccination three months apart. The training and empowerment of local leaders and local implementers was the key element for success as it fostered the feeling of local ownership of the program and prevented conflicts with other development activities within the villages. We conclude that most animal health programs will increase their odds of success by involving local leaders and by addressing the current challenges facing the farmers. Further assessment on the usefulness of this approach is needed.

Investigation was done to determine the presence of Pasteurella multocida and related species in free ranging chickens and ducks, dogs, cats and pigs in three climatic zones (cool, warm and hot) of rural Morogoro, Tanzania. A total of 153 isolates of P. multocida ssp. multocida and related species were obtained by direct culture on blood agar, selective medium and mouse inoculation. P. multocida ssp. multocida was isolated from 0.7% of chickens and 7% of ducks. In dogs and cats, P. multocida ssp. multocida was isolated from 1 and 68%, respectively. One isolate of Pasteurella gallinarum was isolated from a duck. Other species obtained were; P. multocida ssp. septica, Pasteurella stomatis and taxon 16 from dogs and cats, while Pasteurella dagmatis and Pasteurella canis were found in dogs only.
