Abstract

Mesenchymal stem cells (MSCs) isolated from several adult human tissues are reported to be a promising tool for regenerative medicine. In order to broaden the array of tools for therapeutic application, we isolated a new population of cells from adult human testis termed gonadal stem cells (GSCs). GSCs express CD105, CD166, CD73, CD90, STRO-1 and lack hematopoietic markers CD34, CD45, and HLA-DR which are characteristic identifiers of MSCs. In addition, GSCs express pluripotent markers Oct4, Nanog, and SSEA-4. GSCs propagated for at least 64 population doublings and exhibited clonogenic capability. GSCs have a broad plasticity and the potential to differentiate into adipogenic, osteogenic, and chondrogenic cells. These studies demonstrate that GSCs are easily obtainable stem cells, have growth kinetics and marker expression similar to MSCs, and differentiate into mesodermal lineage cells. Therefore, GSCs may be a valuable tool for therapeutic applications.

Abstract

Surround cinema and 3D stereoscopy represent the future of the multimedia experience. They place the viewer in the middle of the action by increasing the immersion and interaction at a level people could only dream of a few years ago. Attend a live event or simply having the possibility to open a window to another location is possible by streaming and making the experience fully immersive is the novelty. In this paper we present a method we developed to solve the numerous technical challenges hidden behind the streaming of 3D stereoscopy immersive content, from acquisition to display: combination of several cameras, merge of the signals, synchronization of multiple sources and setting up of a surround cinema. Our proof of concept installation is made of two cameras equipped with fisheye lenses, streaming over LAN network to a digital dome.

Abstract

We report here our results in a multi-sensor setup reproducing the conditions of an automated focused ultrasound surgery environment. The aim is to continuously predict the position of an internal organ (here the liver) under guided and non-guided free breathing, with the accuracy required by surgery. We have performed experiments with 16 healthy human subjects, two of those taking part in full-scale experiments involving a 3 Tesla MRI machine recording a volume containing the liver. For the other 14 subjects we have used the optical tracker as a surrogate target. All subjects where volunteers who agreed to participate in the experiments after being thoroughly informed about it. For the MRI sessions we have analyzed semi-automatically offline the images in order to obtain the ground truth, the true position of the selected feature of the liver. The results we have obtained with continuously updated random forest models are very promising, we have obtained good prediction-target correlation coefficients for the surrogate targets (0.71 ± 0.1) and excellent for the real targets in the MRI experiments (over 0.91), despite being limited to a lower model update frequency, once every 6.16 seconds.

Abstract

Research Question: Can federated learning be used to develop an artificial intelligence (AI) model for evaluating oocyte competence using two-dimensional images of denuded oocytes in metaphase II prior to intracytoplasmic sperm injection (ICSI)?, Results: The oocyte AI model demonstrated area under the curve (AUC) up to 0.65 on two blind test datasets. High sensitivity for predicting competent oocytes (83-88%) was offset by lower specificity (26-36%). Exclusion of confounding biological variables (male factor infertility and maternal age ≥35 years) improved AUC up to 14%, primarily due to increased specificity. AI score correlated with size of the zona pellucida and perivitelline space, and ooplasm appearance. AI score also correlated with blastocyst expansion grade and morphological quality. The sum of AI scores from oocytes in group culture images predicted the formation of two or more usable blastocysts (AUC 0.77)., Conclusion: An AI model to evaluate oocyte competence was developed using federated learning, representing an essential step in protecting patient data. The AI model was significantly predictive of oocyte competence, as defined by usable blastocyst formation, which is a critical factor for IVF success. Potential clinical utility ranges from selective oocyte fertilization to guiding treatment decisions regarding additional rounds of oocyte retrieval., Design: In total, 10,677 oocyte images with associated metadata were collected prospectively by eight IVF clinics across six countries. AI training used federated learning, where data were retained on regional servers to comply with data privacy laws. The final AI model required a single image as input to evaluate oocyte competence, which was defined by the formation of a usable blastocyst (≥expansion grade 3 by day 5 or 6 post ICSI)., (Copyright © 2024 The Authors. Abstract

Objective: To develop a simplified polymerase chain reaction (PCR) protocol on single cells for the purpose of preimplantation genetic diagnosis. Also to evaluate a new thermal cycler, RoboCycler 40 (Stratagene, La Jolla, CA), for reducing the time to complete PCR amplification., Design: PCR amplification without DNA purification or reamplification of a 149 base pair (bp) segment of the human Y chromosome was used as a model. The assay was tested in human fetal cells, single lymphocytes and single human blastomeres., Results: Amplification of the 149 bp segment using fetal cells was 100% correct. Results on single lymphocytes were concordant in all but one of the 15 male cases. However, 2 of the 25 female cases were identified as male suggesting the occurrence of DNA contamination. Analysis of 61 blastomeres were concordant in 57 cases (93%); results for male blastomeres showed 12% of false negatives. No false positives were detected for female cells. Amplification using the simplified PCR protocol in combination with the RoboCycler was completed in 2 hours., Conclusion: These data show that this PCR assay performed directly, without DNA extraction or purification and without re-amplification is a practical and effective approach for amplification of specific DNA sequences in single cells. Furthermore, the simplified PCR protocol significantly reduced the time to complete DNA amplification. The reduced time is expected to facilitate the management of a routine program for preimplantation genetic diagnosis.
