The invasive rust Austropuccinia psidii, responsible for myrtle rust disease, poses a serious threat to the New Zealand Myrtaceae. Since the 2017 detection of Austropuccinia psidii in Aotearoa / New Zealand, the rust has spread rapidly, resulting in the decline and death of a range of indigenous Myrtaceae, most notably the two species of the endemic genus Lophomyrtus, ramarama (L. bullata) and rōhutu (L. obcordata). While the threat Austropuccinia psidii poses to Lophomyrtus is now widely recognised, the indirect impact the rust has on the associated biota is poorly understood. Very little has been documented about the biota found in association with Lophomyrtus. To rectify this, we undertook a survey of the specimens held in three of the key Aotearoa / New Zealand herbaria that had been collected from Lophomyrtus. This was supplemented by field work in eight sites in western Te Ika a Maui / North Island, and north-western Te Wai Pounamu / South Island of Aotearoa / New Zealand. Although the herbarium searches located few specimens, and field work was limited to a few sample points within the range of Lophomyrtus, we found 221 taxa associated with Lophomyrtus, 176 taxa on ramarama, 81 on rōhutu and one on the naturally occurring hybrid between these two species Lophomyrtus ×ralphii. Of the 176 taxa found on ramarama, 59 are bryophytes (one hornwort, 33 liverworts and 25 mosses), five pteridophytes, 16 spermatophytes and 96 are lichenised mycobiota. Rōhutu supported 81 taxa: comprising one cyanobacterium, one alga, twenty-nine bryophytes (17 liverworts and 12 mosses), four pteridophytes, two spermatophytes and 44 lichenised mycobiota. Wild populations of Lophomyrtus ×ralphii were not investigated, and herbarium searches only disclosed one plant, the mistletoe Korthalsella lindsayi, associated with it. Several lichens and liverworts collected from Lophomyrtus represent potentially new species, and Lepra erythrella is a new addition to the lichenised mycobiota of Aotearoa / New Zealand. None of the putative new species are endemic to Lophomyrtus.

Drones, or unoccupied aircraft systems (UAS), can transform the way scientific information on wildlife populations is collected. UAS surveys produce accurate estimates of ground-nesting seabirds and a variety of waterbirds, but few studies have examined the trade-offs of this methodology for counting cliff-nesting seabirds. In this study, we examined how different UAS survey parameters might influence seabird counts for population monitoring and assessed behavioral responses to aerial surveys for three sub-Arctic seabird taxa in the Gulf of Alaska: common murres (Uria aalge), black-legged kittiwakes (Rissa tridactyla), and pelagic and double-crested cormorants (Phalacrocorax pelagicus and Phalacrocorax auritus). We flew two commercially available models of UAS in planned approaches at different speeds and distances from colonies during incubation and chick-rearing periods. We compared counts from UAS-derived images with those from vessel-based photography and assessed video recordings of individual birds’ behaviors for evidence of disturbance during UAS operations and control phases. Count estimates from UAS images were similar to or higher than those from conventional vessel-based images, and UAS were particularly effective at photographing birds at sites with high cliff walls or complex topography. We observed no significant behavioral responses to the UAS by murres or cormorants, but we did observe flushing by black-legged kittiwakes during UAS flights; most of these birds were not incubating or brooding. At both the colony and individual level, we observed slightly greater responses to the smaller UAS platform and closer approaches. These results inform both species specific and general best practices for research and recreational usage of UAS near cliff-nesting seabird colonies.

Enhancing social inclusion in young people could increase engagement in education, yet few longitudinal studies have examined this relationship. This study aimed to identify whether social inclusion in an Australian adolescent sample predicted high school completion three years later. Using state-representative data from the International Youth Development Study, two waves of the youngest cohort (51.6% female and 94.6% Australian born) during mid-adolescence (n = 825, Mage = 15.99, SD = 0.39) and post-secondary school (n = 809, Mage = 19.03, SD = 0.44) were analyzed. Factor analysis identified a 4-factor structure that represented an overarching social inclusion construct: (1) Citizenship, (2) Connectedness to Community, (3) Connectedness to Family, and (4) Connectedness to and Participation in School. Multivariate regression analyses indicated higher social inclusion levels in mid-adolescence predicted an increased likelihood of high school completion three years later. The implementation of strategies that incorporate the enhancement of social inclusion may improve educational outcomes for young people.

Morphological and single-nucleotide polymorphism data support splitting Pultenaea glabra Benth. into eight species, including one in Victoria, and seven in eastern and northern New South Wales. Six species are newly described, five of which are, like P. glabra, narrow-range endemics within the Greater Blue Mountains World Heritage Area and adjacent sandstone landforms of the Great Dividing Range. The recognition of six new species from what was broadly P. glabra has implications for conservation management, including for P. glabra itself, which has a smaller distribution and more precise habitat requirements than previously thought. One of the new species, P. percussa, is known by a single 1971 gathering only. The occurrence of several narrow-range Blue Mountains endemic Pultenaea species may be explained by the combination of edaphic diversity and topographic complexity, which could act in concert to promote divergences among small, allopatric populations. Hybrids between P. glabra and P. flexilis Sm. are documented for the first time; however, limited evidence for introgression between the two species was observed. All forms of family violence may negatively affect a child’s development. However, research on child maltreatment is primarily focused on the child who is directly maltreated and does not often account for how other children in the family experience the abuse. The central aim of our study was to better understand how children’s direct experience of physical abuse and exposure to physical abuse influence their academic outcomes. Data were taken from the Minnesota Departments of Education and Human Services. The sample was developed from a population-level cohort of 8–10 years old children ( N = 1740) from two groups: Child Protective Service (CPS)-involved (a child who allegedly experienced physical abuse or a child who was exposed to the alleged physical abuse of another child in their household) and the matched comparison. Exposure to intimate partner violence (IPV) was also measured for CPS-involved children. School attendance and academic achievement were examined over 4 years. Descriptive statistics and Generalized Estimating Equations (GEE) were used to answer the three research questions. Over time, declines in attendance for children exposed to physical abuse were significantly greater than those of their matched peers. Exposure to IPV for CPS-involved children resulted in further declines in attendance. Math proficiency of children who experienced physical abuse declined at a significantly faster rate than their matched peers. The decline in reading proficiency of both children who experienced physical abuse and children exposed to physical abuse was more significantly pronounced than that of their matched peers. Differences in math and reading proficiency were eliminated when IPV exposure was taken into account. Child protection workers and school professionals should be aware of negative effects of experiences of and exposures to child maltreatment and work collaboratively to provide academic support, counseling, and other interventions to support children’s academic stability.

The huge worldwide demand for vaccines targeting SARS-CoV-2 has necessitated the continued development of novel improved formulations capable of reducing the burden of the COVID-19 pandemic. Herein, we evaluated novel protein subunit vaccine formulations containing a resistin-trimerized spike antigen, SmT1. When combined with sulfated lactosyl archaeol (SLA) archaeosome adjuvant, formulations induced robust antigen-specific humoral and cellular immune responses in mice. Antibodies had strong neutralizing activity, preventing viral spike binding and viral infection. In addition, the formulations were highly efficacious in a hamster challenge model reducing viral load and body weight loss even after a single vaccination. The antigen-specific antibodies generated by our vaccine formulations had stronger neutralizing activity than human convalescent plasma, neutralizing the spike proteins of the B.1.1.7 and B.1.351 variants of concern. As such, our SmT1 antigen along with SLA archaeosome adjuvant comprise a promising platform for the development of efficacious protein subunit vaccine formulations for SARS-CoV-2.

APOBEC3 (A3) proteins are host-encoded deoxycytidine deaminases that provide an innate immune barrier to retroviral infection, notably against HIV-1. Low levels of deamination are believed to contribute to the genetic evolution of HIV-1, while intense catalytic activity of these proteins can induce catastrophic hypermutation in proviral DNA leading to near-total HIV-1 restriction. So far, little is known about how A3 cytosine deaminases might impact HIV-1 proviral DNA integration sites in human chromosomal DNA. Using a deep sequencing approach, we analyze the influence of catalytic active and inactive APOBEC3F and APOBEC3G on HIV-1 integration site selections. Here we show that DNA editing is detected at the extremities of the long terminal repeat regions of the virus. Both catalytic active and non-catalytic A3 mutants decrease insertions into gene coding sequences and increase integration sites into SINE elements, oncogenes and transcription-silencing non-B DNA features. Our data implicates A3 as a host factor influencing HIV-1 integration site selection and also promotes what appears to be a more latent expression profile.

Women's social experiences can have long-term implications for their offspring's health, but little is known about the potential independent contributions of multiple periods of stress exposures over time. This study examined associations of maternal exposure to adversity in childhood and pregnancy with children's anxiety and depression symptoms in a large, sociodemographically diverse sample. Participants were 1389 mother-child dyads (child age M = 8.83 years; SD = 0.66; 42% Black, 42% White; 6% Hispanic) in the ECHO-PATHWAYS Consortium's three U.S. pregnancy cohorts. Women reported their exposure to childhood traumatic events (CTE) and pregnancy stressful life events (PSLE). Children self-reported on their symptoms of anxiety and depression at age 8-9 years. Regression analyses estimated associations between maternal stressors and children's internalizing problems, adjusting for confounders, and examined child sex as a modifier. Exploratory interaction analyses examined whether geospatially-linked postnatal neighborhood quality buffered effects. Background Community paramedicine programs (i.e., physician-directed preventive care by emergency medical services personnel embedded in communities) offer a novel approach to community-based health care. Project Swaddle, a community paramedicine program for mothers and their infants, seeks to address (directly or through referrals) the physical, mental, social, and economic needs of its participants. The objective of this process evaluation was to describe women’s experiences in Project Swaddle. By understanding their experiences, our work begins to build the foundation for similar programs and future examinations of the efficacy and effectiveness of these approaches. Methods We completed 21 interviews with women living in Indiana (July 2019–February 2020) who were currently participating in or had graduated from Project Swaddle. Interviews were audio-recorded, transcribed, and analyzed using a six-phase approach to thematic analysis. Results Program enrollment was influenced by the community paramedics’ experience and connections, as well as information received in the community from related clinics or organizations. Participants viewed the community paramedic as a trusted provider who supplied necessary health information and support and served as their advocate. In their role as physician extenders, the community paramedics enhanced patient care through monitoring critical situations, facilitating communication with other providers, and supporting routine healthcare. Women noted how community paramedics connected them to outside resources (i.e., other experts, tangible goods), which aimed to support their holistic health and wellbeing. Conclusions Results demonstrate Project Swaddle helped women connect with other healthcare providers, including increased access to mental health services. The community paramedics were able to help women establish care with primary care providers and pediatricians, then facilitate communication with these providers. Women were supported through their early motherhood experience, received education on parenting and taking control of their health, and gained access to resources that met their diverse needs.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has killed over 6 million individuals worldwide and continues to spread in countries where vaccines are not yet widely available or its citizens are hesitant to become vaccinated. Therefore, it is critical to unravel the molecular mechanisms that allow SARS-CoV-2 and other coronaviruses to infect and overtake the host machinery of human cells. Coronavirus replication triggers endoplasmic reticulum (ER) stress and activation of the unfolded protein response (UPR), a key host cell pathway widely believed to be essential for viral replication. We examined the master UPR sensor IRE1α kinase/RNase and its downstream transcription factor effector XBP1s, which is processed through an IRE1α-mediated mRNA splicing event, in human lung-derived cells infected with betacoronaviruses. We found that human respiratory coronavirus OC43 (HCoV-OC43), Middle East respiratory syndrome coronavirus (MERS-CoV), and murine coronavirus (MHV) all induce ER stress and strongly trigger the kinase and RNase activities of IRE1α as well as XBP1 splicing. In contrast, SARS-CoV-2 only partially activates IRE1α through autophosphorylation, but its RNase activity fails to splice XBP1. Moreover, while IRE1α was dispensable for replication in human cells for all coronaviruses tested, it was required for maximal expression of genes associated with several key cellular functions, including the interferon signaling pathway, during SARS-CoV-2 infection. Our data suggest that SARS-CoV-2 actively inhibits the RNase of autophosphorylated IRE1α, perhaps as a strategy to eliminate detection by the host immune system.

Using our strongly immunogenic SmT1 SARS-CoV-2 spike antigen platform, we developed antigens based on the Beta & Delta variants of concern (VOC). These antigens elicited higher neutralizing antibody activity to the corresponding variant than comparable vaccine formulations based on the original reference strain, while a multivalent vaccine generated cross-neutralizing activity in all three variants. This suggests that while current vaccines may be effective at reducing severe disease to existing VOC, variant-specific antigens, whether in a mono- or multivalent vaccine, may be required to induce optimal immune responses and reduce infection against arising variants.

Since 2016, abortion care has undergone several important changes, particularly related to the provision of medical abortion using mifepristone. We aimed to document characteristics of the abortion care workforce in Canada after the update of clinical practice guidelines of mifepristone use for medical abortion.We conducted a national, web-based, anonymized, bilingual (English/French) survey. We collected demographics and clinical care characteristics of physicians and nurse practitioners who provided abortion care in 2019. Between July and December 2020, we distributed the survey through professional organizations, including The College of Family Physicians of Canada and The Society of Obstetricians and Gynaecologists of Canada. We present descriptive statistics.Overall, 465 respondents representing all 10 provinces and 3 territories in Canada completed the survey. Of these, 388 (83.4%), including 30 nurse practitioners, provided first-trimester medical abortion, of which 350 (99.4%) used mifepristone. Two hundred and nineteen (47.1%) respondents provided first-trimester surgical abortion, 109 (23.4%) provided second-trimester surgical abortion and 115 (24.7%) provided second- or third-trimester medical abortion. Half of respondents reported fewer than 5 years of experience with any abortion care. Respondents reported providing a total of 48 509 abortions in 2019, including 32 345 (66.7%) first-trimester surgical abortions and 13 429 (27.7%) first-trimester medical abortions. In Quebec, only 1918 (12.5%) of reported abortions were first-trimester medical abortions. Primary care providers provided 34 540 (71.2%) of the total abortions. First-trimester medical abortions represented 44.4% (in/i= 2334) of all abortions in rural areas, as opposed to 25.6% (in/i= 11 067) in urban areas.The increased availability of medical abortion facilitates abortion access, especially in primary care and rural settings, and where surgical abortion is not available. Rejuvenation of the workforce is a critical contributor to equitable access to abortion services.

Summary Regional and local studies suggest that the Tufted Puffin Fratercula cirrhata in North America is declining in portions of its range. However, whether the overall population is declining, or its range is contracting with little change to the overall population size, is unknown. To examine population trends throughout its North American range, we assembled 11 datasets that spanned 115 years (1905–2019) and included at-sea density and encounter estimates and at-colony burrow and bird counts. We assessed trends for the California Current, Gulf of Alaska, and Bering Sea/Aleutian Islands large marine ecosystems (LME). We found: (1) nearly uniform and long-term declines of Puffins breeding in the California Current ecosystem, with most ecosystem colonies surveyed, (2) declining trends at two large colonies and in one at-sea dataset in the Gulf of Alaska LME, with the fourth smaller colony exhibiting no significant trend, and (3) positive trends at four out of five colonies in the Bering Sea/Aleutian Islands ecosystem complex, with no detectable trend at the fifth very large colony. The general pattern of Tufted Puffin declines across the California Current and Gulf of Alaska LMEs may be attributable to a variety of factors, but additional study is needed to evaluate the relative influence of potential population drivers both independently and synergistically. Potential mechanisms driving population increases in the Bering Sea/Aleutian Islands ecosystem include reduced depredation and bycatch, intrinsic population growth, and immigration. We found strong evidence for declines in two of the three LMEs evaluated representing approximately three quarters of the species’ North American range. This region of decline includes the Gulf of Alaska LME, which contains a significant portion of the species’ estimated total North American population. Despite data limitations, our analysis coupled with more focused and local studies indicates that the Tufted Puffin is a species of conservation concern.

In this study, we examined changes in mothers’ reports of discipline, nurturing, and parenting satisfaction, along with their perceptions of how intimate partner violence (IPV) affected their role as a mother. Data were gathered from 85 women (86% identified as non-Hispanic White; mean age = 31.7 years; average number of children = 2.5) across three data waves. We used a convergent parallel mixed method design to examine women’s parenting experiences. Two standardized scales assessed parenting discipline, nurturance, and satisfaction. Five additional items assessed perceptions of how IPV influenced parenting. Women responded to open-ended questions about perceptions of their mothering role and how IPV influenced this role over time. We found significant changes in discipline scores and women’s ability to have their desired relationship with their children. Nurturing and parenting satisfaction scores did not significantly change over time. Four themes emerged for women’s role as mothers: provider and nurturer, teacher and discipliner, challenges, and importance of the mother role. Seven themes emerged on how IPV influenced mothering: ability to be present, trouble with discipline, overcompensation and overprotective, learned behavior, no impact, finding strength, and self-doubt. These findings contribute to the growing scholarship assessing women’s perceptions of their mothering roles and parenting in the context of IPV. Future research should examine the impact and nuances of parental interference in various contexts and over time.

Using the Intimate Partner Violence Strategies Index (IPVSI), we assessed women’s strategy use and the helpfulness of these strategies in preventing, stopping, or coping with intimate partner violence (IPV). We further examined which strategies women found most and least helpful and why, associations between strategy use and types of IPV experienced, and differences in strategy use and helpfulness by select demographic variables. We used data from Wave 1 of a two-year panel study of women who experienced IPV victimization and received civil legal services. The IPVSI includes 39 strategies across six categories: formal network, legal, safety planning, informal network, resistance, and placating. We collected data on women’s perceptions of the most and least helpful strategies and why, and whether they used other strategies not listed on the IPVSI. We used descriptive, bivariate, and content analyses to answer the research questions. Women used an average of 21 strategies. Placating, resistance, informal network, and legal strategies were the most frequently used; however, women rated resistance and placating strategies as least helpful and informal network strategies as most helpful. IPV types (physical, nonphysical) were differentially associated with the number and types of strategies used. Women expressed why strategies were helpful or unhelpful, along with what other strategies they used. Models of IPV help-seeking have evolved from viewing women as passive to seeing them as actively engaging in many private and public strategies to manage, prevent, and escape violence. Our findings highlight some discrepancies between what is used and what works.

Researchers continue to demonstrate the significant physical and mental health consequences to women who experience intimate partner violence (IPV) and children exposed to IPV. However, more research is needed to examine group interventions that support mothers, their children, and the parent-child relationship in the aftermath of IPV. We analyzed data provided by women who participated in a 12-week parent group. The objectives of the parent group were for mothers to better understand their children’s developmental experience of IPV and to break the secret of domestic violence in their family. The sample included 15 women who completed the group and the baseline, midpoint, and endpoint surveys. Eleven of these women also completed a semi-structured interview three months following the group. Survey items and interview topics focused on group process and outcomes. Women reported the group helped them feel less alone in their experience of IPV and they valued having a space to learn and discuss new concepts that related to their experiences and their children’s experiences of IPV. They reported more confidence in identifying their child’s feelings and being a safe person to whom their children could express their anger. Women also reported they developed greater coping and communication tools, and a strengthened mother-child bond, through group participation. Group interventions are an important format for learning and sharing for some mothers who have experienced IPV, though more evaluation is needed on what components of these group interventions are the most supportive of mothers’ healing and growth.

Other common coronaviruses such as HCoV-OC43 cause less severe respiratory disease. Thus, it is imperative to understand the similarities and differences among these viruses in how each interacts with host cells. We focused here on the inositol-requiring enzyme 1α (IRE1α) pathway, part of the host unfolded protein response to virus-induced stress. We found that while MERS-CoV and HCoV-OC43 fully activate the IRE1α kinase and RNase activities, SARS-CoV-2 only partially activates IRE1α, promoting its kinase activity but not RNase activity. Based on IRE1α-dependent gene expression changes during infection, we propose that SARS-CoV-2 prevents IRE1α RNase activation as a strategy to limit detection by the host immune system.

While the family Schizaeaceae (Schizaeales) represents only about 0.4% of the extant fern species diversity, it differs from other ferns greatly in gross morphologies, niche preferences, and life histories. One of the most notable features in this family is its mycoheterotrophic life style in the gametophytic stage, which appears to be associated with extensive losses of plastid genes. However, the limited number of sequenced plastomes, and the lack of a well-resolved phylogenetic framework of Schizaeaceae, makes it difficult to gain any further insight. Here, with a comprehensive sampling of ~77% of the species diversity of this family, we first inferred a plastid phylogeny of Schizaeaceae using three DNA regions. To resolve the deep relationships within this family, we then reconstructed a plastome-based phylogeny focusing on a selection of representatives that covered all the major clades. From this phylogenomic backbone, we traced the evolutionary histories of plastid genes and examined whether gene losses were associated with the evolution of gametophytic mycoheterotrophy. Our results reveal that extant Schizaeaceae is comprised of four major clades—Microschizaea, Actinostachys, Schizaea, and Schizaea pusilla. The loss of all plastid NADH-like dehydrogenase (ndh) genes was confirmed to have occurred in the ancestor of extant Schizaeaceae, which coincides with the evolution of mycoheterotrophy in this family. For chlorophyll biosynthesis genes (chl), the losses were interpreted as convergent in Schizaeaceae, and found not only in Actinostachys, a clade producing achlorophyllous gametophytes, but also in S. pusilla with chlorophyllous gametophytes. In addition, we discovered a previously undescribed but phylogenetically distinct species hidden in the Schizaea dichotoma complex and provided a taxonomic treatment and morphological diagnostics for this new species—Schizaea medusa. Finally, our phylogenetic results suggest that the current PPG I circumscription of Schizaea is non-monophyletic, and we therefore proposed a three-genus classification moving a subset of Schizaea species sensu PPG I to a third genus—Microschizaea.

Objective Depression and anxiety are comorbid conditions that are disproportionately high among American Indians (AIs) or Alaska Natives. The purpose of this study was to identify potential risk (e.g., low income, intimate partner violence [IPV], adverse childhood experiences [ACEs]) and protective factors (e.g., family resilience, social and community support) related to symptoms of depression and anxiety among AI adults. Method As part of larger exploratory sequential mixed-methods research, the study focused on survey data with 127 AI adults from two Southeastern tribes (n = 117 when missing data were removed). We used the following three-stage hierarchical regression to understand factors related to depressive and anxiety symptoms: (a) demographics, including income; (b) ACEs and IPV; and (c) family resilience, along with family and community support. Results Many participants experienced elevated levels of clinically significant symptoms of depression and anxiety (15% and 20%, respectively). Results indicated lower income was associated with higher depressive and anxiety symptoms. IPV and ACE variables were positively associated with depressive and anxiety symptoms. Family resilience was negatively associated with symptoms of anxiety and depression. Social and community support were associated with symptoms of anxiety. Conclusions The findings provide strong preliminary support for the role of family protective and promotive factors in offsetting symptoms of anxiety and depression. This is contrary to most models of interventions for anxiety and depression focusing on individual psychotherapy rather than promoting family resilience or involvement. Surface modification of nanocarriers enables selective attachment to specific molecular targets within a complex biological environment. Besides the enhanced uptake due to specific interactions, the surface ligands can be utilized for radiolabeling applications for bimodal imaging ensured by positron emission topography (PET) and magnetic resonance imaging (MRI) functions in one source. Herein, we describe the surface functionalization of magnetite (Fe3O4) with folic acid as a target vector. Additionally, the magnetic nanocarriers were conjugated with appropriate ligands for subsequent copper-catalyzed azide-alkyne cycloaddition or carbodiimide coupling reactions to successfully achieve radiolabeling with the PET-emitter 18F. The phase composition (XRD) and size analysis (TEM) confirmed the formation of Fe3O4 nanoparticles (6.82 nm ± 0.52 nm). The quantification of various surface functionalities was performed by Fourier-transform infrared spectroscopy (FT-IR) and ultraviolet-visible microscopy (UV-Vis). An innovative magnetic-HPLC method was developed in this work for the determination of the radiochemical yield of the 18F-labeled NPs. The as-prepared Fe3O4 particles demonstrated high radiochemical yields and showed high cellular uptake in a folate receptor overexpressing MCF-7 cell line, validating bimodal imaging chemical design and a magnetic HPLC system. This novel approach, combining folic acid-capped Fe3O4 nanocarriers as a targeting vector with 18F labeling, is promising to apply this probe for bimodal PET/MR-studies.

Efficient therapies for breast cancer remain elusive because of the lack of strategies for targeted transport and receptor-mediated uptake of synthetic drug molecules by cancer cells. Conjugation of nanoparticles (NPs) with active targeting ligands enabling selective molecular recognition of antigens expressed on the surface of cancer cells is promising for localization and treatment of malignant cells. In this study, covalent attachment of synthetic estrogen 17α-ethynylestradiol on the silica (SiO2) shell of silica-gold NPs (SiO2@Au) was undertaken to improve the cancer-targeting ability of the nano-biotags. Chemical and structural analysis of the bioconjugates examined in solution (UV-vis and ξ-potential) and solid state (Fourier transform infrared spectroscopy, X-ray diffractometry, and transmission electron microscopy) confirmed the identity of the carrier particles and surface-bound ligands. The mesoporous silica shell served as a reservoir for anticancer drugs (doxorubicin and quercetin) and to facilitate covalent attachment of receptor molecules by click chemistry protocols. The chemoselective recognition between the nanoconjugates and cell membranes was successfully demonstrated by the accumulation of nanoprobes in the tumor tissue of mice with subcutaneous breast cancer, whereas healthy cells were unaffected. The drug release studies showed sustained release kinetics over several weeks. These findings elaborate the exceptional selectivity and potential of estrogen-coated nano-biolabels in efficient diagnosis and detection of breast cancer cells.

Cololejeunea reniformis is described as new based on a single collection from Tully Falls National Park in north-east Queensland. Cololejeunea reniformis is similar to C. cairnsiana in the size and the falcate leaf lobes and ampulliform lobules, features which are distinctive among Australian species, and was initially thought to be that species. While C. cairnsiana is an epiphyll, the single known gathering of C. reniformis was found on bark, and the two species differ in a number of micromorphological characters, including details of the vitta, which contain 6-9 cells in a single row in C. cairnsiana and 12-20 cells in 2 or 3 rows in C. reniformis. Cololejeunea reniformis shares many micromorphological features with the south-east Asian C. ensifera, but differs in its smaller size and falcate leaf lobes, in addition to other characters. Thirty-nine Cololejeunea species are now known for Australia.

Danhatchia novaehollandiae and D. australis were separated at species rank due to differences in petal length and flower opening, with the Australian species having smaller tardily opening flowers. From this, flower lengths for Australia and New Zealand are expected to be bi-modally distributed with peaks at c. 3 mm and c. 5 mm respectively. Quantification of flower size found flower length was unimodal, with nearly identical ranges in Australian and New Zealand plants. Flower size variation in Australian and New Zealand Danhatchia specimens has two significant contributing components, inter-individual variation, and ontogenetic variation where flowers increase in size as they age. Dimensions quoted by Edgar and Moore (1970) and Jones and Clements (2018) reflect upper and lower limits on the range of variation in flower size present in both New Zealand and Australia, respectively. Within herbarium material, 20% of flowers on New Zealand specimens, and 40% of flowers on Australian specimens exhibited signs of opening. There was no correlation between flower size and opening, as might be expected if the two species were both present in Australia and/or New Zealand. Neither the biogeographic context, pollination ecology, nor morphological evidence support Danhatchia australis and D. novaehollandiae as distinct species.

It is well known that during ovarian cancer progression, the omentum transforms from a thin lacy organ to a thick tougher tissue. However, the mechanisms regulating this transformation and the implications of the altered microenvironment on ovarian cancer progression remain unclear. To address these questions, the global and local concentrations of collagen I were determined for normal and metastatic human omentum. Collagen I was increased 5.3-fold in omenta from ovarian cancer patients and localized to areas of activated fibroblasts rather than regions with a high density of cancer cells. Transforming growth factor beta 1 (TGFβ1) was detected in ascites from ovarian cancer patients (4 ng/mL), suggesting a potential role for TGFβ1 in the observed increase in collagen. Treatment with TGFβ1 induced fibroblast activation, proliferation, and collagen deposition in mouse omental explants and an in vitro model with human omental fibroblasts. Finally, the impact of increased collagen I on ovarian cancer cells was determined by examining proliferation on collagen I gels formulated to mimic normal and cancerous omenta. While collagen density alone had no impact on proliferation, a synergistic effect was observed with collagen density and heparin-binding epidermal growth factor treatment. These results suggest that TGFβ1 induces collagen deposition from the resident fibroblasts in the omentum and that this altered microenvironment impacts cancer cell response to growth factors found in ascites. IMPACT STATEMENT: Using quantitative analysis of patient samples, in vitro models of the metastatic ovarian cancer microenvironment were designed with pathologically relevant collagen densities and growth factor concentrations. Studies in these models support a mechanism where transforming growth factor β1 in the ascites fluid induces omental fibroblast proliferation, activation, and deposition of collagen I, which then impacts tumor cell proliferation in response to additional ascites growth factors such as heparin-binding epidermal growth factor. This approach can be used to dissect mechanisms involved in microenvironmental modeling in multiple disease applications.

Cryptic bryophyte species exhibit a decoupling in the degree of morphological and molecular divergence, as a result of different processes, from recent divergence to stasis. Here a body of cryptic species literature comprising 110 papers published between 2000 and end 2018 is reviewed. Most studies of cryptic species focused on northern hemispheric taxa, but we do not yet have sufficient studies to assess whether a geographic bias in the distribution of cryptic species exists, and we don’t know how many cryptic bryophyte species there might be globally. Fully two-thirds of all studies on cryptic bryophyte species rested their claims of morphological crypsis on previous taxonomic investigations, without revision of morphology to confirm cryptic species status. There is more than one kind of morphological crypsis, and while quantification of morphological patterns can contribute to our understanding of crypsis this is a widely neglected component. The usage of ‘cryptic species’ as an etymological tool to flag instances where traditional species concepts are deficient devalues the term, and a distinction between genuine crypsis and business as usual revision of species circumscription should be re-established and maintained. Hybridisation is possibly an under-appreciated contributor to cryptic species, but inference of hybridization has been limited by study design. Opportunities exist in the application of geometric morphometric methods and next generation sequencing technologies to overcome intrinsic limitations in traditional morphological and molecular data sources.

Indigenous peoples of the United States are distinct from other ethnic minorities because they have experienced colonization as the original inhabitants. Social and health disparities are connected to a context of historical oppression—the chronic, pervasive, and intergenerational experiences of oppression that, over time, may be normalized, imposed, and internalized into the daily lives of many Indigenous peoples (including individuals, families, and communities). As part of the critical Framework of Historical Oppression, Resilience, and Transcendence (FHORT), in this article, we introduce the Historical Oppression Scale (HOS), a scale assessing internalized and externalized oppression. Our study reports on survey data ( N = 127) from a larger convergent mixed-methodology study with scale items derived from thematic analysis of qualitative data ( N = 436), which informed the resultant 10-item scale. After six cases were removed from the 127 participants who participated in the quantitative component to the study due to missing data across two tribes, the sample size for analysis was 121. Confirmatory factor analysis testing of the hypothesized unidimensional construct indicated acceptable model fit ( X2 = 58.10, [Formula: see text] 1.94, CFI = .98, TLI = .97, RMSEA = .088, 90% CI = .05, .12). Reliability of the 10-item scale was excellent (α = .97) and convergent and discriminant validity were established. The HOS explicates complex associations between historical oppression and health and social disparities and may be an important clinical and research tool in an understudied area.

All studied leafy liverworts have shoots with leaf to underleaf ratios of either 2:1 or 1:1. These ratios are the product of growth by either helical or pendular segmentation of the tetrahedral apical cell. Here we report that Herzogianthus vaginatus has a leaf to underleaf ratio approaching 3:1 on primary shoots, and on secondary shoots the ratio is closer to 4:1. These ratios are incompatible with the simple helical or pendular growth patterns found in other leafy liverworts. Further, the sequence of leaves and underleaves on Herzogianthus shoots is not wholly regular, which is previously unrecorded. We assess potential growth models that may explain the leaf to underleaf ratios and architecture of Herzogianthus shoots, including (1) branch modification, (2) merophyte or leaf-initial duplication and (3) changes in segmentation sequences. We compared leaf sequences produced by pendular and helical segmentation and a range of non-standard models, with sequences observed on shoots of Herzogianthus. Structure allows us to discriminate between the many alternative growth models, because each make testable predictions about leaf and underleaf sequences. We propose that Herzogianthus possesses growth processes unique among leafy liverworts, in which the direction of helication periodically and, sometimes irregularly, reverses. This, in combination with the secondary shoots having different growth patterns from primary shoots, emphasizes that Herzogianthus warrants detailed histological study to test our proposition.
