Post-transcriptional modifications (PTMs) are pivotal in the regulation of gene expression, and pseudouridylation is emerging as a critical player. This modification, facilitated by enzymes such as NOB1 (PNO1), is integral to ribosome biogenesis. PNO1, in collaboration with the NIN1/RPN12 binding protein 1 homolog (NOB1), is vital for the maturation of ribosomes, transitioning 20S pre-rRNA into functional 18S rRNA. Recent studies have highlighted PNO1's potential involvement in cancer progression; however, its underlying mechanisms remain unclear. Relentless growth characterizing cancer underscores the burgeoning significance of epitranscriptomic modifications, including pseudouridylation, in oncogenesis. Given PNO1's emerging role, it is imperative to delineate its contribution to cancer development to identify novel therapeutic interventions. This review summarizes the current literature regarding the role of PNO1 in cancer progression and its molecular underpinnings in oncogenesis. Overexpression of PNO1 was associated with unfavorable prognosis and increased tumor malignancy. At the molecular level, PNO1 facilitates cancer progression by modulating mRNA stability, alternative splicing, and translation efficiency. Its role in pseudouridylation of oncogenic and tumor-suppressor transcripts further underscores its significance in cancer biology. Although disruption of ribosome biogenesis is known to precipitate oncogenesis, the precise mechanisms by which these alterations contribute to cancer remain unclear. This review elucidates the intricate process of ribosomal small subunit maturation, highlighting the roles of crucial ribosomal proteins (RPs) and RNA-binding proteins (RBPs) as well as the positioning and function of NOB1 and PNO1 within the 40S subunit. The involvement of these components in the maturation of the small subunit (SSU) and their significance in the context of cancer therapeutics has been thoroughly explored. PNO1's burgeoning significance in oncology makes it a potential target for cancer therapies. Strategies aimed at modulating PNO1-mediated pseudouridylation may provide new avenues for cancer treatment. Developing non-noble, earth-ample, and stable electrocatalysts are highly anticipated in oxygen-evolution reaction (OER) and hydrogen-evolution reaction (HER) at unique pH conditions. Herein, we have synthesized bimetallic (nickel and iron) zeolite imidazolate framework (ZIF)-based nanofibrous materials via a simple electrospinning (ES) process. The structural stability of the fibrous material is subjected to various calcination conditions. We have elaborated the structural importance of the one-dimensional (1D) fibrous materials in electrocatalytic water-splitting reactions. As a result, NiFe-ZIF-NFs (Nanofibers)-RT (Room Temperature) have delivered a small overpotential of 241 mV at 10 mA cm -2 current density in OER and 290 mV at a fixed current density of 50 mA cm -2 in HER at two different pH conditions with 1 M KOH and 0.5 M H 2 SO 4 , respectively. Furthermore, it exposes the actual surface area of 27.270 m 2 g -1 and a high electrochemical active surface area (ECSA) of 50 μF in OER and 55 μF in HER, which is responsible for the electrochemical performance with better stability. This exceptional activity of the materials is mainly attributed to the structural dependency of the fibrous network through the polymeric architecture.

Out of various available methods, generation of hydrogen by electrocatalytic water splitting is the most accepted one which consists of two half-cell reactions, viz , oxygen evolution reaction (OER) at the anode and hydrogen evolution reaction at the cathode. OER is a complex four-electron transfer process, and to sustain the spontaneous generation of hydrogen at the cathode, it is urgent to develop some earth-abundant, low-cost electrode materials. Recently, use of cobalt-based hydroxide as the electrode substrate has taken much consideration and has been fabricated over various substrates. Because of various structural disorders, internal resistance, and dependence on the electrode, the binder substrate makes their applications limited. Here, in this work, to remove structural disorder and to increase electrical conductivity, we have incorporated silver ions into amorphous Co(OH) 2 , which turns to be a highly active OER electrocatalyst. Also, for the first time, we have developed hydroxide-based materials by using DNA as a stabilizer, and most importantly, using DNA gives an immense opportunity to run long-term OER applications without using an external binder such as nafion. Moreover, for the first time, these DNA-based materials were coated on nickel foam mainly to eliminate the low conductive nature of Ag 2 O. The synthesized catalyst showed a very high OER activity, and to reach 50 mA/cm 2 current density, it needs only 260 mV as overpotential. The amorphous nature of hydroxide-based materials gives a higher opportunity toward the electrolyte to bind on the surface of a catalyst to run the OER with less applied overpotentials.

Background: Pregnancy failure and placenta mediated pregnancy complications affect >25% of pregnancies. Although there is biological plausibility for a procoagulant mechanism underlying some of these events, antithrombotic intervention trials demonstrate limited benefit, possibly through lack of stratification in heterogeneous patient groups. The ANXA5 M2 haplotype is a possible procoagulant biomarker and was tested pragmatically to determine whether this screening and LMWH treatment normalized the outcome for ANXA5 M2 positive couples. This was a pragmatic study that aimed to measure the effectiveness of a testing (for the M2 haplotype) and treatment (LMWH) pathway in routine clinical practice where there is variation between patients. Such a study in couples with fertility problems can inform choices between treatments; it is then the management protocol which is the subject of the investigation, not the individual treatments., Methods: Couples (N=77) with one or both partners ANXA5 M2 positive demonstrated association of this haplotype with adverse IVF outcome. A pragmatic, multicenter, prospective cohort study of ANXA5 M2 haplotype screening, and LWMH treatment following embryo transfer (ET) in 103 IVF couples positive for ANXA5 M2 was performed. They were compared with a group of 1000 contemporaneous randomly selected unscreened and untreated couples undergoing assisted conception, from which 103 matched control couples were derived. The primary outcome measure was live birth incidence. Secondary outcomes were results following embryo transfer (ET) and live birth outcome by gender and M2 carriage, and allelic dose influence., Findings: The tested and treated cohort of ANXA5 M2 carriers achieved a similar live birth rate (37.9%) per ET cycle compared to both the more fertile comparison group (38.5%), and to the 103 matched controls (33.0%). Significantly more treated male carrier only couples had a live birth versus female M2 only (47.7% vs. 25.0% p=0.045)., Interpretation: Pragmatic ANXA5 M5 screening and treatment with LMWH in couples undergoing IVF is associated with similar outcome to couples with more favorable prognostic factors. The difference in live birth outcome for treated male only carrier couples may be consistent with an additional maternal thrombophilic factor that may adversely affect pregnancy, although other mechanisms are possible. Erythema gyratum repens (EGR) is an obligate paraneoplastic eruption that indicates an underlying malignancy, most commonly cancer of the lung or breast. The characteristic clinical finding is a rapidly expanding, mildly scaling dermatosis with a 'wood-grain' pattern. This typical concentric erythema is not seen in all patients. Other findings may include pruritus, palmoplantar keratoderma and/or blood eosinophilia. We report a case of EGR where the diagnosis was suggested less by the wood-grain pattern than by the dynamics of the lesions.
