Classic bladder exstrophy represents the most severe end of all human congenital anomalies of the kidney and urinary tract and is associated with bladder cancer susceptibility. Previous genetic studies identified one locus to be involved in classic bladder exstrophy, but were limited to a restrict number of cohort. Here we show the largest classic bladder exstrophy genome-wide association analysis to date where we identify eight genome-wide significant loci, seven of which are novel. In these regions reside ten coding and four non-coding genes. Among the coding genes is EFNA1, strongly expressed in mouse embryonic genital tubercle, urethra, and primitive bladder. Re-sequence of EFNA1 in the investigated classic bladder exstrophy cohort of our study displays an enrichment of rare protein altering variants. We show that all coding genes are expressed and/or significantly regulated in both mouse and human embryonic developmental bladder stages. Furthermore, nine of the coding genes residing in the regions of genome-wide significance are differentially expressed in bladder cancers. Our data suggest genetic drivers for classic bladder exstrophy, as well as a possible role for these drivers to relevant bladder cancer susceptibility., (© 2022. The Author(s).)

Recently, traces of brominated flame retardants (BFRs) have been detected in black plastic food-contact materials (FCMs), indicating the presence of recycled plastics, mainly coming from waste electric and electronic equipment (WEEE) as BFRs are one of the main additives in electric applications. In order to evaluate efficiently and preliminary in situ the presence of WEEE in plastic FCMs, a generic procedure for the evaluation of WEEE presence in plastic FCMs by using defined parameters having each an associated importance level has been proposed. This can be achieved by combining parameters like overall bromine (Br) and antimony (Sb) content; additive and reactive BFR, rare earth element (REE) and WEEE-relevant elemental content and additionally polymer purity. In most of the cases, the WEEE contamination could be confirmed by combining X-ray fluorescence (XRF) spectrometry and thermal desorption/pyrolysis gas chromatography-mass spectrometry (GC-MS) at first. The Sb and REE content did not give a full confirmation as to the source of contamination, however for Sb the opposite counts: Sb was joined with elevated Br signals. Therefore, Br at first followed by Sb were used as WEEE precursors as both elements are used as synergetic flame-retardant systems. WEEE-specific REEs could be used for small WEEE (sWEEE) confirmation; however, this parameter should be interpreted with care. The polymer purity by Fourier-transform infrared spectrometer (FTIR) and pyrolysis GC-MS in many cases could not confirm WEEE-specific contamination; however, it can be used for purity measurements and for the suspicion of the usage of recycled fractions (WEEE and non-WEEE) as a third-line confirmation. To the best of our knowledge, the addition of WEEE waste to plastic FCMs is illegal; however, due to lack on screening mechanisms, there is still the breakthrough of such articles onto the market, and, therefore, our generic procedure enables the quick and effective screening of suspicious samples.

In order to confirm the possibility that recycled fractions from the waste electrical and electronic equipment (WEEE) stream were illegally entering the European market in black polymeric food-contact articles (FCAs), bromine quantification, brominated flame retardant (BFR) identification combined with WEEE-relevant elemental analysis and polymer impurity analysis were performed. From the 10 selected FCAs, seven samples contained a bromine level ranging from 57 to 5975 mg kg(-)(1), which is lower than expected to achieve flame retardancy. The BFRs that were present were tetrabromobisphenol A (TBBPA), decabromodiphenylether (decaBDE), decabromodiphenylethane (DBDPE) and 1,2-bis(2,4,6-tribromophenoxy)ethane (BTBPE). Typical elements used in electronic equipment and present in WEEE were detected either at trace level or at elevated concentrations. In all cases when bromine was detected at higher concentrations, concurrently antimony was also detected, which confirms the synergetic use of antimony in combination with BFRs. This study describes also the measurement of rare earth elements where combinations of cerium, dysprosium, lanthanum, neodymium, praseodymium and yttrium were detected in four of the seven BFR-positive samples. Additionally, polymer purity was investigated where in all cases foreign polymer fractions were detected. Despite the fact that this study was carried out on a very small amount of samples, there is a significant likelihood that WEEE has been used for the production of FCAs.

We report the infrared spectra of HCl, (HCl)2, and H2O-HCl in liquid helium nanodroplets in the frequency region between 2680 and 2915 cm(-1). For the HCl monomer a line width of 1.0 cm(-1) (H35Cl) corresponding to a lifetime of 5.3 ps was observed. The line broadening indicates fast rotational relaxation similar to that previously observed for HF. For (HCl)2 the free HCl as well as the bound HCl stretching band has been observed. The nu2+ bands of (HCl)2 could be rotationally resolved, and rotational constants were deduced from the spectra. We observed both the allowed and the symmetry forbidden transition. However, the forbidden "broken symmetry" tunneling transition of the mixed dimer shows an intensity that is considerably enhanced compared to the gas phase. Upon the basis of the present measurements we were able to calculate the tunneling splitting in the excited state. The tunneling splitting is found to be reduced by 28% compared to the gas phase. Transitions from the ground state to the Ka=1 level of the free HCl stretch (nu1) are recorded and show considerable line broadening with a line width of 2 cm(-1). The excited state Ka=1 has an additional rotational energy of about 10 cm(-1), thereby allowing fast rotational relaxation by coupling to helium excitations. In addition we observed the HCl stretch of the HCl-H2O dimer, which exhibits an unusually large width (1.7 cm(-1) for H35Cl)) and large red shift (8.5 cm(-1)), compared to the gas-phase values. The large-amplitude motion originating from the libration mode of the HCl-H2O complex is supposed to act as a fast relaxation manifold.

We report the fully analyzed high-resolution spectrum of the carboxylic acid dimer (HCOOH)2 in the gas phase. High-resolution IR spectra in the region of the antisymmetric C-O stretch vibration have been recorded at 1221.0-1226.7 cm(-1). The data could be fit within experimental uncertainty to a rigid rotor Watson S-reduced Hamiltonian. The vibrational frequency of the C-O vibration is 1225.3430(3) cm(-1). On the basis of the measurement of a tunneling splitting of 0.0158(4)cm(-1) for the lower state and 0.0100(3) cm(-1) for the upper state, we determine a proton-transfer time of 1.0 and 1.7 ns for the ground and the vibrationally excited state, respectively.

Purpose: In an attempt to intensify conditioning therapy for bone marrow transplantation of hematologic malignancies, a retrospective three center evaluation of escalating doses of etoposide added to cyclophosphamide and either total body irradiation or busulfan was undertaken., Methods and Materials: Seventy-six patients who received etoposide (25-65 mg/kg) added to cyclophosphamide (60-120 mg/kg) and either total body irradiation (12.0-13.2 Gy) or busulfan (12-16 mg/kg) were evaluable for toxicity. Fifty-one of the evaluable patients received allogeneic transplants, while twenty-six received autologous transplants. A comparative analysis of toxicities according to conditioning regimen, donor source and etoposide dose was made., Results: Similar toxicities were observed among the treatment groups with the exception of more frequent skin (p = 0.003) and life threatening hepatic toxicities (p = 0.01) in the busulfan treated patients. Life threatening or fatal toxicities were not influenced by donor source, either when analyzed by treatment group or etoposide dose. Etoposide at a dose of 60-65 mg/kg in combination with TBI and cyclophosphamide was associated with a significantly increased incidence of life threatening or fatal toxicities compared with a combination using a dose of 25-50 mg/kg (15 of 24 vs. 5 of 20; p = 0.013). The maximally tolerated dose of etoposide in combination with busulfan and cyclophosphamide cannot be definitively established in this analysis in part due to the heterogeneity of the patient population and treatment schemes., Conclusion: Although toxicities with bone marrow transplant preparative regimens containing etoposide in combination with cyclophosphamide and total body irradiation or busulfan were frequently severe, treatment related mortality risk was believed to be acceptably low.
