1. Influence of the placement rate on the fresh concrete lateral pressure in the design of formworks

Abstract

Abstract The fresh concrete lateral pressure generates a load that has great influence in the design of the formworks of columns, walls and lateral faces of the beams.Its prediction must be performed in such a way in order to approach precisely well over the rate values supported by the formworks, avoiding, therefore, the oversizing or undersizing of these temporary structures which represent up to 12% of the total cost of the construction. The calculation of the lateral pressure that fresh concrete exerts on the formworks involves several variables; one of them is the placement rate. The main objective of this work was to study the influence of the placement rate, in situ, on the lateral pressure of fresh concrete calculated from theoretical models presented by norms and international references. For this purpose, measurements of the placement rate were conducted on columns of five building work places at Goiânia city, Goiás, Brazil and it was calculated the maximum pressure supported by their respective formworks from the dimensions and spans between the supports of the components that constituted them. The obtained results indicate that the placement rate in the field observations reach 249 m/h, exceeding the values limited by the theoretical methods of calculation, and that the lateral pressure estimation produced from the placement rate measured in situ exceed the maximum values supported by the formworks up to 604 times. Thus, it was concluded that it is necessary to adjust equations to estimate the lateral pressure of fresh concrete at high rate of concrete placement, according to the current practices in the construction market.

Abstract

desenvolvimento da planta de vinagreira (Hibiscus sabdariffa L.), produzida com diferentes adubações. O experimento teve duração de 120 dias e constou de seis tratamentos T1: Esterco Bovino; T2: Esterco de Ave; T3: NPK (4-14-8); T4: NPK (4-14-8) + cobertura de N; T5: NPK (4-14-8) + cobertura de K; T6: NPK (4-14-8) + cobertura de N e K. A semeadura foi realizada em 13/12/2008, sendo utilizadas bandejas de isopor de 128 células. Foi utilizada uma semente por célula. A irrigação das bandejas foi feita diariamente. A germinação teve início três dias após o plantio (DAP). Aos vinte e dois dias após a semeadura, foram avaliados a altura da planta (AP), a quantidade de frutos por plantas (QFP), matéria seca e matéria verde (MS/MV). Pode concluir-se que as adubações testadas induziram efeitos significativos, destacando-se o T4 que demonstra melhor resultado quando comparado aos demais tratamentos.

Abstract

The aim of this research work is to study the effect of salt mist ageing on the physico-mechanical properties of two ignimbrites from the Canary Islands (Spain). Due to their insular nature and extensive coastlines, these regions are highly susceptible to atmospheric salt aerosols, which is a significant weathering agent for building materials. The ignimbrites analysed are marketed under the names “Azul Lomo Tomás de León” and “Marrón de Abades” and are widely used as building stones. Petrographic, mineralogical and chemical properties were obtained via polarisation microscopy, X-ray diffraction and X-ray fluorescence. The samples were subjected to 60 cycles of a salt mist environment, following relevant European standards. Scanning electron microscopy images of the specimens were analysed along with physical properties, including apparent density, open porosity, water absorption, colourimetry and ultrasound propagation velocity, initially and after every 20 cycles, allowing us to assess their evolution during the ageing test. Mechanical properties were evaluated using uniaxial compressive strength and flexural strength under concentrated load tests on two groups of specimens: one unaged and the other subjected to 60 ageing cycles. With the exception of apparent density, the other properties show a decrease between 5% and 30%. However, open porosity increases and is one of the properties most affected by the ageing test.

Abstract

Among the different terrestrial ecosystems, forests are the most important biomass carbon producers and the ones that store the most standing biomass carbon. Consequently, they are also the major source of biomass for energy. Forest biomass has been used as a fuel from early times, and from the late twentieth century onward, there has been a renewed interest in its use to produce heat and electricity. The interest in forest biomass as an energy source relates to some of its features, such as relative abundance and uniformity worldwide and neutrality of CO2 emissions. Nonetheless, its use is not free of risks, mostly related with the sustainability of the forest systems and their productions. This study reviews the state of the art of the forest sources of biomass for energy, their assessment, their properties as a fuel, as well as the conversion technologies used in the most common energy applications.

Abstract

Assessment and monitoring of forest structure is frequently done with absolute density measures with field forest inventory data and expansion methods. The development of basal area and the number of trees estimation functions with data derived from very high spatial resolution satellite images enable their short-term and cost-effective evaluation, allowing also the estimation for the area not requiring extrapolation methods. The functions of basal area and the number of trees per hectare are based on crown cover obtained with very high spatial resolution satellite images for two evergreen oaks and umbrella pine. The three tree species are especially important in the agroforestry systems of the Mediterranean region. The linear functions fitted for pure stands of the three species and mixed stands of cork and holm oak and of cork oak and umbrella pine showed a better performance for basal area than for the number of trees per hectare. The inclusion of dummy variables for species composition improved the accuracy of the functions.

Abstract

Landscape evaluation and monitoring enable us to understand the interactions between its components and the effects of disturbances (whether they are natural or artificial) in its dynamics. Forests have a wide variability and diversity, and their analysis at the landscape level allows us to evaluate its spatial distribution pattern. This study focused on the analysis of the landscape spatial variability of forest species with data derived from remote sensing and landscape metrics of a case study in Alto Alentejo, Portugal. Sentinel-2 satellite images were used to produce a land use and land cover map with a random forest classification algorithm, where the bands, vegetation and texture indices were the explanatory variables. The obtained land use/cover map has classified five forest classes and one non-forest class. The map was used to evaluate the diversity with eleven composition and configuration landscape diversity metrics for Alto Alentejo and for four sub-regions delimited according to their edaphic-climatic characteristics. The results showed that the land use/cover map had a good precision (a global precision of 89% and a kappa of 86%) and that both Alto Alentejo and its sub-regions had high forest diversity both in composition and configuration.

Abstract

In the context of the cancer-inflammation relationship and the use of natural products as potential antitumor and anti-inflammatory agents, the alkaloid-enriched fraction of Boehmeriacaudata (BcAEF) aerial parts was evaluated. In vitro antiproliferative studies with human tumor cell lines showed high activity at low concentrations. Further investigation on NCI-H460 cells showed an irreversible effect on cell proliferation, with cell cycle arrest at G2/M phase and programmed cell death induction. Molecular docking studies of four alkaloids identified in BcAEF with colchicine’s binding site on β-tubulin were performed, suggesting (−)-C (15R)-hydroxycryptopleurine as the main inductor of the observed mitotic death. In vivo studies showed that BcAEF was able to reduce Ehrlich tumor volume progression by 30 to 40%. Checking myeloperoxidase activity, BcAEF reduced neutrophils migration towards the tumor. The in vivo anti-inflammatory activity was evaluated by chemically induced edema models. In croton oil-induced ear edema and carrageenan (CG)-induced paw edema models, BcAEF reduced edema around 70 to 80% together with inhibition of activation and/or migration of neutrophils to the inflammatory area. All together the results presented herein show BcAEF as a potent antitumor agent combining antiproliferative and anti-inflammatory properties, which could be further explored in (pre)clinical studies.

Abstract

In order to address the global antivenom crisis, novel antivenoms need to present high therapeutic efficacy, broad neutralization ability against systemic and local damage, sufficient safety, and cost-effectiveness. Due to biological characteristics of camelid single-domain antibodies (VHH) such as high affinity, their ability to penetrate dense tissues, and facility for genetic manipulation, their application in antivenoms has expanded considerably. VHHs that are active against the metalloprotease BjussuMP-II from the snake Bothrops jararacussu were selected. After isolation of BjussuMP-II, a camelid was immunized with the purified toxin in order to construct the recombinant phage library. Following a round of biopanning, 52% of the selected clones were able to recognize BjussuMP-II in an ELISA assay. After sequencing, seven sequence profiles were identified. One selected clone (VHH61) showed cross-reactivity to B. brazili venom, but did not recognize the Crotalus and Lachesis genera, indicating specificity for the Bothrops genus. Through in vitro tests, the capacity to neutralize the toxicity triggered by BjussuMP-II was observed. Circular dichroism spectroscopy indicated a robust secondary structure for VHH61, and the calculated melting temperature ( T M ) for the clone was 56.4°C. In silico analysis, through molecular docking of anti-BjussuMP-II VHHs with metalloprotease, revealed their potential interaction with amino acids present in regions critical for the toxin’s conformation and stability. The findings suggest that anti-BjussuMP-II VHHs may be beneficial in the development of next-generation antivenoms.

A pressão lateral do concreto fresco gera uma carga que possui grande influência no dimensionamento das fôrmas de pilares, paredes e faces laterais das vigas. Sua previsão deve ser realizada de modo que se aproxime bem dos valores suportados pelas fôrmas, evitando-se, assim, o superdimensionamento ou subdimensionamento dessas estruturas provisórias que representam até 12% do custo total da obra. O cálculo da pressão lateral que o concreto fresco exerce sobre as fôrmas envolve diversas variáveis, sendo uma delas a velocidade de concretagem. O principal objetivo deste trabalho foi estudar a influência da velocidade de concretagem sobre a pressão lateral do concreto fresco calculada a partir de modelos teóricos apresentados por normas e referências internacionais. Para isso, foram realizadas medições da velocidade de concretagem em pilares de cinco obras de Goiânia e calculadas as pressões máximas suportadas por suas respectivas fôrmas, a partir das dimensões e dos vãos entre os apoios dos elementos que as constituíam. Os resultados obtidos apontam que as velocidades de concretagem em campo chegam a 248,57 m/h, superando os valores limitados pelos métodos teóricos de cálculo, e que as estimativas da pressão realizadas a partir das velocidades medidas in loco ultrapassam os valores máximos suportados pelas fôrmas em até 603,75 vezes. Assim, concluiu-se que é necessária a formulação de equações para estimar a pressão lateral do concreto fresco a altas velocidades de concretagem, conforme as atuais práticas no mercado da construção civil. Abstract

Cellulases are enzymes that degrade cellulosic materials. Cellulose is the most abundant renewable carbon resource on Earth, and cellulases are used in various industrial sectors. Although cellulases are obtained from a variety of sources, this is the first description of cellulolytic activity isolated from a coral metagenomic library. A metagenomic fosmid library of microorganisms associated with the coral Siderastrea stellata, comprising 3552 clones, was screened for cellulolytic activity; this allows access to non-cultivable microorganisms by exploiting the full biotechnological potential. Clones were grown on LB agar plates supplemented with 0.5% carboxymethylcellulose and cellulase positive clones revealed by staining with Congo red. Using this approach, six positive clones with cellulolytic activity were identified. The enzymatic index (EI) of the positive clones was calculated by the ratio between the hydrolysis zone diameter and colony diameter. All positive clones had an EI greater than 1.5. Digestion of the DNA isolated from the six positive clones, using the HindIII restriction endonuclease, revealed different restriction patterns in each clone, indicating that the DNA of each clone is different. There is a growing interest for new cellulolytic enzymes in various industry sectors. Here, we present the initial selection of potential clones for cellulose degradation that could be targets for future studies of enzymatic characterization.

Abstract

The oil obtained from Pterodon pubescens (Leguminosae) seeds are known to display anti-cancer, anti-dermatogenic and anti-nociceptive activitiy. Phytochemical studies have demonstrated that its main constituents are diterpenoids with voucapan skeletons. Considering the potential biological activities of the oil, rapid and efficient methods for assessing its quality would facilitate certification and quality control.To develop a direct mass spectrometric fingerprinting method for the P. pubescens seed oil that would focus on the major diterpenoids constituents, enabling quality control, origin certification and recognition of marker species in commercially available products.Two techniques were used: (i) direct infusion electrospray ionisation (ESI) mass spectrometry after solvent extraction and dilution and (ii) ambient desorption/ionisation via easy ambient sonic-spray ionisation, EASI(+)-MS, performed directly on the seed surface or at a paper surface imprinted with the oil.From a combination of ESI-MS, HRESI-MS and ESI-MS/MS data, 12 diterpenes were characterised, and typical profiles were obtained for the oil extract or the crude oil via both ESI-MS and EASI-MS. These techniques require no or very simple sample preparation protocols and the whole analytical processes with spectra acquisition take just a few minutes.Both techniques, but particularly EASI-MS, provide simple, fast and efficient MS fingerprinting methodologies to characterise the P. pubescens oil with typical (di)terpene profiles being applicable to quality control and certification of authenticity and origin.

22. Kinetic characterization of rat tissue kallikrein using N alpha-substituted arginine 4-nitroanilides and N alpha-benzoyl-L-arginine ethyl ester as substrates

Abstract

Hydrolysis of seven N alpha-substituted L-arginine 4-nitroanilides: benzoyl-arginine p-nitroanilide (Bz-Arg-Nan), tosyl-arginine p-nitroanilide (Tos-Arg-Nan), acetyl-leucyl-arginine p-nitroanilide (Ac-Leu-Arg-Nan), acetyl-phenylalanyl-arginine p-nitroanilide (Ac-Phe-Arg-Nan), benzoyl-phenylalanyl-arginine p-nitroanilide (Bz-Phe-Arg-Nan), tosyl-phenylalanyl-arginine p-nitroanilide (Tos-Phe-Arg-Nan), and D-valyl-leucyl-arginine p-nitroanilide (D-Val-Leu-Arg-Nan), and the N alpha-substituted L-arginine ester: benzoyl-arginine ethyl ester (Bz-Arg-OEt), by rat tissue kallikrein was studied throughout a wide range of substrate concentrations. The enzyme showed a bimodal behavior with all the substrates tested except Tos-Arg-Nan. At low substrate concentrations (10 to 170 microM for p-nitroanilides and 50 to 190 microM for Bz-Arg-OEt) the hydrolysis followed Michaelis-Menten kinetics, but at higher substrate concentrations (150 to 700 microM for p-nitroanilides and 200 to 1800 microM for Bz-Arg-OEt) a deviation from Michaelis-Menten kinetics was observed with a significant decrease in hydrolysis rates. At high concentrations of the p-nitroanilide substrates, partial enzyme inhibition was observed, whereas complete enzyme inhibition was observed with Bz-Arg-OEt at high concentration. The kinetic parameters reported here were calculated from data for substrate concentrations range where the enzyme followed Michaelis-Menten behavior. D-Val-Leu-Arg-Nan (Km = 24 +/- 2 microM; Vmax = 10.42 +/- 0.28 microM/min) was the best substrate tested, followed by Ac-Phe-Arg-Nan (Km = 13 +/- 2 microM; Vmax = 3.21 +/- 0.11 microM/min), while Tos-Arg-Nan (Km = 29 +/- 2 microM; Vmax = 0.10 +/- 0.002 microM/min) was the worst of the tested substrates for rat tissue kallikrein. For the hydrolysis of Bz-Arg-OEt (Km = 125 +/- 15 microM; Vmax = 121.3 +/- 7.6 microM/min), the kinetic parameters using a substrate inhibition model can reasonably account for the observed enzyme behavior, with a Ksi value about 13.6 times larger than the estimated Km value.

Abstract

The aim of this study was to analyse the composition, structure and spatial and temporal patterns of diversity and abundance of the ichthyofauna of the Santa Cruz Reservoir in semi-arid Brazil. Data were collected quarterly at eight sampling locations on the reservoir between February 2010 and November 2011 using gillnets from 12- to 70-mm mesh that were left in the water for 12h00min during the night. We evaluated the composition, structure and assemblage descriptors (Shannon-Wiener diversity index and equitability, respectively) and catch per unit effort by the number (CPUEn) and biomass (CPUEb) of the ichthyofauna. The 6,047 individuals (399,211.6 g) captured represented three orders, ten families and 20 species, of which four belonged to introduced species. The family Characidae was the most abundant with a total of 2,772 (45.8%) individuals captured. The species-abundance curve fit the log-normal model. In the spatial analysis of diversity, there were significant differences between sampling sites in the lacustrine and fluvial regions, and the highest values were found in the lacustrine region. In the temporal analysis of diversity, significant differences were also observed between the rainy and dry seasons, and the higher values were found during the dry season. Equitability followed the same spatiotemporal pattern as diversity. The Spearman correlation was significantly negative between diversity and rainfall. A cluster analysis spatially separated the ichthyofauna into two groups: one group formed by sampling sites in the fluvial region and another group formed by the remainder of the points in the lacustrine region. Both the CPUEn and CPUEb values were higher at point 8 (fluvial region) and during the rainy season. A two-way ANOVA showed that the CPUEn and CPUEb values were spatially and temporally significant. We conclude that the spatial and temporal trends of diversity in the Santa Cruz reservoir differ from those of other Brazilian reservoirs but that the fish community composition and spatiotemporal patterns of abundance were similar.

Abstract

The estimation of vegetation parameters, such as above-ground biomass, with high accuracy using remote sensing data, represents a promising approach. The present study develops models to estimate and map above-ground biomass of Mediterranean Quercus rotundifolia stands using one QuickBird satellite image in pan-sharpened mode, with four multispectral bands (blue, green, red and near infrared) and a spatial resolution of 0.70 m. The satellite image was orthorectified, geometrically and radiometrically corrected. Object-oriented classification methods and multi-resolution segmentation were used to derive a vegetation mask per forest species. Data from forest inventory (24 plots) and vegetation indices (NDVI, EVI, SR and SAVI) derived from high spatial resolution satellite images were used for an area of 133 km2, in southern Portugal. The statistical analysis included correlation, variance analysis and linear regression. The linear regression models fitted included the arithmetic mean and the median values of the vegetation indices per inventory plot as explanatory variables. The overall results of the fitted models show a trend of better performance for those with the median value of the vegetation index as the explanatory variable. The best fitted model (R2 = 75.3) was associated with the Simple Ratio (SR) median value as an explanatory variable. A Quercus rotundifolia above-ground biomass map was produced.
