PurposeCustomer participation (CP) has received considerable interest in the service literature as a way to improve the customer experience and reduce service providers' costs. While its benefits are not in question, there is a paucity of research on potential pitfalls. This paper provides a conceptual foundation to address this gap and develops a comprehensive model of the risks of customer participation in service delivery, integrating research from the marketing, operations and supply chain management, strategy, and information technology fields.Design/methodology/approachThe model is derived deductively by integrating insights from research in marketing, operations and supply chain management, strategy, and information technology.FindingsThis paper identifies three categories of potential risks of CP (i.e. market, operational, and service network) and discusses ways that firms can mitigate these risks. Building on the model, it develops a CP risk assessment tool that managers can use when evaluating increases in CP.Research limitations/implicationsThe conceptual model proposed in this paper can serve as a robust basis for future research in customer participation, particularly in such areas as sharing economy services, service delivery networks, and experiential services. The risk assessment tool offers clear guidelines for managers who are considering an increase in customer participation in their service.Originality/valueThis is the first attempt to conceptually define customer participation risk and develop a comprehensive model of its drivers and strategies to mitigate it. This paper develops a straightforward method for managers to evaluate CP risk.

Serology has provided valuable diagnostic and epidemiological data on antibody responses to SARS-CoV-2 in diverse patient cohorts. Deployment of high content, multiplex serology platforms across the world, including in low and medium income countries, can accelerate longitudinal epidemiological surveys. Here we report multiSero, an open platform to enable multiplex serology with up to 48 antigens in a 96-well format. The platform consists of three components: ELISA-array of printed proteins, a commercial or home-built plate reader, and modular python software for automated analysis (pysero). We validate the platform by comparing antibody titers against the SARS-CoV-2 Spike, receptor binding domain (RBD), and nucleocapsid (N) in 114 sera from COVID-19 positive individuals and 87 pre-pandemic COVID-19 negative sera. We report data with both a commercial plate reader and an inexpensive, open plate reader (nautilus). Receiver operating characteristic (ROC) analysis of classification with single antigens shows that Spike and RBD classify positive and negative sera with the highest sensitivity at a given specificity. The platform distinguished positive sera from negative sera when the reactivity of the sera was equivalent to the binding of 1 ng mL−1 RBD-specific monoclonal antibody. We developed normalization and classification methods to pool antibody responses from multiple antigens and multiple experiments. Our results demonstrate a performant and accessible pipeline for multiplexed ELISA ready for multiple applications, including serosurveillance, identification of viral proteins that elicit antibody responses, differential diagnosis of circulating pathogens, and immune responses to vaccines.

Background A high population level of vaccination is required to control the COVID-19 pandemic, but not all Canadians are convinced of the value and safety of vaccination. Understanding more about these individuals can aid in developing strategies to increase their acceptance of a COVID-19 vaccine. The objectives of this study were to describe COVID-19 vaccine acceptance, hesitancy and refusal rates and associated factors in Saskatchewan, Canada. Methods This is a cross-sequential study that consisted of pooled responses from weighted samples of 9,252 Saskatchewan adults (≥18 years) across nine rounds of data collection between May 4, 2020 and April 3, 2021. The outcome variable was vaccine intention: vaccine acceptance, hesitancy, and refusal. The independent variables were layered into socio-demographic factors, risk of exposure to coronavirus, mitigating behaviours, and perceptions of COVID-19. Data were analyzed using multinomial logistic regression and a classification and regression tree. Results Seventy-six percent of the respondents indicated that they had been or were willing to be vaccinated, 13% had not yet decided, and the remaining 11% said they would not be vaccinated. Factors that increased the likelihood of vaccine refusal and hesitancy were lower education level, financial instability, Indigenous status, and not being concerned about spreading the coronavirus. Perceiving COVID-19 to be more of a threat to one’s community and believing that one had a higher risk of illness or death from COVID-19 decreased the likelihood of both vaccine refusal and hesitancy. Women and newcomers to Canada were more likely to be unsure about getting vaccinated. Respondents who did not plan to be vaccinated were less likely to wear face masks and practice physical distancing. Conclusion While many Canadians have voluntarily and eagerly become vaccinated already, reaching sufficient coverage of the population is likely to require targeted efforts to convince those who are resistant or unsure. Identifying and overcoming any barriers to vaccination that exist within the socio-demographic groups we found were least likely to be vaccinated is a crucial component.

Studies on work–family conflict amongst university faculty members indicate that women experience significantly more conflict in balancing their dual roles than their male counterparts. Research suggests that female faculty may be disadvantaged because of the norms structuring academic environments, which seemingly accommodate the life courses of men. Interestingly, the experience of work–family conflict for graduate students, who are besieged by many of the same environmental forces as female faculty, has been largely ignored within the scholarly literature. In the present study, qualitative responses regarding work–family conflict from 65 academic women (32 faculty; 33 graduate students) from universities and colleges across Canada were submitted to thematic analysis. Results revealed three interconnected themes: masculine workplace norms, the need to choose between work and family, and consequences of work–family conflict. The findings point to the need for academic institutions to critically examine their cultures surrounding motherhood in an effort to provide hospitable environments for faculty and graduate students who are, or who will become, parents.

Militaries are men-dominated and value a heteronormative masculine warrior ideal (Kaplan, 2003). Soldiers, however, are not necessarily heterosexual and men, nor do they always embody the prescribed warrior ideal. How the values related to hyper-masculinity and heteronormativity influence the experiences of gay and lesbian soldiers is an empirical question. In the present study, the Psycho-Social Ethnography of the Commonplace (P-SEC; Gouliquer & Poulin, 2005) methodology was used to investigate how the institutions of heterosexuality and the Canadian military shape gay and lesbian soldiers’ experiences. Interviews with 10 lesbian and 10 gay soldiers were conducted and analysed. The findings reveal that lesbian and gay soldiers face fairly widespread discrimination in the military environment. They must contend with the institutional demands to meet the requirements of a hyper-masculine-heteronormative soldier ideal. Gender differences are highlighted and social policy recommendations are discussed.

Introduction: Prejudice and discrimination against transgender individuals (i.e., transphobia) is pervasive and has been shown to have pernicious effects on the physical (e.g., substance abuse and other self-harm behaviors) and psychological (e.g., depression and suicidal ideation) well-being of those targeted.Aims: To date, a review of the psychometric properties of scales assessing transphobia has not been conducted; this gap compromises researchers' ability to make informed measurement decisions.Methods: In the current study, 61 articles that contained 83 scales were identified, and their adherence to best practices in psychometric testing was evaluated.Results: Most of the transphobia scales included in the current review did not provide sufficient information about item generation and refinement, scale dimensionality, scale score reliability, or validity. Eliciting broadly neutralizing antibodies (bNAbs) against the four dengue virus serotypes (DENV1-4) that are spreading into new territories is an important goal of vaccine design. To define bNAb targets, we characterized 28 antibodies belonging to expanded and hypermutated clonal families identified by transcriptomic analysis of single plasmablasts from DENV-infected individuals. Among these, we identified J9 and J8, two somatically related bNAbs that potently neutralized DENV1-4. Mutagenesis studies showed that the major recognition determinants of these bNAbs are in E protein domain I, distinct from the only known class of human bNAbs against DENV with a well-defined epitope. Phenotypic screening of antigen-specific antibodies in human blood is a common diagnostic test for infectious agents and a correlate of protection after vaccination. In addition to long-lived antibody secreting plasma cells residing in the bone marrow, memory B cells are a latent source of antigen-experienced, long-term immunity that can be found at low frequencies in circulating PBMCs. Assessing the genotype, clonal frequency, quality, and function of antibodies resulting from an individual’s persistent memory B cell repertoire can help inform the success or failure of immune protection. We have applied ELISPOT cell culture methods to functionally expand the memory repertoire from PBMCs and clonally map monoclonal antibodies from this population. We show that combining deep sequencing of stimulated memory B cell repertoires with retrieving single antigen-specific cells is a promising approach in evaluating the latent, functional B cell memory in PBMCs.

Although structural equation modelling (SEM) is a popular analytic technique in the social sciences, it remains subject to misuse. The purposes of this paper are to assist psychologists interested in using SEM by: 1) providing a brief overview of this method; and 2) describing best practice recommendations for testing models and reporting findings. We also outline several resources that psychologists with limited familiarity about SEM may find helpful.

The low-earth orbit environment contains small micrometeoroid and orbital debris (MMOD) particles traveling at characteristic velocities of several kilometers per second. In addition to being a direct threat to astronauts and spacecraft, upon impact with the exterior surface of a space vehicle, these highly energetic MMOD particles can create cut and puncture hazards for astronauts performing extra-vehicular activities (EVA). In this work, we demonstrate that replacing the standard neoprene-coated nylon absorber layers with woven aramid textiles intercalated with colloidal shear thickening fluids, i.e., STF-Armor™, can provide a meaningful enhancement to the cut and puncture resistance of the thermal micrometeoroid garment (TMG). Quasi-static puncture testing is performed using hypodermic needles of varying gauge to simulate the cutting and puncture hazards at deformation rates characteristic of human motion. At equal areal densities, we find that a TMG lay-up containing STF-Armor™ greatly improves puncture protection with a reduction in weight and comparable flexibility.

Researchers (e.g., Faludi, 1991; Schwartz, 1989) in the late 1980s and early 1990s explored and critiqued the phenomenon of the "mommy track" (i.e., prioritising mothering) by noting that professional mothers are relegated to lower-status and lowerpaying jobs because of perceived expectations associated with their parental roles (Cummins, 2012). In academic settings, the discrimination associated with perceived mommy tracking has been put forth to explain, in part, the "leaky pipeline" (van Anders, 2004) and "pyramid problem" (Mason, 2011). The leaky pipeline and pyramid problem are used to describe the tendency for university faculty who are women to occupy the lowest levels of the academic hierarchy. That is, at each successive level within academic institutions, the representation of women decreases (van Anders, 2004). Of the teaching positions at universities within Canada, 40% are occupied by women, almost half of them are mothers (CAUT, 2013), and so parental status may indeed be involved in the disproportionate representation of women at the highest academic ranks.Research has suggested that women faculty perceive the current climate in academic institutions to be less than welcoming to mothers (Castaneda & Isgro, 2013). Armenti (2004) interviewed 19 Canadian women professors who had engaged or anticipated having to engage in strategies to minimise the negative repercussions associated with their motherhood status (e.g., not being hired, negative effect on promotion and tenure reviews), and alleviate disapproval from colleagues. Results from the qualitative analysis revealed that participants with grown children reported having timed their pregnancies to coincide with the end of the academic year, and younger professors who might have started to have children opted to delay pregnancies until they had secured tenure. Overall, the perceived lack of support for academic mothers may cause some women to leave academia, delay tenure or promotions, or accept positions at teaching-focused, as opposed to research-focused, universities to prioritise their families (Damiano-Teixeira, 2006).In an effort to understand the barriers faced by academic women, Comer and Stites-Doe (2006) developed a theoretical model of three factors that complicate (or facilitate) one's ability to balance academic and parental roles. First, the phase of a woman's career (i.e., pre- or posttenure) and the stage of her family can affect a woman's role-balancing capacity. Women professors who have preschool-aged children, and women who are working toward tenure, have been theorized to experience greater difficulty. Second, the extent to which a woman's partner contributes to household and childcare tasks affects a woman's role balancing; specifically, the greater the contributions from one's partner, the better a woman's ability to balance roles. Indeed, compared with live-in support from other relatives, only spousal/partner support has been found to actually decrease the hours mothers work inside the home (Abroms & Goldscheider, 2002). Third, institutional factors (e.g., placement in a research- or teaching-focused university, work-family culture, and presence/absence of campus childcare) can affect women's role balancing.Despite the plethora of variables that could theoretically result in barriers for women, much of the empirical work examining differences between academic women and men has relied extensively on one key metric, namely, their respective publication records. Researchers consistently find that faculty men produce greater research output, and identify the ranks of assistant and full professor where this gap is most noticeable (e.g., Carleton, Parkerson, & Horswill, 2012; Nakhaie, 2002; Stack, 2004). Although number of publications is undoubtedly an important metric to consider, Ward and Wolf-Wendel (2004) caution that discrepancies in academic women's productivity (compared with academic men) may be due to breaks women take to have children. …

Human respiratory syncytial virus (RSV) is a single-stranded RNA virus that causes acute, and occasionally fatal, lower respiratory illness in young infants, the elderly, and immunocompromised patients. Therapeutic interventions able to cut short viral replication and quickly return the airways to normal function are needed. An understanding of antiviral activities and their effects on host defense mechanisms is important for the design of safe and effective therapy. We targeted functionally and temporally distinct steps within the viral life cycle using small-molecule RSV inhibitors and studied their antiviral activities and their effects on innate interferon responses of airway epithelial cells in vitro . Antivirals acting upstream of RSV polymerase activity (i.e., compounds targeting the fusion protein or the nucleoprotein) reduced viral load immediately postinfection and partially attenuated interferon responses. In contrast, antivirals directed to the RSV polymerase demonstrated activity throughout the viral replication cycle and specifically modulated the RIG-I/mitochondrial antiviral signaling protein (MAVS)/TBK1/IRF3/interferon-stimulated gene (ISG) axis, causing either an upregulation or a downregulation of interferon responses, depending on the mechanism of polymerase inhibition. Notably, polymerase inhibition leading to the accumulation of abortive RNA products correlated with the amplification of interferon-stimulated genes to up to 10 times above normal infection levels. Understanding how antiviral activities and their modulation of innate immunity may affect recovery from RSV infection will help guide the development of safe and effective therapies. IMPORTANCE RSV circulates seasonally, causing acute lower respiratory disease. Therapeutic interventions with efficacy throughout the viral replication cycle, rapid viral clearance, and prevention of potentially harmful inflammatory responses are desirable. Compounds targeting the RSV polymerase inhibited virus replication late in the viral life cycle and, depending on the functional domain targeted, either attenuated or amplified RIG-I and downstream interferon pathways in infected cells. These data will help guide the development of safe and effective therapies by providing new molecular evidence that the mechanism of inhibition by an antiviral compound can directly impact innate antiviral immune responses in the airway epithelium.

Little research has been conducted examining women’s perceptions of pornography, with the few studies available focusing almost exclusively on heterosexual and lesbian pornography. While these studies are valuable, anecdotal arguments suggest that gay male pornography may offer women an erotic alternative. This assertion was explored through a series of personal interviews conducted with women (n = 14) who perceive this type of pornography to be erotic. Three brief sequences, extracted from popular titles (i.e., Doing it Together: The Peters Twins, Folsom Filth, Miles and Ashton: Bareback), were reviewed and discussed. The data were thematically analysed with three overarching themes emerging: reasons for watching gay pornography, the importance of pornography being ‘believable,’ and homonegative and femi-negative reactions to the medium. All of Canada's provinces and territories legally recognize the right of gay and lesbian couples to adopt children; however, widespread acceptance of this practice has not been documented. Using an experimental design, with 506 university students, the present study assessed (1) attitudes toward gay, lesbian, and heterosexual adoptive couples; (2) the sex of the child to be adopted; (3) gender role characteristics of the adoptive couple; and (4) predictors of attitudes toward adoption by same-sex couples. Using vignettes describing potential adoptive couples, the results revealed that gay and lesbian couples were rated significantly less favorably than heterosexual couples when asked about outcomes for the adoptive child. Participants were more likely to approve of child placements with lesbian adoptive couples whose gender role characteristics emulated the traditional masculine/feminine dyad as compared to lesbian couples in which both partners displayed feminine characteristics. In this article, we introduce the concept of the service delivery network (SDN) defined as two or more organizations that, in the eyes of the customer, are responsible for the provision of a connected overall service experience. This responds to calls for frameworks recognizing that dyadic service encounters are embedded in the series of experiences customers have with complementary providers as part of the journey to achieve their desired goals. Adopting an SDN perspective presents a dramatically different set of challenges for managers and provides research opportunities challenging the current view of established service concepts. Managers must recognize that to better serve the customer they need to understand the role that they play in the customer-defined service journey and be prepared to coordinate their activities with complementary providers. Participating in helping build and manage the SDN for the customer, or understanding how they fit into customer’s self-designed SDN, becomes a central challenge, often requiring firms to develop a new set of capabilities. The SDN also challenges the way we view many of the core concepts in service research, which are anchored in the dyadic view. This provides considerable opportunity for future inquiry. We present a series of research questions, inspired by the SDN, organized into categories including building cooperative and collaborative networks, customer cocreation, systems thinking, customer relationship management, managing service failure and recovery, building capabilities, and customer-to-customer interactions.

A series of [b]-fused 6,7-diethynylquinoxaline derivatives have been synthesized through an imine condensation strategy to examine the effect of extended benzannelation on the thermal reactivity of enediynes. Absorption and emission spectra of the highly conjugated quinoxalenediynes were red-shifted approximately 100-200 nm relative to those of 1,2-diethynylbenzene. Strong exotherms indicative of enediyne cyclization were observed by differential scanning calorimetry, while solution cyclizations in the presence of 1,4-cyclohexadiene confirmed C(1)-C(6) Bergman cyclization. To provide further insight into Bergman cyclization energetics, computational studies were performed to compare changes in the cyclization enthalpy barrier, reaction enthalpy, and barrier of retro-Bergman ring-opening. Extension of benzannelation from 1,2-diethynylbenzene to either 2,3-diethynylnaphthalene or the 6,7-diethynylquinoxalines had a minimal effect on the cyclization barrier. In comparison, the enthalpies of cyclization were increased upon linearly extended benzannelation, which resulted in reduced barriers to retro-Bergman ring-opening. In addition, the orientation of extended benzannelation was found to have a significant effect on the cyclization endothermicity. In particular, 5,6-diethynylquinoxaline exhibited a 6.9 kcal/mol decrease in cyclization enthalpy compared to 6,7-diethynylquinoxaline due to increased aromatic stabilization energy in the respective angularly versus linearly fused azaacene cyclized products.

The purpose of this study was to determine whether water quality parameters commonly associated with primary productivity may be used to predict the susceptibility of a specific water body to exceed proposed fish consumption advisory limitation of 0.3 mg kg−1. South Dakota currently has nine lakes and impoundments that exceed fish tissue mercury advisory limits of 1.0 mg kg−1 total mercury, far exceeding US Environmental Protection Agency and Food and Drug Administration 0.3 mg kg−1 consumption criteria. Previous studies suggest that increased aquatic productivity may mitigate the effects of biological production and subsequent uptake of methyl mercury through bio-dilution; however, it is uncertain whether these trends may exist within highly alkaline and highly productive aquatic conditions common to South Dakota lakes and impoundments. Water quality parameters and fish tissue mercury data for northern pike and walleye were collected and assessed using existing South Dakota Department of Environment and Natural Resources and Game Fish and Parks data. The data was initially screened using both parametric linear regression and non-parametric Mann–Whitney rank sum comparisons and further assessed using binary logistic regression and stepwise logistic regression methodology. Three separate phosphorus measurements (total, total dissolved, and Trophic State Index) and pH were determined to significantly correlate with increased mercury concentrations for the northern pike-in-impoundments model. However, phosphorus surprisingly was not a strong predictor for the remaining scenarios modeled. For the northern pike-in-natural lakes models, alkalinity was the most significant water quality parameter predicting increased mercury concentrations. Mercury concentrations for the walleye-in-natural lakes models were further influenced by pH and alkalinity. The water quality and fish tissue mercury interrelationships determined within this study suggest aquatic productivity, and consequential eutrophication processes appear to be reasonable indicators of fish tissue mercury susceptibility for aquatic conditions common to South Dakota and highlight the continuing need to minimize eutrophication through effective watershed management strategies.

The VERITAS observatory is an imaging atmospheric Cherenkov telescope array located in southern Arizona and covers an energy range between 100 GeV and 30 TeV. The VERITAS collaboration pursues a rigorous observing program that targets a range of key science objectives in astrophysics and particle physics; the understanding of the origin of cosmic rays, the search for supersymmetric dark matter self-annihilation, illuminating the connection between black holes and relativistic jets and constraints to the cosmological diffuse infrared background. We provide a summary of results from the first two years of observations with the full 4-telecope array reported at RICAP09.

We report on observations of very high energy gamma rays from the shell-type supernova remnant (SNR) Cassiopeia A with the Very Energetic Radiation Imaging Telescope Array System stereoscopic array of four imaging atmospheric Cherenkov telescopes in Arizona. The total exposure time for these observations is 22 hr, accumulated between September and November of 2007. The gamma-ray source associated with the SNR Cassiopeia A was detected above 200 GeV with a statistical significance of 8.3 sigma. The estimated integral flux for this gamma-ray source is about 3% of the Crab-Nebula flux. The photon spectrum is compatible with a power law dN/dE proportional to E(-Gamma) with an index Gamma = 2.61 +/- 0.24(stat) +/- 0.2(sys). The data are consistent with a point-like source. We provide a detailed description of the analysis results and discuss physical mechanisms that may be responsible for the observed gamma-ray emission.

We present evidence that the very high energy (VHE, E > 100 GeV) gamma-ray emission coincident with the supernova remnant IC 443 is extended. IC 443 contains one of the best studied sites of supernova remnant/molecular cloud interaction and the pulsar wind nebula CXOU J061705.3+222127, both of which are important targets for VHE observations. VERITAS observed IC 443 for 37.9 hr during 2007 and detected emission above 300 GeV with an excess of 247 events, resulting in a significance of 8.3 standard deviations (sigma) before trials and 7.5 sigma after trials in a point-source search. The emission is centered at 6(h)16(m)51(s)+22 degrees 30'11 '' (J2000)+/- 0 degrees.03(stat)+/- 0 degrees.08(sys), with an intrinsic extension of 0 degrees.16 +/- 0 degrees.03(stat)+/- 0 degrees.04(sys). The VHE spectrum is well fit by a power law (dN/dE = N(0) x(E/TeV)(-Gamma)) with a photon index of 2.99 +/- 0.38(stat)+/- 0.3(sys) and an integral flux above 300 GeV of (4.63 +/- 0.90(stat)+/- 0.93(sys)) x 10(-12) cm(-2) s(-1). These results are discussed in the context of existing models for gamma-ray production in IC 443.

HESS J0632+057 is one of only two unidentified very-high-energy gamma-ray sources which appear to be point-like within experimental resolution. It is possibly associated with the massive Be star MWC 148 and has been suggested to resemble known TeV binary systems like LS I +61 303 or LS 5039. HESS J0632+057 was observed by VERITAS for 31 hours in 2006, 2008 and 2009. During these observations, no significant signal in gamma rays with energies above 1 TeV was detected from the direction of HESS J0632+057. A flux upper limit corresponding to 1.1% of the flux of the Crab Nebula has been derived from the VERITAS data. The non-detection by VERITAS excludes with a probability of 99.993% that HESS J0632+057 is a steady gamma-ray emitter. Contemporaneous X-ray observations with Swift XRT reveal a factor of 1.8+-0.4 higher flux in the 1-10 keV range than earlier X-ray observations of HESS J0632+057. The variability in the gamma-ray and X-ray fluxes supports interpretation of the ob ject as a gamma-ray emitting binary., Comment: 8 pages, 3 figures, Accepted for publication in The Astrophysical Journal

VERITAS is an array of four, 12-m-diameter, Cherenkov telescopes, designed to explore the very-high-energy gamma-ray sky in the energy band between 100 GeV and 50 TeV. Its construction and commissioning have occurred over the past two years and the array has been taking scientific data with three or more telescopes since November 2006. We present results from observations made with VERITAS during the past observing season, including new results on the distant blazar 1ES 1218 + 304 , the active galaxy M87 and the high-mass X-ray binary system LS I + 61 303. We also describe the plans in place for the coming observing seasons.

Our laboratory studies how the contractile cytoskeleton contributes to the process of phagocytosis. Because of its larger size and ease of manipulation, we chose the macrostomal cell of Tetrahymena vorax as our model for analysis of the distribution of proteins of prominent filamentous structures within the large oral apparatus (OA). Previous work in our laboratory identified centrin as a component of the fine filamentous reticulum (FFR) and actin and tetrin as colocalizing components of the coarse filamentous reticulum (CFR) and cross-connectives (CC) (J. Eukaryot. Microbiol., 51:253–257). Our new data also show that actin coimmunoprecipitates with tetrin proteins, confirming our actin–tetrin colocalization results. Because of its positioning around the cytostome, the actin-containing CFR/CC is a logical candidate for involvement in phagosome “pinch-off” following prey ingestion. We have analyzed this process by employing an assay that uses the addition of calcium to induce phagosome formation. We show that inhibitors of actin are able to block this event, indicating that actin is necessary for phagosome “pinch-off”. The OA also contains precisely arranged arrays of microtubules. We have examined the spatial relationship of the microtubular arrays to the distribution patterns of centrin, actin and tetrin, and found that tubulin and centrin fluorescences overlap in the region of the undulating membrane (UM). Tubulin fluorescence overlaps with actin and tetrin labeling at the inner edge of the CFR, where the CFR and UM converge. In addition to ciliary and oral rib labeling, tubulin antibodies also recognize the outer microtubule bundle (OMB), which delineates the right and posterior boundary of the OA.

The stalked ciliate, Vorticella convallaria, is a good model system to study mechanochemical motility because its contractile organelles (spasmoneme and myonemes) use a mode of contraction that differs from most other eukaryotic motile systems. Since calcium triggers this contraction, we have undertaken the molecular characterization of the calcium-binding proteins associated with these organelles. We have isolated and identified seven unique centrin-like cDNAs from V. convallaria. Each encodes an acidic protein of approximately 20-kDa, containing a unique N-terminus and four potential calcium-binding domains. We predict that each centrin has a distinct function within the cell. To define these functions, we have initiated immunofluorescence localization studies utilizing various anti-centrin antibodies. Western analysis indicates that each antibody recognizes a distinct protein or subset of proteins in Vorticella. Using these antibodies, we have localized centrin to various structures within the cell; myonemes, spasmoneme, and the oral apparatus. Because each of these antibodies recognizes a different protein on Westerern analysis, we conclude that a number of calcium-binding proteins are associated with the contractile organelles. To further characterize this gene family, we have initiated immunolocalization at the ultrastructural level. This will permit subcellular localization of all Vorticella centrins and enable us to dissect the function of this multi-gene family.

This study tests a path analytic model of buyer–supplier relationships, linking the supplier’s level of trust to three categories of inter-firm cooperative behaviors and these behaviors to the buyer’s perception of the relationship’s performance. Data was used from a survey of 164 dyads composed of a purchasing manager and a counterpart in a firm that they identified as their most cooperative suppliers. Higher levels of inter-organizational cooperative behaviors such as shared planning and flexibility in coordinating activities were found to be strongly linked to the supplier’s trust in the buyer firm. However, not all of the types of cooperative behaviors, particularly joint responsibility for problem solving, had significant impacts on the buyer’s perceptions of the relationship’s performance.

Despite many calls for case-based operations management research, the successful publication rate of such articles in top-tier journals has been less than stellar. A five step case-based research and dissemination process is presented. Guidance is given to future researchers for each step in the process. In addition, areas of weakness are examined and discussed in detail. Future potential research questions in operations management considered to be appropriate for the case-based method are highlighted.

We present optical, X-ray, high energy ($\lessapprox 30$ GeV) and very high energy ($\gtrapprox 100$ GeV; VHE) observations of the high-frequency peaked blazar Mrk 421 taken between 2008 May 24 and June 23. A high energy $\gamma$-ray signal was detected by AGILE with \sqrt{TS}=4.5 on June 9--15, with $F(E>100 \mathrm{MeV})= 42^{+14}_{-12}\times 10^{-8}$ photons cm$^{-2}$ s$^{-1}$. This flaring state is brighter than the average flux observed by EGRET by a factor of $\sim$3, but still consistent with the highest EGRET flux. In hard X-rays (20-60 keV) SuperAGILE resolved a 5-day flare (June 9-15) peaking at $\sim$ 55 mCrab. SuperAGILE, RXTE/ASM and Swift/BAT data show a correlated flaring structure between soft and hard X-rays. Hints of the same flaring behavior are also detected in the simultaneous optical data provided by the GASP-WEBT. A Swift/XRT observation near the flaring maximum revealed the highest 2-10 keV flux ever observed from this source, of 2.6 $\times 10^{-9}$ erg cm$^{-2}$ s$^{-1}$ (i.e. > 100 mCrab). A peak synchrotron energy of $\sim$3 keV was derived, higher than typical values of $\sim$0.5-1 keV. VHE observations with MAGIC and VERITAS on June 6-8 show the flux peaking in a bright state, well correlated with the X-rays. This extraordinary set of simultaneous data, covering a twelve-decade spectral range, allowed for a deep analysis of the spectral energy distribution as well as of correlated light curves. The $\gamma$-ray flare can be interpreted within the framework of the synchrotron self-Compton model in terms of a rapid acceleration of leptons in the jet., Comment: accepted for publication in ApJL, 7 pages, 2 figures, 1 table

Objectives Aspirin is recommended for cardiovascular disease (CVD) prevention in patients who are at high risk for CVD. The objective of this study was to compare agreement between two American Diabetes Association-endorsed CVD risk calculators in identifying candidates for aspirin therapy. Methods Adult patients with diabetes mellitus (n = 238) were studied for 1 year in a family medicine clinic. Risk scores were calculated based on the United Kingdom Prospective Diabetes Study Risk Engine and the Atherosclerosis Risk in Communities Coronary Heart Disease Risk Calculator. Analyses included χ(2), κ scores, and logistic regressions. Results The Atherosclerosis Risk in Communities Coronary Heart Disease Risk Calculator identified 50.4% of patients as high risk versus 23.5% by the United Kingdom Prospective Diabetes Study Risk Engine. κ score for agreement identifying high-risk status was 0.3642. Among patients at high risk, African Americans (odds ratio [OR] 0.45, 95% confidence interval [CI] 0.24-0.86) and those with uncontrolled diabetes (OR 0.30, 95% CI 0.16-0.56) had lower odds of disagreement, whereas nonsmokers had higher odds (OR 2.98, 95% CI 1.57-5.69). Among patients at low risk, women (OR 3.83, 95% CI 1.64-8.91), African Americans (OR 5.96, 95% CI 3.07-11.59), and those with high high-density lipoprotein (OR 2.82, 95% CI 1.48-5.37) showed greater odds of disagreement. Conclusions Improved risk assessment methods are needed to identify patients with diabetes mellitus who benefit from aspirin for the primary prevention of CVD. Prospective trials are needed to provide additional evidence for aspirin use in this population.

Supplier alliances have been widely touted but there are probably a limited number of situations where they are applicable. This paper develops a model of important factors that define which suppliers offer the best choices for pursuing alliance-like relationships. Some factors relate to the technology of the goods or services being sourced; others relate to the ability to develop mutual goodwill trust with the targeted supplier. The model was developed through a combination of extensive literature reviews and a series of interviews with managers in 15 manufacturing and service firms.

Spontaneous clearance of hepatitis C virus (HCV) is rare in immunocompromised patients, such as those who have undergone organ transplantation. It has been recognized that patients receiving liver transplantation for HCV-related disease have decreased graft and patient survival compared with those transplanted for other etiologies. There is a growing trend toward treating HCV recurrence aggressively after liver transplantation. For other organ transplant recipients with concurrent HCV, treatment is not often an option, given the high rates of graft rejection and loss secondary to interferon and its immunomodulatory effects. Although spontaneous clearance of HCV has been reported in recipients of solitary liver and renal transplants, a common factor arising in these cases has been previous exposure to interferon. To date, no reports of spontaneous clearance of HCV RNA have been reported in a multiorgan transplant recipient. A case of spontaneous clearance of HCV RNA in an immunocompromised patient, within five months of simultaneous liver and kidney retransplantation is described. Importantly, this patient had no previous exposure to interferon.

When a functional layout is converted to a cellular layout, the cell design is generally based on a static picture of production volume and part mix, but manufacturing environments face ongoing changes in these parameters. It is expected that, eventually, changes in production volume and part mix will cause a deterioration in cell performance to the point that a cell’s machine layout must be redesigned, marking the end of the cell’s life cycle. Tests the existence of cell life cycles and performance deterioration attributed to changes in production volume and part mix through an exploratory field study which was undertaken at 15 firms using cellular manufacturing. Finds that cell life cycles did exist, but usually either in anticipation of declining cell performance ‐ rather than in reaction to it ‐ or in anticipation of potential improvements in cell performance due to changes in the marketplace.

We extended the sensitivity of the ELISPOT assay by including an antigen-driven proliferation step prior to a final restimulation with antigen and irradiated antigen presenting cells (APCs). This improved sensitivity made the modified ELISPOT assay better suited to the detection of rare or low frequency T lymphocytes than the standard ELISPOT assay or alternatives such as limiting dilution analysis or in situ hybridization. Use of ELISA-grade plastic or polyvinylidene difluoride (PVDF) plates for the detection of different cytokines improved the signal-to-noise ratio for counting cytokine spots, and use of video computer imaging software improved objective quantitation. Analysis of antigen-reactive peripheral blood mononuclear cells (PBMC) from multiple sclerosis (MS) patients using both the traditional and our modified ELISPOT assay demonstrate a > 10-fold increase in numbers of myelin basic protein (MBP)-responsive T cells detected (an average of less than 1 spot forming cell (SFC) per 2 x 10(5) PBMC with the standard assay compared to 19 SFC per 2 x 10(5) PBMC with the modified assay). In addition, the modified ELISPOT assay could be performed with frozen PBMC, which permitted greater flexibility in sample processing, multiple use of a single sample as an internal standard, and simultaneous analysis of samples collected at different time points. This modified ELISPOT assay has many applications, including analysis of cytokine profiles in rare T cell populations, identification of antigen-responsive individuals as PBMC donors for T lymphocyte cloning or for therapeutic intervention, and assessment of vaccine or therapeutic efficacy as a surrogate clinical marker.

A recent trend in product development is the move to outsource more of the design and development work. Specifically, manufacturing firms are looking to component suppliers to increase their roles from that of supplying specified components to that of sharing in or taking major responsibility for component design and development. Effective teaming of product designers and the allied suppliers is likely to become an increasingly important element in the product innovation process. However, this role may be unfamiliar to both product development staff and component supplier staff. This study examines perceptions of new product designers about the component suppliers they used for the design of critical technology in recent new product development projects. The study focuses on how the suppliers were viewed in terms of their technical capability and how it contributed to the project, as well as their capability to work effectively with the product design team. Results indicate that the cooperativeness of the supplier was probably more influential than its technical competence in influencing the willingness of the product designer to bring the component supplier into future development projects.

Many managers are trying to leverage the skills and resources of key component suppliers to develop high-quality new products faster and at a lower cost. In this exploratory study, we surveyed product-development engineers in 79 small- to medium-sized firms to determine if three techniques commonly used to integrate suppliers into product development increase the supplier's perceived contributions to product development in the eyes of its customer. The techniques studied are: (1) timing of a supplier's involvement; (2) supplier's design responsibility; and (3) communication frequency. Of these, only early supplier involvement was significantly related to the perception of an increased contribution by the supplier. In addition, we found that the supplier's contributions to product development had little practical influence on the overall project technical success. The findings suggest that more research is needed to understand how to use suppliers effectively to improve product-development outcomes.

Sediment cores were collected from ten eutrophic lakes in South Dakota to determine the lateral extent of sediment-bound mercury (Hg) concentrations and to assess the relationship between watershed and land use characteristics with lake Hg fish tissue consumption advisory (>1.0 mg Hg/kg fish tissue) status. Advisory lakes were characterized as having higher sediment Hg and organic matter and lower total sulfur content compared to non-advisory lakes, and results highlight the importance of sulfide and organic carbon availability associated with potential Hg methylation biogeochemical processes. Advisory lakes generally had higher percentage of areal development and grasslands, higher catchment to lake area, and lower percentage of wetlands compared to non-advisory lakes. These results signify the importance of minimizing watershed sediment transport and associated organic carbon loading as effective Hg fish tissue lake management strategies.

Background Few studies have investigated the service needs of persons who inject drugs (PWID) who live in less populated regions of Canada. With access to fewer treatment and harm reduction services than those in more urban environments, the needs of PWID in smaller centres may be distinct. As such, the present study examined the needs of PWID in Prince Edward Island (PEI), the smallest of Canada's provinces. Methods Eight PWID were interviewed about the services they have accessed, barriers they faced when attempting to access these services, and what services they need that they are not currently receiving. Results Participants encountered considerable barriers when accessing harm reduction and treatment services due to the limited hours of services, lengthy wait times for treatment, and shortage of health care practitioners. They also reported experiencing considerable negativity from health care practitioners. Participants cited incidences of stigmatisation, and they perceived that health care practitioners received insufficient training related to drug use. Recommendations for the improvement of services are outlined. Conclusions The findings indicate that initiatives should be developed to improve PWID's access to harm reduction and treatment services in PEI. Additionally, health care practitioners should be offered sensitisation training and improved education on providing services to PWID. The findings highlight the importance of considering innovative alternatives for service provision in regions with limited resources.

Investigates the change in requirements to sustain supplier alliances as the relationship matures. Notes that the trend to outsource more materials and services has made firms more reliant on their sources of supply. This has led to a move towards increasingly collaborative supplier relationships, referred to as supplier alliances. Widespread use of these relationships is new and relatively little is known about the attributes that may promote success or failure. A recent paper concludes that such “partnership” relationships tend to expand and be more successful with each year following establishment. Tests this assumption using a longitudinal survey of 41 buyer‐supplier relationships. Finds that the purchasing firms perceived some improvements in benefits from their alliance‐like relationships but not significantly more than those which maintained traditional relationships. Also finds that the factors leading to improved benefits changed significantly as the alliance matured, with specific information flows becoming key. Proposes a profile of the relative importance of these factors over time.

Recent interest in alternative purchasing arrangements has centered on “supplier alliances,” an approach used to establish broader roles in the buyer-supplier relationship. Prior research about strategic supplier alliances has focused on the appropriate environments for developing these relationships and the potential benefits of such a strategy. This article addresses a different issue: implementation difficulties caused by supplier discontent. Information about this issue has been generated from a survey covering a wide array of firms. Responding buyers were requested to nominate exemplary suppliers for a follow-up survey. Responses from matched pairs of buyers and suppliers subsequently were compared for differences in their perceptions of their alliance. The degree to which their views differed was evaluated against several key hypotheses. This research interprets and suggests the management implications of the results.
