The energy supplied by the high-forage diets used in organic farming may be insufficient to meet the requirements of dairy cattle. However, few studies have considered this problem. The present study aimed to analyze the composition of the diets and the nutritional status (focusing on the energy–protein balance of the diets) of dairy cattle reared on organic farms in northern Spain, which are similar to other organic farming systems in temperate regions. Exhaustive information about diets was obtained from organic (ORG) and representative conventional grazing (GRZ) and conventional no-grazing (CNG) farms. Samples of feed from the respective farms were analyzed to determine the composition. Overall, the diets used on the ORG farms were very different from those used on the CNG farms, although the difference was not as evident for GRZ. The CNG farms were characterized by a higher total dry matter intake with a high proportion of concentrate feed, maize silage and forage silage. By contrast, on ORG and GRZ farms, the forage, pasture and fibre intake were the most important variables. The ration used on ORG farms contained a significantly higher percentage of ADF and lower organic matter (OM) content than the rations used in both of the conventional farming systems, indicating that the diets in the former were less digestible. Although the protein concentration in the diets used on the grazing farms (ORG and GRZ) was higher than those used on CNG farms, the protein intake was similar. The results indicated an imbalance between energy and protein due to the low level of energy provided by the ORG diets, suggesting that more microbial protein could be synthesized from the available rumen-degraded dietary nitrogen if rumen-fermentable OM was not limiting. The imbalance between energy and protein led to a reduced amount of total digestible protein reaching the intestine and a lower milk yield per kilogram of CP intake on the ORG farms. In order to improve the protein use efficiency and consequently to reduce the loss of nitrogen to the environment, organic farming should aim to increase the energy content of cattle diets by improving forage quality and formulating rations with more balanced combinations of forage and grain.

The present study aimed to evaluate trace mineral status of organic dairy herds in northern Spain and the sources of minerals in different types of feed. Blood samples from organic and conventional dairy cattle and feed samples from the respective farms were analysed by inductively coupled plasma mass spectrometry to determine the concentrations of the essential trace elements (cobalt (Co), chromium (Cr), copper (Cu), iron (Fe), iodine (I), manganese (Mn), molybdenum (Mo), nickel (Ni), selenium (Se) and zinc (Zn)) and toxic trace elements (arsenic (As), cadmium (Cd), mercury (Hg) and lead (Pb)). Overall, no differences between organic and conventional farms were detected in serum concentrations of essential and toxic trace elements (except for higher concentrations of Cd on the organic farms), although a high level of inter-farm variation was detected in the organic systems, indicating that organic production greatly depends on the specific local conditions. The dietary concentrations of the essential trace elements I, Cu, Se and Zn were significantly higher in the conventional than in the organic systems, which can be attributed to the high concentration of these minerals in the concentrate feed. No differences in the concentrations of trace minerals were found in the other types of feed. Multivariate chemometric analysis was conducted to determine the contribution of different feed sources to the trace element status of the cattle. Concentrate samples were mainly associated with Co, Cu, I, Se and Zn (i.e. with the elements supplemented in this type of feed). However, pasture and grass silage were associated with soil-derived elements (As, Cr, Fe and Pb) which cattle may thus ingest during grazing.

This study was designed to assess the value of seaweeds from the Galician coast as a source of minerals (especially iodine (I) but also other micro-minerals) in organic dairy cattle. It was conducted in an organic dairy farm in the Lugo province that typically represents the organic milk production in NW Spain. The animal’s diet consisted mainly of local forage (at pasture or as hay and silage in the winter) and 5 kg of purchased concentrate/day per animal (representing 23.5% of feed intake). Based on the mineral composition of the diet, the physiological requirements and the EU maximum authorised levels in feed, a supplement composed by Sea Lettuce (Ulva rigida) (as flakes, 80%), Japanese Wireweed (Sargasum muticum) (flakes, 17.5%) and Furbelows (Saccorhiza polyschides) (powder, 2.5%) was formulated to give 100 g/animal per day. Sixteen Holstein Friesian lactating cows were randomly selected and assigned to the control (n=8) and algae-supplemented groups (n=8). Both groups had exactly the same feeding and management with the exception of the algae supplement, which was mixed with the concentrate feed and given to the animals at their morning milking for 10 weeks. Heparinised blood (for plasma analysis) and milk samples were collected at 2-week intervals and analysed for toxic and trace element concentrations by inductively coupled plasma-mass spectrometry or inductively coupled plasma-optical emission spectrometry. The algae supplement significantly improved the animals’ mineral status, particularly I and selenium that were low on the farm. However, the effect of the algae supplement on the molybdenum status in cattle needs further investigation because of its great relevance on copper metabolism in ruminants. The I supply deserves special attention, since this element is at a very high concentration in brown-algae species and it is excreted in the milk proportionally to its concentration in plasma concentrations (mean±s.e. in the algae-supplemented and control groups were 268±54 and 180±42 µg/l, respectively).

The aim of this paper is to evaluate at a histopathological level the effect of the most commonly used copper (Cu) supplementation (15 mg/kg dry matter (DM)) in the liver of intensively reared beef cattle. This was done by a histochemistry evaluation of (i) the antioxidant capacity in the liver – by the determination of metallothioneins (MT) and superoxide dismutase (SOD) expression – as well as (ii) the possible induction of oxidative damage – by the determination of inducible nitric oxide synthase (iNOS), nitrotyrosine (NITT), malondialdehyde (MDA) and 8-oxoguanine (8-oxo) – that (iii) could increase apoptotic cell death – determined by cytochrome-c (cyto-c), caspase 1 (casp1) and terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL). Liver samples from Cu-supplemented (15 mg Cu sulphate/kg DM, n = 5) and non-supplemented calves (n = 5) that form part of other experiments to evaluate Cu status were collected at slaughter and processed for immunohistochemistry and TUNEL. MT expression was diffuse and SOD showed slight changes although without statistical significance. iNOS and NITT positive (+) cells significantly increased, mainly around the central veins in the animals from the Cu-supplemented group, whereas no differences were appreciated for the rest of the oxidative stress and apoptosis markers. Under the conditions of this study, which are the conditions of the cattle raised in intensive systems in NW Spain and also many European countries, routinely Cu supplementation increased the risk of the animals to undergo subclinical Cu toxicity, with no significant changes in the Cu storage capacity and the antioxidant defensive system evaluated by MT and SOD expression, but with a significant and important increase of oxidative damage measured by iNOS and NITT. The results of this study indicated that iNOS and NITT could be used as early markers of initial pathological changes in the liver caused by Cu supplementation in cattle, although more studies in cattle under different levels of Cu supplementation are needed.

The overall aim of the present study was to analyse and compare organic beef cattle farming in Spain with intensive and conventional systems. An on-farm study comparing farm management practices and animal health was carried out. The study also focussed on a slaughterhouse analysis by comparing impacts on the safety and quality of the cattle products. Twenty-four organic and 26 conventional farms were inspected, and farmers responded to a questionnaire that covered all basic data on their husbandry practices, farm management, veterinary treatments and reproductive performance during 2007. Furthermore, data on the hygiene and quality of 244, 2596 and 3021 carcasses of calves from organic, intensive and conventional farms, respectively, were retrieved from the official yearbook (2007) of a slaughterhouse. Differences found between organic and conventional farms across the farm analysis did not substantially reflect differences between both farm types in the predominant diseases that usually occur on beef cattle farms. However, calves reared organically presented fewer condemnations at slaughter compared with intensive and to a lesser extent with conventionally reared calves. Carcass performance also reflected differences between farm type and breed and was not necessarily better in organic farms.

Los costes asociados a la seguridad y salud laboral han sido ampliamente estudiados en diversas actividades industriales. Sin embargo, los modelos tradicionales aplicados para su estudio no resultan efectivos en las obras de construcción. Por otra parte, considerando que son las obras de construcción las que registran las mayores tasas de siniestralidad laboral, y que los accidentes llevan asociados importantes costes, no solo para la sociedad sino también para las empresas en los que ocurren, el objetivo de esta investigación se ha centrado en analizar el alcance que presenta el sistema de gestión de los costes de seguridad en las empresas de construcción. Para ello, se ha llevado a cabo un estudio empírico, al objeto de analizar las posibles relaciones existentes entre el coste de prevención y el coste de los accidentes ocasionado en estas empresas, a partir de una muestra de 40 obras de construcción, en las que se han producido un total de 178 accidentes. En dicho estudio se ha puesto de manifiesto que en las fases de obra de mayor riesgo identificadas, estructuras y movimiento de tierras, se asumen los mayores costes de prevención, a pesar de que también se obtienen los peores datos de siniestralidad.

This paper examines how variations in the height and health of Mexicans during the second half of the twentieth century reflect the evolution of economic inequality, as its effects have repercussions on the health and nutritional conditions of the population. The average height of Mexican adults had a modest increase with respect to the possibilities of human plasticity. These anthropometric variations were the result of the incorporation of advances in science and technology leading to improved standards of living among the population. Body changes were impacted by dietary habits, urbanization, and government policies supporting food production and distribution.

Recent ecotoxicological studies have indicated mercury (Hg) contamination in aquatic ecosystems in the Amazon Basin. Although Hg contamination can be associated with small-scale gold mining, the soils of the Amazon region have naturally high Hg concentrations, and can be transported to aquatic ecosystems via deforestation and mining activities. Biomagnification of Hg can pose risks to the local human population; therefore, its concentration in fish tissues must be monitored consistently. Fast and sensitive Hg determination is required for continuously monitoring ecosystems impacted by mineral exploration. The direct mercury analyzer (DMA-80) is widely used for determining total Hg levels in tissue samples; it is fast and cost-effective, without requiring sample preparation. Here, we determined the sensitivity and specificity of Hg detection accomplished using DMA-80, and whether these results are reliable compared to those obtained using Inductively Coupled Plasma Mass Spectrometer (ICP-MS), which is the gold standard. We obtained 106 paired dried samples of muscle tissue from fish species occupying different trophic levels in the Lower Amazon region, and analyzed them using both equipment (DMA-80 and ICP-MS). The results obtained using DMA-80 had an overall Hg mean of 1.90 ± 0.18 mg/kg which was higher (p < 0.05) than the mean of those obtained using ICP-MS (1.55 ± 0.13 mg/kg). Linear regression analysis comparing the Hg levels obtained using both devices was within the 95% prediction interval, and a high coefficient of correlation showed agreement between the devices (r = 0.979; 0.069 to 0.986, 95% CI). Bland-Altman analysis showed that DMA-80 had a positive bias of 6.5% in relation to ICP-MS, which is more evident in samples with high Hg concentrations. The main objective was to determine the chemical, phytochemical, fatty acid and mineral profiles of three commercially relevant brown macroalgae ( Laminaria digitata , Laminaria hyperborea and Ascophyllum nodosum ) collected each season for two years off the west coast of Ireland. All the chemical, phytochemical, fatty acid and minerals analysed varied significantly depending on the macroalgal species, season and year of collection. Overall, the protein contents of macroalgae were negatively correlated with carbohydrate content. Protein (2-11%) was at its highest during winter and/or spring, decreasing to a minimum during summer and/or autumn. The three macroalgal species analysed in this study had clearly differentiated fatty acid profiles. The concentration of fatty acids was higher in A. nodosum compared with both Laminaria species. The mineral profile of the three macroalgal species was rich in essential metals, particularly Ca, Mg and P, while the levels of I were approximately 9- to 10-fold higher in both Laminaria spp. compared with A. nodosum . The levels of toxic metals (Cd, Hg and Pb) in all the macroalgal species studied were low in the current study; while the levels of total As were high (49-64 mg/kg DW macroalgae) compared with previous reports.

Anorectal manometry is a diagnostic technique used to investigate the correct mechanical performance of the internal anal sphincter (IAS). By distending the rectal ampulla while recording changes in the luminal pressure, this method allows for characterizing the anorectal reflex. It can also provide, indirectly, information about the electrical activity of the IAS. In this study, seventeen neonates having 24-hour delayed passage of meconium or presenting distal intestinal obstruction symptoms underwent anorectal manometry to discard Hirschsprung's disease. All patients had normal anorectal reflex. The time delay between stimulation of the rectal ampulla and the relaxation of the anal canal was studied. The average period of the pressure fluctuations was 5.44 ± 0.13s. The overall duration of the relaxation time was 9.71 ± 0.21s. The maximum lag between the onset of the stimulus and the relaxation of the IAS was 2.90 s, and was achieved when the stimulus was applied following a local maximum of the pressure wave. The existence of a refractory period during the suprathreshold depolarization of smooth muscle cells can explain the evidence of a temporal delay between the stimulus and the mechanical response. In occasions, relaxation appeared first distally. This phenomenon can be explained by the arrangement and morphology of bipolar cells, which may evidence the anisotropic propagation of the mechanical activity. These data may contribute to depict the alterations in excitability underlying the relaxation reflex by means of manometric recording of the anal canal.

This article presents analyses of soil and environmental information for the Darro River basin (Granada-Spain) preliminary to its hydrological and forestry restoration. These analyses were carried out using a geographical information system (GIS) and employing a new procedure that adapts hydrological forest-restoration methods. The complete analysis encompasses morphological conditions, soil and climate characteristics as well as vegetation and land use. The study investigates soil erosion in the basin by using Universal Soil Loss Equation (USLE) and by mapping erosion fragility units. The results are presented in a set of maps and their analysis, providing the starting point for river basin management and the hydrological and forestry-restoration project that was approved at the end of 2015. The presence of soft substrates (e.g. gravel and sand) indicates that the area is susceptible to erosion, particularly the areas that are dominated by human activity and have little soil protection. Finally, land use and vegetation cover were identified as key factors in the soil erosion in the basin. According to the results, river authorities have included several measures in the restoration project aimed at reducing the erosion and helping to recover the environmental value of this river basin and to include it in recreation possibilities for the community of Granada. A protein extract from the brown seaweed Himanthalia elongata (Linnaeus) S. F. Gray was prepared and its functional properties, colour and amino acid composition were assessed for its potential future use by the food industry. The total content of amino acids was determined as 54.02±0.46gaminoacids/kg dry weight, with high levels of the essential amino acids lysine and methionine. SDS-PAGE showed 5 protein bands with molecular weights of 71.6, 53.7, 43.3, 36.4 and 27.1kDa. The water holding capacity and oil holding capacity were determined as 10.27±0.09gH 2 O/g and 8.1±0.07goil/g respectively. Foaming activity and stability were higher at alkaline pH values. The emulsifying capacity and stability of the extract varied depending on the pH and oil used. Following a request from the European Commission, the EFSA Panel on Additives and Products or Substances used in Animal Feed (FEEDAP) was asked to deliver a scientific opinion on the safety and efficacy of Calsporin ® when used in feed for sows and piglets. The additive contains viable spores of a single strain of Bacillus subtilis . This species is considered by EFSA to be suitable for the qualified presumption of safety (QPS) approach to safety assessment, which requires the identity of the strain to be conclusively established, evidence that the strain is not toxigenic and that it does not show resistance to antibiotics of human and veterinary importance. The strain was found to meet the criteria for the QPS approach in the context of previous opinions and since concerns are not expected from other components of the additive, Calsporin ® is presumed safe for all target species, consumers and the environment. In a previous opinion, the FEEDAP Panel concluded that Calsporin ® is not a dermal/eye irritant or a skin sensitiser but should be considered a potential respiratory sensitiser. Use of the additive in feed for sows and suckling piglets will not introduce hazards for users not already considered. Three trials performed with sows over a complete reproductive cycle and their offspring showed positive effects on zootechnical parameters of sows and piglets. Therefore, Calsporin ® has the potential to improve one or more performance parameters of sows and suckling piglets when supplemented to both sows and suckling piglets at 3 × 10 8  CFU/kg of complete feedingstuffs. In the view of the FEEDAP Panel, the effects observed can predominantly be traced back to the administration of the additive to pregnant/lactating sows. Introduction: The evaluation of symptom association between gastroesophageal reflux and cardiorespiratory events in preterm infants remains unclear. This paper describes a conservative approach to decision-making of anti-reflux surgery through symptom association analysis., Methods: Forty-three neonates with potentially reflux-related cardiorespiratory symptoms underwent synchronized esophageal impedance-pH and cardiorespiratory monitoring. Three indices were considered to evaluate symptom association, the symptom index (SI), the symptom sensitivity index (SSI) and the symptom association probability (SAP). A conservative strategy was adopted regarding the decision of anti-reflux surgery, and therefore, patients were scheduled for laparoscopic Nissen fundoplication if the three indices showed a positive assessment of symptom association. Retrospectively, these indices and the binomial symptom index (BSI) were contrasted against the decision of anti-reflux surgery using different windows of association., Results: Thirteen patients showed positive symptom association but only two underwent anti-reflux surgery. The SI and the SSI showed an increasing trend with the width of the window of association. The SAP was affected randomly by slightly altering the windowing parameters. The BSI showed the best performance with the two-minute window (κ =0.78) CONCLUSIONS: The pathology under study is known to improve with maturity. However, the severity of cardiorespiratory symptoms may threaten the neonate's life and therefore, in some occasions, invasive treatments must be considered to protect life. The BSI provides a good prediction of a combination of positive SI, SSI and SAP, which may improve clinical decisions. Previous contributions suggested that gastroesophageal reflux can be modeled in terms of a Poisson process. This study aims to provide empirical data to validate this statement in pediatric patients so that computational models can be broadly used as an alternative for research. A retrospective review of 63 pediatric patients who underwent 24-hour impedance-pH monitoring to discard gastroesophageal reflux disease was conducted in this study. Patients were grouped by age as preterm (21), infants (21), and children (21). All the tracings were analyzed by a trained physician who identified the reflux entry and the bolus clearance time of each episode. The time between reflux episodes was tested against three probability distributions (gamma, exponential and inverse Gaussian) whereas the bolus clearance time was tested against a normal probability distribution. Parameters were estimated using the maximum likelihood method. The Kolmogorov-Smirnov test and the Kullback-Leibler divergence were computed to evaluate the goodness of fit. One-way analysis of variance was applied to compare results along the three groups. Exponential fitting for inter-reflux time was successful in 90.48% of children older than 1 year. The overall reflux rate was 57.58 reflux episodes per day whereas the mean bolus clearance time ranged between 10.87 in preterm subjects and 12.05 in children, showing a good Gaussian fitting. The time between reflux episodes can be modeled in terms of a Poisson process in non tube-fed patients, whereas the bolus clearance time follows a normal distribution in all cases., (© 2014 International Society for Diseases of the Esophagus.)

Gastroesophageal monitoring is limited to 96 hours by the current technology. This work presents a computational model to investigate symptom association in gastroesophageal reflux disease with larger data samples proving important deficiencies of the current methodology that must be taking into account in clinical evaluation. A computational model based on Monte Carlo analysis was implemented to simulate patients with known statistical characteristics Thus, sets of 2000 10-day-long recordings were simulated and analyzed using the symptom index (SI), the symptom sensitivity index (SSI), and the symptom association probability (SAP). Afterwards, linear regression was applied to define the dependency of these indexes with the number of reflux, the number of symptoms, the duration of the monitoring, and the probability of association. All the indexes were biased estimators of symptom association and therefore they do not consider the effect of chance: when symptom and reflux were completely uncorrelated, the values of the indexes under study were greater than zero. On the other hand, longer recording reduced variability in the estimation of the SI and the SSI while increasing the value of the SAP. Furthermore, if the number of symptoms remains below one-tenth of the number of reflux episodes, it is not possible to achieve a positive value of the SSI. A limitation of this computational model is that it does not consider feeding and sleeping periods, differences between reflux episodes or causation. However, the conclusions are not affected by these limitations. These facts represent important limitations in symptom association analysis, and therefore, invasive treatments must not be considered based on the value of these indexes only until a new methodology provides a more reliable assessment., (© 2012 Copyright the Authors. Background: The evaluation of symptom association in gastroesophageal reflux is an open problem. The scientific literature reports important deficiencies and clinicians are claiming a new methodology. This article provides an optimal method for the evaluation of symptom association, the binomial symptom index (BSI)., Methods: A mathematical description of the BSI was presented for the study of association and causality. A total of n = 850,000 patients were simulated using a Monte Carlo model to perform a two-way sensitivity analysis. The average and the standard deviation of the BSI were evaluated in groups of 5000 patients with the same values of the reflux rate, symptom rate, association ratio, window of association, and monitoring time in order to contrast their influence on the estimator., Key Results: The BSI decreased with the number of reflux episodes when there was association, and remained constant and below 40% when there was not. The standard deviation was no higher than 40% and decreased with the reflux or symptom rates, and more sharply with the monitoring time, reaching approximately 0% for 50 days. A window length matching the characteristic reflux-symptom lag maximized the overall BSI and minimized its dispersion. Twenty-four hour and 96-h monitorings allowed detecting association ratios of 50% and 25%, respectively., Conclusions & Inferences: The BSI is a simple and reliable index for the evaluation of symptom association that considers all the parameters under analysis. Gastroesophageal reflux (GER) disease is a serious complication of the upper gastrointestinal tract. Cardiorespiratory symptoms such as apnea, oxygen desaturation and bradycardia may be related to GER. Thus, the recommended diagnostic methodology in pediatric patients requires 24-h synchronized esophageal and cardiorespiratory monitoring. However, there is no computer tool available for this purpose and therefore, researchers and physicians are forced to seek for customized solutions. This paper presents an open source computer program for the analysis of symptom association. It allows a convenient visualization of the biological signals and implements the three main metrics for symptom association, that is, the symptom index, the symptom sensitivity index and the symptom association probability. This software represents a flexible solution and will facilitate caregivers an easy assessment of the existence of temporal association between GER and cardiorespiratory episodes. Purpose: Hirschsprung disease (HSCR) is a developmental disorder caused by a failure of neural crest cells to migrate, proliferate, and/or differentiate during the enteric nervous system development. It presents a multifactorial, nonmendelian pattern of inheritance, with several genes playing some role in its pathogenesis. Its major susceptibility gene is the RET protooncogene, which encodes a receptor tyrosine kinase activating several key signaling pathways in the enteric nervous system development. Given the pivotal role of RET in HSCR, the genes encoding their ligands (GDNF, NRTN, ARTN, and PSPN) are also good candidates for the disease., Methods: We have performed a case-control study using Taqman technology to evaluate 10 polymorphisms within these genes, as well as haplotypes comprising them, as susceptibility factors for HSCR., Results: No differences were found in the allelic frequencies of the variants or in the haplotype distribution between patients and controls. In addition, no particular association was detected of the variants/haplotypes to any demographic/clinical parameters within the group of patients., Conclusion: These data would be consistent with the lack of association between these polymorphisms and HSCR, although they do not permit to completely discard a possible role of other variants within these genes in the disease. Moreover, because the gene-by-gene approach does not take into account the polygenic nature of HSCR disease, it would be interesting to investigate sets of variants in many other different susceptibility loci described for HSCR, which may permit to consider possible interactions among susceptibility genes.

Hirschsprung disease (HSCR) is a congenital disorder caused by a failure of neural crest cells to migrate, proliferate, and/or differentiate during the enteric nervous system (ENS) development. The requirement of the NTF-3/TrkC signaling for the proper development of the ENS, together with the evidences presented by animal models, led us to investigate the involvement of NTF-3 gene in HSCR. We performed both a mutational screening of NTF-3 and a complete evaluation of 3 polymorphisms as genetic susceptibility factors for HSCR. We identified a novel sequence variant, G76R, present in 2 different patients and absent in controls. We postulate that this variation could generate a lack of mature functional NTF-3 proteins in neural crest cell precursors; thus, altering the NTF-3/TrkC signaling pathway and influencing in the adequate ENS development. Although these results do not provide complete assurance of the involvement of this gene in HSCR, given the polygenic nature of the disease and its etiology, investigation of the genes encoding protein members of the signaling pathways governing the ENS development could provide new key findings in the elucidation of this complex disease.

In the last few years, the detection of free radical damage and the body's defences against it have become increasingly important in clinical medicine as a complementary tool in the evaluation of metabolic status. The aim of this study was to evaluate, under field conditions, the anti-oxidant status of healthy cows during late pregnancy and lactation onset using two parameters: (1) plasma levels of malondialdehyde (MDA), a degradation product of lipid peroxidation, and (2) total antioxidant status (TAS). Results were compared with those obtained in another group of cows with lesser metabolic demands. We also investigated possible relationships between antioxidant status markers and other relevant blood parameters. Our results confirmed the characteristic metabolic changes associated with late pregnancy and early lactation. MDA and TAS provided an accurate reflection of the internal physiological status of the animal. The data indicated increased lipid peroxidation around parturition, but with wide individual variations that may be attributable not only to the physiological stage but also to unknown factors that will have to be further considered in future studies.
