In this study we present the results of an emission study of different phytoplankton samples in aqueous media treated with elevated ozone levels. Halocarbon measurements show that the samples tested released bromoform and different iodocarbons, including iodomethane, iodochloromethane and diiodomethane. Iodide and iodate levels in the liquid phase were representative of concentrations of surface water in a natural environment. Measurement of volatile iodine (I2) emissions from two diatom samples (Mediopyxis helysia and Porosira glacialis) and the background sample (F/2 medium from filtered natural seawater) showed that the quantity of evolved I2 depends on the ozone concentration in the air. This behaviour was assumed to be caused by the oxidation reaction mechanism of iodide with ozone. The I2 emission flux agrees with model calculations at different iodide concentrations. The I2 emission of a natural plankton concentrate sample was, however, very low compared to other samples and showed no dependence on ozone. The reason for this was shown to be the low iodide concentration in the algal suspension, which seems to be the limiting factor in the oxidative formation of I2.

Based on the results of a pilot study in 2007, which found high mixing ratios of molecular iodine (I2) above the intertidal macroalgae (seaweed) beds at Mweenish Bay (Ireland), we extended the study to nine different locations in the vicinity of Mace Head Atmospheric Research Station on the west coast of Ireland during a field campaign in 2009. The mean values of chem{I_2} mixing ratio found above the macroalgae beds at nine different locations ranged from 104 to 393 ppt, implying a high source strength of I2. Such mixing ratios are sufficient to result in photochemically driven coastal new-particle formation events. Mixing ratios above the Ascophyllum nodosum and Fucus vesiculosus beds increased with exposure time: after 6 h exposure to ambient air the mixing ratios were one order of magnitude higher than those initially present. This contrasts with the emission characteristics of Laminaria digitata, where most I2 was emitted within the first half hour of exposure. Discrete in situ measurements (offline) of I2 emission from ambient air-exposed chamber experiments of L. digitata, A. nodosum and F. vesiculosus substantially supported the field observations. Further online and time-resolved measurements of the I2 emission from O3-exposed macroalgal experiments in a chamber confirmed the distinct I2 emission characteristics of A. nodosum and F. vesiculosus compared to those of L. digitata. The emission rates of A. nodosum and F. vesiculosus were comparable to or even higher than L. digitata after the initial exposure period of ~20–30 min. We suggest that A. nodosum and F. vesiculosus may provide an important source of photolabile iodine in the coastal boundary layer and that their impact on photochemistry and coastal new-particle formation should be reevaluated in light of their longer exposure at low tide and their widespread distribution.

In this work, we describe a simple diffusion capillary device for the generation of various organic test gases. Using a set of basic equations the output rate of the test gas devices can easily be predicted only based on the molecular formula and the boiling point of the compounds of interest. Since these parameters are easily accessible for a large number of potential analytes, even for those compounds which are typically not listed in physico-chemical handbooks or internet databases, the adjustment of the test gas source to the concentration range required for the individual analytical application is straightforward. The agreement of the predicted and measured values is shown to be valid for different groups of chemicals, such as halocarbons, alkanes, alkenes, and aromatic compounds and for different dimensions of the diffusion capillaries. The limits of the predictability of the output rates are explored and observed to result in an underprediction of the output rates when very thin capillaries are used. It is demonstrated that pressure variations are responsible for the observed deviation of the output rates. To overcome the influence of pressure variations and at the same time to establish a suitable test gas source for highly volatile compounds, also the usability of permeation sources is explored, for example for the generation of molecular bromine test gases.

Knowledge of the composition and emission rates of iodine-containing volatiles from major widespread seaweed species is important for modeling the impact of halogens on gas-phase atmospheric chemistry, new particle formation, and climate. In this work, we present the application of mass spectrometric techniques for the quantification of short-lived iodine-containing volatiles emitted by eight different seaweeds from the intertidal zone of Helgoland, Germany. A previously developed online time-of-flight aerosol mass spectrometric method was used to determine I(2) emission rates and investigate temporally resolved emission profiles. Simultaneously, iodocarbons were preconcentrated on solid adsorbent tubes and quantified offline using thermodesorption-gas chromatography-mass spectrometry. The total iodine content of the seaweeds was determined using microwave-assisted tetramethylammonium hydroxide extraction followed by inductively coupled-plasma mass spectrometry analysis. The highest total iodine content was found in the Laminariales, followed by the brown algae Ascophyllum nodosum, Fucus vesiculosus, Fucus serratus, and both red algae Chondrus crispus and Delesseria sanguinea. Laminariales were found to be the strongest I(2) emitters. Time series of the iodine release of Laminaria digitata and Laminaria hyperborea showed a strong initial I(2) emission when first exposed to air followed by an exponential decline of the release rate. For both species, I(2) emission bursts were observed. For Laminaria saccharina und F. serratus, a more continuous I(2) release profile was detected, however, F. serratus released much less I(2). A. nodosum and F. vesiculosus showed a completely different emission behavior. The I(2) emission rates of these species were slowly increasing with time during the first 1 to 2 h until a more or less stable I(2) emission rate was reached. The lowest I(2) emission rates were detected for the red algae C. crispus and D. sanguinea. Total iodocarbon emission rates showed almost the same general trend, however, the total iodocarbon emission rates were about one to two orders of magnitude lower than those of molecular iodine, demonstrating that I(2) is the major iodine containing volatile released by the investigated seaweed species. In addition, a clear dependency of iodocarbon emission from the ozone level (0-150 ppb O(3)) was found for L. digitata.

Here we present a new application of a time-of-flight aerosol mass spectrometer (TOF-AMS) for the measurement of atmospheric trace gases in real-time. Usually, TOF-AMS instruments are not sensitive to gas-phase species due to the aerodynamic particle focusing inlet system which reduces the gas phase species by a factor of about 10(7) relative to the particle phase. This efficient removal of the gas phase and the resulting high relative enrichment of particles is one reason for the very high sensitivity of TOF-AMS instruments for particle phase compounds (detection limits in the sub-μg/m(3)-range for online measurements with 1 min integration time), which allows application of the instruments even under clean atmospheric conditions. Here we use artificially generated particles as sampling probes to transfer selected atmospheric trace gases into the particle phase before entering the AMS (gaseous compound trapping in artificially generated particles-AMS, GTRAP-AMS). The sampling probe particles are mixed with the gaseous analytes upstream of the TOF-AMS in a 0.5 L flow tube. As an exemplary application of the method, the measurement of trace levels of gaseous molecular iodine is demonstrated. α-Cyclodextrin (α-CD/NH(4)Br) particles are used as selective sampling probes to transfer molecular iodine into the AMS. A detection limit in the subparts-per-billion (sub-ppb) range was achieved. The method was compared to a recently developed off-line method that combines denuder sampling of gaseous I(2) and gas chromatography/mass spectrometry (GC/MS) analysis. To demonstrate the usability of the method, temporally resolved I(2) emission profiles from a brown algae species (Laminaria saccharina) under exposure of ambient ozone levels were investigated. Total I(2) release rates of 36.5 pmol min(-1) grams fresh weight (gFW)(-1) at 100 pbb O(3) and 33.4 pmol min(-1) gFW(-1) at 50 ppb O(3) were obtained within the first hour of ozone exposure., (© 2011 American Chemical Society)

Neuronal morphogenesis, the growth and arborization of neuronal processes, is an essential component of brain development. Two important but seemingly disparate components regulating neuronal morphology have previously been described. In the hippocampus, neurotrophins, particularly brain-derived neurotrophic factor (BDNF) and neurotrophin-3 (NT3), act to enhance cell growth and branching, while activity-induced branching was shown to be dependent upon intracellular beta-catenin. We now describe a molecular link between NT3 stimulation and beta-catenin increase in developing neurons and demonstrate that this process is required for the NT3-mediated increase in process branching. Here, we show that beta-catenin is rapidly increased specifically in growth cones following NT3 stimulation. This increase in beta-catenin is protein synthesis dependent and requires the activity of cytoplasmic polyadenylation element-binding protein-1 (CPEB1), an mRNA-binding protein that regulates mRNA translation. We find that CPEB1 protein binds beta-catenin mRNA in a CPE-dependent manner and that both localize to growth cones of developing hippocampal neurons. Both the NT3-mediated rapid increase in beta-catenin and process branching are abolished when CPEB1 function is inhibited. In addition, the NT3-mediated increase in beta-catenin in growth cones is dependent upon internal calcium and the activity of CaMKII (calcium/calmodulin-dependent kinase II). Together, these results suggest that CPEB1 regulates beta-catenin synthesis in neurons and may contribute to neuronal morphogenesis.

The ability of neurons to modify synaptic connections is critical for proper brain development and function in the adult. It is now clear that changes in synaptic strength are often accompanied by changes in synaptic morphology. This synaptic plasticity can be maintained for varying lengths of time depending on the type of neuronal activity that first induced the changes. Long-term synaptic plasticity requires the synthesis of new proteins, and one mechanism for the regulation of experience-induced protein synthesis in neurons involves cytoplasmic polyadenylation element binding protein (CPEB1). CPEB1 can bidirectionally regulate mRNA translation, first repressing translation, and then activating translation after the phosphorylation of two critical residues (T171 and S177). To determine the full extent of CPEB1-mediated protein synthesis in synaptic function, we engineered a line of mice expressing CPEB1 with these phosphorylation sites mutated to alanines (mCPEB1-AA) exclusively in cerebellar Purkinje neurons (PNs). Thus, mRNAs bound by mCPEB1-AA would be held in a translationally dormant state. We show that mCPEB1-AA localizes to synapses in cerebellum and resulted in a loss of protein synthesis-dependent phase of parallel fiber-PN long-term depression. This was accompanied by a change in spine number and spine length that are likely attributable in part to the dysregulation of IRSp53, a protein known to play a role in synaptic structure. Finally, mCPEB1-AA mice displayed a significant impairment of motor coordination and a motor learning delay.

Long-term synaptic plasticity is both protein synthesis-dependent and synapse-specific. Therefore, the identity of the newly synthesized proteins, their localization, and mechanism of regulation are critical to our understanding of this process. Tissue plasminogen activator (tPA) is a secreted protease required for some forms of long-term synaptic plasticity. Here, we show tPA activity is rapidly increased in hippocampal neurons after glutamate stimulation. This increase in tPA activity corresponds to an increase in tPA protein synthesis that results from the translational activation of mRNA present at the time of stimulation. Furthermore, the mRNA encoding tPA is present in dendrites and is rapidly polyadenylated after glutamate stimulation. Both the polyadenylation of tPA mRNA and the subsequent increase in tPA protein is dependent on metabotropic glutamate receptor (mGluR) activation. A similar mGluR-dependent increase in tPA activity was detected after stimulation of a synaptic fraction isolated from the hippocampus, suggesting tPA synthesis is occurring in the synaptodendritic region. Finally, we demonstrate that tPA mRNA is bound by the mRNA-binding protein CPEB (cytoplasmic polyadenylation element binding protein-1), a protein known to regulate mRNA translation via polyadenylation. These results indicate that neurons are capable of synthesizing a secreted protein in the synaptic region, that mGluR activation induces mRNA polyadenylation and translation of specific mRNA, and suggest a model for synaptic plasticity whereby translational regulation of an immediate early gene precedes the increase in gene transcription.
