Reliable neutron star mass measurements are key to determining the equation-of-state of cold nuclear matter, but these are rare. "Black Widows" and "Redbacks" are compact binaries consisting of millisecond pulsars and semi-degenerate companion stars. Spectroscopy of the optically bright companions can determine their radial velocities, providing inclination-dependent pulsar mass estimates. While inclinations can be inferred from subtle features in optical light curves, such estimates may be systematically biased due to incomplete heating models and poorly-understood variability. Using data from the Fermi Large Area Telescope, we have searched for gamma-ray eclipses from 49 spider systems, discovering significant eclipses in 7 systems, including the prototypical black widow PSR B1957$+$20. Gamma-ray eclipses require direct occultation of the pulsar by the companion, and so the detection, or significant exclusion, of a gamma-ray eclipse strictly limits the binary inclination angle, providing new robust, model-independent pulsar mass constraints. Understanding the regulation of photosynthetic light harvesting and electron transfer is of great importance to efforts to improve the ability of the electron transport chain to supply downstream metabolism. A central regulator of the electron transport chain is ATP synthase, the molecular motor that harnesses the chemiosmotic potential generated from proton-coupled electron transport to synthesize ATP. ATP synthase is regulated both thermodynamically and post-translationally, with proposed phosphorylation sites on multiple subunits. In this study we focused on two N-terminal serines on the catalytic subunit β in tobacco (Nicotiana tabacum), previously proposed to be important for dark inactivation of the complex to avoid ATP hydrolysis at night. Here we show that there is no clear role for phosphorylation in the dark inactivation of ATP synthase. Instead, mutation of one of the two phosphorylated serine residues to aspartate to mimic constitutive phosphorylation strongly decreased ATP synthase abundance. We propose that the loss of N-terminal phosphorylation of ATPβ may be involved in proper ATP synthase accumulation during complex assembly.

Precision timing of millisecond pulsars in binary systems enables observers to detect the relativistic Shapiro delay induced by space time curvature. When favourably aligned, this enables constraints to be placed on the component masses and system orientation. Here we present the results of timing campaigns on seven binary millisecond pulsars observed with the 64-antenna MeerKAT radio telescope that show evidence of Shapiro delay: PSRs~J0101$-$6422, J1101$-$6424, J1125$-$6014, J1514$-$4946, J1614$-$2230, J1732$-$5049, and J1909$-$3744. Evidence for Shapiro delay was found in all of the systems, and for three the orientations and data quality enabled strong constraints on their orbital inclinations and component masses. For PSRs~J1125$-$6014, J1614$-$2230 and J1909$-$3744, we determined pulsar masses to be $M_{\rm p} = 1.68\pm 0.17 \, {\rm M_{\odot}} $, $1.94\pm 0.03 \, {\rm M_{\odot}} $ and $1.45 \pm 0.03 \, {\rm M_{\odot}}$, and companion masses to be $M_{\rm c} = 0.33\pm 0.02 \, {\rm M_{\odot}} $, $0.495\pm 0.005 \, {\rm M_{\odot}} $ and $0.205 \pm 0.003 \, {\rm M_{\odot}}$, respectively. This provides the first independent confirmation of PSR~J1614$-$2230's mass, one of the highest known. The Shapiro delays measured for PSRs~J0101$-$6422, J1101$-$6424, J1514$-$4946, and J1732$-$5049 were only weak, and could not provide interesting component mass limits. Despite a large number of millisecond pulsars being routinely timed, relatively few have accurate masses via Shapiro delays. We use simulations to show that this is expected, and provide a formula for observers to assess how accurately a pulsar mass can be determined. Multi-element interferometers such as MeerKAT, which observe with high time resolution and have a wide field of view, provide an ideal opportunity to perform real-time, untargeted transient and pulsar searches. However, because of data storage limitations, it is not always feasible to store the baseband data required to image the field of a discovered transient or pulsar. This limits the ability of surveys to effectively localize their discoveries and may restrict opportunities for follow-up science, especially of one-off events like some fast radio bursts. Here, we present a novel maximum-likelihood estimation approach to localizing transients and pulsars detected in multiple MeerKAT tied-array beams at once, which we call tied-array beam localization, as well as a Python implementation of the method named SeeKAT. We provide real-world examples of SeeKAT’s use as well as a Monte Carlo analysis to show that it is capable of localizing single pulses detected in beamformed MeerKAT data to (sub)arcsec precision.

Background: Peripheral artery disease (PAD) results in exercise-induced ischemia in leg muscles. 31 Phosphorus (P) magnetic resonance spectroscopy demonstrates prolonged phosphocreatine recovery time constant after exercise in PAD but has low signal to noise, low spatial resolution, and requires multinuclear hardware. Chemical exchange saturation transfer (CEST) is a quantitative magnetic resonance imaging method for imaging substrate (CEST asymmetry [CEST asym ]) concentration by muscle group. We hypothesized that kinetics measured by CEST could distinguish between patients with PAD and controls. Methods: Patients with PAD and age-matched normal subjects were imaged at 3T with a transmit-receive coil around the calf. Four CEST mages were acquired over 24-second intervals. The subjects then performed plantar flexion exercise on a magnetic resonance imaging-compatible ergometer until calf exhaustion. Twenty-five CEST images were obtained at end exercise. Regions of interest were drawn around individual muscle groups, and (CEST asym ) decay times were fitted by exponential curve to CEST values. In 10 patients and 11 controls, 31 P spectra were obtained 20 minutes later after repeat exercise. Five patients and 5 controls returned at a mean of 1±1 days later for repeat CEST studies. Results: Thirty-five patients with PAD (31 male, age 66±8 years) and 29 controls (11 male, age 63±8 years) were imaged with CEST. The CEST asym decay times for the whole calf (341±332 versus 153±72 seconds; P asym decay and phosphocreatine recovery time constant was good. Conclusions: CEST is a magnetic resonance imaging method that can distinguish energetics in patients with PAD from age-matched normal subjects on a per muscle group basis. CEST agrees reasonably well with the gold standard 31 P magnetic resonance spectroscopy. Moreover, CEST has higher spatial resolution, creates an image, and does not require multinuclear hardware and thus may be more suitable for clinical studies in PAD.

The purpose was to explore interrelations between factors related to engagement in physical activity in inactive adults with knee pain.Inactive adults with knee pain (In this cohort (age = 60.9 ± 8.6 years; 22 females), self-reported physical activity was 26.3 ± 46.8 min/week. Factors related to physical activity were grouped into domains of physical status, psychological status, environment, knowledge, and resources. It was seen that the interrelations between a person and their environment, as well as, between impairments and everyday responsibilities influenced engagement in physical activity. Females were more likely to identify physical and psychological status, social expectations, and lack of knowledge as barriers. Males indicated a preference for using mobile technologies to overcome barriers.Interplay of various barriers and facilitators is related to engagement in physical activity in inactive older adults with knee pain. Interventions to promote physical activity should address these interrelations and sex differences.Implications for rehabilitationInterrelations between individual factors related to engagement in physical activity and sex differences in these factors are present in inactive adults with knee pain.Interventions to improve physical activity should be implemented by addressing factors and interrelations between factors related to physical activity in inactive adults with knee pain.Interventions to address low levels of physical activity in adults with knee pain should take into account sex differences.

We present the first 2.5 years of data from the MeerKAT Pulsar Timing Array (MPTA), part of MeerTime, a MeerKAT Large Survey Project. The MPTA aims to precisely measure pulse arrival times from an ensemble of 88 pulsars visible from the Southern Hemisphere, with the goal of contributing to the search, detection and study of nanohertz-frequency gravitational waves as part of the International Pulsar Timing Array. This project makes use of the MeerKAT telescope, and operates with a typical observing cadence of two weeks using the L-band receiver that records data from 856-1712 MHz. We provide a comprehensive description of the observing system, software, and pipelines used and developed for the MeerTime project. The data products made available as part of this data release are from the 78 pulsars that had at least $30$ observations between the start of the MeerTime programme in February 2019 and October 2021. These include both sub-banded and band-averaged arrival times, as well as the initial timing ephemerides, noise models, and the frequency-dependent standard templates (portraits) used to derive pulse arrival times. After accounting for detected noise processes in the data, the frequency-averaged residuals of $67$ of the pulsars achieved a root-mean-square residual precision of $< 1 \mu \rm{s}$. We also present a novel recovery of the clock correction waveform solely from pulsar timing residuals, and an exploration into preliminary findings of interest to the international pulsar timing community. We present the polarization profiles of 22 pulsars in the globular cluster 47 Tucanae using observations from the MeerKAT radio telescope at UHF-band (544-1088 MHz) and report precise values of dispersion measure (DM) and rotation measure (RM). We use these measurements to investigate the presence of turbulence in electron density and magnetic fields. The structure function of DM shows a break at $\sim 30$ arcsec ($\sim 0.6$ pc at the distance of 47 Tucanae) that suggests the presence of turbulence in the gas in the cluster driven by the motion of wind-shedding stars. On the other hand, the structure function of RM does not show evidence of a break. This non-detection could be explained either by the limited number of pulsars or by the effects of the intervening gas in the Galaxy along the line of sight. Future pulsar discoveries in the cluster could help confirm the presence and localise the turbulence., Accepted for publication in MNRAS, 14 pages, 6 figures

Triose-phosphate utilization (TPU) limits the maximum rate at which plants can photosynthesize. However, TPU is almost never found to be limiting photosynthesis under ambient conditions for plants. This, along with previous results showing adaptability of TPU at low temperature, suggest that TPU capacity is regulated to be just above the photosynthetic rate achievable under the prevailing conditions. A set of experiments were performed to study the adaptability of TPU capacity when plants are acclimated to elevated CO concentrations. Plants held at 1500 ppm CO were initially TPU limited. After 30 hours they no longer exhibited TPU limitations but they did not elevate their TPU capacity. Instead, the maximum rates of carboxylation and electron transport declined. A timecourse of regulatory responses was established. A step increase of CO first caused PSI to be oxidized but after 40 s both PSI and PSII had excess electrons as a result of acceptor-side limitations. Electron flow to PSI slowed and the proton motive force increased. Eventually, non-photochemical quenching reduced electron flow sufficiently to balance the TPU limitation. Over several minutes rubisco deactivated contributing to regulation of metabolism to overcome the TPU limitation.

Serious neurological illnesses are associated with significant palliative care needs, including symptom management, complex decision-making, support for caregivers, and end-of-life care. While all neurologists are responsible for the provision of primary palliative care, there is an increasing need for trained neurologists with expertise in palliative medicine to manage refractory symptoms, mitigate conflict around goals of care, and provide specialized end-of-life care. This has led to the emergence of neuropalliative care as a subspecialty. There are different ways to acquire palliative care skills, incorporate them into one's neurology practice, and develop a neuropalliative career. We interviewed three leaders in the field of neuropalliative care, Dr. Robert Holloway, Dr. Jessica McFarlin, and Dr. Janis Miyasaki, who are all neurologists with different subspecialties and training pathways working in academic centers. They share their career paths, their advice for neurology trainees interested in pursuing a career in neuropalliative care, and their thoughts on the future of the field.

Hidradenitis suppurativa is a chronic inflammatory condition that presents a challenging reconstructive problem for plastic surgeons.The authors performed a retrospective chart review of hidradenitis suppurativa patients managed with surgical excision between 2005 and 2020 at Brigham and Women's Hospital and Tulane University Medical Center. Operative cases associated with the same hospitalization were organized into treatment episodes and assessed for patient demographics, operative techniques, and outcomes.A total of 181 patients, 435 cases and 316 treatment episodes (Brigham and Women's Hospital, n = 269; Tulane University Medical Center, n = 47), were identified across two diverse institutions. Their respective series showed comparable patient demographics, and 94 percent of the combined episodes achieved wound closure and healing during the study period. Several techniques of closure were identified, including immediate closure and site-specific methods, such as an expedited staged closure using internal negative-pressure wound therapy as a temporary bridge, "recycled" skin grafting, and repurposing iodoform wicks as an adjunct wound healing therapy to immediate closure.This large multi-institutional retrospective chart review on the plastic surgical management of hidradenitis suppurativa demonstrates that surgery is an effective therapy for hidradenitis suppurativa and captures a diversity of site-specific techniques that may serve as a foundation for future prospective studies and evidence-based guidelines for the use of various techniques to optimize patients' surgical outcomes.Therapeutic, IV.

NUT carcinoma (NC) is a rare subtype of squamous cell carcinoma defined by NUTM1 rearrangements encoding NUT fusion oncoproteins (the most frequent fusion partner being BRD4 ) that carries a very poor prognosis, with most patients dying in under 1 year. Only rare primary thyroid NCs have been reported. Here, we evaluated a series of 14 cases. The median patient age at diagnosis was 38 years (range: 17 to 72 y). Eight of 13 cases with slides available for review (62%) showed a morphology typical of NC, whereas 5 (38%) had a non-NC-like morphology, some of which had areas of cribriform or fused follicular architecture resembling a follicular cell-derived thyroid carcinoma. For cases with immunohistochemistry results, 85% (11/13) were positive for NUT on biopsy or resection, though staining was significantly decreased on resection specimens due to fixation; 55% (6/11) were positive for PAX8, and 54% (7/13) for TTF-1. Tumors with a non-NC-like morphology were all positive for PAX8 and TTF-1. The fusion partner was known in 12 cases: 9 (75%) cases had a NSD3-NUTM1 fusion, and 3 (25%) had a BRD4-NUTM1 fusion. For our cohort, the 2-year overall survival (OS) was 69%, and the 5-year OS was 58%. Patients with NC-like tumors had a significantly worse OS compared with that of patients with tumors with a non-NC-like morphology ( P =0.0462). Our study shows that NC of the thyroid can mimic other thyroid primaries, has a high rate of NSD3 - NUTM1 fusions, and an overall more protracted clinical course compared with nonthyroid primary NC.

The discovery of pulsars opened a new research field that allows studying a wide range of physics under extreme conditions. More than 3,000 pulsars are currently known, including especially more than 200 of them studied at gamma-ray frequencies. By putting recent insights into the pulsar magnetosphere in a historical context and by comparing them to key observational features at radio and high-energy frequencies, we show the following: ▪ Magnetospheric structure of young energetic pulsars is now understood. Limitations still exist for old nonrecycled and millisecond pulsars. ▪ The observed high-energy radiation is likely produced in the magnetospheric current sheet beyond the light cylinder. ▪ There are at least two different radio emission mechanisms. One operates in the inner magnetosphere, whereas the other one works near the light cylinder and is specific to pulsars with the high magnetic field strength in that region. ▪ Radio emission from the inner magnetosphere is intrinsically connected to the process of pair production, and its observed properties contain the imprint of both the geometry and propagation effects through the magnetospheric plasma. We discuss the limitations of our understanding and identify a range of observed phenomena and physical processes that still await explanation in thefuture. This includes connecting the magnetospheric processes to spin-down properties to explain braking and possible evolution of spin orientation, building a first-principles model of radio emission and quantitative connections with observations.
