Antarctica's Lambert Glacier drains about one-sixth of the ice from the East Antarctic Ice Sheet and is considered stable due to the strong buttressing provided by the Amery Ice Shelf. While previous projections of the sea-level contribution from this sector of the ice sheet have predicted significant mass loss only with near-complete removal of the ice shelf, the ocean warming necessary for this was deemed unlikely. Recent climate projections through 2300 indicate that sufficient ocean warming is a distinct possibility after 2100. This work explores the impact of parametric uncertainty on projections of the response of the Lambert–Amery system (hereafter “the Amery sector”) to abrupt ocean warming through Bayesian calibration of a perturbed-parameter ice-sheet model ensemble. We address the computational cost of uncertainty quantification for ice-sheet model projections via statistical emulation, which employs surrogate models for fast and inexpensive parameter space exploration while retaining critical features of the high-fidelity simulations. To this end, we build Gaussian process (GP) emulators from simulations of the Amery sector at a medium resolution (4–20 km mesh) using the Model for Prediction Across Scales (MPAS)-Albany Land Ice (MALI) model. We consider six input parameters that control basal friction, ice stiffness, calving, and ice-shelf basal melting. From these, we generate 200 perturbed input parameter initializations using space filling Sobol sampling. For our end-to-end probabilistic modeling workflow, we first train emulators on the simulation ensemble and then calibrate the input parameters using observations of the mass balance, grounding line movement, and calving front movement with priors assigned via expert knowledge. Next, we use MALI to project a subset of simulations to 2300 using ocean and atmosphere forcings from a climate model for both low- and high-greenhouse-gas-emission scenarios. From these simulation outputs, we build multivariate emulators by combining GP regression with principal component dimension reduction to emulate multivariate sea-level contribution time series data from the MALI simulations. We then use these emulators to propagate uncertainty from model input parameters to predictions of glacier mass loss through 2300, demonstrating that the calibrated posterior distributions have both greater mass loss and reduced variance compared to the uncalibrated prior distributions. Parametric uncertainty is large enough through about 2130 that the two projections under different emission scenarios are indistinguishable from one another. However, after rapid ocean warming in the first half of the 22nd century, the projections become statistically distinct within decades. Overall, this study demonstrates an efficient Bayesian calibration and uncertainty propagation workflow for ice-sheet model projections and identifies the potential for large sea-level rise contributions from the Amery sector of the Antarctic Ice Sheet after 2100 under high-greenhouse-gas-emission scenarios.

Some ocean modeling studies have identified a potential tipping point from a low to a high basal melt regime beneath the Filchner–Ronne Ice Shelf (FRIS), Antarctica, with significant implications for subsequent Antarctic ice sheet mass loss. To date, investigation of the climate drivers and impacts of this possible event have been limited because ice-shelf cavities and ice-shelf melting are only now starting to be included in global climate models. Using a global ocean–sea-ice configuration of the Energy Exascale Earth System Model (E3SM) that represents both ocean circulations and melting within ice-shelf cavities, we explore freshwater triggers (iceberg melt and ice-shelf basal melt) of a transition to a high-melt regime at FRIS in a low-resolution (30 km in the Southern Ocean) global ocean–sea-ice model. We find that a realistic spatial distribution of iceberg melt fluxes is necessary to prevent the FRIS melt regime change from unrealistically occurring under historical-reanalysis-based atmospheric forcing. Further, improvement of the default parameterization for mesoscale eddy mixing significantly reduces a large regional fresh bias and weak Antarctic Slope Front structure, both of which precondition the model to melt regime change. Using two different stable model configurations, we explore the sensitivity of FRIS melt regime change to regional ice-sheet freshwater fluxes. Through a series of sensitivity experiments prescribing incrementally increasing melt rates from the smaller, neighboring ice shelves in the eastern Weddell Sea, we demonstrate the potential for an ice-shelf melt “domino effect” should the upstream ice shelves experience increased melt rates. The experiments also reveal that modest ice-shelf melt biases in a model, especially at coarse ocean resolution where narrow continental shelf dynamics are not well resolved, can lead to an unrealistic melt regime change at downstream ice shelves. Thus, we find that remote connections between melt fluxes at different ice shelves are sensitive to baseline model conditions. Our results highlight both the potential and the peril of simulating prognostic Antarctic ice-shelf melt rates in a low-resolution global model.

The Antarctic Ice Sheet represents the largest source of uncertainty in future sea level rise projections, with a contribution to sea level by 2100 ranging from −5 to 43 cm of sea level equivalent under high carbon emission scenarios estimated by the recent Ice Sheet Model Intercomparison for CMIP6 (ISMIP6). ISMIP6 highlighted the different behaviors of the East and West Antarctic ice sheets, as well as the possible role of increased surface mass balance in offsetting the dynamic ice loss in response to changing oceanic conditions in ice shelf cavities. However, the detailed contribution of individual glaciers, as well as the partitioning of uncertainty associated with this ensemble, have not yet been investigated. Here, we analyze the ISMIP6 results for high carbon emission scenarios, focusing on key glaciers around the Antarctic Ice Sheet, and we quantify their projected dynamic mass loss, defined here as mass loss through increased ice discharge into the ocean in response to changing oceanic conditions. We highlight glaciers contributing the most to sea level rise, as well as their vulnerability to changes in oceanic conditions. We then investigate the different sources of uncertainty and their relative role in projections, for the entire continent and for key individual glaciers. We show that, in addition to Thwaites and Pine Island glaciers in West Antarctica, Totten and Moscow University glaciers in East Antarctica present comparable future dynamic mass loss and high sensitivity to ice shelf basal melt. The overall uncertainty in additional dynamic mass loss in response to changing oceanic conditions, compared to a scenario with constant oceanic conditions, is dominated by the choice of ice sheet model, accounting for 52 % of the total uncertainty of the Antarctic dynamic mass loss in 2100. Its relative role for the most dynamic glaciers varies between 14 % for MacAyeal and Whillans ice streams and 56 % for Pine Island Glacier at the end of the century. The uncertainty associated with the choice of climate model increases over time and reaches 13 % of the uncertainty by 2100 for the Antarctic Ice Sheet but varies between 4 % for Thwaites Glacier and 53 % for Whillans Ice Stream. The uncertainty associated with the ice–climate interaction, which captures different treatments of oceanic forcings such as the choice of melt parameterization, its calibration, and simulated ice shelf geometries, accounts for 22 % of the uncertainty at the ice sheet scale but reaches 36 % and 39 % for Institute Ice Stream and Thwaites Glacier, respectively, by 2100. Overall, this study helps inform future research by highlighting the sectors of the ice sheet most vulnerable to oceanic warming over the 21st century and by quantifying the main sources of uncertainty.

Multi-meter sea level rise (SLR) is thought to be possible within the next few centuries, with most of the uncertainty originating from the Antarctic land ice contribution. One source of uncertainty relates to the ice sheet model initialization. Since ice sheets have a long response time (compared to other Earth system components such as the atmosphere), ice sheet model initialization methods can have significant impacts on how the ice sheet responds to future forcings. To assess this, we generated 25 different ice sheet spin-ups, using the Community Ice Sheet Model (CISM) at a 4 km resolution. During each spin-up, we varied two key parameters known to impact the sensitivity of the ice sheet to future forcing: one related to the sensitivity of the ice shelf melt rate to ocean thermal forcing (TF) and the other related to the basal friction. The spin-ups all nudge toward observed thickness and enforce a no-advance calving criterion, such that all final spin-up states resemble observations but differ in their melt and friction parameter settings. Each spin-up was then forced with future ocean thermal forcings from 13 different CMIP6 models under the Shared Socioeconomic Pathway (SSP)5-8.5 emissions scenario and modern climatological surface mass balance data. Our results show that the effects of the ice sheet and ocean parameter settings used during the spin-up are capable of impacting multi-century future SLR predictions by as much as 2 m. By the end of this century, the effects of these choices are more modest, but still significant, with differences of up to 0.2 m of SLR. We have identified a combined ocean and ice parameter space that leads to widespread mass loss within 500 years (low friction and high melt rate sensitivity). To explore temperature thresholds, we also ran a synthetically forced CISM ensemble that is focused on the Amundsen region only. Given certain ocean and ice parameter choices, Amundsen mass loss can be triggered with thermal forcing anomalies between 1.5 and 2 ∘C relative to the spin-up. Our results emphasize the critical importance of considering ice sheet and ocean parameter choices during spin-up for SLR predictions and suggest the importance of including glacial isostatic adjustment in ice sheet simulations.

Humboldt Glacier, northern Greenland, has retreated and accelerated through the 21st century, raising concerns that it could be a significant contributor to future sea-level rise. We use a data-constrained ensemble of three-dimensional higher-order ice sheet model simulations to estimate the likely range of sea-level rise from the continued retreat of Humboldt Glacier. We first solve for basal traction using observed ice thickness, bed topography, and ice surface velocity from the year 2007 in a PDE-constrained (partial differential equation) optimization. Next, we impose calving rates to match mean observed retreat rates from winter 2007–2008 to winter 2017–2018 in a transient calibration of the exponent in the power-law basal friction relationship. We find that power-law exponents in the range of 1/7–1/5 – rather than the commonly used 1/3–1 – are necessary to reproduce the observed speedup over this period. We then tune an iceberg calving parameterization based on the von Mises stress yield criterion in another transient-calibration step to approximate both observed ice velocities and terminus position in 2017–2018. Finally, we use the range of basal friction relationship exponents and calving parameter values to generate the ensemble of model simulations from 2007–2100 under three climate forcing scenarios from CMIP5 (two RCP8.5 forcings, Representative Concentration Pathway) and CMIP6 (one SSP5-8.5 forcing, Shared Socioeconomic Pathway). Our simulations predict 5.2–8.7 mm of sea-level rise from Humboldt Glacier, significantly higher than a previous estimate (∼ 3.5 mm) and equivalent to a substantial fraction of the 40–140 mm predicted by ISMIP6 from the whole Greenland Ice Sheet. Our larger future sea-level rise prediction results from the transient calibration of our basal friction law to match the observed speedup, which requires a semi-plastic bed rheology. In many simulations, our model predicts the growth of a sizable ice shelf in the middle of the 21st century. Thus, atmospheric warming could lead to more retreat than predicted here if increased surface melt promotes hydrofracture of the ice shelf. Our data-constrained simulations of Humboldt Glacier underscore the sensitivity of model predictions of Greenland outlet glacier response to warming to choices of basal shear stress and iceberg calving parameterizations. Further, transient calibration of these parameterizations, which has not typically been performed, is necessary to reproduce observed behavior. Current estimates of future sea-level rise from the Greenland Ice Sheet could, therefore, contain significant biases.

Subglacial hydrology is a leading control on basal friction and the dynamics of glaciers and ice sheets. At low discharge, subglacial water flows through high-pressure, sheet-like systems that lead to low effective pressures. However, at high discharge, subglacial water melts the overlying ice into localized channels that efficiently remove water from the bed, thereby increasing effective pressure and basal friction. Recent observations suggest channelized subglacial flow exists beneath Thwaites Glacier, yet it remains unclear if stable channelization is feasible in West Antarctica, where surface melting is nonexistent and water at the bed is limited. Here, we use the MPAS-Albany Land Ice model to run a suite of over 130 subglacial hydrology simulations of Thwaites Glacier across a wide range of physical parameter choices to assess the likelihood of channelization. We then narrow our range of viable simulations by comparing modeled water thicknesses to previously observed radar specularity content, which indicates flat, spatially extensive water bodies at the bed. In all of our data-compatible simulations, stable channels reliably form within 100–200 km of the grounding line and reach individual discharge rates of 35–110 m3 s−1 at the ice–ocean boundary. While only one to two channels typically form across the 200 km width of the glacier in our simulations, their high efficiency drains water across the entire lateral extent of the glacier. We posit the large catchment size of Thwaites Glacier, its funnel-like geometry, and high basal melt rates together accumulate enough water to form stable channels. No simulations resembled observed specularity content when channelization was disabled. Our results suggest channelized subglacial hydrology has two consequences for Thwaites Glacier dynamics: (i) amplifying submarine melting of the terminus and ice shelf while (ii) simultaneously raising effective pressure within 100 km of the grounding line and increasing basal friction. The distribution of effective pressure implied from our modeling differs from parameterizations typically used in large-scale ice sheet models, suggesting the development of more process-based parameterizations may be necessary.

Using a numerical ice flow model, we study changes in ice shelf buttressing and grounding-line flux due to localized ice thickness perturbations, a proxy for localized changes in sub-ice-shelf melting. From our experiments, applied to idealized (MISMIP+) and realistic (Larsen C) ice shelf domains, we identify a correlation between a locally derived buttressing number on the ice shelf, based on the first principal stress, and changes in the integrated grounding-line flux. The origin of this correlation, however, remains elusive from the perspective of a theoretical or physically based understanding. This and the fact that the correlation is generally much poorer when applied to realistic ice shelf domains motivate us to seek an alternative approach for predicting changes in grounding-line flux. We therefore propose an adjoint-based method for calculating the sensitivity of the integrated grounding-line flux to local changes in ice shelf geometry. We show that the adjoint-based sensitivity is identical to that deduced from pointwise, diagnostic model perturbation experiments. Based on its much wider applicability and the significant computational savings, we propose that the adjoint-based method is ideally suited for assessing grounding-line flux sensitivity to changes in sub-ice-shelf melting.

Ice flow models of the Antarctic ice sheet are commonly used to simulate its future evolution in response to different climate scenarios and assess the mass loss that would contribute to future sea level rise. However, there is currently no consensus on estimates of the future mass balance of the ice sheet, primarily because of differences in the representation of physical processes, forcings employed and initial states of ice sheet models. This study presents results from ice flow model simulations from 13 international groups focusing on the evolution of the Antarctic ice sheet during the period 2015–2100 as part of the Ice Sheet Model Intercomparison for CMIP6 (ISMIP6). They are forced with outputs from a subset of models from the Coupled Model Intercomparison Project Phase 5 (CMIP5), representative of the spread in climate model results. Simulations of the Antarctic ice sheet contribution to sea level rise in response to increased warming during this period varies between −7.8 and 30.0 cm of sea level equivalent (SLE) under Representative Concentration Pathway (RCP) 8.5 scenario forcing. These numbers are relative to a control experiment with constant climate conditions and should therefore be added to the mass loss contribution under climate conditions similar to present-day conditions over the same period. The simulated evolution of the West Antarctic ice sheet varies widely among models, with an overall mass loss, up to 18.0 cm SLE, in response to changes in oceanic conditions. East Antarctica mass change varies between −6.1 and 8.3 cm SLE in the simulations, with a significant increase in surface mass balance outweighing the increased ice discharge under most RCP 8.5 scenario forcings. The inclusion of ice shelf collapse, here assumed to be caused by large amounts of liquid water ponding at the surface of ice shelves, yields an additional simulated mass loss of 28 mm compared to simulations without ice shelf collapse. The largest sources of uncertainty come from the climate forcing, the ocean-induced melt rates, the calibration of these melt rates based on oceanic conditions taken outside of ice shelf cavities and the ice sheet dynamic response to these oceanic changes. Results under RCP 2.6 scenario based on two CMIP5 climate models show an additional mass loss of 0 and 3 cm of SLE on average compared to simulations done under present-day conditions for the two CMIP5 forcings used and display limited mass gain in East Antarctica.

We present the result of the third Marine Ice Sheet Model Intercomparison Project, MISMIP+. MISMIP+ is intended to be a benchmark for ice-flow models which include fast sliding marine ice streams and floating ice shelves and in particular a treatment of viscous stress that is sufficient to model buttressing, where upstream ice flow is restrained by a downstream ice shelf. A set of idealized experiments first tests that models are able to maintain a steady state with the grounding line located on a retrograde slope due to buttressing and then explore scenarios where a reduction in that buttressing causes ice stream acceleration, thinning, and grounding line retreat. The majority of participating models passed the first test and then produced similar responses to the loss of buttressing. We find that the most important distinction between models in this particular type of simulation is in the treatment of sliding at the bed, with other distinctions – notably the difference between the simpler and more complete treatments of englacial stress but also the differences between numerical methods – taking a secondary role.

The sea level contribution of the Antarctic ice sheet constitutes a large uncertainty in future sea level projections. Here we apply a linear response theory approach to 16 state-of-the-art ice sheet models to estimate the Antarctic ice sheet contribution from basal ice shelf melting within the 21st century. The purpose of this computation is to estimate the uncertainty of Antarctica's future contribution to global sea level rise that arises from large uncertainty in the oceanic forcing and the associated ice shelf melting. Ice shelf melting is considered to be a major if not the largest perturbation of the ice sheet's flow into the ocean. However, by computing only the sea level contribution in response to ice shelf melting, our study is neglecting a number of processes such as surface-mass-balance-related contributions. In assuming linear response theory, we are able to capture complex temporal responses of the ice sheets, but we neglect any self-dampening or self-amplifying processes. This is particularly relevant in situations in which an instability is dominating the ice loss. The results obtained here are thus relevant, in particular wherever the ice loss is dominated by the forcing as opposed to an internal instability, for example in strong ocean warming scenarios. In order to allow for comparison the methodology was chosen to be exactly the same as in an earlier study (Levermann et al., 2014) but with 16 instead of 5 ice sheet models. We include uncertainty in the atmospheric warming response to carbon emissions (full range of CMIP5 climate model sensitivities), uncertainty in the oceanic transport to the Southern Ocean (obtained from the time-delayed and scaled oceanic subsurface warming in CMIP5 models in relation to the global mean surface warming), and the observed range of responses of basal ice shelf melting to oceanic warming outside the ice shelf cavity. This uncertainty in basal ice shelf melting is then convoluted with the linear response functions of each of the 16 ice sheet models to obtain the ice flow response to the individual global warming path. The model median for the observational period from 1992 to 2017 of the ice loss due to basal ice shelf melting is 10.2 mm, with a likely range between 5.2 and 21.3 mm. For the same period the Antarctic ice sheet lost mass equivalent to 7.4 mm of global sea level rise, with a standard deviation of 3.7 mm (Shepherd et al., 2018) including all processes, especially surface-mass-balance changes. For the unabated warming path, Representative Concentration Pathway 8.5 (RCP8.5), we obtain a median contribution of the Antarctic ice sheet to global mean sea level rise from basal ice shelf melting within the 21st century of 17 cm, with a likely range (66th percentile around the mean) between 9 and 36 cm and a very likely range (90th percentile around the mean) between 6 and 58 cm. For the RCP2.6 warming path, which will keep the global mean temperature below 2 ∘C of global warming and is thus consistent with the Paris Climate Agreement, the procedure yields a median of 13 cm of global mean sea level contribution. The likely range for the RCP2.6 scenario is between 7 and 24 cm, and the very likely range is between 4 and 37 cm. The structural uncertainties in the method do not allow for an interpretation of any higher uncertainty percentiles. We provide projections for the five Antarctic regions and for each model and each scenario separately. The rate of sea level contribution is highest under the RCP8.5 scenario. The maximum within the 21st century of the median value is 4 cm per decade, with a likely range between 2 and 9 cm per decade and a very likely range between 1 and 14 cm per decade.

Ice sheet numerical modeling is an important tool to estimate the dynamic contribution of the Antarctic ice sheet to sea level rise over the coming centuries. The influence of initial conditions on ice sheet model simulations, however, is still unclear. To better understand this influence, an initial state intercomparison exercise (initMIP) has been developed to compare, evaluate, and improve initialization procedures and estimate their impact on century-scale simulations. initMIP is the first set of experiments of the Ice Sheet Model Intercomparison Project for CMIP6 (ISMIP6), which is the primary Coupled Model Intercomparison Project Phase 6 (CMIP6) activity focusing on the Greenland and Antarctic ice sheets. Following initMIP-Greenland, initMIP-Antarctica has been designed to explore uncertainties associated with model initialization and spin-up and to evaluate the impact of changes in external forcings. Starting from the state of the Antarctic ice sheet at the end of the initialization procedure, three forward experiments are each run for 100 years: a control run, a run with a surface mass balance anomaly, and a run with a basal melting anomaly beneath floating ice. This study presents the results of initMIP-Antarctica from 25 simulations performed by 16 international modeling groups. The submitted results use different initial conditions and initialization methods, as well as ice flow model parameters and reference external forcings. We find a good agreement among model responses to the surface mass balance anomaly but large variations in responses to the basal melting anomaly. These variations can be attributed to differences in the extent of ice shelves and their upstream tributaries, the numerical treatment of grounding line, and the initial ocean conditions applied, suggesting that ongoing efforts to better represent ice shelves in continental-scale models should continue.

A collection of scientific analyses, metrics, and visualizations for robust validation of ice sheet models is presented using the Land Ice Verification and Validation toolkit (LIVVkit), version 2.1, and the LIVVkit Extensions repository (LEX), version 0.1. This software collection targets stand-alone ice sheet or coupled Earth system models, and handles datasets and analyses that require high-performance computing and storage. LIVVkit aims to enable efficient and fully reproducible workflows for postprocessing, analysis, and visualization of observational and model-derived datasets in a shareable format, whereby all data, methodologies, and output are distributed to users for evaluation. Extending from the initial LIVVkit software framework, we demonstrate Greenland ice sheet simulation validation metrics using the coupled Community Earth System Model (CESM) as well as an idealized stand-alone high-resolution Community Ice Sheet Model, version 2 (CISM2), coupled to the Albany/FELIX velocity solver (CISM-Albany or CISM-A). As one example of the capability, LIVVkit analyzes the degree to which models capture the surface mass balance (SMB) and identifies potential sources of bias, using recently available in situ and remotely sensed data as comparison. Related fields within atmosphere and land surface models, e.g., surface temperature, radiation, and cloud cover, are also diagnosed. Applied to the CESM1.0, LIVVkit identifies a positive SMB bias that is focused largely around Greenland's southwest region that is due to insufficient ablation.

We describe and evaluate version 2.1 of the Community Ice Sheet Model (CISM). CISM is a parallel, 3-D thermomechanical model, written mainly in Fortran, that solves equations for the momentum balance and the thickness and temperature evolution of ice sheets. CISM's velocity solver incorporates a hierarchy of Stokes flow approximations, including shallow-shelf, depth-integrated higher order, and 3-D higher order. CISM also includes a suite of test cases, links to third-party solver libraries, and parameterizations of physical processes such as basal sliding, iceberg calving, and sub-ice-shelf melting. The model has been verified for standard test problems, including the Ice Sheet Model Intercomparison Project for Higher-Order Models (ISMIP-HOM) experiments, and has participated in the initMIP-Greenland initialization experiment. In multimillennial simulations with modern climate forcing on a 4 km grid, CISM reaches a steady state that is broadly consistent with observed flow patterns of the Greenland ice sheet. CISM has been integrated into version 2.0 of the Community Earth System Model, where it is being used for Greenland simulations under past, present, and future climates. The code is open-source with extensive documentation and remains under active development.

We introduce MPAS-Albany Land Ice (MALI) v6.0, a new variable-resolution land ice model that uses unstructured Voronoi grids on a plane or sphere. MALI is built using the Model for Prediction Across Scales (MPAS) framework for developing variable-resolution Earth system model components and the Albany multi-physics code base for the solution of coupled systems of partial differential equations, which itself makes use of Trilinos solver libraries. MALI includes a three-dimensional first-order momentum balance solver (Blatter–Pattyn) by linking to the Albany-LI ice sheet velocity solver and an explicit shallow ice velocity solver. The evolution of ice geometry and tracers is handled through an explicit first-order horizontal advection scheme with vertical remapping. The evolution of ice temperature is treated using operator splitting of vertical diffusion and horizontal advection and can be configured to use either a temperature or enthalpy formulation. MALI includes a mass-conserving subglacial hydrology model that supports distributed and/or channelized drainage and can optionally be coupled to ice dynamics. Options for calving include eigencalving, which assumes that the calving rate is proportional to extensional strain rates. MALI is evaluated against commonly used exact solutions and community benchmark experiments and shows the expected accuracy. Results for the MISMIP3d benchmark experiments with MALI's Blatter–Pattyn solver fall between published results from Stokes and L1L2 models as expected. We use the model to simulate a semi-realistic Antarctic ice sheet problem following the initMIP protocol and using 2 km resolution in marine ice sheet regions. MALI is the glacier component of the Energy Exascale Earth System Model (E3SM) version 1, and we describe current and planned coupling to other E3SM components.

We propose a new ice sheet model validation framework – the Cryospheric Model Comparison Tool (CmCt) – that takes advantage of ice sheet altimetry and gravimetry observations collected over the past several decades and is applied here to modeling of the Greenland ice sheet. We use realistic simulations performed with the Community Ice Sheet Model (CISM) along with two idealized, non-dynamic models to demonstrate the framework and its use. Dynamic simulations with CISM are forced from 1991 to 2013, using combinations of reanalysis-based surface mass balance and observations of outlet glacier flux change. We propose and demonstrate qualitative and quantitative metrics for use in evaluating the different model simulations against the observations. We find that the altimetry observations used here are largely ambiguous in terms of their ability to distinguish one simulation from another. Based on basin-scale and whole-ice-sheet-scale metrics, we find that simulations using both idealized conceptual models and dynamic, numerical models provide an equally reasonable representation of the ice sheet surface (mean elevation differences of

Ice temperature profiles from the Greenland Ice Sheet contain information on the deformation history, past climates and recent warming. We present full-depth temperature profiles from two drill sites on a flow line passing through Swiss Camp, West Greenland. Numerical modeling reveals that ice temperatures are considerably higher than would be expected from heat diffusion and dissipation alone. The possible causes for this extra heat are evaluated using a Lagrangian heat flow model. The model results reveal that the observations can be explained with a combination of different processes: enhanced dissipation (strain heating) in ice-age ice, temperate paleo-firn, and cryo-hydrologic warming in deep crevasses.

The rapid drainage of supraglacial lakes introduces large pulses of meltwater to the subglacial environment and creates moulins, surface-to-bed conduits for future melt. Introduction of water to the subglacial system has been shown to affect ice flow, and modeling suggests that variability in water supply and delivery to the subsurface play an important role in the development of the subglacial hydrologic system and its ability to enhance or mitigate ice flow. We developed a fully automated method for tracking meltwater and rapid drainages in large (> 0.125 km2) perennial lakes and applied it to a 10 yr time series of ETM+ and MODIS imagery of an outlet glacier flow band in West Greenland. Results indicate interannual variability in maximum coverage and spatial evolution of total lake area. We identify 238 rapid drainage events, occurring most often at low (< 900 m) and middle (900–1200 m) elevations during periods of net filling or peak lake coverage. We observe a general progression of both lake filling and draining from lower to higher elevations but note that the timing of filling onset, peak coverage, and dissipation are also variable. Lake coverage is sensitive to air temperature, and warm years exhibit greater variability in both coverage evolution and rapid drainage. Mid-elevation drainages in 2011 coincide with large surface velocity increases at nearby GPS sites, though the relationships between ice-shed-scale dynamics and meltwater input are still unclear.

Abstract Background KDM6A, encoding a histone demethylase, is one of the top ten mutated epigenetic cancer genes. The effect of mutations on its structure and function are however poorly characterized. Methods Database search identified nonsense and missense mutations in the N-terminal TPR motifs and the C-terminal, catalytic JmjC domain, but also in the intrinsically disordered region connecting both these two well-structured domains. KDM6A variants with cancer-derived mutations were generated using site directed mutagenesis and fused to eGFP serving as an all-in-one affinity and fluorescence tag to study demethylase activity by an ELISA-based assay in vitro, apoptosis by FACS, complex assembly by Co-immunoprecipitation and localization by microscopy in urothelial cells and apoptosis by FACS. Results Independent of the mutation and demethylase activity, all KDM6A variants were detectable in the nucleus. Truncated KDM6A variants displayed changes in complex assemblies affecting (1) known interactions with the COMPASS complex component RBBP5 and (2) KDM6A-DNA associated assemblies with the nuclear protein Nucleophosmin. Some KDM6A variants induced a severe cellular phenotype characterized by multiple acute effects on nuclear integrity, namely, release of nuclear DNA into the cytoplasm, increased level of DNA damage indicators RAD51 and p-γH2A.X, and mitosis defects. These damaging effects were correlated with increased cell death. Conclusion These observations reveal novel effects of pathogenic variants pointing at new specific functions of KDM6A variants. The underlying mechanisms and affected pathways have to be investigated in future research to understand how tumor cells cope with and benefit from KDM6A truncations.
