1. Cell type and sex specific mitochondrial phenotypes in iPSC derived models of Alzheimer’s disease

Abstract

IntroductionMitochondrial dysfunction is observed in Alzheimer’s disease (AD). Altered mitochondrial respiration, cytochrome oxidase (COX) Vmax, and mitophagy are observed in human subjects and animal models of AD. Models derived from induced pluripotent stem cells (iPSCs) may not recapitulate these phenotypes after reprogramming from differentiated adult cells.MethodsWe examined mitochondrial function across iPSC derived models including cerebral organoids, forebrain neurons, and astrocytes. iPSCs were reprogrammed from fibroblasts either from the University of Kansas Alzheimer’s Disease Research Center (KU ADRC) cohort or purchased from WiCell. A total of four non-demented and four sporadic AD iPSC lines were examined. Models were subjected to mitochondrial respiration analysis using Seahorse XF technology, spectrophotometric cytochrome oxidase (COX) Vmax assays, fluorescent assays to determine mitochondrial mass, mitochondrial membrane potential, calcium, mitochondrial dynamics, and mitophagy levels. AD pathological hallmarks were also measured.ResultsiPSC derived neurons and cerebral organoids showed reduced COX Vmax in AD subjects with more profound defects in the female cohort. These results were not observed in astrocytes. iPSC derived neurons and astrocytes from AD subjects had reduced mitochondrial respiration parameters with increased glycolytic flux. iPSC derived neurons and astrocytes from AD subjects showed sex dependent effects on mitochondrial membrane potential, mitochondrial superoxide production, and mitochondrial calcium. iPSC derived neurons from AD subjects had reduced mitochondrial localization in lysosomes with sex dependent effects on mitochondrial mass, while iPSC derived astrocytes from female AD subjects had increased mitochondrial localization to lysosomes. Both iPSC derived neurons and astrocytes from AD subjects showed altered mitochondrial dynamics. iPSC derived neurons had increased secreted Aβ, and sex dependent effects on total APP protein expression. iPSC derived astrocytes showed sex dependent changes in GFAP expression in AD derived cells.ConclusionOverall, iPSC derived models from AD subjects show mitochondrial phenotypes and AD pathological hallmarks in a cell type and sex dependent manner. These results highlight the importance of sex as a biological variable in cell culture studies.

2. Mercury contamination of an introduced generalist fish of intermediate trophic level

Abstract

Mercury contamination is a global issue because mercury concentrations in aquatic systems are influenced by both natural and anthropogenic pathways, including the burning of fossil fuels and flooding during hydroelectric development. Mercury biomagnifies in aquatic ecosystems, leading to higher concentrations in piscivore fishes than those at lower trophic levels. Here, liver and muscle total mercury (THg) concentrations in black crappiePomoxis nigromaculatusfrom three lakes in southeastern Manitoba, Canada were related to age, morphology and physiological traits to better understand the dynamics of mercury accumulation in an introduced generalist fish species. Black crappie liver and muscle samples from Big Whiteshell Lake (relatively large lake, 17.5 km2; n=30), Caddy Lake (small lake surrounded by wetlands, 3.1 km2; n=42) and Lac du Bonnet (river widening influenced by hydroelectric dams, 84.0 km2; n=29) were analyzed for THg content. These THg concentrations were then compared to black crappie mercury concentrations in other Canadian water bodies to assess within species relative contamination levels, as well as to mercury concentrations in other freshwater fishes to examine biomagnification. Age and size had strong positive correlations (r>0.60) with muscle mercury concentrations. No evidence of acute point source contamination was found in the study area when compared to black crappie muscle mercury concentrations in other water bodies, and tissue THg concentration was not correlated with a reduction in gonadosomatic index (GSI) or hepatosomatic index (HSI). Analysis of liver THg in addition to muscle THg revealed the possible impacts of seasonal and ontogenetic differences in diet on exposure. Furthermore, THg analysis of liver and muscle tissue showed how generalist foraging techniques of black crappie may curb the progressively greater mercury exposure and resultant physiological consequences expected from ontogenetic diet shifts from invertebrates to fishes. Although there appeared to be temporally varied levels of mercury exposure (i.e., liver THg) by sex, there was no sex effect observed in long-term accumulation in the muscle. Flood risk is believed to be a key driver of differences in black crappie THg concentrations between lakes in the region. Black crappie bioaccumulated less mercury at age than primary piscivore species in the region. These results will help foster a better understanding of mercury biomagnification in boreal shield lakes within a region impacted by legacy mercury.

Abstract

Membrane polarization occurs in sediments with different surface area of capillaries (pores) and is regarded as a slow type of polarization. This phenomenon is the foundation of the well known methods of induced polarization (IP): time domain and frequency domain induced polarization. The characteristic parameters of induced polarization which are required for studying physical properties of rocks are measured in the laboratory. Data measured in the laboratory confirmed the distinctions of IP processes at time-on and time-off. Additionally linear dependence of voltage and applied current is not always observed. This paper presents the first step of studying: theoretical consideration for time-on and mathematical modeling of membrane polarization, ion concentrations of electrolyte in the pores of different models of pores space, and arising voltage. The problem of concentration of ions along the pores can be solved using the diffusion equation with specified initial and boundary conditions. Reduced boundary conditions for time-on show that transient concentrations at the boundaries are linear with time. It allows obtaining the analytical solution for this equation. Mathematical modeling has been performed for different combinations of pores. It is shown that if electrical current flows from the pores with greater transfer numbers to the pores with smaller transfer numbers, an excess of ions will be observed at this boundary. If the difference of transfer numbers is negative, there is a decrease in the concentration of ions at the vicinity of the boundary. This decrease will continue until the concentration at this boundary reaches zero. In this case the galvanic chain will be interrupted and electrical current flowing through the sample does not penetrate to this cell. The duration of the process of ions distribution in the pore and time of blockage t0 is proportional to the radii of contacted pores and inversely proportional to the transfer number difference and square of the current flowing through this cell. It was shown by both laboratory measurement and field processes that induced polarization relates to low porous rocks with small transfer number differences.

Abstract

The technique of nuclear emulsions for high-energy physics experiments is being revived, thanks to the remarkable progress in measurement automation achieved in the past years. The present paper describes the features and performances of the European Scanning System, a last-generation automatic microscope working at a scanning speed of 20 cm 2 / h . The system has been developed in the framework of the OPERA experiment, designed to unambigously detect ν μ → ν τ oscillations in nuclear emulsions.

Abstract

A new approach to modeling the seismoelectric (seismo-electrokinetic) effect is presented. It is based on the concept that at a specific time each location on the wave front that proceeds through the medium imparts equal pressure on the solid matrix and pore space. Consequently two fundamental equations contribute to the basis for the calculation of the electrokinetic effect (EKE): the first describes the relative displacement of particles in the sediment at the arrivals of the seismic wave and second, the transient streaming potential, induced in a pore, when the pressure pulse is applied to a cylindrical pore. To calculate the EKE on the surface the following are required: calculate the pressure gradient in a single pore caused by a seismic wave, the transfer from EKE arising in a single pore to the porous media, integrate the EKE signals all over the sediment space or layer and access the electrokinetic coefficients. A new algorithm has been developed. Being able to model field data mathematically shows the viability of this approach and allows the estimation of permeability of layers. To obtain a realistic geological and groundwater model test site, several geophysical methods were applied to a small area (shallow refraction seismic, DC resistivity, SP, FDEM, magnetic gradiometry). Comparing the EKE result modelled with the new algorithm with the above methods shows that the seismo-electrokinetic method can be successfully used for the exploration for ground water at shallow depths.

Abstract

The design, construction, and performance of a linear radio-frequency ion trap (RFQ) intended for use in the Enriched Xenon Observatory (EXO) are described. EXO aims to detect the neutrinoless double-beta decay of $^{136}$Xe to $^{136}$Ba. To suppress possible backgrounds EXO will complement the measurement of decay energy and, to some extent, topology of candidate events in a Xe filled detector with the identification of the daughter nucleus ($^{136}$Ba). The ion trap described here is capable of accepting, cooling, and confining individual Ba ions extracted from the site of the candidate double-beta decay event. Abstract

Asymmetries in quasi-elastic 3 He ( e → , e ′ p ) have been measured at a momentum transfer of 0.67 (GeV/ c ) 2 and are compared to a calculation which takes into account relativistic kinematics in the final state and a relativistic one-body current operator. With an exact solution of the Faddeev equation for the 3 He -ground state and an approximate treatment of final state interactions in the continuum good agreement is found with the experimental data.

Abstract

Precise data on the neutron magnetic form factor G_{mn} have been obtained with measurements of the ratio of cross sections of D(e,e'n) and D(e,e'p) up to momentum transfers of Q^2 = 0.9 (GeV/c)^2. Data with typical uncertainties of 1.5% are presented. Abstract

Polarization transfer in the 4He ( e → ,e′ p → ) 3 H reaction at a Q2 of 0.4 (GeV/c)2 was measured at the Mainz Microtron MAMI. The ratio of the transverse to the longitudinal polarization components of the ejected protons was compared with the same ratio for elastic ep scattering. The results are consistent with a recent fully relativistic calculation which includes a predicted medium modification of the proton form factor based on a quark–meson coupling model.

Abstract

We have measured the neutron electric form factor ${G}_{\mathrm{en}}$ via ${}^{3}\stackrel{\ensuremath{\rightarrow}}{\mathrm{He}}(\stackrel{\ensuremath{\rightarrow}}{e}{,e}^{\ensuremath{'}}n)\mathrm{pp}$ at ${Q}^{2}\phantom{\rule{0ex}{0ex}}=\phantom{\rule{0ex}{0ex}}0.67(\mathrm{GeV}/c{)}^{2}$ using the 3-spectrometer facility of the A1 Collaboration at the Mainz Microtron and a dedicated neutron detector. High pressure polarized ${}^{3}\stackrel{\ensuremath{\rightarrow}}{\mathrm{He}}$ gas was used as a target of polarized neutrons. ${G}_{\mathrm{en}}$ is determined from the ratio of the asymmetries ${A}_{\ensuremath{\perp}}{/A}_{\ensuremath{\parallel}}$ measured in quasifree kinematics with the target spin perpendicular and parallel to the momentum transfer. Abstract

The OPERA neutrino detector at the underground Gran Sasso Laboratory (LNGS) was designed to perform the first detection of neutrino oscillations in appearance mode, through the study of nu_mu to nu_tau oscillations. The apparatus consists of a lead/emulsion-film target complemented by electronic detectors. It is placed in the high-energy, long-baseline CERN to LNGS beam (CNGS) 730 km away from the neutrino source. In August 2006 a first run with CNGS neutrinos was successfully conducted. A first sample of neutrino events was collected, statistically consistent with the integrated beam intensity. After a brief description of the beam and of the various sub-detectors, we report on the achievement of this milestone, presenting the first data and some analysis results., Submitted to the New Journal of Physics

The use of nuclear emulsions in very large physics experiments is now possible thanks to the recent improvements in the industrial production of emulsions and to the development of fast automated microscopes. Abstract

We studied the reaction {sup 12}C(e,e{sup '}p) in quasielastic kinematics at momentum transfers between 0.6 and 1.8 (GeV/c){sup 2} covering the single-particle region. From this the nuclear transparency factors are extracted using two methods. The results are compared to theoretical predictions obtained using a generalization of Glauber theory described in this paper. Furthermore, the momentum distribution in the region of the 1s-state up to momenta of 300 MeV/c is obtained from the data and compared to the correlated basis function theory and the independent-particle shell model.

Abstract

We have carried out an (e,e'p) experiment at high momentum transfer and in parallel kinematics to measure the strength of the nuclear spectral function S(k,E) at high nucleon momenta k and large removal energies E. This strength is related to the presence of short-range and tensor correlations, and was known hitherto only indirectly and with considerable uncertainty from the lack of strength in the independent-particle region. Abstract

The electric form factor of the neutron was determined from measurements of the (d) over right arrow((e) over right arrow ,e(`)n)p reaction for quasielastic kinematics. Polarized electrons were scattered off a polarized deuterated ammonia ((ND3)-N-15) target in which the deuteron polarization was perpendicular to the momentum transfer. The scattered electrons were detected in a magnetic spectrometer in coincidence with neutrons in a large solid angle detector. We find G(E)(n)=0.0526+/-0.0033(stat)+/-0.0026(sys) and 0.0454+/-0.0054+/-0.0037 at Q(2)=0.5 and 1.0 (GeV/c)(2), respectively.

Abstract

We report the first measurement using a solid polarized target of the neutron electric form factor G(n)(E) via d-->(e-->,e(')n)p. G(n)(E) was determined from the beam-target asymmetry in the scattering of longitudinally polarized electrons from polarized deuterated ammonia ( 15ND3). The measurement was performed in Hall C at Thomas Jefferson National Accelerator Facility in quasifree kinematics with the target polarization perpendicular to the momentum transfer. The electrons were detected in a magnetic spectrometer in coincidence with neutrons in a large solid angle segmented detector. We find G(n)(E) = 0.04632+/-0.00616(stat)+/-0.00341(syst) at Q2 = 0.495 (GeV/c)(2).

Abstract

In the frame of the A1-Collaboration at the Mainz Microtron a test measurement of doubly polarized 3He(e,e′n) scattering from a high pressure target was performed in July aiming for the determination of the neutron electric form factor Gen at high momentum transfer (Q2=0.7(GeV/c)2). Due to the small value of Gen compared to Gmn a preferred procedure is to determine the asymmetry in the exclusive quasi elastic scattering of polarized electrons (P≈70%, I⩾2μA) from polarized 3He. The scattered electrons are detected in a high resolution magnetic spectrometer while the scattering angles of the outgoing neutrons are measured in a plastic scintillator. In this reaction the polarized 3He nucleus serves as an effective polarized neutron target. Because of the large magnetic field gradients caused by the spectrometer and limited space at the target place, the 3He gas is polarized elsewhere and transported to the target place in specially prepared glass cells. The glass cells are designed for high pressure (up to 10 bar) and with thin windows to prevent background. To reduce the relaxation due to the magnetic field gradients a μ-metal shielded guiding field of 4 Gauss is used. The guiding field is generated by three independent coils which permit the rotation of the target spin in any direction desired, especially perpendicular and parallel with respect to q. So we are able to take the ratio of the asymmetries A⊥/A∥, which in first order depends only on kinematical factors and the ratio Gen/Gmn. The method of metastable optical pumping is used to polarize the 3He at about 1 mbar with a LNA laser (≈6W). We have studied the performance of a new algorithm for electron/pion separation in an Emulsion Cloud Chamber (ECC) made of lead and nuclear emulsion films. The software for separation consists of two parts: a shower reconstruction algorithm and a Neural Network that assigns to each reconstructed shower the probability to be an electron or a pion. The performance has been studied for the ECC of the OPERA experiment [1]. The $e/\pi$ separation algorithm has been optimized by using a detailed Monte Carlo simulation of the ECC and tested on real data taken at CERN (pion beams) and at DESY (electron beams). The algorithm allows to achieve a 90% electron identification efficiency with a pion misidentification smaller than 1% for energies higher than 2 GeV. Abstract

The OPERA neutrino oscillation experiment has been designed to prove the appearance of ντ in a nearly pure νμ beam (CNGS) produced at CERN and detected in the underground Hall C of the Gran Sasso Laboratory, 730 km away from the source. In OPERA, τ leptons resulting from the interaction of ντ are produced in target units called bricks made of nuclear emulsion films interleaved with lead plates. The OPERA target contains 150000 of such bricks, for a total mass of 1.25 kton, arranged into walls interleaved with plastic scintillator strips. The detector is split into two identical supermodules, each supermodule containing a target section followed by a magnetic spectrometer for momentum and charge measurement of penetrating particles. Real time information from the scintillators and the spectrometers provide the identification of the bricks where the neutrino interactions occurred. The candidate bricks are extracted from the walls and, after X-ray marking and an exposure to cosmic rays for alignment, their emulsion films are developed and sent to the emulsion scanning laboratories to perform the accurate scan of the event. In this paper, we review the design and construction of the detector and of its related infrastructures, and report on some technical performances of the various components. The construction of the detector started in 2003 and it was completed in Summer 2008. The experiment is presently in the data taking phase. 29. Male and Female Perceptions of Supervision During Strength Training.

Abstract

A cross-sectional survey was distributed to 1322 members of a 1-on-1 personalized strength training studio. A total of 366 respondents ( n = 134 male and n = 232 female), all aged over 20 years, reported considerable training experience, with 55% of the males and 42% of the females reporting 5+ years of experience. The data were analyzed and reported descriptively with differences >5% identified based on the use of a 5-point Likert scale, the sample size, and the nature of the observations. Disparities between the males and females were identified; the males reported higher perceptions of managing effort, technique, and programming without supervision compared to the females. Safety was noted as being more important to the females compared to the males. Qualitatively, additional themes were raised including an analogy of the personal relationship between the trainer and trainee being similar to that between medical professionals and patients. This was validated where the participants discussed their adaptations from supervised strength training for maintaining quality of life in aging and recovering from medical conditions and injury. The data are discussed in the context of a previous body of literature suggesting males falsely report higher levels of confidence in tasks compared to females, particularly in relation to effort, role models, and verbal encouragement. We posit that the greater confidence expressed by males at least partially explains the greater engagement in strength training practices by males compared to females, as well as explaining the higher level of participation in supervised strength training by females compared to males. This research proves beneficial for strength training practitioners in enhancing their understanding and expectations of clients, as well as hopefully proving insightful in engaging more people in strength training.

Abstract

Background: Dietary recommendations in inflammatory bowel disease (IBD) are inconclusive, and patients may follow restrictive diets with increased risk of malnutrition. The aim of this study was to compare dietary intakes and nutritional status in men and women with newly diagnosed IBD with a general population sample, and to investigate whether intakes were in line with the Nordic Nutrition Recommendations., Methods: This was a cross-sectional study including adults≥ 40 years with IBD from the Inflammatory Bowel Disease in South-Eastern Norway (IBSEN) III cohort study. A validated food frequency questionnaire (FFQ) was used in dietary data collection, and a sample from the seventh survey of the Tromsø Study was included as a comparison group., Results: A total of 227 men and women with IBD were included. IBD patients had higher intake of grain products, sweetened beverages, energy, fat and polyunsaturated fat (PUFA), but lower intake of dairy products, alcohol and iodine compared to adults from the comparison sample ( p  < 0.01). Intakes of saturated fat and carbohydrates in both genders, and vitamin D in women were not within recommended levels. Anemia and hypoalbuminemia were more prevalent in IBD patients than in the comparison sample., Conclusions: Dietary intakes in newly diagnosed IBD patients were mostly in line with Nordic Nutrition Recommendations. Higher proportion of IBD patients exceeded recommended allowances of fat and added sugar than the comparison sample. Abstract

Objectives: Dietary environmental impact in a Norwegian adult population was estimated for six environmental impact categories. Moreover, environmental benefits of scenario diets complying with the Norwegian Food-Based Dietary Guidelines (FBDG) and the EAT-Lancet reference diet were assessed., Design: The current diet of Norwegian adults was estimated according to 24-h dietary recall data from a national dietary surveillance survey (Norkost 3). Scenario diets were modelled to represent the Norwegian FBDG and the EAT-Lancet healthy reference diet. Dietary environmental impact in terms of global warming potential, freshwater and marine eutrophication, terrestrial acidification, water use and transformation and use of land was estimated for the current and scenario diets using environmental impact data representative of the Norwegian market. Significant associations between impact and gender/educational attainment were assessed at P < 0·05., Setting: Norway., Participants: Adults ( n =1787) aged 18-70 years who participated in the Norkost 3 survey (2010-2011)., Results: Environmental impact varied significantly by gender and educational attainment. The food groups contributing most to environmental impact of Norwegian diets were meat, dairy, beverages, grains and composite dishes. Compared with the current Norwegian diet, the FBDG scenario reduced impacts from 2 % (freshwater eutrophication) to 32 % (water use), while the EAT-Lancet scenario reduced impacts from 7 % (marine eutrophication) to 61 % (land use). The EAT-Lancet scenario resulted in 3-48 % larger reductions in impact than the FBDG scenario., Conclusions: The Norwegian FBDG, while not as environmentally friendly as the EAT-Lancet reference diet, can still be an important tool in lessening environmental burden of Norwegian diets.

Abstract

Mercury contamination is a global issue because mercury concentrations in aquatic systems are influenced by both natural and anthropogenic pathways. Here, liver and muscle total mercury (THg) concentrations in black crappie Pomoxis nigromaculatus from three boreal lakes in southeastern Manitoba, Canada, were related to age, morphology and physiological traits to better understand the dynamics of mercury accumulation in an introduced generalist fish species. These THg concentrations were then compared to black crappie mercury concentrations in other Canadian water bodies and to mercury concentrations in other freshwater fishes in southeastern Manitoba. Age and size had strong positive correlations (P < 0.001, r ≥ 0.60) with muscle mercury concentrations. No evidence of acute point source contamination or physiological impairment in black crappie was found in the study area. Analysis of liver THg revealed the possible impacts of seasonal and ontogenetic differences in diet on exposure. Furthermore, THg analysis of liver and muscle tissue showed how generalist foraging may curb the progressively greater mercury exposure and resultant physiological consequences expected from ontogenetic diet shifts in black crappie. Although there appeared to be temporally varied levels of mercury exposure (i.e., liver THg) by sex, there was no sex effect observed in long-term mercury accumulation in the muscle. Black crappie bioaccumulated less mercury at age than primary piscivore species in the region. These results will help foster a better understanding of mercury biomagnification within a region impacted by legacy mercury., (© 2023. 33. Differentiation of Intracerebral Tumor Entities with Quantitative Contrast Attenuation and Iodine Mapping in Dual-Layer Computed Tomography.

Abstract

Purpose: To investigate if quantitative contrast enhancement and iodine mapping of common brain tumor (BT) entities may correctly differentiate between tumor etiologies in standardized stereotactic CT protocols. Material and Methods: A retrospective monocentric study of 139 consecutive standardized dual-layer dual-energy CT (dlDECT) scans conducted prior to the stereotactic needle biopsy of untreated primary brain tumor lesions. Attenuation of contrast-enhancing BT was derived from polyenergetic images as well as spectral iodine density maps (IDM) and their contrast-to-noise-ratios (CNR) were determined using ROI measures in contrast-enhancing BT and healthy contralateral white matter. The measures were correlated to histopathology regarding tumor entity, isocitrate dehydrogenase (IDH) and MGMT mutation status. Results: The cohort included 52 female and 76 male patients, mean age of 59.4 (±17.1) years. Brain lymphomas showed the highest attenuation (IDM CNR 3.28 ± 1,23), significantly higher than glioblastoma (2.37 ± 1.55, p < 0.005) and metastases (1.95 ± 1.14, p < 0.02), while the differences between glioblastomas and metastases were not significant. These strongly enhancing lesions differed from oligodendroglioma and astrocytoma (Grade II and III) that showed IDM CNR in the range of 1.22−1.27 (±0.45−0.82). Conventional attenuation measurements in DLCT data performed equally or slightly superior to iodine density measurements. Conclusion: Quantitative attenuation and iodine density measurements of contrast-enhancing brain tumors are feasible imaging biomarkers for the discrimination of cerebral tumor lesions but not specifically for single tumor entities. CNR based on simple HU measurements performed equally or slightly superior to iodine quantification.

Abstract

Background: Adequate sleep is vital for health and quality of life. People with stroke and a concomitant sleep disorder may have poorer outcomes than those without a sleep disorder., Objective: To systematically evaluate the published literature to determine the impact of sleep disorders on physical, functional recovery at the activity and participation level after stroke., Methods: A systematic review was conducted using PubMed, CINAHL, Scopus, and PsycINFO. Studies were selected that reported outcomes on physical, functional recovery at the activity and participation levels in participants with stroke and a diagnosed sleep disorder. A meta-analysis was performed on included studies that reported Barthel Index (BI) and modified Rankin Scale (mRS) scores. Results: A total of 33 studies were included in the systematic review with 9 of them in the meta-analysis. The mean mRS score was 0.51 points higher in participants with stroke and sleep disorders versus participants with stroke without sleep disorder [95% CI: 0.23-0.78]. The mean BI score was 10.2 points lower in participants with stroke and sleep disorders versus participants with stroke without sleep disorder [95% CI: -17.9 to -2.6]., Conclusions: People with stroke and a sleep disorder have greater functional limitations and disability than those without a sleep disorder. Rehabilitation professionals should screen their patients with stroke for potential sleep disorders and further research is needed to develop sleep and rehabilitation interventions that can be delivered in combination. Objective: We aimed to determine optimal window settings for conventional polyenergetic (PolyE) and virtual monoenergetic images (MonoE) derived from abdominal portal venous phase computed tomography (CT) examinations on a novel dual-layer spectral-detector CT (SDCT)., Methods: From 50 patients, SDCT data sets MonoE at 40 kiloelectron volt as well as PolyE were reconstructed and best individual window width and level values manually were assessed separately for evaluation of abdominal arteries as well as for liver lesions. Via regression analysis, optimized individual values were mathematically calculated. Subjective image quality parameters, vessel, and liver lesion diameters were measured to determine influences of different W/L settings., Results: Attenuation and contrast-to-noise values were significantly higher in MonoE compared with PolyE. Compared with standard settings, almost all adjusted W/L settings varied significantly and yielded higher subjective scoring. No differences were found between manually adjusted and mathematically calculated W/L settings., Conclusions: PolyE and MonoE from abdominal portal venous phase SDCT examinations require appropriate W/L settings depending on reconstruction technique and assessment focus.

Objectives: To investigate the utilization of virtual mono-energetic images (MonoE) at low kiloelectron volt (keV) levels derived from a dual-layer spectral detector CT (SDCT) for the assessment of abdominal arteries in venous contrast phase scans using arterial phase imaging as an internal reference standard., Materials and Methods: A total of 50 patients who received arterial and venous phase imaging of the abdomen on a SDCT system were included in this study. Absolute attenuation, noise, signal- and contrast to noise ratios (SNR; CNR) as well as arterial diameters in defined landmarks were assessed. In arterial phase, conventional reconstructions (CR ART ) as well as MonoE ART at 40keV and in venous phase, conventional reconstructions (CR VEN ) as well as MonoE VEN at 70 and 40keV were investigated and intra-individual comparisons were performed. If an artery stenosis (10 patients) was present, the degree of stenosis was assessed according to the system of the North American Symptomatic Carotid Endarterectomy Trial (NASCET)., Results: MonoE 40keV yielded significantly higher attenuation values (in arterial as well as in venous phase) compared to CR ART (p<0.001) while noise levels were substantially low. This resulted in markedly superior SNR and CNR in large vessel compared to CR ART . Luminal diameters were significantly smaller in MonoE 40keV in both contrast phases compared to CR ART (p<0.001), whereas no significant differences were found between both MonoE reconstructions (p≥0.92). The degree of vessel stenosis was significantly higher in MonoE 40keV of both contrast phases compared to CR ART (p≥0.02)., Conclusion: MonoE at low keV of venous contrast phase scans derived from a novel SDCT are suitable for the assessment of arteries in the abdomen and subsequent stenosis assessment. However, MonoE at 40keV constantly showed significant smaller luminal diameters than the corresponding conventional reconstructions (including the reference standard). This is possibly due to an improved differentiation of the vessel lumen from the wall and raises the question, which imaging technique should be used as an appropriate reference standard for vascular SDCT imaging studies., (Copyright © 2017 Elsevier B.V. Objectives: To quantitatively and qualitatively assess abdominal arterial and venous phase contrast-enhanced spectral detector computed tomography (SDCT) virtual mono-energetic (MonoE) datasets in comparison to conventional CT reconstructions provided by the same system., Materials and Methods: Conventional and MonoE images at 40-120 kilo-electron volt (keV) levels with a 10 keV increment as well as 160 and 200 keV were reconstructed in abdominal SDCT datasets of 55 patients. Attenuation, image noise, and contrast- / signal-to-noise ratios (CNR, SNR) of vessels and solid organs were compared between MonoE and conventional reconstructions. Two readers assessed contrast conditions, detail visualization, overall image quality and subjective image noise with both, fixed and adjustable window settings., Results: Attenuation, CNR and SNR of vessels and solid organs showed a stepwise increase from high to low keV reconstructions in both contrast phases while image noise stayed stable at low keV MonoE reconstruction levels. Highest levels were found at 40 keV MonoE reconstruction (p<0.001), respectively. Solid abdominal organs showed a stepwise decrease from low to high energy levels in regard to attenuation, CNR and SNR with significantly higher values at 40 and 50 keV, compared to conventional images. The 70 keV MonoE was comparable to conventional poly-energetic reconstruction (p≥0.99). Subjective analysis displayed best image quality for the 70 keV MonoE reconstruction level in both phases at fixed standard window presets and at 40 keV if window settings could be adjusted., Conclusion: SDCT derived low keV MonoE showed markedly increased CNR and SNR values due to constantly low image noise values over the whole energy spectrum from 40 to 200 keV.

Objectives: To evaluate image quality parameters of virtual mono-energetic (MonoE) and conventional (CR) imaging derived from a dual-layer spectral detector CT (DLCT) in oncological follow-up venous phase imaging of the chest and comparison with conventional multi-detector CT (CR MDCT ) imaging., Materials and Methods: A total of 55 patients who had oncologic staging with conventional CT and DLCT of the chest in venous phase were included in this study. Established image quality parameters were derived from all datasets in defined thoracic landmarks. Attenuation, image noise, and signal-/contrast- to noise ratios (SNR, CNR) were compared between CR DLCT and MonoE as well as CR MDCT imaging. Two readers performed subjective image analysis., Results: CR MDCT showed significant lower attenuation values compared to CR DLCT and MonoE at 40-70keV (p≤0.05). Moreover, MonoE at 40-70keV revealed significantly higher attenuations values compared to CR DLCT (p<0.001). Noise was statistically lower in CR MDCT compared with CR DLCT and MonoE at 40keV (11.4±2.3 HU vs. 12.0±3.1 HU vs. 11.7±5.2 HU; p<0.001). In contrast, all MonoE levels showed significantly lower noise levels compared to CR DLCT (p<0.001). SNR was not significantly different between CR MDCT and CR DLCT (13.5±3.7 vs. 14.4±5.3; p>0.99). SNR values were significantly increased for MonoE at 40-80keV compared to CR MDCT and CR DLCT (p<0.001). CR DLCT and MonoE (40-70keV) from DLCT revealed significantly higher CNR values than CR MDCT (p<0.001). In subjective analysis, MonoE at 40keV surpassed all other image reconstructions except for noise in MonoE at 70 keV., Conclusion: In dual-layer spectral detector CT, MonoE at low keV showed superior image quality compared to conventional images derived from the same system and may therefore be added to clinical routine imaging protocols. Abstract

Purpose: This phase II study evaluated the safety and efficacy of weekly docetaxel and capecitabine in patients with metastatic breast cancer., Patients and Methods: Thirty-nine patients with metastatic breast cancer received 30 mg/m2 of docetaxel on days 1, 8, and 15 in combination with capecitabine 800 mg/m2 twice daily on days 1-21, repeated every 28 days., Results: The median number of treatment cycles was 4 (range, 1-20 cycles). Grade 3 toxicities per patient were asthenia (n = 7; 18%), diarrhea (n = 7; 18%), nausea/vomiting (n = 5; 13%), stomatitis (n = 5; 13%), neutropenia (n = 5; 13%), and hand-foot syndrome (n = 4; 10%). There were only 2 grade 4 toxicities, febrile neutropenia and pulmonary embolism. The overall response rate was 44% (95% confidence interval (CI), 28%-60%), median duration of response was 9.1 months (95% CI, 6.2-12 months), and median time to progression was 5.5 months (95% CI, 3.7-7.3 months)., Conclusion: Weekly docetaxel with capecitabine was active with acceptable toxicities. Additional trials to define the optimal schedule of docetaxel and capecitabine are justified.

Abstract

Purpose: To evaluate the safety and efficacy of bevacizumab and weekly docetaxel as first- or second-line therapy in patients with metastatic breast cancer (MBC)., Patients and Methods: Twenty-seven MBC patients received i.v. bevacizumab at 10 mg/kg on days 1 and 15 in combination with i.v. docetaxel 35 mg/m2 on days 1, 8, and 15 of a 28-day cycle. Primary end points were to assess toxicity, overall response rate, and progression-free survival. A secondary end point was to assess the relationship between plasma endothelial and cell adhesion markers and clinical outcomes., Results: One-hundred fifty-eight treatment cycles were administered with a median of six cycles (range 1-15 cycles) per patient. The most common grade 4 toxicities per patient were as follows: 2 (7%)-pulmonary embolus, 1 (4%)-febrile neutropenia, and 1 (4%)-infection; grade 3 toxicities were 4 (15%)-neutropenia, 4 (15%)-fatigue, 2 (7%)-neuropathy, 2 (7%)-athralgias, 2 (7%)-stomatitis, 1 (7%)-pleural effusion, and 1 (4%)-hypertension. The overall response rate was 52% [95% confidence interval (95% CI), 32-71%], median response duration was 6.0 months (95% CI, 4.6-6.5 months), and the median progression-free survival was 7.5 months (95% CI, 6.2-8.3 months). In hypothesis-generating univariate and limited multivariate analyses, E-selectin was statistically significantly associated with response to the combination., Conclusion: Bevazicumab in combination with weekly docetaxel is active with acceptable toxicities in MBC. Additional studies evaluating E-selectin as a marker of response to bevacizumab-containing chemotherapy are warranted.

Abstract

Purpose: In most neoadjuvant chemotherapy regimens, the taxane is administered either in combination with an anthracycline or after an anthracycline-containing regimen. We sought to test the feasibility, safety, and determine the pathological complete response (pCR) rate of administering docetaxel first followed by epirubicin as neoadjuvant chemotherapy in women with clinical stage II, III breast cancer., Patients and Methods: Twenty-five women with newly diagnosed clinical stage IIB (n = 10), IIIA (n = 5), or IIIB (n = 10) received 3 cycles of docetaxel 100 mg/M2 intravenously (IV) every 3 weeks followed by 3 cycles of epirubicin 100 mg/M2 IV every 3 weeks. pCR was defined as the absence of invasive cancer in the breast at definitive surgery., Results: The median primary tumor size was 6 cm (range 1-12 cm), and 13 (52%) women had clinically palpable axillary lymph nodes. Patients received 149 of the 150 planned cycles of docetaxel and epirubicin without treatment delays, and only 3 (12%) patients had a dose reduction of docetaxel. Seven (28%) patients experienced febrile neutropenia, and 9 (36%) patients had grade 3 non-hematological toxicities with diarrhea being the most frequent in 3 (12%) patients. Six (24%) patients had pCR in the breast. Analysis of pre- and post-docetaxel biopsies from a subset of patients documented taxane-induced activation of mitogen-activated and stress-activated protein kinase pathways., Conclusion: Neoadjuvant docetaxel followed by epirubicin is well tolerated and active in breast cancer. To our knowledge, this is first description of docetaxel-induced activation of mitogen-activated and stress-activated protein kinase pathways in human breast cancer.

We have carried out an (e,e'p) experiment at high momentum transfer and in parallel kinematics to measure the strength of the nuclear spectral function S(k,E) at high nucleon momenta k and large removal energies E. This strength is related to the presence of short-range and tensor correlations, and was known hitherto only indirectly and with considerable uncertainty from the lack of strength in the independent-particle region. This experiment locates by direct measurement the correlated strength predicted by theory.

Abstract

The electric form factor of the neutron was determined from measurements of the d-->(e-->,e'n)p reaction for quasielastic kinematics. Polarized electrons were scattered off a polarized deuterated ammonia (15ND3) target in which the deuteron polarization was perpendicular to the momentum transfer. The scattered electrons were detected in a magnetic spectrometer in coincidence with neutrons in a large solid angle detector. We find G(n)(E)=0.0526+/-0.0033(stat)+/-0.0026(sys) and 0.0454+/-0.0054+/-0.0037 at Q(2)=0.5 and 1.0 (GeV/c)(2), respectively.

Abstract

Several neuropeptides are secreted in high amounts in pediatric tumors such as neuroblastoma and have been used as markers of residual or recurrent disease. Plasma levels of neuropeptides might be expected to change during development, but have not been determined in normal children. We have obtained fresh plasma from cord blood of six full-term infants and from peripheral blood in 41 healthy children, ages 1 month to 21 years. Levels of six neuropeptides, vasoactive intestinal peptide (VIP), somatostatin, gastrin releasing peptide (GRP), substance P, pancreastatin and neuropeptide Y (NPY) were measured by radioimmunoassay along with insulin-like growth factor-1 (IGF-1) whose plasma levels are known to vary during development. A child with neuroblastoma was treated with the somatostatin analogue, octreotide, and the effect on plasma neuropeptides quantified. Octreotide doses of 2-3 microg/kg daily resulted in a 40-60% decrease in plasma levels of IGF-1, pancreastatin and GRP. These results are the first publication of plasma neuropeptide levels in normal children.

Abstract

Purpose: Capecitabine and docetaxel have demonstrated preclinical antitumor synergy. This synergy is thought to occur from docetaxel-mediated upregulation of thymidine phosphorylase (dThdPase), an enzyme responsible for the relative tumor selectivity of capecitabine. On the basis of the time-dependency and transiency for this upregulation, we performed a phase I study of capecitabine in combination with weekly docetaxel. We hypothesized that weekly docetaxel would result in sustained dThdPase expression and that capecitabine administration at times of maximum dThdPase upregulation would increase the therapeutic index for this combination., Patients and Methods: Patients with advanced solid malignancies received docetaxel on days 1, 8, and 15, and capecitabine bid on days 5 to 18, every 4 weeks. Docetaxel was fixed at 36 mg/m(2)/wk, whereas capecitabine was escalated in successive patients cohorts., Results: Sixteen patients received 77 courses at capecitabine doses from 950 to 1,500 mg/m(2)/d. The most common toxicities were hand-foot syndrome, diarrhea, nausea/vomiting, and asthenia. Grades 3 to 4 hematologic toxicities were infrequent and no treatment-related hospitalizations occurred. Three of three patients treated at 1,500/36 mg/m(2) capecitabine/docetaxel developed grade 3 hand-foot syndrome or diarrhea during either their first or second course, whereas only two of 13 patients at 1,250/36 mg/m(2) doses developed significant toxicity. Antitumor responses (n = 7) occurred in patients with hepatocellular, non-small-cell lung, and chemotherapy-refractory breast, bladder, and colorectal carcinomas. Prolonged stabilizations occurred in patients with metastatic mesothelioma (n = 2), chemorefractory non-small-cell lung carcinoma, and bronchioloalveolar carcinoma., Conclusion: Capecitabine in combination with weekly docetaxel is well tolerated. Recommended doses are capecitabine 1,250 mg/m(2)/d (625 mg/m(2) bid) with docetaxel 36 mg/m(2)/wk. The acceptable toxicity profile in this dose schedule, and the antitumor activity observed, warrant further evaluation of this regimen.

Abstract

We report the first measurement using a solid polarized target of the neutron electric form factor G(n)(E) via d-->(e-->,e(')n)p. G(n)(E) was determined from the beam-target asymmetry in the scattering of longitudinally polarized electrons from polarized deuterated ammonia ( 15ND3). The measurement was performed in Hall C at Thomas Jefferson National Accelerator Facility in quasifree kinematics with the target polarization perpendicular to the momentum transfer. The electrons were detected in a magnetic spectrometer in coincidence with neutrons in a large solid angle segmented detector. We find G(n)(E) = 0.04632+/-0.00616(stat)+/-0.00341(syst) at Q2 = 0.495 (GeV/c)(2).
