1. Implementation of 26 Gy in five fractions over 1 week adjuvant radiotherapy for breast cancer: Prospective report of acute skin toxicity and consideration of resource implications

Abstract: Purpose: In March 2020, a 1-week adjuvant breast radiotherapy schedule, 26 Gy in 5 fractions, was adopted to reduce the risk of COVID19 for staff and patients. This study quantifies acute toxicity rates and the effect on linac capacity. Materials and methods: This is a report of consecutive patients receiving ultrafractionated breast radiotherapy ( ± sequential boost) Mar–Aug 2020. Virtual consultations assessed acute skin toxicity during treatment and weeks 1, 2, 3 and 4 post treatment using CTCAE V5 scoring criteria. The number of linac minutes saved was estimated accounting for boost and DIBH use. Results: In total, 128/135 (95%) patients, including 31/33 boost patients, completed at least 3/5 assessments. 0/128 (0%) reported moist desquamation not confined to skin folds or minor bleeding (Grade 3), 41/128 (32%) reported brisk erythema, moist desquamation confined to skin folds or breast swelling (Grade 2), 62/128 (48%) reported faint erythema or dry desquamation (Grade 1) as their worst skin toxicity, with the remaining 20% reporting no skin toxicity. The highest prevalence of grade 2 toxicity occurred week 1 following treatment (20%), reducing to 3% by week 4. There was no difference in toxicity between those who received a boost versus not (p = 1.00). Delivering this schedule to 135 patients over six months saved 21,300 linac minutes and 1485 hospital visits compared to a 3-week schedule. Conclusion: Rapidly implementing ultrahypofractionated breast radiotherapy is feasible and acute toxicity rates are acceptable even when followed by boost.

Abstract: The IPED predictive pedestal code has been used to determine the critical gradients for the onset of 1) separatrix ballooning modes and 2) global peeling-ballooning modes as a function of plasma shaping. This results in a scaling of the onset threshold of separatrix ballooning modes as a function of elongation and triangularity $\alpha_\textrm{edge,crit} = 0.64\kappa^{2.2}(1+\delta)^{0.9}$ , while the critical gradient for global peeling-balloonig modes increases as ${\approx}\alpha_\textrm{edge,crit}^{1.5}$ . This implies that operational space for a ballooning unstable separatrix and stable peeling-ballooning modes exists at sufficiently high shaping. Applying a collisional broadnening based scaling of the separatrix gradients allows the critical separatrix density, required to drive the separatrix ballooning mode, to be derived from global plasma parameters for any operational scenario on any device. Evaluations for ASDEX Upgrade, JET, and the ITER 15 MA baseline plasma predict critical separatrix densities of 0.3–0.4 $ {n_\mathrm{GW}}$ for QCE access, making the QCE an attractive operational scenario for fusion devices.

Abstract: Experiments on ASDEX Upgrade (AUG) in 2021 and 2022 have addressed a number of critical issues for ITER and EU DEMO. A major objective of the AUG programme is to shed light on the underlying physics of confinement, stability, and plasma exhaust in order to allow reliable extrapolation of results obtained on present day machines to these reactor-grade devices. Concerning pedestal physics, the mitigation of edge localised modes (ELMs) using resonant magnetic perturbations (RMPs) was found to be consistent with a reduction of the linear peeling-ballooning stability threshold due to the helical deformation of the plasma. Conversely, ELM suppression by RMPs is ascribed to an increased pedestal transport that keeps the plasma away from this boundary. Candidates for this increased transport are locally enhanced turbulence and a locked magnetic island in the pedestal. The enhanced D-alpha (EDA) and quasi-continuous exhaust (QCE) regimes have been established as promising ELM-free scenarios. Here, the pressure gradient at the foot of the H-mode pedestal is reduced by a quasi-coherent mode, consistent with violation of the high-n ballooning mode stability limit there. This is suggestive that the EDA and QCE regimes have a common underlying physics origin. In the area of transport physics, full radius models for both L- and H-modes have been developed. These models predict energy confinement in AUG better than the commonly used global scaling laws, representing a large step towards the goal of predictive capability. A new momentum transport analysis framework has been developed that provides access to the intrinsic torque in the plasma core. In the field of exhaust, the X-Point Radiator (XPR), a cold and dense plasma region on closed flux surfaces close to the X-point, was described by an analytical model that provides an understanding of its formation as well as its stability, i.e., the conditions under which it transitions into a deleterious MARFE with the potential to result in a disruptive termination. With the XPR close to the divertor target, a new detached divertor concept, the compact radiative divertor, was developed. Here, the exhaust power is radiated before reaching the target, allowing close proximity of the X-point to the target. No limitations by the shallow field line angle due to the large flux expansion were observed, and sufficient compression of neutral density was demonstrated. With respect to the pumping of non-recycling impurities, the divertor enrichment was found to mainly depend on the ionisation energy of the impurity under consideration. In the area of MHD physics, analysis of the hot plasma core motion in sawtooth crashes showed good agreement with nonlinear 2-fluid simulations. This indicates that the fast reconnection observed in these events is adequately described including the pressure gradient and the electron inertia in the parallel Ohm’s law. Concerning disruption physics, a shattered pellet injection system was installed in collaboration with the ITER International Organisation. Thanks to the ability to vary the shard size distribution independently of the injection velocity, as well as its impurity admixture, it was possible to tailor the current quench rate, which is an important requirement for future large devices such as ITER. Progress was also made modelling the force reduction of VDEs induced by massive gas injection on AUG. The H-mode density limit was characterised in terms of safe operational space with a newly developed active feedback control method that allowed the stability boundary to be probed several times within a single discharge without inducing a disruptive termination. Regarding integrated operation scenarios, the role of density peaking in the confinement of the ITER baseline scenario (high plasma current) was clarified. The usual energy confinement scaling ITER98( p,y ) does not capture this effect, but the more recent H20 scaling does, highlighting again the importance of developing adequate physics based models. Advanced tokamak scenarios, aiming at large non-inductive current fraction due to non-standard profiles of the safety factor in combination with high normalised plasma pressure were studied with a focus on their access conditions. A method to guide the approach of the targeted safety factor profiles was developed, and the conditions for achieving good confinement were clarified. Based on this, two types of advanced scenarios (‘hybrid’ and ‘elevated’ q -profile) were established on AUG and characterised concerning their plasma performance.

Abstract: Within the 9th European Framework programme, since 2021 EUROfusion is operating five tokamaks under the auspices of a single Task Force called ‘Tokamak Exploitation’. The goal is to benefit from the complementary capabilities of each machine in a coordinated way and help in developing a scientific output scalable to future largre machines. The programme of this Task Force ensures that ASDEX Upgrade, MAST-U, TCV, WEST and JET (since 2022) work together to achieve the objectives of Missions 1 and 2 of the EUROfusion Roadmap: i) demonstrate plasma scenarios that increase the success margin of ITER and satisfy the requirements of DEMO and, ii) demonstrate an integrated approach that can handle the large power leaving ITER and DEMO plasmas. The Tokamak Exploitation task force has therefore organized experiments on these two missions with the goal to strengthen the physics and operational basis for the ITER baseline scenario and for exploiting the recent plasma exhaust enhancements in all four devices (PEX: Plasma EXhaust) for exploring the solution for handling heat and particle exhaust in ITER and develop the conceptual solutions for DEMO. The ITER Baseline scenario has been developed in a similar way in ASDEX Upgrade, TCV and JET. Key risks for ITER such as disruptions and run-aways have been also investigated in TCV, ASDEX Upgrade and JET. Experiments have explored successfully different divertor configurations (standard, super-X, snowflakes) in MAST-U and TCV and studied tungsten melting in WEST and ASDEX Upgrade. The input from the smaller devices to JET has also been proven successful to set-up novel control schemes on disruption avoidance and detachment.

Abstract: In 2021 JET exploited its unique capabilities to operate with T and D–T fuel with an ITER-like Be/W wall (JET-ILW). This second major JET D–T campaign (DTE2), after DTE1 in 1997, represented the culmination of a series of JET enhancements—new fusion diagnostics, new T injection capabilities, refurbishment of the T plant, increased auxiliary heating, in-vessel calibration of 14 MeV neutron yield monitors—as well as significant advances in plasma theory and modelling in the fusion community. DTE2 was complemented by a sequence of isotope physics campaigns encompassing operation in pure tritium at high T-NBI power. Carefully conducted for safe operation with tritium, the new T and D–T experiments used 1 kg of T (vs 100 g in DTE1), yielding the most fusion reactor relevant D–T plasmas to date and expanding our understanding of isotopes and D–T mixture physics. Furthermore, since the JET T and DTE2 campaigns occurred almost 25 years after the last major D–T tokamak experiment, it was also a strategic goal of the European fusion programme to refresh operational experience of a nuclear tokamak to prepare staff for ITER operation. The key physics results of the JET T and DTE2 experiments, carried out within the EUROfusion JET1 work package, are reported in this paper. Progress in the technological exploitation of JET D–T operations, development and validation of nuclear codes, neutronic tools and techniques for ITER operations carried out by EUROfusion (started within the Horizon 2020 Framework Programme and continuing under the Horizon Europe FP) are reported in (Litaudon et al Nucl. 6. Active control of Alfvén eigenmodes by external magnetic perturbations with different spatial spectra

Abstract: Alfvén eigenmodes have been suppressed and excited in tokamak plasmas by (just) modifying the poloidal spectra of externally applied static magnetic perturbations. This effect is observed experimentally when toroidal spectra of n = 2, n = 4 as well as a mixed spectrum of n = 2 and n = 4 is applied. Under the n = 2 magnetic perturbations, the modes are excited or suppressed by modifying the coil phasing between the upper and the lower set of coils. Regardless of the absolute rotation, an even parity for the n = 4 perturbation is observed to reduce the amplitude of the Alfvénic instabilities, while an odd parity amplifies it. To combine the stabilizing (and destabilizing) effect of n = 2 and n = 4, a mixed spectrum is applied, finding similar reduction (and amplification) trends. However, the impact on the mode amplitude is more subtle, due to the reduced coil current required for a mixed spectrum. The signal level on the fast-ion loss detector is sensitive to the applied poloidal spectrum, which is consistent with Hamiltonian full-orbit modelling of an edge resonant transport layer activated by the 3D perturbative fields. An internal redistribution of the fast-ion population is induced, modifying the phase-space gradients driving the Alfvénic instabilities, and ultimately determining their existence. The calculated edge resonant layers for both n = 2 and n = 4 toroidal spectra are consistent with the observed suppressed and excited phases. Moreover, hybrid kinetic-magnetohydrodynamic (MHD) simulations reveal that this edge resonant transport layer overlaps in phase-space with the population responsible for the fast-ion drive. The results presented here may help to control fast-ion driven Alfvénic instabilities in future burning plasmas with a significant fusion born alpha particle population.

Abstract: ASDEX Upgrade has developed multiple massive gas injection (MGI) scenarios to investigate runaway electron (RE) dynamics. During the current quench of the MGI induced disruptions, Alfvénic activity is observed in the 300–800 kHz range. With the help of a mode tracing algorithm based on Fourier spectrograms, mode behaviour was classified for 180 discharges. The modes have been identified as global Alfvén eigenmodes using linear gyrokinetic MHD simulations. Changes in the Alfvén continuum during the quench are proposed as explanation for the strong frequency sweep observed. A systematic statistical analysis shows no significant connection of the mode characteristics to the dynamics of the subsequent runaway electron beams. In our studies, the appearance and amplitude of the modes does not seem to affect the potential subsequent runaway beam. Beyond the scope of the 180 investigated dedicated RE experiments, the Alfvénic activity is also observed in natural disruptions with no RE beam forming.

8. Effect of symmetry-breaking on the MHD edge stability limit of tokamak plasmas

Abstract: We demonstrate for the first time that symmetry-breaking in tokamak plasmas reduces the window of stable edge operational space, leading to a lower achievable edge pressure gradient. Predictive simulations with the linear magnetohydrodynamic stability code CASTOR3D show a reduction of the critical pressure gradient by up to 30%, in agreement with experimental observations. The analysis has been extended to experimental plasmas considering, for the first time, ion diamagnetic drift effects in realistic non-axisymmetric (3D) tokamak geometry. The 3D geometry in plasmas with edge localised modes (ELMs) increases the growth rate of edge instabilities compared to the axisymmetric case. Further reducing the edge pressure gradient eliminates the instabilities corresponding to an experiment with suppressed ELMs, reproducing the empirically observed threshold for ELM suppression. Our findings highlight the importance of predictions of the ELM occurrence in full 3D geometry for future tokamak devices with intentionally and unintentionally broken axisymmetry.

Abstract: Recent results from MAST Upgrade are presented, emphasising understanding the capabilities of this new device and deepening understanding of key physics issues for the operation of ITER and the design of future fusion power plants. The impact of MHD instabilities on fast ion confinement have been studied, including the first observation of fast ion losses correlated with Compressional and Global Alfvén Eigenmodes. High-performance plasma scenarios have been developed by tailoring the early plasma current ramp phase to avoid internal reconnection events, resulting in a more monotonic q profile with low central shear. The impact of m / n = 3/2, 2/1 and 1/1 modes on thermal plasma confinement and rotation profiles has been quantified, and scenarios optimised to avoid them have transiently reached values of normalised beta approaching 4.2. In pedestal and ELM physics, a maximum pedestal top temperature of ∼350 eV has been achieved, exceeding the value achieved on MAST at similar heating power. Mitigation of type-I ELMs with n = 1 RMPs has been observed. Studies of plasma exhaust have concentrated on comparing conventional and Super-X divertor configurations, while X-point target, X-divertor and snowflake configurations have been developed and studied in parallel. In L-mode discharges, the separatrix density required to detach the outer divertors is approximately a factor 2 lower in the Super-X than the conventional configuration, in agreement with simulations. Detailed analysis of spectroscopy data from studies of the Super-X configuration reveal the importance of including plasma-molecule interactions and D _2 Fulcher band emission to properly quantify the rates of ionisation, plasma-molecule interactions and volumetric recombination processes governing divertor detachment. In H-mode with conventional and Super-X configurations, the outer divertors are attached in the former and detached in the latter with no impact on core or pedestal confinement.

10. Exploring the physics of a high-performance H-mode scenario with small ELMs at low collisionality in JET with Be/W wall

Abstract: A new H-mode regime at low density and low edge safety factor ( q _95 = 3.2, with $I_\mathrm{p}$ = 3 MA) that combines high energy confinement, stationary conditions for density and radiation and small Edge Localized Modes (ELMs) have been found in JET with Be/W wall. Such a regime is achieved by operating without external gas puffing, leading to a decrease in the edge density and a substantial increase in rotation and ion temperature in both the pedestal and the core region. Transport modelling shows a reduction of the turbulence, which starts in the pedestal region and extends into the plasma core, and outward impurity convection, consistent with the improved energy confinement and the lack of W accumulation observed in those conditions. In addition, large type I ELMs, typically found in gas-fuelled plasmas, are replaced by smaller and more frequent ELMs, whose appearance is correlated with a substantial reduction of the pedestal density and its gradient. Pedestals in this operating regime are stable to peeling–ballooning modes, consistent with the lack of large ELMs. This is in contrast to results in unfuelled JET-C plasmas that typically operated at higher pedestal densities and developed low frequency, large type I ELMs, thus pointing to the low density as one of the critical parameters for accessing this small ELMs regime in JET. This small ELMs regime exhibits the same low pedestal collisionality ( $\nu_{\mathrm{e},\mathrm{ped}}^*\sim0.1$ ) expected in ITER and operates at low q _95 , thus making it different from other small ELMs regimes that are typically obtained at higher q _95 and higher pedestal collisionality. These features make this newly developed H-mode regime in JET with Be/W wall a valuable tool for exploring the underlying transport, the different mechanisms of turbulence stabilization, as well as the physics associated with the appearance of small ELMs in high-temperature plasmas at ITER relevant pedestal collisionality.

Abstract: For its initial operational phase, ITER has until recently considered using non-nuclear hydrogen (H) or helium (He) plasmas to keep nuclear activation at low levels. To this end, the Tokamak Exploitation Task Force of the EUROfusion Consortium carried out dedicated experimental campaigns in He on the ASDEX Upgrade (AUG) and JET tokamaks in 2022, with particular emphasis put on the ELMy H-mode operation and plasma-wall interaction processes as well as comparison to H or deuterium (D) plasmas. Both in pure He and mixed He + H plasmas, H-mode operation could be reached but more effort was needed to obtain a stable plasma scenario than in H or D. Even if the power threshold for the LH transition was lower in He, entering the type-I ELMy regime appeared to require equally much or even more heating power than in H. Suppression of ELMs by resonant magnetic perturbations was studied on AUG but was only possible in plasmas with a He content below 19%; the reason for this unexpected behaviour remains still unclear and various theoretical approaches are being pursued to properly understand the physics behind ELM suppression. The erosion rates of tungsten (W) plasma-facing components were an order of magnitude larger than what has been reported in hydrogenic plasmas, which can be attributed to the prominent role of He ^2+ ions in the plasma. For the first time, the formation of nanoscale structures (W fuzz) was unambiguously demonstrated in H-mode He plasmas on AUG. However, no direct evidence of fuzz creation on JET was obtained despite the main conditions for its occurrence being met. The reason could be a delicate balance between W erosion by ELMs, competition between the growth and annealing of the fuzz, and coverage of the surface with co-deposits.

Abstract: Disruptions lead to a rapid loss of thermal and magnetic energy and can cause large heat loads, mechanical forces, and the excitation of a beam of relativistic runaway electrons. The operation of tokamaks at high energy and plasma current requires the use of a mitigation system to limit such detrimental effects. Mitigation techniques rely mainly on the injection of a large amount of impurities to radiate the majority of the thermal and magnetic energies. Heat loads and electro-magnetic (EM) forces as well as their toroidal asymmetries can be greatly reduced by such measures. In this paper, a theory is lined out to explain the reduction of the global vertical force based on large toroidal halo currents that keep the current centroid stationary in the midplane. As a consequence, the vertical current moment, which is linked to the EM-force, is reduced. The theory is backed up by experimental observations in shattered pellet injection mitigated vertical displacement event experiments in ASDEX Upgrade (AUG) and JET as well as by 2D simulations with the extended MHD code JOREK. Scans in the boundary heat flux are carried out to estimate the correct scrape-off layer temperature and the influence of the fraction of conducted energy. Finally, predictive simulations for ITER confirm the reduction of the vertical force by the injection of impurities.

13. Parameter dependencies of the separatrix density in low triangularity L-mode and H-mode JET-ILW plasmas

Abstract: The midplane electron separatrix density, n _e,sep , in JET-ILW L-mode and H-mode low triangularity deuterium fuelled plasmas exhibits a strong explicit dependence on the averaged outer divertor target electron temperature, n _e,sep ∼ T _e,ot ^−1/2 . This dependence is reproduced by analytic reversed two point model (rev-2PM), and arises from parallel pressure balance, as well as the ratio of the power and momentum volumetric loss factors, (1 − f _cooling )/(1 − f _mom-loss ). Quantifying the influence of the (1 − f _cooling ) and (1 − f _mom-loss ) loss factors on n _e,sep has been enabled by measurement estimates of these quantities from L-mode density (fueling) ramps in the outer horizontal, VH(C), and vertical target, VV, divertor configurations. Rev-2PM n _e,sep estimates from the extended H-mode and more limited L-mode datasets are recovered to within ±25% of the measurements, with a scaling factor applied to account for use of $\left\langle {{T_{{\text{e,ot}}}}} \right\rangle $ , an averaged quantity, rather than flux tube resolved target values. Both the (1 − f _cooling ) and (1 − f _mom-loss ) trends and recovery of n _e,sep using the rev-2PM formatting are reproduced in EDGE2D-EIRENE L-mode-like and H-mode-like density scan simulations. The general lack of a divertor configuration effect in the JET-ILW n _e,sep trends can be attributed to a significant influence of main chamber recycling, which has been shown in the EDGE2D-EIRENE results to moderate n _e,sep with respect to changes in divertor neutral leakage imposed by changes in the divertor configuration. The unified n _e,sep vs $\left\langle {{T_{{\text{e,ot}}}}} \right\rangle $ trends can, however, be broken if large modifications to the divertor geometry (e.g. complete removal of the outer divertor baffle structure) are introduced in the model. The more pronounced high-field side high density region formation in the VH(C) configuration with reduced clearance to the separatrix does not appear to have a significant influence on the outer midplane separatrix and pedestal parameters when mapped to $\left\langle {{T_{{\text{e,ot}}}}} \right\rangle $ , although conditions at the inner midplane could not be assessed.

Abstract: We study localized edge turbulence in the ASDEX Upgrade tokamak that appears if resonant magnetic perturbations (RMP) are applied to suppress edge localized modes (ELMs) in the high confinement mode. The concomitant density fluctuations are detected by microwave reflectometry at the outboard midplane. Two modes can be distinguished, (a) a broadband fluctuation below a threshold of the RMP field amplitude, and (b) a narrow-band quasi-coherent mode (QCM) above the threshold. The broadband fluctuation is toroidally spread out but disappears at the toroidal position of maximum E × B shear in the gradient region. Temporal and spatial correlation along field lines of the midplane density fluctuation and the divertor particle flux suggests that this mode is producing significant particle transport across the gradient region and into the divertor, hence contributing to the plasma density reduction that is often observed when applying RMP fields (the so-called ‘pump-out’ effect). The QCM is also toroidally localized, its radial extent grows with increasing RMP field amplitude, and leads to further increased divertor particle flux compared to the broadband mode. Our observations suggest that both modes not only play an important role in keeping the plasma density stationary in the absence of ELMs but also to reduce the plasma pressure such that the plasma edge becomes stable against ELMs.

Abstract: The quasi-continuous exhaust (QCE) regime, formerly known as either type-II ELM or small ELM regime is studied in ASDEX Upgrade. The regime is a natural type-I ELM-free H-mode. The operational space of QCE discharges in ASDEX Upgrade with respect to their separatrix conditions and their power exhaust capabilities are presented. A significant broadening of the power fall-off length is observed, correlating to an increased separatrix density and pressure. Moreover, the possible reactor relevance of this regime is demonstrated by expanding the operational space to low edge safety factor and demonstrating the benign tungsten impurity behaviour. A discharge without any type-I ELM from start to end reaching a partially detached divertor at high normalised energy confinement time is presented.

16. 2D and 3D studies of the X-divertor configuration in the future upper divertor of ASDEX upgrade

Abstract: ASDEX Upgrade is planning the installation of a pair of in-vessel coils in order to study alternative divertor configurations, like an X-divertor (XD) or a Snowflake (SF) in a machine with a high heating power compared to its size. Two designs for the technical realization of these coils are currently proposed, one including z-shaped joints and one with a single spiral conductor. The magnetic fields produced by these conductors intrinsically have a non-axisymmetric component. In particular in the XD configuration, where very shallow field line incidence angles occur, these 3D fields have a strong impact on the power distribution on the outer divertor plate. The 2D and 3D plasma transport is studied by the SOLPS and initial EMC3-EIRENE code simulations, respectively, in order to support the decision making processes of the engineering design. Although several aspects still require further attention, this paper gives a status report. The strong toroidal asymmetries observed in the power deposition pattern for the spiral conductor seem to be strongly reduced, when highly dissipative divertor conditions are reached. The asymmetry for the z-joint design is substantially smaller than the spiral conductor even for low-dissipative conditions. 17. On the ion and electron temperature recovery after the ELM-crash at ASDEX upgrade

Abstract: The access to fast measurements, i.e. Δt ≈ 100 µs, of the ions and the electrons during an entire edge localized cycle (ELM) reveals asymmetries in the recovery of the maximum edge gradients. Different magnetic fluctuations are found to correlate with the saturation of the edge ion temperature (Ti), electrons temperature (Te) and density (ne) gradients. In particular, while ∇Ti and ∇ne clamp roughly 3.0 ms after the ELM-crash together with the onset of mid-frequency (f ≲ 50 kHz) magnetic fluctuations, ∇Te recovers to the pre-ELM conditions only after 7.0 ms and saturates with the appearance of high frequency fluctuations (f ≈ 200 kHz). The effect of electron temperature gradient modes (ETGs) and of energy losses induced by ionization of neutrals are discussed as possible reasons for the delayed recovery of ∇Te. The onset and the suppression of ETGs qualitatively follow the requirements of an increased electron heat transport. However, gyro-kinetic simulations are necessary to quantify the impact of ETGs. On the other hand, the impact of the neutral ionization during the density build-up as an electron energy loss channel is measured to be small compared to the total electron energy. 18. Impact of the new TCV baffled divertor upgrade on pedestal structure and performance

Abstract: A new set of carbon tiles, neutral beam heating optics and gas baffles were installed on TCV during the baffled divertor upgrade in early 2019. The installation of the baffles allows a deconvolution of the roles of main chamber and divertor neutral pressure on the H-mode pedestal structure. This physical barrier allows relatively high neutral pressures to be constrained to the divertor, thus preventing neutrals from entering the main chamber and potentially degrading core confinement. This study presents the experimentally measured and modelled pedestal heights and structure for a series of H-mode discharges prior to and after this upgrade.Increased pedestal performance at high divertor neutral pressure was observed after the baffled divertor upgrade. This was consistent across all triangularities and outer target locations investigated and is attributed to higher pedestal top temperatures being maintained at high gas injection rates. ASTRA simulations indicated beam heating power coupled to the plasma did not significantly vary after the baffled divertor upgrade or as a function of divertor neutral gas pressure. Analysis of the pedestal structure exposed a strong correlation between pedestal performance and the density pedestal position prior to and after the baffled divertor upgrade. The baffled divertor upgrade limited the outward shift of the density pedestal, thus maintaining higher pedestal performance at high divertor neutral pressures. Stability analysis indicated the majority of discharges studied were within 25% of the stability boundary. No correlation was found between the distance from the stability boundary and pedestal performance or structure. Comparison with the EPED1 model indicated that TCV discharges do not have a fixed dependence between pedestal βθand pedestal width. A large variation in the EPED1 relating parameter was observed and found to vary with the density pedestal position.

Abstract: An overview of results of negative triangularity (NT) studies from the ASDEX Upgrade tokamak (AUG) is given. Moderate values of the triangularity in the range of δ _average ≈ −0.2 have been obtained. Results from both unfavorable and favorable magnetic configuration in terms of high-confinement mode (H-mode) access are presented. In unfavorable configuration, the ion grad B drift points away from the active x-point, which usually results in a factor of 2 higher power thresholds to enter H-mode. In unfavorable configuration, low-confinement mode (L-mode) operation is confirmed at high auxiliary heating input powers $ > 10$ MW at low collisionality. Under these conditions, plasmas with positive triangularity (PT) are usually in H-mode. Highest plasma energies have been obtained using a combination of neutral beam injection, electron- and ion cyclotron resonance heating, where values of the plasma beta of β _N ≈ 1.7 have been reached in L-mode. While discharges with mixed auxiliary heating power input show moderate confinement on L-mode levels, plasmas heated with electron cyclotron resonance heating show H-mode like confinement. Operation in favorable configuration, however, leads to H-mode with type-I edge localized modes (ELMs). The power required for the L–H transition is comparable to the typical L–H power threshold in PT cases ( $< 2.6$ MW), and good confinement of typical PT H-modes is obtained. The ELM frequency is demonstrated to reach 700 Hz. These results show a clear effect of the NT configuration on the ELM behavior in H-modes, and that on AUG, unfavorable configuration is an essential ingredient if H-mode operation is to be avoided in NT plasmas. So far, in all AUG NT plasmas, power degradation is observed. Initial gyrokinetic simulations of the edge plasma in unfavorable configuration reveal the ion-temperature gradient mode as the fastest growing mode with a subdominant trapped-electron-mode.

Abstract: Magnetic skyrmions are topological spin textures that exhibit classical or quantum quasiparticle behavior. A substantial amount of research has occurred in this field, both because of their unique electromagnetic properties and potential application for future nonvolatile memory storage applications, as well as fundamental questions on their topology and unique magnetic phases. Here, we investigate the fluctuation properties of a magnetic Fe/Gd skyrmion lattice, using short-pulsed x rays. We first measure spontaneous fluctuations of the skyrmion lattice phase and find an inherent, collective mode showing an underdamped oscillation with a relaxation of a couple of nanoseconds. Further observations track the response towards the continuous phase transition and a “critical-like” slowing down of fluctuations is observed well before the critical point. These results suggest that the skyrmion lattice phase never fully freezes into a static crystal. This constant state of fluctuation indicates that the physics of topological magnetic phases may have more in common with high-temperature superconductors with disorder.

21. The high field side high density region in SOLPS-modeling of nitrogen-seeded H-modes in ASDEX Upgrade

Abstract: The understanding of divertor physics and the evolution of divertor detachment is crucial for developing the capability to model power exhaust in current experiments and reliably predict it for future fusion devices. In simulations of ASDEX Upgrade, an experimentally observed region of high density in the high field side scrape-off layer has been recovered. Validated modeling with SOLPS5.0 shows that a detailed match of the high field side scrape-off layer plasma is not only important for local plasma parameters, but can lead to strong changes in global parameters. Drifts play a crucial role in lower-single null discharges with forward toroidal field (∇B→-drift pointing down). Their inclusion changes the spatial extent as well as the radial and poloidal gradients of the high field side high density. Adapted transport coefficients that take into account core fueling by plasma diffusion due to the presence of the high field side density and drift-driven radial fluxes now reconcile the modeled deuterium compression ratio, divertor neutral density, neutral radiation levels and deuterium fueling rates with experimental measurements. The onset of strong volume recombination in the simulations now allows to remove the previously necessary increase of perpendicular transport in the inner divertor from the simulations. The possibility of operating the standard lower single-null (SN) and double-null (DN) would be preserved. Potential effects to reduce the peak parallel- and/or perpendicular heat flux are predicted from a simple geometrical-diffusive model as well as by numerical EMC3-EIRENE simulations for pure deuterium attached conditions with spatially constant diffusion coefficients. Abstract: During June-October 2022, the SARS-CoV-2 Omicron BA.5 sublineage accounted for most of the sequenced viral genomes in the United States, with further Omicron sublineage diversification through November 2022.* Bivalent mRNA vaccines contain an ancestral SARS-CoV-2 strain component plus an updated component of the Omicron BA.4/BA.5 sublineages. On September 1, 2022, a single bivalent booster dose was recommended for adults who had completed a primary vaccination series (with or without subsequent booster doses), with the last dose administered ≥2 months earlier (1). During September 13-November 18, the VISION Network evaluated vaccine effectiveness (VE) of a bivalent mRNA booster dose (after 2, 3, or 4 monovalent doses) compared with 1) no previous vaccination and 2) previous receipt of 2, 3, or 4 monovalent-only mRNA vaccine doses, among immunocompetent adults aged ≥18 years with an emergency department/urgent care (ED/UC) encounter or hospitalization for a COVID-19-like illness.

Abstract: Over the last 50 years, access to new data and analytical tools has expanded the study of analytical paleobiology, contributing to innovative analyses of biodiversity dynamics over Earth's history. Despite—or even spurred by—this growing availability of resources, analytical paleobiology faces deep-rooted obstacles that stem from the need for more equitable access to data and best practices to guide analyses of the fossil record. Recent progress has been accelerated by a collective push toward more collaborative, interdisciplinary, and open science, especially by early-career researchers. Here, we survey four challenges facing analytical paleobiology from an early-career perspective: (1) accounting for biases when interpreting the fossil record; (2) integrating fossil and modern biodiversity data; (3) building data science skills; and (4) increasing data accessibility and equity. We discuss recent efforts to address each challenge, highlight persisting barriers, and identify tools that have advanced analytical work. Given the inherent linkages between these challenges, we encourage discourse across disciplines to find common solutions. We also affirm the need for systemic changes that reevaluate how we conduct and share paleobiological research.

29. Structural maturation of SYCP1-mediated meiotic chromosome synapsis by SYCE3

Abstract: In meiosis, a supramolecular protein structure, the synaptonemal complex (SC), assembles between homologous chromosomes to facilitate their recombination. Mammalian SC formation is thought to involve hierarchical zipper-like assembly of an SYCP1 protein lattice that recruits stabilizing central element (CE) proteins as it extends. Here we combine biochemical approaches with separation-of-function mutagenesis in mice to show that, rather than stabilizing the SYCP1 lattice, the CE protein SYCE3 actively remodels this structure during synapsis. We find that SYCP1 tetramers undergo conformational change into 2:1 heterotrimers on SYCE3 binding, removing their assembly interfaces and disrupting the SYCP1 lattice. SYCE3 then establishes a new lattice by its self-assembly mimicking the role of the disrupted interface in tethering together SYCP1 dimers. SYCE3 also interacts with CE complexes SYCE1–SIX6OS1 and SYCE2–TEX12, providing a mechanism for their recruitment. Thus, SYCE3 remodels the SYCP1 lattice into a CE-binding integrated SYCP1–SYCE3 lattice to achieve long-range synapsis by a mature SC.

30. The Return of Fossils Removed Under Colonial Rule

Abstract: Debates on the restitution of colonial loot usually focus on art, antiquities, religious artefacts, and similar objects. Many fossils of considerable scientific and cultural value were also removed under colonial rule, yet they rarely feature in these discussions despite being classified as cultural objects. This article seeks to shed light on the colonial removal of fossils and explore potential avenues for their return under public international law. Instead of focusing on the (il-)legality of colonial takings, we argue that the right to access culture has developed from the right to participate in cultural life in Article 15(1)(a) of the International Covenant on Economic, Social and Cultural Rights (ICESCR), which provides, if not a solid legal basis, a valuable set of arguments for former colonies requesting the return of fossils looted from their countries/territories of origin. The case study of the negotiations on the return of the Broken Hill skull before the UNESCO Intergovernmental Committee for Promoting the Return of Cultural Property to its Countries of Origin or its Restitution in Case of Illicit Appropriation (ICPRCP) highlights the potential of this mechanism of dispute resolution with respect to fossils.

31. Publication pressure threatens the integrity of palaeontological research

Abstract: Publications are the de facto currency of academia. Academics, palaeontologists included, are judged by publication metrics, which usually include the impact factor of the journal in which they publish, the number of publications, and the number of citations. However, in the race to publish in high-impact journals and the pressure to increase research productivity, sometimes corners are cut, leading to an increase in scientific and other types of misconduct. In this paper, we demonstrate how ethical, and even legal, transgressions within palaeontology, particularly regarding the provenance of fossil specimens, are inherently related to the pressure faced by academic researchers to publish “novel” studies in high-impact journals. We note how papers in high-impact journals follow a consistent theme of either showcasing novel evidence or methods, or describing charismatic or unusual specimens, often dinosaur-related. We examine notable recent examples of these papers that have been linked to ethical and legal problems, which have ironically been brought to light by virtue of the paper being widely publicised as highly “impactful”. Finally, we discuss the importance of developing an ethical framework for scientific publishing, which currently does not exist; only a handful of professional palaeontological bodies, including societies and journals, have independently developed policies to combat these issues. As long as the culture of “publish or perish” lingers in palaeontology, there will be no incentive for more ethical research that better serves both science and society to prevail.

Abstract: We investigated invasive group A Streptococcus epidemiology in Idaho, USA, during 2008-2019 using surveillance data, medical record review, and emm (M protein gene) typing results. Incidence increased from 1.04 to 4.76 cases/100,000 persons during 2008-2019. emm 1, 12, 28, 11, and 4 were the most common types, and 2 outbreaks were identified. We examined changes in distribution of clinical syndrome, patient demographics, and risk factors by comparing 2008-2013 baseline with 2014-2019 data. Incidence was higher among all age groups during 2014-2019. Streptococcal toxic shock syndrome increased from 0% to 6.4% of cases (p = 0.02). We identified no differences in distribution of demographic or risk factors between periods. Results indicated that invasive group A Streptococcus is increasing among the general population of Idaho. Ongoing surveillance of state-level invasive group A Streptococcus cases could help identify outbreaks, track regional trends in incidence, and monitor circulating emm types.

Abstract: Within the broader contexts of language maintenance, revitalisation and use, schools have an important role to play in the early transmission of minority or minoritized languages. Through effective classroom-based practices, teachers can offer rich, continuous linguistic experiences for the young people they teach that can lead to long-term, sustained engagement with language over time. How effective a given approach or practice may be in delivering a linguistic outcome in different contexts depends to a large extent on the skills and competence of the teacher involved (Fitzpatrick et al., 2018, p. 59), and the skills and competence of the teacher are usually rooted in their early experiences as student teachers. Initial teacher education (ITE) programmes are therefore central to the delivery of effective classroom-based practices and fundamental in generating autonomous teachers who can explore critically the different pedagogical approaches as they pertain to minority language contexts. This paper provides a comparative overview of the current issues and challenges facing ITE for primary education in two distinct minority language settings, namely Wales and Ireland. Both settings provide Welsh-medium or Irish-medium immersion education, as well as English-medium education offering Welsh or Irish as statutory subjects up to age 16. Both contexts also offer ITE programmes through the medium of Welsh or Irish, catering specifically for those student teachers wishing to teach in immersion settings. Both contexts face a number of shared challenges in terms of the content, design and delivery of an effective ITE programme for the context. We therefore highlight areas in which resources and methodologies could be shared amongst all those involved in ITE where minority language maintenance is a key component of education.

Abstract: The Omicron variant (B.1.1.529) of SARS-CoV-2, the virus that causes COVID-19, was first identified in the United States in November 2021, with the BA.1 sublineage (including BA.1.1) causing the largest surge in COVID-19 cases to date. 35. Effects of pneumococcal conjugate vaccines on reducing the risk of respiratory disease associated with coronavirus infection

Abstract: Pneumococcal conjugate vaccines (PCVs) provide protection against vaccine-type pneumococcal disease in both children and adults. Growing evidence suggests that PCVs also reduce pneumonia and lower respiratory tract infections (LRTIs) more broadly, including protecting against viral-associated respiratory diseases. In this short narrative review, we highlight clinical studies investigating whether PCVs might have a role in reducing coronavirus disease, both those caused by endemic human coronaviruses (HCoVs) and severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2). These studies include two randomized controlled trials assessing HCoV-associated pneumonia, one each in children and older adults, and two observational studies of PCV13 effectiveness against HCoV-associated LRTI and COVID-19 in adults. We discuss possible mechanisms for PCV protection including preventing viral pneumococcal co-infections and the possibility that pneumococci in the upper respiratory tract might modify the host immune response to SARS-CoV-2. Lastly, we identify knowledge gaps and further questions on the potential role of PCVs during the COVID-19 pandemic.

36. Therapeutic immunization with a whole cell vaccine reduces pneumococcal nasopharyngeal density, shedding, and middle ear infection in mice

Abstract: Pneumococcal Conjugate Vaccines (PCVs) have substantially reduced the burden of disease caused byStreptococcus pneumoniae(the pneumococcus). However, protection is limited to vaccine serotypes, and when administered to children who are colonized with pneumococci at the time of vaccination, immune responses to the vaccine are blunted. Here, we investigate the potential of a killed whole cell pneumococcal vaccine (WCV) to reduce existing pneumococcal carriage and mucosal disease when given therapeutically to infant mice colonized with pneumococci. We show that a single dose of WCV reduced pneumococcal carriage density in an antibody-dependent manner. Therapeutic vaccination induced robust immune responses to pneumococcal surface antigens CbpA, PspA (family 1) and PiaA. In a co-infection model of otitis media, a single dose of WCV reduced pneumococcal middle ear infection. Lastly, in a two-dose model, therapeutic administration of WCV reduced nasal shedding of pneumococci. Taken together, our data demonstrate that WCV administered in colonized mice reduced pneumococcal density in the nasopharynx and the middle ear, and decreased shedding. A vaccine with similar properties in children would be beneficial in low and middle-income settings where pneumococcal carriage is high.ImportanceAlthough typically asymptomatic, pneumococcal carriage plays an essential role in transmission and the development of disease. Pneumococcal Conjugate Vaccines (PCVs) have reduced the burden of pneumococcal disease worldwide. However, their use has increased carriage and disease caused by non-vaccine serotypes, prompting investigations into serotype-independent pneumococcal vaccines. An additional limitation of PCVs is immune hypo-responsiveness to vaccines in children carrying pneumococci at the time of vaccination. Therefore, there is great interest in next generation vaccines such as whole cell vaccines. In this study we investigate a pneumococcal whole cell vaccine (WCV) for it effect on carriage in mice that are already colonized at the time of vaccination. We show that this ‘therapeutic’ vaccination of mice can reduce pneumococcal carriage density, shedding and infection of the middle ear. Our study suggests that WCV could be beneficial in high burden settings where carriage at the time of vaccination is more common.

Abstract: Due to advancements in systemic targeted and immunotherapies resulting in improved disease control and overall survival, and the increasing use of computed tomography and spine magnetic resonance imaging surveillance, the number of patients presenting with both asymptomatic and symptomatic spinal metastases is increasing. The need for versatile tumour ablative local management strategies, beyond the limits afforded by conventional palliative external beam radiation therapy (cEBRT), is increasingly more important. Stereotactic body radiation therapy (SBRT) was developed to meet such a need. This highly conformal technique allows the delivery of high biologically effective doses of radiation to the vertebral target, while controlling the differential dose exposure to the adjacent critical neural tissue. Identifying patients with painful spine metastases who would gain the most benefit from this important therapeutic option can be challenging. Here we summarise the randomised evidence specific to spine SBRT, comparing cEBRT with SBRT for pain control in patients with spine metastases in the palliative setting to better understand the role of spine SBRT in modern oncological spinal care.

Abstract: Background Following historically low influenza activity during the 2020–2021 season, the United States saw an increase in influenza circulating during the 2021–2022 season. Most viruses belonged to the influenza A(H3N2) 3C.2a1b 2a.2 subclade. Methods We conducted a test-negative case-control analysis among adults ≥18 years of age at 3 sites within the VISION Network. Encounters included emergency department/urgent care (ED/UC) visits or hospitalizations with ≥1 acute respiratory illness (ARI) discharge diagnosis codes and molecular testing for influenza. Vaccine effectiveness (VE) was calculated by comparing the odds of influenza vaccination ≥14 days before the encounter date between influenza-positive cases (type A) and influenza-negative and severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2)–negative controls, applying inverse probability-to-be-vaccinated weights, and adjusting for confounders. Results In total, 86 732 ED/UC ARI-associated encounters (7696 [9%] cases) and 16 805 hospitalized ARI-associated encounters (649 [4%] cases) were included. VE against influenza-associated ED/UC encounters was 25% (95% confidence interval (CI), 20%–29%) and 25% (95% CI, 11%–37%) against influenza-associated hospitalizations. VE against ED/UC encounters was lower in adults ≥65 years of age (7%; 95% CI, −5% to 17%) or with immunocompromising conditions (4%; 95% CI, −45% to 36%). Conclusions During an influenza A(H3N2)-predominant influenza season, modest VE was observed. These findings highlight the need for improved vaccines, particularly for A(H3N2) viruses that are historically associated with lower VE.

Abstract: An overview of results of negative triangularity (NT) studies from the ASDEX Upgrade tokamak (AUG) is given. Moderate values of the triangularity in the range of δ average ≈ −0.2 have been obtained. Results from both unfavorable and favorable magnetic configuration in terms of high-confinement mode (H-mode) access are presented. In unfavorable configuration, the ion grad B drift points away from the active x-point, which usually results in a factor of 2 higher power thresholds to enter H-mode. In unfavorable configuration, low-confinement mode (L-mode) operation is confirmed at high auxiliary heating input powers 10$?> > 10 MW at low collisionality. Under these conditions, plasmas with positive triangularity (PT) are usually in H-mode. Highest plasma energies have been obtained using a combination of neutral beam injection, electron- and ion cyclotron resonance heating, where values of the plasma beta of β N ≈ 1.7 have been reached in L-mode. While discharges with mixed auxiliary heating power input show moderate confinement on L-mode levels, plasmas heated with electron cyclotron resonance heating show H-mode like confinement. Operation in favorable configuration, however, leads to H-mode with type-I edge localized modes (ELMs). The power required for the L–H transition is comparable to the typical L–H power threshold in PT cases ( < 2.6 MW), and good confinement of typical PT H-modes is obtained. The ELM frequency is demonstrated to reach 700 Hz. These results show a clear effect of the NT configuration on the ELM behavior in H-modes, and that on AUG, unfavorable configuration is an essential ingredient if H-mode operation is to be avoided in NT plasmas. So far, in all AUG NT plasmas, power degradation is observed. Initial gyrokinetic simulations of the edge plasma in unfavorable configuration reveal the ion-temperature gradient mode as the fastest growing mode with a subdominant trapped-electron-mode.

Abstract: Background Community-acquired pneumonia is an important cause of morbidity and mortality in adults. Approximately one-third of pneumonia cases can be attributed to the pneumococcus. Pneumococcal conjugate vaccines (PCVs) protect against colonisation with vaccine-type serotypes. The resulting decrease in transmission of vaccine serotypes leads to large indirect effects. There are limited data from developing countries demonstrating the impact of childhood PCV immunisation on adult pneumonia. There are also insufficient data available on the burden and severity of all-cause pneumonia and respiratory syncytial virus (RSV) in adults from low resource countries. There is currently no recommendation for adult pneumococcal vaccination with either pneumococcal polysaccharide vaccine or PCVs in Mongolia. We describe the protocol developed to evaluate the association between childhood 13-valent PCV (PCV13) vaccination and trends in adult pneumonia. Methods PCV13 was introduced into the routine childhood immunisation schedule in Mongolia in a phased manner from 2016. In March 2019 we initiated active hospital-based surveillance for adult pneumonia, with the primary objective of evaluating trends in severe hospitalised clinical pneumonia incidence in adults 18 years and older in four districts of Ulaanbaatar. Secondary objectives include measuring the association between PCV13 introduction and trends in all clinically-defined pneumonia, radiologically-confirmed pneumonia, nasopharyngeal carriage of S. pneumoniae and pneumonia associated with RSV or influenza. Clinical questionnaires, nasopharyngeal swabs, urine samples and chest radiographs were collected from enrolled patients. Retrospective administrative and clinical data were collected for all respiratory disease-related admissions from January 2015 to February 2019. Discussion Establishing a robust adult surveillance system may be an important component of monitoring the indirect impact of PCVs within a country. Monitoring indirect impact of childhood PCV13 vaccination on adult pneumonia provides additional data on the full public health impact of the vaccine, which has implications for vaccine efficiency and cost-effectiveness. Adult surveillance in Mongolia will contribute to the limited evidence available on the burden of pneumococcal pneumonia among adults in low- and middle-income countries, particularly in the Asia-Pacific region. In addition, it is one of the few examples of implementing prospective, population-based pneumonia surveillance to evaluate the indirect impact of PCVs in a resource-limited setting.

We compared vaccine effectiveness (VE) against medically attended COVID-19 of 2-3 mRNA and 1-2 viral-vector vaccine doses between IC and non-IC adults.MethodsUsing a test-negative design among eight VISION Network sites, VE against laboratory-confirmed COVID-19–associated emergency department (ED) or urgent care (UC) events and hospitalizations from 26 August-25 December 2021 was estimated separately among IC and non-IC adults and among specific IC condition subgroups. Vaccination status was defined using number and timing of doses. VE for each status (versus unvaccinated) was adjusted for age, geography, time, prior positive test result, and local SARS-CoV-2 circulation.ResultsWe analyzed 8,848 ED/UC events and 18,843 hospitalizations among IC patients and 200,071 ED/UC events and 70,882 hospitalizations among non-IC patients. Among IC patients, 3-dose mRNA VE against ED/UC (73% [95% CI: 64-80]) and hospitalization (81% [95% CI: 76-86]) was lower than that among non-IC patients (ED/UC: 94% [95% CI: 93-94]; hospitalization: 96% [95% CI: 95-97]). Similar patterns were observed for viral-vector vaccines. Transplant recipients had lower VE than other IC subgroups.ConclusionsDuring B.1.617.2 (Delta) variant predominance, IC adults received moderate protection against COVID-19–associated medical events from three mRNA doses, or one viral-vector dose plus a second dose of any product. However, protection was lower in IC versus non-IC patients, especially among transplant recipients, underscoring the need for additional protection among IC adults.Key pointsDuring Delta variant predominance, immunocompromised (IC) adults received moderate protection against COVID-19-associated medical events from three mRNA doses, but IC patients, especially transplant recipients, were less protected than non-IC patients, underscoring the need for additional protection beyond the primary series.

Abstract: Persons with moderate-to-severe immunocompromising conditions might have reduced protection after COVID-19 vaccination, compared with persons without immunocompromising conditions (1-3). On August 13, 2021, the Advisory Committee on Immunization Practices (ACIP) recommended that adults with immunocompromising conditions receive an expanded primary series of 3 doses of an mRNA COVID-19 vaccine. ACIP followed with recommendations on September 23, 2021, for a fourth (booster) dose and on September 1, 2022, for a new bivalent mRNA COVID-19 vaccine booster dose, containing components of the BA.4 and BA.5 sublineages of the Omicron (B.1.1.529) variant (4). 48. Scleromochlus and the early evolution of Pterosauromorpha

Abstract: Pterosaurs, the first vertebrates to evolve powered flight, were key components of Mesozoic terrestrial ecosystems from their sudden appearance in the Late Triassic until their demise at the end of the Cretaceous1–6. However, the origin and early evolution of pterosaurs are poorly understood owing to a substantial stratigraphic and morphological gap between these reptiles and their closest relatives6, Lagerpetidae7. Scleromochlus taylori, a tiny reptile from the early Late Triassic of Scotland discovered over a century ago, was hypothesized to be a key taxon closely related to pterosaurs8, but its poor preservation has limited previous studies and resulted in controversy over its phylogenetic position, with some even doubting its identification as an archosaur9. Here we use microcomputed tomographic scans to provide the first accurate whole-skeletal reconstruction and a revised diagnosis of Scleromochlus, revealing new anatomical details that conclusively identify it as a close pterosaur relative1 within Pterosauromorpha (the lagerpetid + pterosaur clade). Scleromochlus is anatomically more similar to lagerpetids than to pterosaurs and retains numerous features that were probably present in very early diverging members of Avemetatarsalia (bird-line archosaurs). These results support the hypothesis that the first flying reptiles evolved from tiny, probably facultatively bipedal, cursorial ancestors1.

Abstract: ImportanceRecent sublineages of the SARS-CoV-2 Omicron variant, including BA.4 and BA.5, may be associated with greater immune evasion and less protection against COVID-19 following vaccination.ObjectiveTo evaluate the association between COVID-19 mRNA vaccination with 2, 3, or 4 doses among immunocompetent adults and the risk of medically attended COVID-19 illness during a period of BA.4/BA.5 predominant circulation; to evaluate the relative severity of COVID-19 in hospitalized cases across Omicron BA.1, BA.2/BA.2.12.1, and BA.4/BA.5 sublineage periods.Setting, Design and ParticipantsTest-negative study of adults with COVID-19-like illness (CLI) and molecular testing for SARS-CoV-2 conducted in 10 states from December 16, 2021, to August 20, 2022.ExposuremRNA COVID-19 vaccination.Main Outcomes and MeasuresEmergency department/urgent care encounters, hospitalizations, and admission to the intensive care unit (ICU) or in-hospital death. The adjusted odds ratio (OR) for the association between prior vaccination and medically attended COVID-19 was used to estimate VE, stratified by care setting and vaccine doses (2, 3, or 4 doses vs 0 doses as reference group). Among hospitalized case-patients, demographic and clinical characteristics and in-hospital outcomes including ICU admission and death were compared across sublineage periods.ResultsBetween June 19 – August 20, 2022, 82,229 ED/UC and 21,007 hospital encounters were included for the BA.4/BA.5 vaccine effectiveness analysis. Among adults hospitalized with CLI, the adjusted odds ratio (OR) was 0.75 (95% CI: 0.68-0.83) for receipt of 2 vaccine doses at ≥150 days after receipt, 0.32 (95% CI: 0.20-0.50) for a third dose 7-119 days after receipt, and 0.64 (95% CI: 0.58-0.71) for a third dose ≥120 days (median 235 days) after receipt for cases vs controls. For COVID-19-associated hospitalization, among patients ages ≥65 years 7-59 and ≥60 days (median 88 days) after a fourth dose, ORs were 0.34 (95% CI: 0.25-0.47) and 0.43 (95% CI: 0.34-0.56), respectively. Among hospitalized cases, ICU admission and/or in-hospital death occurred in 21.4% during the BA.1 vs 14.7% during the BA.4/BA.5 period (standardized mean difference: 0.17).ConclusionVE against medically attended COVID-19 illness decreased over time since last dose; receipt of one or two booster doses increased effectiveness over a primary series alone.KEY POINTSQuestionWhat is the association between receipt of first-generation COVID-19 mRNA vaccines and medically attended COVID-19 during Omicron BA.4/BA.5 sublineage predominance?FindingsThis test-negative analysis included 82,229 emergency department or urgent care encounters and 21,007 hospitalizations for COVID-19-like illness. 50. The early diversification of ray-finned fishes (Actinopterygii): hypotheses, challenges and future prospects

Abstract: Actinopterygii makes up half of living vertebrate diversity, and study of fossil members during their Palaeozoic rise to dominance has a long history of descriptive work. Although research interest into Palaeozoic actinopterygians has increased in recent years, broader patterns of diversity and diversity dynamics remain critically understudied. Past studies have investigated macroevolutionary trends in Palaeozoic actinopterygians in a piecemeal fashion, variably using existing compendia of vertebrates or literature-based searches. Here, we present a comprehensive occurrence-based dataset of actinopterygians spanning the whole of the Palaeozoic. We use this to produce the first through-Palaeozoic trends in genus and species counts for Actinopterygii. Diversity through time generally tracks metrics for sampling, while major taxonomic problems pervading the Palaeozoic actinopterygian record obscure diversity trends. Many described species are concentrated in several particularly problematic 'waste-basket' genera, hiding considerable morphological and taxonomic diversity. This taxonomic confusion also feeds into a limited understanding of phylogenetic relationships. A heavy sampling bias towards Europe and North America exists in both occurrence databases and available phylogenetic matrices, with other regions underrepresented despite yielding important data. Scrutiny of the extent to which spatial biases influence the actinopterygian record is lacking, as is research on other forms of bias. Low richness in some time periods may be linked to geological biases, while the effects of taphonomic biases on Palaeozoic actinopterygians have not yet been investigated. Efforts are already underway both to redescribe poorly defined taxa and to describe taxa from underrepresented regions, helping to address taxonomic issues and accuracy of occurrence data. New methods of sampling standardisation utilising up-to-date occurrence databases will be critical in teasing apart biological changes in diversity and those resulting from bias. Lastly, continued phylogenetic work will enable the use of phylogenetic comparative methods to elucidate the origins of actinopterygian biogeography and subsequent patterns of radiation throughout their rise to dominate aquatic faunas.
