Abstract

A continuous random vector (X,Y) uniquely determines a copula C:[0,1]2→[0,1] such that when the distribution functions of X and Y are properly composed into C, the joint distribution function of (X,Y) results. A copula is said to be D4-invariant if its mass distribution is invariant with respect to the symmetries of the unit square. A D4-invariant copula leads naturally to a family of measures of concordance having a particular form, and all copulas generating this family are D4-invariant. The construction examined here includes Spearman’s rho and Gini’s measure of association as special cases.

2. Baited remote underwater video sample less site attached fish species along a subsea pipeline compared to a remotely operated vehicle

Abstract

Context Remotely operated vehicles (ROVs) are routinely used to inspect oil and gas infrastructure for industry’s operational purposes and scientists utilise this video footage to understand how fish interact with these structures. Aim This study aims to clarify how fish abundance data obtained from ROV video compares to that collected using baited remote underwater video (BRUV). Method This study compares fish assemblages observed using an industry ROV and BRUVs along a pipeline located in 130-m water depth in north-west Australia. Key results Both methods recorded 22 species of fish, however each method observed 15 unique species. The fish assemblage recorded by each method was statistically different at all sites. Differences in the fish assemblages correlated with the caudal fin aspect ratio of each species: the mean caudal fin aspect ratio of fish recorded using BRUVs was 2.81, compared to 1.87 for ROV observations. Conclusions We interpret this to indicate differences in site attachment, with site-attached species having generally lower caudal fin aspect ratios that are associated with slower swimming speeds with a burst and glide pattern. Implications Our results show that these remote video methods predominantly sample different fish assemblages and demonstrates how different sampling methods can provide different insights into fish interactions with subsea infrastructure.

Abstract

Efficacious monitoring of fish stocks is critical for efficient management. Multibeam acoustic cameras, that use sound-reflectance to generate moving pictures, provide an important alternative to traditional video-based methods that are inoperable in turbid waters. However, acoustic cameras, like standard video monitoring methods, produce large volumes of imagery from which it is time consuming and costly to extract data manually. Deep learning, a form of machine learning, can be used to automate the processing and analysis of acoustic data. We used convolutional neural networks (CNNs) to detect and count fish in a publicly available dual-frequency identification sonar (DIDSON) dataset. We compared three types of detections, direct acoustic, acoustic shadows, and a combination of direct and shadows. The deep learning model was highly reliable at detecting fish to obtain abundance data using acoustic data. Model accuracy for counts-per-image was improved by the inclusion of shadows (F1 scores, a measure of the model accuracy: direct 0.79, shadow 0.88, combined 0.90). Model accuracy for MaxN per video was high for all three types of detections (F1 scores: direct 0.90, shadow 0.90, combined 0.91). Our results demonstrate that CNNs are a powerful tool for automating underwater acoustic data analysis. Given this promise, we suggest broadening the scope of testing to include a wider range of fish shapes, sizes, and abundances, with a view to automating species (or ‘morphospecies’) identification and counts.

5. Echocardiography-derived septal curvature correlated with invasive hemodynamics in pediatric pulmonary hypertension

Abstract

Right ventricular function and afterload are associated with clinical outcomes in pulmonary hypertension (PH). MRI-derived interventricular septal curvature has been associated with invasive hemodynamics in PH patients. This study sought to determine the relationship of echocardiography derived septal curvature with invasive hemodynamics in pediatric PH patients.A single center chart review identified 56 pediatric patients with PH and 50 control patients with adequate echocardiography to assess septal curvature within one month of initial cardiac catheterization. Echocardiographic indices of septal flattening including end-systolic eccentricity index (EIPH patients had a median right ventricular systolic pressure of 64 mmHg (interquartile range (IQR) 48-81), mean pulmonary artery pressure of 44 mmHg (IQR 32-57), pulmonary vascular resistance of 7.9 iWU (IQR 4.8-12.9), and pulmonary capillary wedge pressure of 10 mmHg (IQR 8-12). Patients with PH had higher EIs and EIEchocardiography derived septal curvature is a non-invasive marker of ventricular afterload and adverse outcomes.

Abstract

Helminth parasites infect over 2 billion people worldwide resulting in huge global health and economic burden. Helminths typically stimulate Type 2 immune responses and excel at manipulating or suppressing host-immune responses resulting in chronic infections that can last for years to decades. Alongside the importance for the development of helminth treatments and vaccines, studying helminth immunity has unraveled many fundamental aspects of Type 2 immunity and immune regulation with implications for the treatment of autoimmunity and Type 2-mediated diseases, such as allergies. Here we describe the maintenance and use of Litomosoides sigmodontis, a murine model for studying host-parasite interactions, Type 2 immunity, and vaccines to tissue-dwelling filarial nematodes, which in humans cause lymphatic filariasis (e.g., Brugia malayi) and onchocerciasis (Onchocerca volvulus).

Abstract

International government guidance recommends patient and public involvement (PPI) to improve the relevance and quality of research. PPI is defined as research being carried out ‘with’ or ‘by’ patients and members of the public rather than ‘to’, ‘about’ or ‘for’ them (http://www.invo.org.uk/). Patient involvement is different from collecting data from patients as participants. Ethical considerations also differ. PPI is about patients actively contributing through discussion to decisions about research design, acceptability, relevance, conduct and governance from study conception to dissemination. Occasionally patients lead or do research. The research methods of PPI range from informal discussions to partnership research approaches such as action research, co-production and co-learning. This article discusses how researchers can involve patients when they are applying for research funding and considers some opportunities and pitfalls. It reviews research funder requirements, draws on the literature and our collective experiences as clinicians, patients, academics and members of UK funding panels.

Abstract

Survivors of brain tumors are faced with a high risk for a wide range of cognitive problems and learning difficulties. These problems are caused by the lesion itself and its surgical removal as well as by the treatments to follow (chemo- and/or radiation therapy). A few recent studies have indicated that children with brain tumors (BT) might exhibit cognitive problems already at diagnosis, i.e. before the start of any medical treatment. The aim of the present study was to investigate the "baseline" neuropsychological profile in children with BT in comparison to children with an oncological diagnosis not involving the central nervous system (CNS). 20 children with BT and 27 children with an oncological disease without involvement of the CNS (age range: 6.1 to 16.9 years) were evaluated with an extensive battery of neuropsychological tests tailored to the patient's age. Furthermore, the child and its parents completed self-report questionnaires about emotional functioning and quality of life. In both groups, tests were administered before any therapeutic intervention such as surgery, chemotherapy or irradiation. Groups were comparable regarding age, gender and social economic status. Compared to the CG, patients with BTs performed significantly worse in tests of working memory, verbal memory and attention. In contrast the areas of perceptual reasoning, processing speed and verbal comprehension were preserved at this time. Younger children with BT were especially disadvantaged. Compared to aged matched children with malignancies not involving the CNS and older BT patients the young BT patients showed deficits in attention, working memory and verbal memory measures. Our results highlight the need for cognitive assessments and interventions early in the treatment process in order to minimize or even prevent academic difficulties as patients return to school.

Abstract

SUMMARY Aims: To compare the efficacy, safety and tolerability of an omeprazole/amoxycillin (OA) dual therapy Helicobacter pylori eradication regimen with an omeprazole/amoxycillin/metronidazole (OAM) triple therapy regimen. Methods: In this double-blind trial, conducted in 19 hospitals, 119 patients with symptomatic duodenal ulcer disease were randomized to receive either 14 days treatment with omeprazole 40 mg daily, amoxycillin 500 mg t.d.s. and placebo followed by a further 14 days’treatment with omeprazole 20 mg daily (n= 59) or 14 days treatment with omeprazole 40 mg daily, amoxycillin 500 mg t.d.s., and metronidazole 400 mg t.d.s., followed by a further 14 days’treatment with omeprazole 20 mg daily (n= 60). H. pylori status was assessed by 13C-urea breath test at entry and at 4 weeks post-treatment. Results: H. pylori infection was eradicated in 46% of the OA treated patients and in 92% of the OAM treated patients, a mean difference of 46% (P < 0.0001, 95% CI for the difference: + 30 to + 62). In only one patient was the duodenal ulcer not endoscopically healed after 4 weeks of treatment (OA 100%; OAM 98% healed). There were no significant differences in speed of symptom relief or improvement in symptoms between the two groups. Both regimens were well tolerated, with 96% of patients completing the course, and only one patient withdrawing due to an adverse event. The only side-effect with a significantly higher incidence in the OAM group was diarrhoea, which occurred in 36% of patients compared to 16% of patients in the OA group (P < 0.05). Conclusions: A regimen consisting of omeprazole 40 mg daily, amoxycillin 500 mg t.d.s. and metronidazole 400 mg t.d.s. for 14 days gives an appreciably higher H. pylori eradication rate than omeprazole and amoxycillin alone, with acceptable tolerability.

In 1984 Scarsini introduced a set of axioms for measures of concordance of ordered pairs of continuous random variables. We exhibit an extension of these axioms to ordered n-tuples of continuous random variables, n ≥ 2. We derive simple properties of such measures, give examples, and discuss the relation of the extended axioms to multivariate measures of concordance previously discussed in the literature.

Abstract

A survey was made of the prevalence, diagnosis and treatment of dermatological conditions in small animals in general practice in the UK. Out of 3707 small animal consultations in general practice that were observed and recorded, 795 (21·4 per cent) involved animals that had a dermatological problem. In dogs and exotic species, pruritus was the most common presenting sign, accounting for 30 to 40 per cent of the dermatological consultations. In cats, cutaneous swellings were the most common presentation (36 per cent). A diagnosis or recommendation for treatment was made on the basis of the presenting clinical signs and physical examination alone in 576 (72 per cent) of the cases, and various diagnostic tests were performed in the other cases. In dogs, parasitic infestations, bacterial infections and neoplasia accounted for the majority of the diagnoses. In cats, parasites and bacterial infections were the most common. In exotic species, parasites accounted for over 80 per cent of the dermatological diagnoses. In dogs, the most common final diagnoses were otitis, pyoderma, anal sac impaction, flea infestation and atopic dermatitis. In cats, abscesses, flea infestation, and otitis were the most common diagnoses. In exotic species, the most common diagnosis was an unspecified mite infestation. Systemic antibiotics were prescribed in 196 cases (25 per cent), systemic glucocorticoids were prescribed in 162 cases (20 per cent) and treatment with an ectoparasiticide was prescribed in 167 cases (21 per cent). Abstract

This paper describes the redesign of the HP turbine of the Rolls-Royce Trent 500 engine, making use of nonaxisymmetric end walls. The original, datum turbine used conventional axisymmetric end walls, while the vane and (shrouded) rotor aerofoil profiles were nominally the same for the two designs. Previous research on the large-scale, low-speed linear cascade at Durham University (see Hartland et al., 1998, “Non-Axisymmetric End Wall Profiling in a Turbine Cascade,” ASME 98–GT-525), had already demonstrated significant potential for reducing turbine secondary losses using nonaxisymmetric end walls-by about one third. This paper shows how a methodology was derived from the results of this research and applied to the design of the single-stage Trent 500-HP turbine (model rig). In particular, the application of a new linear design system for the parametric definition of these end wall shapes (described in Harvey et al., 1999, “Non-Axisymmetric Turbine End Wall Design: Part I Three-Dimensional Linear Design System,” ASME 99–GT-337) is discussed in detail.

Abstract

We present intermediate-resolution spectra for the powerful radio galaxy Cygnus A taken with the ISIS spectrograph on the William Herschel Telescope. The spectra show complex emission-line profiles across the entire extent of the narrow-line region (NLR) along the radio axis (PA 105°), with line splittings (Δ V ∼ 300 km s - 1 ) at radial distances of 1-2 kpc to both the north-east and south-west, and a combination of broad (500 < FWHM < 800 km s - 1 ) and narrow (FWHM < 300 km s - 1 ) components within a radial distance of 1 kpc of the nucleus. The wide spectral coverage of the data allows us for the first time to measure the reddening, physical conditions and ionization of the kinematic subcomponents individually. Only the broad component detected in the near-nuclear regions shows evidence for significant reddening over and above that due to the Galaxy. The level of reddening of the nuclear broad component is consistent with the idea that the extinction associated with the kiloparsec-scale dust lane induces a mild (≤factor 2) anisotropy in the total emission-line flux, although there is no evidence for a compact, high-density inner narrow-line region in this source. Following correction for reddening, we find a remarkable degree of uniformity in the physical conditions and ionization state between the various spatial and kinematic subcomponents, suggesting a common ionization mechanism for all the subcomponents, despite the differences in kinematics. We use diagnostic diagrams to demonstrate that photoionization by the active galactic nucleus is the dominant ionization mechanism. However, a combination of matter- and radiation-bounded photoionized components is required to explain the strength of the [Fe VIII] λ6087 line and the high electron temperature measured from the [O III] (5007 + 4959)/4363 diagnostic ratio. A major outstanding issue for our understanding of the NLR gas in this and other radio galaxies is how the uniformity in the ionization and physical conditions can be maintained in a mixed photoionized medium across a range of spatial scales and kinematic subcomponents.

Abstract

Whooping cough is a highly contagious respiratory disease caused by Bordetella pertussis (B. pertussis). T helper 17 (Th17) cells have a central role in the resolution of the infection. Emerging studies document that type I interferons (IFNs) suppress Th17 differentiation and interleukin (IL)-17 responses in models of infection and chronic inflammation. As plasmacytoid dendritic cells (pDCs) are a major source of type I IFNs, we hypothesize that during B. pertussis infection in mice, pDC-derived IFNα inhibits a rapid increase in Th17 cells. We found that IFNα-secreting pDCs appear in the lungs during the early stages of infection, while a robust rise of Th17 cells in the lungs is detected at 15 days post-infection or later. The presence of IFNα led to reduced Th17 differentiation and proliferation in vitro. Furthermore, in vivo blocking of IFNα produced by pDCs during infection with B. pertussis infection resulted in early increase of Th17 frequency, inflammation, and reduced bacterial loads in the airways of infected mice. Taken together, the experiments reported here describe an inhibitory role for pDCs and pDC-derived IFNα in modulating Th17 responses during the early stages of B. pertussis infection, which may explain the prolonged nature of whooping cough.

Abstract

Atypical teratoid rhabdoid tumor of the brain is a rare embryonal tumor of infancy which has become more widely recognized due to specific immunostaining now more routinely available. These tumors still carry a poor prognosis and no standard therapy is currently available. However, the recent development of aggressive multimodality strategies seems to be associated with improved survival rates. In this chapter, we provide a review of our current knowledge on the demographics and epidemiology, pathology, and molecular characteristics. We review each individual therapeutic modality used (surgery, conventional chemotherapy, high-dose chemotherapy with stem cell rescue, intrathecal chemotherapy, and radiation) and the contribution of each to outcome when available. We provide an overview of the most recent therapeutic trials published and discuss the future options in development.

Abstract

Continuous high-quality fibers of yttria-doped zirconia with 4 and 8 mol% yttria were blow spun using an inorganic sol precursor. The fibers were heated and analyzed using scanning electron microscopy, x-ray diffraction, differential thermal/thermogravimetric analysis, and surface area/porosity by nitrogen adsorption. The fibers were of a single-phase cubic structure after crystallization at 529 °C and typically 3.3–5.5 or 7.5–11.5 μm in diameter depending on the composition. Samples of fibers were heated to various temperatures and their friability index and creep properties analyzed using the bend stress relaxation technique and compared to other fibers of similar composition prepared by different solution techniques and commercially available fibers.

Abstract

A Kjeldahl digestion method normally used for total nitrogen (N) was compared with NaOH fusion and dry ashing methods for determination of total phosphorus (P). Overall there was good correlation between the methods (r2=0.996). Results from the digestion method were on average 3.7% lower than the fusion and dry ashing methods, but there were significant differences between methods as shown by the regression analysis. The digestion method proved suitable for analysis of highly organic soils and some soils from the Pacific Islands. It was also useful for estimation of total ? in well‐weathered soils such as Oxisols and in A horizons, but not suitable for less weathered mineral soils.

Abstract

Background: Despite widespread use of aminosalicylates as maintenance treatment for ulcerative colitis (UC), patients still report troublesome symptoms, often nocturnally. Aim: To compare the efficacy and safety of balsalazide (Colazide) with mesalazine (Asacol) in maintaining UC remission. Methods: A randomized, double-blind comparison of balsalazide 3 g daily (1.04 g 5-ASA) and mesalazine 1.2 g daily for 12 months, in 99 (95 evaluable) patients in UC remission. Results: Balsalazide patients experienced more asymptomatic nights (90% vs. 77%, P = 0.0011) and days (58% vs. 50%, N.S.) during the first 3 months. Abstract

There is environmental concern about the increasing land application of dairy shed effluent in New Zealand. To minimize groundwater contamination by applied nutrients and pathogens it is desirable to avoid preferential flow through topsoils. Our objective was to determine an irrigation rate that retained applied effluent in the topsoil of two commonly irrigated New Zealand soils. In a field experiment, well drained Horotiu soils and poorly drained Te Kowhai soils were irrigated with a pyranine dye/KBr tracer solution at four rates (5, 10, 15, 20 mm/h) using a drip-type rainfall simulator. Twenty minutes after irrigation ceased the soil was excavated horizontally at 25 or 50 mm intervals, and photographed under UV light until no further fluorescence was visible. Each layer was also analysed for bromide content, without subsampling. The wetting pattern was uneven in both soils as leachate moved preferentially through worm channels and structural cracks. Preferential flow was greatest in the Te Kowhai soil and increased at faster application rates. Dye fluorescence was curvilinearly related to bromide concentration. At both study sites, maintaining the irrigation rate at ≤ 10 mm/h minimized leachate movement into the subsoil. Pulsed irrigation at faster rates was not retained in the topsoil.

Background: There is need for an evidence-based comparison of clinical management strategies to provide the rationale for selection of a particular therapeutic approach to treatment. Ideal dyspepsia treatment should quickly and conveniently alleviate patient symptoms whilst also minimizing the use of healthcare resources. Aim: To examine dyspepsia symptom relief over 16 weeks and compare an omeprazole clinical management strategy with a commonly used combination of antacid–alginate followed by H2-antagonist. Methods: Seven hundred and twenty-five patients participated in this randomized, open, parallel group comparison over 16 weeks. Abstract

Background & Aims: Aminosalicylates are widely used in the treatment of ulcerative colitis (UC). Balsalazide is a novel mesalamine prodrug, activated by colonic bacteria. The aim of this study was to compare the efficacy and safety of balsalazide with that of a pH-dependent formulation of mesalamine in active UC. Methods: A randomized, double-blind study was performed comparing balsalazide, 6.75 g daily, with mesalamine, 2.4 g daily, administered for 4, 8, or 12 weeks to 101 (99 evaluable) patients with symptomatic, sigmoidoscopically verified UC. Results: More patients treated with balsalazide achieved symptomatic remission after 2 (64% [balsalazide] vs. 43% [mesalamine]), 4 (70% vs. 51%), 8 (78% vs. 45%), and 12 weeks (88% vs. 57%) and complete remission (none/mild symptoms, sigmoidoscopy grade 0/1, no rectal steroid use within 4 days) after 4 (38% vs. 12%), 8 (54% vs. 22%), and 12 weeks (62% vs. 37%). Patients taking balsalazide experienced more asymptomatic days (4 weeks, 24% vs. 14%) and achieved the first asymptomatic day more rapidly (median, 10 vs. 25 days). Fewer patients in the balsalazide group reported adverse events (48% vs. 71%); four serious adverse events occurred in the mesalamine group. Abstract

The organic carbon in New Zealand's vegetation and soils has been estimated for 1992 from updated national databases of vegetative cover and soil carbon. These databases were augmented by inclusion of vegetation and soils information for Stewart Island, and addition of carbon estimates for upland and high‐country soils of South Island. Plant biomass estimates from literature were combined with the Vegetative Cover Map of New Zealand to give an estimate of 2420 Mt carbon for vegetation carbon above and below ground, including litter and humus. More than 80% of this carbon occurs in indigenous forested ecosystems on less than 26% of the land area, with only about 5% in planted forests. Soil organic carbon estimates for two depth ranges (0–0.25 m, 0–1 m) were derived from the N.Z. Land Resource Inventory, the Soil Map of Stewart Island, and the National Soils Database. We have investigated the use of a microwave cavity (Labwell AB, Sweden) to improve the radiochemical yield of 2-[ 18 F]fluoro-2-deoxyglucose (2-[ 18 F]FDG). After characterizing the heating properties of the cavity, three steps of the Hamacher 2-[ 18 F]FDG synthesis which require heating—azeotropic distillation of the target water, nucleophilic substitution, and hydrolysis of the product—were investigated separately. The average radiochemical yield of 2-[ 18 F]FDG for the microwave synthesis, using the phase transfer reagent tetrabutylammonium bicarbonate, was 62 ± 4% (72 ± 5%, decay corrected, synthesis time=31 min).

Abstract

Levels of cadmium (Cd) in New Zealand pastoral soils have increased due to Cd impurities in applied fertilisers. As there is little information on the interaction of Cd with soil mineral‐organic matter complexes, the sorption of Cd by complexes of kaolinite with humic acid has been investigated. Sorption was measured at pH and ionic strength values typically found for solutions of pastoral soils in New Zealand. Sorption increased with the content of humic acid in the complex, and as the pH of the medium was raised from 4.2 to 6.3. Sorption was also influenced by the ionic strength of the ambient solution, notably by the nature of the cation in the added electrolyte. The experimental data were interpreted in terms of the effect of solution pH and ionic composition on the charge characteristics of kaolinite and humic acid. These factors, in turn, influence clay particle association as well as the clay‐humic and metal‐humic interaction.

Abstract

In vitro and in situ experimental models that are descriptive of drug absorption in vivo are valuable tools in the discovery of new chemical entities that are bioavailable after oral administration. The specific objective of the study was to compare the intestinal permeabilities obtained in the three absorption models for consistency, and to assess the utility of the models in predicting the fraction of dose absorbed in human studies. The intestinal absorption models that were compared are widely used: the rat in situ single-pass intestinal perfusion system, the rat everted intestinal ring method, and monolayers of human colon adenocarcinoma cell line (CACO-2). The models were compared using small molecular reference compounds, as well as a series of peptidomimetic (PM) analogs. Each model had strong potential for estimating the fraction absorbed. For small organic molecules, excellent correlation was observed when permeabilities from CACO-2 cells and perfusions, or everted rings and perfusions, were compared. Weaker correlation was observed between everted rings and CACO-2 cells. Permeabilities for the set of reference compounds and PMs were positively correlated between any two of the three systems. Variance between correlations for reference compounds and PMs are likely due to structural features and physicochemical properties that are unique to the latter class of compounds. The results support caution in extrapolating correlations based on findings with small organic molecules to the behavior of complex peptidomimetics. Corroboration of permeabilities with two methods of determination is a useful cross-validation of experimental systems, as well as producing a reliable permeability assessment. CACO-2 cell monolayers and rat single-pass intestinal perfusion combine the highest correlation between systems, most defined relationship with fraction absorbed in humans, and experimental logistics in-line with discovery candidates.

Abstract

Background/Aims: Dilatation combined with subsequent pharmacological control of gastroesophageal reflux represents a logical but poorly documented approach to the management of benign esophageal stricture. This large trial (366 patients) aimed to assess whether omeprazole as the most effective available medication for gastroesophageal reflux disease prevents recurrent stricture formation. Methods: Patients (n = 366) were randomized in a double-blind study to undergo either omeprazole (20 mg once daily; 180 evaluable patients) or ranitidine therapy (150 mg twice daily; 185 evaluable patients) for 1 year after dilatation to 12–18-mm diameter (36–54F gauge). Subsequently, endoscopy and dilatation were performed when clinically indicated and endoscopy on completion. Symptoms were assessed at clinic visits every 3 months and using weekly diary cards. Results: Fewer patients undergoing omeprazole therapy required redilatation compared with those on ranitidine (43 of 143 [30%] vs. 66 of 143 [46%] by 12 months; P P P P P Conclusions: Omeprazole, 20 mg once daily, was more effective than ranitidine, 150 mg twice daily, as prophylaxis against stricture recurrence and in providing symptom relief.
