Acetic acid (HOAc) and diethylenetriaminepentaacetic acid (DTPA) single extraction agents were evaluated as screening methods to estimate the mobility of some trace elements in coastal sediments from Algeciras Bay. Sediments’ total metal concentrations of most heavy metals were found to be high around the areas impacted by anthropogenic activities such a sewage, atmospheric deposition and industrial activities, with notable values for As, Ni, Cr, Pb and Cd. The order of significant extraction efficiencies obtained with DTPA, were as follows: Pb (25.65%), Cu (19.78%), Cd (14.80%) and Zn (11.25%), while those obtained with HOAc were: Mn (33.00%), Tl (24.88%), Pb (18.99%), Cd (13.59%) and Co (11.78%). The comparison between the risk assessment codes (RAC) and the percent metal extractable fractions provided results of serious concern. Very high risk values of Cu, Zn, Cd and Pb extracts in DTPA were observed near the metallurgical industry, with Mn and Tl in HOAc extracts showing high risk values near the same industrial area and harbour activities. Sediments’ total metal concentrations were compared with the Low Alert-Level (LAL) sediment quality guidelines, where Co, Pb, Zn and Ni in both extractants and Cd and Cu in DTPA as well as Tl extracted in HOAc exceeded the LAL values respectively. The Spearman Rank test showed positive correlations between Co, Cu, Ni and Zn in DTPA extracts and their corresponding total metal concentrations, with Co, Cr, Fe, Ni, Tl and Zn in HOAc and total concentrations showing positive correlations. Furthermore, higher positive correlations were found between both extraction methods for Co (ρ = 0.797), Cu (ρ = 0.777), Ni (ρ = 0.789) and Zn (ρ = 0.942), indicating comparable potential extraction efficiencies between these extractants for these metals in the sediment studied. Biotic samples from Algeciras Bay (South-west Europe) were studied to assess its ecological status, complementing the previous abiotic monitoring of trace metals in water and sediments. This bay is a densely populated area with intense port traffic and is highly industrialised with metal inputs. To study the impact of this, Zn, Cd, Pb, and Cu contents were determined in tissues of benthic ( Solea senegalensis ) and benthopelagic species ( Scorpaena porcus , Trigloporus lastoviza, and Diplodus sargus sargus ). Notable levels of Zn and Cu were found in the liver and gills of all fish species. Compared to international muscle guidelines, Pb sometimes exceeded the most restrictive values, outstanding S. porcus with 27% of samples above the permissible value. Metal pollution indexes revealed that the liver and gills of benthic species were more affected by metal pollution than benthopelagic species, especially in most industrialised sites. Particularly, S. senegalensis presented a higher accumulation factor from sediment of Cd and Cu in the liver (30.1 and 345.1), probably due to the close interaction as benthic species. Among the species studied, S. senegalensis and D. sargus sargus proved to be the best representative and useful bioindicators of metal-polluted environments as this bay. The results were consistent with the findings from the abiotic samples.

The ecological status of Algeciras Bay (South-west Europe), highly influenced by anthropogenic activities, was assessed by monitoring Zn, Cd, Pb, and Cu in water and sediment samples. Total contents and metal fractions with different availabilities and their spatial-seasonal distribution were determined. The trend in water and sediment contents were Zn > Pb ≈ Cu > Cd, without significant seasonal variations. Sites 3 and 4, closest to industrial activities, had the highest metal concentrations, mainly in sediments. Cd showed low partition coefficient in water, indicating higher bioavailability. Total metal content in sediments exceeded the threshold effect level for Cu and were close to Pb. The BCR procedure revealed the highest availabilities for Cd and Pb, due to its higher content in exchangeable and reducible fractions. Higher Pb levels (21.4 ± 5.1 mg/kg) were found in sediments of this bay compared with other ecosystems. Pollution indexes for sediment quality revealed that site 3 was the most polluted (CF = 7.12 and I geo = 2.25). For an integrative study of the ecological status of this significant bay, these results have been complemented with the metal evaluation in benthic and benthopelagic fish tissues in Ecological status of Algeciras Bay, in a highly anthropised area in south-west Europe, through metal assessment - Part II: Biotic samples .

The aim of this work was to compare the potential induction of oxidative stress and the antioxidant enzymatic response after a short-term waterborne exposure to copper (Cu) and arsenic (As) with that of the nanoparticles (NPs) of these elements (Cu-NPs and As-NPs) in fish larvae of the species Dicentrarchus labrax . Larvae were grouped in several tanks and exposed to different concentrations of contaminants (0 to 10 mg/L) for 24 or 96 h under laboratory conditions. Copper and arsenic concentrations were analysed in larval tissues using ICP-MS. A set of oxidative stress biomarkers, including the levels of hydroperoxides (HPs), and superoxide dismutase (SOD) and catalase (CAT) activities were assessed. The trace element concentrations (mg/kg d.w.) in larvae ranged as follows: 3.28-6.67 (Cu at 24 h) and 2.76-3.42 (Cu at 96 h); 3.03-8.31 (Cu-NPs at 24 h) and 2.50-4.86 (Cu-NPs at 96 h); 1.92-3.45 (As at 24 h) and 2.22-4.71 (As at 96 h); and 2.19-8.56 (As-NPs at 24 h) and 1.75-9.90 (As-NPs at 96 h). In Cu tests, the oxidative damage (ROOH levels) was induced from 0.1 mg/L at both exposure times, while for Cu-NPs, this damage was not observed until 1 mg/L, which was paralleled by concomitant increases in SOD activity. The CAT activity was also increased but at lower metal concentrations (0.01 mg/L and 0.1 mg/L for both chemical forms). No oxidative damage was observed for As or As-NPs after 24 h, but it was observed for As after 96 h of treatment with 0.01 mg/L. A decrease in SOD activity was observed for As after 24 h, but it turned out to be increased after 96 h. However, As-NPs did not alter SOD activity. The CAT activity was stimulated only at 96 h by As and at 24 h by As-NPs. Therefore, the two chemical forms of Cu exhibited a higher bioaccumulation and toxicity potential as compared to those of As. Importantly, the association of both Cu and As in NPs reduced the respective trace metal bioaccumulation, resulting also in a reduction in the toxic effects (mortality and biochemical). Furthermore, the assessment of oxidative stress-related biomarkers in seabass larvae appears to be a useful tool for biomonitoring environmental-occurring trace elements.

The design, characterization, and analytical application of a green optical sensor for the selective determination of Fe(II) ions is proposed. The sensor is based on the immobilization of the chromogenic reagent picolinaldehyde salicyloylhydrazone (SHPA) within a polymer inclusion membrane. To reduce solvent usage, the reagent was synthesized using a green mechanochemical procedure. The components for sensor preparation were optimized with a sequential simplex method and the optimal composition was found to be 0.59 g cellulose triacetate (base polymer), 0.04 g SHPA (chemosensor reagent), 4.9 mL dibutyl phthalate (plasticizer), and 38 mL dichloromethane (solvent). The conditions of iron analysis were also optimized resulting in pH 6 for aqueous solution, 90 min exposure time and 10 min short-term stability. The optical sensor showed a linear range from the limit of detection (0.48 µmol L -1 ) to 54 µmol L -1 Fe(II). The precision of the method was found to be 1.44% and 1.19% for 17.9 and 45 µmol L -1 Fe(II), respectively. The characteristics of the sensor allowed the design of a Fe(II)/Fe(III) speciation scheme. The methodology was successfully applied to the determination of iron in food preservatives, food additives, and dietary supplement. Additionally, the Fe speciation scheme was successfully applied to an agricultural fertilizer.

A disposable colour-changeable optical sensor based on an interesting polymer inclusion-membrane (PIM) was designed to determine Cd(II) ions in aqueous medium. The Schiff base 2-acetylpyridine benzoylhydrazone (2-APBH) immobilised on the polymer membrane was used as a sensing molecule. The amounts of the PIM components were optimised by a 3 2 fractional factorial design with two central points and two blocks. The best optical sensor composition consisted of 2.5 g of poly(vinylchloride) (PVC) as a base polymer, 3 mL of tributyl phosphate (TBP) as a plasticiser, and 0.02 g of 2-APBH as a reagent. The sensor showed a good linear response in the range from 0.02 mg L -1 (limit of detection) to 1 mg L -1 of Cd(II) under the following experimental conditions: pH 9.5 (adjusted using ammonium chloride buffer solution at 0.337 mol L -1 ), 60 min of exposure time plus 2 min of sonication (pulses at 2 s intervals), and 10 min of short-term stability. The relative standard deviation of the method was determined to be 4.04% for 0.4 mg L -1 of Cd(II). The optical sensor was successfully applied to the determination of Cd(II) in natural-water and art-paint samples.

A global assessment of arsenic (As), nickel (Ni), cobalt (Co) and chromium (Cr) was performed in environmental samples from an important industrial bay. Different fractions of water, sediments and tissues from four species of fish were analysed. Samples were collected from selected sampling sites during four consecutive samplings in spring and autumn seasons, in order to evaluate concentrations and their possible correlations among the aquatic compartments. While a higher availability of Cr and Ni was found in water, Co and As were the most available elements in sediments. In fish, the liver was the tissue with the highest proportion of As and Co, and gills showed the highest concentrations of Ni and Cr. Significance differences were observed among sites showing the pollution sources. In sediments, high correlations were found between total Co content and the most available fractions. Total Ni content highly correlated with the oxidisable fraction, while Cr total content tightly correlated with the least available fractions. Quality guideline values for sediments were frequently exceeded. In sediments and biota, concentrations were slightly higher than in other ecosystems, indicating that maritime, industrial and urban activities are affecting this type of ecosystem with great anthropogenic influence.

A single-use optical sensor was designed for Zn(II) determination based on the immobilisation of the colorimetric reagent 2-acetylpyridine benzoylhydrazone (2-APBH) in a polymer inclusion membrane (PIM) adhered on the surface of an inert rectangular strip of polyester (Mylar). Different components for the membrane preparation were tested and those resulting in membrane with good appearance, proper physical and optical properties and ease of preparation were selected. Factorial design 2 3 with three replicates of the central point was applied for the optimisation of the membrane composition. The optimal composition consisted of 2.5 g of poly(vinyl chloride) (PVC), 4 mL of tributyl phosphate (TBP) and 0.04 g of 2-APBH. The optode showed a linear dynamic range from 0.03 (detection limit) to 1 mg L -1 of Zn(II) ions with a response time of 30 min in aqueous solution at pH 6 and a relative standard deviation of 3.90% for 0.4 mg L -1 of Zn(II). The sensor exhibited good selectivity to Zn(II) over other commonly ions. A tandem system based on the coupling of a bulk liquid membrane and a flow injection analysis for the separation, preconcentration and spectrophotometric determination of copper in saline water is presented. The ligand pyridine-2-acetaldehyde benzoylhydrazone has been used as a carrier in the liquid membrane as well as a spectrophotometric reagent for UV-VIS detection. Simultaneous and sequential experimental designs were used to optimise the chosen variables of each technique, respectively. The metal was separated and preconcentrated from the sample with an efficiency of 100.5 ± 0.9% and a metal preconcentration factor of 16.1. The on-line FIA determination was accomplished after metal complexation by the reagent at pH 3. A linear response was obtained in a range from 6.9 to 984.5 µg L -1 Cu(II), providing a detection limit of 1.8 µg L -1 . Saline matrix and other metal ions were not cause of interferences with relative errors below 4.6% for 50 µg L -1 of Cu(II) determination. The proposed tandem system was successfully tested using a TMDA-62 certified reference material providing a relative error of + 1.9%; it was also applied to the Cu(II) determination in coastal seawater samples with low relative errors ranging from - 3.8% to 0.0% (using DPASV as reference method)., (Copyright © 2018 Elsevier B.V. A comprehensive approach to chemical accumulation and biological effects of short-term Cu exposure in juveniles of European seabass (Dicentrarchus labrax) has been achieved. Fish were exposed to 0.01-10mgL -1 nominal Cu concentrations for 24-96h. Metal concentrations in water and gills, liver, muscle and brain tissues were studied along with oxidative stress biomarkers (superoxide dismutase, catalase, glutathione peroxidase, lipid peroxidation). Induction of oxidative damage was observed in all the organs with differential antioxidant responses; gills appearing as the most sensitive from low environmentally water Cu concentrations as 0.01mgL -1 . Histopathological alterations were also observed in liver and gills, even without a significant Cu accumulation. The results show that the combination of oxidative stress parameters, particularly lipid peroxidation and glutathione peroxidase activities, and histopathological alterations provide a good model fish and reliable early biomarkers for monitoring Cu pollution in seawater and might call for the protection agencies to revise the Cu environmental standards., (Copyright © 2017 Elsevier Ltd. Trace metals (Cd, Co, Cu, Fe, Mn, Ni, Pb, Zn) were determined in two operationally defined fractions (HCl and pyrite) in sediments from Ensenada and El Sauzal harbors (Mexico). The HCl fraction had significantly higher metal concentrations relative to the pyrite fraction in both harbors, underlining the weak tendency of most trace metals to associate with pyrite. Exceptionally, Cu was highly pyritized, with degrees of trace metal pyritization (DTMP) >80% in both harbors. Dissolved Fe flux measurements combined with solid phase Fe sulfide data indicated that 98 mt of Fe are precipitated as iron sulfides every year in Ensenada Harbor. These Fe sulfides (and associated trace metals) will remain preserved in the sediments, unless they are perturbed by dredging or sediment resuspension. Calculations indicate that dredging activities could export to the open ocean 0.20±0.13 to (0.30±0.56)×10(3) mt of Cd and Cu, respectively, creating a potential threat to marine benthic organisms. Degrees of pyritization (DOP) values in Ensenada and El Sauzal harbors were relatively low (<25%) while degrees of sulfidization (DOS) were high (~50%) because of the contribution of acid volatile sulfide. DOP values correlated with DTMP values (p≤0.001), indicating that metals are gradually incorporated into pyrite as this mineral is formed. Significant correlations were also found between DTMP values and -log(Ksp(MeS)/Ksp(pyr)) for both harbors, indicating that incorporation of trace metals into the pyrite phase is a function of the solubility product of the corresponding metal sulfide. A simple technique for the isolation, concentration and matrix simplification of Cu(II) ion in natural waters is proposed. This method has been developed using poly(styrene-divinylbenzene) membrane disks modified with the synthesized ligand pyridoxal salicyloylhydrazone (PSH). The retained ions on the disks were eluted with 10 mL 1 mol L(-1) HNO(3) and measured by flame atomic absorption spectrometry at 324.8 nm. The influence of pH, amount of ligand, type and amount of eluent for the stripping of copper ion from the membrane over extraction efficiency were evaluated. Extraction efficiencies >99% were obtained by elution of the disks with minimal amount of solvent with a preconcentration factor up to 100. The precision of the method for 10 replicate measurements of aqueous solutions containing 40 μg L(-1) Cu(II) was 1.5% at significance level of 95%. The breakthrough volume for 5 μg of Cu(II) was found to be 1000 mL and the detection limit of the method was 4 ng L(-1) of metal ion. A study to determine total and mobile heavy metals concentrations in sediments from Algeciras Bay was performed and pollution hotspots were identified. The effects on aquatic organisms were established using sediment quality guidelines (SQGs). Ni and Cr exceeded the effect range medium and low levels, respectively, around industrial area. Potential toxicity of metals was determined by diethylenetriaminepenta-acetic acid (DTPA) extractions and low alert levels of Co, Cu, Zn, Cd, Ni and Pb were exceeded at most sampling sites. Three pollution indicators were used showing significant values for As, Ni, Cr, Pb and Cd. Sediment speciation using the sequential extraction BCR procedure was carried out, being Cd, Zn, Pb and As the most available metals. Principal component, cluster and ANOVA analyses were performed in order to assess the sources of metals and the influence of seasonality and anthropogenic activities on the sediment quality. Two principal component analysis (PCA) factors were obtained identifying the sampling sites affected by anthropogenic activities; Ni-Cr and Zn-Cu-V clusters were also obtained associated with stainless steel and petrochemical industrial activities. ANOVA showed the outstanding sites because of total metal concentration and significant differences among sampling sites by the acid extractable and reducible fractions for all metals except for Ba and V., (Copyright © 2011 Elsevier B.V.
