To improve the efficiency and durability of gas turbine components, advancements are needed in cooling technologies. To accomplish this task, some manufacturers are turning to additive manufacturing (AM), as it offers the ability to both rapidly iterate on component design, as well as incorporate unique internal cooling structures directly into parts. As one example, wavy microchannels can be readily integrated into turbine components. This study investigates wavy channels of varying channel amplitude and wavelength through experimental measurements of heat transfer and pressure loss. In addition to experimental testing, CFD predictions were made to identify internal flow features that impacted performance. Five channel geometries were integrated into test coupons that were additively manufactured out of Hastelloy-X using direct metal laser sintering. True coupon geometric characteristics and wall roughness values were captured non-destructively using computed tomography (CT) scans. Geometric analyses indicated that coupons were reproduced accurately with minimal deviation from design intent. Experimental results indicated that decreasing the channel wavelength and increasing the channel amplitude resulted in substantial increases in both bulk friction factor and Nusselt number with respect to the nominal case and were scaled using a relative waviness parameter. CFD simulations predicted significant mixing of flow in the cases with the smallest wavelength and greatest amplitude.

Molecular dynamics (MD) simulations provide an in silico method to study the structure of lipid-based formulations (LBFs) and the incorporation of poorly water-soluble drugs within such formulations. In order to validate the ability of MD to effectively model the properties of LBFs, this work investigates the well-known cyclosporine A formulations, Sandimmune® and Neoral®. Sandimmune® exhibits poor dispersibility and its absorption from the gastrointestinal tract is enhanced when administered after food, whereas Neoral® disperses comparatively well and shows no food effect. MD simulations were performed of both LBFs to investigate the differences observed in fasted and fed conditions. These conditions were also tested using an in vitro experimental model of dispersion and digestion. These MD simulations were able to show that the food effect observed for Sandimmune® can be explained by large changes in drug solubilization on addition of bile. In contrast, Neoral® is well dispersed in water or in simulated fasted conditions, and this dispersion is relatively unchanged on moving to fed conditions. These differences were confirmed using dispersion and digestion in vitro experimental model. The current data suggests that MD simulations are a potential method to model the fate of LBFs in the gastrointestinal tract, predict their dispersion and digestion, investigate behaviour of APIs within the formulations, and provide insights into the clinical performance of LBFs.

Introduction: The purpose of this study was to determine if pharmacological treatment could increase progenitor cell proliferation in the Sub-ventricular Zone of aged rats. Previous work had shown that increasing progenitor cell proliferation in this region correlated well (R2=0.78; p= 0.0007) with functional recovery in a damaged corpus callosum (white matter tract), suggesting that progenitor cell proliferation results in oligodendrocytes in this region. Methods: 10 month old male and female Sprague Dawley rats were fed the drugs for 30 days in cookie dough, then immunocytochemistry was performed on coronal brain sections, using Ki67 labeling to determine progenitor cell proliferation. Results: Female rats showed low endogenous (control) progenitor cell proliferation, significantly different from male rats (P Discussion: The three drug combinations (fluoxetine, simvastatin, and ascorbic acid, patent # 9,254,281) led to ~ 4 fold increase in progenitor cell proliferation in females, while male progenitor cell proliferation was highest with 50 mg/kg ascorbic acid. However, the ascorbic acid increase in proliferation appears to be only on the sides of the ventricles, which is not the region that normally gives rise to oligodendrocytes. Conclusion: There are innate gender differences in progenitor cell proliferation at the Sub-Ventricular Zone at middle age in rats, possibly due to the loss of estrogen in females. We also see notable gender differences in progenitor cell proliferation in the Sub ventricular Zone in response to common drugs, such as fluoxetine, simvastatin and Vitamin C (ascorbic acid).

Additive manufacturing (AM) provides designers with the freedom to implement many designs that previously would have been costly or difficult to traditionally manufacture. This experimental study leverages this freedom and evaluates several different pin shapes integrated into pin fin arrays of a variety of spacings. Test coupons were manufactured out of Hastelloy-X using direct metal laser sintering (DMLS) and manufacturer recommended process parameters. After manufacturing, internal surface roughness and as-built accuracy were quantified using Computed Tomography (CT) scans. Results indicated that pin fins were all moderately undersized, and that there was significant surface roughness on all interior surfaces. Experimental data indicated that diamond shaped pins were found to have the highest heat transfer of the tested shapes, but triangle shaped pins pointed into the flow incurred the smallest pressure drop. Modifications to the streamwise spacing of the pins had little impact on the friction factor, but did increase heat transfer with increasing pin density. Prior Nusselt number correlations found in literature underestimated heat transfer and pressure loss relative to what was measured resulting from the AM roughness. A new correlation was developed accounting for AM roughness on pin fin arrays.

Bacille Calmette-Guerin (BCG) is a potential tool in the control of Mycobacterium bovis in European badgers (Meles meles). A five year Test and Vaccinate or Remove (TVR) research intervention project commenced in 2014 using two BCG strains (BCG Copenhagen 1331 (Years 1-3/ BadgerBCG) and BCG Sofia SL2222 (Years 4-5). Badgers were recaptured around 9 weeks after the Year 5 vaccination and then again a year later. The Dual-Path Platform (DPP) Vet TB assay was used to detect serological evidence of M. bovis infection. Of the 48 badgers, 47 had increased Line 1 readings (MPB83 antigen) between the Year 5 vaccination and subsequent recapture. The number of BCG Sofia vaccinations influenced whether a badger tested positive to the recapture DPP VetTB assay Line 1 (p 0.001) while the number of BadgerBCG vaccinations did not significantly affect recapture Line 1 results (p = 0.59). Line 1 relative light units (RLU) were more pronounced in tests run with sera than whole blood. The results from an in_house MPB83 ELISA results indicated that the WB DPP VetTB assay may not detect lower MPB83 IgG levels as well as the serum DPP VetTB assay. Changes in interferon gamma assay (IFN-γ) results were seen in 2019 with significantly increased CFP-10 and PPDB readings. Unlike BadgerBCG, BCG Sofia induces an immune response to MPB83 (the immune dominant antigen in M. bovis badger infection) that then affects the use of immunodiagnostic tests. The use of the DPP VetTB assay in recaptured BCG Sofia vaccinated badgers within the same trapping season is precluded and caution should be used in badgers vaccinated with BCG Sofia in previous years. The results suggest that the DPP VetTB assay can be used with confidence in badgers vaccinated with BadgerBCG as a single or repeated doses.

Adolescence is a critical period of development with increased sensitivity toward psychological stressors. Many psychiatric conditions emerge during adolescence and animal studies have shown that that acute stress has long-term effects on hypothalamic-pituitary-adrenal axis function and behavior. We recently demonstrated that acute stress produces long-term electrophysiological changes in locus coeruleus and long-lasting anxiety-like behavior in adolescent male rats. Based on prior reports of increased stress sensitivity during adolescence and increased sensitivity of female locus coeruleus toward corticotropin releasing factor, we hypothesized that the same acute stressor would cause different behavioral and physiological responses in adolescent female and adult male and female rats one week after stressor exposure. In this study, we assessed age and sex differences in how an acute psychological stressor affects corticosterone release, anxiety-like behavior, and locus coeruleus physiology at short- and long-term intervals. All groups of animals except adult female responded to stress with elevated corticosterone levels at the acute time point. One week after stressor exposure, adolescent females showed decreased firing of locus coeruleus neurons upon current injection and increased exploratory behavior compared to controls. The results were in direct contrast to changes observed in adolescent males, which showed increased anxiety-like behavior and increased spontaneous and induced firing in locus coeruleus neurons a week after stressor exposure. Adult males and females were both behaviorally and electrophysiologically resilient to the long-term effects of acute stress. Therefore, there may be a normal developmental trajectory for locus coeruleus neurons which promotes stress resilience in adults, but stressor exposure during adolescence perturbs their function. Furthermore, while locus coeruleus neurons are more sensitive to stressor exposure during adolescence, the effect varies between adolescent males and females. These findings suggest that endocrine, behavioral, and physiological responses to stress vary among animals of different age and sex, and therefore these variables should be taken into account when selecting models and designing experiments to investigate the effects of stress. These differences in animals may also allude to age and sex differences in the prevalence of various psychiatric illnesses within the human population.

Cyclosporins are natural or synthetic undecapeptides with a wide range of actual and potential pharmaceutical applications. Several members of the cyclosporin compound family have remarkably high passive membrane permeabilities that are not well-described by simple structural metrics. Here we review experimental studies of cyclosporin structure and permeability, including cyclosporin-metal complexes. We also discuss models for the conformation-dependent permeability of cyclosporins and similar compounds. Finally, we identify current knowledge gaps in the literature and provide recommendations regarding future avenues of exploration.

Introduction The utilization of local or regional anaesthesia for manipulation of nasal fractures (MNF) avoids the need for general anaesthetic (GA), and the risk associated with instrumentation of the airway during the COVID-19 pandemic. Furthermore, MNF under local anaesthetic (LA) provides similar results with regards to cosmesis and patient satisfaction. We present our experience of performing MNF under LA during the COVID-19 pandemic. Method A single-centre, prospective study of all patients undergoing MNF under LA was conducted (13th July/20–11thSeptember/20). Following reduction, pain scores and patient satisfaction surveys were administered. Results A total of 25 patients (M/F:16/9, median age, 25.6yr (14-52yr)) were enrolled. The majority of patients received either one or two instillations of LA (n = 19, 76%). Pain reported during the MNF procedure was 4.4/10, whilst pain during LA administration was reported as 3.2/10. 80% of patients felt instillation of LA was less painful than expected. 88% of respondents tolerated the LA well, and only 8% would have opted for general anaesthetic. 24 (96%) participants were happy with the cosmetic result. Discussion MNF under LA is a safe and effective alternative to MNF under GA. More literature is needed to define the best method of administering LA prior to performing MNF.

Immunosuppressive ability in human MSC donors has been shown to be variable and may be a limiting factor in MSC therapeutic efficacy in vivo. The importance of cytokine activation of mesenchymal stromal cells (MSCs) to facilitate their immunosuppressive function is well established. This study sought to further understand the interactions between MSCs and the commonly used calcineurin inhibitor cyclosporine A (CsA). The existing literature regarding approaches that use MSCs and cyclosporine are conflicting regarding the effect of CsA on MSC potency and function. Here, we clearly demonstrate that when added at the same time as MSCs, CsA negatively affects MSC suppression of T cell proliferation. However, licencing MSCs with IFNγ before addition of CsA protects MSCs from this negative effect. Notably, adding CsA to MSCs after IFNγ pre-stimulation enhances MSC production of IDO. Mechanistically, we identified that CsA reduces SOCS1 expression to facilitate enhanced IDO production in IFNγ pre-stimulated MSCs. Importantly, CsA exposure to IFNγ pre-stimulated MSC before administration, significantly enhanced the potency of MSCs in a human relevant humanised mouse model of acute Graft versus Host Disease. In summary, this study identified a novel licencing strategy to enhance MSC potency in vitro and in vivo. Background Forelimb Asymmetry Test is a simple test of motor function, using exploration behavior of a rat in a novel environment and counting the number of times that a rat touches the wall with either forepaw. Our lab has noticed, however, that there appears to be an increased number of fingertip touches to the wall following a stroke in the impaired forelimb. New method We counted the number of times that the animal either laid its palm flat against the wall of the chamber or touched the wall with only its fingertips, for both the left and right forepaws. We also separated bouts of exploration, so we could clearly determine if fingertip touches normally were associated with a transition from resting state to exploration state. Results and comparison with existing methods Fishers exact test indicated that there were significant differences in the way that the animals touched the wall pre-stroke compared to post-stroke, with more fingertip touches occurring post-stroke. Counting palm touches as normal and fingertip touches as abnormal increases the sensitivity of the Forelimb Asymmetry analysis and gives a good correlation with the contralateral functional deficits determined by Montoya Staircase post-stroke. If we counted every wall touch as normal (palm touches and fingertip touches), we see a loss of sensitivity and a poor correlation with contralateral function as determined by Montoya Staircase. Conclusions This refinement of the Forelimb Asymmetry analysis improves correlation with Montoya Staircase contralateral function after stroke.

The aim of this quality improvement project was to improve hepatitis A and B vaccination rates by 20% in patients listed for liver transplant by improving vaccine tracking, ordering/scheduling practices, out-of-network vaccinations, and flagging electronic health records. We audited 101 records; 44 patients were vaccine-deficient. Combined vaccine completion rates improved 94.6% (44.7%-87.0%) after implementing the educational and health record interventions. We believe the results of this study have implications for improving survival and decreasing complication rates for patients with chronic liver disease.

Objectives: To develop three different training drill classification systems for Australian Rules football using physical and skill-related data.Methods: Forty professional Australian footballers wore 10 Hz Global Positioning System units for six matches and 17 training sessions including 35 drills. High intensity running per minute, metres per minute and high-intensity running as a percentage of total distance were obtained to represent each drill's physical requirements. Velocity at kick (moving or stationary), time in possession (greater or less than 2 s) and the presence of pressure were coded for each kick to represent the constraints associated with each drill.Results: For the first system, two k-means clustering algorithms were run on physical and skill data separately to identify similarities between drills. To determine whether glycaemic control and the prevalence of microvascular complications in Waikato children/youth with type 1 diabetes (T1D) has changed since 2003.A retrospective review was performed of clinical records of children and youth with T1D who were under the care of the Waikato Paediatric and Young Adult Diabetes Services between March 2016 and March 2017. Comparisons were made to published data from the same service in 2003.Despite a more than two-fold increase in insulin-pump therapy since 2003, glycaemic control was not significantly improved in either children or youth. However, since 2003 there has been a significant reduction in the prevalence of diabetic retinopathy (24.6% vs 6.0%; P=0.003) and nephropathy (6.0% vs 25.4%; P=0.002), while symptomatic diabetic neuropathy remains rare. This reduction occurred despite a significant increase in obesity and hypertension, and no significant difference in the rates of dyslipidaemia or smoking.There has been a marked reduction in microvascular complications in Waikato youth and young adults with type 1 diabetes, but the reasons for the reduction are not clear given there has been no significant improvements in glycaemic control.

Cocaine addiction is a chronic, relapsing disorder. Stress and cues related to cocaine are two common relapse triggers. We have recently shown that exposure to repeated restraint stress during early withdrawal accelerates the time-dependent intensification or “incubation” of cue-induced cocaine craving that occurs during the first month of withdrawal, although craving ultimately plateaus at the same level observed in controls. These data indicate that chronic stress exposure during early withdrawal may result in increased vulnerability to cue-induced relapse during this period. Previous studies have shown that chronic stress exposure in drug-naïve rats increases neuronal activity in the basolateral amygdala (BLA), a region critical for behavioral responses to stress. Given that glutamatergic projections from the BLA to the nucleus accumbens are critical for the incubation of cue-induced cocaine craving, we hypothesized that cocaine withdrawal and chronic stress exposure produce separate increases that additively increase BLA neuronal activity. To assess this, we conducted in vivo extracellular single-unit recordings from the BLA of anesthetized adult male rats following cocaine or saline self-administration (6 h/day for 10 days) and repeated restraint stress or control conditions on withdrawal days (WD) 6–14. Recordings were conducted from WD15 to WD20. Interestingly, cocaine exposure alone increased the spontaneous firing rate in the BLA to levels observed following chronic stress exposure in drug-naïve rats. Chronic stress exposure during cocaine withdrawal further increased firing rate. These studies may identify a potential mechanism by which both cocaine and chronic stress exposure drive cue-induced relapse vulnerability during abstinence.

Spoilage caused by yeasts is a constant, widespread problem in the beverage industry that can result in major economic losses. Fruit juices provide an environment that allows the proliferation of yeast. Some factories in South Africa are not equipped with laboratory facilities to identify spoilage yeasts and outsourcing becomes a prolonged process which obstructs corrective action planning. This study aimed to establish yeast diversity and apply a rapid method for preliminary identification of spoilage yeasts associated with a small-scale fruit juice bottling factory. Yeast population in the factory was determined by isolation from the production environment, process equipment and spoiled products. PCR-RFLP analysis targeting the 5.8S-ITS region and D1/D2 sequencing was used for identification. A total of 207 yeasts belonging to 10 different genera ( Candida, Lodderomyces, Wickerhamomyces, Yarrowia, Zygosaccharomyces, Zygoascus, Cryptococcus, Filobasidium, Rhodotorula/Cystobasidium and Trichosporon) were isolated and identified from the production environment and processing equipment. Candida intermedia, C. parapsilosis and Lodderomyces elongisporus were widely distributed in the factory. Zygosaccharomyces bailii, Z. bisporus, Zygoascus hellenicus and Saccharomyces cerevisiae were isolated from the spoiled products. The data provided a yeast control panel that was used successfully to identify unknown yeasts in spoiled products from this factory using polymerase chain reaction-restriction length polymorphism (PCR-RFLP) comparative analysis.

Cocaine addiction is a devastating public health epidemic that continues to grow. Studies focused on identifying biological factors influencing cocaine craving and relapse vulnerability are necessary in order to promote abstinence in recovering drug users. Sex and ovarian hormones are known to influence cocaine addiction liability and relapse vulnerability in both humans and rodents. Previous studies have investigated sex differences in the time-dependent intensification or “incubation” of cue-induced cocaine craving that occurs during withdrawal from extended-access cocaine self-administration and have identified changes across the rat reproductive cycle (estrous cycle). Female rats in the estrus stage of the cycle (Estrus Females), the phase during which ovulation occurs, show an increase in the magnitude of incubated cue-induced cocaine craving compared to females in all other phases of the estrous cycle (Non-Estrus Females). Here we extend these findings by assessing incubated craving across the estrous cycle during earlier (withdrawal day 1 and 15) and later withdrawal periods (withdrawal day 48). We found that this increase in the magnitude of incubated craving during estrus (Estrus Females) is present on withdrawal day 15 but not withdrawal day 1 and further increases by withdrawal day 48. No difference in the magnitude of incubated craving was observed between Males and Non-Estrus Females. Our data indicate that the effects of hormonal fluctuations on cue-induced cocaine craving intensify during the first month and a half of withdrawal, showing an interaction between abstinence length, estrous cycle fluctuations, and cocaine craving. SIGNIFICANCE STATEMENT Cocaine addiction is a chronic, relapsing condition. Females have been historically understudied in preclinical research studies but understanding how biological factors like sex and ovarian hormones influence relapse vulnerability is critical in our understanding of how to reduce craving and promote abstinence in recovering users of both sexes. Here we characterized changes in the intensification or “incubation” of cue-induced cocaine craving during earlier and later withdrawal from extended-access cocaine self-administration between male and female rats and across the estrous cycle. Our results indicate an interaction between estrous cycle fluctuations and time-dependent changes in the underlying craving state and lay the foundation for future studies focused on investigating cellular mechanisms driving sex differences in relapse vulnerability.

Aim To assess the proportion of patients with gout who achieve target serum urate levels, the drug regime required and the reasons for failing to do so. Methods We reviewed the files of all patients with gout who presented to a gout-oriented rheumatology practice between January 2010 and September 2014. Results Two hundred and thirty patients agreed to commence urate lowering therapy (ULT); 73% achieved their urate target, including 74% with non-tophaceous gout (target ≤ 0.36 mmol/L) and 71% with tophi (target ≤ 0.30 mmol/L). Of the 62 who failed to reach target, in 61 it was due to non-adherence and in one due to inefficacy. Conclusion Adherence remains the major challenge to successful long-term gout management.

Sea surface salinity (SSS) variability during the 1997–1998 El Nino event and the failed 2012–2013 and 2014–2015 El Nino events is explored using a combination of observations and ocean reanalyses. Previously, studies have mainly focused on the sea surface temperature (SST) and sea surface height (SSH) variability. This analysis utilizes salinity data from Argo and the Simple Ocean Data Assimilation (SODA) reanalysis to examine the SSS variability. Advective processes and evaporation minus precipitation (E−P) variability is understood to influence SSS variability. Using surface wind, surface current, evaporation, and precipitation data, we analyze the causes for the observed SSS variability during each event. Barrier layer thickness and upper level salt content are also examined in connection to subsurface salinity variability. Both advective processes and E-P variability are important during the generation and onset of a successful El Nino, while a lack of one or both of these processes leads to a failed ENSO event.

In team-sport, physical and skilled output is often described via aggregate parameters including total distance and number of skilled involvements. However, the degree to which these output change throughout a team-sport match, as a function of time, is relatively unknown. This study aimed to identify and describe segments of physical and skilled output in team-sport matches with an example in Australian Football. The relationship between the number of change points and level of similarity was also quantified. A binary segmentation algorithm was applied to the velocity time series, collected via wearable sensors, of 37 Australian football players (age: 23 ± 4 years, height: 187 ± 8 cm, mass: 86 ± 9 kg). A change point quotient of between 1 and 15 was used. For these quotients, descriptive statistics, spectral features and a sum of skilled involvements were extracted. Segment similarity for each quotient was evaluated using a random forest model. The strongest classification features in the model were spectral entropy and skewness. Offensive and defensive involvements were the weakest features for classification, suggesting skilled output is dependent on match circumstances. The methodology presented may have application in comparing the specificity of training to matches and designing match rotation strategies.

Patients with these inflammatory conditions are often treated with immunosuppressive drugs, however we do not fully understand how MSC and immunosuppressive drugs interact and whether or not they have complimentary or inhibitory effects on each other. Communities in the Okanagan Valley, Canada are increasingly under threat from forest fires due to climate change and expanding urban development into fire interface zones. The effects of forest fires are not always quantifiable ‘hard' impacts. The fluid and chaotic ‘soft' impacts can have a profound effect on the collective consciousness of the people living close to the fires. To make sense of these impacts and understand where and when these forest fires have taken place, the authors have developed and implemented a Geoweb tool to support citizen-to-citizen dialogue and tell the stories of these impacts. This article will explore the interlinked ‘chaos' that exists between forest fires, GIS and volunteered geographic information, using a Geoweb focused case study from the Okanagan Valley, and argue that the Geoweb offers an unprecedented opportunity for citizen-citizen interaction and combines many types of dissimilar and unstructured data into a unified whole.

Maps were historically used as tools of the elite to maintain and expand power and control. The development of participatory mapmaking and the geoweb have opened new avenues for broader citizen engagement and therefore challenge traditional power dynamics. This chapter analyzes three examples and presents experiential learning around participatory processes and VGI contributions. Specifically we explore who is contributing their information, what are their motivations and incentives, in what ways do users interact with available technologies, and how is this contributing to change? We conclude by discussing the roles of motivations, the type of contribution, organizational capacity and leadership, and objectives. In comparing and contrasting these case studies we examine the individual and organizational dynamics of engagement, and how this can better inform the discourse about VGI.

Background The Institute of Medicine has called for actions to understand and target sex‐related differences in care and outcomes for out‐of‐hospital cardiac arrest patients. We assessed changes in bystander and first‐responder interventions and outcomes for males versus females after statewide efforts to improve cardiac arrest care. Methods and Results We identified out‐of‐hospital cardiac arrests from North Carolina (2010–2014) through the CARES (Cardiac Arrest Registry to Enhance Survival) registry. Outcomes for men versus women were examined through multivariable logistic regression analyses adjusted for (1) nonmodifiable factors (age, witnessed status, and initial heart rhythm) and (2) nonmodifiable plus modifiable factors (bystander cardiopulmonary resuscitation and defibrillation before emergency medical services), including interactions between sex and time (ie, year and year 2 ). Of 8100 patients, 38.1% were women. From 2010 to 2014, there was an increase in bystander cardiopulmonary resuscitation (men, 40.5%–50.6%; women, 35.3%–51.8%; P for each P =0.007; women, 8.5%–23.7%, P =0.004). From 2010 to 2014, the unadjusted predicted probability of favorable neurologic outcome was higher and increased more for men (men, from 6.5% [95% confidence interval (CI), 5.1–8.0] to 9.7% [95% CI, 8.1–11.3]; women, from 6.3% [95% CI, 4.4–8.3] to 7.4% [95% CI, 5.5–9.3%]); while adjusted for nonmodifiable factors, it was slightly higher but with a nonsignificant increase for women (from 9.2% [95% CI, 6.8–11.8] to 10.2% [95% CI, 8.0–12.5]; men, from 5.8% [95% CI, 4.6–7.0] to 8.4% [95% CI, 7.1–9.7]). Adding bystander cardiopulmonary resuscitation and defibrillation before EMS (modifiable factors) did not substantially change the results. Conclusions Bystander and first‐responder interventions increased for men and women, but outcomes improved significantly only for men. Additional strategies may be necessary to improve survival among female cardiac arrest patients.

Recently epidemiological studies suggest females lose neuroprotection from neurodegenerative diseases as they go through menopause. It has been hypothesized that this neuroprotection is hormone-dependent. The current study characterized cell signaling molecules downstream of estrogen receptor beta that are known to play a role in memory, PKC, ERK, and connexin-43, in regions of the brain associated with memory decline in an attempt to elucidate significant changes that occur post-estrus. Total whole cell lysates were compared to isolated mitochondrial protein because mitochondrial function is known to be altered during aging. As hypothesized, protein concentrations differed depending on age, gender, and brain region. Additionally, many of these changes occurred within mitochondria but not within whole cell lysates indicating that these are epigenetic alterations. These findings accentuate the complexity of aging and provide insight into the gender-specific cellular processes that occur throughout this process.
