Copyright © 2023 The Author(s). Background: Sleep disturbance is common following hospital admission both for COVID-19 and other causes. The clinical associations of this for recovery after hospital admission are poorly understood despite sleep disturbance contributing to morbidity in other scenarios. We aimed to investigate the prevalence and nature of sleep disturbance after discharge following hospital admission for COVID-19 and to assess whether this was associated with dyspnoea. Methods: CircCOVID was a prospective multicentre cohort substudy designed to investigate the effects of circadian disruption and sleep disturbance on recovery after COVID-19 in a cohort of participants aged 18 years or older, admitted to hospital for COVID-19 in the UK, and discharged between March, 2020, and October, 2021. Participants were recruited from the Post-hospitalisation COVID-19 study (PHOSP-COVID). Follow-up data were collected at two timepoints: an early time point 2–7 months after hospital discharge and a later time point 10–14 months after hospital discharge. Sleep quality was assessed subjectively using the Pittsburgh Sleep Quality Index questionnaire and a numerical rating scale. Sleep quality was also assessed with an accelerometer worn on the wrist (actigraphy) for 14 days. Participants were also clinically phenotyped, including assessment of symptoms (ie, anxiety [Generalised Anxiety Disorder 7-item scale questionnaire], muscle function [SARC-F questionnaire], dyspnoea [Dyspnoea-12 questionnaire] and measurement of lung function), at the early timepoint after discharge. Actigraphy results were also compared to a matched UK Biobank cohort (non-hospitalised individuals and recently hospitalised individuals). Multivariable linear regression was used to define associations of sleep disturbance with the primary outcome of breathlessness and the other clinical symptoms. PHOSP-COVID is registered on the ISRCTN Registry (ISRCTN10980107). Findings: 2320 of 2468 participants in the PHOSP-COVID study attended an early timepoint research visit a median of 5 months (IQR 4–6) following discharge from 83 hospitals in the UK. Data for sleep quality were assessed by subjective measures (the Pittsburgh Sleep Quality Index questionnaire and the numerical rating scale) for 638 participants at the early time point. Sleep quality was also assessed using device-based measures (actigraphy) a median of 7 months (IQR 5–8 months) after discharge from hospital for 729 participants. After discharge from hospital, the majority (396 [62%] of 638) of participants who had been admitted to hospital for COVID-19 reported poor sleep quality in response to the Pittsburgh Sleep Quality Index questionnaire. A comparable proportion (338 [53%] of 638) of participants felt their sleep quality had deteriorated following discharge after COVID-19 admission, as assessed by the numerical rating scale. Device-based measurements were compared to an age-matched, sex-matched, BMI-matched, and time from discharge-matched UK Biobank cohort who had recently been admitted to hospital. Compared to the recently hospitalised matched UK Biobank cohort, participants in our study slept on average 65 min (95% CI 59 to 71) longer, had a lower sleep regularity index (–19%; 95% CI –20 to –16), and a lower sleep efficiency (3·83 percentage points; 95% CI 3·40 to 4·26). Similar results were obtained when comparisons were made with the non-hospitalised UK Biobank cohort. Overall sleep quality (unadjusted effect estimate 3·94; 95% CI 2·78 to 5·10), deterioration in sleep quality following hospital admission (3·00; 1·82 to 4·28), and sleep regularity (4·38; 2·10 to 6·65) were associated with higher dyspnoea scores. Poor sleep quality, deterioration in sleep quality, and sleep regularity were also associated with impaired lung function, as assessed by forced vital capacity. Depending on the sleep metric, anxiety mediated 18–39% of the effect of sleep disturbance on dyspnoea, while muscle weakness mediated 27–41% of this effect. Interpretation: Sleep disturbance following hospital admission for COVID-19 is associated with dyspnoea, anxiety, and muscle weakness. Due to the association with multiple symptoms, targeting sleep disturbance might be beneficial in treating the post-COVID-19 condition. Background: Most studies of immunity to SARS-CoV-2 focus on circulating antibody, giving limited insights into mucosal defences that prevent viral replication and onward transmission. We studied nasal and plasma antibody responses one year after hospitalisation for COVID-19, including a period when SARS-CoV-2 vaccination was introduced. Methods: In this follow up study, plasma and nasosorption samples were prospectively collected from 446 adults hospitalised for COVID-19 between February 2020 and March 2021 via the ISARIC4C and PHOSP-COVID consortia. IgA and IgG responses to NP and S of ancestral SARS-CoV-2, Delta and Omicron (BA.1) variants were measured by electrochemiluminescence and compared with plasma neutralisation data. Findings: Strong and consistent nasal anti-NP and anti-S IgA responses were demonstrated, which remained elevated for nine months (p < 0.0001). Nasal and plasma anti-S IgG remained elevated for at least 12 months (p < 0.0001) with plasma neutralising titres that were raised against all variants compared to controls (p < 0.0001). Of 323 with complete data, 307 were vaccinated between 6 and 12 months; coinciding with rises in nasal and plasma IgA and IgG anti-S titres for all SARS-CoV-2 variants, although the change in nasal IgA was minimal (1.46-fold change after 10 months, p = 0.011) and the median remained below the positive threshold determined by pre-pandemic controls. Samples 12 months after admission showed no association between nasal IgA and plasma IgG anti-S responses (R = 0.05, p = 0.18), indicating that nasal IgA responses are distinct from those in plasma and minimally boosted by vaccination. Interpretation: The decline in nasal IgA responses 9 months after infection and minimal impact of subsequent vaccination may explain the lack of long-lasting nasal defence against reinfection and the limited effects of vaccination on transmission. Photonic waveguides with triangular cross section are being investigated for material systems such as diamond, glasses and gallium nitride, which lack easy options to create conventional rectangular nanophotonic waveguides. The design rules for optical elements in these triangular waveguides, such as couplers and gratings, are not well established. Here we present simulations of elements designed to couple light into, and out of, triangular waveguides from the vertical direction, which can be implemented with current angled-etch fabrication technology. The devices demonstrate coupling efficiencies approaching \unit{50}{\%} for light focused from a high numerical aperture objective. The implementation of such couplers will enable fast and efficient testing of closely spaced integrated circuit components., 7 pages, 5 figures

How the cerebral cortex operates near a critical phase transition point for optimum performance. Individual neurons have limited computational powers, but when they work together, it is almost like magic. Firing synchronously and then breaking off to improvise by themselves, they can be paradoxically both independent and interdependent. This happens near the critical point: when neurons are poised between a phase where activity is damped and a phase where it is amplified, where information processing is optimized, and complex emergent activity patterns arise. The claim that neurons in the cortex work best when they operate near the critical point is known as the criticality hypothesis. In this book John Beggs—one of the pioneers of this hypothesis—offers an introduction to the critical point and its relevance to the brain. Drawing on recent experimental evidence, Beggs first explains the main ideas underlying the criticality hypotheses and emergent phenomena. He then discusses the critical point and its two main consequences—first, scale-free properties that confer optimum information processing; and second, universality, or the idea that complex emergent phenomena, like that seen near the critical point, can be explained by relatively simple models that are applicable across species and scale. Finally, Beggs considers future directions for the field, including research on homeostatic regulation, quasicriticality, and the expansion of the cortex and intelligence. An appendix provides technical material; many chapters include exercises that use freely available code and data sets.

The present study is part of a larger project aiming to more closely integrate theory with empirical research into dynamic risk. It seeks to generate empirical findings with the dynamic risk factors contained in the Violence Risk Scale—Sexual Offense version (VRS-SO) that might constrain and guide the further development of Thornton’s theoretical model of dynamic risk. Two key issues for theory development are (a) whether the structure of pretreatment dynamic risk factors is the same as the structure of the change in the dynamic risk factors that occurs during treatment, and (b) whether theoretical analysis should focus on individual dynamic items or on the broader factors that run through them. Factor analyses and item-level prediction analyses were conducted on VRS-SO pretreatment, posttreatment, and change ratings obtained from a large combined sample of men ( Ns = 1,289–1,431) convicted and treated for sexual offenses. Results indicated that the latent structure of pretreatment dynamic risk was best described by a three-factor model while the latent structure of change items was two dimensional. Prediction analyses examined the degree to which items were predictive beyond prediction obtained from the broader factor that they loaded on. Results showed that for some items, their prediction appeared to be largely carried by the three broad factors. In contrast, other items seem to operate as funnels through which the broader factors’ predictiveness flowed. Implications for theory development implied by these results are identified.

We examined the incremental contributions of static and dynamic sexual violence risk assessment in a multisite sample of 1,289 men treated for sexual offending. The study extends validation work that established new risk categories and recidivism estimates for the Violence Risk Scale–Sexual Offense version (VRS-SO), using the risk assessment common language (CL) framework. Different rates of sexual recidivism were observed at different thresholds of static risk (Static-99R) as a function of dynamic risk and treatment change, particularly for men who were actuarially above or well above average risk (Levels IVa and IVb, respectively). A framework integrating CL risk levels for Static-99R and VRS-SO dynamic scores into overall CL risk levels is presented. We discuss implications for dynamic sexual violence risk assessment regarding the language used for risk communication and the importance of dynamic risk instruments in sexual violence evaluations, particularly when credible agents of risk change may be present.

Neural information processing is widely understood to depend on correlations in neuronal activity. However, whether correlation is favorable or not is contentious. Here, we sought to determine how correlated activity and information processing are related in cortical circuits. Using recordings of hundreds of spiking neurons in organotypic cultures of mouse neocortex, we asked whether mutual information between neurons that feed into a common third neuron increased synergistic information processing by the receiving neuron. We found that mutual information and synergistic processing were positively related at synaptic timescales (0.05–14 ms), where mutual information values were low. This effect was mediated by the increase in information transmission—of which synergistic processing is a component—that resulted as mutual information grew. However, at extrasynaptic windows (up to 3,000 ms), where mutual information values were high, the relationship between mutual information and synergistic processing became negative. In this regime, greater mutual information resulted in a disproportionate increase in redundancy relative to information transmission. These results indicate that the emergence of synergistic processing from correlated activity differs according to timescale and correlation regime. In a low-correlation regime, synergistic processing increases with greater correlation, and in a high-correlation regime, synergistic processing decreases with greater correlation., Author Summary In the present work, we address the question of whether correlated activity in functional networks of cortical circuits supports neural computation. To do so, we combined network analysis with information theoretic tools to analyze the spiking activity of hundreds of neurons recorded from organotypic cultures of mouse somatosensory cortex. We found that, at timescales most relevant to direct neuronal communication, neurons with more correlated activity predicted greater computation, suggesting that correlated activity does support computation in cortical circuits. Importantly, this result reversed at timescales less relevant to direct neuronal communication, where even greater correlated activity predicted decreased computation. Thus, the relationship between correlated activity and computation depends on the timescale and the degree of correlation in neuronal interactions.

Surgical excision remains the most commonly utilized treatment for superficial basal cell carcinoma (sBCC). In the era of cost containment of healthcare, the rising incidence of BCC and the high cost of excision require a continuous search for efficient and cost-effective management.Examine the feasibility of the diagnosis and treatment of low-risk sBCC in a single visit.Retrospective chart review of sBCCs diagnosed and treated in a single visit.The study identified 151 histologically confirmed sBCCs in 86 patients over a 5-year period, 93 (61.6%) cases of which were diagnosed as low-risk sBCC and treated in a single appointment. The majority of the cases (Diagnosis and treatment of sBCC in a single visit is an efficient and cost-effective management option for those who are proficient in identifying low-risk sBCC.

Prenatal cannabis exposure (PCE) influences human brain development, but it is challenging to model PCE using animals and current cell culture techniques. Here, we developed a one-stop microfluidic platform to assemble and culture human cerebral organoids from human embryonic stem cells (hESC) to investigate the effect of PCE on early human brain development. By incorporating perfusable culture chambers, air-liquid interface, and one-stop protocol, this microfluidic platform can simplify the fabrication procedure, and produce a large number of organoids (169 organoids per 3.5 cm x 3.5 cm device area) without fusion, as compared with conventional fabrication methods. These one-stop microfluidic assembled cerebral organoids not only recapitulate early human brain structure, biology, and electrophysiology but also have minimal size variation and hypoxia. Under on-chip exposure to the psychoactive cannabinoid, delta-9-tetrahydrocannabinol (THC), cerebral organoids exhibited reduced neuronal maturation, downregulation of cannabinoid receptor type 1 (CB1) receptors, and impaired neurite outgrowth. Moreover, transient on-chip THC treatment also decreased spontaneous firing in microfluidic assembled brain organoids. This one-stop microfluidic technique enables a simple, scalable, and repeatable organoid culture method that can be used not only for human brain organoids, but also for many other human organoids including liver, kidney, retina, and tumor organoids. This technology could be widely used in modeling brain and other organ development, developmental disorders, developmental pharmacology and toxicology, and drug screening.

Background: The risk factors for recovery from COVID-19 dyspnoea are poorly understood. We investigated determinants of recovery from dyspnoea in adults with COVID-19 and compared these to determinants of recovery from non-COVID-19 dyspnoea. Methods: We used data from two prospective cohort studies: PHOSP-COVID (patients hospitalised between March 2020 and April 2021 with COVID-19) and COVIDENCE UK (community cohort studied over the same time period). PHOSP-COVID data were collected during hospitalisation and at 5-month and 1-year follow-up visits. COVIDENCE UK data were obtained through baseline and monthly online questionnaires. Dyspnoea was measured in both cohorts with the Medical Research Council Dyspnoea Scale. We used multivariable logistic regression to identify determinants associated with a reduction in dyspnoea between 5-month and 1-year follow-up. Findings: We included 990 PHOSP-COVID and 3309 COVIDENCE UK participants. We observed higher odds of improvement between 5-month and 1-year follow-up among PHOSP-COVID participants who were younger (odds ratio 1.02 per year, 95% CI 1.01–1.03), male (1.54, 1.16–2.04), neither obese nor severely obese (1.82, 1.06–3.13 and 4.19, 2.14–8.19, respectively), had no pre-existing anxiety or depression (1.56, 1.09–2.22) or cardiovascular disease (1.33, 1.00–1.79), and shorter hospital admission (1.01 per day, 1.00–1.02). Similar associations were found in those recovering from non-COVID-19 dyspnoea, excluding age (and length of hospital admission). Interpretation: Factors associated with dyspnoea recovery at 1-year post-discharge among patients hospitalised with COVID-19 were similar to those among community controls without COVID-19. We present our recent research on color centers in Aluminum Gallium Nitride which emit single photons up to room temperature. The mature processing technology which is available for group-III-nitrides and the host material’s optical transparency in the visible and infra-red opens up the possibility of novel applications in nanophotonics and quantum devices. We are working to create suspended photonic devices, including waveguides and photonic crystal cavities, which we will show can guide and enhance the color center emission.

Functional networks of cortical neurons contain highly interconnected hubs, forming a rich-club structure. However, the cell type composition within this distinct subnetwork and how it influences large-scale network dynamics is unclear. Using spontaneous activity recorded from hundreds of cortical neurons in orbitofrontal cortex of awake behaving mice and from organotypic cultures, we show that the rich-club is disproportionately composed of inhibitory neurons, and that inhibitory neurons within the rich-club are significantly more synchronous than other neurons. At the population level, neurons in the rich-club exert higher than expected Granger causal influence on overall population activity at a broad range of frequencies compared to other neurons. Finally, neuronal avalanche duration is significantly correlated with the fraction of rich neurons that participate in the avalanche. Together, these results suggest an unexpected role of a highly connected, inhibition-rich subnetwork in driving and sustaining activity in local cortical networks.SIGNIFICANCE STATEMENTIt is widely believed that the relative abundance of excitatory and inhibitory neurons in cortical circuits is roughly 4:1. This relative abundance has been widely used to construct numerous cortical network models. Here we show that contrary to this notion, a sub-network of highly connected hub neurons (rich-club) consists of a higher abundance of inhibitory neurons compared to that found in the entire network or the non-rich subnetwork. Inhibitory hub neurons contribute to higher synchrony within the rich club compared to the rest of the network. Strikingly, higher activation of the inhibition-dominated rich club strongly correlates with longer avalanches in cortical circuits. Our findings reveal how network topology combined with cell-type specificity orchestrates population wide activity in cortical microcircuits.

The hypothesis that living neural networks operate near a critical phase transition point has received substantial discussion. This “criticality hypothesis” is potentially important because experiments and theory show that optimal information processing and health are associated with operating near the critical point. Despite the promise of this idea, there have been several objections to it. While earlier objections have been addressed already, the more recent critiques of Touboul and Destexhe have not yet been fully met. The purpose of this paper is to describe their objections and offer responses. Their first objection is that the well-known Brunel model for cortical networks does not display a peak in mutual information near its phase transition, in apparent contradiction to the criticality hypothesis. In response I show that it does have such a peak near the phase transition point, provided it is not strongly driven by random inputs. Their second objection is that even simple models like a coin flip can satisfy multiple criteria of criticality. This suggests that the emergent criticality claimed to exist in cortical networks is just the consequence of a random walk put through a threshold. In response I show that while such processes can produce many signatures criticality, these signatures (1) do not emerge from collective interactions, (2) do not support information processing, and (3) do not have long-range temporal correlations. Because experiments show these three features are consistently present in living neural networks, such random walk models are inadequate. Nevertheless, I conclude that these objections have been valuable for refining research questions and should always be welcomed as a part of the scientific process.

Much evidence seems to suggest cortex operates near a critical point, yet a single set of exponents defining its universality class has not been found. In fact, when critical exponents are estimated from data, they widely differ across species, individuals of the same species, and even over time, or depending on stimulus. Interestingly, these exponents still approximately hold to a dynamical scaling relation. Here we show that the theory of quasicriticality, an organizing principle for brain dynamics, can account for this paradoxical situation. As external stimuli drive the cortex, quasicriticality predicts a departure from criticality along a Widom line with exponents that decrease in absolute value, while still holding approximately to a dynamical scaling relation. Objective. Many neural systems display spontaneous, spatiotemporal patterns of neural activity that are crucial for information processing. While these cascading patterns presumably arise from the underlying network of synaptic connections between neurons, the precise contribution of the network’s local and global connectivity to these patterns and information processing remains largely unknown. Approach. Here, we demonstrate how network structure supports information processing through network dynamics in empirical and simulated spiking neurons using mathematical tools from linear systems theory, network control theory, and information theory. Main results. In particular, we show that activity, and the information that it contains, travels through cycles in real and simulated networks. Significance. Broadly, our results demonstrate how cascading neural networks could contribute to cognitive faculties that require lasting activation of neuronal patterns, such as working memory or attention.

The two visual streams hypothesis is a robust example of neural functional specialization that has inspired countless studies over the past four decades. According to one prominent version of the theory, the fundamental goal of the dorsal visual pathway is the transformation of retinal information for visually-guided motor behavior. To that end, the dorsal stream processes input using absolute (or veridical) metrics only when the movement is initiated, necessitating very little, or no, memory. Conversely, because the ventral visual pathway does not involve motor behavior (its output does not influence the real world), the ventral stream processes input using relative (or illusory) metrics and can accumulate or integrate sensory evidence over long time constants, which provides a substantial capacity for memory. In this study, we tested these relations between functional specialization, processing metrics, and memory by training identical recurrent neural networks to perform either a viewpoint-invariant object classification task or an orientation/size determination task. The former task relies on relative metrics, benefits from accumulating sensory evidence, and is usually attributed to the ventral stream. The latter task relies on absolute metrics, can be computed accurately in the moment, and is usually attributed to the dorsal stream. To quantify the amount of memory required for each task, we chose two types of neural network models. Using a long-short-term memory (LSTM) recurrent network, we found that viewpoint-invariant object categorization (object task) required a longer memory than orientation/size determination (orientation task). Additionally, to dissect this memory effect, we considered factors that contributed to longer memory in object tasks. First, we used two different sets of objects, one with self-occlusion of features and one without. Second, we defined object classes either strictly by visual feature similarity or (more liberally) by semantic label. The models required greater memory when features were self-occluded and when object classes were defined by visual feature similarity, showing that self-occlusion and visual similarity among object task samples are contributing to having a long memory. The same set of tasks modeled using modified leaky-integrator echo state recurrent networks (LiESN), however, did not replicate the results, except under some conditions. This may be because LiESNs cannot perform fine-grained memory adjustments due to their network-wide memory coefficient and fixed recurrent weights. In sum, the LSTM simulations suggest that longer memory is advantageous for performing viewpoint-invariant object classification (a putative ventral stream function) because it allows for interpolation of features across viewpoints. The results further suggest that orientation/size determination (a putative dorsal stream function) does not benefit from longer memory. These findings are consistent with the two visual streams theory of functional specialization. A central component of information processing in both biological and artificial neural networks is their ability to perform synergistic integration–a type of computation. We established previously that synergistic integration varies directly with the strength of feedforward information flow. However, the relationships between both recurrent and feedback information flow and synergistic integration remain unknown. To address this, we analyzed the spiking activity of hundreds of neurons in organotypic cultures of mouse cortex. We asked how empirically observed synergistic integration–determined from partial information decomposition–varied with local functional network structure that was categorized into motifs with varying recurrent and feedback information flow. We found that synergistic integration was elevated in motifs with greater recurrent information flow beyond that expected from the local feedforward information flow. Feedback information flow was interrelated with feedforward information flow and was associated with decreased synergistic integration. We have developed an inductively coupled plasma etching technique using a Faraday cage to create suspended gallium-nitride devices in a single step. The angle of the Faraday cage, gas mix, and chamber condition define the angle of the etch and the cross-sectional profile, which can feature undercut angles of up to 45°. We fabricate singly- and doubly-clamped cantilevers of a triangular cross section and show that they can support single optical modes in the telecom C-band.

To understand how neural circuits process information, it is essential to identify the relationship between computation and circuit organization. Rich clubs, highly interconnected sets of neurons, are known to propagate a disproportionate amount of information within cortical circuits. Here, we test the hypothesis that rich clubs also perform a disproportionate amount of computation. To do so, we recorded the spiking activity of on average ∼300 well-isolated individual neurons from organotypic cortical cultures. We then constructed weighted, directed networks reflecting the effective connectivity between the neurons. For each neuron, we quantified the amount of computation it performed based on its inputs. We found that rich-club neurons compute ∼160% more information than neurons outside of the rich club. The amount of computation performed in the rich club was proportional to the amount of information propagation by the same neurons. This suggests that in these circuits, information propagation drives computation. In total, our findings indicate that rich-club organization in effective cortical circuits supports not only information propagation but also neural computation., Author Summary Here we answer the question of whether rich-club organization in functional networks of cortical circuits supports neural computation. To do so, we combined network analysis with information theoretic tools to analyze the spiking activity of hundreds of neurons recorded from organotypic cultures of mouse somatosensory cortex. We found that neurons in rich clubs computed significantly more than neurons outside of rich clubs, suggesting that rich clubs do support computation in cortical circuits. Indeed, the amount of computation that we found in the rich clubs was proportional to the amount of information they propagate, suggesting that in these circuits, information propagation drives computation.

The present study examined the discrimination and calibration properties of Violence Risk Scale–Sexual Offense version (VRS-SO) risk and change scores for sexual and violent recidivism as a function of age at release, on a combined sample of 1,287 men who had attended sexual offense-specific treatment services. The key aim was to examine to what extent VRS-SO scores can accurately discriminate recidivists from nonrecidivists among older cohorts, and if the existing age-related adjustments in the instrument adequately correct for increasing age. VRS-SO risk and change scores showed consistent properties of discrimination for sexual recidivism across the age cohorts, via area under the curve and Cox regression survival analysis, as demonstrated through fixed effects meta-analysis. Calibration analyses, employing logistic regression, demonstrated that age at release was consistently incrementally predictive of violent, but not sexual, recidivism after controlling for individual differences on static and dynamic risk factors. E/O index analyses demonstrated that predicted rates of sexual recidivism from VRS-SO scores, particularly when employed with Static-99R, were not significantly different from those observed among age cohorts; however, calibration was weaker for general violence. Implications for use of the VRS-SO in sexual recidivism risk assessment with older offenders are discussed.

Whether the brain operates at a critical “tipping” point is a long standing scientific question, with evidence from both cellular and systems-scale studies suggesting that the brain does sit in, or near, a critical regime. Neuroimaging studies of humans in altered states of consciousness have prompted the suggestion that maintenance of critical dynamics is necessary for the emergence of consciousness and complex cognition, and that reduced or disorganized consciousness may be associated with deviations from criticality. Unfortunately, many of the cellular-level studies reporting signs of criticality were performed in non-conscious systems (in vitro neuronal cultures) or unconscious animals (e.g. anaesthetized rats). Here we attempted to address this knowledge gap by exploring critical brain dynamics in invasive ECoG recordings from multiple sessions with a single macaque as the animal transitioned from consciousness to unconsciousness under different anaesthetics (ketamine and propofol). We use a previously-validated test of criticality: avalanche dynamics to assess the differences in brain dynamics between normal consciousness and both drug-states. Propofol and ketamine were selected due to their differential effects on consciousness (ketamine, but not propofol, is known to induce an unusual state known as “dissociative anaesthesia”). Our analyses indicate that propofol dramatically restricted the size and duration of avalanches, while ketamine allowed for more awake-like dynamics to persist. In addition, propofol, but not ketamine, triggered a large reduction in the complexity of brain dynamics. All states, however, showed some signs of persistent criticality when testing for exponent relations and universal shape-collapse. Further, maintenance of critical brain dynamics may be important for regulation and control of conscious awareness., Author summary Here we explore how different anaesthetic drugs change the nature of brain dynamics, using neural activity recorded from sub-dural electrophysiological arrays implanted in a macaque brain. Previous research has suggested that loss of consciousness under anaesthesia is associated with a movement away from critical brain dynamics, towards a less flexible regime. When comparing ketamine and propofol, two anaesthetics with largely different effects on consciousness, we find that propofol, but not ketamine, produces a dramatic reduction in the complexity of brain activity and restricts the range of scales where critical dynamics are plausible. These results suggest that maintenance of critical dynamics may be important for regulation and control of conscious awareness.

Detecting synaptic connections using large-scale extracellular spike recordings presents a statistical challenge. While previous methods often treat the detection of each putative connection as a separate hypothesis test, here we develop a modeling approach that infers synaptic connections while incorporating circuit properties learned from the whole network. We use an extension of the Generalized Linear Model framework to describe the cross-correlograms between pairs of neurons and separate correlograms into two parts: a slowly varying effect due to background fluctuations and a fast, transient effect due to the synapse. We then use the observations from all putative connections in the recording to estimate two network properties: the presynaptic neuron type (excitatory or inhibitory) and the relationship between synaptic latency and distance between neurons. Constraining the presynaptic neuron’s type, synaptic latencies, and time constants improves synapse detection. In data from simulated networks, this model outperforms two previously developed synapse detection methods, especially on the weak connections. We also apply our model to in vitro multielectrode array recordings from mouse somatosensory cortex. Here our model automatically recovers plausible connections from hundreds of neurons, and the properties of the putative connections are largely consistent with previous research.New & NoteworthyDetecting synaptic connections using large-scale extracellular spike recordings is a difficult statistical problem. Perfluorooctanesulfonic acid (PFOS) is a persistent organic pollutant with worldwide bioaccumulation due to a very long half-life. Perfluorooctanesulfonic acid exposure results in significant hepatic effects including steatosis, proliferation, hepatomegaly, and in rodents, carcinogenesis. The objective of this study was to determine whether PFOS exposure exacerbates nonalcoholic fatty liver disease and nonalcoholic steatohepatitis pathogenesis. Eight-week-old male C57BL/6 J mice (n = 5 per group) were fed ad libitum normal chow diet (ND) alone, 60% high-fat diet (HFD) alone, ND + PFOS, and HFD + PFOS (0.0001% w/w (1 mg/kg) of PFOS) for 6 weeks. Both HFD alone and the ND + PFOS treatment induced significant adiposity and hepatomegaly, but the HFD + PFOS treatment showed a marked protection. Oil Red O staining and quantitative analysis of hepatic lipid content revealed increased hepatic steatosis in ND + PFOS and in HFD alone fed mice, which was prevented in HFD + PFOS treatment. Further studies revealed that ND + PFOS treatment significantly affected expression of lipid trafficking genes to favor steatosis, but these changes were absent in HFD + PFOS group. Specifically, expression of CD36, the major lipid importer in the cells, and peroxisome proliferator-activated receptor gamma (PPARγ), its major regulator, were induced in HFD + no treatment (NT) and ND + PFOS-fed mice but remained unchanged in HFD + PFOS mice. In conclusion, these data indicate that coadministration of PFOS with HFD mitigates steatosis and hepatomegaly induced by HFD and that by PFOS fed in ND diet via regulation of cellular lipid import machinery. These findings suggest dietary lipid content be considered when performing risk management of PFOS in humans and the elucidation of PFOS-induced hepatotoxicity.

The present study sought to develop updated risk categories and recidivism estimates for the Violence Risk Scale-Sexual Offense version (VRS-SO; Wong, Olver, Nicholaichuk, & Gordon, 2003-2017), a sexual offender risk assessment and treatment planning tool. The overarching purpose was to increase the clarity and accuracy of communicating risk assessment information that includes a systematic incorporation of new information (i.e., change) to modify risk estimates. Four treated samples of sexual offenders with VRS-SO pretreatment, posttreatment, and Static-99R ratings were combined with a minimum follow-up period of 10-years postrelease (N = 913). Logistic regression was used to model 5- and 10-year sexual and violent (including sexual) recidivism estimates across 6 different regression models employing specific risk and change score information from the VRS-SO and/or Static-99R. A rationale is presented for clinical applications of select models and the necessity of controlling for baseline risk when utilizing change information across repeated assessments. Information concerning relative risk (percentiles) and absolute risk (recidivism estimates) is integrated with common risk assessment language guidelines to generate new risk categories for the VRS-SO. The manufacture of carbon fibers includes an oxidative electrochemical process that represents an untapped source of potential surface manipulation beyond simple installation of oxygen containing functional groups. In this work we show that applying an oxidative treatment, at much lower oxidative potentials (+1.75 V) compared to industry standards, can result in simultaneous surface modification and oxidation resulting in substantial increases in interfacial shear strength, up to 112%, relative to pristine carbon fibers. The optimal treatment window for this process is 10 min, offering synergistic oxidation and surface grafting interactions with epoxy resin. This treatment is easily scalable, amenable to a continuous oxidation process, and can be adapted to existing infrastructure currently used in carbon fiber manufacture.

Attachment of amine-bearing molecules, denoted surface grafted molecules (SGMs), onto the surface of carbon fibre has been previously shown to enhance interfacial interactions at the carbon fibre/epoxy interface. However, the design principles inherent to optimising this enhancement are not yet established. Here, we investigate the influence of SGM design criteria on the interfacial mechanical response for three types of SGM via predictions based on molecular dynamics simulations, and by experimental measurements. The SGMs are covalently grafted to the graphitic fibre surface via in situ generated and decomposed phenyl diazo salts. All three SGMs possess a phenyl ring as the surface attachment point, and differ by the number and/or position (meta or para) of the amine-terminated side-chain(s) attached to the ring. The single-chain meta-substituted SGM produces the least interfacial enhancement, while the double-chain meta-substituted SGM enhances the interfacial shear strength by 29% relative to the control. In contrast, the single-chain para-SGM performs almost comparably to the double-chain meta-SGM. Our modelling predictions recover this trend and offer molecular-scale explanations for these findings, providing guidance in the design of effective surface-tailoring strategies to realise enhancements in the shear response of carbon fibre–epoxy interfaces.

CD8 T-cell lymphomas comprise a wide spectrum of lymphomas, many which have yet to be formally classified. We present a case of a 43-year-old woman with an enlarging tumor distal to the distal interphalangeal joint of the fourth finger, compressing the underlying nail matrix. Magnetic resonance imaging showed bony involvement of the underlying distal phalanx. Histology showed a dense epidermotropic and pandermal infiltrate composed of medium-sized, uniformly pleomorphic lymphocytes with cleaved nuclei, which raised the possibility of primary cutaneous CD8 aggressive epidermotropic cytotoxic T-cell lymphoma. However, the patient's clinical photograph was inconsistent with this diagnosis. Other diagnoses, such as primary cutaneous acral CD8 T-cell lymphoma-a provisional entity, were also considered but did not capture all the features of this patient's lymphoma. We propose to classify this case as a primary cutaneous CD8 T-cell lymphoma, an indolent and locally aggressive form.
