Abstract: The IceCube Neutrino Observatory instruments about 1 km3 of deep, glacial ice at the geographic South Pole. It uses 5160 photomultipliers to detect Cherenkov light emitted by charged relativistic particles. An unexpected light propagation effect observed by the experiment is an anisotropic attenuation, which is aligned with the local flow direction of the ice. We examine birefringent light propagation through the polycrystalline ice microstructure as a possible explanation for this effect. The predictions of a first-principles model developed for this purpose, in particular curved light trajectories resulting from asymmetric diffusion, provide a qualitatively good match to the main features of the data. This in turn allows us to deduce ice crystal properties. Since the wavelength of the detected light is short compared to the crystal size, these crystal properties include not only the crystal orientation fabric, but also the average crystal size and shape, as a function of depth. By adding small empirical corrections to this first-principles model, a quantitatively accurate description of the optical properties of the IceCube glacial ice is obtained. In this paper, we present the experimental signature of ice optical anisotropy observed in IceCube light-emitting diode (LED) calibration data, the theory and parameterization of the birefringence effect, the fitting procedures of these parameterizations to experimental data, and the inferred crystal properties.

Abstract: Abstract High-energy tau neutrinos are rarely produced in atmospheric cosmic-ray showers or at cosmic particle accelerators, but are expected to emerge during neutrino propagation over cosmic distances due to flavor mixing. When high energy tau neutrinos interact inside the IceCube detector, two spatially separated energy depositions may be resolved, the first from the charged current interaction and the second from the tau lepton decay. We report a novel analysis of 7.5 years of IceCube data that identifies two candidate tau neutrinos among the 60 “High-Energy Starting Events” (HESE) collected during that period. The HESE sample offers high purity, all-sky sensitivity, and distinct observational signatures for each neutrino flavor, enabling a new measurement of the flavor composition. The measured astrophysical neutrino flavor composition is consistent with expectations, and an astrophysical tau neutrino flux is indicated at 2.8 $$\sigma $$ σ significance.

Abstract: Abstract The reconstruction of event-level information, such as the direction or energy of a neutrino interacting in IceCube DeepCore, is a crucial ingredient to many physics analyses. Algorithms to extract this high level information from the detector’s raw data have been successfully developed and used for high energy events. In this work, we address unique challenges associated with the reconstruction of lower energy events in the range of a few to hundreds of GeV and present two separate, state-of-the-art algorithms. One algorithm focuses on the fast directional reconstruction of events based on unscattered light. The second algorithm is a likelihood-based multipurpose reconstruction offering superior resolutions, at the expense of larger computational cost.

Abstract: Abstract Adopting the Standard Halo Model (SHM) of an isotropic Maxwellian velocity distribution for dark matter (DM) particles in the Galaxy, the most stringent current constraints on their spin-dependent scattering cross-section with nucleons come from the IceCube neutrino observatory and the PICO-60 $$\hbox {C}_3\hbox {F}_8$$ C 3 F 8 superheated bubble chamber experiments. The former is sensitive to high energy neutrinos from the self-annihilation of DM particles captured in the Sun, while the latter looks for nuclear recoil events from DM scattering off nucleons. Although slower DM particles are more likely to be captured by the Sun, the faster ones are more likely to be detected by PICO. Recent N-body simulations suggest significant deviations from the SHM for the smooth halo component of the DM, while observations hint at a dominant fraction of the local DM being in substructures. We use the method of Ferrer et al. (JCAP 1509: 052, 2015) to exploit the complementarity between the two approaches and derive conservative constraints on DM-nucleon scattering. Our results constrain $$\sigma _{\mathrm{SD}} \lesssim 3 \times 10^{-39} \mathrm {cm}^2$$ σ SD ≲ 3 × 10 - 39 cm 2 ( $$6 \times 10^{-38} \mathrm {cm}^2$$ 6 × 10 - 38 cm 2 ) at $$\gtrsim 90\%$$ ≳ 90 % C.L. for a DM particle of mass 1 TeV annihilating into $$\tau ^+ \tau ^-$$ τ + τ - ( $$b\bar{b}$$ b b ¯ ) with a local density of $$\rho _{\mathrm{DM}} = 0.3~\mathrm {GeV/cm}^3$$ ρ DM = 0.3 GeV / cm 3 . The constraints scale inversely with $$\rho _{\mathrm{DM}}$$ ρ DM and are independent of the DM velocity distribution.

Abstract: Abstract The Neutrino Mass Ordering (NMO) remains one of the outstanding questions in the field of neutrino physics. One strategy to measure the NMO is to observe matter effects in the oscillation pattern of atmospheric neutrinos above $$\sim 1\,\mathrm {GeV}$$ ∼1GeV , as proposed for several next-generation neutrino experiments. Moreover, the existing IceCube DeepCore detector can already explore this type of measurement. We present the development and application of two independent analyses to search for the signature of the NMO with three years of DeepCore data. These analyses include a full treatment of systematic uncertainties and a statistically-rigorous method to determine the significance for the NMO from a fit to the data. Both analyses show that the dataset is fully compatible with both mass orderings. For the more sensitive analysis, we observe a preference for normal ordering with a p-value of $$p_\mathrm {IO} = 15.3\%$$ pIO=15.3% and $$\mathrm {CL}_\mathrm {s}=53.3\%$$ CLs=53.3% for the inverted ordering hypothesis, while the experimental results from both analyses are consistent within their uncertainties. Since the result is independent of the value of $$\delta _\mathrm {CP}$$ δCP and obtained from energies $$E_\nu \gtrsim 5\,\mathrm {GeV}$$ Eν≳5GeV , it is complementary to recent results from long-baseline experiments. These analyses set the groundwork for the future of this measurement with more capable detectors, such as the IceCube Upgrade and the proposed PINGU detector.

Abstract: Using data from the IceCube Neutrino Observatory, we searched for high-energy neutrino emission from the gravitational-wave events detected by the advanced LIGO and Virgo detectors during their third observing run. We did a low-latency follow-up on the public candidate events released during the detectors’ third observing run and an archival search on the 80 confident events reported in the GWTC-2.1 and GWTC-3 catalogs. An extended search was also conducted for neutrino emission on longer timescales from neutron star containing mergers. Follow-up searches on the candidate optical counterpart of GW190521 were also conducted. We used two methods; an unbinned maximum likelihood analysis and a Bayesian analysis using astrophysical priors, both of which were previously used to search for high-energy neutrino emission from gravitational-wave events. No significant neutrino emission was observed by any analysis, and upper limits were placed on the time-integrated neutrino flux as well as the total isotropic equivalent energy emitted in high-energy neutrinos.

Abstract: Abstract The IceCube Collaboration has observed a high-energy astrophysical neutrino flux and recently found evidence for neutrino emission from the blazar TXS 0506$$+$$ + 056. These results open a new window into the high-energy universe. However, the source or sources of most of the observed flux of astrophysical neutrinos remains uncertain. Here, a search for steady point-like neutrino sources is performed using an unbinned likelihood analysis. The method searches for a spatial accumulation of muon-neutrino events using the very high-statistics sample of about 497,000 neutrinos recorded by IceCube between 2009 and 2017. The median angular resolution is $$\sim 1^\circ $$ ∼1∘ at 1 TeV and improves to $$\sim 0.3^\circ $$ ∼0.3∘ for neutrinos with an energy of 1 PeV. Compared to previous analyses, this search is optimized for point-like neutrino emission with the same flux-characteristics as the observed astrophysical muon-neutrino flux and introduces an improved event-reconstruction and parametrization of the background. The result is an improvement in sensitivity to the muon-neutrino flux compared to the previous analysis of $$\sim 35\%$$ ∼35% assuming an $$E^{-2}$$ E-2 spectrum. The sensitivity on the muon-neutrino flux is at a level of $$E^2 \mathrm {d} N /\mathrm {d} E = 3\cdot 10^{-13}\,\mathrm {TeV}\,\mathrm {cm}^{-2}\,\mathrm {s}^{-1}$$ E2dN/dE=3·10-13TeVcm-2s-1 . No new evidence for neutrino sources is found in a full sky scan and in an a priori candidate source list that is motivated by gamma-ray observations. Furthermore, no significant excesses above background are found from populations of sub-threshold sources. The implications of the non-observation for potential source classes are discussed.

Abstract: Abstract With the observation of high-energy astrophysical neutrinos by the IceCube Neutrino Observatory, interest has risen in models of PeV-mass decaying dark matter particles to explain the observed flux. We present two dedicated experimental analyses to test this hypothesis. One analysis uses 6 years of IceCube data focusing on muon neutrino ‘track’ events from the Northern Hemisphere, while the second analysis uses 2 years of ‘cascade’ events from the full sky. Known background components and the hypothetical flux from unstable dark matter are fitted to the experimental data. Since no significant excess is observed in either analysis, lower limits on the lifetime of dark matter particles are derived: we obtain the strongest constraint to date, excluding lifetimes shorter than $$10^{28}\hbox { s}$$ 1028s at 90% CL for dark matter masses above $$10~\hbox {TeV}$$ 10TeV .

10. Duodenal intussusception of the remnant stomach after biliopancreatic diversion: a case report

Abstract: Abstract Background We present a rare case of an antegrade intussusception of the remnant stomach four years after a biliopancreatic diversion. Case presentation A 55-year-old female patient presented with epigastric pain in our emergency room. Laboratory parameters showed an anemia as well as elevated transaminases and hyperbilirubinemia. The CT scan showed an intussusception of the remnant stomach into the duodenum followed by cholestasis. At laparotomy the remnant stomach was resected. Conclusion Bowel obstruction and intussusception after bariatric surgery are a rare but often unrecognized complication. Sonography as well as a CT scan should be performed. The exploratory laparoscopy however is the most valuable diagnostic tool in patients with suspected intussusception, due to the high rate of non-specific symptoms and misinterpreted radiographic investigations.

Abstract: Abstract IceCube is a neutrino observatory deployed in the glacial ice at the geographic South Pole. The $$\nu _\mu $$ νμ energy unfolding described in this paper is based on data taken with IceCube in its 79-string configuration. A sample of muon neutrino charged-current interactions with a purity of 99.5% was selected by means of a multivariate classification process based on machine learning. The subsequent unfolding was performed using the software Truee. The resulting spectrum covers an E$$_\nu $$ ν -range of more than four orders of magnitude from 125 GeV to 3.2 PeV. Compared to the Honda atmospheric neutrino flux model, the energy spectrum shows an excess of more than $$1.9\,\sigma $$ 1.9σ in four adjacent bins for neutrino energies $$E_\nu \ge 177.8\,\text {TeV}$$ Eν≥177.8TeV . The obtained spectrum is fully compatible with previous measurements of the atmospheric neutrino flux and recent IceCube measurements of a flux of high-energy astrophysical neutrinos.

Abstract: Abstract We present a search for a neutrino signal from dark matter self-annihilations in the Milky Way using the IceCube Neutrino Observatory (IceCube). In 1005 days of data we found no significant excess of neutrinos over the background of neutrinos produced in atmospheric air showers from cosmic ray interactions. We derive upper limits on the velocity averaged product of the dark matter self-annihilation cross section and the relative velocity of the dark matter particles $$\langle \sigma _{\text {A}}v\rangle $$ ⟨ σ A v ⟩ . Upper limits are set for dark matter particle candidate masses ranging from 10 GeV up to 1 TeV while considering annihilation through multiple channels. This work sets the most stringent limit on a neutrino signal from dark matter with mass between 10 and 100 GeV, with a limit of $$1.18\cdot 10^{-23}\text { cm}^3\text {s}^{-1}$$ 1.18 · 10 - 23 cm 3 s - 1 for 100 GeV dark matter particles self-annihilating via $$\tau ^+\tau ^-$$ τ + τ - to neutrinos (assuming the Navarro–Frenk–White dark matter halo profile).

Abstract: Abstract We present results from an analysis looking for dark matter annihilation in the Sun with the IceCube neutrino telescope. Gravitationally trapped dark matter in the Sun’s core can annihilate into Standard Model particles making the Sun a source of GeV neutrinos. IceCube is able to detect neutrinos with energies >100 GeV while its low-energy infill array DeepCore extends this to >10 GeV. This analysis uses data gathered in the austral winters between May 2011 and May 2014, corresponding to 532 days of livetime when the Sun, being below the horizon, is a source of up-going neutrino events, easiest to discriminate against the dominant background of atmospheric muons. The sensitivity is a factor of two to four better than previous searches due to additional statistics and improved analysis methods involving better background rejection and reconstructions. The resultant upper limits on the spin-dependent dark matter-proton scattering cross section reach down to $$1.46\times 10^{-5}$$ 1.46×10-5 pb for a dark matter particle of mass 500 GeV annihilating exclusively into $$\tau ^{+}\tau ^{-}$$ τ+τ- particles. These are currently the most stringent limits on the spin-dependent dark matter-proton scattering cross section for WIMP masses above 50 GeV.

Abstract: Astrophysical sources of gravitational waves, such as binary neutron star and black hole mergers or core-collapse supernovae, can drive relativistic outflows, giving rise to non-thermal high-energy emission. High-energy neutrinos are signatures of such outflows. The detection of gravitational waves and high-energy neutrinos from common sources could help establish the connection between the dynamics of the progenitor and the properties of the outflow. We searched for associated emission of gravitational waves and high-energy neutrinos from astrophysical transients with minimal assumptions using data from Advanced LIGO from its first observing run O1, and data from the A ntares and IceCube neutrino observatories from the same time period. We focused on candidate events whose astrophysical origins could not be determined from a single messenger. We found no significant coincident candidate, which we used to constrain the rate density of astrophysical sources dependent on their gravitational-wave and neutrino emission processes.

Abstract: Persons with drug-resistant epilepsy may benefit from epilepsy surgery and should undergo presurgical testing to determine potential candidacy and appropriate intervention. Institutional expertise can influence use and availability of evaluations and epilepsy surgery candidacy. This census survey study aims to examine the influence of geographic region and other center characteristics on presurgical testing for medically intractable epilepsy.We analyzed annual report and supplemental survey data reported in 2020 from 206 adult epilepsy center directors and 136 pediatric epilepsy center directors in the United States. Test utilization data were compiled with annual center volumes, available resources, and US Census regional data. We used Wilcoxon rank-sum, Kruskal-Wallis, and chi-squared tests for univariate analysis of procedure utilization. Multivariable modeling was also performed to assign odds ratios (ORs) of significant variables.The response rate was 100% with individual element missingness 11% across 342 observations undergoing univariate analysis. A total of 278 complete observations were included in the multivariable models, and significant regional differences were present. For instance, compared to centers in the South, those in the Midwest used neuropsychological testing (OR = 2.87, 95% confidence interval [CI] = 1.2-6.86; p = .018) and fluorodeoxyglucose-positron emission tomography (OR = 2.74, 95% CI = = 1.14-6.61; p = .025) more commonly. For centers in the Northeast (OR = .46, 95% CI = .23-.93; p = .031) and West (OR = .41, 95% CI = .19-.87; p = .022), odds of performing single-photon emission computerized tomography were lower by nearly 50% compared to those in the South. Center accreditation level, demographics, volume, and resources were also associated with varying individual testing rates.Presurgical testing for drug-resistant epilepsy is influenced by US geographic region and other center characteristics. These findings have potential implications for comparing outcomes between US epilepsy centers and may inject disparities in access to surgical treatment.

Abstract: Background and ObjectiveNearly one-third of persons with epilepsy will continue having seizures despite trialing multiple antiseizure medications. Epilepsy surgery may be beneficial in these cases, and evaluation at a comprehensive epilepsy center is recommended. Numerous palliative and potentially curative approaches exist, and types of surgery performed may be influenced by center characteristics. This article describes epilepsy center characteristics associated with epilepsy surgery access and volumes in the United States.MethodsWe analyzed National Association of Epilepsy Centers 2019 annual report and supplemental survey data obtained with responses from 206 adult epilepsy center directors and 136 pediatric epilepsy center directors in the United States. Surgical treatment volumes were compiled with center characteristics, including US Census region. We used multivariable modeling with zero-inflated Poisson regression models to present ORs and incidence rate ratios of receiving a given surgery type based on center characteristics.ResultsThe response rate was 100% with individual element missingness less than 4% across 352 observations undergoing univariate analysis. Multivariable models included 319 complete observations. Significant regional differences were present. The rates of laser interstitial thermal therapy (LITT) were lower at centers in the Midwest (incidence rate ratio [IRR] 0.74, 95% CI 0.59–0.92;p= 0.006) and Northeast (IRR 0.77, 95% CI 0.61–0.96;p= 0.022) compared with those in the South. Conversely, responsive neurostimulation implantation rates were higher in the Midwest (IRR 1.45, 95% CI 1.1–1.91;p= 0.008) and West (IRR 1.91, 95% CI 1.49–2.44;p< 0.001) compared with the South. Center accreditation level, institution type, demographics, and resources were also associated with variations in access and rates of potentially curative and palliative surgical interventions.DiscussionEpilepsy surgery procedure volumes are influenced by US epilepsy center region and other characteristics. These variations may affect access to specific surgical treatments for persons with drug resistant epilepsy across the United States.

Abstract: Background and ObjectivesPersons with epilepsy, especially those with drug resistant epilepsy (DRE), may benefit from inpatient services such as admission to the epilepsy monitoring unit (EMU) and epilepsy surgery. The COVID-19 pandemic caused reductions in these services within the US during 2020. This article highlights changes in resources, admissions, and procedures among epilepsy centers accredited by the National Association of Epilepsy Centers (NAEC).MethodsWe compared data reported in 2019, prior to the COVID-19 pandemic, and 2020 from all 260 level 3 and level 4 NAEC accredited epilepsy centers. Data were described using frequency for categorical variables and median for continuous variables and were analyzed by center level, center population category, and geographical location. Qualitative responses from center directors to questions regarding the impact from COVID-19 were summarized utilizing thematic analysis. Responses from the NAEC center annual reports as well as a supplemental COVID-19 survey were included.ResultsEMU admissions declined 23% (-21,515) in 2020, with largest median reductions in level 3 centers [-55 admissions (-44%)] and adult centers [-57 admissions (-39%)]. The drop in admissions was more substantial in the East North Central, East South Central, Mid Atlantic, and New England US Census divisions. Survey respondents attributed reduced admissions to re-assigning EMU beds, restrictions on elective admissions, reduced staffing, and patient reluctance for elective admission. Treatment surgeries declined by 371 cases (5.7%), with the largest reduction occurring in VNS implantations [-486 cases (-19%)] and temporal lobectomies [-227 cases (-16%)]. All other procedure volumes increased, including a 35% (54 cases) increase in corpus callosotomies.DiscussionIn the US, access to care for persons with epilepsy declined during the COVID-19 pandemic in 2020. Adult patients, those relying on level 3 centers for care, and many persons in the eastern half of the US were most affected.

Abstract: Pregnancy presents a unique risk to chemical researchers due to their occupational exposures to chemical, equipment, and physical hazards in chemical research laboratories across science, engineering, and technology disciplines. Understanding "risk" as a function of hazard, exposure, and vulnerability, this review aims to critically examine the state of the science for the risks and associated recommendations (or lack thereof) for pregnant researchers in chemical laboratories (labs). Commonly encountered hazards for pregnant lab workers include chemical hazards (organic solvents, heavy metals, engineered nanomaterials, and endocrine disruptors), radiation hazards (ionizing radiation producing equipment and materials and nonionizing radiation producing equipment), and other hazards related to the lab environment (excessive noise, excessive heat, psychosocial stress, strenuous physical work, and/or abnormal working hours). Lab relevant doses and routes of exposure in the chemical lab environment along with literature and governmental recommendations or resources for exposure mitigation are critically assessed. The specific windows of vulnerability based on stage of pregnancy are described for each hazard, if available. Finally, policy gaps for further scientific research are detailed to enhance future guidance to protect pregnant lab workers.

Abstract: An effective (early) mobilization in COVID-19 intensive care patients with ECMO treatment is very important. Sedation, extracorporeal procedures with the danger of circuit malfunction, large lumen ECMO cannulas with a risk of dislocation and a very severe neuromuscular weakness are factors that could deem mobilization beyond stage 1 of the ICU mobility score (IMS) in some cases difficult or impossible; however, early mobilization is a key point of the ABCDEF bundle to counteract pulmonary complications, neuromuscular dysfunction and enable recovery. The case of a 53-year-old, previously healthy and active male patient with a severe and complicated course of COVID-19 and pronounced ICU-acquired weakness is described. While receiving ECMO the patient could be mobilized using a robotic system. Due to severe and rapidly progressing pulmonary fibrosis, additional low-dose methylprednisolone therapy (Meduri protocol) was implemented. Under this multimodal treatment the patient was successfully weaned from the ventilator and decannulated. Robotic-assisted mobilization has the potential to be a novel and safe therapeutic option for a customized and highly effective mobilization in ECMO patients.Die qualitativ hochwertige (Früh)Mobilisation ist auch bei schwer kranken (COVID-19-)Intensivpatienten mit ECMO-Therapie unbedingt erstrebenswert. Sedierung, blutflusssensibles extrakorporales Verfahren, großlumige ECMO-Kanülen mit Dislokationsrisiko und besonders eine bei COVID-19-Patienten häufig ausgeprägt auftretende neuromuskuläre Schwäche lassen eine Mobilisation über die Stufe 1 des IMS aber teilweise nicht zu. Dabei ist die Frühmobilisation ein wesentlicher Baustein eines Maßnahmenbündels (ABCDEF), um einer pulmonalen Funktionsstörung und neuromuskulärer Dysfunktion im Sinne einer ICUAW (intensiv care unit-acquired weakness) entgegenzuwirken und eine Regeneration zu ermöglichen.Es wird der Fall eines 53-jährigen, bis dato gesunden und sportlich aktiven Patienten geschildert, der bei schwerem und kompliziertem COVID-19-Verlauf mit ausgeprägter ICUAW bei noch laufender ECMO- und Katecholamintherapie mittels Mobilisationsrobotik behandelt werden konnte. Bei schwerem und rasch progredientem pulmonalem Fibrosierungsprozess gelangen unter zusätzlicher Anwendung einer niedrigdosierten, prolongierten Methylprednisolontherapie (Meduri-Schema) letztlich die Beatmungsentwöhnung und Dekanülierung.Robotische Assistenzsysteme könnten eine neuartige, praktikable und sichere Therapieoption für eine individuell angepasste, schonende und effektivitätssteigernde Mobilisation für diese Patientengruppe sein.

Abstract: Femtosecond lasers at fixed wavelengths above 1,000 nm are powerful, stable and inexpensive, making them promising sources for two-photon microscopy. Biosensors optimized for these wavelengths are needed for both next-generation microscopes and affordable turn-key systems. Here we report jYCaMP1, a yellow variant of the calcium indicator jGCaMP7 that outperforms its parent in mice and flies at excitation wavelengths above 1,000 nm and enables improved two-color calcium imaging with red fluorescent protein-based indicators. jYCaMP1, a yellow variant of the calcium indicator jGCaMP7, enables fast multicolor two-photon imaging at excitation wavelengths above 1,000 nm for use with popular ytterbium-doped fiber and modelocked semiconductor lasers.

Abstract: Individuals differ widely in their drug-craving behaviors. One reason for these differences involves sleep. Sleep disturbances lead to an increased risk of substance use disorders and relapse in only some individuals. While animal studies have examined the impact of sleep on reward circuitry, few have addressed the role of individual differences in the effects of altered sleep. There does, however, exist a rodent model of individual differences in reward-seeking behavior: the sign/goal-tracker model of Pavlovian conditioned approach. In this model, only some rats show the key behavioral traits associated with addiction, including impulsivity and poor attentional control, making this an ideal model system to examine individually distinct sleep-reward interactions. Here, we describe how the limbic neural circuits responsible for individual differences in incentive motivation overlap with those involved in sleep-wake regulation, and how this model can elucidate the common underlying mechanisms. Consideration of individual differences in preclinical models would improve our understanding of how sleep interacts with motivational systems, and why sleep deprivation contributes to addiction in only select individuals.

Abstract: Background and Objectives:Infantile spasms (IS) are early childhood seizures with potentially devastating consequences. Standard therapies (adrenocorticotropic hormone [ACTH], high-dose prednisolone, and vigabatrin) are strongly recommended as the first treatment for IS. While this recommendation comes without preference for one standard therapy over another, early remission rates are higher with hormone therapy (ACTH and high-dose prednisolone) when compared to vigabatrin. Using quality improvement (QI) methodology that included hormone therapy as the first treatment, we sought to increase the percentage of children with new onset non-tuberous sclerosis complex (TSC)-associated IS achieving 3-month electroclinical remission from a mean of 53.8% to >70%.Methods:Observational cohort study using a consecutive sample at a single academic tertiary care hospital comparing a prospective intervention cohort (5/2019 – 1/2022, N = 57) to a retrospective baseline cohort (11/2015 – 4/2019, N = 67). Our initiative addressed key drivers such as the routine use of vigabatrin over hormone therapy as first treatment and the common initiation of a second treatment after 14 days for initial non-responders. We included consecutive children without TSC presenting with new onset IS diagnosed and treated between 2 – 24 months of age. We displayed our primary outcome and process measures as control charts in which the centerline is the quarterly (previous three months) mean based on statistical process control methodology.Results:QI interventions that included the standardization of hormone therapy as the first treatment resulted in higher rates of 3-month remission, rising from 53.8% (baseline cohort) to 75.9% (intervention cohort). Process measure results included an increased rate of children receiving hormone therapy as first treatment (mean 44.6% to 100%) as well as a decreased number of days to both clinical follow-up after first treatment (mean of 16.3 to 12.6 days) and starting a second treatment within 14 days for initial non-responders (mean of 36.3 to 17.2 days).Discussion:For children with IS, improved rates of 3-month electroclinical remission can be achieved with QI methodology. Implementation of similar QI initiatives at other centers may likewise improve local remission rates.

Abstract: The astrophysical journal / 2 930, L24 (2022). doi:10.3847/2041-8213/ac67d8, We present the first comprehensive search for high-energy neutrino emission from high- and low-mass X-ray binaries conducted by IceCube. Galactic X-ray binaries are long-standing candidates for the source of Galactic hadronic cosmic rays and neutrinos. The compact object in these systems can be the site of cosmic-ray acceleration, and neutrinos can be produced by interactions of cosmic rays with radiation or gas, in the jet of a microquasar, in the stellar wind, or in the atmosphere of the companion star. We study X-ray binaries using 7.5 yr of IceCube data with three separate analyses. In the first, we search for periodic neutrino emission from 55 binaries in the Northern Sky with known orbital periods. In the second, the X-ray light curves of 102 binaries across the entire sky are used as templates to search for time-dependent neutrino emission. Finally, we search for time-integrated emission of neutrinos for a list of 4 notable binaries identified as microquasars. In the absence of a significant excess, we place upper limits on the neutrino flux for each hypothesis and compare our results with theoretical predictions for several binaries. Abstract: SUMMARYMonitoring spiking activity across large neuronal populations at behaviorally relevant timescales is critical for understanding neural circuit function. Unlike calcium imaging, voltage imaging requires kilohertz sampling rates which reduces fluorescence detection to near shot noise levels. High-photon flux excitation can overcome photon-limited shot noise but photo-bleaching and photo-damage restricts the number and duration of simultaneously imaged neurons. We investigated an alternative approach aimed at low two-photon flux, voltage imaging below the shot noise limit. This framework involved developing: a positive-going voltage indicator with improved spike detection (SpikeyGi); an ultra-fast two-photon microscope for kilohertz frame-rate imaging across a 0.4×0.4mm2 field of view, and; a self-supervised denoising algorithm (DeepVID) for inferring fluorescence from shot-noise limited signals. Through these combined advances, we achieved simultaneous high-speed, deep-tissue imaging of more than one hundred densely-labeled neurons over one hour in awake behaving mice. This demonstrates a scalable approach for voltage imaging across increasing neuronal populations.

We describe a flexible cellular resolution two-photon microscope capable of simultaneous video rate acquisition of four independently targetable brain regions spanning an approximate five-millimeter field of view. With this system, we demonstrate the ability to measure calcium activity across mouse sensorimotor cortex at behaviorally relevant timescales., Functional brain imaging with two-photon microscopy is limited by a tradeoff between imaging area and acquisition speed. Abstract: We address the problem of optimal size-selective exploitation in an age-structured fish population model by systematically examining how density and size dependency in growth, mortality and fecundity affect optimal harvesting patterns when judged against a set of fisheries objectives. The study offers five key insights. First, while minimum-length limits often maximize the biomass yield, exploitation using harvest slots (i.e. regulations that protect both immature and very large individuals) can generate within 95% of maximum yield; harvest slots also generally maximize the number of fish that are harvested. Second, density dependence in growth and size-dependent mortality predict more liberal optimal size limits than those derived under assumptions of no density and size dependence. Third, strong density dependence in growth maximizes the catch of trophy fish only when modest harvest is introduced; the same holds for numbers harvested, when the stock–recruitment function follows the Ricker type. Fourth, the inclusion of size-dependent maternal effects on fecundity or egg viability has only limited effects on optimal size limits, unless the increase in fecundity with mass (“hyperallometry”) is very large. However, large hyperallometry in fecundity shifts the optimal size limit for biomass yield from the traditional minimum-length limit to a harvest slot. Fifth, harvest slots generally provide the best compromises among multiple objectives. We conclude that harvest slots, or more generally dome-shaped selectivity to harvest, can outperform the standard minimum-length selectivity. The exact configuration of optimal size limits crucially depends on objectives, local fishing pressure, the stock–recruitment function, and the density and size dependency of growth, mortality and fecundity. Abstract: Mitigating the negative impacts of invasive lionfish (Pterois volitans/miles complex) is a top priority for marine reef fisheries management, with human removals considered the most viable approach to population control. Control efforts via diver spearfishing removals have annually removed tens of thousands of lionfish throughout their invasive range, but the effectiveness of removal efforts to remove 100% or achieve target lionfish densities in a given reef system has not been fully evaluated. Accounting for detection and removal efficacy is necessary for developing and evaluating lionfish management targets, as population- and community-level effects of lionfish removals may be diminished by undetected lionfish remaining in the system. This study quantified lionfish detection, catchability, and removal efficiency to evaluate the effectiveness of lionfish surveys and removal efforts on northern Gulf of Mexico (nGOM) artificial and natural reefs. Detection was assessed during crepuscular and midday time periods via diver and remotely operated vehicle (ROV) video surveys, with covariates for time of day and survey methodology assessed using generalized linear mixed models. Catchability and removal efficiency were estimated via depletion models based on serial removals via spearfishing on 6 artificial reefs and 9 natural reefs. A priori, we had expected lionfish detection to be higher during crepuscular periods given lionfish in the Caribbean and in their native range have been shown to forage more actively away from reefs then. However, we found lionfish detection was not significantly different between midday and crepuscular periods. Survey methodology affected detection, with 24% fewer lionfish being detected via ROV surveys versus diver surveys at artificial reefs and 72% fewer lionfish detected via ROV surveys at natural reefs. Therefore, density estimates on nGOM natural reefs, which constitute of >99% of the region’s habitat, may be higher than previously reported and problematic for lionfish management. Mean catchability for spearfishing lionfish was 0.88 on artificial reefs and 0.69 on natural reefs standardized for area. Abstract: Fisheries resources in the U.S. Gulf of Mexico (GoM) are under increasing pressure from both natural and anthropogenic stressors that have potentially broad effects on the ecosystem and introduce considerable uncertainty into management outcomes. To address these issues, more holistic, ecosystem-based tools are needed to inform decision-making. A Scoping Workshop with scientists, managers, and other stakeholders was held to identify and prioritize challenges in the GoM that could be addressed using ecosystem models, and how best to incorporate those models into the existing fisheries assessment and management framework. Challenges identified were associated with uncertainty in stock assessments, environmental stressors, multi-species reference points, invasive species, habitat effects, spatial management, and forage fisheries. Short-term priorities included those that address critical assumptions in stock assessments and inform imminent decision making, whereas long-term priorities were those associated with environmental stressors and novel management approaches. This information is intended to guide future ecosystem modeling efforts and help advance ecosystem based fisheries management in the region.

41. Atlantic tarpon (Megalops atlanticus) nursery habitats: evaluation of habitat quality and broad-scale habitat identification

Abstract: Coastal habitats are threatened worldwide by habitat loss and degradation. These habitats play a crucial role as fish nurseries. Unfortunately, it is difficult to determine the impact of habitat degradation for many species because data are lacking on early life history metrics including growth (0.07 ± 0.04 SE mm/day in this study), survival (apparent annual survival 0.007 (95% CI: 0.001–0.033 in this study), emigration (27% in this study) and the spatial extent and condition of these habitats. The juvenile life stage of Atlantic tarpon (Megalops atlanticus), an economically important species in the Caribbean, sub-tropical and tropical Atlantic, and Gulf of Mexico, depends upon wetlands and marshes. A mark-recapture study designed to measure juvenile tarpon growth in an altered mangrove habitat in Florida (USA) found that juvenile tarpon exhibited slow growth and emigrated at small sizes. The low scores on these metrics, in combination with a broad knowledge gap on the extent and condition of juvenile tarpon habitats in Florida, caused concern about the conservation prospects for tarpon and the fishery it supports. To provide information necessary to formulating an effective conservation plan for tarpon, we used citizen science to identify juvenile tarpon habitats and to characterize them as natural or altered (a first-level measure of direct, physical habitat change). A comparison of angler reports and habitat assessments with scientific field assessments showed that using anglers is an efficient and effective means of identifying juvenile tarpon habitats and providing a first-level assessment of habitat condition. This study provides a baseline for ongoing and future habitat conservation and restoration efforts for juvenile tarpon and other species that also use these habitats as nurseries.

Abstract: A recent time-integrated analysis of a catalog of 110 candidate neutrino sources revealed a cumulative neutrino excess in the data collected by IceCube between April 6, 2008 and July 10, 2018. This excess, inconsistent with the background hypothesis in the Northern hemisphere at the $3.3~\sigma$ level, is associated with four sources: NGC 1068, TXS 0506+056, PKS 1424+240 and GB6 J1542+6129. This letter presents two time-dependent neutrino emission searches on the same data sample and catalog: a point-source search that looks for the most significant time-dependent source of the catalog by combining space, energy and time information of the events, and a population test based on binomial statistics that looks for a cumulative time-dependent neutrino excess from a subset of sources. Compared to previous time-dependent searches, these analyses enable a feature to possibly find multiple flares from a single direction with an unbinned maximum-likelihood method. M87 is found to be the most significant time-dependent source of this catalog at the level of $1.7~\sigma$ post-trial, and TXS 0506+056 is the only source for which two flares are reconstructed. The binomial test reports a cumulative time-dependent neutrino excess in the Northern hemisphere at the level of $3.0~\sigma$ associated with four sources: M87, TXS 0506+056, GB6 J1542+6129 and NGC 1068.

Abstract: It has been argued that the dopaminergic system is involved in the attribution of motivational value to reward predictive cues as well as prediction error. To evaluate, dopamine neurons were recorded from male rats performing a Pavlovian approach task containing cues that have both “predictive” and “incentive” properties. All animals learned the predictive nature of the cue (illuminated lever entry into cage), but some also found the cue to be attractive and were motivated toward it (“sign-trackers,” STs). “Goal-trackers” (GTs) predominantly approached the location of reward receptacle. Rats were implanted with tetrodes for neural electrophysiological recordings in the ventral tegmental area. Cells were characterized by spike waveform shape and firing rate. Firing rates and magnitudes of responses in relation to Pavlovian behaviors, cue presentation, and reward delivery were assessed. We identified 103 dopamine and 141 nondopamine neurons. GTs and STs both showed responses to the initial lever presentation (CS1) and lever retraction (CS2). However, higher firing rates were sustained during the lever interaction period only in STs. Further, dopamine cells of STs showed a significantly higher proportion of cells responding to both CS1 and CS2. These are the first results to show that neurons from the VTA encode both predictive and incentive cues, support an important role for dopamine neurons in the attribution of incentive salience to reward-paired cues, and underscore the consequences of potential differences in motivational behavior between individuals.SIGNIFICANCE STATEMENTThis project serves to determine whether dopamine neurons encode differences in cued approach behaviors and incentive salience. How neurons of the VTA affect signaling through the NAcc and subsequent dopamine release is still not well known. All cues that precede a reward are predictive in nature. Some, however, also have incentive value, in that they elicit approach toward them. We quantified the attribution of incentive salience through cue approach behavior and cue interaction, and the corresponding magnitude of VTA neural firing. We found dopamine neurons of the VTA encode strength of incentive salience of reward cues. This suggests that dopamine neurons specifically in the VTA encode motivation.

Abstract: Journal of Instrumentation 15(06), P06032 (2020). doi:10.1088/1748-0221/15/06/P06032, We describe an improved in-situ calibration of the single-photoelectron charge distributions for each of the in-ice Hamamatsu Photonics R7081-02[MOD] photomultiplier tubes in the IceCube Neutrino Observatory. The characterization of the individual PMT charge distributions is important for PMT calibration, data and Monte Carlo simulation agreement, and understanding the effect of hardware differences within the detector. Abstract: Defensive system activation promotes heightened perception of threat signals, and excessive attention to threat signals has been discussed as a contributory factor in the etiology of anxiety disorders. However, a mechanistic account of attentional modulation during fear-relevant processes, especially during fear generalization remains elusive. To test the hypothesis that social fear generalization prompts sharpened tuning in the visuocortical representation of social threat cues, 67 healthy participants underwent differential fear conditioning, followed by a generalization test in which participants viewed faces varying in similarity with the threat-associated face. We found that generalization of social threat sharpens visuocortical tuning of social threat cues, whereas ratings of fearfulness showed generalization, linearly decreasing with decreasing similarity to the threat-associated face. Moreover, individuals who reported greater anxiety in social situations also showed heightened sharpened tuning of visuocortical neurons to facial identity cues, indicating the behavioral relevance of visuocortical tuning during generalization learning.

Abstract: Resilience thinking has generated much interest among scientific communities, yet most resilience concepts have not materialized into management applications. We believe that using resilience concepts to characterize systems and the social and ecological processes affecting them is a way to integrate resilience into better management decisions. This situation is exemplified by inland recreational fisheries, which represent complex socioecological systems that face unpredictable and unavoidable change. Making management decisions in the context of resilience is increasingly important given mounting environmental and anthropogenic perturbations to inland systems. Herein, we propose a framework that allows resilience concepts to be better incorporated into management by (i) recognizing how current constraints and management objectives focus on desired or undesired systems (specific fish and anglers), (ii) evaluating the state of a system in terms of how both social and ecological forces enforce or erode the desired or undesired system, (iii) identifying the resilience-stage cycles a system state may undergo, and (iv) determining the broad management strategies that may be viable given the system state and resilience stage. We use examples from inland recreational fisheries to illustrate different system state and resilience stages and synthesize several key results. Across all combinations of socioecological forces, five common types of viable management strategies emerge: (i) adopt a different management preference or focus, (ii) change stakeholder attitudes or behaviors via stakeholder outreach, (iii) engage in (sometimes extreme) biological intervention, (iv) engage in fishery intervention, and (v) adopt landscape-level management approaches focusing on achieving different systems in different waters. We then discuss the challenges and weaknesses of our approach, including specifically the cases in which there are multiple strong social forces (i.e., stakeholders holding competing objectives or values) and situations where waters are not readily divisible, such as rivers or great lakes, and in which spatial separation of competing objectives will be difficult. We end with our vision of how we believe these types of operationalized resilience approaches could improve or transform inland recreational fisheries management.

Abstract: Spearfishing is currently the primary approach for removing invasive Indo-Pacific lionfish (Pterois volitans/miles complex) to mitigate their impacts to western Atlantic marine ecosystems. However, a substantial portion of lionfish spawning biomass is beyond the depth limits of SCUBA. Innovative technologies may offer an alternative means to target lionfish and allow for the development of a deepwater lionfish trap fishery, but the removal efficiency and potential environmental impacts of traps have not been evaluated. We tested a collapsible non-containment trap (the ‘Gittings trap’) near artificial reefs in the northern Gulf of Mexico. A total of 327 lionfish and 28 native fishes (4 of which regulated species) recruited to traps (i.e., number of fish observed within the trap footprint) during 82 trap sets, catching 144 lionfish and 29 native species. Lionfish recruitment was highest for single (versus paired) traps deployed 10X higher for lionfish versus native fishes. Gittings traps did not move on the bottom during two major storm events; however, further testing will be necessary to test trap movement with surface buoys. Additional research should also focus on design and operational modifications to improve Gittings trap deployment success (68% successfully opened on the seabed) and reduce lionfish escapement (56% escaped from traps upon retrieval). While removal efficiency for lionfish (12–24%) was far below that of spearfishing, study results demonstrate Gittings traps are suitable for future testing on deepwater natural reefs, which constitute >90% of the region’s reef habitat.

Abstract: The Neutrino Mass Ordering (NMO) remains one of the outstanding questions in the field of neutrino physics. One strategy to measure the NMO is to observe matter effects in the oscillation pattern of atmospheric neutrinos above ∼1GeV, as proposed for several next-generation neutrino experiments. Moreover, the existing IceCube DeepCore detector can already explore this type of measurement. We present the development and application of two independent analyses to search for the signature of the NMO with three years of DeepCore data. These analyses include a full treatment of systematic uncertainties and a statistically-rigorous method to determine the significance for the NMO from a fit to the data. Both analyses show that the dataset is fully compatible with both mass orderings. For the more sensitive analysis, we observe a preference for normal ordering with a p-value of pIO= 15.3 % and CL s= 53.3 % for the inverted ordering hypothesis, while the experimental results from both analyses are consistent within their uncertainties. Since the result is independent of the value of δCP and obtained from energies Eν≳5GeV, it is complementary to recent results from long-baseline experiments.
