Abstract Background Since the beginning of SARS-CoV2 pandemic, the mortality rate among elderly patients (60–90 years) has been around 50%, so age has been a determining factor of a worse COVID-19 prognosis. Associated with age, the thymic function involution and depletion plays an important role, that could be related to a dysregulated and ineffective innate and adaptive immune response against SARS-CoV2. Our study aims to further in vitro effect of human Thymosin-alpha-1 (α1Thy) treatment on the immune system in population groups with different thymic function levels in the scenario of SARS-CoV2 infection. Results Activation markers such as CD40, CD80 and TIM-3 were upregulated in α1Thy presence, especially in plasmacytoid dendritic cells (pDCs) and, with increased TNFα production was observed compared to untreated condition. Co-cultures of CD4 + and CD8 + T cells with DCs treated with α1Thy in response to SARS-CoV2 peptides showed a decrease in the cytokine production compared to the condition without α1Thy pre-treated. A decrease in CD40L activation co-receptor expression in CD8 + LTs was also observed, as well as an increase in PD1 in CD4 + TLs expression in both age groups. In fact, there are no age-related differences in the immunomodulatory effect of the hormone, and it seems that effector memory and terminally differentiated memory T lymphocyte subsets were the most actively influenced by the immunomodulatory α1Thy effect. Finally, the polyfunctionality measured in SARS-CoV2 Specific-T cells response was maintained in α1Thy presence in total and memory subpopulations CD4 + and CD8 + T-cells, despite decreased proinflammatory cytokines production. Conclusion The hormone α1Thy could reduce, through the modulation of DCs, the amount of proinflammatory cytokines produced by T cells. Moreover, α1Thy improve lymphocyte functionality and could become a beneficial therapeutic alternative as an adjuvant in SARS-CoV2 treatment either in the acute phase after infection or reinfection. In addition, the effect on the T immune response means that α1Thy can be incorporated into the vaccination regimen, especially in the most immunologically vulnerable individuals such as the elderly. Abstract Background The absence of an effective treatment and vaccine in HIV-1 pandemic place preventive strategies such as safety and effective microbicide development as a central therapeutic approach to control HIV-1 pandemic nowadays. Results Studies of cytotoxicity, immune population status, inflammation or tissue damage and mainly prophylactic inhibition of HIV-1 infection in vaginal human explants demonstrate the biosafety and effectivity of G2-S16 dendrimer. Human explants treated with G2-S16 dendrimer or treated and HIV-1 infected do not presented signs of irritation, inflammation, immune activation or T cell populations deregulation. Conclusions Herein we conclude that G2-S16 dendrimer has demonstrated sufficient efficacy, biosafety, effectivity and behavior in the closest to the real-life condition model represented by the human healthy donor vaginal tissue explants, to raise G2-S16 dendrimer as a promising candidate to clinical trials to develop an effective microbicide against HIV-1 infection. The mucosa of the female reproductive tract must reconcile the presence of commensal microbiota and the transit of exogenous spermatozoa with the elimination of sexually transmitted pathogens. In the vagina, neutrophils are the principal cellular arm of innate immunity and constitute the first line of protection in response to infections or injury. Neutrophils are absent from the vaginal lumen during the ovulatory phase, probably to allow sperm to fertilize; however, the mechanisms that regulate neutrophil influx to the vagina in response to aggressions remain controversial. We have used mouse inseminations and infections of Neisseria gonorrhoeae, Candida albicans, Trichomonas vaginalis, and HSV-2 models. We demonstrate that neutrophil infiltration of the vaginal mucosa is distinctively contingent on the ovarian cycle phase and independent of the sperm and pathogen challenge, probably to prevent sperm from being attacked by neutrophils. Neutrophils extravasation is a multi-step cascade of events, which includes their adhesion through selectins (E, P and L) and integrins of the endothelial cells. We have discovered that cervical endothelial cells expressed selectin-E (SELE, CD62E) to favor neutrophils recruitment and estradiol down-regulated SELE expression during ovulation, which impaired neutrophil transendothelial migration and orchestrated sperm tolerance. Progesterone up-regulated SELE to restore surveillance after ovulation.

Abstract Background Chronic pelvic pain syndrome (CPPS) is a multifactorial disorder that affects 5.7% to 26.6% of women and 2.2% to 9.7% of men, characterized by hypersensitivity of the central and peripheral nervous system affecting bladder and genital function. People with CPPS have much higher rates of psychological disorders (anxiety, depression, and catastrophizing) that increase the severity of chronic pain and worsen quality of life. Myofascial therapy, manual therapy, and treatment of trigger points are proven therapeutic options for this syndrome. This study aims to evaluate the efficacy of capacitive resistive monopolar radiofrequency (CRMRF) at 448 kHz as an adjunct treatment to other physiotherapeutic techniques for reducing pain and improving the quality of life of patients with CPPS. Methods This triple-blind (1:1) randomized controlled trial will include 80 women and men with CPPS. Participants will be randomized into a CRMRF activated group or a CRMRF deactivated group and receive physiotherapeutic techniques and pain education. The groups will undergo treatment for 10 consecutive weeks. At the beginning of the trial there will be an evaluation of pain intensity (using VAS), quality of life (using the SF-12), kinesiophobia (using the TSK-11), and catastrophism (using the PCS), as well as at the sixth and tenth sessions. Discussion The results of this study will show that CRMRF benefits the treatment of patients with CPPS, together with physiotherapeutic techniques and pain education. These results could offer an alternative conservative treatment option for these patients. Trial registration ClinicalTrials.gov NCT03797911 . Registered on 8 January 2019.

Abstract Background The appearance of resistance against new treatments and the fact that HIV-1 can infect various cell types and develop reservoirs and sanctuaries makes it necessary to develop new therapeutic approaches to overcome those failures. Results Studies of cytotoxicity, genotoxicity, complexes formation, stability, resistance, release and particle size distribution confirmed that G2-SN15-PEG, G3-SN31-PEG, G2-SN15-PEG-FITC and G3-SN31-PEG-FITC dendrimers can form complexes with miRNAs being biocompatible, stable and conferring protection to these nucleic acids. Confocal microscopy and flow cytometry showed effective delivery of these four dendrimers into the target cells, confirming their applicability as delivery systems. Dendriplexes formed with the dendrimers and miRNAs significantly inhibited HIV-1 infection in PBMCs. Conclusions These dendrimers are efficient delivery systems for miRNAs and they specifically and significantly improved the anti-R5-HIV-1 activity of these RNA molecules. Graphic Abstract

This paper explores the education of prospective teachers with regard to the Theory of Didactical Situations when they engage in lesson study. We particularly focus on studying how a lesson study process oriented towards the Theory of Didactical Situations contributes to increasing prospective teachers' self-efficacy to plan and teach lessons based on such theory, reducing the gap between theory and practice. Prior to the study, we will discuss how the theoretical postulates assumed in lesson study affect crucial aspects of the process. The study is implemented with 47 prospective early childhood education teachers. We also consider another group of 47 prospective teachers that were engaged in practicum at the time the lesson study process took place. Using a quasi-experimental methodology based on a questionnaire developed ad hoc that captures the particularities of the didactic paradigm assumed, the results of our study show that both lesson study and practicum lead to a statistically significant increase in future teachers' self-efficacy to plan and teach lessons in line with the paradigm assumed. However, size effect measures show that the increase observed in the lesson study group is significantly higher, which supports the benefits of lesson study in initial teacher education. We discuss what the features of lesson study are that could be related to this increase. Finally, we sketch new lines of research connected with the benefits of lesson study versus other teacher education experiences like practicum, as well as with links between self-efficacy and knowledge growth in lesson study.

Abstract Background Human cytomegalovirus (HCMV) is a worldwide infection, causing different troublesome in immunosupressed patients and very related to Human Immunodeficiency Virus 1 (HIV-1) infection, mainly in developing countries, with a co-infection rate of 80% in Africa. The high cost of present treatments and the lack of routinely tests in these countries urge the necessity to develop new molecules or strategies against HCMV. The new treatments should be low-cost and capable of avoiding the emerging problem of resistant virus. Nanoparticles play an important role in several viral infections. Our main focus is to study the potential activity of polyanionic carbosilane dendrimers (PDC), which are hyperbranched molecules with several sulfonate or sulfate groups in their periphery, against different viruses. Results We studied the activity of G1-S4, G2-S16 and G2-S24P PDCs in MRC-5 cell line against HCMV infection by several plaque reduction assays. Our results show that dendrimers present good biocompatibility at the concentrations tested (1–50 µM) for 6 days in cell culture. Interestingly, both G2-S16 and G2-S24P showed a remarked inhibition at 10 µM against HCMV infection. Results on attachment and virucidal assays indicated that the inhibition was not directed to the virus or the virus-cell attachment. However, results of time of addition, showed a longer lasting activity of these dendrimers in comparison to ganciclovir, and the combination of G2-S16 or G2-S24P with ganciclovir increases the HCMV inhibition around 90 %. Conclusions Nanotechnology, in particular polyanionic carbosilane dendrimers, have proved their potential application against HCMV, being capable of inhibiting the infection by themselves or enhancing the activity of ganciclovir, the actual treatment. These compounds represent a low-cost approach to fight HCMV infections.

Abstract Background NRTIs-sparing regimens exert favourable profiles on T-cell homeostasis associated parameters. Our aim was to analyze the effect of NRTIs sparing regimen (NRTI-sparing-cART) vs NRTIs-containing regimen (NRTI-cART), on T-cell homeostasis associated parameters in naive HIV-infected patients. Methods Biomarkers of cell survival (CD127) and replicative senescence (CD57), were measured by multiparametric flow cytometry for T-cell phenotyping on peripheral blood mononuclear cells (PBMCs) samples just before (baseline) and after 48 weeks of undetectable viral load in patients on NRTI-sparing-cART (N = 13) and NRTI-cART (N = 14). After 48 weeks a subgroup of patients (n = 5) on NRTI-cART switched to NRTI-sparing-cART for another additional 48 weeks. In vitro assays were performed on PBMCs from HIV-uninfected healthy donors exposed or not to HIV. To analyze the independent factors associated with type of cART bivariate and stepwise multivariate analysis were performed after adjusting for basal CD4+, CD8+ and nadir CD4+ T-cell counts. Results After 48 weeks of a NRTI-sparing-cART vs NRTI-cART patients have higher effector memory (EM) CD4+ CD127+ T-cell levels, lower EM CD4+ CD57+ T-cell levels, higher CD8+ CD127+ T-cell levels, lower CD8+ CD57+ T-cell levels and higher memory CD8+ T-cell levels. This effect was confirmed in the subgroup of patients who switched to NRTI-sparing-cART. In vitro assays confirmed that the deleterious effect of a NRTIs-containing regimen was due to NRTIs. Conclusions The implementation of NRTI-sparing regimens, with a favourable profile in CD127 and CD57 T-cell expression, could benefit cART-patients. These results could have potential implications in a decrease in the number of Non-AIDS events.

Lung injuries are increasingly prevalent due to various diseases causing alveolar damage, potentially leading to respiratory disorders. This study employed an incubation method to develop nano-encapsulated ibuprofen within a chitosan matrix for targeted pulmonary therapy. The encapsulation was successful without altering the molecular structure of chitosan, and a 500 mg dose was identified as optimal through lung tissue cell viability and histological analysis. The controlled release mechanism of this formulation ensures targeted delivery to the lungs, reducing inflammation and promoting alveolar regeneration. This approach highlights the importance of dose optimization and presents a promising strategy to enhance the efficacy and safety of pulmonary treatments.

Introduction: Osteoarthritis is a disabling pathology characterised by joint pain and stiffness. A prevalence of coxarthrosis of 7.4% is reported in our country. Total hip joint replacement is indicated in advanced stages, a procedure that is not free of complications, the most frequent being prosthetic dislocation, which can be prevented with dual mobility systems. The following study aims to determine the rate of complications and clinical outcomes in dual mobility systems in primary coxarthrosis. Materials and methods: A retrospective study included 120 cases in 114 patients diagnosed with grade III coxarthrosis, mean age was 62.43 years, with a mean follow-up of 4.5 years. Joint replacement was performed by Hardinge approach. All cases were assessed clinically using the Harris Hip Score (HHS) and radiologically to demonstrate mid-term results. Results: The preoperative value on the HHS scale had a mean of 56.45, postoperative at one month 74.23; 6 months 85.40; 1 year 94.01 and at 5 years 94.84 points, representing a functional improvement of 17.78 postoperative month; 28.95 at 6 months postoperative; 37.56 at one year postoperative and 38.39 points at 5 years postoperative. A complication rate of 3.44%; 0.86% of complications were associated with the prosthetic components. Conclusion: The dual mobility system should be considered as a therapeutic option in primary hip joint replacement due to excellent functional results and low complication rates. Evidence level: IV. En nuestro país se reporta una prevalencia de coxartrosis del 7,4%. El reemplazo articular total de cadera está indicado en etapas avanzadas, procedimiento que no está exento de complicaciones, la más frecuente es la luxación protésica, que puede ser prevenida con sistemas de doble movilidad (DM). El siguiente estudio tiene como objetivo determinar la tasa de complicaciones y los resultados clínicos en los sistemas de DM en coxartrosis primaria. Materiales y métodos: Estudio retrospectivo donde se incluyeron 120 casos en 114 pacientes con diagnóstico de coxartrosis grado III, la media de edad fue de 62,43 años, con seguimiento medio de 4,5 años. El reemplazo articular se realizó mediante abordaje de Hardinge. Todos los casos fueron valorados clínicamente mediante la escala de Harris Hip Score (HHS) y radiológicamente para demostrar los resultados a mediano plazo. Resultados: El valor preoperatorio en la escala HHS tuvo una media de 56,45, posquirúrgica al mes 74,23; seis meses de 85,40; un año de 94,01 y a los cinco años de 94,84 puntos, representando una mejoría funcional de 17,78 mes posquirúrgico; 28,95 a los seis meses posquirúrgicos; 37,56 al año posquirúrgico y 38,39 puntos a los cinco años posquirúrgicos. Una tasa de complicaciones de 3,44%; 0,86% de las complicaciones se asociaron a los componentes protésicos. Conclusión: El sistema de DM debe ser considerado como una opción terapéutica en reemplazo articular primario de cadera debido a los resultados funcionales excelentes y bajas tasas de complicaciones. IntroductionIndividuals with subjective cognitive decline (SCD) express concern about self-perceived cognitive decline despite no objective impairment and are at higher risk of developing Alzheimer’s disease. Despite documented links between SCD and repetitive negative thinking (RNT), the specific impact of RNT on brain integrity and cognition in exacerbating the SCD condition remains unclear. We aimed to investigate the influence of RNT on global cognition and brain integrity, and their interrelationships among healthy middle-aged and older adults experiencing SCD.MethodsOut of 616 individuals with neuroimaging and neuropsychological data available, 89 (mean age = 56.18 years; 68.54% females) met SCD criteria. Eighty-nine non-SCD individuals matched by age, sex, and education were also selected and represented the control group (mean age = 56.09 years; 68.54% females). Global cognition was measured using the preclinical Alzheimer’s cognitive composite (PACC5), which includes dementia screening, episodic memory, processing speed, and category fluency tests. RNT was calculated through three questionnaires assessing intrusive thoughts, persistent worry, and rumination. We generated cortical thickness (CTh) maps and quantified the volume of white matter lesions (WML) in the whole brain, as grey and white matter integrity measures, respectively.ResultsSCD individuals exhibited higher RNT scores, and thinner right temporal cortex compared to controls. No differences were observed in PACC5 and WML burden between groups. Only the SCD group demonstrated positive associations in the CTh-PACC5, CTh-RNT, and WML-RNT relationships.DiscussionIn this cross-sectional study, RNT was exclusively associated with brain integrity in SCD. Even though our findings align with the broader importance of investigating treatable psychological factors in SCD, further research may reveal a modulatory effect of RNT on the relationship between cognition and brain integrity in SCD.

In Mediterranean oak-savannas (known as dehesas in Spain), it is important to better understand the interactions between vegetation structure and local climate regulation at scales relevant to farm management and the dominant hydrological regime. This study evaluated the water use patterns of dehesa vegetation patches (open grasslands, lowland grasslands with high soil moisture, tree + grass, and riparian vegetation), estimating actual evapotranspiration (ET). We used different models, previously validated in the area, that integrate remotely sensed data. They apply (a) a soil water balance (Kc-FAO56), (b) a surface energy balance (ALEXI/DisALEXI, and SEBS), and (c) a sharpening algorithm (STARFM), obtaining products at multiple spatial resolutions (30 m, 1 km, 5 km). The conceptual and operational differences between the methodologies reinforce the idea of a combined application of models. We demonstrated the need for high spatial and temporal resolution for on-farm livestock management due to the importance of the grasslands layer. This scale is crucial to determine the grass's emergence/drying cycle, which is key for livestock feeding planning. In humid/denser areas that provide essential ecosystem services (e.g., refuge, pasture rotation), transpiration rates are higher throughout the year and were underestimated when coarser spatial scale data was used. Over the typical system (grass with dispersed trees), the ET maps at low spatial resolution reflected the water use trends, and all models correlated well. Higher differences were found when comparing the models' performance over open grasslands.

Abstract During the preparation of fixed prosthesis (including individual bridges and crowns) it is important to select the materials that have the best features and properties to predict a successful clinical treatment. The objective of this study was to determine if the chemical and structural characteristics could cause to increase the fracture resistance, we used four bis-acryl resins Luxatemp, Protemp, Structur and Telio. Three-points bending by Flexural test were performed in ten bars and they were carried out to compare with Anova test. In addition, the bis-acryl resins were analyzed by scanning electron microscopy, to analyze microstructure and morphology and the molecular structure were performed by Infrared Spectroscopy through Attenuated Total Reflectance. A higher flexural strength was found in Luxatemp and Structur with, no significant differences between this study groups. Regarding Protemp and Telio, these study groups showed a lower flexural strength when were compared with Luxatemp and Structur. These results corroborate SEM and ATR analysis because Luxatemp sample showed a regular size particle on the surface and chemically presents a long cross-linkage polymer chain. The presence of CO3, SiO2 and N–H groups as a fillers particle interacting with OH groups cause a higher flexural strength compared with another groups.

Durante los últimos décadas ha habido un desarrollo de los intermetálicos, sobre todo por aplicaciones estructurales a alta temperatura en aplicaciones aeroespaciales, donde, por su rigidez alta, en combinación con una resistencia mecánica y ductilidad razonable, su buena estabilidad estructural y resistencia a la oxidación, han sido vistos como versiones avanzadas y mejoradas de las aleaciones metálicas como, por ejemplo, las superaleaciones a base de nitrógeno y las aleaciones de titanio. Se discute el desarrollo importante durante las últimas décadas, y también los nuevos desarrollos probables durante los próximos años. Se podrían ver los intermetálicos como versiones mejoradas de los cerámicos.

Durante los últimos años se ha prestado mucha atención a la familia de intermetálicos Fe-Al, puesto que estos constituyen un considerable potencial como materiales de ingeniería en aplicaciones a temperaturas intermedias o altas, sobre todo en casos donde se necesita alta resistencia a la oxidación o corrosión. A pesar del considerable esfuerzo desarrollado para obtener aleaciones con mejores propiedades, su resistencia mecánica a alta temperatura no es muy elevada. Se discutirán los aspectos que contribuyen a la baja resistencia mecánica a temperatura elevada en función de la estructura de dislocaciones y los mecanismos de anclaje que operan en este intermetálico. Se considerarán, también, maneras alternativas para mejorar la resistencia a temperatura elevada mediante la modificación de la microestructura y la incorporación de partículas de segunda fase.

Antecedentes: En los pacientes con lesiones de menisco se presentan de forma concomitante lesiones condrales en el 51% de los casos; la asociación entre la gravedad de la lesión condral y el mayor riesgo de presentar osteoartritis está claramente establecida. Sin embargo, hay escasa información acerca de las características que se asocian a la gravedad de la lesión condral en sujetos con lesiones de menisco mediales y laterales de forma aislada; subgrupo en el que el tratamiento preventivo no está establecido. Objetivo: Identificar las características del paciente con lesión aguda simple de menisco asociadas con la gravedad de la lesión condral. Método: En un estudio transversal se incluyeron sujetos con lesión aguda simple de menisco, se recabó la información de variables clínicas y se evaluó la gravedad de la lesión condral de acuerdo con los criterios de Outerbridge. Resultados: De 70 pacientes, 23 (32.9%) presentaban lesión de tipo I, 34 (48.6%) de tipo II y 13 (18.5%) de tipo III. Los sujetos mayores de 35 años (OR: 3.400; IC95%: 1.198-9.650) y aquellos con etiología degenerativa (OR: 9.333; IC95%: 1.144-76.161) mostraron mayor riesgo de presentar lesiones graves. Conclusiones: La gravedad de la lesión condral en sujetos con lesiones simples de menisco se asocia principalmente a la etiología.

Abstract The complexity of the agroecosystem can also be assessed by the different land uses in the system and the surroundings, being a relevant way to assess the heterogeneity of the landscape and the effects on the community of interest, in this case, entomofauna. Thus, the objective of this work is to verify how the use of soil in the surroundings of Chilean lettuce horticultural systems, in the Coquimbo Region, alters the entomological community of the crop. Insect sampling was conducted (February 2021 to March 2022) using yellow pan traps. Two sites will be sampled on each of the seven studied lettuce crops. Land use and land cover classes were defined: Forests, water bodies, shrub vegetation, grasslands, barren lands, impermeable surfaces, and urban areas. After land use and land cover classification, buffers of 500 to 5,000 m were created around each data collection point. For data analysis, the percentages of land use of different classes were compared with the ecological attributes: Abundance of insects, abundance of insect pests, richness of entomological families and types of oral apparatus (licker-sucker, mandible, picker-sucker, and sucker). Land uses at different distances from horticultural systems affected the entomological community.

IntroductionUnderstanding the impact of different lifestyle trajectories on health preservation and disease risk is crucial for effective interventions.MethodsThis study analyzed lifestyle engagement over five years in 3,013 healthy adults aged 40-70 from the Barcelona Brain Health Initiative using K-means clustering. Nine modifiable risk factors were considered, including cognitive, physical, and social activity, vital plan, diet, obesity, smoking, alcohol consumption, and sleep. Self-reported diagnoses of new diseases at different time-points after baseline allowed to explore the association between these five profiles and health outcomes.ResultsThe data-driven analysis classified subjects into five lifestyle profiles, revealing associations with health behaviors and risk factors. Those exhibiting high scores in health-promoting behaviors and low-risk behaviors, demonstrate a reduced likelihood of developing diseases (p < 0.001). In contrast, profiles with risky habits showed distinct risks for psychiatric, neurological, and cardiovascular diseases. Participant’s lifestyle trajectories remained relatively stable over time.DiscussionOur findings have identified risk for distinct diseases associated to specific lifestyle patterns. These results could help in the personalization of interventions based on data-driven observation of behavioral patterns and policies that promote a healthy lifestyle and can lead to better health outcomes for people in an aging society.

Introduction: Gamification consists of the use of games in non-playful contexts. It is widely employed in the motor rehabilitation of neurological diseases, but mainly in adult patients. The objective of this review was to describe the use of gamification in the rehabilitation of children and adolescents with neuromotor impairment. Methods: We performed a systematic review of clinical trials published to date on the MEDLINE (PubMed), Scielo, SCOPUS, Dialnet, CINAHL, and PEDro databases, following the PRISMA protocol. The methodological quality of the studies identified was assessed using the PEDro scale. Results: From a total of 469 studies, 11 clinical trials met the inclusion criteria. We analysed the gamification systems used as part of the rehabilitation treatment of different neuromotor conditions in children and adolescents. Cerebral palsy was the most frequently studied condition (6 studies), followed by developmental coordination disorder (3), neurological gait disorders (1), and neurological impairment of balance and coordination (1). Conclusion: The use of gamification in rehabilitation is helpful in the conventional treatment of neuromotor disorders in children and adolescents, with increased motivation and therapeutic adherence being the benefits with the greatest consensus among authors. While strength, balance, functional status, and coordination also appear to improve, future research should aim to determine an optimal dosage. Resumen: Introducción: La ludificación consiste en emplear el juego en contextos no lúdicos. Su uso en la rehabilitación motora de patologías neurológicas está muy extendido, pero sobre todo en pacientes adultos. El objetivo de esta revisión fue describir el uso de la ludificación en los tratamientos de rehabilitación en niños y adolescentes con afectación neuromotora. Métodos: Se realizó una revisión sistemática de ensayos clínicos en diferentes bases de datos: Medline (a través de Pubmed), Scielo, SCOPUS, Dialnet, Cinahl y PEDro de la literatura científica publicada hasta la fecha siguiendo el protocolo PRISMA. La calidad metodológica de los estudios identificados se evaluó a través de la escala PEDro. Resultados: De un total de 469 estudios localizados se seleccionaron 11 ensayos clínicos que cumplieron los criterios de inclusión. Se analizaron los sistemas de ludificación utilizados como parte del tratamiento rehabilitador en distintas afecciones neuromotoras en niños y adolescentes. La Parálisis Cerebral fue la afección con mayor número de estudios (n = 6) seguida del Trastorno del Desarrollo de la Coordinación (n = 3). También se estudió la alteración de la marcha por causa neurológica (n = 1) y la alteración del equilibrio y coordinación por causa neurológica (n = 1). Conclusión: El uso de la ludificación en rehabilitación aporta beneficios al tratamiento convencional de las alteraciones neuromotoras en niños y adolescentes siendo el incremento de la motivación y adherencia terapéutica los que mayor consenso han alcanzado entre autores. Fuerza, equilibrio, funcionalidad y coordinación son otras variables analizadas que, si bien sugieren mejoras, necesitarían futuras investigaciones para determinar una óptima dosificación.

Introduction. Vascular endothelial growth factor (VEGF), an angiogenic factor secreted by type II pneumocytes, could play a role in congenital diaphragmatic hernia (CDH) pathogenesis. Animal studies suggest that VEGF accelerates lung growth. Aim. To quantify VEGF on fetal lungs in a nitrofen rat model for CDH and to analyze the effect of tracheal occlusion (TO) in VEGF in fetal lung rats after nitrofen and in control rats not exposed to nitrofen. Methods. Pregnant rats received nitrofen on day 9.5 of gestation. Fetuses were divided into 2 groups: those that underwent TO on day 20 and those that did not. On day 21, fetuses were delivered, and the lungs were dissected for subsequent VEGF quantification. Results. CDH was detected in 43% of the fetuses that received nitrofen. Fetuses with CDH showed significantly reduced lung weight/fetal weight ratio and lower VEGF levels than the remainder. A higher VEGF value was observed after TO. Conclusions. VEGF protein was significantly lower in fetuses with CDH. TO induced a significant increase in VEGF compared to the fetuses that did not undergo TO. Although not statistically significant, we observed higher VEGF levels in fetuses with CDH and TO compared to fetuses with CDH and no further intervention.

Studies performed in North America, Africa, and South America have led to the isolation of four new species of Kastovskya, a filamentous cyanobacterial genus that before this manuscript had only one species, Kastovskya adunca from Chile. Kastovskya nitens and K. viridissima were isolated from soils on San Nicolas Island, K. sahariensis was isolated from hypolithic habitats from the Sahara Desert in Algeria, and K. circularithylacoides was isolated from hypolithic habitats in Chile. The molecular analyses are corroborated by morphological data, morphometric analysis, and ecological and biogeographical considerations for robust polyphasic descriptions of all taxa. The peculiar transatlantic distribution of this genus bears similarity to other taxa in recently published studies and is in agreement with a hypothesis suggesting that cyanobacteria in Africa may disperse to the Americas on dust particles during windstorms. This work is unusual in that species in a single rare cyanobacterial genus with a disjunct distribution are described simultaneously from three continents. The 16S rRNA gene analyses performed for this study also revealed that another recent genus, Arizonema, is clearly a later synonym of Symplocastrum. This issue is resolved here with the collapsing of the type species Arizonema commune into Symplocastrum flechtnerae.

Introduction Prosocial behaviors are voluntary behaviors that are performed for the benefit of other people and promote harmonious relationships with others. This type of enhanced behavior could reduce physical and verbal aggressive acts in adolescents. Objectives analyze the association between aggressive and prosocial behaviors in adolescents Methods The study was non-experimental of a transactional - correlational type, two evaluation instruments validated in the context were applied to 500 adolescents attending school in the department of Córdoba. The type of sampling was non-probabilistic. Results A Pearson correlation was performed, previously verifying the normality of the data, which showed a statistically significant, negative association between the prosocial behaviors and the aggressive behaviors of those evaluated (Table 1). Introduction Empathy is an essential skill in the doctor-patient relationship since it contributes to improve aspects of health care and patient satisfaction. Nevertheless, burnout research projects have been developed in recent years. Objectives To examine the predictive capacity that empathy has on burnout syndrome in health professionals. Methods A non-experimental, cross-sectional design was proposed. The type of study was correlational-descriptive since it was sought out to explore a functional relation through the prognosis of a criterion variable. Sample: 200 (100 female and 100 male). Results First, the variance of cognitive and Affective Empathy was dug out in the emotional exhaustation criterion scale. Results accounted for 15% of variability in emotional exhaustation. (Corrected R 2 = .15, F = 17,56, p = 0,00). The best predictor of emotional exhaustation refers to Cognitive Empathy. (B = -.27, p = 0.00). It does not seem that Affective Empathy acts as a predictor variable of Emotional Exhaustation. (Table 1). Introduction According to Bisquerra Alzina (2003),competencies are defined as a set of knowledge, capabilities, skills and attitudes, necessary to understand, express and regulate emotional phenomena appropriately and which are fundamental in the teaching profesion since they are closely related to students´performance and mental health. Objectives compare socio-emotional skills in two groups of participants: female and male Methods A non-experimental,cross-sectional design was proposed for this study. The scope of this research is descriptive, in the sense,that it seeks to establish measures in regard to specific variables. Sample (100 female and 100 male). Results Results revealed that the evaluated teachers show average level of socio-emotional competencies, (Table 1).The highest scores were encountered in relation to the optimism competence. It suggests that teachers have the ability to obtain favorable balances from adverse situations presented in their daily lives. Introduction Emotional competencies, according to Bisquerra Alzina & Escoda (2007), refer to “knowledge, skills and attitudes necessary to understand, express and appropriately regulate emotional phenomena” (p. 22) in the management of emotions with oneself and with the other. Objectives Measure the emotional competencies Empathy, Emotional Expression and Emotional Regulation in health professionals in healthcare centers. Methods Quantitative descriptive. The Inventory of Emotional Competencies for Adults (Mikulic, Crespi, Radusky, 2015) was applied to 30 participants (doctor, psychologist, nurse, dentist). Results The grouped measurements show skills at a medium and high level. Table 1. In the strategy for management, support, increase and suppression of the current affective state to self-soothe and find a state of relaxation, it is at an average value between medium and high (table 1.) In the ability to start and maintain conversations, express one’s own thoughts and feelings clearly, both in verbal and non-verbal communication, and demonstrate to others that they have been well understood, the level is mostly medium (table 1.) Conclusions The levels of emotional competencies evaluated are mostly in the middle in the assessment by dimensions, empathy registered a lower level in contrast to other dimensions.Taking into consideration professional practice, response to organic and mental human vulnerability, it is a field for promoting the well-being of the health professional. The activity-regulated cytoskeleton-associated protein (Arc) is a complex regulator of synaptic plasticity in glutamatergic neurons. Understanding its molecular function is key to elucidate the neurobiology of memory and learning, stress regulation, and multiple neurological and psychiatric diseases. The recent development of anti-Arc nanobodies has promoted the characterization of the molecular structure and function of Arc. This study aimed to validate two anti-Arc nanobodies, E5 and H11, as selective modulators of the human Arc N-lobe (Arc-NL), a domain that mediates several molecular functions of Arc through its peptide ligand binding site. The structural characteristics of recombinant Arc-NL-nanobody complexes were solved at atomic resolution using X-ray crystallography. Both anti-Arc nanobodies bind specifically to the multi-peptide binding site of Arc-NL. Isothermal titration calorimetry showed that the Arc-NL-nanobody interactions occur at nanomolar affinity, and that the nanobodies can displace a TARPγ2-derived peptide from the binding site. Thus, both anti-Arc-NL nanobodies could be used as competitive inhibitors of endogenous Arc ligands. Differences in the CDR3 loops between the two nanobodies indicate that the spectrum of short linear motifs recognized by the Arc-NL should be expanded. We provide a robust biochemical background to support the use of anti-Arc nanobodies in attempts to target Arc-dependent synaptic plasticity. Function-blocking anti-Arc nanobodies could eventually help unravel the complex neurobiology of synaptic plasticity and allow to develop diagnostic and treatment tools.

Background As the world population continues to age, the prevalence of neurological diseases, such as dementia, poses a significant challenge to society. Detecting cognitive impairment at an early stage is vital in preserving and enhancing cognitive function. Digital tools, particularly mHealth, offer a practical solution for large-scale population screening and prompt follow-up assessments of cognitive function, thus overcoming economic and time limitations. Objective In this work, two versions of a digital solution called Guttmann Cognitest ® were tested. Methods Two hundred and one middle-aged adults used the first version (Group A), while 132 used the second one, which included improved tutorials and practice screens (Group B). This second version was also validated in an older age group (Group C). Results This digital solution was found to be highly satisfactory in terms of usability and feasibility, with good acceptability among all three groups. Specifically for Group B, the system usability scale score obtained classifies the solution as the best imaginable in terms of usability. Conclusions Guttmann Cognitest ® has been shown to be effective and well-perceived, with a high potential for sustained engagement in tracking changes in cognitive function.

Fluid inclusions in stalagmites are becoming increasingly important for paleoclimate research. Within this framework, noble gas thermometry, based on noble gases dissolved in water from fluid inclusions, provides quantitative estimations of cave air paleotemperature. Two major issues of Noble Gas Temperature (NGT) determination on speleothems are (1) the potential lack of enough water for the analysis and (2) the presence of trapped gas not dissolved in water that can be released during the analysis from biphasic or all-gas fluid inclusions, as its contribution to the bulk noble gas signal can hinder NGT results. Although the step-crushing method helps to reduce the second issue, it also decreases the amount of water available for the calculations. In order to obtain reliable NGT results with low uncertainties, a major challenge is still to reach a balance between sufficient water for analysis and a small amount of “atmospheric” gas. The difficulty is that the extraction process cannot be standardized since it strongly depends on the type of sample. The objective of this work is to investigate how the characteristics of the speleothem can determine the adequacy of the extraction process. For this purpose, we consider a stalagmite from a Mediterranean cave that consists of columnar elongated calcite and contains a significant quantity of fluid inclusions, which suggests good potential for NGT analysis. Results, however, were poorly satisfactory. Trying to understand the source of the problems, an integrated study of petrography and petrophysical features was performed. The samples were found to be different depending on the stage of coalescence of crystals and thus separated into “open” and “closed” fabrics. Classic petrographic analysis and non-destructive (nuclear magnetic resonance) techniques were used to characterize the type and amount of fluid inclusions present in both types of fabrics. The study indicates that the closed fabric (total coalescence of calcite crystals) has most water trapped in water-filled, small intracrystalline fluid inclusions that usually contain very little gas. This fabric is very suitable for NGT determination, but since the amount of water is quite small, the sample should be crushed in only one step with a large number of beats to break all the inclusions. In contrast, samples with open fabric (partial coalescence of calcite crystals) contain a higher amount of water and, also, gas-filled large intercrystalline fluid inclusions. For this fabric, step-crushing of the sample is necessary. However, the low amount of water left for the second and third crushings could lead to flawed NGT results. Thus, we suggest modifying the method to get rid of part of the gas in the first crushing while leaving enough water for the following steps. This work shows the importance of characterizing speleothems and fluid inclusions, including their petrography and petrophysical characteristics, before starting NGT analysis, allowing the selection of the most favorable samples and the customization of the step-crushing procedure.

Experiments on ASDEX Upgrade (AUG) in 2021 and 2022 have addressed a number of critical issues for ITER and EU DEMO. A major objective of the AUG programme is to shed light on the underlying physics of confinement, stability, and plasma exhaust in order to allow reliable extrapolation of results obtained on present day machines to these reactor-grade devices. Concerning pedestal physics, the mitigation of edge localised modes (ELMs) using resonant magnetic perturbations (RMPs) was found to be consistent with a reduction of the linear peeling-ballooning stability threshold due to the helical deformation of the plasma. Conversely, ELM suppression by RMPs is ascribed to an increased pedestal transport that keeps the plasma away from this boundary. Candidates for this increased transport are locally enhanced turbulence and a locked magnetic island in the pedestal. The enhanced D-alpha (EDA) and quasi-continuous exhaust (QCE) regimes have been established as promising ELM-free scenarios. Here, the pressure gradient at the foot of the H-mode pedestal is reduced by a quasi-coherent mode, consistent with violation of the high-n ballooning mode stability limit there. This is suggestive that the EDA and QCE regimes have a common underlying physics origin. In the area of transport physics, full radius models for both L- and H-modes have been developed. These models predict energy confinement in AUG better than the commonly used global scaling laws, representing a large step towards the goal of predictive capability. A new momentum transport analysis framework has been developed that provides access to the intrinsic torque in the plasma core. In the field of exhaust, the X-Point Radiator (XPR), a cold and dense plasma region on closed flux surfaces close to the X-point, was described by an analytical model that provides an understanding of its formation as well as its stability, i.e., the conditions under which it transitions into a deleterious MARFE with the potential to result in a disruptive termination. With the XPR close to the divertor target, a new detached divertor concept, the compact radiative divertor, was developed. Here, the exhaust power is radiated before reaching the target, allowing close proximity of the X-point to the target. No limitations by the shallow field line angle due to the large flux expansion were observed, and sufficient compression of neutral density was demonstrated. With respect to the pumping of non-recycling impurities, the divertor enrichment was found to mainly depend on the ionisation energy of the impurity under consideration. In the area of MHD physics, analysis of the hot plasma core motion in sawtooth crashes showed good agreement with nonlinear 2-fluid simulations. This indicates that the fast reconnection observed in these events is adequately described including the pressure gradient and the electron inertia in the parallel Ohm’s law. Concerning disruption physics, a shattered pellet injection system was installed in collaboration with the ITER International Organisation. Thanks to the ability to vary the shard size distribution independently of the injection velocity, as well as its impurity admixture, it was possible to tailor the current quench rate, which is an important requirement for future large devices such as ITER. Progress was also made modelling the force reduction of VDEs induced by massive gas injection on AUG. The H-mode density limit was characterised in terms of safe operational space with a newly developed active feedback control method that allowed the stability boundary to be probed several times within a single discharge without inducing a disruptive termination. Regarding integrated operation scenarios, the role of density peaking in the confinement of the ITER baseline scenario (high plasma current) was clarified. The usual energy confinement scaling ITER98( p,y ) does not capture this effect, but the more recent H20 scaling does, highlighting again the importance of developing adequate physics based models. Advanced tokamak scenarios, aiming at large non-inductive current fraction due to non-standard profiles of the safety factor in combination with high normalised plasma pressure were studied with a focus on their access conditions. A method to guide the approach of the targeted safety factor profiles was developed, and the conditions for achieving good confinement were clarified. Based on this, two types of advanced scenarios (‘hybrid’ and ‘elevated’ q -profile) were established on AUG and characterised concerning their plasma performance.

In 2021 JET exploited its unique capabilities to operate with T and D–T fuel with an ITER-like Be/W wall (JET-ILW). This second major JET D–T campaign (DTE2), after DTE1 in 1997, represented the culmination of a series of JET enhancements—new fusion diagnostics, new T injection capabilities, refurbishment of the T plant, increased auxiliary heating, in-vessel calibration of 14 MeV neutron yield monitors—as well as significant advances in plasma theory and modelling in the fusion community. DTE2 was complemented by a sequence of isotope physics campaigns encompassing operation in pure tritium at high T-NBI power. Carefully conducted for safe operation with tritium, the new T and D–T experiments used 1 kg of T (vs 100 g in DTE1), yielding the most fusion reactor relevant D–T plasmas to date and expanding our understanding of isotopes and D–T mixture physics. Furthermore, since the JET T and DTE2 campaigns occurred almost 25 years after the last major D–T tokamak experiment, it was also a strategic goal of the European fusion programme to refresh operational experience of a nuclear tokamak to prepare staff for ITER operation. The key physics results of the JET T and DTE2 experiments, carried out within the EUROfusion JET1 work package, are reported in this paper. Progress in the technological exploitation of JET D–T operations, development and validation of nuclear codes, neutronic tools and techniques for ITER operations carried out by EUROfusion (started within the Horizon 2020 Framework Programme and continuing under the Horizon Europe FP) are reported in (Litaudon et al Nucl. Fusion accepted), while JET experience on T and D–T operations is presented in (King et al Nucl. Fusion submitted).

MAST-U is equipped with on-axis and off-axis neutral beam injectors (NBI), and these external sources of super-Alfvénic deuterium fast-ions provide opportunities for studying a wide range of phenomena relevant to the physics of alpha-particles in burning plasmas. The MeV range D-D fusion product ions are also produced but are not confined. Simulations with the ASCOT code show that up to 20% of fast ions produced by NBI can be lost due to charge exchange (CX) with edge neutrals. Dedicated experiments employing low field side (LFS) gas fuelling show a significant drop in the measured neutron fluxes resulting from beam-plasma reactions, providing additional evidence of CX-induced fast-ion losses, similar to the ASCOT findings. Clear evidence of fast-ion redistribution and loss due to sawteeth (ST), fishbones (FB), long-lived modes (LLM), Toroidal Alfvén Eigenmodes (TAE), Edge Localised Modes (ELM) and neoclassical tearing modes (NTM) has been found in measurements with a Neutron Camera (NCU), a scintillator-based Fast-Ion Loss Detector (FILD), a Solid-State Neutral Particle Analyser (SSNPA) and a Fast-Ion Deuterium- α (FIDA) spectrometer. Unprecedented FILD measurements in the range of 1–2 MHz indicate that fast-ion losses can be also induced by the beam ion cyclotron resonance interaction with compressional or global Alfvén eigenmodes (CAEs or GAEs). These results show the wide variety of scenarios and the unique conditions in which fast ions can be studied in MAST-U, under conditions that are relevant for future devices like STEP or ITER.

An overview is presented of the progress since 2021 in the construction and scientific programme preparation of the Divertor Tokamak Test (DTT) facility. Licensing for building construction has been granted at the end of 2021. Licensing for Cat. A radiologic source has been also granted in 2022. The construction of the toroidal field magnet system is progressing. The prototype of the 170 GHz gyrotron has been produced and it is now under test on the FALCON facility. The design of the vacuum vessel, the poloidal field coils and the civil infrastructures has been completed. The shape of the first DTT divertor has been agreed with EUROfusion to test different plasma and exhaust scenarios: single null, double null, X-divertor and negative triangularity plasmas. A detailed research plan is being elaborated with the involvement of the EUROfusion laboratories.

We present the first validated synthetic diagnostic for fast ion loss detectors (FILDs) in the Wendelstein 7-X (W7-X) stellarator. This model has been developed on, and validated against experimental data from, a FILD provided by the National Institute for Fusion Science (NIFS-FILD), with potential future applicability to the existing Faraday Cup FILD (FC-FILD) on W7-X as well as the scintillating FILD (S-FILD) currently under development. A workflow combining Monte Carlo codes BEAMS3D and ASCOT5 is used to track fast ions produced by neutral beam injection from the moment of ionization until they are thermalized or lost from the last closed flux surface, and from there to a virtual plane which serves as a projection of the entrance aperture to the FILD. Simulations in ASCOT5 are analyzed via a geometric method to determine the probability of transmission through the FILD aperture and onto the detector as a function of normalized momentum, pitch angle, gyrophase, and position at the virtual plane. This probability is then applied to the simulated ions arriving from the plasma, producing a simulated signal from a computationally tractable number of simulated fast ions. Simulated signals are presented for two W7-X experiments with neutral beam injection and quantitatively compared with experimental measurements from the NIFS-FILD diagnostic. An estimate of the frequency of charge-exchange with neutral particles in the edge is performed, and it is found that this process may have a significant impact on the measured signals.

Observations of enhanced fast-ion losses during edge localized modes (ELMs) have been reported in the ASDEX Upgrade tokamak, revealing losses above the injection energy. This suggests that fast ions can be accelerated and lost due to the ELMs. Recent analysis of the ELM-induced losses suggests that the fast ions are lost due to a resonant interaction with the electromagnetic perturbation during the ELM crash. The fast-ion transport and acceleration during ELMs is modelled using electromagnetic fields computed using the hybrid kinetic-MHD code MEGA, while fast-ion full orbits are tracked with the ASCOT code. Time-evolving 3D electromagnetic fields have been implemented in ASCOT to compute fast-ion orbits in the presence of fast MHD events such as ELMs. The simulations successfully reproduce a field-aligned pattern of the losses on the tokamak wall and the formation of an accelerated population in the lost fast-ion distribution, while they predict an accelerated population in the confined distribution. A parametric study of the fast-ion constants of motion suggests a resonant interaction between the fast-ions and the electromagnetic fields arising during the ELM crash. In the case of fast-ion acceleration, the perpendicular electric perturbation, with scales smaller than the fast-ion gyroradius, breaks magnetic moment conservation and resonantly modifies the fast-ion energy.

The impact of electron cyclotron current drive (ECCD)-driven current on toroidicity-induced Alfvén eigenmodes (TAEs) in experiments on the AUG tokamak is investigated numerically. The dynamical evolution of the plasma profiles and equilibria are modelled with European transport solver, while ion cyclotron resonance heating-accelerated H-minority ions exciting TAEs are assessed with the PION code. TAEs, their drive and damping are computed with the codes CASTOR and CASTOR-K. In the set of discharges analysed, two groups of TAEs are observed, differing in frequency and radial location. Experimental observations show that when counter-ECCD is applied the higher frequency group of approximately 150 kHz is suppressed, while the lower frequency modes of 125 kHz are amplified. When co-ECCD is applied, depending on the location of the ECCD current deposition layer, both groups of TAE can be suppressed. Numerical calculations of energetic particle drive and thermal plasma damping show that neither one effect could explain the variety of the experimental observations. The fine balance between the drive, sensitive to the TAE position, and the radiative and continuum damping effects could only explain the experiment if the effects are considered all together.
