Tropospheric ozone results from in situ chemical formation and stratosphere–troposphere exchange (STE), with the latter being more important in the middle and upper troposphere than in the lower troposphere. Ozone photochemical formation is nonlinear and results from the oxidation of methane and non-methane hydrocarbons (NMHCs) in the presence of nitrogen oxide (NOx=NO+NO2). Previous studies showed that O3 short- and long-term trends are nonlinearly controlled by near-surface anthropogenic emissions of carbon monoxide (CO), volatile organic compounds (VOCs), and nitrogen oxides, which may also be impacted by the long-range transport (LRT) of O3 and its precursors. In addition, several studies have demonstrated the important role of STE in enhancing ozone levels, especially in the midlatitudes. In this article, we investigate tropospheric ozone spatial variability and trends from 2005 to 2019 and relate those to ozone precursors on global and regional scales. We also investigate the spatiotemporal characteristics of the ozone formation regime in relation to ozone chemical sources and sinks. Our analysis is based on remote sensing products of the tropospheric column of ozone (TrC-O3) and its precursors, nitrogen dioxide (TrC-NO2), formaldehyde (TrC-HCHO), and total column CO (TC-CO), as well as ozonesonde data and model simulations. Our results indicate a complex relationship between tropospheric ozone column levels, surface ozone levels, and ozone precursors. While the increasing trends of near-surface ozone concentrations can largely be explained by variations in VOC and NOx concentration under different regimes, TrC-O3 may also be affected by other variables such as tropopause height and STE as well as LRT. Decreasing or increasing trends in TrC-NO2 have varying effects on TrC-O3, which is related to the different local chemistry in each region. We also shed light on the contribution of NOx lightning and soil NO and nitrous acid (HONO) emissions to trends of tropospheric ozone on regional and global scales.

Abstract In the present study, we developed and validated an experimental life support system (ELSS) designed to investigate coral reef associated bacterial communities. The microcosms in the ELSS consisted of coral reef sediment, synthetic seawater, and specimens of five benthic reef species. These included two hard corals Montipora digitata and Montipora capricornis, a soft coral Sarcophyton glaucum, a zoanthid Zoanthus sp., and a sponge Chondrilla sp.. Physicochemical parameters and bacterial communities in the ELSS were similar to those observed at shallow coral reef sites. Sediment bacterial evenness and higher taxonomic composition were more similar to natural-type communities at days 29 and 34 than at day 8 after transfer to the microcosms, suggesting microbial stabilization after an initial recovery period. Biotopes were compositionally distinct but shared a number of ASVs. At day 34, sediment specific ASVs were found in hosts and visa versa. Transplantation significantly altered the bacterial community composition of M. digitata and Chondrilla sp., suggesting microbial adaptation to altered environmental conditions. Altogether, our results support the suitability of the ELSS developed in this study as a model system to investigate coral reef associated bacterial communities using multi-factorial experiments.

Objective: We aimed at investigating the incidence and influencing factors of sarcopenia in patients with stage IIIB-IV non-small cell lung cancer (NSCLC). Patients and Methods: A total of 152 patients with IIIB-IV NSCLC were enrolled and their body composition were measured. Results: The incidence of sarcopenia in IIB-IV NSCLC patients was 40.13%, which related to age, diabetes mellitus, body mass index (BMI), and other body component. Besides, age and diabetes mellitus were independent risk factors, while high BMI was opposite. Conclusions: Patients with IIIB-IV NSCLC generally have decreased muscle mass, it’s necessary to early evaluate the muscle mass and to provide early nutritional intervention.

Moving target tracking technology based on Unmanned Aerial Vehicles (UAV) is widely used in many fields such as automatic inspection and emergency response. The existing moving target tracking methods usually have the problems of large computation and low tracking efficiency. Limited by the computing power of the UAV platform, real-time tracking and analysis of multiple targets based on the video data collected by UAV platform is a difficult task. In this paper, we proposed a novel Target Specific Filtering Tracking with Memory (TSFMTrack) method designed for UAV-based real-time tracking tasks, which involves a Tracklet Filtering Module (TFM) for capturing object appearance features and a Tracklet Matching Module (TMM) for bounding box association in each frame. By experimental comparison with other State-Of-The-Art (SOTA) methods on popular MOT and UAV tracking datasets, the TSFMTrack have shown obvious advantages in accuracy, computational efficiency and reliability. Furthermore, we deployed the TSFMTrack on the brain-inspired chip Lynchip KA200, the experimental results have shown that the TSFMTrack is effective on edge computational platform and suitable for UAV real-time tracking tasks.

The cloud condensation nuclei (CCN) activity of black carbon (BC) particles importantly determines their impacts on cloud microphysics and atmospheric lifetime. This process is crucially influenced by the number of hygroscopic coating materials that BC acquire during the ageing process. It remains a challenge for ambient measurements to capture this process and link this with CCN activity of BC. Here, we directly measured the droplet activation diameter (D50) and activation fraction of BC-containing (BCc) particles (Fact,BC) in suburban Beijing using coupled measurements of size-resolved number concentrations of CCN at configured water supersaturation (SS) and BCc particles. The number concentration of BCc particles was found to peak at diameter 180–210 nm after acquiring coatings, larger than that for all particles (50–150 nm). Consequently, the initially smaller BC particles become enlarged and more hygroscopic, thereby exhibiting CCN activities than other particles. The Fact,BC increased from 42 % to 69 % in number and from 67 % to 85 % in mass as SS increased from 0.1 % to 0.2 % but tended to reach a plateau when SS > 0.2 %. Notably, Fact,BC and D50 linearly correlated with equivalent photochemical age, at a rate of +2 % h−1 and −3 nm h−1, respectively. The results suggest BCc particles from anthropogenic sources can readily serve as CCN at a relatively low SS, and more than half of the BC population can be activated within a few hours, indicating that the surface-sourced BC can efficiently incorporate into clouds and potentially exert important indirect radiative impacts.

Background: Artificial urinary sphincter (AUS) placement remains the gold-standard treatment for post-prostatectomy urinary incontinence (PPUI), despite their need for periodic surgical revision. Objective: To understand the experiences of patients who undergo repeat AUS revisions. Design: Mixed design including quantitative surveys and qualitative interviews for thematic analysis. Methods: Men with ⩾2 revisions were collected from a single-institution, retrospective database of AUS patients. Participants were interviewed about their prostatectomy, incontinence, AUS placement, and revisions. A survey was administered utilizing validated tools (e.g., Decision Regret Scale (DRS), Incontinence Impact Questionnaire-7) for quantitative analysis. Interview transcripts were used for qualitative thematic analysis. Results: Of 26 respondents, 20 completed the interview. Twenty-three men completed the survey. The mean DRS score for prostatectomy was 24 (standard deviation (SD) = 27), indicating low regret. Median Incontinence Impact Questionnaire score was 54 (SD = 27), with 70% of participants describing their PPUI as “severe.” Participants experienced a significant decrease in daily pad usage with AUS placement (5.5 pre-AUS vs 1.4 post-AUS, p

Ferroelectric nematic liquid crystals exhibit unique non-linear optical properties, with the potential to become transformative materials for photonic applications. A promising direction relies on the fabrication of tailored polar orientational patterns via photoalignment, thus shaping the non-linear optical susceptibility through thin slabs of the ferroelectric fluid. Here, we explore the fabrication of 2D periodic SHG active arrays in ferroelectric nematic fluids, for different materials, cell thicknesses and motifs. Based on polarizing optical microscopy observations in combination with optical simulations, second harmonic generation microscopy and interferometry, the 3D structure of the motifs is revealed. Two different 2D periodic patterns are explored, showing that the balance between flexoelectric and electrostatic energy can lead to different domain structures, an effect which is rooted in the difference between the flexoelectric properties of the materials. It is shown that by combining the surface-inscribed alignment with different spontaneous degrees of twist, 2D SHG active arrays can be obtained in the micrometre scale, in which adjacent areas exhibit maximum SHG signals at opposite angles.

Summary:. Pediatric hand burns are difficult to treat, with thin tissue with critical structures close to the skin and the small scale of the anatomy of children’s hands. Additionally, pediatric burns can be challenging due to the concern for donor-site morbidity and the paucity of donor sites when reconstructing these wounds. In this report, we discuss the successful application of a piscine-derived acellular dermal matrix in a 13-month-old child with deep partial thickness and full-thickness burns to the right upper extremity. She had excellent long term cosmetic results and function by 3 years postoperatively, including full extension and flexion of all digits in her right hand.

Land–atmosphere (L–A) interactions are important for understanding convective processes, climate feedbacks, the development and perpetuation of droughts, heatwaves, pluvials, and other land-centered climate anomalies. Local L–A coupling (LoCo) metrics capture relevant L–A processes, highlighting the impact of soil and vegetation states on surface flux partitioning and the impact of surface fluxes on boundary layer (BL) growth and development and the entrainment of air above the BL. A primary goal of the Climate Process Team in the Coupling Land and Atmospheric Subgrid Parameterizations (CLASP) project is parameterizing and characterizing the impact of subgrid heterogeneity in global and regional Earth system models (ESMs) to improve the connection between land and atmospheric states and processes. A critical step in achieving that aim is the incorporation of L–A metrics, especially LoCo metrics, into climate model diagnostic process streams. However, because land–atmosphere interactions span timescales of minutes (e.g., turbulent fluxes), hours (e.g., BL growth and decay), days (e.g., soil moisture memory), and seasons (e.g., variability in behavioral regimes between soil moisture and latent heat flux), with multiple processes of interest happening in different geographic regions at different times of year, there is not a single metric that captures all the modes, means, and methods of interaction between the land and the atmosphere. And while monthly means of most of the LoCo-relevant variables are routinely saved from ESM simulations, data storage constraints typically preclude routine archival of the hourly data that would enable the calculation of all LoCo metrics. Here, we outline a reasonable data request that would allow for adequate characterization of sub-daily coupling processes between the land and the atmosphere, preserving enough sub-daily output to describe, analyze, and better understand L–A coupling in modern climate models. A secondary request involves embedding calculations within the models to determine mean properties in and above the BL to further improve characterization of model behavior. Higher-frequency model output will (i) allow for more direct comparison with observational field campaigns on process-relevant timescales, (ii) enable demonstration of inter-model spread in L–A coupling processes, and (iii) aid in targeted identification of sources of deficiencies and opportunities for improvement of the models.

An animal's skin provides a first point of contact with the sensory environment, including noxious cues that elicit protective behavioral responses. Nociceptive somatosensory neurons densely innervate and intimately interact with epidermal cells to receive these cues, however the mechanisms by which epidermal interactions shape processing of noxious inputs is still poorly understood. Here, we identify a role for dendrite intercalation between epidermal cells in tuning sensitivity of Drosophila larvae to noxious mechanical stimuli. In wild-type larvae, dendrites of nociceptive class IV da neurons intercalate between epidermal cells at apodemes, which function as body wall muscle attachment sites, but not at other sites in the epidermis. From a genetic screen we identified miR-14 as a regulator of dendrite positioning in the epidermis: miR-14 is expressed broadly in the epidermis but not in apodemes, and miR-14 inactivation leads to excessive apical dendrite intercalation between epidermal cells. We found that miR-14 regulates expression and distribution of the epidermal Innexins ogre and Inx2 and that these epidermal gap junction proteins restrict epidermal dendrite intercalation. Finally, we found that altering the extent of epidermal dendrite intercalation had corresponding effects on nociception: increasing epidermal intercalation sensitized larvae to noxious mechanical inputs and increased mechanically evoked calcium responses in nociceptive neurons, whereas reducing epidermal dendrite intercalation had the opposite effects. Altogether, these studies identify epidermal dendrite intercalation as a mechanism for mechanical coupling of nociceptive neurons to the epidermis, with nociceptive sensitivity tuned by the extent of intercalation.

Due to a high spatial angle resolution and low circuit cost of massive hybrid analog and digital (HAD) multiple-input multiple-output (MIMO), it is viewed as a valuable green communication technology for future wireless networks. Integrating the massive HAD-MIMO with direction of arrival (DOA) will provide an even ultra-high performance of DOA measurement, which can the fully-digital (FD) MIMO. However, phase ambiguity is a challenge issue for a massive HAD-MIMO DOA estimation. In this paper, we consider three parts: detection, estimation, and Cramer-Rao lower bound (CRLB). First, a multi-layer-neural-network (MLNN) detector is proposed to infer the existence of emitters. Then, a two-layer HAD (TLHAD) MIMO structure is proposed to estimate the DOA and eliminate phase ambiguity using only one time block. Simulation results show that the proposed MLNN detector is much better than both the existing generalized likelihood ratio test (GRLT) and the ratio of maximum eigen-value (Max-EV) to minimum eigen-value (R-MaxEV-MinEV) in terms of detection probability. Additionally, the proposed TLHAD structure can achieve the corresponding CRLB.

Abstract The aim of this study is to systematically appraise the evidence on available full thickness 3D gingival and mucosal models (3D culture in scaffold base system) and their application in periodontal and peri-implant research. This study involved a systematic review of twenty-two studies obtained from searching from five electronic databases: MEDLINE-OVID, EMBASE, EBSCOhost, Web of Science Core Collection and LILACS, as well as a hand search of eligible articles up to September 2022. A total of 2338 studies were initially identified, after removal of duplicates (573), abstracts/title selection (1765), and full text screening (95), twenty-two studies were included, thirty-seven models were identified. Several cellular markers were reported by the studies included. The expression of keratinocytes differentiation markers (K4, K5, K10, K13, K14, K16, K17, K18, K19, involucrin, laminin5), proliferation marker (Ki67, CD90), and vimentin, Type I, II and IV collagen produced by fibroblasts were investigated in thirty models. No quantitative analyses were performed, and results of the review confirmed a substantial level of heterogeneity across experiments. In conclusion, there is currently insufficient evidence to conclude that the available 3D gingival and mucosal models can entirely recapitulate the human gingival tissue/mucosa and provide a useful research tool for periodontal and peri-implant research. This review also highlighted the lack of a standardized protocol to construct and characterize 3D gingival models. A new protocol is proposed for the characterization of in vitro gingival models for future research.

The light-absorbing organic aerosol (OA), known as brown carbon (BrC), has important radiative impacts; however, its sources and evolution after emission remain to be elucidated. In this study, the light absorption at multiple wavelengths, mass spectra of OA and microphysical properties of black carbon (BC) were characterized at a typical suburban environment in Beijing. The absorption of BC is constrained by its size distribution and mixing state, and the BrC absorption is obtained by subtracting the BC absorption from the total aerosol absorption. Aerosol absorption was further apportioned to BC, primary BrC and secondary BrC by applying the least correlation between secondary BrC and BC. The multilinear regression analysis on the factorized OA mass spectra indicated that the OA from traffic and biomass burning emission contributed to primary BrC. Importantly, the moderately oxygenated OA (O / C = 0.62) was revealed to highly correlate with secondary BrC. These OA had higher nitrogen content, in line with the nitrogen-containing functional groups detected by the Fourier transform infrared spectrometer. The photochemical processes were found to reduce the mass absorption cross section (MAC) of primary OA, reducing its contribution to total absorption by 20 %, at the same time increasing MAC for secondary OA, which showed a 30 % enhancement in contribution to total absorbance, implying the concurrent whitening and darkening of BrC. This provides field evidence that the photochemically produced secondary nitrogen-containing OA can considerably compensate for some bleaching effect on the primary BrC, hereby causing radiative impacts.

The data of debris-flow events between 2004 and 2021 in the Shenmu area Taiwan are presented and discussed in this paper. A total of 20 debris-flow events were observed in 18 years. Debris flows in the Shenmu area usually occurred in the Aiyuzi Stream during the rainy season, May to September, and about once per year between 2009 and 2017. The rainfall thresholds from the observed data are proposed to be Imax⁡ (maximum hourly rainfall), R24 (24 h accumulated rainfall), and Rt (effective accumulated rainfall) of 9, 23, and 67.8 mm, respectively. The rainfall data also imply that the trend curves of intensity–duration (I–D) were different before and after 2009, which is due to the extreme rainfall event of Typhoon Morakot in 2009. The I–D curve obtained from the post-2009 data is proposed as the baseline of the debris-flow I–D relationship in the study area. The extreme rainfall event also influenced the flow speed (average 14.3 m s−1 before 2010 and 4.46 m s−1 after 2010) and the occurrence frequency of debris flows (1.83 times per year before 2009 and 0.75 times after 2009). Recent findings have indicated that the ground surface vibrational signals of debris flows are potentially useful for debris-flow early warning in terms of accumulated energy, and the characteristic frequency of debris flows in the study area was below 40 Hz. The dataset and the rainfall thresholds in this study permit comparison with other monitored catchments and are useful for global debris-flow datasets.

The COVID-19 pandemic, driven by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has spurred an urgent need for effective therapeutic interventions. The spike glycoprotein of the SARS-CoV-2 is crucial for infiltrating host cells, rendering it a key candidate for drug development. By interacting with the human angiotensin-converting enzyme 2 (ACE2) receptor, the spike initiates the infection of SARS-CoV-2. Linoleate is known to bind the spike glycoprotein, subsequently reducing its interaction with ACE2. However, the detailed mechanisms underlying the protein-ligand interaction remain unclear. In this study, we characterized the pathways of ligand dissociation and the conformational changes associated with the spike glycoprotein by using ligand Gaussian accelerated molecular dynamics (LiGaMD). Our simulations resulted in eight complete ligand dissociation trajectories, unveiling two distinct ligand unbinding pathways. The preference between these two pathways depends on the gate distance between two α-helices in the receptor binding domain (RBD) and the position of the N-linked glycan at N343. Our study also highlights the essential contributions of K417, N121 glycan, and N165 glycan in ligand unbinding, which are equally crucial in enhancing spike-ACE2 binding. We suggest that the presence of the ligand influences the motions of these residues and glycans, consequently reducing accessibility for spike-ACE2 binding. These findings enhance our understanding of ligand dissociation from the spike glycoprotein and offer significant implications for drug design strategies in the battle against COVID-19.

The ongoing destruction of habitats in the tropics accelerates the current rate of species extinction. Range-restricted species are exceptionally vulnerable, yet we have insufficient knowledge about their protection. Species’ current distributions, range sizes, and protection gaps are crucial to determining conservation priorities. Here, we identified priority range-restricted bird species and their conservation hotspots in the Northern Andes. We employed maps of the Area of Habitat (AOH), that better reflect their current distributions than existing maps. AOH provides unprecedented resolution and maps a species in the detail essential for practical conservation actions. We estimated protection within each species’ AOH and for the cumulative distribution of all 335 forest-dependent range-restricted birds across the Northern Andes. For the latter, we also calculated protection across the elevational gradient. We estimated how much additional protection community lands (Indigenous and Afro-Latin American lands) would contribute if they were conservation-focused. AOHs ranged from 8 to 141,000 km2. We identified four conservation priorities based on cumulative species richness: the number of AOHs stacked per unit area. These priorities are high-resolution mapped representations of Endemic Bird Areas for the Tropical Andes that we consider critically important. Protected areas cover only 31% of the cumulative AOH, but community lands could add 19% more protection. Sixty-two per cent of the 335 species have ranges smaller than their published estimates, yet IUCN designates only 23% of these as Threatened. We identified 50 species as top conservation priorities. Most of these concentrate in areas of low protection near community lands and at middle elevations where, on average, only 34% of the land is protected. We highlight the importance of collaborative efforts among stakeholders: governments should support private and community-based conservation practices to protect the region with the most range-restricted birds worldwide.

Osteosarcoma is a rare type of bone cancer, and half of the cases affect children and adolescents younger than 20 years of age. Despite intensive efforts to improve both chemotherapeutics and surgical management, the clinical outcome for metastatic osteosarcoma remains poor. Transforming growth factor β (TGF-β) is one of the most abundant growth factors in bones. The TGF-β signaling pathway has complex and contradictory roles in the pathogenesis of human cancers. TGF-β is primarily a tumor suppressor that inhibits proliferation and induces apoptosis of premalignant epithelial cells. In the later stages of cancer progression, however, TGF-β functions as a metastasis promoter by promoting tumor growth, inducing epithelial-mesenchymal transition (EMT), blocking antitumor immune responses, increasing tumor-associated fibrosis, and enhancing angiogenesis. In contrast with the dual effects of TGF-β on carcinoma (epithelial origin) progression, TGF-β seems to mainly have a pro-tumoral effect on sarcomas including osteosarcoma (mesenchymal origin). Many drugs that target TGF-β signaling have been developed: neutralizing antibodies that prevent TGF-β binding to receptor complexes; ligand trap employing recombinant Fc-fusion proteins containing the soluble ectodomain of either type II (TβRII) or the type III receptor ((TβRIII), preventing TGF-β from binding to its receptors; antisense nucleotides that reduce TGF-β expression at the transcriptional/translational level; small molecule inhibitors of serine/threonine kinases of the type I receptor (TβRI) preventing downstream signaling; and vaccines that contain cell lines transfected with TβRII antisense genes, or target furin convertase, resulting in reduced TGF-β signaling.TGF-β antagonists have been shown to have effects on osteosarcoma in vitro and in vivo. One of the small molecule TβRI inhibitors, Vactosertib, is currently undergoing a phase 1/2 clinical trial to evaluate its effect on osteosarcoma. Several phase 1/2/3 clinical trials have shown TGF-β antagonists are safe and well tolerated. For instance, Luspatercept, a TGF-β ligand trap, has been approved by the FDA for the treatment of anemia associated with myeloid dysplastic syndrome (MDS) with ring sideroblasts/mutated SF3B1 with acceptable safety. Clinical trials evaluating the long-term safety of Luspatercept are in process.

ABSTRACT: A large number of female goats are needed for the dairy goat industry; therefore, the development of a method to ensure the birth of more females than males in a single pregnancy will lead to economic benefits. Increasing the number of X-sperm would be an effective way to increase the proportion of female offspring. In this study, goat semen was incubated at pH 7.4 in alkaline diluent combined with resiquimod (R848) and the number of X-sperm was enriched by the swim-up method. The percentage of X-sperm was determined using the double TaqMan qPCR method. Sperm total motility, progressive motility, average path velocity, straight-line velocity, and curvilinear velocity were measured using a computer-aided sperm analysis system, and the functional parameters of the sperm plasma membrane, the acrosome, mitochondrial activity, ATP content, and reactive oxygen species levels were also measured. Lastly, the ratio of female embryos was determined by in vitro fertilization, and the number of female kids and the pregnancy rate of does was assessed by artificial insemination. The results showed that dilution of semen in an alkaline buffer containing R848 could enrich the number of X-sperm to 85.57% ± 3.27%. The progressive motility, average path velocity, straight-line velocity, curvilinear velocity, mitochondrial activity, and ATP level of the collected X-sperm-enriched semen were significantly reduced, but its total motility, plasma membrane, and acrosome were not affected. The in vitro fertilization experiments showed that the rate of female embryo production using X-sperm-rich seminal fluid could reach 83.25% (174/209), which was significantly higher than the proportion of female embryos in the control group, 47.71% ± 1.80% (104/218). As determined by artificial insemination, the number of female kids in the test group increased by 62.79% (243/387), which was significantly higher than that in the control group (47.65%, 193/405). There was no significant difference in pregnancy rate between the test group and the control group (71.71% vs. 78.48%). Therefore, this study demonstrated that use of a pH 7.4 diluent containing R848 is a simple and effective method of X-sperm enrichment for dairy goat production. Its application would allow does to produce more female offspring for herd expansion and milk production.

Summary: Tumor-associated macrophages (TAMs) are integral to the development of complex tumor microenvironments (TMEs) and can execute disparate cellular programs in response to extracellular cues. However, upstream signaling processes underpinning this phenotypic plasticity remain to be elucidated. Here, we report that concordant AXL-STAT3 signaling in TAMs is triggered by lung cancer cells or cancer-associated fibroblasts in the cytokine milieu. This paracrine action drives TAM differentiation toward a tumor-promoting “M2-like” phenotype with upregulation of CD163 and putative mesenchymal markers, contributing to TAM heterogeneity and diverse cellular functions. One of the upregulated markers, CD44, mediated by AXL-IL-11-pSTAT3 signaling cascade, enhances macrophage ability to interact with endothelial cells and facilitate formation of primitive vascular networks. We also found that AXL-STAT3 inhibition can impede the recruitment of TAMs in a xenograft mouse model, thereby suppressing tumor growth. These findings suggest the potential application of AXL-STAT3-related markers to quantitatively assess metastatic potential and inform therapeutic strategies in lung cancer.

ObjectivesFetal alcohol spectrum disorder (FASD) captures the broad range of emotional, cognitive, behavioral, and congenital abnormalities associated with maternal alcohol consumption, and women living in resource-limited settings may be higher risk. This study aims to examine knowledge, attitudes, practices, and beliefs (KAPB) of women in Leyte, The Philippines regarding prenatal alcohol consumption.MethodsOne hundred postpartum women were recruited from a birth cohort in Leyte. A prenatal alcohol use KAPB survey was constructed in Waray, the local language. The survey was administered in June-September 2019. Descriptive statistics, chi-squared test, and Fisher's exact test were used to analyze responses.ResultsSeventy-five percent of subjects reported drinking tuba, a local palm wine, during pregnancy. Most participants (75%) did not believe tuba contained alcohol. Vincristine is a widely used chemotherapeutic drug for the treatment of multiple malignant diseases that causes a dose-limiting peripheral neurotoxicity. There is no clinically effective preventative treatment for vincristine-induced sensory peripheral neurotoxicity (VIPN), and mechanistic details of this side effect remain poorly understood. We hypothesized that VIPN is dependent on transporter-mediated vincristine accumulation in dorsal root ganglion neurons. Using a xenobiotic transporter screen, we identified OATP1B3 as a neuronal transporter regulating the uptake of vincristine. In addition, genetic or pharmacological inhibition of the murine orthologue transporter OATP1B2 protected mice from various hallmarks of VIPN — including mechanical allodynia, thermal hyperalgesia, and changes in digital maximal action potential amplitudes and neuronal morphology — without negatively affecting plasma levels or antitumor effects of vincristine. Finally, we identified α-tocopherol from an untargeted metabolomics analysis as a circulating endogenous biomarker of neuronal OATP1B2 function, and it could serve as a companion diagnostic to guide dose selection of OATP1B-type transport modulators given in combination with vincristine to prevent VIPN. Collectively, our findings shed light on the fundamental basis of VIPN and provide a rationale for the clinical development of transporter inhibitors to prevent this debilitating side effect.

Since the first plasma realized in 2020, a series of key systems on HL-3 (known as HL-2M before) tokamak have been equipped/upgraded, including in-vessel components (the first wall, lower divertor, and toroidal cryogenic/water-cooling/baking/glow discharge systems, etc.), auxiliary heating system of 11 MW, and 28 diagnostic systems (to measure the plasma density, electron temperature, radiation, magnetic field, etc.). Magnet field systems were commissioned firstly for divertor plasma discharges. During the 2nd experimental campaign of HL-3 tokamak, several great progresses have been achieved. Firstly, the successful operation with plasma current larger than 1 MA was achieved under a divertor configuration. Secondly, the advanced divertor concept with two distinct snowflake configurations was realized. It is found that the distribution of ion saturation current and heat flux on bottom plate becomes wide due to magnetic surface expansion, demonstrating the advantage of such configuration in the heat flux mitigation. In addition, using the combination of NBI, ECRH and LHCD, the standard sawtoothing high confinement mode of megampere plasma was firstly accessed on the HL-3. The successful commissioning of HL-3 is beneficial for the initial operation of ITER.

The Earth's land surface features spatial and temporal heterogeneity over a wide range of scales below those resolved by current Earth system models (ESMs). State-of-the-art land and atmosphere models employ parameterizations to represent their subgrid heterogeneity, but the land–atmosphere coupling in ESMs typically operates on the grid scale. Communicating the information on the land surface heterogeneity with the overlying atmospheric boundary layer (ABL) remains a challenge in modeling land–atmosphere interactions. In order to account for the subgrid-scale heterogeneity in land–atmosphere coupling, we implement a new coupling scheme in the Energy Exascale Earth system model version 1 (E3SMv1) that uses adjusted surface variances and covariance of potential temperature and specific water content as the lower boundary condition for the atmosphere model. The new lower boundary condition accounts for both the variability of individual subgrid land surface patches and the inter-patch variability. The E3SMv1 single-column model (SCM) simulations over the Atmospheric Radiation Measurement (ARM) Southern Great Plain (SGP) site were performed to assess the impacts. We find that the new coupling parameterization increases the magnitude and diurnal cycle of the temperature variance and humidity variance in the lower ABL on non-precipitating days. The impacts are primarily attributed to subgrid inter-patch variability rather than the variability of individual patches. These effects extend vertically from the surface to several levels in the lower ABL on clear days. We also find that accounting for surface heterogeneity increases low cloud cover and liquid water path (LWP). These cloud changes are associated with the change in cloud regime indicated by the skewness of the probability density function (PDF) of the subgrid vertical velocity. In precipitating days, the inter-patch variability reduces significantly so that the impact of accounting for surface heterogeneity vanishes. These results highlight the importance of accounting for subgrid heterogeneity in land–atmosphere coupling in next-generation ESMs.

The clean wall condition in tokamak devices can be achieved by utilizing the active chemical property of lithium (Li). This also exposes Li to air contamination even when protected by inert gas. To determine whether air contamination affects the retention of hydrogen isotopes in Li, the desorption performances of deuterium (D2) in liquid Li with D/Li ratios of 4.1 mol.%, 2.5 mol.%, and 0.6 mol.% were compared. At a D/Li ratio as low as 0.6 mol.%, the results revealed a significant change in the desorption temperature of D2 by Li contaminants. The ratio of contaminants to D2 in Li was discovered to be important. In this regard, the influence of N2, O2, and H2O (accounting for 15 mol.% of D2) on the desorption performance of D2 in liquid Li was discussed. Both H2O and N2 preabsorbed in liquid Li could react with D2, leading to the changed desorption temperature of D2. Notably, the H2O present in Li consumed all of D2 via reaction, whereas the same amount of N2 as H2O could only react with a part of D2. However, the effect of O2 on the desorption temperature and the effect of impurities on the desorption amount of D2 were not obvious. The impacts of the findings of this work on the retention and extraction of hydrogen isotopes in Li may be further explored to prompt the application of Li in fusion devices.

Bacterial secondary metabolites exhibit diverse remarkable bioactivities and are thus the subject of study for different applications. Recently, the individual effectiveness of tripyrrolic prodiginines and rhamnolipids against the plant-parasitic nematode Heterodera schachtii, which causes tremendous losses in crop plants, was described. Notably, rhamnolipid production in engineered Pseudomonas putida strains has already reached industrial implementation. However, the non-natural hydroxyl-decorated prodiginines, which are of particular interest in this study due to a previously described particularly good plant compatibility and low toxicity, are not as readily accessible. In the present study, a new effective hybrid synthetic route was established. This included the engineering of a novel P. putida strain to provide enhanced levels of a bipyrrole precursor and an optimization of mutasynthesis, i.e., the conversion of chemically synthesized and supplemented monopyrroles to tripyrrolic compounds. Subsequent semisynthesis provided the hydroxylated prodiginine. The prodiginines caused reduced infectiousness of H. schachtii for Arabidopsis thaliana plants resulting from impaired motility and stylet thrusting, providing the first insights on the mode of action in this context. Furthermore, the combined application with rhamnolipids was assessed for the first time and found to be more effective against nematode parasitism than the individual compounds. To obtain, for instance, 50% nematode control, it was sufficient to apply 7.8 μM hydroxylated prodiginine together with 0.7 μg/ml (~ 1.1 μM) di-rhamnolipids, which corresponded to ca. ¼ of the individual EC50 values. In summary, a hybrid synthetic route toward a hydroxylated prodiginine was established and its effects and combinatorial activity with rhamnolipids on plant-parasitic nematode H. schachtii are presented, demonstrating potential application as antinematodal agents. Dynamic induction control (DIC) has proven to be an effective method of increasing the power output for a wind farm in both simulation studies and wind tunnel experiments. By pitching the blades of a wind turbine periodically, the recovery of the low-velocity wake is accelerated, thereby increasing the energy available to downstream turbines. The wake itself of a turbine operating with DIC has not yet been studied experimentally. This paper presents a wind tunnel experiment where the wake of a wind turbine under periodic excitation is investigated. Using three-dimensional particle image velocimetry, the velocity field behind the turbine was reconstructed. Analysis of the velocity fields indicated that the available power in the wake increases when using DIC. This increase was partially due to a lower average thrust force experienced by the turbine with DIC. However, a large difference was seen between measurement results and actuator disk theory, indicating enhanced recovery of the wake is contributing to the increased energy. Instantaneous measurements visualizing the development of blade tip vortices also showed how the location of vortex breakdown, which is directly related to re-energizing the wake, shifts over time with DIC. We believe this shifting location is contributing to the enhanced wake recovery of DIC, providing more energy to downstream wind turbines.

Ozone (O3) dry deposition is a major O3 sink. As a follow-up study of Huang et al. (2021), we quantify the impact of satellite soil moisture (SM) on model representations of this process when different dry-deposition parameterizations are implemented, based on which the implications for interpreting O3 air pollution levels and assessing the O3 impacts on human and ecosystem health are provided. The SM data from NASA's Soil Moisture Active Passive mission are assimilated into the Noah-Multiparameterization (Noah-MP) land surface model within the NASA Land Information System framework, semicoupled with Weather Research and Forecasting model with online Chemistry (WRF-Chem) regional-scale simulations covering the southeastern US. Major changes in the modeling system used include enabling the dynamic vegetation option, adding the irrigation process, and updating the scheme for the surface exchange coefficient. Two dry-deposition schemes are implemented, i.e., the Wesely scheme and a “dynamic” scheme, in the latter of which dry-deposition parameterization is coupled with photosynthesis and vegetation dynamics. It is demonstrated that, when the dynamic scheme is applied, the simulated O3 dry-deposition velocities vd and their stomatal and cuticular portions, as well as the total O3 fluxes Ft, are larger overall; vd and Ft are 2–3 times more sensitive to the SM changes due to the data assimilation (DA). Further, through case studies at two forested sites with different soil types and hydrological regimes, we highlight that, applying the Community Land Model type of SM factor controlling stomatal resistance (i.e., β factor) scheme in replacement of the Noah-type β factor scheme reduced the vd sensitivity to SM changes by ∼75 % at one site, while it doubled this sensitivity at the other site. Referring to multiple evaluation datasets, which may be associated with variable extents of uncertainty, the model performance of vegetation, surface fluxes, weather, and surface O3 concentrations shows mixed responses to the DA, some of which display land cover dependency. Finally, using model-derived concentration- and flux-based policy-relevant O3 metrics as well as their matching exposure–response functions, the relative biomass/crop yield losses for several types of vegetation/crops are estimated to be within a wide range of 1 %–17 %. Their sensitivities to the model's dry-deposition scheme and the implementation of SM DA are discussed.

Mural painting is one of the carriers expressing history and culture. Due to the natural and anthropogenic factors, the salt in mural painting and environment is enriched in the surface layer with temperature change. It will induce irreversible diseases such as crispy alkali, which is not conducive to the survival of mural painting in the present. An efficient and non-destructive method to detect salt in murals is of great importance. Therefore, we proposed a method to predict the soluble salt content of mural paintings based on hyperspectral techniques. First, simulated samples with different salt concentrations were measured by a special spectroradiometer to acquire their spectra. Next, breakpoint correction and average smoothing preprocessing are performed and the data set is divided. Then, the spectra were enhanced by continuum removal (CR) and the logarithm of reciprocal (LR). The salt concentration was correlated with the spectra to extract 10 characteristic bands. Finally, the salt content prediction model was established by simple linear regression (SLR) and multiple linear regression (MLR). The accuracy of the model was evaluated with the coefficient of determination R2, root mean square error RMSE, and relative percent deviation RPD. The experimental results show that the best inversion fit is based on the combination of the CR-MLR model at the strong correlation bands of 420nm, 584nm, and 2379nm (Calibration Set R2 = 0.846, RMSE = 0.138, and RPD = 3.240). This paper provides a new technical means for the non-destructive detection of salt content in murals.

Written information on the surface of cultural relics can record important historical events. Due to the influence of natural and human factors, the surface of cultural relics fades and the words are difficult to identify. Take advantage of the hyperspectral data image and spectral unity and wide spectral range, a cultural relics surface handwriting enhancement method based on manifold learning and mixed pixel decomposition was proposed. First, the minimum noise fraction (MNF) transformation was carried out on the hyperspectral image, and then the top 10 bands were selected for inverse MNF transformation to reduce noise of the hyperspectral image. Then, the reconstructed image was dimensionally reduced by locally linear embedding (LLE) to obtain a gray image with the maximum amount of information. At the same time, the spectral features of the handwriting and background area in the reconstructed image were analysed. The automatic target generation process (ATGP) was adopted for endmember extraction on the reconstructed image to identify the endmember of handwriting. The abundance map of handwriting area was obtained by the fully constrained least squares (FCLS). Finally, the gray image and the abundance map of the handwriting region were weighted together to obtain the handwriting enhanced image. The true color image was synthesized from the reconstructed image, Then the true color image and the handwriting enhancement image were fused to obtain the handwritting fusion image. The hyperspectral image of a faded text in Shuozhou City, Shanxi Province, China, was used as an example for verification, and the results showed that the method can effectively enhance the text on the surface of the artifacts.

The size-resolved physiochemical properties of aerosols determine their atmospheric lifetime, cloud interactions and the deposition rate on the human respiratory system; however most atmospheric composition studies tend to evaluate these properties in bulk. This study investigated size-resolved constituents of aerosols on mass and number basis, and their droplet activation properties, by coupling a suite of online measurements with an aerosol aerodynamic classifier (AAC) based on aerodynamic diameter (Da) in Pinggu, a suburb of Beijing. While organic matter accounted for a large fraction of mass, a higher contribution of particulate nitrate at larger sizes (Da>300 nm) was found under polluted cases. By considering the mixing state of refractory-black-carbon-containing particles (rBCc) and composition-dependent densities, aerosols including rBCc were confirmed to be nearly spherical at Da>300 nm. Importantly, the number fraction of rBCc was found to increase with Da at all pollution levels. The number fraction of refractory black carbon (rBC) is found to increase from ∼3 % at ∼90 nm to ∼15 % at ∼1000 nm, and this increasing rBC number fraction may be caused by the coagulation during atmospheric ageing. The droplet activation diameter at a water supersaturation of 0.2 % was 112±6 and 193±41 nm for all particles with Da smaller than 1 µm (PM1) and rBCc respectively. As high as 52±6 % of rBCc and 50±4 % of all PM1 particles in number could be activated under heavy pollution due to enlarged particle size, which could be predicted by applying the volume mixing of substance hygroscopicity within rBCc. As rBCc contribute to the quantity of aerosols at larger particle size, these thickly coated rBCc may contribute to the radiation absorption significantly or act as an important source of cloud condensation nuclei (CCN). This size regime may also exert important health effects due to their higher deposition rate.

Realistic simulation of the Earth's mean-state climate remains a major challenge, and yet it is crucial for predicting the climate system in transition. Deficiencies in models' process representations, propagation of errors from one process to another, and associated compensating errors can often confound the interpretation and improvement of model simulations. These errors and biases can also lead to unrealistic climate projections and incorrect attribution of the physical mechanisms governing past and future climate change. Here we show that a significantly improved global atmospheric simulation can be achieved by focusing on the realism of process assumptions in cloud calibration and subgrid effects using the Energy Exascale Earth System Model (E3SM) Atmosphere Model version 1 (EAMv1). The calibration of clouds and subgrid effects informed by our understanding of physical mechanisms leads to significant improvements in clouds and precipitation climatology, reducing common and long-standing biases across cloud regimes in the model. The improved cloud fidelity in turn reduces biases in other aspects of the system. Furthermore, even though the recalibration does not change the global mean aerosol and total anthropogenic effective radiative forcings (ERFs), the sensitivity of clouds, precipitation, and surface temperature to aerosol perturbations is significantly reduced. This suggests that it is possible to achieve improvements to the historical evolution of surface temperature over EAMv1 and that precise knowledge of global mean ERFs is not enough to constrain historical or future climate change. Cloud feedbacks are also significantly reduced in the recalibrated model, suggesting that there would be a lower climate sensitivity when it is run as part of the fully coupled E3SM. This study also compares results from incremental changes to cloud microphysics, turbulent mixing, deep convection, and subgrid effects to understand how assumptions in the representation of these processes affect different aspects of the simulated atmosphere as well as its response to forcings. We conclude that the spectral composition and geographical distribution of the ERFs and cloud feedback, as well as the fidelity of the simulated base climate state, are important for constraining the climate in the past and future.

Aerosol–planetary boundary layer (PBL) interaction has been proposed as a key mechanism for stabilizing the atmosphere and exacerbating surface air pollution. Although the understanding of this process has progressed enormously, its magnitude and impact remain uncertain and vary widely concerning aerosol types, vertical distributions, synoptic conditions, etc. In this study, our primary interest is to distinguish the aerosol–PBL interaction of absorbing and scattering aerosols under contrasting synoptic patterns and aerosol vertical distributions. Detailed in situ aircraft (King Air 350) measurements and online coupled model Weather Research and Forecasting with Chemistry (WRF-Chem) simulations are explored over the North China Plain (NCP). Furthermore, a long-term PBL stability trend from 1980 to 2020 over the NCP is also investigated. The aircraft measurements and surface observations show that the surface air pollution over the city of Baoding on 3 January is heavier than that on 4 January 2020. In addition, the aerosols are restricted to the low layer on 3 January, whereas the aerosols mix more homogeneously in an upwards direction on 4 January. Thus, we focus on the 2 d with distinct synoptic circumstances, PBL stability, and aerosol vertical distributions over the NCP. According to the WRF-Chem modeling, the synoptic pattern over Baoding differs between the 2 d. The prevailing wind direction is opposite with a southwesterly wind on 3 January and a northeasterly wind on 4 January. The results indicate that the synoptic condition may affect the PBL thermal structure, thus affecting the aerosol vertical distribution. Additionally, the sensitive numerical experiments reveal that the light-absorbing and light-scattering aerosols have different effects on altering the PBL thermal structure. The inhibition effect of scattering aerosols on the PBL appears to be independent of the aerosol height distribution and solely depends on its concentration. However, the aerosol–PBL feedback of absorbing aerosols is highly dependent on its vertical distribution. Besides the 2 d case investigation, the analysis of the modeling results for nearly 1 month from 3 to 30 January 2020 in Baoding yields a more robust and representative conclusion. Our analysis highlights that we should principally concentrate on controlling the emissions of scattering aerosols under the stable stratification, while cooperating to control the emissions of scattering and absorbing aerosols in an unstable stratification. Moreover, the long-term interannual variation in the PBL stability shows a strong correlation with the East Asian winter monsoon, which seems to be valuable in determining which pollutants to target in different monsoon years and attaining more precise air pollution control. Based on the numerical simulations and observational constraints, a concept scheme description has been concluded to deepen our recognition of the interactions between thermodynamic stability and aerosols within the PBL over the NCP region.

Real-time wall conditioning and edge-localized mode (ELM) suppression through real-time boron powder injection into high-confinement mode discharges of the Experimental Advanced Superconducting Tokamak (EAST) have been investigated. The boron powder injection effectively conditioned the plasma-facing components, as shown by the reductions in low-Z and high-Z impurities and the global recycling level in the whole inner vessel, and suppressed the strong magnetohydrodynamic (MHD) activity in the plasma core. The improved wall conditions also resulted in an earlier transition from L- to H-mode. Boron powder injection also effectively suppressed ELM in the lower single null configuration with BT in the favorable direction while leaving the plasma density and stored energy largely unaffected.

We hereby present a simple process, called in situ two-end diffusion (TED), to reach coercivity enhancement with almost unchanged energy product for hot deformed (HD) NdFeB magnets. Coercivity of the HD magnets is increased from 15.1 to 18.7 kOe for the magnet TED with (Nd0.75Pr0.25)70Cu30 (NdPrCu), which is larger than 17.6 kOe for the magnet directly doped with 2 wt. % NdPrCu alloy. Besides, the magnet TED with NdPrCu exhibits good (00L) texture and therefore could support high (BH)max of 43.0 MGOe. In addition, the magnet TED with Ce70Cu30 (CeCu) exhibit superior magnetic properties of Br = 12.8 kG, iHc = 18.0 kOe and (BH)max = 40.7 MGOe to those of traditional doping with CeCu (Br = 11.7 kG, iHc = 16.4 kOe and (BH)max = 34.2 MGOe). For TED process, low-melting CeCu can infiltrate into the center of the magnet, and most Ce distribute at grain boundary. The distribution of Ce and Cu at grain boundary can reduce the magnetization of grain boundary phase, strengthen the decoupling effect between grains and contribute to the coercivity enhancement. This work demonstrates TED with low-melting HRE-free alloys is a simple method to effectively enhance coercivity and keep high Br and (BH)max for HD NdFeB magnets.
