Abstract

Objective Systemic lupus is a chronic autoimmune disease characterised by its phenotypic heterogeneity. Neutropaenia is a frequent event in SLE occurring in 20%–40% of patients depending on the threshold value of neutrophil count. On a daily basis, the management of neutropaenia in SLE is difficult with several possible causes. Moreover, the infectious consequences of neutropaenia in SLE remain not well defined.Methods 998 patients from the Lupus BioBank of the upper Rhein (LBBR), a large German and French cohort of patients with SLE, mostly of Caucasian origin (83%), were included in this study. Neutropaenia was considered when neutrophil count was below 1800×106/L. An additional analysis of detailed medical records was done for 65 LBBR patients with neutropaenia.Results 208 patients with neutropaenia (21%) were compared with 779 SLE patients without neutropaenia. Neutropaenia in SLE was significantly associated with thrombocytopaenia (OR 4.11 (2.57–10.3)), lymphopaenia (OR 4.41 (2.51–11.5)) and low C3 (OR 1.91 (1.03–4.37)) in multivariate analysis. 65 representative patients with neutropaenia were analysed. Neutropaenia was moderate to severe in 38%, chronic in 31%, and both severe and chronic in 23% of cases. Moderate to severe and chronic neutropaenia were both associated with lymphopaenia and thrombopaenia. Chronic neutropaenia was also associated anti-Ro/SSA antibodies and moderate to severe neutropaenia with oral ulcers.Conclusion This study is to date the largest cohort to describe neutropaenia in SLE. Neutropaenia displays a strong association with other cytopaenias, suggesting a common mechanism. Chronic neutropaenia is associated with anti-Ro/SSA antibodies with or without identified Sjögren’s disease.

Abstract

We show that a hierarchy of topological phases in one dimension—a topological Devil’s staircase—can emerge at fractional filling fractions in interacting systems, whose single-particle band structure describes a topological or a crystalline topological insulator. Focusing on a specific example in the BDI class, we present a field-theoretical argument based on bosonization that indicates how the system, as a function of the filling fraction, hosts a series of density waves. Subsequently, based on a numerical investigation of the low-lying energy spectrum, Wilczek–Zee phases, and entanglement spectra, we show that they are symmetry protected topological phases. In sharp contrast to the non-interacting limit, these topological density waves do not follow the bulk-edge correspondence, as their edge modes are gapped. We then discuss how these results are immediately applicable to models in the AIII class, and to crystalline topological insulators protected by inversion symmetry. Our findings are immediately relevant to cold atom experiments with alkaline-earth atoms in optical lattices, where the band structure properties we exploit have been recently realized.

Abstract

We study the equilibrium properties of the one-dimensional disordered Bose–Hubbard model by means of a gauge-adaptive tree tensor network variational method suitable for systems with periodic boundary conditions. We compute the superfluid stiffness and superfluid correlations close to the superfluid to glass transition line, obtaining accurate locations of the critical points. By studying the statistics of the exponent of the power-law decay of the correlation, we determine the boundary between the superfluid region and the Bose glass phase in the regime of strong disorder and in the weakly interacting region, not explored numerically before. In the former case our simulations are in agreement with previous Monte Carlo calculations.

Abstract

Abstract The eruption of the Hunga-Tonga volcano in the South Pacific Ocean on January 15, 2022, at about 4:15 UTC, generated a violent explosion, which created atmospheric pressure disturbances in the form of Rayleigh-Lamb waves detected all over the globe. Here we discuss the observation of the Hunga-Tonga shock-wave performed at the Ny-Ålesund Research Station on the Spitsbergen island, by the detectors of the PolarquEEEst experiment and their ancillary sensors. Online pressure data as well as the results of dedicated offline analysis are presented and discussed in details. Results include wave arrival times, wave amplitude measurements and wave velocity calculation. We observed five passages of the shock wave with a significance larger than 3 $$\sigma$$ σ and an amplitude up to 1 hPa. The average propagation velocity resulted to be (308 ± 0.6) m/s. Possible effects of the atmospheric pressure variation associated with the shock-wave multiple passages on the cosmic-ray rate at ground level are also investigated. We did not find any significant evidence of this effect.

Abstract

Abstract This paper describes the simulation framework of the extreme energy events (EEE) experiment. EEE is a network of cosmic muon trackers, each made of three multi-gap resistive plate chambers (MRPC), able to precisely measure the absolute muon crossing time and the muon integrated angular flux at the ground level. The response of a single MRPC and the combination of three chambers have been implemented in a GEANT4-based framework (GEMC) to study the telescope response. The detector geometry, as well as details about the surrounding materials and the location of the telescopes have been included in the simulations in order to realistically reproduce the experimental set-up of each telescope. A model based on the latest parametrization of the cosmic muon flux has been used to generate single muon events. After validating the framework by comparing simulations to selected EEE telescope data, it has been used to determine detector parameters not accessible by analysing experimental data only, such as detection efficiency, angular and spatial resolution.

Abstract

Objective: To use Multi-Criteria Decision Analysis (MCDA) to determine weights for eleven criteria in order to prioritize COVID-19 non-critical patients for admission to hospital in healthcare settings with limited resources. Methods: The MCDA was applied in two main steps: specification of criteria for prioritizing COVID-19 patients (and levels within each criterion); and determination of weights for the criteria based on experts’ knowledge and experience in managing COVID-19 patients, via an online survey. Criteria were selected based on available COVID-19 evidence with a focus on low- and middle-income countries (LMICs). Results: The most important criteria (mean weights, summing to 100%) are: PaO2 (16.3%); peripheral O2 saturation (15.9%); chest X-ray (14.1%); Modified Early Warning Score-MEWS (11.4%); respiratory rate (9.5%); comorbidities (6.5%); living with vulnerable people (6.4%); body mass index (5.6%); duration of symptoms before hospital evaluation (5.4%); CRP (5.1%); and age (3.8%). Conclusions: At the beginning of a new pandemic, when evidence for disease predictors is limited or unavailable and effective national contingency plans are difficult to establish, the MCDA prioritization model could play a pivotal role in improving the response of health systems.

Abstract

Abstract The goal of the PolarquEEEst experiment was to measure the cosmic charged particle rate at latitudes greater than 66$$^{\circ }$$ ∘ N, where no systematic and accurate measurements at sea level have ever been performed. A latitude range well above the Arctic Circle was explored on board of a sailboat, up to the unprecedented northernmost value of $$82^{\circ }07^{\prime }$$ 82∘07′ N. In this paper a description of the experimental set-up is reported, then the procedures for calibration and data analysis are described in detail. The results show that the rate measured in this latitude range stays constant within a novel accuracy of $$\pm 1$$ ±1 %.

Abstract

Microtubules are one of the three components of the cell cytoskeleton. They are hollow wires, with a diameter of about 25 nm, formed by 13 laterally associated protofilaments composed by dimers of α and β-tubulin. Microtubules are dynamic structures which undergo constant modifications by shrinking and elongating in a phenomenon called treadmilling. Microtubules intervene in various fundamental aspects of the biology of a cell. They contribute to determine the shape of a cell, play a role in the cell movement, and in the intracellular transport of organelles during motion and mitotic chromosome segregation. Despite the relevance of the processes mediated by microtubules most studies on the effects of ionizing radiations focus their attention on the damages delivered to DNA. In this paper we attempt to assess the effects borne by IRs to the microtubules network as a biological target. In this study we irradiated Hs 578Bst cells (a no-cancer, no-immortalized, human breast epithelial cell line) with an 8 Gy single dose of either X-rays or protons. After the irradiated cells fixation, the microtubules were imaged by means of stochastic optical reconstruction microscopy to characterize the network disruption. In our results, Microtubules fibers integrity appears to not have been significantly affected at the administered dose of protons and X-rays, nonetheless we observed differences in the MT network distribution and fiber curvatures.

11. Influencia de la frecuencia de ordeñes en el nivel de producción y composición láctea en ovejas de la raza pampinta

Abstract

Con el objetivo de determinar la influencia que tiene la frecuencia de ordeñe en el rendimiento y composición láctea de ovejas de la raza Pampinta, se realizó un ensayo en el que se siguió un diseño completamente aleatorizado con un factor (tipo de ordeñe) con dos tratamientos (un ordeñe diario contra dos ordeñes diarios) y con medidas repetidas en el tiempo (tres fechas para la variable producción y dos fechas para la variable composición láctea). Se utilizaron cinco animales por tratamiento (n=5) y cada animal se mantuvo identificado para poder hacerle un seguimiento en tiempo. La suma de los tres controles lecheros dio una producción total de 16,66 litros para el tratamiento doble ordeñe contra 13,83 litros del tratamiento único ordeñe, Los valores promedio de los dos tratamientos fueron los siguientes: 0,922 Its/día para el tratamiento único ordeñe y 1,11° Its/día para el tratamiento doble ordene. Si bien en este trabajo no se encontraron diferencias con significancia estadística entre los tratamientos, existen dos factores fundamentales que afectan la magnitud de la respuesta al doble ordeñe como son, el potencial lechero y la etapa de la curva de lactación. Animales con alto potencial lechero responden en menor grado que aquellos con bajo potencial y a medida que avanzamos en la lactancia tenemos menos respuesta al doble ordeñe. Tampoco se encontraron diferencias significativas estadísticamente entre los tratamientos en lo que respecta a composición láctea, tanto para los contenidos de grasa como para los contenidos de extracto seco. 13. A study of the inter- and intra-operator variability on echocardiographic measurements in dogs

Abstract

Standard transthoracic echocardiography is considered the non-invasive gold standard for the diagnosis of most cardiac diseases. Defining reproducibility, repeatability, and reliability of this exam is imperative to reduce errors in clinical evaluations. The present study aimed at: 1) evaluating the reproducibility and repeatability of 15 echocardiographic parameters in dogs by analyzing measurements obtained from several operators with different levels of experience and comparing them to the ones obtained from two board-certified operators (gold standards - GSs); 2) assessing whether different formative paths have an influence on the variability of the echocardiographic measurements. Fifty-one operators have been included in this study, along with two diplomates of the European College of Veterinary Internal Medicine - Cardiology. Ten dogs were enrolled, 5 Golden Retrievers and 5 Cavalier King Charles Spaniels. Echocardiographic examination was performed on each dog by one GS and several operators on the same day. Results show that M-mode measurements of the interventricular septum and the left ventricular free wall are the least reliable ones since they show a high deviation from the GS and a poor inter-operator reproducibility. Differently, M-mode-obtained internal diameters of the left ventricle in systole and diastole, and measurements of the aortic annulus and root are the most reliable echocardiographic parameters, demonstrating that all the operators can correctly assess left ventricular systolic function and dilation, and evaluate the aortic valve. Furthermore, a specialist clinical activity, more than the acquired theoretical knowledge, affects the reliability of the echocardiographic examination by reducing the difference from the GS’ measurements.

Abstract

After its successful campaign of measurements beyond the Polar Arctic Circle, the PolarquEEEst experiment measured the cosmic charged particle rate at sea level in a latitude interval between 35$$^{\circ }$$ ∘ N and 82$$^{\circ }$$ ∘ N. In this paper, these measurements are described and the corresponding results are discussed.

Abstract

BackgroundPediatric acute-onset neuropsychiatric syndrome (PANS) is characterized by a wide spectrum of symptoms, including the onset of obsessive-compulsive disorder and/or severely restricted food intake, associated with emotional symptoms, behavioral symptoms, developmental regression, and somatic symptoms. Among the possible triggering agents, infectious agents have been extensively explored. More recently, sporadic case reports describe a possible association between PANS and SARS-CoV-2 infection but data on clinical presentation and treatment are still scarce.MethodsWe describe a case series (10 children) with acute onset or relapse of PANS symptoms after SARS-CoV-2 infection. Standardized measures (CBCL, CPRS, C-GAS, CGI-S, Y-BOCS, PANSS, and YGTSS) were used to describe the clinical picture. The efficacy of a pulse treatment with steroids for three consecutive months was assessed.ResultsOur data suggest that the clinical presentation of the COVID-19-triggered PANS is largely similar to that reported in typical PANS, including acute onset, with OCD and/or eating disorders, and associated symptoms. Our data suggest that treatment with corticosteroids may be beneficial for both global clinical severity and global functioning. No serious adverse effects were observed. Both OCD symptoms and tics consistently improved. Among psychiatric symptoms, affective and oppositional symptoms appeared more sensitive to the steroid treatment than the other symptoms.ConclusionOur study confirms that COVID-19 infection in children and adolescents could trigger acute-onset neuropsychiatric symptoms. Thus, in children and adolescents with COVID-19, a specific neuropsychiatric follow-up should be routinely included. Even if a small sample size and a follow-up with only two points (baseline and endpoint, after 8 weeks) limit the conclusions, it seems that steroid treatment in the acute phase may be beneficial and well tolerated.

Abstract

Background Understanding of the physiological effects of chytridiomycosis is crucial to worldwide amphibian conservation. Therefore, we analyzed the cardiac function of two anuran species (Xenopus laevis and Physalaemus albonotatus) with different susceptibilities to infection by the causative agent of chytridiomycosis, Batrachochytrium dendrobatidis (hereafter Bd). Methods We analyzed the in situ heart rate (fH - bpm), relative ventricular mass (RVM -%), and Ca2+ handling in heart of Bd infected animals compared to uninfected controls of both study species. Results Bd infection resulted in a 78% decrease in contraction force values in P. albonotatus when compared to the less susceptible X. laevis. This negative effect was even more evident (82%) for the cardiac pumping capacity. The time to reach peak tension was 125% longer in P. albonotatus than in X. laevis, and cardiac relaxation was 57% longer. Discussion These results indicate a delay in the cardiac cycle of P. albonotatus on a beat-to-beat basis, which was corroborated by the bradycardia observed in situ. In summary, Bd-sensitive species present impaired cardiac function, which could be a factor in mortality risk. The more pronounced effects of Bd in P. albonotatus may not only result from electrolyte imbalance, as previously reported, but also could be an effect of toxins produced by Bd. For X. laevis, the ability to promote cardiac adjustments seems to be an important homeostatic feature that allows greater tolerance to chytridiomycosis. This study provides new physiological mechanisms underlying the tolerance or susceptibility of amphibian species to chytridiomycosis, which determine their adaptability to survive in the affected environments.

Mucormycosis are a group of infections that affect principally immunocompromised host and have a high mortality. Liposomal amphotericin B is the first-line treatment with combined surgical removal of the infectious focus. We report the case of 67-year-old man with idiopathic granulocytic aplasia and a cutaneous lesion caused by Rhizopus arrhizus treated with isavuconazole. Abstract

The existence of independent, yet time correlated, Extensive Air Showers (EAS) has been discussed over the past years, with emphasis on possible physical mechanisms that could justify their observation. The detector network of the Extreme Energy Events (EEE) Collaboration, with its approximately 60 cosmic ray telescopes deployed over the Italian territory, has the potential to search for such events, employing different analysis strategies. In this paper, we have analyzed a set of EEE data, corresponding to an approximately five month observation period, searching for multi-coincidence events among several far telescopes, within a time window of 1 ms. Events with up to 12 coincident telescopes have been observed. Results were compared to expectations from a random distribution of events and discussed with reference to the relativistic dust grain hypothesis.

Abstract

Abstract. Members of the European Society for Immunodeficiencies (ESID) and other colleagues have updated themulti-stage expert-opinion-based diagnostic protocol for non-immunologists incorporating newly defined primary immunodeficiency diseases (PIDs). The protocol presented here aims to increase the awareness of PIDs among doctors working in different fields. Prompt identification of PID is important for prognosis, but this may not be an easy task. The protocol therefore starts from the clinical presentation of the patient. Because PIDs may present at all ages, this protocol is aimed at both adult and paediatric physicians. The multi-stage design allows cost-effective screening for PID of the large number of potential cases in the early phases, with more expensive tests reserved for definitive classification in collaboration with a specialist in the field of immunodeficiency at a later stage.

Abstract

A muon telescope of the Extreme Energy Events (EEE) Project is made by 3 Multigap Resistive Plate Chambers (MRPC). The MRPCs have been fluxed so far with a standard mixture composed by 98% C2H2F4 and 2% SF6, that are GreenHouse Gases (GHG) phasing out of production. In order to reduce GHG emissions, without affecting MRPC excellent performance, the EEE Collaboration is currently studying alternative mixtures environmentally and economically sustainable. MRPCs performance with new gas mixtures, in terms of resolution (time and space), efficiency, tracking capability and stability, are currently under investigation and will be briefly described in this contribution.

Abstract

The whole Extreme Energy Events (EEE) array is composed of 61 telescopes installed in Italian High Schools, built and operated by students and teachers, constantly supervised by researchers. The muon telescope of the EEE Project is made by 3 Multigap Resistive Plate Chambers (MRPC). The unconventional working sites are a unique test field for checking the robustness and the low-ageing features of the MRPC technology for particle tracking and timing purposes. The MRPCs are fluxed with a standard mixture (98% C2H2F4 - 2% SF6) of greenhouse gases (GHG) phasing out of production. The EEE Collaboration is currently studying alternative mixtures environmentally and economically sustainable. The EEE Collaboration actions to reduce the Global Warming Potential (GWP) in the MRPC array of the EEE experiment are progressing.

Abstract

The Extreme Energy Events Project (EEE) represents a breakthrough in outreach activities in Cosmic Ray Physics: high school students are protagonists of an experiment to measure Extensive Air Showers at ground. They start their experience at CERN with the construction of the three high performing Multigap Resistive Plate Chambers constituting the telescope that is then installed inside their school; then they take care of the telescope operation and data analysis. Presently 60 telescopes are installed in Italy and, since 2014, coordinated data taking have been performed during each school year providing a huge amount of candidate muon tracks. Every year hundreds of students and teachers are involved in the activities directly correlated to EEE. The COVID-19 pandemic has strongly affected the experimental activities of the EEE Project. However in the last two years the online activities were strengthened, with an intense programme of collaboration meetings, masterclasses, and hugely successful topical seminars. Starting from the fall of 2021, the improvement of epidemiological situation made it possible to start some of the EEE activities in presence.

28. Lower Extremity Stress Fractures in the National Basketball Association, 2013-2014 Through 2018-2019

Abstract

Background: Players in the National Basketball Association (NBA) are at risk for lower extremity stress fractures, partly because of the sport’s high-intensity demand on the lower body. Purpose: To provide insight on the identification and management of potential risk factors associated with lower extremity stress fractures in NBA athletes. Study Design: Case series; Level of evidence, 4. Methods: A retrospective study was conducted using the NBA electronic medical record database for all players who were on an NBA roster for ≥1 game from the 2013-2014 through 2018-2019 seasons. Player characteristics, games missed, and treatment methodology were independently analyzed. Results were presented as incidence per 1000 player-games. Results: There were 22 stress fractures identified in 20 NBA players over the course of 6 years, with an average of 3.67 stress fractures per year and an incidence of 0.12 stress fractures per 1000 player-games. Most stress fractures occurred in the foot (17/22), and 45% (10/22) of stress fractures were treated surgically, with the most common site of operation being the navicular. On average, approximately 37 games and 243 days were missed per stress fracture injury. There was no significant difference in time to return to play between high-risk stress fractures treated operatively versus nonoperatively (269.2 vs 243.8 days; P = .82). Conclusion: The overall incidence of stress fractures in NBA players was 0.12 per 1000 player-games, and a high percentage of players returned to NBA activity after the injury. There was a relatively even distribution between high-risk stress fractures treated operatively and nonoperatively. When comparing high-risk stress fractures treated operatively to ones treated nonoperatively, no significant difference in average time to return to play in the NBA was found.

Abstract

The search for long-range correlations among air showers is one of the main goal of the Extreme Energy Events (EEE) Project. The existence of such events has only been supposed theoretically through several physical mechanisms, the most convincing being the so-called GZ effect, based on the photodisintegration of a heavy primary nucleus in the solar field. Even with a large detector coverage, current rate expectations are of few events per year. To measure time correlations among distant air showers, sparse arrays of detection stations spread over large areas are needed. A very limited number of experimental setups can perform this measurement and few experimental results have been reported over the past years. Started in 2004 the EEE project is a network of about 60 cosmic muons tracking telescopes made by 3 wide area Multigap Resistive Plate Chambers (MRPCs), sensitive to the direction of incident charged cosmic particles. The telescopes are distributed over the whole Italian territory, thus making the EEE array an ideal tool for the detection of long-range time correlations among extensive air showers. In this paper we will describe the analysis strategies adopted to search for such rare correlation events, together with the results obtained analysing the full statistics collected by the EEE telescopes.

31. Exploring Interacting Topological Insulators with Ultracold Atoms: The Synthetic Creutz-Hubbard Model

Abstract

Understanding the robustness of topological phases of matter in the presence of strong interactions and synthesizing novel strongly correlated topological materials lie among the most important and difficult challenges of modern theoretical and experimental physics. In this work, we present a complete theoretical analysis of the synthetic Creutz-Hubbard ladder, which is a paradigmatic model that provides a neat playground to address these challenges. We give special attention to the competition of correlated topological phases and orbital quantum magnetism in the regime of strong interactions. These results are, furthermore, confirmed and extended by extensive numerical simulations. Moreover, we propose how to experimentally realize this model in a synthetic ladder made of two internal states of ultracold fermionic atoms in a one-dimensional optical lattice. Our work paves the way towards quantum simulators of interacting topological insulators with cold atoms.

Abstract

Due to their efficiency, tracking capabilities and long-term operational conditions, Multigap Resistive Plate Chambers (MRPC) may be used in a stable location in coincidence with additional detectors fixed to other parts of a civil building, to detect relative displacements of different parts of the building due to long term deformations of the structure. In this contribution we report the results of two different measurements carried out in the Department of Physics and Astronomy of the University of Catania (with three floors above the ground and an underground basement), using one of the MRPC telescopes of the Extreme Energy Events project, in coincidence with two scintillator-based muon telescopes located in various positions inside the building. Experimental results obtained by these measurements campaigns over a period of about three months are reported, together with a discussion about the performance of the technique. The sensitivity of the method is also discussed by means of Monte Carlo simulations.

Abstract

This paper describes the simulation framework of the Extreme Energy Events (EEE) experiment. EEE is a network of cosmic muon trackers, each made of three Multi-gap Resistive Plate Chambers (MRPC), able to precisely measure the absolute muon crossing time and the muon integrated angular flux at the ground level. The response of a single MRPC and the combination of three chambers have been implemented in a GEANT4-based framework (GEMC) to study the telescope response. The detector geometry, as well as details about the surrounding materials and the location of the telescopes have been included in the simulations in order to realistically reproduce the experimental set-up of each telescope. A model based on the latest parametrization of the cosmic muon flux has been used to generate single muon events. After validating the framework by comparing simulations to selected EEE telescope data, it has been used to determine detector parameters not accessible by analysing experimental data only, such as detection efficiency, angular and spatial resolution., Comment: 16 pages, 25 figures, 3 tables, submitted to EPJC

Abstract

The PolarquEEEst scientific programme consists in a series of measurements of the cosmic ray flux up to the highest latitudes, well beyond the Polar Article Circle. It started in Summer 2018, when one telescope for cosmic rays was installed on a sailboat leaving from North Iceland, to circumnavigate the Svalbard archipelago and land in Tromsø. It collected data up to 82N, measuring with unprecedented precision the charged particle rate at sea level in these regions. During Fall of the same year and Spring 2019 the PolarquEEEst programme continued with a series of measurements performed using the same detector, which took place first in Italy, with the southernmost point reached at Lampedusa, and then in Germany, with the goal to measure the dependence of cosmic charged particle rate with latitude. Then, in May 2019, the PolarquEEEst collaboration accomplished another important result, installing a cosmic ray observatory for the detection of secondary cosmic muons at Ny Alesund, at 79N, made of three independent identical detectors positioned a few hundred meters from each other, and synchronized in order to operate together as a network. This configuration allows high precision measurements never performed before at these latitudes on a long term, also interesting for their connection with environmental phenomena. Here the various missions will be presented, and the latest results from the measurements performed will be shown.

36. POS0400 MODULATION OF HUMAN EARLY B CELL DEVELOPMENT THROUGH TARGETED DEGRADATION OF IKAROS AND AIOLOS WITH IBERDOMIDE

Abstract

BackgroundB differentiation in the bone marrow (BM) is impaired in patients carrying mutation in the IKFZ1 gene, coding for Ikaros a zinc-finger transcription factor. High Ikaros expression is on the contrary associated with systemic lupus erythematosus[1] and multiple myeloma[2]. Targeted treatment with iberdomide, a cereblon modulator which enhances degradation of Ikaros and Aiolos, is under clinical investigation in multiple myeloma patients and systemic lupus erythematosus. However, consequences of the treatment on human early B cell development remain elusive. Immature B cells develop in the BM from hematopoietic stem cells. An intricate network of transcription factors regulates the maturation process. Ikaros and Aiolos regulate gene expression during B cell development. As reported in mice, Ikaros is essential for the commitment to the lymphoid lineage and later, together with Aiolos, ensures the transition from pre-BII large to pre-BII small cells.ObjectivesInvestigate the effect of iberdomide (CC-220) on human early B cell development simulated in vitro.MethodsWe tested the impact of iberdomide on short term culture of BM-derived lymphocytes and in a unique in vitro modeling of early B cell development starting from cord blood (CB)- CD34+ progenitors [3, 4]. We used multi-dimensional spectra flow cytometry (17-color pan-el) to dissect early B cell subpopulations.ResultsIberdomide treatment led to enhanced degradation of Ikaros and Aiolos in both BM- and CB-derived cultures. Addition of iberdomide early (day 7) to the CB-derived culture impaired the specification to the lymphoid lineage and later also the commitment to the B cell lineage. These observations were confirmed by reduced E2A and PAX5 gene expression, respectively. Treatment with iberdomide on B cell precursors (pro- and pre-B cells, day 28 of culture) on one side it enhanced the proliferation of early progenitors resulting in increased amount of CD10+CD38+ lymphoid-committed cells. On the other side, it resulted in a accumulation of pre-B cells and inefficient development of immature B cells.ConclusionIberdomide impairs the commitment to the lymphoid lineage by enhancing Ikaros’ degrada-tion. When targeting already committed B cells, iberdomide treatment undermines the transition of pre-BII large to pre-BII small cells due to increased Aiolos’ degradation, conse-quently impairing the development of immature B cells. Abstract

Amphibians have been facing a pandemic caused by the deadly fungus Batrachochytrium dendrobatidis (Bd). Although studies have elucidated cutaneous and homeostatic disturbances, it is still unknown if the hepatic function can be affected or if hepatic effects differ among host species. Thus, we evaluated the effects of an experimental Bd infection on the liver (histopathology and the hepatosomatic index) of 2 anuran species (Xenopus laevis and Physalaemus albonotatus) with different susceptibilities to Bd infection and compared them to uninfected controls. Bd infection increased the melanomacrophage cell area and induced leukocyte infiltration in both species. The effects were more pronounced in the sensitive species, P. albonotatus, which showed severe reduction in glycogen stores and liver atrophy, due to energetic imbalance. Hepatocytes of P. albonotatus also showed ballooning degeneration (vacuolization), which could lead to cell death and liver failure. Our results provide evidence that although the sensitive species showed more severe effects, the tolerant species also had hepatic responses to the infection. These findings indicate that hepatic function can play an important role in detoxification and in immune responses to chytridiomycosis, and that it may be used as a new biomarker of health status in chytrid infections.

Abstract

Inborn errors of immunity (IEI) are a genetically heterogeneous group of disorders with a broad clinical spectrum. Identification of molecular and functional bases of these disorders is important for diagnosis, treatment, and an understanding of the human immune response. We identified 6 unrelated males with neutropenia, infections, lymphoproliferation, humoral immune defects, and in some cases bone marrow failure associated with 3 different variants in the X-linked gene TLR8, encoding the endosomal Toll-like receptor 8 (TLR8). Abstract

The Extreme Energy Events (EEE) experiment, dedicated to the study of secondary cosmic rays, is arguably the largest detector system in the world implemented by Multigap Resistive Plate Chambers. The EEE network consists of 60 telescopes distributed over all the Italian territory; each telescope is made of three MRPCs and allows to reconstruct the trajectory of cosmic muons with high efficiency and optimal angular resolution. A distinctive feature of the EEE network is that almost all telescopes are housed in High Schools and managed by groups of students and teachers, who previously took care of their construction at CERN. This peculiarity is a big plus for the experiment, which combines the scientific relevance of its objectives with effective outreach activities. The unconventional location of the detectors, mainly in standard classrooms of school buildings, with heterogeneous maintenance conditions and without controlled temperature and dedicated power lines, is a unique test field to verify the robustness, the low aging characteristics and the long-lasting performance of MRPC technology for particle monitoring and timing. Abstract

Due to their penetration capability, cosmic muons may provide a way to monitor the alignment and possible long term deformations of large structures, such as historical or other civil buildings. The basic idea behind this possibility is to look for any misalignment between position-sensitive detectors, fixed to different parts of the structure, relative to the original alignment condition. In this paper we discuss the possibility of employing Multigap Resistive Plate Chambers (MRPC) as tracking devices, operating them in coincidence with additional detectors without tracking capability. One of the MRPC telescopes (size 158× 82 cm2) of the Extreme Energy Events (EEE) project, installed in the underground floor at the Department of Physics and Astronomy in Catania, was used together with a 40× 60 cm2 scintillator-based detector, located at about 16 m vertical distance, on the third floor of the same building. Coincidence measurements were carried out over a period of about two months by shifting the position of the smaller detector, to mimic the movement of the structure. Plans for future studies with different detectors and under different geometrical configurations are also discussed.

Abstract

The PolarquEEEst scientific programme consists in a series of measurements of the cosmic ray flux up to the highest latitudes. It started in Summer 2018, when three telescopes made out of scintillators readout by SiPMs were built and installed in Italy, Norway and on a sailboat leaving from North Island, to circumnavigate the Svalbard archipelago and land in Tromsø. They collected data on a latitude range from 44° N up to 82° N, with a dense sampling of the Northernmost interval. The PolarquEEEst mission continued afterwards with a series of measurements in Italy, Southward reaching Lampedusa, and in Germany. In May 2019 the PolarquEEEst collaboration accomplished another important result, installing a cosmic ray observatory for the detection of secondary cosmic muons at Ny Alesund, at 79° N, made of three independent identical detectors positioned a few hundred meters from each other, and synchronized in order to operate together as a network. The configuration used will allow high precision measurements never performed before at these latitudes on a long term, also interesting for their connection with environmental phenomena. The network will also complement the existing stations for the detection of cosmic neutrons at the Svalbard archipelago, enlarging by far the physics scope that is possible to pursue in this field at this peculiar location. Here the various missions are presented, and some preliminary results from the measurements performed are shown.

Abstract

The Extreme Energy Events (EEE) Project employs Multi-gap Resistive Plate Chamber (MRPC) for studying the secondary cosmic ray muons in Extensive Air Showers. The array consists of about 60 tracking detectors, sparse on Italian territory and at CERN. The MRPCs are flowed with a gas mixture based on $C_2H_2F_4$ and $SF_6$, both of which are fluorinated greenhouse gases with a high environmental impact on the atmosphere. Due to the restrictions imposed by the European Union, these gases are being phased out of production and their cost is largely increasing. The EEE Collaboration started a campaign to reduce the gas emission from its array with the aim of containing costs and decreasing the experiment global warming impact. One method is to reduce the gas rate in each EEE detector. Another is to develop a gas recirculation system, whose a first prototype has been installed at one of the EEE stations located at CERN. Jointly a parallel strategy is focused on searching for environmental friendly gas mixtures which are able to substitute the standard mixture without affecting the MRPC performance. Abstract

Cosmic ray muons are a penetrating component of extensive air showers created in the Earth atmosphere by the interaction of highly energetic primary particles, mostly protons, which continuously bombard our Planet. The secondary cosmic radiation is the result of the complex interplay between the production cross section and the interaction mechanisms with the atmosphere (including the energy loss, multiple scattering and particle decay). Cosmic muons have been considered since several decades as a powerful probe to exploit our environment, from muography of volcanoes to absorption radiography of possible hidden rooms inside large structures, such as Pyramids, to the detection of high-Z illicit nuclear materials inside containers and many other applications of social interest. This paper discusses the possibility to employ the Multigap Resistive Plate Chambers (MRPC) of the Extreme Energy Events (EEE) Project as muon tracking detectors to monitor the long term stability of civil buildings and structures when used in conjunction with additional detectors. For this application the average direction of the cosmic muon tracks passing through the MRPC telescope and an additional detector located some distance apart in the same building may be reconstructed with good precision and any small variation over long time acquisition periods may be monitored. The performance of such setup is discussed and experimental results from first coincidence measurements obtained with a 40 × 60 cm2 scintillator detector operated in the same building with one of the EEE telescopes, at about 15 m vertical distance from it, are presented. Simple Monte Carlo and GEANT simulations were also carried out to evaluate typical acceptance values for the operating conditions employed so far, to extrapolate to other geometrical configurations, and to evaluate multiple scattering effects.

Abstract

EEE is an extended cosmic ray observatory, covering more than 10 degrees in latitude and longitude. The relative distances between clusters of telescopes reached the 1200 km, allowing the search for rare long distance correlations between cosmic showers. On the other side each EEE telescope is capable of measuring the flux of secondary particles, opening to the study of low energy phenomena such as solar activity and Forbush decreases. An introduction to the observaotry and updates on the main scientific results are presented. Last updates on the searches for rare correlated showers, with a set of long distance correlation candidate events are also reported.

Abstract

The Extreme Energy Events (EEE) Project is mainly devoted to the study of the secondary cosmic ray radiation by using muon tracker telescopes made of three Multigap Resistive Plate Chambers (MRPC) each. The experiment consists of a telescope network mainly distributed across Italy, hosted in different building structures pertaining to high schools, universities and research centers. Therefore, the possibility to take into account the effects of these structures on collected data is important for the large physics programme of the project. A simulation tool, based on GEANT4 and using GEMC framework, has been implemented to take into account the muon interaction with EEE telescopes and to estimate the effects on data of the structures surrounding the experimental apparata.A dedicated event generator producing realistic muon distributions, detailed geometry and microscopic behavior of MRPCs have been included to produce experimental-like data. The comparison between simulated and experimental data, and the estimation of detector resolutions is here presented and discussed.

Abstract

A simulation tool based on GEMC framework to describe the MRPC telescope of the Extreme Energy Events (EEE) Project is presented. The EEE experiment is mainly devoted to the study of the secondary cosmic muons by using MRPC telescope distributed in high schools and research centres in Italy and at CERN. This takes into account the muon interactions with EEE telescopes and the structures surrounding the experimental apparata; it consists of a dedicated event generator producing realistic muon distribution and a detailed geometry description of the detector. Microscopic behaviour of MRPCs has been included to produce experimental-like data. Abstract

The Extreme Energy Events (EEE) Project is an experiment aimed at the detection of secondary cosmic ray muons. It consists of a sparse array of about 60 telescopes, based on Multigap Resistive Plate Chambers (MRPCs), mostly distributed throughout the Italian territory, mainly in high schools locations, and at CERN. The telescopes are now operational and taking data since more than ten years with a high duty cycle and detection efficiency. The analysis activity is currently in progress and focused on several items, including the detailed study of the muon ux under dfferent conditions, its connection with atmospheric and solar events, the detection of extensive air showers and the search for long distance correlations between different extensive air showers. In this paper an overall description of the experiment will be given, together with its educational fallout. The operation of the whole array is also discussed by showing the most recent results obtained from the analysis of the collected data.
