Entecavir treatment failure can be observed in compliant patients despite an absence of detectable resistance mutations by Pol/RT Sanger sequencing. We hypothesized that these unexplained treatment failures could rely on other mechanisms of viral resistance, especially on mutations selected outside of the Pol/RT domain. Partial virological response to entecavir was observed in three patients treated with immunosuppressive drugs, without selection of Pol/RT resistance mutations. Mutations selected in the whole HBV genome during entecavir treatment and potentially associated with resistance were searched for using deep sequencing and characterized using a phenotypic resistance assay. Mutations Q206K (pre-core/core), Q120K (pre-S1/pre-S2, T-cell epitope) and A300E (spacer domain) were selected during entecavir treatment in patient #1 but were not associated with an increased level of resistance to entecavir or an increase in HBV replication capacity. Core promoter mutations T1753G, A1762T and G1764A were present as major mutations before and after treatment in patient #1. HBs Ag immune escape mutations were present as major mutations before and after treatment in patients #2 (sK122R, sT126I, sP127S and sG145R) and #3 (sM133I). We demonstrated that PVR to entecavir does not require selection of any resistance mutation in the whole HBV genome. Our results demonstrate that major mutations can be selected outside of the Pol/RT domain before or during entecavir treatment. Treatment adaptation after hepatitis B virus (HBV) treatment failure relies on genotypic resistance testing. However, the results of such tests are not always consistent with treatment response. These discrepancies may be due to differences in resistance levels between isolates with the same genotypic resistance testing profiles. We explored this hypothesis by investigating six cases of entecavir treatment failure with an integrative strategy combining genotypic and phenotypic resistance testing, medical record review and therapeutic drug monitoring. Among isolates with genotypic reduced susceptibility to entecavir, one displayed a higher level of resistance to entecavir (mean fold change in entecavir IC 50 of 1 508 ± 531 vs. 318 ± 53, p = 0.008). This isolate harbored a substitution (rt250L) at a position reported to be associated with resistance (rt250V). Reversion to wild-type amino acid at this position partially restored susceptibility to entecavir, confirming that the rt250L mutation was responsible for the high level of resistance to entecavir. This is the first description of entecavir treatment failure associated with selection of the rt250L mutation without other entecavir resistance mutations. One isolate with genotypic resistance to entecavir, harboring the rt173L mutation, displayed a lower level of resistance than the other, harboring the rt202G mutation (mean fold change of 323 ± 124 vs. 6 036 ± 2 100, p = 0.20). These results suggest that isolates harboring the rt250L mutations should be considered resistant to entecavir, whereas isolates harboring the rt173L mutations should be considered to display reduced susceptibility to entecavir. An integrative approach to antiviral drug resistance in HBV would provide a more accurate assessment of entecavir treatment failures and help to improve the accuracy of genotypic testing algorithms., (Copyright © 2020 Elsevier B.V. Single-walled carbon nanotubes can be spun in a polyvinyl alcohol stream to produce nanocomposite fibres. We use a facile ester linking between both elements to create improved fibres which exhibit outstanding enhancements in the absence of post-processing stages, providing a promising alternative based on a chemical method.

Residual stress in polymers arises from the freezing of unstable molecular conformations. Residual stress is critical because its relaxation can cause shrinkage, defects, and fractures of polymer materials. The storage of stress is purposely enhanced to develop shape memory materials. Unfortunately, the storage of mechanical stress is still poorly controlled and understood. An approach to sense the storage of stress based on the spectroscopic response of carbon nanotubes is explored. The Raman response of nanotubes exhibits a variable sensitivity to strain when embedded in polymers that have experienced different thermal and mechanical treatments. An assembly of packed and oriented rodlike particles exhibit anisotropic physical properties. We investigate in the present work the anisotropic conductivity of films made of intrinsically conducting rods. These films are obtained from more or less ordered carbon nanotube liquid crystals. Their orientational order parameter is measured by polarized Raman spectroscopy. A relationship between the anisotropy of surface conductivity and orientational order parameter is determined. The experimental results are accounted for by a model that takes into account the number of intertube contacts and density of conductive pathways in different directions, as introduced by J. Fischer et al. for magnetically aligned nanotubes. We find that a good agreement, without any fitting parameter, of the proposed model and experiments is obtained when we consider a two-dimensional (2D) Gaussian distribution of the nanotube orientation. The conductivities parallel and perpendicular to the nematic director differ by almost an order of magnitude. This anisotropy is much greater than that of conventional dielectric liquid crystals, where the behavior is governed by the mobility anisotropy of ionic current carriers. The present results do not depend on the intrinsic properties of the nanotubes and are expected to be relevant for other assemblies of conducting rodlike particles, such as metallic or semi-conducting nanowires and ribbons.

Many synthetic or natural fibers are produced via the transformation of a liquid solution into a solid filament, which allows the wet processing of high molecular weight polymers, proteins, or inorganic particles. Synthetic wet-spun fibers are used in our everyday life from clothing to composite reinforcement applications. Spun fibers are also common in nature. Silk solidification results from the coagulation of protein solutions. The chemical phenomena involved in the formation of all these classes of fibers can be quite different but they all share the same fundamental transformation from a liquid to a solid state. The solidification process is critical because it governs the production rate and the strength that fibers can sustain to be drawn and wound. An approach is proposed in this work to investigate the kinetics of fiber solidification. This approach consists in circulating solidifying fibers in the extensional flow of a surrounding liquid. Such as polymers in extensional flows, the fibers break if resultant drag forces exceed the fiber tensile strength. The solidification kinetics of nanotube composite fibers serves as a validation example of this approach. The method could be extended to other systems and advance thereby the science and technology of fiber and textile materials. It is also a way to directly visualize the scission of chain-like systems in extensional flows.

Poor electron transfer and slow mass transport of substrates are significant rate-limiting steps in electrochemical systems. It is especially true in biological media, in which the concentrations and diffusion coefficients of substrates are low, hindering the development of power systems for miniaturized biomedical devices. In this study, we show that the newly engineered porous microwires comprised of assembled and oriented carbon nanotubes (CNTs) overcome the limitations of small dimensions and large specific surface area. Their improved performances are shown by comparing the electroreduction of oxygen to water in saline buffer on carbon and CNT fibres. Under air, and after several hours of operation, we show that CNT microwires exhibit more than tenfold higher performances than conventional carbon fibres. Consequently, under physiological conditions, the maximum power density of a miniature membraneless glucose/oxygen CNT biofuel cell exceeds by far the power density obtained for the current state of art carbon fibre biofuel cells.

We present a detailed study of the influence of pH on the dispersion and film-forming properties of poly(acrylic acid)-stabilized carbon nanotubes. Poly(acrylic acid) (PAA) is a weak polyelectrolyte, with a pH-responsive behavior in aqueous solution. We obtain quantitative UV-visible measurements to show that the amount of polyelectrolyte in optimal pH conditions is weak, showing a good efficiency of the polymer as a carbon nanotube dispersing agent. The best dispersion conditions are achieved at pH 5, a value close to the pK(a) of PAA. Apart from this tenuous pH value, the PAA is not efficient at stabilizing nanotubes and atomic force microscopy allows us to explain the delicate balance between the PAA adsorption and the suspension stability. This study finally permits optimal conditions for making homogeneous and conductive composite films to be determined.

Lyotropic nematic aqueous suspensions of single-wall carbon nanotubes can be uniformly aligned in thin cells by shearing. Homogeneous anisotropic thin films of nanotubes can be prepared by drying the nematic. Optical transmission between parallel or crossed polarizers is measured and described in order to estimate the dichroic ratio. The order parameter is measured using polarized Raman spectroscopy and found to be quite weak due to entanglement of the nanotubes and/or to an intrinsic viscoelastic behavior of the nanotube suspensions.

Shape-memory polymers can revert to their original shape when they are reheated. The stress generated by shape recovery is a growing function of the energy absorbed during deformation at a high temperature; thus, high energy to failure is a necessary condition for strong shape-memory materials. We report on the properties of composite nanotube fibers that exhibit this particular feature. We observed that these composites can generate a stress upon shape recovery up to two orders of magnitude greater than that generated by conventional polymers. In addition, the nanoparticles induce a broadening of the glass transition and a temperature memory with a peak of recovery stress at the temperature of their initial deformation.

As-produced carbon nanotubes often contain a fraction of impurities such as metal catalysts, inorganic supports, and carbon by-products. These impurities can be partially removed by using acidic dissolution. The resulting nanotube materials have to be dried to form a powder. The processability of nanotubes subjected to regular (thermal vaporisation) drying is particularly difficult because capillary forces pack and stick the nanotubes irreversibly, which limits their dispersability in polymeric matrices or solvents. We show that this dramatic limitation can be circumvented by using freeze-drying instead of regular-drying during nanotube purification process. In this case, the nanotubes are trapped in frozen water which is then sublimated. As a result the final powder is significantly less compact and, more important, the nanotubes can be easily dispersed with no apparent aggregates, thereby greatly enhancing their processability, e.g., they can be used to make homogeneous composites and fibers. Results from coagulation spinning from water-based dispersions of regularly-dried and freeze-dried nanotubes are compared. We also show that freeze-dried materials, in contrast to regularly-dried materials, can be dissolved in organic polar solvents using alkali-doped nanotubes. High resolution TEM and XRD analysis demonstrate that the nanotube structure and quality are not affected at the nanoscale by freeze-drying treatments.

We report the spontaneous liquid crystal phase separation of nanotubes (single-walled carbon nanotubes, SWNTs) stabilized in aqueous biological (hyaluronic acid, HA) solutions. Sonication of SWNTs in solutions of HA produced well-dispersed single-phase isotropic dispersions which, over time, phase separated into dispersions containing birefringent nematic domains in equilibrium with an isotropic phase. The time required for phase separation to occur was shown to depend on the concentration of SWNT and HA, with the attractive interactions between the SWNT and HA shifting the onset of the phase separation toward lower concentration. This phase separation is accompanied by an increase in the dispersion viscosity with this increase qualitatively matching the degree of phase separation. The formation of ordered phases in biological media can offer wide opportunities for processing conducting biomaterials with aligned and oriented domains.

The hydrolysis of distannylated compounds in which the tin atoms are linked by an organic spacer has been studied under microemulsion conditions using dynamic light scattering and infrared spectroscopy. The experiments provided evidence that the growth of hybrid material particles occurs in the aqueous phase, outside the organic phase of the microemulsion. The growth rates of the particles were found to be strongly dependent on the nature of the spacers, a polymethylene chain inducing the fastest process. This different behavior was explained by a slower condensation process rather than a slower hydrolysis. The high surface areas measured for the hybrid materials could be explained by a possible coating of the hybrid particles by surfactant molecules, thus preventing either their growth or their aggregation.

We report a new hot-drawing process for treating wet-spun composite fibers made of single- and multiwall carbon nanotubes and poly(vinyl alcohol). As shown in previous reports, untreated composite nanotube fibers exhibit a very large strain-to-failure, and their toughness, which is the energy needed to break the fibers, exceeds that of any other known materials. However, untreated composite nanotube fibers absorb a very small amount of energy at low strain and become degraded in humid conditions. In this work, we use hot-drawing treatments, a concept inspired from textile technologies, to improve the properties of nanotube/PVA fibers. This treatment yields a crystallinity increase of the PVA and an unprecedented degree of alignment of the nanotubes. These structural modifications lead to a markedly improved energy absorption at low strain and make the fibers resistant to moisture. Hot-drawn nanotube/PVA fibers hold great potential for a number of applications such as bulletproof vests, protective textiles, helmets, and so forth.

Percolation is a statistical concept that describes the formation of an infinite cluster of connected particles or pathways. Lowering the percolation threshold is a critical issue to achieve light and low-cost conductive composites made of an insulating matrix loaded with conductive particles. This has interest for applications where charge dissipation and electrical conductivity are sought in films, coatings, paints, or composite materials. One route to decreasing the loading required for percolation is to use rod-like particles. Theoretical predictions indicate that this may also be achieved by altering the interaction potential between the particles. Although percolation may not always respond monotonically to interactions, the use of adhesive rods can be expected to be an ideal combination. By using a system made of carbon nanotubes in an aqueous surfactant solution, we find that very small attraction can markedly lower the percolation threshold. The strength of this effect can thereby have direct technological interest and explain the large variability of experimental results in the literature dealing with the electrical behavior of composites loaded with conducting rods.

Hybrid materials where layers of tin oxide alternate with layers of hydrophobic organic chains were prepared by the hydrolysis of distannylated compounds containing an organic chain alpha,omega-disubstituted by tripropynylstannyl groups. In the case of an aliphatic chain, hydrolysis under microemulsion conditions led to the organization of the corresponding hybrid. These hydrolysis conditions also induced a high surface area and a defined mesoporosity in the hybrid. When a mixed aromatic-aliphatic spacer was used, weak hydrophobic interactions between the spacers were sufficient to generate the same type of organization in the corresponding material.
