In response to paradigm shifts in education, teacher education institutions have increasingly adopted flexible learning modalities such as online courses, blended learning approaches, and personalized instruction to meet the diverse needs of students and prepare educators for the demands of modern classrooms. This study used a mixed-method research design to gain a comprehensive understanding of the flexible teaching and learning modalities it brought to mathematics teacher educators (MTEs) and learners in a state university in West Philippines. Since ethical considerations preclude random sampling, the study comprised nonrandom participants using purposive sampling, where 189 learners were surveyed, and six MTEs and 16 learners were interviewed. A researcher-made interview guide was prepared to describe the flexible teaching and learning modalities and opportunities of the MTEs. Also, a researcher-made questionnaire was used to describe the learners' general assessment of the flexible teaching and learning modalities in mathematics education. Quantitative data were gathered online, while qualitative data were gathered through interviews. Results showed that flexible teaching-learning approaches have implications for education, overcoming barriers, and promoting engagement. The findings highlight the importance of technology integration, inclusive assessment modalities, infrastructure support, and professional development opportunities in enhancing the quality and accessibility of flexible teaching and learning, ultimately leading to inclusive educational environments. Therefore, the educational institution must adopt flexible instructional modalities, emphasize diverse assessment methods, and personalize the learning experience by integrating online platforms and various assessments for effective mathematics instruction.

In this research work, the feasibility of using fusel oil, a by-product of the sugar–alcohol industry, as an LVLC solvent in blends with straight vegetable oils (SVOs) and diesel was investigated. Concretely, diesel/fusel oil/sunflower oil (D/FO/SO) and diesel/fusel oil/castor oil (D/FO/CO) triple blends were prepared and characterized by measuring the most important physicochemical properties, i.e., viscosity, density, cold flow properties, flash point and cetane number. An appreciable improvement in cold flow values has been achieved with triple blends, without compromising properties such as calorific value and cetane number. Likewise, the triple blends meet the viscosity and density requirements specified by the European quality standard EN 14214 and the American standard ASTM D6751. After characterization, the triple blends were used on a diesel engine, evaluating different parameters such as power output, opacity, exhaust emissions (CO and NOx) and consumption at different engine loads. The results indicate that as the biofuel content in the blend increases, engine power decreases while fuel consumption rises. Nevertheless, the values obtained with D/FO/CO are better than those for D/FO/SO and are also very similar to those of fossil diesel. Regarding opacity values and NOx emissions obtained with the utilization of the triple blends, they are lower than those produced by diesel. However, in the case of CO emissions, it depends on the type of oil used, with the samples prepared with castor oil exhibiting the best results.

Abstract The principal effect controlling the oxygen affinity of vertebrate haemoglobins (Hbs) is the allosteric switch between R and T forms with relatively high and low oxygen affinity respectively. Uniquely among jawed vertebrates, crocodilians possess Hb that shows a profound drop in oxygen affinity in the presence of bicarbonate ions. This allows them to stay underwater for extended periods by consuming almost all the oxygen present in the blood-stream, as metabolism releases carbon dioxide, whose conversion to bicarbonate and hydrogen ions is catalysed by carbonic anhydrase. Despite the apparent universal utility of bicarbonate as an allosteric regulator of Hb, this property evolved only in crocodilians. We report here the molecular structures of both human and a crocodilian Hb in the deoxy and liganded states, solved by cryo-electron microscopy. We reveal the precise interactions between two bicarbonate ions and the crocodilian protein at symmetry-related sites found only in the T state. No other known effector of vertebrate Hbs binds anywhere near these sites.

Multiple myeloma (MM), a cancer of the bone marrow, is categorized as the second most common hematological malignancy of adults in the Western world. Despite dramatic improvements in immunotherapies in the field of cancers, MM immunotherapy has not been promising until now. Recent clinical studies of immune checkpoint inhibitor therapy, either alone or in combination with anticancer drugs, showed excessive side effects or low efficacy, particularly in advanced MM patients. In this context, lymphocyte levels of exhaustion markers play a pivotal role in the MM tumor microenvironment (TME). Hence in the present review, the mechanisms relevant to MM of five inhibitory molecules including T-cell immunoreceptor with Ig and ITIM domains (TIGIT), T-cell immunoglobulin, and mucin domain 3 (Tim-3), lymphocyte activation gene-3 (LAG-3), V-domain Ig Suppressor of T-cell activation and killer immunoglobulin-like receptors along with bispecific T-cell antibodies (BsAbs) will be discussed. Further, we summarized the underlying biology of these checkpoints in cancer and their rapidly emerging role in pathways in MM along with presenting recent clinical trials in context.

The Philippines, frequently affected by typhoons, faces the hazard of storm surges. This study examined the Coupled-Ocean-Atmosphere-Wave-Sediment Transport Modeling System (COAWST) to simulate Typhoon Karding's September 2022 storm surge. COAWST integrates the Regional Ocean Modeling System (ROMS), Simulating WAves Nearshore (SWAN), and Weather Research and Forecasting (WRF) coupled using the Model Coupling Toolkit (MCT). Four setups were analyzed: i. ROMS only, ii. ROMS-SWAN, iii. ROMS-WRF, and iv. ROMS-SWAN-WRF, focusing on four variables: a. Surface air pressure, b. Wind speed, c. Free-surface elevation, and d. Significant wave height. Results show that the ROMS-WRF and ROMS-SWAN-WRF setups accurately simulated Typhoon Karding's track with minimal positional error and wind speed. However, the models overestimated the typhoon's minimum air pressure with p-biases of 7.74% (i and ii), 4.1% (iii), and 3.9% (iv), and RMSE values of 68.529 hPa (i and ii), 36.744 hPa (iii), and 36.789 hPa (iv). Additionally, water levels were underestimated, with RMSE ranging from 0.31 to 0.35 meters and p-biases from −72.56% to −154.89% at Baler, Aurora validation point. At the Real, Quezon validation point, RMSE and p-bias ranged from 0.30 to 0.34 meters and −84.80% to −166.44%, respectively. Nonetheless, the models were able to simulate the storm surge and significant wave height at Baler and Real points similar to recorded data, with setup iv performing best in storm surge simulation. In summary, COAWST may be employed for typhoon simulations, with coupling being able to increase accuracy.

This study evaluates the performance of biofuels created from triple blends of fossil diesel, sunflower or castor oil (SVOs), and 2-Ethylhexyl Nitrate (EHN), a low-viscosity, high-cetane (LVHC) solvent. EHN reduces the viscosity of SVOs to enable their use in conventional diesel engines without compromising fuel properties. The results show that the power output from these blends is similar to or greater than that of fossil diesel, with comparable fuel consumption. Furthermore, the blends significantly reduce emissions of carbon monoxide (CO) and soot, though NOx emissions are slightly higher due to the nitrogen content in EHN. However, NOx levels remain within permissible limits. The substitution of fossil diesel could be further enhanced if EHN were produced using green hydrogen and lignocellulosic biomass, making it a renewable and sustainable biofuel component. These findings support the potential of EHN/SVO biofuel blends to replace a significant portion of fossil diesel in conventional diesel engines while maintaining performance and reducing harmful emissions, except for a slight increase in NOx.

IntroductionSevere forms of short bowel syndrome (SBS) resulting in chronic intestinal failure (IF) have limited therapeutic options, all of which are associated with significant morbidities. Spring-mediated distraction enterogenesis (SMDE) uses an intraluminal self-expanding spring to generate mechanical force to induce intestinal stretching and sustained axial growth, providing a promising novel approach for patients with SBS. Previous studies have established this method to be safe and effective in small and large animal models. However, SMDE has previously not been implemented in a large, clinically relevant animal model.MethodsJuvenile mini-Yucatan pigs with 75% of their small intestine resected had intraluminal springs placed after an initial adaptive period. Morphological and histological assessments were performed on SMDE segments compared to the control region of the intestine undergoing normal adaptive responses to resection.ResultsWhile the initial histologic adaptive response observed following resection was attenuated after a month, the SMDE segments instead augmented these adaptive changes. Specifically, intestinal length increased 2-fold in SMDE segments, and the widths of the epithelial, muscularis, and serosal layers were enhanced in SMDE compared with control segments of the same animal. This data suggests that morphologic intestinal adaptation may be enhanced with SMDE in the setting of SBS.DiscussionHere we demonstrate the successful and reproducible implementation of SMDE in a large animal model in the setting of prior intestinal resection, making SMDE a viable and novel approach for SBS to be explored further.

Malaria is an infectious disease caused by Plasmodium spp. parasites, with widespread drug resistance to most antimalarial drugs. We report the development of two 3D-QSAR models based on comparative molecular field analysis (CoMFA), comparative molecular similarity index analysis (CoMSIA), and a 2D-QSAR model, using a database of 349 compounds with activity against the P. falciparum 3D7 strain. The models were validated internally and externally, complying with all metrics (q2 > 0.5, r2test > 0.6, r2m > 0.5, etc.). The final models have shown the following statistical values: r2test CoMFA = 0.878, r2test CoMSIA = 0.876, and r2test 2D-QSAR = 0.845. The models were experimentally tested through the synthesis and biological evaluation of ten quinoline derivatives against P. falciparum 3D7. The CoMSIA and 2D-QSAR models outperformed CoMFA in terms of better predictive capacity (MAE = 0.7006, 0.4849, and 1.2803, respectively). The physicochemical and pharmacokinetic properties of three selected quinoline derivatives were similar to chloroquine. Finally, the compounds showed low cytotoxicity (IC50 > 100 µM) on human HepG2 cells. These results suggest that the QSAR models accurately predict the toxicological profile, correlating well with experimental in vivo data.

IntroductionPaneth cells are critically important to intestinal health, including protecting intestinal stem cells, shaping the intestinal microbiome, and regulating host immunity. Understanding Paneth cell biology in the immature intestine is often modeled in rodents with little information in larger mammals such as sheep. Previous studies have only established the distribution pattern of Paneth cells in healthy adult sheep. Our study aimed to examine the ontogeny, quantification, and localization of Paneth cells in fetal and newborn lambs at different gestational ages and with perinatal transient asphyxia. We hypothesized that ovine Paneth cell distribution at birth resembles the pattern seen in humans (highest concentrations in the ileum) and that ovine Paneth cell density is gestation-dependent.MethodsIntestinal samples were obtained from 126–127 (preterm, with and without perinatal transient asphyxia) and 140–141 (term) days gestation sheep. Samples were quantified per crypt in at least 100 crypts per animal and confirmed as Paneth cells through in immunohistochemistry.ResultsPaneth cells had significantly higher density in the ileum compared to the jejunum and were absent in the colon.DiscussionExposure to perinatal transient asphyxia acutely decreased Paneth cell numbers. These novel data support the possibility of utilizing ovine models for understanding Paneth cell biology in the fetus and neonate.

Nowadays, there is an intense debate in the European Union (EU) regarding the limits to achieve the European Green Deal, to make Europe the first climate-neutral continent in the world. In this context, there are also different opinions about the role that thermal engines should play. Furhermore, there is no clear proposal regarding the possibilities of the use of green hydrogen in the transport decarbonization process, even though it should be a key element. Thus, there are still no precise guidelines regarding the role of green hydrogen, with it being exclusively used as a raw material to produce E-fuels. This review aims to evaluate the possibilities of applying the different alternative technologies available to successfully complete the process already underway to achieve Climate Neutrality by about 2050, depending on the maturity of the technologies currently available, and those anticipated to be available in the coming decades.

To provide technical and economical solutions regarding management of plastic waste, which is constantly increasing worldwide, this study addresses the possibility of using plastic oils (PO) obtained from these plastic wastes as biofuels. To this end, the replacement of the fossil diesel employed in internal combustion diesel engines with triple diesel/PO/vegetable oil mixtures has been investigated. Sunflower (SO) and castor oil (CO) mixed with PO in the most appropriate proportion are evaluated as pure vegetable oils (SVO). Thus, diesel/PO/SVO triple blends were prepared, characterized, and then tested on a diesel engine operating as electricity generator, evaluating power output, consumption, and exhaust emissions. The obtained results show that, with the incorporation of relatively small quantities of pure, non-edible vegetable oils, in double mixtures of PO/SO and PO/CO, an effective alternative fuel for transport is obtained, that allows for 100% of fossil diesel to be replaced. In fact, with these double PO/SVO biofuel mixtures, higher engine power values and lower consumption levels are obtained than those achieved with fossil diesel. Regarding exhaust emissions, these are produced with a slightly greater opacity than with fossil diesel, but there are lower values of carbon gases as a whole (CO + CO2) and in NOx gases.

The eggplant fruit and shoot borer (EFSB) (Leucinodes orbonalis Guenée) is a devastating lepidopteran pest of eggplant (Solanum melongena L.) in the Philippines. Management of an insect pest like the EFSB requires an understanding of its biology, evolution, and adaptations. Genomic resources provide a starting point for understanding EFSB biology, as the resources can be used for phylogenetics and population structure studies. To date, genomic resources are scarce for EFSB; thus, this study generated its complete mitochondrial genome (mitogenome). The circular mitogenome is 15,244 bp-long. It contains 37 genes, namely 13 protein-coding, 22 tRNA, and 2 rRNA genes, and has conserved noncoding regions, motifs, and gene syntenies characteristic of lepidopteran mitogenomes. Some protein-coding genes start and end with non-canonical codons. The tRNA genes exhibit a conserved cloverleaf structure, with the exception in trnS1. Partitioned phylogenetic analysis using 72 pyraloids generated highly supported maximum likelihood and Bayesian inference trees revealing expected basal splits between Crambidae and Pyralidae, and Spilomelinae and Pyraustinae. Spilomelinae was recovered to be paraphyletic, with the EFSB robustly placed before the split of Spilomelinae and Pyraustinae. Overall, the EFSB mitogenome resource will be useful for delineations within Spilomelinae and population structure analysis.

Checkpoint markers and immune checkpoint inhibitors have been increasingly identified and developed as potential immunotherapeutic targets in various human cancers. Despite valuable efforts to discover novel immune checkpoints and their ligands, the precise roles of their therapeutic functions, as well as the broad identification of their counterpart receptors, remain to be addressed. In this context, it has been suggested that various putative checkpoint receptors can be induced upon activation. In the tumor microenvironment, T cells, as crucial immune response against malignant diseases as well as other immune central effector cells, such as natural killer cells, are regulated via co-stimulatory or co-inhibitory signals from immune or tumor cells. Studies have shown that exposure of T cells to tumor antigens upregulates the expression of inhibitory checkpoint receptors, leading to T-cell dysfunction or exhaustion. Although targeting immune checkpoint regulators has shown relative clinical efficacy in some tumor types, most trials in the field of cancer immunotherapies have revealed unsatisfactory results due to de novo or adaptive resistance in cancer patients. To overcome these obstacles, combinational therapies with newly discovered inhibitory molecules or combined blockage of several checkpoints provide a rationale for further research. Moreover, precise identification of their receptors counterparts at crucial checkpoints is likely to promise effective therapies. In this review, we examine the prospects for the application of newly emerging checkpoints, such as T-cell immunoglobulin and mucin domain 3, lymphocyte activation gene-3, T-cell immunoreceptor with Ig and ITIM domains (TIGIT), V-domain Ig suppressor of T-cell activation (VISTA), new B7 family proteins, and B- and T-cell lymphocyte attenuator, in association with immunotherapy of malignancies. In addition, their clinical and biological significance is discussed, including their expression in various human cancers, along with their roles in T-cell-mediated immune responses.

NEC remains one of the most common causes of mortality and morbidity in preterm infants. Animal models of necrotizing enterocolitis (NEC) have been crucial in improving our understanding of this devastating disease and identifying biochemical pathways with therapeutic potential. The pathogenesis of NEC remains incompletely understood, with no specific entity that unifies all infants that develop NEC. Therefore, investigators rely on animal models to manipulate variables and provide a means to test interventions, making them valuable tools to enhance our understanding and prevent and treat NEC. The advancements in molecular analytic tools, genetic manipulation, and imaging modalities and the emergence of scientific collaborations have given rise to unique perspectives and disease correlates, creating novel pathways of investigation. A critical review and understanding of the current phenotypic considerations of the highly relevant animal models of NEC are crucial to developing novel therapeutic and preventative strategies for NEC.

Plants are not only used as energy and nutrient resources for herbivores. Plants can be ingested because of their activity against host parasites and other pathogens. This so-called medicinal role of plants is well reported in ethnopharmacology and under-reported in wild animals. More studies on wild animals are needed because any plant in the world contains bioactive compounds, and probably all plants, no matter how toxic they are, experience herbivory. For example, we tested the activity of extracts and essential oils from Papaver rhoeas and Echium plantagineum against a selection of laboratory pathogens because Great bustards Otis tarda preferred these plants during the mating season, with male fecal droppings showing a higher frequency of P. rhoeas particles than the fecal droppings of females. We hypothesized that P. rhoeas could be helpful for males in the mating season if any part of this plant harbors bioactivity against parasites and other pathogens. Males’ immune system is weakened during the mating season because of their investment in secondary sexual characters and sexual display. As a first exploration of the bioactivity of these plants, we evaluated extracts of both plants against a sample of laboratory models, including a flagellated protozoon (Trichomonas gallinae), a nematode (Meloidogyne javanica) and a fungus (Aspergillus niger). Non-polar and polar extracts of the aerial parts of P. rhoeas, especially the extracts of flowers and capsules, and the extracts of leaves and flowers of E. plantagineum showed activity against nematodes and trichomonads. The bioactivity of plants against parasites could explain the foraging behavior of stressed animals. The chemical communication underpinning the capacity of fauna to recognize those plants is far less known.

Assessment of serological Plasmodium falciparum–specific antibodies in highly endemic areas provides valuable information about malaria status and parasite exposure in the population. Although serological evidence of Plasmodium exposure is commonly determined by Plasmodium-specific immunoglobulin G (IgG) levels; IgM and IgA are likely markers of malaria status that remain relatively unexplored. Previous studies on IgM and IgA responses have been based on their affinity for single antigens with shortage of immune responses analysis against the whole Plasmodium proteome. Here, we provide evidence of how P. falciparum infection triggers the production of specific IgM and IgA in plasma and its relationship with parasite density and changes in hematological parameters. A total of 201 individuals attending a hospital in Breman Asikuma, Ghana, were recruited into this study. Total and P. falciparum–specific IgM, IgA, and IgG were assessed by ELISA and examined in relation to age (0–5, 14–49, and ≥50 age ranges); infection (submicroscopic vs. microscopic malaria); pregnancy and hematological parameters. Well-known IgG response was used as baseline control. P. falciparum–specific IgM and IgA levels increased in the population with the age, similarly to IgG. These data confirm that acquired humoral immunity develops by repeated infections through the years endorsing IgM and IgA as exposure markers in endemic malaria regions. High levels of specific IgA and IgM in children were associated with microscopic malaria and worse prognosis, because most of them showed severe anemia. This new finding shows that IgM and IgA may be used as diagnostic markers in this age group. We also found an extremely high prevalence of submicroscopic malaria (46.27% on average) accompanied by IgM and IgA levels indistinguishable from those of uninfected individuals. These data, together with the observed lack of sensitivity of rapid diagnostic tests (RDTs) compared to PCR, invoke the urgent need to implement diagnostic markers for submicroscopic malaria. Overall, this study opens the potential use of P. falciparum–specific IgM and IgA as new serological markers to predict malaria status in children and parasite exposure in endemic populations. The difficulties in finding markers of submicroscopic malaria are highlighted, emphasizing the need to explore this field in depth.

Considering the renewable electricity production using sustainable technologies, such as solar photovoltaics or wind turbines, it is essential to have systems that allow for storing the energy produced during the periods of lower consumption as well as the energy transportation through the distribution network. Despite hydrogen being considered a good candidate, it presents several problems related to its extremely low density, which requires the use of very high pressures to store it. In addition, its energy density in volumetric terms is still clearly lower than that of most liquid fuels. These facts have led to the consideration of ammonia as an alternative compound for energy storage or as a carrier. In this sense, this review deals with the evaluation of using green ammonia for different energetic purposes, such as an energy carrier vector, an electricity generator and E-fuel. In addition, this study has addressed the latest studies that propose the use of nitrogen-derived compounds, i.e., urea, hydrazine, ammonium nitrate, etc., as alternative fuels. In this study, the possibility of using other nitrogen-derived compounds, i.e., an update of the ecosystem surrounding green ammonia, has been assessed, from production to consumption, including storage, transportation, etc. Additionally, the future challenges in achieving a technical and economically viable energy transition have been determined.

Here, we report the chemical composition and optical properties of the fine particles (PM2.5) and water-soluble organic carbon (WSOC) of these particles. Additionally, the potential sources of WSOC emission were determined through the study on fluorescence excitation–emission matrix spectra and parallel factor analysis (EEM-PARAFAC). Samples were collected in an urban site of the Monterrey Metropolitan Area in Mexico during summer and winter and characterized using attenuated total reflectance-Fourier-transform infrared spectroscopy (ATR-FTIR), ultraviolet-visible-near infrared-diffuse reflectance spectroscopy (UV–Vis-NIR-DRS), fluorescence spectroscopy, X-ray photoelectron spectroscopy (XPS), and X-ray diffraction (XRD) techniques. The ATR-FTIR analyses allowed the identification of inorganic ions (e.g., CO32−, SO42−, and NO3−), organic functional groups [e.g., carbonyls (C=O), organic hydroxyl (C-OH), carboxylic acid (COOH)], and aromatic and unsaturated aliphatic hydrocarbons. The results obtained by XRD and XPS revealed the presence of organic and inorganic chemical species in PM2.5. The diffuse reflectance spectra of PM2.5 provided the absorption bands in the UV region for CaSO4, CaCO3, and aluminosilicates. The absorption coefficient at 365 nm (Abs365) and Ångström absorption exponent (AAE) values obtained for the aqueous extracts suggest that many of the water-soluble organic compounds corresponded to brown carbon (BrC) chromophores. The mass absorption efficiency values at 365 nm (MAE365) were higher in the winter than summer samples, suggesting the presence of more BrC compounds in the winter samples. The fluorescence indices combined with EEM-PARAFAC analysis showed that the WSOC fraction was mainly composed of humic-like substances (HULIS) which are both of terrestrial and microbial origin.

Abstract Incomplete non-sterile immunity to malaria is attained in endemic regions after recurrent infections by a large percentage of the adult population, who carry the malaria parasite asymptomatically. Although blood-stage Plasmodium falciparum rapidly elicits IgG responses, the target antigens of partially protective and non-protective IgG antibodies as well as the basis for the acquisition of these antibodies remain largely unknown. We performed IgG-immunomics to screen for P. falciparum antigens and to identify epitopes associated with exposure and clinical disease. Sera from malaria cases identified five prevalent antigens recognized by all analyzed patients’ IgGs. Epitope mapping of them, using adult and children sera samples from an endemic malaria region in Ghana segregated into patients with positive or negative subclinical detection of P. falciparum, revealed binding specificity for two 20-mer immunodominant antigenic regions within the START-related lipid transfer protein and the protein disulfide isomerase PDI8. These 20-mer epitopes challenged with sera samples from children under 5 years old displayed specific IgG binding in those with detectable parasitemia, even at subclinical level. These results suggest that humoral response against START and PDI8 antigens may be triggered at submicroscopic parasitemia levels in children and may eventually be used to differentially diagnose subclinical malaria in children.

Metallocarboxypeptidases are zinc-dependent peptide-hydrolysing enzymes involved in several important physiological and pathological processes. They have been a target of growing interest in the search for natural or synthetic compound binders with biomedical and drug discovery purposes, i.e., with potential as antimicrobials or antiparasitics. Given that marine resources are an extraordinary source of bioactive molecules, we screened marine invertebrates for new inhibitory compounds with such capabilities. In this work, we report the isolation and molecular and functional characterization of NpCI, a novel strong metallocarboxypeptidase inhibitor from the marine snail Nerita peloronta. NpCI was purified until homogeneity using a combination of affinity chromatography and RP-HPLC. It appeared as a 5921.557 Da protein with 53 residues and six disulphide-linked cysteines, displaying a high sequence similarity with NvCI, a carboxypeptidase inhibitor isolated from Nerita versicolor, a mollusc of the same genus. The purified inhibitor was determined to be a slow- and tight-binding inhibitor of bovine CPA (Ki = 1.1·× 10−8 mol/L) and porcine CPB (Ki = 8.15·× 10−8 mol/L) and was not able to inhibit proteases from other mechanistic classes. Importantly, this inhibitor showed antiplasmodial activity against Plasmodium falciparum in an in vitro culture (IC50 = 5.5 μmol/L), reducing parasitaemia mainly by inhibiting the later stages of the parasite’s intraerythrocytic cycle whilst having no cytotoxic effects on human fibroblasts. Interestingly, initial attempts with other related proteinaceous carboxypeptidase inhibitors also displayed similar antiplasmodial effects. Coincidentally, in recent years, a metallocarboxypeptidase named PfNna1, which is expressed in the schizont phase during the late intraerythrocytic stage of the parasite’s life cycle, has been described. Given that NpCI showed a specific parasiticidal effect on P. falciparum, eliciting pyknotic/dead parasites, our results suggest that this and related inhibitors could be promising starting agents or lead compounds for antimalarial drug discovery strategies.

The current and projected impacts of climate change are shaped by unprecedented rates of change in environmental conditions. These changes likely mismatch the existing coping capacities of organisms within-generations and impose challenges for population resilience across generations. To better understand the impacts of projected scenarios of climate change on organismal fitness and population maintenance, it is crucial to consider and integrate the proximate sources of variability of plastic and adaptive responses to environmental change in future empirical approaches. Here we explore the implications of considering: (a) the variability in different time-scale events of climate change; (b) the variability in plastic responses from embryonic to adult developmental stages; (c) the importance of considering the species life-history traits; and (d) the influence of trans-generational effects for individual survival and population maintenance. Finally, we posit a list of future challenges with questions and approaches that will help to elucidate knowledge gaps, to better inform conservation and management actions in preserving ecosystems and biodiversity.

Alzheimer’s disease (AD) is a complex disorder characterized by impaired neurotransmission in cholinergic and monoaminergic neurons, which, in combination with the accumulation of misfolded proteins and increased oxidative stress, leads to the typical features of the disease at the biomolecular level. Given the limited therapeutic success of approved drugs, it is imperative to explore rationally supported therapeutic approaches to combat this disease. The search for novel scaffolds that bind to different receptors and inhibit AD disease-related enzymes could lead to new therapeutic solutions. Here, we describe N-hydroxy-N-propargylamide hybrids 1–6, which were designed by combining the structures of Contilisant—a multifunctional anti-AD ligand—and ferulic acid, a natural antioxidant with various other biological activities. Among the synthesized compounds, we identified compound 4 as a micromolar inhibitor of hAChE with a potent radical-scavenging capacity comparable to resveratrol and Trolox. In addition, compound 4 chelated copper(II) ions associated with amyloid β pathology, mitochondrial dysfunction, and oxidative stress. The promising in vitro activity combined with favorable drug-like properties and predicted blood-brain barrier permeability make compound 4 a multifunctional ligand that merits further studies at the biochemical and cellular levels.

After a century of constant failure to produce an in vitro culture of the most widespread human malaria parasite Plasmodium vivax, recent advances have highlighted the difficulties to provide this parasite with a healthy host cell to invade, develop, and multiply under in vitro conditions. The actual level of understanding of the heterogeneous populations of cells—framed under the name ‘reticulocytes’—and, importantly, their adequate in vitro progression from very immature reticulocytes to normocytes (mature erythrocytes) is far from complete. The volatility of its individual stability may suggest the reticulocyte as a delusory cell, particularly to be used for stable culture purposes. Yet, the recent relevance gained by a specific subset of highly immature reticulocytes has brought some hope. Very immature reticulocytes are characterized by a peculiar membrane harboring a plethora of molecules potentially involved in P. vivax invasion and by an intracellular complexity dynamically changing upon its quick maturation into normocytes. We analyze the potentialities offered by this youngest reticulocyte subsets as an ideal in vitro host cell for P. vivax.

Despite the current knowledge of the devastating effects of external exposure to crude oil on animal mortality, the study of developmental, transgenerational effects of such exposure has received little attention. We used the king quail as an animal model to determine if chronic dietary exposure to crude oil in a parental population would affect morpho-physiological phenotypic variables in their immediate offspring generation. Adult quail were separated into three groups: (1) Control, and two experimental groups dietarily exposed for at least 3 weeks to (2) Low (800 PAH ng/g food), or (3) High (2,400 PAH ng/g food) levels of crude oil. To determine the parental influence on their offspring, we measured metabolic and respiratory physiology in exposed parents and in their non-exposed eggs and hatchlings. Body mass and numerous metabolic (e.g., O2 consumption, CO2 production) and respiratory (e.g., ventilation frequency and volume) variables did not vary between control and oil exposed parental groups. In contrast, blood PO2, PCO2, and SO2 varied among parental groups. Notably, water loss though the eggshell was increased in eggs from High oil level exposed parents. Respiratory variables of hatchlings did not vary between populations, but hatchlings obtained from High oil-exposed parents exhibited lower capacities to maintain body temperature while exposed to a cooling protocol in comparison to hatchlings from Low- and Control-derived parents. The present study demonstrates that parental exposure to crude oil via diet impacts some aspects of physiological performance of the subsequent first (F1) generation.

RNA sequencing was used to assemble transcriptome data for Philippine-reared silkworm and compare gene expression profiles of strains reared in high- and low-temperature environments. RNA was isolated from the silk glands of fifth instar larvae and mRNA-enriched libraries were sequenced using Illumina NextSeq 500. Transcriptome reads were assembled using reference-based and de novo assemblers, and assemblies were evaluated using different metrics for transcriptome quality, including the read mapping rate, E90N50, RSEM-eval, and the presence of single-copy orthologs. All transcriptome assemblies were able to reconstruct >40,000 transcripts. Differential expression analysis found 476 differentially expressed genes (DEGs; 222 upregulated, 254 downregulated) in strains reared in different temperatures. Among the top DEGs were myrosinase, heat shock proteins, serine protease inhibitors, dehydrogenases, and regulators of the juvenile hormone. Validation of some of the top DEGs using qPCR supported the findings of the in silico analysis. GO term enrichment analysis reveals an overrepresentation of GO terms related to nucleotide metabolism and biosynthesis, lipid and carbohydrate metabolic processes, regulation of transcription, nucleotide binding, protein binding, metal binding, catalytic activity, oxidoreductase activity, and hydrolase activity. The data provided here will serve as a resource for improving local strains and increasing silk production of Philippine-reared B. mori strains.

En este artículo, analizamos determinados elementos de las reformas educativas de los años noventa, el papel de las Tecnologías de la Información y la Comunicación (TIC) en educación y la adopción de un modelo de enseñanza-aprendizaje por competencias. Los objetivos del mismo son analizar estos elementos relacionados con las mencionadas reformas y proponer una serie de principios clave para orientar la enseñanza hacia una sociedad del aprendizaje. Concluimos que las instituciones educativas escolares pueden ser consideradas, en su naturaleza y modo de funcionamiento, instituciones propias de la modernidad, cuyo alumnado, sin embargo, presenta y demanda rasgos de la postmodernidad.

Este artículo muestra una experiencia de intervención educativa innovadora donde el alumnado acostumbraba solo a memorizar para aprobar el examen. El objetivo de este trabajo es exponer un proceso de enseñanza-aprendizaje y evaluación innovador basado en competencias desde una perspectiva integradora. Adicionalmente, observar y describir el “comportamiento” y las actitudes de los agentes implicados (profesorado y, fundamentalmente, alumnado), aplicando una metodología descriptiva. Para el desarrollo de la experiencia se aplican estrategias participativas y una evaluación integrada dentro de un enfoque de enseñanza-aprendizaje por competencias y se hace uso de hobbies e intereses del alumnado para plantear la asignatura de Innovación e investigación educativa en Educación Primaria con el alumnado de segundo año del Grado de Magisterio en Educación Primaria. Los resultados presentan un cambio de actitud del alumnado hacia las competencias de pensamiento analítico, reflexivo, crítico y creativo y hacia el trabajo autónomo y grupal y la motivación por aprender y ser competente, en un marco de negociación del currículum y el sistema de evaluación.

Many countries are immersed in several strategies to reduce the carbon dioxide (CO2) emissions of internal combustion engines. One option is the substitution of these engines by electric and/or hydrogen engines. However, apart from the strategic and logistical difficulties associated with this change, the application of electric or hydrogen engines in heavy transport, e.g., trucks, shipping, and aircrafts, also presents technological difficulties in the short-medium term. In addition, the replacement of the current car fleet will take decades. This is why the use of biofuels is presented as the only viable alternative to diminishing CO2 emissions in the very near future. Nowadays, it is assumed that vegetable oils will be the main raw material for replacing fossil fuels in diesel engines. In this context, it has also been assumed that the reduction in the viscosity of straight vegetable oils (SVO) must be performed through a transesterification reaction with methanol in order to obtain the mixture of fatty acid methyl esters (FAMEs) that constitute biodiesel. Nevertheless, the complexity in the industrial production of this biofuel, mainly due to the costs of eliminating the glycerol produced, has caused a significant delay in the energy transition. For this reason, several advanced biofuels that avoid the glycerol production and exhibit similar properties to fossil diesel have been developed. In this way, “green diesels” have emerged as products of different processes, such as the cracking or pyrolysis of vegetable oil, as well as catalytic (hydro)cracking. In addition, some biodiesel-like biofuels, such as Gliperol (DMC-Biod) or Ecodiesel, as well as straight vegetable oils, in blends with plant-based sources with low viscosity have been described as renewable biofuels capable of performing in combustion ignition engines. After evaluating the research carried out in the last decades, it can be concluded that green diesel and biodiesel-like biofuels could constitute the main alternative to addressing the energy transition, although green diesel will be the principal option in aviation fuel.
