The Bermuda petrel Pterodroma cahow is an island endemic seabird that belongs to the Procellariiformes, one of the most endangered orders of birds. Historical records suggest a significant population size decline following human settlement in Bermuda, bringing the species to near extinction. Since the 1950’s the population has been recovering aided by the implementation of an ongoing conservation plan. However, it still faces several threats, and negative genetic effects resulting from that drastic decline are to be expected, including inbreeding and genetic drift. We studied genetic diversity and levels of inbreeding, and their effects on individual fitness and mating choice. We also tested for a genetic signature of the recent demographic bottleneck. For this, we analysed variation in thousands of nuclear single nucleotide polymorphisms derived from double digest restriction site associated DNA sequencing and one mitochondrial gene (cytochrome oxidase I). The results revealed that the Bermuda petrel suffered a recent genetic bottleneck and shows low mitochondrial diversity compared to other petrel species. Conversely, nuclear diversity was similar to that of other endangered petrels. Inbreeding levels were not high overall, although some individuals were highly inbred. However, we found no evidence that individual inbreeding or relatedness between mates affected hatching success, or that mate choice is influenced by kinship in this very small population.

The White-faced Storm Petrel (WFSP) Pelagodroma marina has a widespread distribution, although virtually nothing is known about their feeding ecology and distributions at-sea. To describe their foraging areas, a total of 77 birds were equipped with 1 g-GPS loggers on Selvagem Grande, Madeira, Portugal (30° 09′ N, 15° 52′ W), during the 2018 and 2019 breeding seasons. We also assessed the diet of WFSP by analysing 17 faecal samples from chicks and 1 regurgitation from an adult using DNA metabarcoding techniques. Additionally, we collected body feathers from ten WFSP chicks to determine mercury concentration. WFSP fed mainly in deep oceanic waters, travelling up to 400 km from the colony, and did not concentrate in any well-defned, population-level foraging hotspots. Some individuals foraged along the edge of the shelf, near the African coast and the Canary Islands, especially during chick rearing. The duration of foraging trips and the total distance travelled, were, on average, 5.1 days and 723 km during the incubation period and 3.0 days and 578 km during chick rearing. The diet of WFSP was dominated by fsh and cephalopods (crustacean prey were not detected), with Myctophidae (FO=71%) representing the main fsh family. WFSP often consume mesopelagic fsh, in line with their preference for deep oceanic waters and with a small diference in at sea behavior (i.e., travel speed) between the diurnal and nocturnal period. The relatively high concentrations of mercury accumulated in body feathers of WFSP chicks (3.45±1.44 mg kg−1 dry weight; range 1.68–6.01 mg kg−1) support the idea that WFSP raise their chicks mostly on mesopelagic prey from deep pelagic areas. Olfactory receptors (ORs), encoded by the largest vertebrate multigene family, enable the detection of thousands of unique odorants in the environment and consequently play a critical role in species survival. Here, we advance our knowledge of OR gene evolution in procellariiform seabirds, an avian group which relies on the sense of olfaction for critical ecological functions. We built a cosmid library of Cory’s Shearwater (Calonectris borealis) genomic DNA, a model species for the study of olfaction-based navigation, and sequence OR gene-positive cosmid clones with a combination of sequencing technologies. We identified 220 OR open reading frames, 20 of which are full length, intact OR genes, and found a large ratio of partial and pseudogenes to intact OR genes (2:1), suggestive of a dynamic mode of evolution. Phylogenetic analyses revealed that while a few genes cluster with those of other sauropsid species in a γ (gamma) clade that predates the divergence of different avian lineages, most genes belong to an avian-specific γ-c clade, within which sequences cluster by species, suggesting frequent duplication and/or gene conversion events. We identified evidence of positive selection on full length γ-c clade genes. These patterns are consistent with a key role of adaptation in the functional diversification of olfactory receptor genes in a bird lineage that relies extensively on olfaction.

Dinamizaram-se sessões semanais com cerca de 60 participantes, entre os 20 e os 90 anos, organizados em 6 grupos heterogéneos. Desenvolveram-se actividades de leitura e escrita, baseadas no Método Paulo Freire, na Pedagogia da Autonomia, Andragogia e Aprendizagem Autodirigida, nos afectos e na literacia da vida de cada um. O projecto participa no Círculo de Alfabetização da APCEP, desenvolve uma comunidade de prática e oferece formação em alfabetização de adultos.

Trace elements' concentration in the ocean is fast growing and is a source of major concern. Being charismatic and at the top of food chains, seabirds are often used as biological monitors of contaminants. We studied the concentration of trace elements in blood of black-browed albatross from the Falklands Islands, which we here show, by tracking with geolocators, forage over most of the Patagonian Shelf. Levels of trace elements were measured in males and females from two different islands. Blood concentrations of trace elements were not significantly different between islands, which is consistent with observations from foraging behavior revealing that birds from both islands foraged in broadly the same areas in the months before sampling. Arsenic and selenium concentrations in females were higher than in males. Sex-related differences in the concentration of these elements may be related to unknown slight differences in diet or to differences in assimilation between sexes. These results provide reference values for monitoring elemental contamination in the Patagonian Shelf Large Marine Ecosystem using black-browed albatrosses, one of the most abundant top predators and a suitable sentinel for the region's environmental health. Diet studies are crucial for understanding the ecology and evolution of species, as well as for establishing appropriate conservation and management strategies. However, they remain methodologically challenging due to variation between seasons, sites, sexes or age groups and even variation between individuals. Due to method-specific characteristics and biases, a combination of existing techniques can overcome the inherent limitations of each technique and provide a more accurate and broad picture of species’ food preferences. Here, we examine diet information obtained using three different assessment methods to better understand the trophic ecology of the European Roller Coracias garrulus, a species targeted by conservation measures in Europe. First, we analysed regurgitated pellets and video recordings to report the diet composition of adult and nestling Rollers, respectively. Secondly, we used stable isotope analysis (SIA) to investigate adult sexual diet segregation as well as to confirm the main findings regarding adult and nestling diets obtained through conventional methods. Based on the analysis of pellets, the diet of adult Rollers was dominated by Coleoptera, while camera images revealed that the diet of nestlings was dominated by Orthoptera, mainly grasshoppers and bush crickets. Blood isotopic signatures of adult and nestling Rollers confirmed the results obtained through pellet and video recording techniques. Of the 45 three methods, pellet analysis contained the most comprehensive trophic information regarding the detectable prey spectrum and prey species contribution, and also provided basic diet information to inform the SIA. Our results also highlight the potential of SIA for assessing intra-specific variation in diet by sampling individuals of known age and sex, which is often unfeasible through conventional approaches. SIA analysis showed no differences in δ13C and δ15N ratios of blood between males and females and a high degree of overlap amongst isotopic niches, suggesting no sex-specific partitioning in resource use. Overall, we showed that the combination of different methods could be used to gain new and clearer insights into avian trophic ecology that are essential for informing habitat management aiming to improve availability of foraging resources.

The apparent scarcity or absence of blood parasites in some avian groups, such as seabirds, has been related to intrinsic and extrinsic factors including host immunological capacity, host-parasite assemblage, and ecological parameters, but also to reduced sensitivity of some methods to detect low parasite prevalence/intensities of infection. Here, we examined the haemosporidian parasite prevalence in a breeding population of Cory's shearwater Calonectris diomedea borealis, a long-distance migrant seabird, nesting in the Macaronesian region, in the Eastern Atlantic. Previous studies on Calonectris diomedea complex were based on small sample sizes providing weak evidence for a lack of infections by haemoparasites. Here, we investigated the presence of both parasite infections in C. d. borealis and larvae of potential mosquito vectors on the area. By employing a PCR-based assay, we extensively examined the prevalence of blood parasites belonging to the genera Plasmodium, Haemoproteus, and Leucocytozoon in 286 individuals from different life stages (i.e., chicks, immatures, sabbatical, and breeding adults), facing their specific energetic trade-offs (immunological functions vs. life history activities). We sampled immatures and adult shearwaters, of different sexes, ages, and migratory origins, from two sub-colonies. None of the sampled individuals were infected by these parasites, supporting the hypothesis that there was no in situ or ex situ transmission of vector-borne parasites in marine habitats irrespective of host's life stage and in spite of the presence of the potential Plasmodium vector Culiseta longiareolata breeding in the area. These results suggest that the lack of transmission of haemosporidian parasites on Selvagem Grande may be related to the lack of suitable dipteran vectors at the study sites, which may result from the geographic isolation of this area.

Procellariiform seabirds are among the avian species with the fastest rates of extinction due to interactions with fisheries and introduction of alien predators to the breeding colonies. Conservation and management policies targeting populations of these species must include information on colony demographics and levels of isolation and genetic markers go a long way toward providing reliable estimates of these parameters. To this end, we report isolation and characterization of 14 anonymous nuclear loci, with average length of 657 bp, in the pelagic seabird White-faced Storm-petrel Pelagodroma marina, a species for which there is virtually no genetic information available. These loci, initially isolated from a genomic library built from P. marina, were further tested, for a range of conditions, in 7 other species representing all Procellariiform families. We found high levels of cross-species amplification success, varying between 79% and 86% in representatives of Diomedeidae, Procellariidae, Pelecanoididae, and other Hydrobatidae. We also sequenced 11 loci for 22 P. marina individuals and report higher levels of anonymous genetic variation (π = 0.002), with an average of 1 single nucleotide polymorphism every 100 bp surveyed, relative to the levels found on a typically variable intron in avian species. These markers will be a valuable tool in future population genetics and phylogenetic studies, particularly of nonmodel seabird species.

Predation is a major ecological and evolutionary driver of natural populations, greatly influencing fitness and behaviour of prey species. Small, long-lived petrels are vulnerable to predation at the breeding colonies and are expected to evolve behavioural strategies to minimize predation risks. Using an automatic nest monitoring system and nightly aerial counts, we examined the effect of vegetation cover and moonlight on colony attendance patterns and levels of burrow activity of breeding thin-billed prions, Pachyptila belcheri, on New Island, Falkland Islands. We further investigated how these parameters were related to predation by Falkland skuas. We monitored up to 32 nests in two habitats, one with Tussock grass and one with low vegetation cover. Individuals in both areas were more active at the nest before hatching, and those breeding in the low cover habitat were more active and arrived at the colony earlier, which might reflect an effect of reaction time over predation risk. Nocturnal activity peaks shifted in time as the season progressed, indicating behavioural adjustments to sunrise hours. Moon phase did not affect attendance and activity levels of breeders in either habitat or overall aerial activity, but influenced arrival time at the colony during chick-rearing, individuals arriving later in periods of full moon. Skua capture rates were positively correlated with aerial and nest activity but not with overall breeder attendance and were unaffected by moon phase. Thin-billed prions activity budgets are influenced by environmental parameters that affect their likelihood of being predated.

The major histocompatibility complex encodes molecules that present foreign peptides to T cells of the immune system. The peptide binding region (PBR) of these molecules is among the most polymorphic regions found in vertebrate taxa. Genomic cloning approaches are improving our understanding of the evolution of this multigene family in nonmodel avian groups. By building a cosmid library, a new MHC class II B gene, Pabe-DAB1, was isolated and characterized at the genomic level in a sub-Antarctic seabird, the thin-billed prion (Pachyptila belcheri). Pabe-DAB1 exhibits the hallmark structural features of functional MHC class II loci. Direct sequencing of the PBR encoding exon in a panel of prions revealed significantly higher levels of genetic diversity compared to two noncoding neutral loci, with most alleles differing by at least one replacement substitution in the peptide binding codons. We estimated evolutionary dynamics for Pabe-DAB1 using a variety of Bayesian and other approaches. Evidence for balancing selection comes from a spatially variable ratio of nonsynonymous-to-synonymous substitutions (mean d N/d S = 2.87) in the PBR, with sites predicted to be functionally relevant exhibiting the highest ω values. We estimate the population recombination rate to be approximately 0.3 per site per generation, indicating an important role for recombination in generating polymorphism at this locus. Pabe-DAB1 is among the few avian class II loci characterized at the genomic level and with a known intron-exon structure, a feature that greatly facilitated the amplification and sequencing of a single MHC locus in this species.

Long-distance dispersal plays a critical role in population dynamics, particularly in species that occupy fragmented habitats, but it is seldom detected and investigated. The pelagic seabird Swinhoe's Storm-petrel, Oceanodroma monorhis, breeds exclusively in the NW Pacific. Individuals have been regularly observed in the Atlantic Ocean since the 1980s, but breeding has never been confirmed. In this study, we searched for evidence of breeding of Swinhoe's Storm-petrels on Selvagem Grande Island, NE Atlantic, between 2007 and 2013. During this period, six individuals were captured, sexed and characterized molecularly for two mitochondrial loci, cytochrome oxydase I and the control region, to confirm species identity, survey genetic diversity and estimate evolutionary relationships within the Hydrobatinae. These individuals were confirmed to be Swinhoe's Storm-petrels, and all except one are females. Phylogenetic analyses suggest sister relationship with Matsudaira's Storm-petrel and dismiss misidentifications with other dark rump species. Patterns of genetic variation suggest that dispersal occurred likely by more than a single female. Despite the record of a pair duetting in a burrow, breeding could not be confirmed. Swinhoe's Storm-petrels are regularly occurring at Selvagem Grande, but capture/recapture patterns suggest that a possible breeding population is small and likely not self-sustaining. In seabirds, long-distance dispersal events may facilitate colonization of new habitats created in the context of predicted climate change impacts on the marine ecosystems. Seabirds laying a single egg per season can regulate breeding investment during prelaying through size and composition of the egg, and during postlaying by adjusting parental care. Both stages can influence offspring growth and survival. Although likely not independent, attempts to measure the relative contributions of egg size per se and parental quality to offspring fitness are rare. To unravel the effects of both variables, we performed a cross-fostering experiment in thin-billed prions, where pairs laying large or small eggs were randomly given either size of eggs to raise. Nestling growth was followed through to fledging. Hatchlings from large eggs were structurally larger and heavier. Egg size effects, for the first time shown to last throughout the nestling period, influenced chick growth parameters and fledging probability. Large-egg chicks reached a significantly higher maximum growth rate and peak mass than chicks hatched from small eggs. There was also a relationship between the size of the egg laid by the foster parents and growth of their fostered offspring. Chicks raised by high-quality parents were heavier and in better body condition shortly after hatching, when mortality is high, and were provisioned at a frequency that minimized mass fluctuations during growth. Significant year effects and year*egg size/parental quality interactions suggest that contributions of egg size and parental quality to offspring fitness are contingent on interannual environmental variability. We discussed maintenance of egg size polymorphism in this population, where egg size repeatability was found to be high.

The evolutionary mechanisms underlying the geographic distribution of gene lineages in the marine environment are not as well understood as those affecting terrestrial groups. The continuous nature of the pelagic marine environment may limit opportunities for divergence to occur and lineages to spatially segregate, particularly in highly mobile species. Here, we studied the phylogeography and historical demography of two tropically distributed, pelagic seabirds, the Madeiran Storm-petrel Oceanodroma castro, sampled in the Azores, Madeira, Galapagos and Japan, and its sister species Monteiro's Storm-petrel O. monteiroi (endemic to the Azores), using a multi-locus dataset consisting of 12 anonymous nuclear loci and the mitochondrial locus control region. Both marker types support the existence of four significantly differentiated genetic clusters, including the sampled O. monteiroi population and three populations within O. castro, although only the mitochondrial locus suggests complete lineage sorting. Multi-locus coalescent analyses suggest that most divergence events occurred within the last 200,000years. The proximity in divergence times precluded robust inferences of the species tree, in particular of the evolutionary relationships of the Pacific populations. Despite the great potential for dispersal, divergence among populations apparently proceeded in the absence of gene flow, emphasizing the effect of non-physical barriers, such as those driven by the paleo-oceanographical environments, philopatry and local adaptation, as important mechanisms of population divergence and speciation in highly mobile marine species. In view of the predicted climate change impacts, future changes in the demography and evolutionary dynamics of marine populations might be expected.

Small burrowing petrels nesting on islands rarely survive introductions of mammalian predators. On New Island, a population of around two million pairs of thin-billed prions nests despite the presence of introduced ship rats, house mice and feral cats. Understanding the mechanisms of such coexistence is important, as it is important to establish a baseline for future monitoring. To do this, prion breeding success was determined for 7 years and in several habitats. Breeding success was high, except for the small fraction of the population that nests in tussock Poa flabellata stands, where several lines of evidence suggest significant predation by rats. Such high breeding success possibly resulted from predator swamping in this highly seasonal environment. This study suggests that introduced mammals do not currently depress thin-billed prion breeding success on New Island. However, cats and rodents might have future harmful effects if external factors depressed the prion population or allow a significant population growth of predators on New Island.

The conservation status of seabird populations has been deteriorating at a fast pace in recent decades. Studies of population genetic variation provide important insights regarding the evolutionary history of species, but also crucial information for conservation and management strategies. We report isolation and characterization of twelve anonymous nuclear loci in the seabird Madeiran Storm-Petrel Oceanodroma castro. These loci, on average 625 base pair (bp) long, showed high amplification success in eight other species representing all Procellariiform families. A polymorphism survey of these loci in O. castro from the Selvagem Grande I. summer population found an average anonymous genetic diversity of π = 0.0027 (1 SNP per 87 bp). Compared to the other small petrel populations breeding in Selvagem Grande I., O. castro shows the highest level of nuclear genetic diversity, despite its smaller census size. These markers will provide a valuable resource in future evolutionary and conservation studies of seabird species.

Analytical methods that apply coalescent theory to multilocus data have improved infer-ences of demographic parameters that are critical to understanding population divergenceand speciation. In particular, at the early stages of speciation, it is important to implementmodels that accommodate conflicting gene trees, and benefit from the presence of sharedpolymorphisms. Here, we employ eleven nuclear loci and the mitochondrial control regionto investigate the phylogeography and historical demography of the pelagic seabirdWhite-faced Storm-petrel (Pelagodroma marina)bysamplingsubspeciesacrossitsanti-tropical distribution. Groups are all highly differentiated: global mitochondrial ΦST= 0.89(P < 0.01) and global nuclear ΦSTvaries between 0.22 and 0.83 (all P < 0.01). The completelineage sorting of the mitochondrial locus between hemispheres is corroborated by approx-imately half of the nuclear genealogies, suggesting a long-term antitropical divergence inisolation. Coalescent-based estimates of demographic parameters suggest that hemisphericdivergence of P. marina occurred approximately 840 000 ya (95% HPD 582 000–1 170 000),in the absence of gene flow, and divergence within the Southern Hemisphere occurred190000ya(95%HPD96000–600 000), both probably associated with the profound palaeo-oceanographic changes of the Pleistocene. A fledgling sampled in St Helena (tropicalSouth Atlantic) suggests recent colonization from the Northern Hemisphere. Despite thegreat potential for long-distance dispersal, P. marina antitropical groups have beenevolving as independent, allopatric lineages, and divergence is probably maintained byphilopatry coupled with asynchronous reproductive phenology and local adaptation. Recently there has been evidence that contaminants other than organophosphate and carbamate pesticides may inhibit the activity of the enzyme acetylcholinesterase (AChE) both under in vitro and in vivo conditions. In this study we investigated the in vitro effect of three detergents \[dodecyl benzyl sulphonate (DBS), sodium dodecyl sulphate (SDS) and a mixture commonly used as domestic detergent (X)] and three metals \[molybdenum, barium and chromium (VI)] on AChE activity of Mytilus galloprovincialis haemolymph. All the detergents tested significantly inhibited the activity of the enzyme. The lowest observed effect concentrations were equal to 12 5 for DBS and 50 mg l-1 for SDS and X. Among the metals, molybdenum and barium had no effect on AChE activity, whereas chromium (VI) significantly depressed the activity of the enzyme at concentrations equal to or higher than 25 mg l-1. These results suggest that the use of AChE as a specific biomarker for organophosphate and carbamate pesticides should be questioned and that the use of this enzyme as a biomarker could be extended.

Most deep-water chondrichthyans occurring off mainland Portugal are distributed worldwide. There are many closely related species with similar morphology, and the lack of diagnostic characters that unambiguously allow species discrimination has led to extensive identification problems. Here, DNA barcodes were used to evaluate the suitability of cytochrome c oxidase I (COI) to identify deep-water chondrichthyans and to further contribute to the worldwide molecular databases currently in use. The sample was composed of 40 individuals from the families Chimaeridae, Centrophoridae, Somniosidae, Etmopteridae, Oxynotidae, Dalatiidae, Scyliorhinidae, Mitsukurinidae, Chlamydoselachidae and Hexanchidae. COI proved to be adequate for species identification, with almost all putative species recovered as well supported monophyletic clades, with low intraspecific variation. Sequence matches against the BOLD database provided evidence for the existence of identification problems particularly within the genera Hydrolagus, Galeus and Centrophorus. Present results suggest that three different Centrophorus species occur off the Portuguese mainland, C. squamosus, C. granulosus and C. uyato. The sample also included an albino Deania calcea and a C. squamosus specimen without dermal denticles. This study updates the current knowledge on diversity and distribution of deep-water chondrichthyans occurring along the Portuguese coast, and provides an essential tool for species identification and consistency of taxonomic designations worldwide.

In this study a comparison was made of the results obtained in Daphnia magna chronic bioassays after first-brood release and after 21 days of exposure, using inhibition of normal reproduction and growth as effect criteria and EC10, EC20, EC50, no-observed-effect concentration (NOEC), and lowest-observed-effect concentration (LOEC) as statistical parameters. Test substances were sodium bromide (NaBr), 3,4-dichloroaniline (DCA), cadmium, and parathion. For NaBr, DCA, and cadmium, toxicity evaluated after the first-brood release was similar to toxicity evaluated after 21 days, using reproduction as end point. Parathion did not affect either reproduction or growth. Thus, LC50, NOEC, and LOEC were calculated using mortality as the endpoint for parathion. Results indicate that the period until release of the first brood is sufficient to predict the toxicity of some chemicals to D. magna. Values estimated on the basis of a logistic model (EC10, EC20, and EC50) were more appropriate than NOECs and LOECs for evaluating toxicity of the test substances. Furthermore, classic endpoints used for the evaluation of chronic toxicity (inhibition of normal reproduction and growth) may not be adequate to evaluate the sublethal toxicity of compounds that induce cumulative effects leading to mortality within the test period and causing no observable effects on the reproduction and growth of the species. Endpoints indicative of biochemical stress or effects on specific targets of the test compound may be useful in sublethal toxicity evaluation.

Procellariiformes are well known for their low rates of food provisioning to their slow-growing chicks. In some species, the patterns of food delivery to chicks have been deduced from changes in their weight, obtained from periodic weighings. However, the behaviour of individual parents cannot be resolved using this method. In this study, we used a periodic weighing protocol with Cory's shearwater chicks on Selvagem Grande island, in the northeast Atlantic. In addition, we used an automatic logging system to examine the attendance of individual parents. In 1997, the chicks were fed infrequently, and were in significantly poorer condition, than in other years and at other colonies. This suggests that the adults were experiencing some difficulties in finding an adequate food supply close to the colony. Under these conditions, individual parents adopted a dual provisioning strategy, making both short and long foraging trips, a previously undescribed behaviour in any northern hemisphere petrel species. Although meals delivered to chicks were larger after long trips than after short trips, the average amount of food provisioned per day spent at sea decreased with increasing trip length, and so chicks did not benefit from longer trips. This finding suggests that long trips can be used to restore the adult's body condition, presumably depleted during short trips as shown previously for some petrels and albatrosses. The adoption of this flexible foraging strategy, which differs from the uniform intervals observed in Cory's shearwaters experiencing situations of 'normal' food abundance, may represent a mechanism through which breeding birds compromise between the needs of their chicks and the maintenance of their own body condition. We studied the diet of the Common Tern (Sterna hirundo) in several islands of the Azores during 1994, based on the analysis of 777 food pellets. The diet of Common Terns was based on a few species of small pelagic fish. Trumpet Fish (Macroramphosus spp.) dominated the diet in terms of both frequency of occurrence (84%) and numerical frequency (69%). This species, together with Boarfish (Capros aper), Blue Jack Mackerel (Trachurus picturatus) and lantern fish (Families Myctophidae, Sternoptychidae, Diretmidae), accounted for more than 96% of the prey taken by Common Terns on all the islands.
