Protein components of the spliceosome are highly conserved in eukaryotes and can influence several steps of the gene expression process. RSR-2, the Caenorhabditis elegans ortholog of the human spliceosomal protein SRm300/SRRM2, is essential for viability, in contrast to the yeast ortholog Cwc21p. We took advantage of mutants and RNA interference (RNAi) to study rsr-2 functions in C. elegans, and through genetic epistasis analysis found that rsr-2 is within the germline sex determination pathway. Intriguingly, transcriptome analyses of rsr-2(RNAi) animals did not reveal appreciable splicing defects but instead a slight global decrease in transcript levels. We further investigated this effect in transcription and observed that RSR-2 colocalizes with DNA in germline nuclei and coprecipitates with chromatin, displaying a ChIP-Seq profile similar to that obtained for the RNA Polymerase II (RNAPII). Consistent with a novel transcription function we demonstrate that the recruitment of RSR-2 to chromatin is splicing-independent and that RSR-2 interacts with RNAPII and affects RNAPII phosphorylation states. Proteomic analyses identified proteins associated with RSR-2 that are involved in different gene expression steps, including RNA metabolism and transcription with PRP-8 and PRP-19 being the strongest interacting partners. PRP-8 is a core component of the spliceosome and PRP-19 is the core component of the PRP19 complex, which interacts with RNAPII and is necessary for full transcriptional activity. Taken together, our study proposes that RSR-2 is a multifunctional protein whose role in transcription influences C. elegans development.

Lake Caviahue (37° 50 ‘S and 71° 06’ W; Patagonia, Argentina) is an extreme case of a glacial, naturally acidic, aquatic environment (pH ~ 3). Knowledge of the bacterial communities in the water column of this lake, is incipient, with a basal quantification of the bacterioplankton abundance distribution in the North and South Basins of Lake Caviahue, and the described the presence of sulfur and iron oxidizing bacteria in the lake sediments. The role that bacterioplankton plays in nutrient utilization and recycling in this environment, especially in the phosphorus cycle, has not been studied. In this work, we explore this aspect in further depth by assessing the diversity of pelagic, littoral and sediment bacteria, using state of the art molecular methods and identifying the differences and commonalties in the composition of the cognate communities. Also, we investigate the interactions between the sediments of Lake Caviahue and the microbial communities present in both sediments, pore water and the water column, to comprehend the ecological relationships driving nutrient structure and fluxes, with a special focus on carbon, nitrogen, and phosphorus. Two major environmental patterns were observed: (a) one distinguishing the surface water samples due to temperature, Fe2+, and electrical conductivity, and (b) another distinguishing winter and summer samples due to the high pH and increasing concentrations of N-NH4+, DOC and SO42−, from autumn and spring samples with high soluble reactive phosphorus (SRP) and iron concentrations. The largest bacterial abundance was found in autumn, alongside higher levels of dissolved phosphorus, iron forms, and increased conductivity. The highest values of bacterial biomass were found in the bottom strata of the lake, which is also where the greatest diversity in microbial communities was found. The experiments using continuous flow column microcosms showed that microbial growth over time, in both the test and control columns, was accompanied by a decrease in the concentration of dissolved nutrients (SRP and N-NH4+), providing proof that sediment microorganisms are active and contribute significantly to nutrient utilization/mobilization.

The destructive impact of fungi in agriculture and animal and human health, coincident with increases in antifungal resistance, underscores the need for new and alternative drug targets to counteract these trends. Cellular metabolism relies on many intermediates with intrinsic toxicity and promiscuous enzymatic activity generates others. Fuller knowledge of these toxic entities and their generation may offer opportunities of antifungal development. From this perspective our observation of media-conditional lethal metabolism in respiratory mutants of the opportunistic fungal pathogen Candida albicans was of interest. C. albicans mutants defective in NADH:ubiquinone oxidoreductase (Complex I of the electron transport chain) exhibit normal growth in synthetic complete medium. In YPD medium, however, the mutants grow normally until early stationary phase whereupon a dramatic loss of viability occurs. Upwards of 90% of cells die over the subsequent four to six hours with a loss of membrane integrity. The extent of cell death was proportional to the amount of BactoPeptone, and to a lesser extent, the amount of yeast extract. YPD medium conditioned by growth of the mutant was toxic to wild-type cells indicating mutant metabolism established a toxic milieu in the media. Conditioned media contained a volatile component that contributed to toxicity, but only in the presence of a component of BactoPeptone. Fractionation experiments revealed purine nucleosides or bases as the synergistic component. GC-mass spectrometry analysis revealed acetal (1,1-diethoxyethane) as the active volatile. This previously unreported and lethal synergistic interaction of acetal and purines suggests a hitherto unrecognized toxic metabolism potentially exploitable in the search for antifungal targets.

BackgroundIn Latin America (LA), the prevalence of dementia is expected to triple to 150 million people by 2050. The 2020 Lancet Commission report identified several modifiable dementia risk factors, yet few social and environmental factors, most relevant to vulnerable regions of LA, were highlighted in this report. We sought to assess the epidemiology of neurocognitive disorders (NCD) in Puente Piedra, one of the most socially and economically vulnerable districts of Lima, the capital of Peru.MethodologyThis was a cross-sectional door-to-door observational study that used two-stage household sampling. One young adult (30–59 years) and one older adult (>60 years) per household were enrolled. We collected demographic, clinical, and neurocognitive data. Addenbrooke’s Cognitive Examination (young adults) and the RUDAS-PE (older adults) were used, classifying participants as cognitively normal, possible mild NCD, or possible major NCD.ResultsWe enrolled 247 participants (median age 46 years; 67% female). One-fourth had not completed secondary school and more than 50% completed only secondary school. Most participants were housewives (46%) and 21% did not have health insurance. The overall prevalence of possible NCD was 30% (25.6 and 41.8% among younger adults and older adults, respectively). Among younger adults, those ages 55–59 years more frequently had NCD (70%) compared to younger age ranges. Among older adults, only 3 subjects (4.5%) had major NCD.ConclusionWe found a high frequency of possible NCDs in a socially and economically vulnerable community in Lima, Peru, with younger adults showing levels of NCD higher than expected. Our findings support the need for health systems to incorporate cognitive screenings programs for NCD in younger ages. Future research on NCD would include younger populations, particularly in vulnerable communities.

Background: Salicornia neei is a halophyte plant that has been proposed for use in the phytoremediation of the saline wastewater generated by land-based aquaculture. To identify the molecular mechanisms related to ammonium response, we analyzed the transcriptome of S. neei in response to growth in saline water containing 3 mM ammonium. Results: The RNA sequencing generated a total of 14,680,108 paired-end reads from the control and stressed conditions. De novo assembly using the CLC Genomic Workbench produced 86,020 transcripts and a reference transcriptome with an N50 of 683 base pair.A total of 45,327 genes were annotated, representing 51.2% of the contig predicted from de novo assembly. As regards differentially expressed genes, a total of 9,140 genes were differentially expressed in response to ammonium in saline water; of these, 7,396 could be annotated against functional databases. The upregulated genes were mainly involved in cell wall biosynthesis, transmembrane transport and antiporter activities, including biological Kyoto Encyclopedia of Genes and Genomes, pathways linked to the biosynthesis of secondary metabolites, plant hormone signal transduction, autophagy, and nitrogen metabolism. In addition, a set of 72 genes was directly involved in ammonium metabolism, including glutamine synthetase 1, glutamate synthase 1, and ferredoxin-dependent glutamate synthase chloroplastic. Conclusions: Our results support the hypothesis that an ammonium detoxification system mediated by glutamine and glutamate synthase was activated in S. neei when exposed to ammonium and saline water. Introducción: conocer la seguridad de las drogas actualmente disponibles para el tratamiento de las enfermedades reumáticas es muy importante al momento de tomar decisiones terapéuticas objetivas e individualizadas en la consulta médica diaria. Asimismo, datos de la vida real amplían el conocimiento revelado por los ensayos clínicos. Objetivos: describir los eventos adversos (EA) reportados, estimar su frecuencia e identificar los factores relacionados con su desarrollo. Materiales y métodos: se utilizaron datos BIOBADASAR, un registro voluntario y prospectivo de seguimiento de EA de tratamientos biológicos y sintéticos dirigidos en pacientes con enfermedades reumáticas inmunomediadas. Los pacientes son seguidos hasta la muerte, pérdida de seguimiento o retiro del consentimiento informado. Para este análisis se extrajeron datos recopilados hasta el 31 de enero de 2023. Resultados: se incluyó un total de 6253 pacientes, los cuales aportaron 9533 ciclos de tratamiento, incluyendo 3647 (38,3%) ciclos sin drogas modificadoras de la enfermedad biológicas y sintéticas dirigidas (DME-b/sd) y 5886 (61,7%) con DME-b/sd. Dentro de estos últimos, los más utilizados fueron los inhibidores de TNF y abatacept. Se reportaron 5890 EA en un total de 2701 tratamientos (844 y 1857 sin y con DME-b/sd, respectivamente), con una incidencia de 53,9 eventos cada 1000 pacientes/año (IC 95% 51,9-55,9). Estudiar la teoría del cuerpo en la arquitectura no sólo implica el cuestionamiento de su significado en la disciplina, también pone en entredicho las representaciones tras la producción y reproducción de los discursos hegemónicos que lo constituyen y guían. ¿Cómo se ha vinculado la teoría y práctica de la arquitectura al cuerpo? ¿Qué cuerpos teorizan sobre los cuerpos? Una revisión crítica y feminista de resulta vital a la hora de desmenuzar los viejos paradigmas disciplinares invitando a repensar en cuerpo y su representatividad en el desarrollo de la producción teórica en la arquitectura.

The sensitivity of some taxonomic groups to environmental conditions allows for their use as ecological state indicators. In ecological communities, this analysis is of interest to know which species or taxa are indicators of particular environment conditions, taking into account their presence in a set of sampling units. There are different approaches to estimate indicator species, in this work we develop a new tool for this task identifying indicator units defined as species (or taxa) whose observed frequencies are not uniform between environments or groups of sample units. The package provides methods to estimate the indicator species, the belonging of new samples to a given environment, and returns the frequency of taxonomic units in a selected combination of environmental factors as an estimation of the ecological niche. Previous studies have revealed that norrin can reverse vascular endothelial-growth-factor (VEGF)-induced permeability in a β-catenin-dependent pathway. Here, we have explored the contribution of disheveled-1 (DVL1) in norrin-induced blood-retinal barrier (BRB) restoration. We provide evidence that in addition to canonical signaling, DVL1 promotes tight junction (TJ) stabilization through a novel, non-canonical signaling pathway involving direct claudin-5 (CLDN5) binding. Immunofluorescence staining of rat retinal cross-sections showed enriched expression of DVL1 and 3 at endothelial capillaries and co-localization with CLDN5 and ZO-1 at the TJ complex in primary bovine retinal endothelial cells (BRECs). Barrier properties of BRECs were determined via measurements of trans-endothelial electrical resistance (TEER) or permeability to 70 kDa RITC-dextran. These studies demonstrated that norrin restoration of barrier properties after VEGF treatment required DVL1 as an siRNA knockdown of Dvl1 but not Dvl2 or Dvl3, reduced basal barrier properties and ablated norrin-induced barrier restoration. However, loss of Dvl1 did not decrease β-catenin signaling activity as measured by Axin2 mRNA expression, suggesting the contribution of a non-canonical pathway. DVL and TJ protein interactions were analyzed via co-immunoprecipitation of endogenous protein in BRECs, which demonstrated that DVL1 interacts with both CLDN5 and ZO-1, while DVL3 interacts only with ZO-1. These interactions were most abundant after inducing BRB restoration by treating BRECs with VEGF and norrin. DVL has previously been shown to form intramolecular bindings between the C-terminal PDZ-binding motif (PDZ-BM) with an internal PDZ domain. Co-transfection of HEK293 cells with DVL1 and CLDN5 or relevant mutants revealed that DVL1 interacts with CLDN5 through the DVL PDZ domain binding, CLDN5 PDZ-BM, in competition with DVL1 PDZ-BM, since DVL/CLDN5 interaction increases with deletion of the DVL1 PDZ-BM and decreases by co-expressing the C-terminal fragment of DVL1 containing the PDZ-BM or through deletion of CLDN5 PDZ-BM. In BREC cells, transfection of the C-terminal fragment of DVL1 downregulates the expression of CLDN5 but does not affect the expression of other proteins of the TJs, including ZO-1, occludin, CLDN1 or VE-cadherin. Blocking DVL1/CLDN5 interaction increased basal permeability and prevented norrin induction of barrier properties after VEGF. Combined with previous data, these results demonstrate that norrin signals through both a canonical β-catenin pathway and a non-canonical signaling pathway by which DVL1 directly binds to CLDN5 to promote barrier properties.

Digital leisure has become the main reason young people make use of the Internet and social media. Previous research shows the danger of certain activities in the online environment. Of particular concern are those of a recreational nature, which are more socially accepted by young people; among them one can find: online gambling and betting, online shopping and eGames, and the consumption of content on social media. This study aims to identify the behavioural and psychographic variables which impact the probability that young people will perceive the danger of these risky activities. We have carried out a descriptive and causal investigation with non-experimental cross-cutting analysis through a computer-assisted phone survey on a sample of 1500 young people aged between 18 and 35. The results show that all the activities are perceived as dangerous by the majority of those questioned, but a large percentage of young people do not perceive any risk in online gambling, betting and eGames. We have determined several psychographic and behavioural variables to help predict the perception of risk among young people to help define formal and informal policies for reducing their vulnerability in the event of the inappropriate use of the studied activities.

The study of human intestinal biology in healthy and diseased conditions has always been challenging. Primary obstacles have included limited tissue accessibility, inadequate in vitro maintenance and ethical constrains. The development of three-dimensional organoid cultures has transformed this entirely. Intestinal organoids are self-organized three-dimensional structures that partially recapitulate the identity, cell heterogeneity and cell behaviour of the original tissue in vitro. This includes the capacity of stem cells to self-renew, as well as to differentiate towards major intestinal lineages. Therefore, over the past decade, the use of human organoid cultures has been instrumental to model human intestinal development, homeostasis, disease, and regeneration. Intestinal organoids can be derived from pluripotent stem cells (PSC) or from adult somatic intestinal stem cells (ISC). Both types of organoid sources harbour their respective strengths and weaknesses. In this mini review, we describe the applications of human intestinal organoids, discussing the differences, advantages, and disadvantages of PSC-derived and ISC-derived organoids.

Abstract A feasibility analysis of tertiary treatment for Organic Liquid Agricultural Waste is presented using filamentous algae belonging to the genus Cladophora sp. as an alternative to chemical tertiary treatment. The main advantages of tertiary treatments that use biological systems are the low cost investment and the minimal dependence on environmental variables. In this work we demonstrate that filamentous algae reduces the nutrient load of nitrate (circa 75%) and phosphate (circa 86%) from the organic waste effluents coming from dairy farms after nine days of culture, with the added advantage being that after the treatment period, algae removal can be achieved by simple procedures. Currently, the organic wastewater is discarded into fields and local streams. However, the algae can acquire value as a by-product since it has various uses as compost, cellulose, and biogas. A disadvantage of this system is that clean water must be used to achieve enough water transparency to allow algae growth. Even so, the nutrient reduction system of the organic effluents proposed is friendly to the ecosystem, compared to tertiary treatments that use chemicals to precipitate and collect nutrients such as nitrates and phosphates.

Abstract Objective Distal sensory polyneuropathy (DSP) and neuropathic pain are important clinical concerns in virally suppressed people with HIV. We determined how these conditions evolved, what factors influenced their evolution, and their clinical impact. Methods Ambulatory, community‐dwelling HIV seropositive individuals were recruited at six research centers. Clinical evaluations at baseline and 12 years later determined neuropathy signs and distal neuropathic pain (DNP). Additional assessments measured activities of daily living and quality of life (QOL). Factors potentially associated with DSP and DNP progression included disease severity, treatment, demographics, and co‐morbidities. Adjusted odds ratios were calculated for follow‐up neuropathy outcomes. Results Of 254 participants, 21.3% were women, 57.5% were non‐white. Mean baseline age was 43.5 years. Polyneuropathy prevalence increased from 25.7% to 43.7%. Of 173 participants initially pain‐free, 42 (24.3%) had incident neuropathic pain. Baseline risk factors for incident pain included unemployment (OR [95% CI], 5.86 [1.97, 17.4]) and higher baseline body mass index (BMI) (1.78 [1.03, 3.19] per 10‐units). Participants with neuropathic pain at follow‐up had significantly worse QOL and greater dependence in activities of daily living than those who remained pain‐free. Interpretation HIV DSP and neuropathic pain increased in prevalence and severity over 12 years despite high rates of viral suppression. The high burden of neuropathy included disability and poor life quality. However, substantial numbers remained pain‐free despite clear evidence of neuropathy on exam. Protective factors included being employed and having a lower BMI. Implications for clinical practice include promotion of lifestyle changes affecting reversible risk factors.

Canine leishmaniosis is a challenge in veterinary medicine and no drug to date has achieved parasite clearance in dogs. Histone deacetylase inhibitors are a drug class widely used in cancer chemotherapy. We have successfully used O-alkyl hydroxamates (vorinostat derivatives) in the treatment of a laboratory model of visceral leishmaniasis without showing toxicity. In order to test the effectiveness of a particular compound, MTC-305, a parallel-group, randomized, single-centre, exploratory study was designed in naturally infected dogs. In this clinical trial, 18 dogs were allocated into 3 groups and were treated with either meglumine antimoniate (104 mg SbV/kg), MTC-305 (3.75 mg/kg) or a combination of both using a lower MTC-305 dose (1.5 mg/kg) through a subcutaneous route for 2 treatment courses of 30 days, separated by a 30-day rest period. After treatment, a follow-up time of 4 months was established. Parasite burden in bone marrow, lymph node and peripheral blood were quantified through qPCR. Antibody titres were determined through an immunofluorescence antibody test, and cytokine expression values were calculated through RT-qPCR. Treatment safety was evaluated through the assessment of haematological and biochemical parameters in blood, weight, and gastrointestinal alterations. Assessment was carried out before, between and after treatment series. Treatment with MTC-305 was effective at reducing parasite burdens and improving the animals’ clinical picture. Dogs treated with this compound did not present significant toxicity signs. These results were superior to those obtained using the reference drug, meglumine antimoniate, in monotherapy. These results would support a broader clinical trial, optimised dosage, and an expanded follow-up stage to confirm the efficacy of this drug.

We carried out the physicochemical characterization of extreme environments of acidic geothermal springs of the Copahue Thermal Complex, and isolated and cultivated algae used for therapeutic and medicinal purposes. Ecophysiological aspects, algal photosynthetic responses and potential toxicity of sediments were analysed. At the 15 sampling sites (pH: 2.0-6.7, conductivity: 283-3230 µS/cm, temperature: 22-60 °C), 11 Cyanobacteria species of a total of 24 algae were identified. The species richness was low with true inhabitants of highly acidic waters: Cyanidium caldarium, Euglena mutabilis, Chlamydomonas acidophila, Achnanthidium minutissimum, and Eunotia exigua, and cosmopolitan species of thermal springs: Mastigocladus laminosus, Leptolyngbia boryana and Phormidium tergestinum. All the species were well adapted to low light levels (15-55 µmol photon.m-2.s-1), and photosynthetic responses were similar to those in comparable environments. pH and temperature were important factors in algal distribution, and N:P relationship suggests that P is the limiting nutrient for algal growth. Fractions controlling P availability in the muds were those bound to organic matter, and Fe/Al oxyhydroxides. Healing muds have the ability to precipitate metals and would not be toxic. Cadmium and other potentially toxic metals were in very low concentrations and pose no risk for the human use as healing muds and bathing.

Los niños están presentes cada vez más en determinados medios digitales, donde los mensajes comerciales apelan directamente a su participación, facilitando expresar su identidad y establecer relaciones con sus iguales, que se convierten en líderes de opinión. El objetivo de este estudio exploratorio inicial y pionero en el formato digital en España es conocer la percepción que tienen las niñas sobre otras niñas sexualizadas en la publicidad de moda y en Internet, y los valores que asocian a ellas. La metodología empleada ha sido un estudio exploratorio cualitativo a una muestra de niñas españolas entre 8 y 11 años que corresponde al público objetivo de marcas de moda cuya publicidad ha sido categorizada en distintos grados como sexualizante. Los resultados señalan que las niñas rechazan aquellas imágenes de las modelos cuando son representadas de una forma más sexualizada porque no se corresponde con la vida real y no parece un estilo elegido sino impuesto. Además, las entrevistadas asocian la combinación de escenarios, posturas y gestos sexualizantes con rasgos de la personalidad que entienden como negativos –egoísta, desafiante, rebelde, aislada, triste-y, exponen el temor de que se normalice el uso de un tipo de ropa, maquillaje y comportamientos no acordes con su edad. A la luz de los resultados, se recomienda la reflexión ética de publicitarios que usan el estilo transgresor adulto en moda infantil y el estudio de imágenes de niños.

Abstract The diet of arthropodophagous bats can be influenced by several factors. Furthermore, its study is critical for understanding their role in the ecosystem as regulators of arthropod abundance. The aim of this study was to analyze the diet of 12 species beloging to two families of arthropodophagous bats from the Yungas Forests, Northwestern Argentina. We also evaluated differences in diet between well-preserved and disturbed sites, sexes, and seasons. The specimens were collected with mist nets in eight different localities, four well-preserved and four disturbed sites of the Yungas Forests. Through the analysis of feces, arthropod remains were identified until the lowest possible taxonomic level. Volume and frequency of occurrence percentages for each food item and the niche breadth for the species were estimated. A total of 475 samples from 12 species were analyzed and their diet contained arthropods from eight orders and seven families; the highest number of consumed arthropod orders were registered for Eptesicus diminutus. A low niche breadth was recorded in general for all species. The diet was significantly influenced by season and site characteristics only in E. diminutus and E. furinalis, respectively. This showed that bats could modify its diet according to the different habitats and seasons.

Las redes sociales plantean para los menores tanto situaciones de riesgo como respuestas positivas hacia su empoderamiento. El fenómeno social de la sexualización infantil, ya rechazado para las mujeres –por las connotaciones de discriminación, rol pasivo y tolerancia a la violencia–, se ha generalizado en Internet con aparente impunidad por parte de las marcas y medios que reproducen esas imágenes y débil concienciación entre las niñas y los padres que las publican en redes sociales. Ante la llamada de reguladores, padres e investigadores para proteger los derechos de la infancia, los objetivos de esta investigación son conocer el estado de la cuestión sobre la sexualización de la infancia en el entorno digital y recoger algunas propuestas de alfabetización mediática. La metodología utilizada ha sido la revisión de la literatura sobre sexualización, publicidad digital, comercialización de la infancia e infancia y moda en las bases de datos de revistas académicas y bibliotecas universitarias. Las conclusiones arrojan la necesaria contribución de prescriptores de opinión, medios, padres, educadores y anunciantes para evitar reducir la sexualidad a la sexualización y tomar conciencia del impacto de las imágenes comerciales que pueden generar conductas de riesgo.

Diferentes países han generado políticas para controlar la formación docente mediante regulaciones y rendición de cuentas desde los años 90. Sin embargo, existe escasa y contradictoria evidencia sobre los efectos de estas políticas. En Chile, la rendición de cuentas y la desregulación operan simultáneamente en la formación docente. En 2016 se aprobó la política de Sistema de Desarrollo Profesional Docente, que incluye la acreditación obligatoria de programas, el aumento gradual de la selectividad, la renovación de los estándares, y la implementación de pruebas diagnósticas para estudiantes de pedagogía. Este estudio cualitativo exploratorio analiza, a través de entrevistas semiestructuradas, cómo 40 directores de programas de pedagogía de 10 universidades chilenas ponen en práctica —interpretan y responden a— esta política. Utilizando un análisis de contenido cualitativo, se identificaron cuatro grupos de interpretaciones y respuestas, cuyas diferencias pueden entenderse con base en dimensiones contextuales que median las concepciones de profesionalismo, justicia y relaciones de poder expresadas por los participantes. Este estudio contribuye a comprender los elementos que influyen en la puesta en práctica de políticas de formación docente, a expandir la escasa literatura empírica en esta área, y a informar a quienes toman decisiones de política y forman docentes.

BackgroundDiagnosis of toxoplasmic encephalitis (TE) is challenging under the best clinical circumstances. The poor clinical sensitivity of quantitative polymerase chain reaction (qPCR) for Toxoplasma in blood and CSF and the limited availability of molecular diagnostics and imaging technology leaves clinicians in resource-limited settings with few options other than empiric treatment.Methology/principle findingsHere we describe proof of concept for a novel urine diagnostics for TE using Poly-N-Isopropylacrylamide nanoparticles dyed with Reactive Blue-221 to concentrate antigens, substantially increasing the limit of detection. After nanoparticle-concentration, a standard western blotting technique with a monoclonal antibody was used for antigen detection. Limit of detection was 7.8pg/ml and 31.3pg/ml of T. gondii antigens GRA1 and SAG1, respectively. To characterize this diagnostic approach, 164 hospitalized HIV-infected patients with neurological symptoms compatible with TE were tested for 1) T. gondii serology (121/147, positive samples/total samples tested), 2) qPCR in cerebrospinal fluid (11/41), 3) qPCR in blood (10/112), and 4) urinary GRA1 (30/164) and SAG1 (12/164). GRA1 appears to be superior to SAG1 for detection of TE antigens in urine. Fifty-one HIV-infected, T. gondii seropositive but asymptomatic persons all tested negative by nanoparticle western blot and blood qPCR, suggesting the test has good specificity for TE for both GRA1 and SAG1. In a subgroup of 44 patients, urine samples were assayed with mass spectrometry parallel-reaction-monitoring (PRM) for the presence of T. gondii antigens. PRM identified antigens in 8 samples, 6 of which were concordant with the urine diagnostic.Conclusion/significancesOur results demonstrate nanoparticle technology's potential for a noninvasive diagnostic test for TE. Moving forward, GRA1 is a promising target for antigen based diagnostics for TE.

Introduction: COVID-19 represents a serious threat to mental health worldwide. The aim of this study is to identify changes in adolescent psychiatry treatment demand in a tertiary hospital in Madrid during the first month (March 11 to April 11) after the pandemic declaration by the World Health Organization (WHO). We hypothesized that fear of contagion within COVID-19 may deter people from asking for psychiatric care.Method: The current study is retrospective, observational, and transversal. We reviewed the clinical records of 89 adolescents who went to the Emergency Room (ER) or were hospitalized at the Acute Inpatient Unit (AIU) at the Puerta de Hierro University Hospital-Majadahonda (PHUH-M) between March 11 and April 11. Socio-demographic, clinical, and demand variables were included in the study. Chi-square or Fisher exact tests were performed to compare categorical variables. We used the U Mann-Whitney test to compare quantitative variables. The level of statistical significance was set at p< 0.05. Analyses were conducted using SPSS v11.0.Results: The number of adolescents demanding psychiatric care at the ER dropped from 64 adolescents in 2019 to 25 in 2020. Similarly, psychiatric demand collapsed from 31 to 18 patients when comparing 2019 and 2020. Furthermore, the average hospital stay in 2020 trended toward a decrease when compared to 2019 (8.94 ± 4.87 vs. 14.32 ±10.23, p = 0.08). Self-injurious thoughts and behaviors were the most predominant reasons for consultation at both ER and AIU.Conclusion: The demand for adolescent psychiatric care decreased in the first month after the declaration of the pandemic. Our findings may be explained by (1) the fear of contagion, (2) the strict confinement measures, and (3) the initial shock as an adaptive reaction described in other disasters. Further studies are needed.

La violencia de la actual crisis social acontecida en Chile a partir del 18 de octubre de 2019 ha generado graves consecuencias en la ciudadanía y, en específico, sobre los diversos cuerpos de las personas manifestantes y presentes en el espacio público. Golpes, torturas, abusos sexuales, además del uso indiscriminado de balines y lanzamiento de bombas lacrimógenas a quemarropa construyen una nueva cartografía sobre el territorio: la de las corporalidades violentadas por organismos de seguridad del Estado. El siguiente artículo explora desde un ámbito urbano las relaciones corporales de manifestación y represión que tuvieron lugar en Plaza de la de Dignidad (antigua Plaza Italia). Por medio del ejercicio cartográfico, se develan las experiencias situadas: el surgimiento de nuevas corporalidades activas en el espacio y de sus repercusiones físico-simbólicas desplegadas en el entorno construído. Para ello se hará una revisión de los antecedentes recopilados en torno a las manifestaciones, tales como artículos de prensa, redes sociales, fotografías, testimonios, los que abrirán camino a la construcción de una cartografía corporal de la plaza y sus alrededores.

El presente artículo es parte de una investigación mayor que responde a los desafíos del Proyecto PMI-EXA-PNNI 01-17 DIUMCE-UMCE, desarrollado en el marco del fortalecimiento de la formación de profesores que buscó mecanismos para respaldar el logro de las competencias pedagógicas inscritas en las secuencias temporales de los diseños curriculares de las carreras de pedagogía en Chile. Mediante un enfoque cualitativo interpretativo, se realiza un análisis documental para establecer las relaciones semánticas y de contenidos entre un modelo teórico chileno, que se atreve a innovar en las competencias, el Marco para la Buena Enseñanza (MBE) y los Estándares de Desempeño Docente. Los resultados dan cuenta de un correlato entre el modelo chileno y los marcos ministeriales, en lo referido a la acción profesional; no obstante, existen otros elementos de igual importancia para el fortalecimiento de la Formación Inicial Docente (FID), asociados con las competencias sociales, de comunicación y de vocación laboral que están ausentes en el Marco para la Buena Enseñanza y los Estándares de Desempeño Docente Pedagógicos, a tener en cuenta.

Consumers are influenced by a multitude of stimuli, which affect their behaviour and guide their pref-erences towards a particular product. Ethnocentric tendencies are one of these stimuli, understood as consumers’ positive attitudes towards goods produced in their own region rather than those from other regions. From this perspective, the current study describes ethnocentric tendencies and identifies olive oil consumer preferences from Lisbon (Portugal). CETSCALE and the conjoint analysis technique have been used for this purpose. Results show that Portuguese olive oil consumers exhibit a strong ethnocentric ten-dency but the price attribute is also key. Nonetheless, there exists a less ethnocentric segment, on which foreign business strategy can be focused. On this segment, consumers attach lower relative importance to the price and higher relative importance to differentiated-quality parameters: oil type (extra virgin), bottle (glass) and production system (organic).

Abstract Fleas of Argentina are receiving renewed systematic interest, but the identification of many species associated with small mammals can be problematic. We review the taxonomy of the flea genus Agastopsylla including the re-description of two species and one subspecies, and designate neotype and neallotype for Agastopsylla hirsutior, neotype for Agastopsylla nylota nylota from the “Colección Mamíferos Lillo Anexos” (CMLA), Universidad Nacional de Tucumán, Argentina, and neotype and neallotype for Agastopsylla pearsoni from the Natural History Museum (London, U.K.). Additionally, a key to identification of the species of Agastopsylla and a distribution map of the species of the genus are included.

Este trabajo tiene como finalidad determinar si los libros de texto de la asignatura de Historia del 4º curso de ESO ponen de manifiesto la educación no sexista por la que aboga la ley educativa en vigor. Para ello, se ha llevado a cabo una investigación basada en los libros de texto empleados con mayor frecuencia en la ciudad de Sevilla, correspondientes con las editoriales Vicens Vives y Anaya. Se pretendía tomar conciencia de la inclusión o ausencia de personajes femeninos en los libros de texto en cuestión, establecer si se producía una discriminación hacia el colectivo femenino y analizar cómo se representan en ellos a las mujeres. Se ha elaborado un instrumento metodológico cuantitativo y otro cualitativo, cuyos resultados nos han permitido determinar la persistencia de una educación sexista. Hemos reflexionado acerca de si sería necesario realizar una revisión de los mismos desde una perspectiva de género.

Background: The history of immune suppression, especially CD4 nadir, has been shown to be a strong predictor of HIV-associated neurocognitive disorders (HAND). However, the potential mechanism of this association is not well understood. Methods: High resolution structural MRI images and neuropsychological data were obtained from fifty-nine HIV+ adults (mean age, 56.5 ± 5.8) to investigate the correlation between CD4 nadir and cortical thickness. Results: Low CD4 nadir was associated with widespread cortical thinning, especially in the frontal and temporal regions, and global mean cortical thickness correlated with CD4 nadir. In addition, worse global neurocognitive function was associated with bilateral frontal cortical thinning, and the association largely persisted (especially in the left frontal cortex) in the subset of participants who did not meet HAND criteria. Conclusions: These results suggest that low CD4 nadir may be associated with widespread neural injury in the brain, especially in the frontal and temporal regions. The diffuse neural injury might contribute to the prevalence and the phenotypes of HAND, as well as the difficulty treating HAND due to a broad network of brain regions affected. Low CD4 nadir related neural injury to the frontal cortex might contribute to subtle neurocognitive impairment/decline, even in the absence of HAND diagnosis.
