In recent years, corporate social responsibility has emerged as a pivotal factor for corporations in cultivating a favorable public perception. Despite this, many companies remain uncertain about the integration and rationale behind incorporating it into their core business strategies. This study aims to discern dissimilarities in implementing corporate social responsibility initiatives between Slovak and Czech companies, addressing the triple bottom line framework. This approach is a metric for evaluating a company’s achievement across three dimensions: social, environmental, and economic. The study is predicated on a questionnaire survey conducted among a sample of 180 Slovak and 180 Czech companies operating as subsidiaries of multinational corporations as typical implementers of corporate social responsibility activities in emerging economies. Following the triple bottom line framework, three distinct sets of variables were formulated to account for social, environmental, and economic activities using the Mann-Whitney U Test to ascertain statistically significant disparities between Slovak and Czech companies. The findings indicate that the surveyed companies from monitored countries exhibited no notable distinctions in environmental endeavors. However, discrepancies were discerned in economic activities, specifically relating to post-sales or post-warranty customer services, which was significant with a small effect size (0.19). In social activities, significant differences were observed concerning adherence to the principle of equal opportunities in the workplace. Based on these outcomes, it is recommended that future investigations delve more extensively into the monitored parameters within the Visegrad countries. Purpose – find differences in the core principles of implementation of CSR (Corporate Social Responsibility) practices in subsidiaries of foreign multinationals in Slovak and Czech Republic, two similar yet different countries. Research methodology – the questionnaire was distributed to local daughter companies of foreign multinational enterprises selected on the basis of stratified randomization. The sample: 360 enterprises (180 from each country). Mann‐Whitney U Test was used for the analysis (non-normally distributed data) of the differences between Slovak and Czech companies in their CSR implementation. Findings – differences among the two countries in the implementation of CSR exist in few factors due to their joint history. Research limitations – the representativeness of companies connected with foreign mother companies in the two countries cannot be computed: no data are available. To overcome this issue, the same number of monitored entities was analysed to so as to make further statistical analysis possible. Practical implications – the results can be used by business entities, which may benefit from comparing themselves against other players on the market or against the average situation on the monitored markets. Moreover, teaching future managers and promoters CSR is of significant importance. Originality/Value – the work sheds lights on CSR practices in Czechia and Slovakia, two countries in which there is a lack of studies on the topic of CSR.

In spite of geographical and culture differences between examined countries, there can be found similarities in consumer behavior of men and woman and also the similar tendencies on the dairy product market. In the last decade different fields of science concerns with the topic of gender differences more frequently. The article is based on a research of consumers' overall attitude to dairy products in Slovakia and Russia. The important role of gender differences underlines the outcomes of the questionnaire survey. Kruskal-Wallis test and Bonferroni correction was applied to verify the hypothesis whether there is a dependency between gender of the respondents and their attitude while choosing the dairy products in both countries. Analysis showed that in both countries women tend to buy dairy products more often than men. Also consumers prefer more domestic products, but Russian not as significantly as Slovakian. In both countries consumers consider the price of dairy products as high, but they don”™t outline the price as the most important factor when choosing dairy products. At the same time, both genders consider quality as the most important factor. These results indicate the existence of a niche at the Russian market, which could be used by Slovak dairy producers who can possibly penetrate Russian market. In addition, the similar marketing strategy for both, Slovak and Russian market can be used if the advertisement will be sensitively focused on the gender.

Current system of direct payments in Slovakia can be described as per hectare payment. It is based on the logical assumption that the more land farmers cultivates, the more support they need. However, it seems like this principle works differently among EU member states. Historically, Slovakia is a country with the largest farms in EU 28. This extreme big physical farm size is here connected with the lowest output among EU 28 and simultaneously Slovak farms display also lowest efficiency. Paper examine generally accepted assumption that the more hectares farms utilize, the more subsidies they should receive, to help achieve more output. Research is based on the mutual pair combined correlation analysis, which examined relationship between utilized agricultural area, total output and total subsidies. Surprisingly just the relationship between total subsidies and total output was proved to be positive and in a moderate manner. Relation of total subsidies and utilised agricultural area, respectively total output and utilised agricultural area show the inverse relationship. In spite of the development in most European countries, Slovak outcomes indicates that the more subsidies farmers receive, the less output they achieve. This paradox can be caused by the actual Common Agricultural Policy system of subsidies remuneration which is not necessary suitable for whole EU 28 on the same level.

Pancreatic ductal adenocarcinoma (PDAC) is among the deadliest cancers worldwide, primarily due to its robust desmoplastic stroma and immunosuppressive tumor microenvironment (TME), which facilitate tumor progression and metastasis. In addition, fibrous tissue leads to sparse vasculature, high interstitial fluid pressure, and hypoxia, thereby hindering effective systemic drug delivery and immune cell infiltration. Thus, remodeling the TME to enhance tumor perfusion, increase drug retention, and reverse immunosuppression has become a key therapeutic strategy. In recent years, targeting epigenetic pathways has emerged as a promising approach to overcome tumor immunosuppression and cancer progression. Moreover, the progress in nanotechnology has provided new opportunities for enhancing the efficacy of conventional and epigenetic drugs. Nano-based drug delivery systems (NDDSs) offer several advantages, including improved drug pharmacokinetics, enhanced tumor penetration, and reduced systemic toxicity. Smart NDDSs enable precise targeting of stromal components and augment the effectiveness of immunotherapy through multiple drug delivery options. This review offers an overview of the latest nano-based approaches developed to achieve superior therapeutic efficacy and overcome drug resistance. We specifically focus on the TME and epigenetic-targeted therapies in the context of PDAC, discussing the advantages and limitations of current strategies while highlighting promising new developments. By emphasizing the immense potential of NDDSs in improving therapeutic outcomes in PDAC, our review paves the way for future research in this rapidly evolving field.

Decitabine (DAC), a DNA methyltransferase (DNMT) inhibitor, is tested in combination with conventional anticancer drugs as a treatment option for various solid tumors. Although epigenome modulation provides a promising avenue in treating resistant cancer types, more studies are required to evaluate its safety and ability to normalize the aberrant transcriptional profiles. As deoxycytidine kinase (DCK)-mediated phosphorylation is a rate-limiting step in DAC metabolic activation, we hypothesized that its intracellular overexpression could potentiate DAC’s effect on cell methylome and thus increase its therapeutic efficacy. Therefore, two breast cancer cell lines, JIMT-1 and T-47D, differing in their molecular characteristics, were transfected with a DCK expression vector and exposed to low-dose DAC (approximately IC20). Although transfection resulted in a significant DCK expression increase, further enhanced by DAC exposure, no transfection-induced changes were found at the global DNA methylation level or in cell viability. In parallel, an integrative approach was applied to decipher DAC-induced, methylation-mediated, transcriptomic reprogramming. Besides large-scale hypomethylation, accompanied by up-regulation of gene expression across the entire genome, DAC also induced hypermethylation and down-regulation of numerous genes in both cell lines. Interestingly, TET1 and TET2 expression halved in JIMT-1 cells after DAC exposure, while DNMTs’ changes were not significant. The protein digestion and absorption pathway, containing numerous collagen and solute carrier genes, ranking second among membrane transport proteins, was the top enriched pathway in both cell lines when hypomethylated and up-regulated genes were considered. Moreover, the calcium signaling pathway, playing a significant role in drug resistance, was among the top enriched in JIMT-1 cells. Although low-dose DAC demonstrated its ability to normalize the expression of tumor suppressors, several oncogenes were also up-regulated, a finding, that supports previously raised concerns regarding its broad reprogramming potential. Importantly, our research provides evidence about the involvement of active demethylation in DAC-mediated transcriptional reprogramming.

Due to abundant stroma and extracellular matrix, accompanied by lack of vascularization, pancreatic ductal adenocarcinoma (PDAC) is characterized by severe hypoxia. Epigenetic regulation is likely one of the mechanisms driving hypoxia-induced epithelial-to-mesenchymal transition (EMT), responsible for PDAC aggressiveness and dismal prognosis. To verify the role of DNA methylation in this process, we assessed gene expression and DNA methylation changes in four PDAC cell lines. BxPC-3, MIA PaCa-2, PANC-1, and SU.86.86 cells were exposed to conditioned media containing cytokines and inflammatory molecules in normoxic and hypoxic (1% O2) conditions for 2 and 6 days. Cancer Inflammation and Immunity Crosstalk and Human Epithelial to Mesenchymal Transition RT² Profiler PCR Arrays were used to identify top deregulated inflammatory and EMT-related genes. Their mRNA expression and DNA methylation were quantified by qRT-PCR and pyrosequencing. BxPC-3 and SU.86.86 cell lines were the most sensitive to hypoxia and inflammation. Although the methylation of gene promoters correlated with gene expression negatively, it was not significantly influenced by experimental conditions. However, DNA methyltransferase inhibitor decitabine efficiently decreased DNA methylation up to 53% and reactivated all silenced genes. These results confirm the role of DNA methylation in EMT-related gene regulation and uncover possible new targets involved in PDAC progression.
