Limited research examines the process of leading innovative school change from the perspective of principals. In this case study, three school principals implement a novel school model, franchise model schools. Using interviews, the researchers examined the successes and challenges principals experienced while implementing and leading this change. Data analysis revealed three significant challenges leading franchise model schools: (1) low teacher morale at the inception of the model; (2) time management; and (3) alignment among franchise campuses. However, three significant successes surfaced. Franchise principals were able to increase (1) community engagement, (2) autonomy and creativity, and (3) the sharing of best practices. Authors recommend more research context-specific to the school's culture and a principal's level of experience in leading change. Researchers should identify successful innovative schools and examine what change models were used by those principals.

The purpose of this study is to examine the network effect of school principals as it relates to school improvement. Network practices of school principals are compared to an innovative practice for improving networking practices. Through descriptive statistics and chi-square goodness of fit, we illustrate the difference between what school principals do concerning their networking practices for school improvement compared to an innovative ideal approach for using network working for school improvement. Findings indicate there is a statistically significant difference between school principals' networking practices in comparison to ideal networking practices for school improvement. There are also differences between who school principals seek out for ideas and who they seek out for feedback concerning their school improvements. Further discussion informs how the next generation of school principals can be equipped with innovative skills for tackling 21st-century school improvement issues.

This study examines how financial literacy education (FLE) was implemented at the school and classroom levels. A total of 32 teachers and 16 school leaders from 16 public schools across different regions in the Philippines were interviewed. Using a comparative multiple case study approach, the findings indicated that (1) teachers' implementation in the classroom was influenced by the various learner, learning, and contextual factors and that (2) school leaders confronted major implementation challenges related to staff development, instructional supervision, administration, and leadership functions despite recognizing the value of FLE. Despite implementation challenges, the study also revealed teachers' and school leaders' positive attitudes toward incorporating FLE into the K-12 curriculum. Theoretically, the findings highlight the value of teacher and school leader sense-making in successfully implementing any curriculum innovation and crafting the FLE framework. Implications for curriculum implementation and policymaking are discussed.

Insect vectors transmit many plant viruses of agricultural importance. In most cases, these viruses manipulate their vectors' behavior and movement leading to vector settling and feeding preferences that influence virus spread. The latent period within the insect vector is also crucial in virus transmission during vector feeding, however is assumed to be negligible in previous studies. This paper proposes a plant-virus model with a focus on vector settling preference for virus-infected plants and considers the latent periods in both the plant and insect vector populations by introducing them as time delays. We analyze the existence and local stability of the equilibrium solutions using the basic reproduction number of the proposed model which is inversely related to the saturation parameter due to vector preference. We investigate the effects of the time delays on the local stability of equilibria and the possible emergence of stable limit-cycle solutions. Moreover, our theoretical results are corroborated using numerical continuation and bifurcation analysis to provide more insights into how the latent periods and vector preference affect the system's dynamics. Particularly, we show numerically that increasing the saturation parameter due to preferential settling reduces the number of new infections, if not eradicates the infection in the system.

This qualitative study, grounded in descriptive phenomenology, investigated the lived experiences of students studying in a private school under the Central Visayan Institute Foundation – Dynamic Learning Program (CVIF-DLP) framework. Through eight focus group discussions involving 62 participants analyzed with the use of thematic analysis, the research explored the advantages, disadvantages, and transformative potential of the CVIF-DLP approach outlined to the three learning domains. Findings revealed that students focus first on the tangible aspects of learning such as learning activity sheet and writing. They also believed that CVIF-DLP has limited special impact on their physical aspects but affects greatly their cognitive and affective aspects. Despite the varied perspectives and encountered challenges, students agreed that the program helps them improve intellectually and personally. It revealed the complex dynamics shaping student learning journeys, emphasizing the need for ongoing program refinement to address student needs effectively. The study offered valuable insights into educational innovation and pedagogical practices, guiding future efforts to enhance the educational experiences of the implementers of CVIF-DLP.

Marine co-location, i.e., multiple fixed ocean activities operating in the same place and at the same time, can maximize the space- and resource-use efficiency in crowded seascapes. While interest grows, commercial use is nascent and the collective benefits or limitations of co-locating aquatic food and clean energy remains scattered throughout the literature. In this study, we synthesize multinational findings of co-location scientific publications (N = 102) to better understand the patterns and knowledge gaps at the co-located ocean food-energy nexus. We track and compare food (aquaculture) and energy (tidal, offshore wind, and wave) co-located ocean activities, noting the focus (e.g., ecological), motivation (e.g., impact/risk), and assessment type (e.g., modeling), as well as nine key metrics of interest (depth, distance from shore, aquaculture yield, etc.), mainly for aquaculture co-location. We found the number of annual co-location publications increased over time and space but are largely concentrated in the North Sea (n = 39). We also found about half of publications include aquaculture, one-third of publications report at least one metric – reporting aquaculture yield was particularly rare (n = 1) – and few studies focused on impact/risk (n = 7). However, conducting a targeted post-hoc evaluation of North Sea gray literature (N = 61), due to this region’s importance in the field, showed more coverage of impacts/risk (e.g., liability) and similar attention to aquaculture. Of the scientific papers that did report metrics, the ranges of depth and distance exceeded those reported for standalone sectors, indicating co-location could be facilitating a “push” of ocean activities into farther offshore and/or deeper exposed waters. Ultimately, while aquaculture is commonly cited in the co-location literature, the shortage of metrics, like aquaculture yield, and possible impact/risk evaluations – though gray literature can provide critical insights – emphasizes the need for knowledge sharing and modeling to address and explore the uncertainty, especially for co-located aquaculture production. This study provides a needed snapshot of marine co-location, particularly in emerging regions, highlighting gaps in understanding aquaculture-energy potential in the oceans.

Abstract Background Variation in immune response to COVID-19 vaccines is observed among different ethnicities. We aimed to describe the reinfection rates, change in antibody titers, and adverse events among Filipinos. Methods This is a secondary analysis of a cohort study of 307 participants within one year of having COVID-19 infection. We measured COVID-19 antibody levels at pre-determined timepoints (Days 21, 90, 180, 270, and 360 from initial infection). We monitored for COVID-19 symptoms and obtained details on COVID-19 vaccination. An adjudication committee classified the participants as probable, possible, or unlikely COVID-19 reinfection. We determined the probable reinfection rate, adverse events, and the geometric mean titer (GMT) ratio of pre- and post-vaccination antibody levels according to type and brand of COVID-19 vaccine. Results At the end of the follow-up period, 287 (93.5%) out of 307 study participants were fully vaccinated, 1 was partially vaccinated (0.3%), and 19 were unvaccinated (6.2%). Among the fully vaccinated participants, those given mRNA vaccines had the lowest reinfection rate (19.2 cases/100 person-years, 95% CI 9.6, 38.4), followed by viral vector vaccines (29.8 cases/100 person-years, 95% CI 16.9, 52.4). We observed the highest reinfection rate among those given inactivated virus vaccines (32.7 cases/100 person-years, 95% CI 23.6, 45.3). The reinfection rate was 8.6 cases/100 person-years (95% CI 4.1, 17.9) for unvaccinated participants and 3.6 cases/100 person-years (95% CI 0.5, 25.3) for partially vaccinated participants. We observed the largest rise in antibody titers among those given mRNA vaccines (GMT ratio 288.5), and the smallest rise among those given inactivated virus vaccines (GMT ratio 16.7). We observed the highest percentage of adverse events following immunization with viral vector vaccines (63.8%), followed by mRNA vaccines (62.7%), and the lowest for inactivated virus vaccines (34.7%). No serious adverse events were reported. Conclusion Vaccinees given the mRNA vaccines had the lowest reinfection rate and the highest rise in antibody titers. Vaccinees given inactivated virus vaccines had the highest reinfection rate, smallest rise in antibody titers, and lowest percentage of adverse events. The small sample size and imbalanced distribution of the type of vaccines received limits the external generalizability of our results. Epithelial tissue regeneration may be favored if the tissue receives both therapeutic agents such as recombinant human epidermal growth factor (rhEGF) and, simultaneously, antibacterial materials capable of reducing the risk of infections. Herein, we synthesized silver nanoparticles (AgNPs), which are well-known antibacterial materials, and impregnate them with rhEGF in order to study a bio-nanomaterial of potential interest for epithelial tissue regeneration. A suspension of Ag NPs is prepared by the chemical reduction method, employing sodium citrate as both a reducer and capping agent. The AgNPs suspension is mixed with a saline solution containing rhEGF, producing rhEGF-coated Ag NPs with rhEGF loadings between 0.1 and 0.4% w/w. ELISA assays of supernatants demonstrate that, in all studied cases, over 90% of the added rhEGF forms part of the coating, evidencing a high efficiency in impregnation. During the preparation of rhEGF-coated Ag NPs, no significant changes are observed on the nanoparticles, which are characterized by UV-Vis spectroscopy, transmission electron microscopy (TEM) and infrared spectroscopy. The liberation of rhEGF in vitro was followed for 72 h, finding that approximately 1% of rhEGF that is present is released. The rhEGF-coated AgNPs shows antibacterial activity against E. coli, although such activity is decreased with respect to that observed from naked AgNPs. Having confirmed the possibility of simultaneously liberating rhEGF and reducing the proliferation of bacteria, this work helps to support the use of rhEGF-loaded metallic nanoparticles for tissue regeneration.

Despite improvements in cardiopulmonary resuscitation (CPR), survival and neurologic recovery after cardiac arrest remain poor due to ischemia and subsequent reperfusion injury. As the likelihood of survival and favorable neurologic outcome decreases with increasing severity of ischemia during CPR, developing methods to measure the magnitude of ischemia during resuscitation is critical for improving overall outcomes. Cerebral oximetry, which measures regional cerebral oxygen saturation (rSO2) by near‐infrared spectroscopy, has emerged as a potentially beneficial marker of cerebral ischemia during CPR. In numerous preclinical and clinical studies, higher rSO2 during CPR has been associated with improved cardiac arrest survival and neurologic outcome. There is also emerging evidence that this can be integrated with electroencephalogram (EEG) monitoring to provide a bimodal system of brain monitoring during CPR. In this method’s review, we discuss the feasibility, application, and implications of this integrated monitoring approach, highlighting its significance for improving clinical outcomes in cardiac arrest management and guiding future research directions.

Individuals with Parkinson’s disease (PD) have a higher risk of developing dementia compared to age-matched controls. Rapid eye movement sleep behavior disorder (RBD) and hyposmia can influence symptoms severity. We report associations between polysomnography-assessed sleep architecture, olfactory identification, and cognition. Twenty adults with early-stage PD (mean age 69 ± 7.9; 25% female) completed cognitive assessments, the Brief Smell Identification Test (BSIT), and overnight in-clinic polysomnography. A global cognitive score was derived from principal component analysis. Linear regression models examined associations between sleep variables, BSIT performance, and cognition. Cognitive performance was compared between participants with and without RBD. Deep sleep attainment (β ± SE: 1.18 ± 0.45, p = .02) and olfactory identification (0.37 ± 0.12, p = .01) were associated with better cognition. Light sleep, REM sleep, arousal index, and sleep efficiency were not (all p > .05). Participants with RBD had significantly worse cognition ( t -test = −1.06 ± 0.44, p = .03) compared to those without RBD; none entered deep sleep. Deep sleep attainment was associated with better memory (1.20 ± 0.41, p = .01) and executive function (2.94 ± 1.13, p = .02); sleep efficiency was associated with executive function (0.05 ± 0.02, p = .02). These findings suggest interrelationships between lack of deep sleep, hyposmia, and poorer cognition in PD, particularly among individuals with RBD. Assessing these markers together may improve early identification of high-risk individuals and access to interventions.

Abstract Background Preclinical studies highlight the importance of endogenous cannabinoids (endocannabinoids; eCBs) in neurodegeneration. Yet, prior observational studies focused on limited outcome measures and assessed only few eCB compounds while largely ignoring the complexity of the eCB system. We examined the associations of multiple circulating eCBs and eCB-like molecules with early markers of neurodegeneration and neuro-injury and tested for effect modification by sex. Methods This exploratory cross-sectional study included a random sample of 237 dementia-free older participants from the Framingham Heart Study Offspring cohort who attended examination cycle 9 (2011–2014), were 65 years or older, and cognitively healthy. Forty-four eCB compounds were quantified in serum, via liquid chromatography high-resolution mass spectrometry. Linear regression models were used to examine the associations of eCB levels with brain MRI measures (i.e., total cerebral brain volume, gray matter volume, hippocampal volume, and white matter hyperintensities volume) and blood biomarkers of Alzheimer’s disease and neuro-injury (i.e., total tau, neurofilament light, glial fibrillary acidic protein and Ubiquitin C-terminal hydrolase L1). All models were adjusted for potential confounders and effect modification by sex was examined. Results Participants mean age was 73.3 ± 6.2 years, and 40% were men. After adjustment for potential confounders and correction for multiple comparisons, no statistically significant associations were observed between eCB levels and the study outcomes. However, we identified multiple sex-specific associations between eCB levels and the various study outcomes. For example, high linoleoyl ethanolamide (LEA) levels were related to decreased hippocampal volume among men and to increased hippocampal volume among women (β ± SE = − 0.12 ± 0.06, p = 0.034 and β ± SE = 0.08 ± 0.04, p = 0.026, respectively). Conclusions Circulating eCBs may play a role in neuro-injury and may explain sex differences in susceptibility to accelerated brain aging. Particularly, our results highlight the possible involvement of eCBs from the N-acyl amino acids and fatty acid ethanolamide classes and suggest specific novel fatty acid compounds that may be implicated in brain aging. Furthermore, investigation of the eCBs contribution to neurodegenerative disease such as Alzheimer’s disease in humans is warranted, especially with prospective study designs and among diverse populations, including premenopausal women.

Background: Tibial stress fracture (TSF) is an overuse running injury with a long recovery period. While many running studies refer to biomechanical risk factors for TSF, only a few have compared biomechanics in runners with TSF to controls. The aim of this systematic review and meta-analysis was to evaluate biomechanics in runners with TSF compared to controls. Methods: Electronic databases PubMed, Web of Science, SPORTDiscus, Scopus, Cochrane, and CINAHL were searched. Risk of bias was assessed and meta-analysis conducted for variables reported in 3 or more studies. Results: The search retrieved 359 unique records, but only the 14 that compared runners with TSF to controls were included in the review. Most studies were retrospective, 2 were prospective, and most had a small sample size (5–30 per group). Many variables were not significantly different between groups. Meta-analysis of peak impact, active, and braking ground reaction forces found no significant differences between groups. Individual studies found larger tibial peak anterior tensile stress, peak posterior compressive stress, peak axial acceleration, peak rearfoot eversion, and hip adduction in the TSF group. Conclusion: Meta-analysis indicated that discrete ground reaction force variables were not statistically significantly different in runners with TSF compared to controls. In individual included studies, many biomechanical variables were not statistically significantly different between groups. However, many were reported by only a single study, and sample sizes were small. We encourage additional studies with larger sample sizes of runners with TSF and controls and adequate statistical power to confirm or refute these findings.

The purpose of this exploratory study was to examine school principals’ tendencies and practices that bring forth innovative ideas for school improvement. School principals (N = 124) responded to a survey, which included an open-ended question, on their innovative practices toward school improvement. With the Innovator’s DNA framework as a basis for generating innovation for school improvement, path analysis was used to assess how well school principals’ thinking and practices aligned with the framework. School principals believed it was important to help generate innovation through idea networking, being open-minded, and challenging the status quo. However, through path analysis, school principals’ improvement areas associated with the Innovator’s DNA framework were identified. School systems should help principals develop innovative problem-solving skills for school improvement by applying the Innovator’s DNA framework in their professional development opportunities. More research is needed that examines if and how school systems foster school innovation leadership for principals.

Abstract Objective Expression of glial fibrillary acidic protein (GFAP), a marker of reactive astrocytosis, colocalizes with neuropathology in the brain. Blood levels of GFAP have been associated with cognitive decline and dementia status. However, further examinations at a population‐based level are necessary to broaden generalizability to community settings. Methods Circulating GFAP levels were assayed using a Simoa HD‐1 analyzer in 4338 adults without prevalent dementia from four longitudinal community‐based cohort studies. The associations between GFAP levels with general cognition, total brain volume, and hippocampal volume were evaluated with separate linear regression models in each cohort with adjustment for age, sex, education, race, diabetes, systolic blood pressure, antihypertensive medication, body mass index, apolipoprotein E ε4 status, site, and time between GFAP blood draw and the outcome. Associations with incident all‐cause and Alzheimer's disease dementia were evaluated with adjusted Cox proportional hazard models. Meta‐analysis was performed on the estimates derived from each cohort using random‐effects models. Results Meta‐analyses indicated that higher circulating GFAP associated with lower general cognition (ß = −0.09, [95% confidence interval [CI]: −0.15 to −0.03], p = 0.005), but not with total brain or hippocampal volume (p > 0.05). However, each standard deviation unit increase in log‐transformed GFAP levels was significantly associated with a 2.5‐fold higher risk of incident all‐cause dementia (Hazard Ratio [HR]: 2.47 (95% CI: 1.52–4.01)) and Alzheimer's disease dementia (HR: 2.54 [95% CI: 1.42–4.53]) over up to 15‐years of follow‐up. Interpretation Results support the potential role of circulating GFAP levels for aiding dementia risk prediction and improving clinical trial stratification in community settings.

Objetivo: Evaluar las competencias logradas por estudiantes de medicina en la realización de una apendicectomía abierta y jareta en el modelo de simulación creado, evaluar el grado de satisfacción de su uso y determinar sus costos. Método: Estudio preexperimental de grupo único, prospectivo y longitudinal. Se evaluaron las competencias logradas por 24 alumnos de pregrado de medicina en la realización de una apendicectomía abierta y jareta en el simulador, mediante las pautas OSATS (Objective Structured Assessment of Technical Skills) a través de enseñanza virtual. Se realizó una encuesta a los alumnos para evaluar el simulador y se determinaron sus costos. Resultados: Hubo un incremento significativo en las pautas OSATS de las competencias logradas de 7 (pre-test) a 26 ± 5.71 puntos (pos-test final) (p = 0.0001) y una reducción del tiempo operatorio de 12 ± 3.81 minutos (primer pos-test) a 8 ± 2.02 minutos (pos-test final) (p = 0.0001). El 41% de los alumnos estuvieron totalmente satisfechos con los logros obtenidos y el 59% parcialmente satisfechos. El costo del simulador fue de 4.64 dólares americanos. Conclusiones: Los estudiantes lograron una mejora de sus competencias en la técnica quirúrgica. Este modelo de simulación es de bajo costo y presenta un nivel adecuado en la satisfacción de los logros obtenidos por los alumnos.

Objectives: Validate a questionnaire to establish the perception of difficulties in the development of research projects in the students of the research area of ​​the Faculty of Human Medicine of the Peruvian University Los Andes. Methods: Validation study of a questionnaire designed and based on a Likert scale applied to the students of the subjects of Research process, Thesis seminar I and Thesis seminar II with stratified random probability sampling, content validity analysis and construct by expert judgment and statistical analysis of reliability with Cronbach's alpha. Results: The sample consisted of 85 students who agreed to be part of the study, of which 64,70% belonged to the female gender with an average age of 23 ± 5 years, subject of thesis seminar I 36,47%, Process research 34,12%, thesis seminar II 29,41%; 64,71% expressed difficulties in preparing the research project in formulating the theoretical framework and methodology, 62,35% in writing the bibliography and annexes, 61,18% in posing the problem and formulating hypotheses, in definition of the title 58,82% and in preparation of the administration of the project plan 47,06%. Cronbach's alpha reliability was 0,969. Conclusions: The questionnaire is reliable and valid, it has high internal consistency and valid criteria.

Efficacy data on two malaria vaccines, RTS,S and R21, targeting Plasmodium falciparum circumsporozoite protein (PfCSP), are encouraging. Efficacy may be improved by induction of additional antibodies to neutralizing epitopes outside of the central immunodominant repeat domain of PfCSP. We designed four rPfCSP-based vaccines in an effort to improve the diversity of the antibody response. We also evaluated P. falciparum merozoite surface protein 8 (PfMSP8) as a malaria-specific carrier protein as an alternative to hepatitis B surface antigen. We measured the magnitude, specificity, subclass, avidity, durability, and efficacy of vaccine-induced antibodies in outbred CD1 mice. In comparison to N-terminal- or C-terminal-focused constructs, immunization with near full-length vaccines, rPfCSP (#1) or the chimeric rPfCSP/8 (#2), markedly increased the breadth of B cell epitopes recognized covering the N-terminal domain, junctional region, and central repeat. Both rPfCSP (#1) and rPfCSP/8 (#2) also elicited a high proportion of antibodies to conformation-dependent epitopes in the C-terminus of PfCSP. Fusion of PfCSP to PfMSP8 shifted the specificity of the T cell response away from PfCSP toward PfMSP8 epitopes. Challenge studies with transgenic Plasmodium yoelii sporozoites expressing PfCSP demonstrated high and consistent sterile protection following rPfCSP/8 (#2) immunization. Of note, antibodies to conformational C-terminal epitopes were not required for protection. These results indicate that inclusion of the N-terminal domain of PfCSP can drive responses to protective, repeat, and non-repeat B cell epitopes and that PfMSP8 is an effective carrier for induction of high-titer, durable anti-PfCSP antibodies.

Abstract Background Diagnostic accuracy of real‐time, quantitative PCR (qPCR) assays to quantify virulent Rhodococcus equi using rectal swab samples has not been systematically evaluated. Objective To evaluate the accuracy of qPCR of rectal swab samples to differentiate foals with pneumonia from healthy foals of similar age from the same environment. Animals One hundred privately owned foals born in 2021 from 2 farms in New York. Methods An incident case‐control study design was used. Rectal swabs were collected from all foals diagnosed with R. equi pneumonia at 2 horse‐breeding farms (n = 47). Eligible pneumonia cases (n = 39) were matched by age to up to 2 healthy (n = 53) control foals; rectal swabs were collected from control foals on the day of diagnosis of the index case. DNA was extracted from fecal swabs and the concentration of virulent R. equi (ie, copy numbers of the virulence‐associated protein A gene [vapA] per 100 ng fecal DNA) was estimated by qPCR. Results The area under the ROC curve for qPCR of fecal swabs was 83.7% (95% CI, 74.9‐92.6). At a threshold of 14 883 copies of vapA per 100 ng fecal DNA, specificity of the assay was 83.0% (95% CI, 71.7‐92.4) and sensitivity was 79.5% (95% CI, 66.7‐92.3). Conclusions and Clinical Importance Although fecal concentrations of virulent R. equi are significantly higher in pneumonic foals than healthy foals of similar age in the same environment, qPCR of rectal swabs as reported here lacks adequate diagnostic accuracy for clinical use.

Abstract Background Intragastric administration of virulent Rhodococcus equi protects foals against subsequent experimental intrabronchial (IB) infection, but it is unknown whether R. equi naturally ingested by foals contributes to their susceptibility to pneumonia. Hypothesis Fecal concentration of virulent R. equi before IB infection with R. equi is positively associated with protection from pneumonia in foals. Animals Twenty‐one university‐owned foals. Methods Samples were collected from experimental studies. Five foals were gavaged with live, virulent R. equi (LVRE) at age 2 and 4 days; the remaining 16 foals were not gavaged with LVRE (controls). Fecal swabs were collected from foals at ages 28 days, immediately before IB infection. Foals were monitored for clinical signs of pneumonia, and fecal swabs were collected approximately 2 weeks after IB infection. Swabs were tested by quantitative PCR for concentration of virulent R. equi (ie, copy numbers of the virulence‐associated protein A gene [vapA] per 100 ng fecal DNA). Background: Mpox is a rare zoonotic disease caused by the Mpox virus. On May 21, 2022, WHO announced the emergence of confirmed Mpox cases in countries outside the endemic areas in Central and West Africa. Methods: This multicentre study was performed through the Infectious Diseases International Research Initiative network. Nineteen collaborating centres in 16 countries participated in the study. Consecutive cases with positive Mpoxv-DNA results by the polymerase chain reaction test were included in the study. Results: The mean age of 647 patients included in the study was 34.5.98.6% of cases were males, 95.3% were homosexual-bisexual, and 92.2% had a history of sexual contact. History of smallpox vaccination was present in 3.4% of cases. The median incubation period was 7.0 days. The most common symptoms and signs were rashes in 99.5%, lymphadenopathy in 65.1%, and fever in 54.9%. HIV infection was present in 93.8% of cases, and 17.8% were followed up in the hospital for further treatment. In the two weeks before the rash, prodromal symptoms occurred in 52.8% of cases. IntroductionInformation on the magnitude and durability of humoral immunity against COVID-19 among specific populations can guide policies on vaccination, return from isolation and physical distancing measures. The study determined the durability of SARS-CoV-2 antibodies after an initial infection among Filipinos in Metro Manila, Philippines, and the extent of protection SARS-CoV-2 antibodies confer against reinfection.MethodsWe conducted a cohort study to monitor the antibody levels of patients diagnosed with COVID-19. Receptor-binding domain (RBD)-specific antibodies were measured at Days 21, 90, 180, 270 and 360. Antibody levels were reported as geometric mean titers (GMT) with geometric standard deviation (GSD). Differences in GMT were tested using Friedman test and Kruskal Wallis test, with Bonferroni multiple comparisons procedure. Adjusted hazard ratios on the development of probable reinfection were estimated using Cox proportional models.ResultsThere were 307 study participants included in the study, with 13 dropouts. Study participants received SARS-CoV-2 vaccines at varying times, with 278 participants (90.5%) fully vaccinated by the end of study. This practice report shares practices from a successful Latinx Studies learning community model at an urban research-intensive university in the Midwestern United States. The learning community model offers a tiered developmental curriculum to support the three different stages of a student’s journey, from enrollment to graduation. The three distinct phases of the learning community model are: a pre-college summer enrichment program, a first- and second-year college scholars program and a third and fourth-year college-to-career component. The model also integrates 10 out of 11 of Kuh’s (2008) high impact practices demonstrated to support student engagement and success for students making a direct transition from high school to university. The authors share gains in student retention and five-year graduation rates as evidence of effectiveness.

Invasive meningococcal disease (IMD) imposes a significant burden on the global community due to its high case fatality rate (4-20%) and the risk of long-term sequelae for one in five survivors. An expert group meeting was held to discuss the epidemiology of IMD and immunization policies in Malaysia, Philippines, Thailand, and Vietnam. Most of these countries do not include meningococcal immunization in their routine vaccination programs, except for high-risk groups such as immunocompromised people and pilgrims. It is difficult to estimate the epidemiology of IMD in the highly diverse Asia-Pacific region, but available evidence indicate serogroup B is increasingly dominant. Disease surveillance systems differ by country. IMD is not a notifiable disease in some of them. Without an adequate surveillance system in the region, the risk and the burden of IMD might well be underestimated. With the availability of new combined meningococcal vaccines and the World Health Organization roadmap to defeat bacterial meningitis by 2030, a better understanding of the epidemiology of IMD in the Asia-Pacific region is needed.

Background: At the time that the mother and baby transit from breastfeeding to weaning, maternal-fetal attachment becomes a crucial basis for maintaining their emotional relationship. Conceptualization is needed to better understand the phenomenon of maternal-infant attachment during the weaning process. Objective: The purpose of this paper is to explore and identify the essence of the term maternal-infant attachment during weaning and to arrive at an operational definition of the concept. Method: Concept analysis following Walker and Avant. Results: The attributes of maternal-infant attachment during the weaning process include (1) consistent and properly timed, (2) dynamic transactional interaction, and (3) secured adjustment. Maternal-infant attachment during the weaning process is brought by maternal sensitivity, closeness and proximity seeking, gentle and positive weaning, and breastfeeding experiences, which later turn into favorable emotion regulation, maturity, self-efficacy, secured bond, less intrusive interaction, resilience, and child’s behavioral and development. Conclusion: This concept analysis provides new insight into maternal-infant attachment during the weaning process that guides the nursing practice. Also, the findings can help develop or improve the models, theories, and instruments collected for maintaining maternal-fetal attachment during the weaning process.

ObjectiveThe objective of this study was to examine associations between level of depressive symptoms in older adult spouse/partner couples and their physical health and social factors (social activity and number of close friends).MethodsUsing data from 116 community-dwelling couples (age 76.2 ± 8.5), we simultaneously analyzed associations between depressive symptoms (Geriatric Depression Scale, range 0–11) and dyadic physical health, engagement in social activities, and connectedness with close friends.ResultsGreater engagement in social activities was associated with fewer depressive symptoms in men, whereas more close friendships were associated with fewer depressive symptoms in women, controlling for partner effects, age, education, and cognitive function, with good model fit. Additionally, more disparate physical health within the couple (latent incongruence score) was associated with greater depressive symptoms in men.DiscussionLess social activity and fewer close friends were associated with depressive symptoms in older adult couples, but may be distinctly influential depending on gender and in the context of the older adult couple's physical health.

Singles are an understudied yet growing segment of the adult population. The current study aims to expand the lens of relationship science by examining the well-being of unpartnered, single adults using latent profile analysis. We recruited singles (N = 4,835) closely matched to the United States census (ages 18-65; 57.5% female; 71.1% White; 14.5% Black; 13.8% Hispanic) for an exploratory cross-sectional survey using five variables that strongly predict well-being (friendship satisfaction, family satisfaction, self-esteem, neuroticism, and extraversion). All five variables significantly predicted life satisfaction for the full sample. Latent profile analyses detected 10 groups (or profiles) of singles. Half of the profiles were happy (above the full sample mean of life satisfaction) and half of the profiles were unhappy (below the mean). Each profile had its own unique patterns relating to personal relationships, self-esteem, and personality traits. The happiest profile had the best relationships, self-esteem, and personality, while the unhappiest profile had the worst relationships, self-esteem, and personality. The profiles in between these two extremes had more nuanced patterns. For example, one relatively happy profile in the middle had high friendship satisfaction but low family satisfaction, while an adjacent profile showed the opposite pattern. Overall, singles who had positive relationships—both with themselves and others—were happiest.

3D bioprinters present techniques that have various applications in the food industry. For this reason, this work aims to compile and review various research works focused on the utilities and advantages of this type of machinery. Where we first mention the basis of these bioprinting techniques and then proceed to highlight the bioethical issues that surround their application in the food industry, analyze the current advantages and disadvantages, the user that has been given in the production of food for astronauts, and also mention some of the research that has been taking place in Latin America and the world. The greatest advantage of 3D bioprinting of food is the speed of production compared to traditional manufacturing methods, allowing one to obtain food with various geometric shapes; it allows control of the nutritional value, and the texture of the product, reduces environmental pollution and has the advantage of being able to take advantage of the greater performance of the materials required for production. Additionally, this technology is considered an alternative production technique that will be used to solve the problem of feeding in places of scarce resources such as space and areas not suitable for animal husbandry.

This review discusses the potential of major histocompatibility complex (MHC) Class II constructs as stroke therapeutics. We focus on the delivery of MHC Class II construct, DRmQ, as a safe and effective treatment for ischemic stroke. DRmQ was observed to attenuate behavioral deficits and decrease microglia activation and proinflammatory cytokines, illustrating its ability to mitigate the secondary cell death following stroke. Similar anti-neuroinflammation treatments, such as transplantation of mesenchymal stem cells and mitochondrial transfers, are briefly discussed to provide further support that sequestration of inflammation stands as a robust therapeutic target for stroke.

Hepatocyte 12-hour rhythms have a role in cellular stress and metabolic functions. Here, the authors demonstrate disrupting the 12-hour clock through deletion of XBP1 is associated with the development of NAFLD as well as disruption of phospholipid composition and the maintenance of lipid homeostasis.

Alzheimer’s disease and related dementias (ADRD) are among the top contributors to disability and mortality in later life. As with many chronic conditions, aging is the single most influential factor in the development of ADRD. Even among older adults who remain free of dementia throughout their lives, cognitive decline and neurodegenerative changes are appreciable with advancing age, suggesting shared pathophysiological mechanisms. In this Review, we provide an overview of changes in cognition, brain morphology, and neuropathological protein accumulation across the lifespan in humans, with complementary and mechanistic evidence from animal models. Next, we highlight selected aging processes that are differentially regulated in neurodegenerative disease, including aberrant autophagy, mitochondrial dysfunction, cellular senescence, epigenetic changes, cerebrovascular dysfunction, inflammation, and lipid dysregulation. We summarize research across clinical and translational studies to link biological aging processes to underlying ADRD pathogenesis. Targeting fundamental processes underlying biological aging may represent a yet relatively unexplored avenue to attenuate both age-related cognitive decline and ADRD. Collaboration across the fields of geroscience and neuroscience, coupled with the development of new translational animal models that more closely align with human disease processes, is necessary to advance novel therapeutic discovery in this realm.

Sprague Dawley (SD) rats are among the most widely used outbred laboratory rat populations. Despite this, the genetic characteristics of SD rats have not been clearly described, and SD rats are rarely used for experiments aimed at exploring genotype-phenotype relationships. In order to use SD rats to perform a genome-wide association study (GWAS), we collected behavioral data from 4,625 SD rats that were predominantly obtained from two commercial vendors, Charles River Laboratories and Harlan Sprague Dawley Inc. Using double-digest genotyping-by-sequencing (ddGBS), we obtained dense, high-quality genotypes at 291,438 SNPs across 4,061 rats. This genetic data allowed us to characterize the variation present in Charles River vs. Harlan SD rats. We found that the two populations are highly diverged (FST > 0.4). Furthermore, even for rats obtained from the same vendor, there was strong population structure across breeding facilities and even between rooms at the same facility. We performed multiple separate GWAS by fitting a linear mixed model that accounted for population structure and using meta-analysis to jointly analyze all cohorts. Our study examined Pavlovian conditioned approach (PavCA) behavior, which assesses the propensity for rats to attribute incentive salience to reward-associated cues. We identified 46 significant associations for the various metrics used to define PavCA. The surprising degree of population structure among SD rats from different sources has important implications for their use in both genetic and non-genetic studies.

OBJECTIVES/GOALS: Determine if the Conserved Transcriptional Response to Adversity transcriptomic profile established in primary blood mononuclear cells (PBMC) of chronically stress caregivers, is present in individuals with early Alzheimer’s disease. Chronic stress is a risk factor for Alzheimer’s, and may be an untapped biomarker for disease risk and pathology. METHODS/STUDY POPULATION: To collect preliminary data on the Conserved Transcriptional Response to Adversity profile in individuals with Alzheimer’s disease, we were able to utilize primary blood mononuclear cell samples from a small open label pilot study called Senolytic Therapy to Modulate the Progression of Alzheimer’s Disease, designed to clear stressed senescent cells. We hypothesized senolytics may beneficially reverse this stress profile. We developed a NanoString assay (measuring 19 inflammatory, 31 type-1 interferon, and 3 antibody synthesis genes) to compare these transcriptomic changes within 4 individuals measured at baseline, post-treatment with an intermittent 12-week senolytic therapy, and at an optional extended post-treatment follow-up time point > 3 months after their post treatment visit. RESULTS/ANTICIPATED RESULTS: There was relative downregulation of expression in transcription in 7 of 19 measured inflammatory genes (FOS, PTGS2, IL8, FOS, Il1b, JUNB, and JUN) in Alzheimer’s disease participants after receiving senolytic treatment (baseline vs. post-treatment). This is consistent with a decrease in the inflammatory arm of the Conserved Transcriptional Response to Adversity profile. These differences were not significant between baseline and the extended follow-up, indicative of a transient effect of senolytic. There were no changes in type 1 interferon or antibody synthesis genes. This data provides preliminary evidence for larger controlled studies to further establish this profile in Alzheimer’s disease, providing exciting evidence for transcript changes that may be reproducible with senolytic therapy. DISCUSSION/SIGNIFICANCE: Literature relevant to Alzheimer’s disease indicates global increases in inflammation paired with deficits in immune response, capturing some genes associated with the Conserved Transcriptional Response to Adversity. This profile may be a useful biomarker for prediction of disease severity or risk of dementia due to chronic stress.

Summary: The 12-h clock coordinates lipid homeostasis, energy metabolism, and stress rhythms via the transcriptional regulator XBP1. However, the biochemical and physiological bases for integrated control of the 12-h clock and diverse metabolic pathways remain unclear. Here, we show that steroid receptor coactivator SRC-3 coactivates XBP1 transcription and regulates hepatic 12-h cistrome and gene rhythmicity. Mice lacking SRC-3 show abnormal 12-h rhythms in hepatic transcription, metabolic functions, systemic energetics, and rate-limiting lipid metabolic processes, including triglyceride, phospholipid, and cardiolipin pathways. Notably, 12-h clock coactivation is not only preserved, with its cistromic activation priming ahead of the zeitgeber cue of light, but concomitant with rhythmic remodeling in the absence of food. These findings reveal that SRC-3 integrates the mammalian 12-h clock, energy metabolism, and membrane and lipid homeostasis and demonstrates a role for the 12-h clock machinery as an active transcriptional mechanism in anticipating physiological and metabolic energy needs and stresses.

Exophiala is a rare fungus that can cause fatal infections in patients who are immunocompromised. This case describes a 55-year-old post-renal transplant patient undergoing active treatment for multiple myeloma who presented to the emergency room with complaints of cough and dyspnea. She was found to have a cavitary lesion in the left upper lobe of her lung, and direct sampling via bronchoscopy revealed an invasive fungal infection with an Exophiala species. Despite initial improvement on antifungal therapy, her clinical course deteriorated requiring pneumonectomy and formation of brain abscess. This case emphasizes the challenge of invasive fungal infections and the importance of broad differentials in patients who are immunocompromised.

There has been extensive discussion of the “Replication Crisis” in many fields, including genome-wide association studies (GWAS). We explored replication in a mouse model using an advanced intercross line (AIL), which is a multigenerational intercross between two inbred strains. We re-genotyped a previously published cohort of LG/J x SM/J AIL mice (F34; n = 428) using a denser marker set and genotyped a new cohort of AIL mice (F39-43; n = 600) for the first time. We identified 36 novel genome-wide significant loci in the F34 and 25 novel loci in the F39-43 cohort. The subset of traits that were measured in both cohorts (locomotor activity, body weight, and coat color) showed high genetic correlations, although the SNP heritabilities were slightly lower in the F39-43 cohort. For this subset of traits, we attempted to replicate loci identified in either F34 or F39-43 in the other cohort. Coat color was robustly replicated; locomotor activity and body weight were only partially replicated, which was inconsistent with our power simulations. We used a random effects model to show that the partial replications could not be explained by Winner’s Curse but could be explained by study-specific heterogeneity. Despite this heterogeneity, we performed a mega-analysis by combining F34 and F39-43 cohorts (n = 1,028), which identified four novel loci associated with locomotor activity and body weight. These results illustrate that even with the high degree of genetic and environmental control possible in our experimental system, replication was hindered by study-specific heterogeneity, which has broad implications for ongoing concerns about reproducibility.

de Siguas, Arequipa Province, Peru. The concentrations used were 2, 4, and 6 kg ha−1 with 4 applications by means of impregnation to the transplant and via drench every 15 days. The statistical design was through a DBCA with factorial arrangement randomly distributed in four blocks. The tests used were the orthogonal contrast test and Duncan’s multiple comparison test (α = 0.05). The findings showed that the treatment concentrations of 4 kg ha−1 and 6 kg ha−1 resulted in a lower incidence of Fusarium oxysporum, at 6.71% and 9.36%, an ABCPE of 637.78 and 950.14 units, and an exportable yield of 66.34 t ha−1 and 65.42 t ha−1, respectively, displaying significant differences from the control. The first application was statistically significant to the treatments with the highest number of applications, showing a greater exportable yield of 67.54 t ha−1. The best interactions between concentrations and applications were 6 kg ha−1 with 1 application, 4 kg ha−1 with 2 applications, and 4 kg ha−1 with 1 application, with exportable yields of 70.66, 69.61, and 69.11 t ha−1, respectively.

El objetivo del trabajo fue mostrar cómo el Sistema de Innovación Agrícola Local (SIAL) en el municipio Perico, provincia Matanzas, Cuba, pudo resolver desafíos productivos a partir de que se detectaron en la Plataforma Multiactoral de Gestión (PMG), lo cual se inició con la problemática de bajo rendimiento en la producción de Ipomoea batatas (boniato) en el polo productivo de La Angelina. Para ello se utilizaron diferentes métodos científicos y técnicas de investigación como: diagnósticos del contexto, encuestas, muestreos de suelo, entre otros. Además, se sistematizaron los resultados que se alcanzaron en el territorio con la implementación de este tipo de enfoque en las diferentes dimensiones; lo que permitió realizar varios ciclos de aprendizaje y, a su vez, contar con un plan de medidas para incrementar los rendimientos del boniato y mejorar el suelo como recurso fundamental e irremplazable, capaz de sustentar los cultivos y la vida en general.

Abstract Introduction The study evaluated if blood markers reflecting diverse biological pathways differentiate clinical diagnostic groups among Hispanic and non‐Hispanic White adults. Methods Within Hispanic (n = 1193) and non‐Hispanic White (n = 650) participants, serum total tau (t‐tau), neurofilament light (NfL), ubiquitin carboxyl‐terminal hydrolase LI, glial fibrillary acidic protein (GFAP), soluble cluster of differentiation‐14, and chitinase‐3‐like protein 1 (YKL‐40) were quantified. Abstract Introduction Cognitive screening measures often lack sensitivity and are hampered by inequities across ethnoracial groups. A multitrait multimethod (MTMM) classification may attenuate these shortcomings. Methods A sample of 7227 participants across the diagnostic spectrum were selected from the National Alzheimer's Coordinating Center cohort. Random forest ensemble methods were used to predict diagnosis across the sample and within Black American (n = 1025) and non‐Hispanic White groups (n = 5263) based on: (1) a demographically corrected Montreal Cognitive Assessment (MoCA), (2) MoCA and Functional Assessment Questionnaire (FAQ), (3) MoCA and FAQ with demographic correction. Results The MTMM approach with demographic correction had the highest diagnostic accuracy for the cognitively unimpaired (area under curve [AUC] [95% confidence interval (CI)]): 0.906 [0.892, 0.920]) and mild cognitive impairment (AUC: 0.835 [0.810, 0.860]) groups and reduced racial disparities. Discussion With further validation, the MTMM approach combining cognitive screening and functional status assessment may serve to improve diagnostic accuracy and extend opportunities for early intervention with greater equity.
