Abstract

Plants have long been used in traditional medicine to treat illnesses. Nevertheless, their chemical diversity requires studies to establish the extract dosage and its safe use. Pseudobombax parvifolium, an endemic species of the Brazilian Caatinga biome, is commonly used in folk medicine, due to its anti-inflammatory properties related to cellular oxidative stress; however, its biological properties have scarcely been studied. In this study, we chemically characterized the P. parvifolium hydroalcoholic bark extract (EBHE) and evaluated its cytotoxic, mutagenic, and preclinical aspects, as well as its antioxidant effect. Our phytochemical analysis revealed a significative total polyphenol content and identified loliolide for the first time in this species. Cytotoxicity, mutagenicity, and acute oral and repeated dose indicated no toxic effects on cell culture, Drosophila melanogaster, and Wistar rat exposure to different EBHE concentrations, respectively. Furthermore, we observed a significant decrease in lipid peroxidation and a mild hypoglycemic and hypolipidemic effect with repeated oral dosing of EBHE. Although there were no significant changes in glutathione content, we did observe a significant increase in superoxide dismutase at a dose of 400 mg/kg and in glutathione peroxidase at doses of 100, 200, and 400 mg/kg. These findings suggest that EBHE has potential as a source of bioactive molecules, and it can be used safely in traditional medicine and in the development of herbal medicines for application in the public health system. Abstract

Conselho Nacional de Desenvolvimento Científico e Tecnológico - CNPq A inflamação é composta de uma reação vascular e outra celular, conferindo diferentes reações de tecidos e células, tanto do ambiente intravascular, como o do ambiente extravascular. À medida que o processo inflamatório ocorre, proteases da coagulação, em especial a trombina (FIIa), são capazes de desencadear diversas respostas celulares na biologia vascular e por isso, frequentemente é observada a ativação de outros sistemas biológicos, levando a complicações, como a trombose. Assim, moléculas que interferem positivamente nesses eventos são modelos interessantes para o desenvolvimento de novas drogas anti-inflamatórias. Neste contexto, destacam-se os metabólitos secundários de plantas medicinais, os quais interagem com diversas proteínas envolvidas em processos biológicos importantes, incluindo inflamação e coagulação. Por essa razão, o presente trabalho teve por objetivo caracterizar química e toxicologicamente os extratos vegetais de duas espécies do Bioma caatinga (Turnera subulata e Licania rigida Benth), bem como, avaliar os potenciais anti-inflamatórios e anticoagulantes desses extratos. Os extratos foram obtidos a partir da coleta e secagem do material vegetal com posterior extração aquosa e alcoólica, sendo em seguida rotaevaporados e liofilizados. Os extratos de Turnera subulata e Licania rigida Benth apresentaram compostos fenólicos como principais fitocomponentes (flavonóis-3-O-glicosilados), bem como, vitexina, ácido ferúlico, ácido octadecatrienóico, alguns aminoácidos e outros. Em relação a toxicidade, todos os extratos estudados apresentaram baixos efeitos toxicológicos, assim como efeitos anticoagulantes capazes de inibir a trombina de forma direta (~80-90%) e de forma indireta através do cofator II da heparina (~70%). Sobre a inflamação, os extratos apresentaram significativo efeito inibitório com redução de citocinas pró-inflamatórias TNF-α e IL-1β (~70-75%) e efeitos no aumento da secreção da IL-10 (~15-20%), além de serem capazes de inibir prostaglandina E2 (~80%) e óxido nítrico (~65%). Assim, os resultados exibidos pelos extratos, apontam esses como alvos farmacológicos com perspectivas para o desenvolvimento de novas drogas anti-inflamatórias. Inflammation is composed of a vascular and a cellular reaction, conferring different tissue and cell responses, both in the intravascular and extravascular environments. As the inflammatory process occurs, the coagulation protease thrombin (FIIa), can trigger several cellular responses in vascular biology and, therefore, activation of other biological systems is often observed, leading to complications such as thrombosis. Thus, molecules that can modulate these events are interesting models for the development of new antiinflammatory drugs. In this context, secondary metabolites of medicinal plants stand out, since they can interact with several proteins involved in important biological processes, including inflammation and coagulation. For this reason, this study aimed to chemically and toxicologically characterize the plant extracts of two species of the Caatinga Biome (Turnera subulata and Licania rigida Benth), as well as to evaluate the anti-inflammatory and anticoagulant potentials of the extracts. The extracts were obtained after the collection and drying of plant material with subsequent aqueous and alcoholic extraction, followed by rotaevaporation and lyophilization. Turnera subulata and Licania rigida Benth extracts presented phenolic compounds as the main phytocomponents (flavonols-3-Oglycosylated), as well as vitexin, ferulic acid, octadecatrienoic acid, amino acids and others. Regarding toxicity, all extracts showed low toxicological effects, as well as anticoagulant potential through direct thrombin inhibition (~80-90%) and indirect, mediated by the heparin cofactor II (~70%). On inflammation, the extracts showed a significant reduction of proinflammatory cytokines TNF-α and IL-1β (~70-75%) and increase effects on IL-10 secretion (~15-20%), in addition to be able to inhibit prostaglandin E2 (~80%) and nitric oxide (~65%). Thus, the results exhibited by the extracts point to these as a pharmacological target with perspectives for the development of new anti-inflammatory drugs.

Abstract

The balance between embolic risk and bleeding represents a clinical challenge in cancer patient treatment, encouraging studies on adjuvant oncologic treatments. Thereby, this study evaluated the in vitro effect of green tea extract (GTE) and epigallocatechin-3-gallate (EGCG) on hemostasis modulation and the antineoplastic effect on melanoma cells (B16-F10) by applying platelet aggregation, angiogenesis and viability cell assays. The results displayed a significant platelet antiaggregant effect, corresponding to 50 and 80% for the extract and EGCG, respectively, compared to the negative control. Furthermore, both GTE and EGCG exhibited antitumor effects by reducing melanoma cell growth by 25 and 50%, respectively, verified by cellular apoptosis. Regarding angiogenesis, these substances inhibited blood vessel formation, reaching about 25% and 99% for GTE and EGCG at 100 μg/mL, respectively. Moreover, TNF-α cell stimulation evidenced VEGF and IL-8 secretion inhibition at 55 and 20% with GTE, while EGCG promoted an inhibition around 78% for both VEGF and IL-8. The results indicate the promising performance of GTE and EGCG as an option for treating cancer and its side effects. Nonetheless, further studies are required to elucidate their action mechanism on clotting, cell death and angiogenesis. [ABSTRACT FROM AUTHOR]
