Genetic markers are important tools for the study of natural and induced genetic variation. Due to the strong increase in the number of genome sequences, an overview of the genomic diversity of many natural strains from individual species could be gained. This allows for the design of markers for flexible use and cost-efficient small scale genetic studies requiring minimal laboratory and bioinformatics effort. Here, we describe 140 single nucleotide polymorphism (SNP) markers with genome-wide distribution that discriminate between the genotype of the common Arabidopsis thaliana reference accession Columbia-0 (Col-0) and the majority of Arabidopsis thaliana accessions that have been sequenced to date. We designed, generated and validated the markers using the kompetitive allele-specific PCR (KASP) technology and made all 140SNPvCol marker assays publicly available through a service provider. Through the integration of available genomic SNP allele information of 1,135 accessions, we found that 120 of these 140 markers could detect non-reference alleles in 647 accessions and more than 100 markers showed non-reference alleles in 1,094 accessions. We further show that the marker set can be used for the verification or identification of accessions of unknown identity. As the KASP methodology is very flexible, an optimal marker subset can be easily selected among the available 140SNPvCol markers presented here to analyze genetic combinations of Col-0 with any other accession.

The transcription factor FLOWERING LOCUS M (FLM) of the model plant Arabidopsis thaliana is an important component to regulate flowering time in response to changing ambient temperature. This thesis describes the natural genetic variation of this gene, which has an important role for adaptation of flowering time during Arabidopsis thaliana evolution. Furthermore, previously unknown regulatory regions of FLM were identified, which contribute to the regulation of FLM gene expression and flowering time. Der Transkriptionsfaktor FLOWERING LOCUS M (FLM) der Modellpflanze Arabidopsis thaliana ist eine wichtige Komponente zur Steuerung des Blühzeitpunkts in Reaktion auf wechselnde Umgebungstemperatur. Diese Thesis beschreibt die natürliche genetische Variation dieses Gens, die eine wichtige Rolle bei dem evolutionären Anpassungsprozess des Blühverhaltens von Arabidopsis thaliana hat. Desweiteren werden bisher unbekannte regulatorische Regionen des Gens beschrieben, die zur Kontrolle der Genexpression von FLM und damit des Blühverhaltens beitragen.

Tiermodelle, Familien-, Zwillings- und Adoptionsstudien belegen, dass genetische Faktoren neben Umwelteinflüssen eine bedeutende Rolle bei der Genese der Alkoholabhängigkeit spielen. Ebenso geht man von einer multifaktoriellen Ätiologie bei der Entstehung des Delirium tremens (DTs) während der Alkoholentgiftung aus. Ziel der Assoziationsstudie war es, einzelne disponierende Gene bzw. serologische Marker der Alkoholabhängigkeit und des DTs zu identifizieren. Dabei wurden mit Hilfe der Polymerasekettenreaktion (PCR) und anschließendem Restriktionsendonukleasenverdau Kandidatengene analysiert, denen eine gewisse Funktionalität zugesprochen wird. Es konnten 208 nach DSM IV alkoholabhängige Patienten und 102 gesunde Kontrollpersonen rekrutiert werden. 82 der alkoholabhängigen Patienten litten während der Alkoholentgiftung unter schweren Entzugserschienungen in Form eines DTs. Weder für den funktionellen Val/Met-Polymorphismus des katecholaminabbauenden Enzyms Katecholamin-O-methyltransferase (COMT) noch für einen 48bp-VNTR-Polymorphismus des Dopaminrezeptors DRD4 konnte eine signifikante Assoziation mit der Alkoholabhängigkeit bzw. dem DT hergestellt werden. Dagegen stellte sich ein signifikanter Zusammenhang zwischen dem DBH*444g/a-Polymorphismus und der DbetaH-Plasmaaktivität dar. Der DBH*444g/a-Polymorphismus ist mit der Diagnose "Alkoholabhängigkeit" assoziiert: das Allel a weist eine signifikant höhere Frequenz in der Gruppe der alkoholabhängigen Patienten auf. Ferner zeigte sich nach mindestens 22 Tagen Abstinenz eine signifikant erniedrigte DbetaH-Plasmaaktivität bei den alkoholabhängigen Patienten. Zwischen den Patienten mit DT und den Patienten mit nur leichten Entzugserscheinungen während der Entgiftung konnte bezüglich des DBH*444g/a-Polymorphismus und der DbetaH-Plasmaaktivität kein signifikanter Unterschied festgestellt werden. Die Daten deuten auf eine Alkoholabhängigkeit-assoziierte Verminderung der DbetaH-Plasmaaktivität hin, die möglicherweise durch den DBH*444g/a-Polymorphismus beeinflusst wird. Nach der vorliegenden Studie ergibt sich kein Hinweis auf eine Beteiligung der untersuchten Polymorphismen für die Ätiologie des DTs. Animal experiments, family studies, twin studies and adoption studies show that both environmental and genetic influences seem to play a critical role in the etiology of alcohol dependence. It also can be assumed that the emergence of delirium tremens (DT) during alcohol withdrawal is caused by various factors. The intention of this association study was to identify predisposing genes and serologic markers for alcohol dependence or DT. We analysed potential functional polymorphisms via polymerase chain reaction (PCR) and restriction fragment length polymorphism assay (RFLP). 208 patients fulfilling the criteria for alcohol dependence according to DSM IV and 102 healthy controls were recruited. 82 patients had a history of alcohol withdrawal DT. The functional Val/Met-polymorphism of the human catechol-O-methyltransferase gene and the 48bp-VNTR-polymorphism of the dopamine D4 receptor were neither significantly associated with the diagnosis of alcoholism nor DT during alcohol withdrawal. The dopamine-beta-hydroxylase (DbetaH) plasma activity turned out to be influenced by the DBH*444g/a-polymorphism. The DBH*444g/a-polymorphism is significantly associated with the diagnosis "alcohol dependence": allele a had a higher frequency in the group of alcohol dependent patients. In addition mean plasma DbetaH activity measured in alcoholic subjects after 22 days of abstinence was significantly lower than that observed in the control group. No difference was found between subjects with DT during withdrawal and subjects with mild withdrawal symptoms regarding plasma DbetaH activity and DBH*444g/a-polymorphism. The study shows an alcoholism-related reduction in plasma DbetaH activity which seems to be influenced by the DBH*444g/a-polymorphism. According to our data none of the analysed polymorphisms contributed to the etiology of the DT.

Gegenstand der Dissertation ist der Zusammenhang zwischen Extraktion (bestimmten Formen der Umstellung bzw. Dislokation von syntaktischen Objekten) und Projektion (dem Aufbau syntaktischer Struktur) im Deutschen, insbesondere im Hinblick auf die Struktur sententialer Kategorien und die Bewegung in satzeinleitende Positionen. Das Deutsche ist charakterisiert durch Asymmetrien zwischen Sätzen mit Verb-Endstellung (VE) und Sätzen mit Verb-Zweitstellung (V2) bzw. Verb-Erststellung (V1), die sich auch auf Bewegungstypen wie Topikalisierung auswirken. Darauf basierend haben sich konkurrierende Ansätze zur kategorialen Struktur der topologischen Satztypen des Deutschen herausgebildet, die eine uniforme oder unterschiedliche Struktur für VE- und V2/V1-Sätze annehmen. Die Dissertation folgt minimalistischen Prämissen zur Merkmalsgetriebenheit von syntaktischer Bewegung und argumentiert für eine Auflösung der kategorialen Unterschiede zu Gunsten einer einheitlichen kategorialen Struktur und einer Erklärung der Asymmetrien auf der Grundlage von Unterschieden im Merkmalsgehalt einer (singulären) sententialen Kategorie. Kapitel 1 skizziert wesentliche Eigenschaften der Satzstruktur und damit zusammenhängender Extraktionskonstruktionen im Deutschen. Kapitel 2 gibt einen Überblick über den allgemeinen Aufbau des minimalistischen Ansatzes und diskutiert konkurrierende derivationell und repräsentationell orientierte Varianten dieses Ansatzes (Chomsky 1995, Haider 1993, Brody 1995). In Kapitel 3 wird ein minimaler und merkmalsbasierter Ansatz zur Projektion der sententialen Struktur und zur Besetzung der satzeinleitenden Positionen im Deutschen vorgeschlagen. Zudem werden die Eigenschaften von zwei Konstruktionen mit komplexen A’-Abbhängigkeiten, der was-w-Konstruktion im Standarddeutschen und einer speziellen Form von Topikalisierungs-/Parasitic Gap-Konstruktionen im Süddeutschen, diskutiert. The dissertation deals with the relation between extraction (certain forms of dislocation of syntactic objects) and projection (the building of syntactic structure) in German, especially with regard to the structure of clausal categories and to movement in clause-initial positions. German is characterized by asymmetries between clauses with verb-final (VE) order and clauses with verb-second (V2) and verb-first (V1) order which also have effects on movement types as, e.g., topicalization. Based on such asymmetries, competing approaches on the categorial structure of (topological) clause types of German have been developed which assume either a uniform or a different structure for VE- and V2/V1-clauses. Following minimalist assumptions on feature-driven movement, it is argued that categorial differences can be abandoned in favour of a uniform categorial structure which explains the asymmetries based on differences in the featural make-up of a (single) clausal category. Chapter 1 sketches out essential properties of German clause structure and related constructions with extraction. Chapter 2 gives an overview of the general structure of the minimalist approach and discusses competing derivational and representational variants of the approach (Chomsky 1995, Haider 1993, Brody 1995). In Chapter 3, a minimal and feature-based approach to clausal projection and movement into clause-initial positions in German is proposed. The remainder of the thesis discusses the properties of two constructions with complex A’-dependencies, namely the partial (wh-) movement construction in Standard German and a special form of topicalization/parasitic gap constructions in Southern German.
