9. Nitric Oxide Promoted the Seed Germination of Cynanchum auriculatum under Cadmium Stress.

Abstract

Cynanchum auriculatum, an early food-medicine homologous plant native to Asia, possesses significant nutritional and health benefits. However, the presence of cadmium (Cd) in the soil poses a hazard to the germination and growth of C. auriculatum. As nitric oxide (NO) plays a vital role in plant resistance to heavy metal stress, we used three different concentrations of SNP treatment during the germination phase, aiming to alleviate the inhibitory effects of Cd stress on the seed germination of C. auriculatum. The results indicated that when compared to seeds treated with SNP concentrations of 0.2 mM and 0.8 mM, C. auriculatum seeds treated with 0.4 mM SNP exhibited an improved germination rate and germination index, as well as longer hypocotyl. Furthermore, in comparison to NOS-like, the SNP application stimulated the production of endogenous NO through NR catalysis. Additional investigations showed that the ABA level decreased while the GA level increased under normal conditions, while the SNP application enhanced the accumulation of both ABA and GA in C. auriculatum seeds under Cd stress. Histochemical staining and biochemical indicators demonstrated that SNP treatment enhanced the enzymatic activity of SOD, POD, and CAT, while inhibiting the production of hydrogen peroxide and superoxide anion. Moreover, SNP treatment resulted in increased α-amylase activity, which facilitated starch hydrolysis and the generation of soluble sugar. Ultimately, the seed vitality of C. auriculatum under Cd stress was promoted. Our findings present a theoretical framework for the application of SNP in the seed germination mechanism of C. auriculatum and establish the groundwork for comprehending the physiological role of NO under Cd stress. 19. Discovery of TAS2R14 Agonists from Platycodon grandiflorumUsing Virtual Screening and Affinity Screening Based on a Novel TAS2R14-Functionalized HEMT Sensor Combined with UPLC–MS Analysis

Abstract

TAS2R14 is of great potential as a therapeutic target against asthma, and the discovery of TAS2R14 agonists can be very valuable for treating this disease. Herein, we developed a strategy using virtual screening and affinity screening based on a fabricated biosensor combined with UPLC–MS analysis to screen TAS2R14 agonists from Platycodon grandiflorum. By ligand-based virtual screening, 16 best-fit candidates were yielded. A novel TAS2R14-functionalized high-electron-mobility transistor (HEMT) sensor was applied to detect and fish out the potential TAS2R14 agonists from P. grandiflorumextracts. Those components captured by the immobilized TAS2R14 were eluted and characterized on UPLC−QTOF MS. As a result, six potential TAS2R14 agonists were screened out and identified. Among them, platycodin L was confirmed to be a special agonist of TAS2R14 for the first time and had an EC50of 15.03 ± 1.15 μM via intracellular calcium mobilization assay (n= 6). The results indicated that the proposed strategy was efficient to discover TAS2R14 agonists from the herb directly.

21. In vitro Investigation of Blood Compatibility of Ti with Oxide Layers of Rutile Structure

Abstract

Structure characteristics of titanium oxide layer on titanium matrix were investigated by Rutherford Backscattering Spectroscopy (RBS), Auger Electron Spectroscopy (AES) and X-ray diffraction. It has been identified that the titanium oxide layers had rutile structure. The blood compatibility of the titanium oxide layers of different thickness was studied by blood clotting time measurement. It was shown that as the thickness of the titanium oxide layers increased, blood compatibility of these layers was obviously improved.
