NCT03450655) including seventy-seven community-dwelling 70-year-old men and women with central obesity defined as > 1 kg visceral adipose tissue for women and > 2 kg for men. Participants randomized to the intervention group were offered a 10-week progressive vigorous interval training program performed three times per week. Control subjects were asked to maintain their daily living and routines throughout the trial. All participants in both groups had received tailored lifestyle recommendations focused on diet and physical activity at one occasion within 12 months prior to trial initiation. Prespecified outcome measures included: changes in HRQoL using the Short Form Health Survey Questionnaire (SF-36), blood pressure; resting heart rate (HR) and blood lipids. All analyses were conducted on an intention-to-treat basis.Results: The intervention resulted in significant effects on the SF-36 mental component summary (MCS) score and the mental health (MH) subscale (P< 0.05 for both), when compared to the control group. Specifically, the intervention group increased their MCS score by 6.3 points (95% confidence interval [CI] = 0.3–12.3) and their MH score by 6.0 points (95% CI = 1.7–10.4) compared to the control group. Abstract

Despite the availabilty of imaging-based and mass-spectrometry-based methods for spatial proteomics, a key challenge remains connecting images with single-cell-resolution protein abundance measurements. Here, we introduce Deep Visual Proteomics (DVP), which combines artificial-intelligence-driven image analysis of cellular phenotypes with automated single-cell or single-nucleus laser microdissection and ultra-high-sensitivity mass spectrometry. DVP links protein abundance to complex cellular or subcellular phenotypes while preserving spatial context. By individually excising nuclei from cell culture, we classified distinct cell states with proteomic profiles defined by known and uncharacterized proteins. In an archived primary melanoma tissue, DVP identified spatially resolved proteome changes as normal melanocytes transition to fully invasive melanoma, revealing pathways that change in a spatial manner as cancer progresses, such as mRNA splicing dysregulation in metastatic vertical growth that coincides with reduced interferon signaling and antigen presentation. Abstract

Here we describe the LifeTime Initiative, which aims to track, understand and target human cells during the onset and progression of complex diseases, and to analyse their response to therapy at single-cell resolution. This mission will be implemented through the development, integration and application of single-cell multi-omics and imaging, artificial intelligence and patient-derived experimental disease models during the progression from health to disease. The analysis of large molecular and clinical datasets will identify molecular mechanisms, create predictive computational models of disease progression, and reveal new drug targets and therapies. The timely detection and interception of disease embedded in an ethical and patient-centred vision will be achieved through interactions across academia, hospitals, patient associations, health data management systems and industry. The application of this strategy to key medical challenges in cancer, neurological and neuropsychiatric disorders, and infectious, chronic inflammatory and cardiovascular diseases at the single-cell level will usher in cell-based interceptive medicine in Europe over the next decade., The LifeTime initiative is an ambitious, multidisciplinary programme that aims to improve healthcare by tracking individual human cells during disease processes and responses to treatment in order to develop and implement cell-based interceptive medicine in Europe.

Abstract

LifeTime aims to track, understand and target human cells during the onset and progression of complex diseases and their response to therapy at single-cell resolution. This mission will be implemented through the development and integration of single-cell multi-omics and imaging, artificial intelligence and patient-derived experimental disease models during progression from health to disease. Analysis of such large molecular and clinical datasets will discover molecular mechanisms, create predictive computational models of disease progression, and reveal new drug targets and therapies. Timely detection and interception of disease embedded in an ethical and patient-centered vision will be achieved through interactions across academia, hospitals, patient-associations, health data management systems and industry. Applying this strategy to key medical challenges in cancer, neurological, infectious, chronic inflammatory and cardiovascular diseases at the single-cell level will usher in cell-based interceptive medicine in Europe over the next decade.

Abstract

This chapter reviews the field of event experience research, identifying major trends and setting out research directions for the future. Experiences are often viewed as the core of events and are therefore a key concern for event management and design. Much research has therefore focused on the way in which experiences are produced and consumed within events, which are often viewed as a special setting outside of everyday life. Recent research has extended to the complete visitor journey, considering what happens before and after the event as well. Future directions for event experience research suggested here include the development of improved measurement tools, examining the linkages between different elements of the event experience, the co-creation of value through event experiences, the development of event careers among visitors and the application of new technologies to event experience research.

Abstract

Complex chromosomal rearrangements (CCRs) are rearrangements involving more than two chromosomes or more than two breakpoints. Whole genome sequencing (WGS) allows for outstanding high resolution characterization on the nucleotide level in unique sequences of such rearrangements, but problems remain for mapping breakpoints in repetitive regions of the genome, which are known to be prone to rearrangements. Hence, multiple complementary WGS experiments are sometimes needed to solve the structures of CCRs. We have studied three individuals with CCRs: Case 1 and Case 2 presented with de novo karyotypically balanced, complex interchromosomal rearrangements (46,XX,t(2;8;15)(q35;q24.1;q22) and 46,XY,t(1;10;5)(q32;p12;q31)), and Case 3 presented with a de novo, extremely complex intrachromosomal rearrangement on chromosome 1. Molecular cytogenetic investigation revealed cryptic deletions in the breakpoints of chromosome 2 and 8 in Case 1, and on chromosome 10 in Case 2, explaining their clinical symptoms. In Case 3, 26 breakpoints were identified using WGS, disrupting five known disease genes. All rearrangements were subsequently analyzed using optical maps, linked-read WGS, and short-read WGS. In conclusion, we present a case series of three unique de novo CCRs where we by combining the results from the different technologies fully solved the structure of each rearrangement. Abstract

The recent advent of methods for high-throughput single-cell molecular profiling has catalyzed a growing sense in the scientific community that the time is ripe to complete the 150-year-old effort to identify all cell types in the human body. The Human Cell Atlas Project is an international collaborative effort that aims to define all human cell types in terms of distinctive molecular profiles (such as gene expression profiles) and to connect this information with classical cellular descriptions (such as location and morphology). An open comprehensive reference map of the molecular state of cells in healthy human tissues would propel the systematic study of physiological states, developmental trajectories, regulatory circuitry and interactions of cells, and also provide a framework for understanding cellular dysregulation in human disease. Here we describe the idea, its potential utility, early proofs-of-concept, and some design considerations for the Human Cell Atlas, including a commitment to open data, code, and community.

Abstract

This chapter considers the concept of 'network value' as one of the important benefits that events can generate. It explores the analysis of value creation from the traditional areas of economic, social and cultural value towards new sources of value that are particularly important in the 'knowledge economy' or the 'network society'. The chapter focuses on the ways in which the Sonar Festival has generated 'network value' for Barcelona, which provides the physical and symbolic hub around which Sonar has developed. It analyses the network value being generated by a specific event, the Sonar Festival, which operates both in a local and a global context. The addition of new nodes to the network has not only increased the total network value, but it has also strengthened the position of Barcelona as the network 'hub', and as the location of the 'original' festival.

Abstract

Most balanced translocations are thought to result mechanistically from nonhomologous end joining or, in rare cases of recurrent events, by nonallelic homologous recombination. Here, we use low-coverage mate pair whole-genome sequencing to fine map rearrangement breakpoint junctions in both phenotypically normal and affected translocation carriers. In total, 46 junctions from 22 carriers of balanced translocations were characterized. Genes were disrupted in 48% of the breakpoints; recessive genes in four normal carriers and known dominant intellectual disability genes in three affected carriers. Finally, seven candidate disease genes were disrupted in five carriers with neurocognitive disabilities (SVOPL, SUSD1, TOX, NCALD, SLC4A10) and one XX-male carrier with Tourette syndrome (LYPD6, GPC5). Breakpoint junction analyses revealed microhomology and small templated insertions in a substantive fraction of the analyzed translocations (17.4%; n = 4); an observation that was substantiated by reanalysis of 37 previously published translocation junctions. Microhomology associated with templated insertions is a characteristic seen in the breakpoint junctions of rearrangements mediated by error-prone replication-based repair mechanisms. Abstract

Global classification of the human proteins with regards to spatial expression patterns across organs and tissues is important for studies of human biology and disease. Here, we used a quantitative transcriptomics analysis (RNA-Seq) to classify the tissue-specific expression of genes across a representative set of all major human organs and tissues and combined this analysis with antibody- based profiling of the same tissues. To present the data, we launch a new version of the Human Protein Atlas that integrates RNA and protein expression data corresponding to 80% of the human protein-coding genes with access to the primary data for both the RNA and the protein analysis on an individual gene level. We present a classification of all human protein-coding genes with regards to tissue-specificity and spatial expression pattern. 38. High-resolution 3D reflection seismic investigation over a quick-clay landslide scar in southwest Sweden

Abstract

Quick-clay landslides often occur in the northern hemisphere in areas that were covered by Pleistocene glaciations. They are particularly common along the shorelines of the Göta River in southwestern Sweden. Characterization of potential landslide areas and identification of features that indicate high risk are necessary to better understand the triggering mechanisms of these events. Therefore, an intensive characterization project was initiated at the Fråstad landslide in Sweden. Part of the characterization program included the acquisition of 3D reflection seismic data to image structures in the normally consolidated sediments, as well as the bedrock topography below the landslide scar. Two seismic horizons within the glacial and postglacial sediments were observed. The shallowest seismic horizon (here, referred to as S1) corresponds to a coarse-grained layer that was previously detected by eight geotechnical boreholes located within the 3D survey area. Discontinuities in S1, mapped by the 3D reflection seismic data, occur across a zone that correlates with the landslide scar boundary, suggesting that this zone may have played a role in triggering and/or in limiting the extension of the landslide. If S1 is truncated by or mixed with clays in this zone, then the outflow of water from the permeable S1 into the clays above may have increased the amount of quick clays above this zone. The increased outflow of water may also have caused a higher pore-water pressure south of the zone, which in turn could have acted as a trigger for the landslide. We evaluated the potential of using the 3D reflection seismic method as a complement to drilling and other geophysical methods when performing landslide site investigations. We also demonstrated the importance of further investigating the relationship between 3D subsurface geometries and landslide development.

Abstract

Identifying the building blocks of mammalian tissues is a precondition for understanding their function. In particular, global and quantitative analysis of the proteome of mammalian tissues would point to tissue-specific mechanisms and place the function of each protein in a whole-organism perspective. We performed proteomic analyses of 28 mouse tissues using high-resolution mass spectrometry and used a mix of mouse tissues labeled via stable isotope labeling with amino acids in cell culture as a "spike-in" internal standard for accurate protein quantification across these tissues. We identified a total of 7,349 proteins and quantified 6,974 of them. Bioinformatic data analysis showed that physiologically related tissues clustered together and that highly expressed proteins represented the characteristic tissue functions. Tissue specialization was reflected prominently in the proteomic profiles and is apparent already in their hundred most abundant proteins. The proportion of strictly tissue-specific proteins appeared to be small. However, even proteins with household functions, such as those in ribosomes and spliceosomes, can have dramatic expression differences among tissues. We describe a computational framework with which to correlate proteome profiles with physiological functions of the tissue. Abstract

Difficulties encountered during the processing of the timelapse 3D land-seismic data at the CO2 geological storage site at Ketzin, Germany, were to a large extent attributed to changes in near-surface velocities. Two workflows for processing of the 4D data were tested. The first one included re-calculation of the refraction static corrections based on new information about the near-surface from first breaks. This workflow showed that the near-surface changes could only imperfectly be resolved by new refraction static corrections. The second workflow included cross-correlation of the traces acquired at the same locations but during different campaigns and calculation of the prestack time shifts between the surveys. Both workflows demonstrated their capability to minimize the time-lapse noise and enhance the time-lapse reservoir signature. They provide similar time-lapse results, except that the cross-correlation workflow is quicker, more accurate and displays less time-lapse noise.

44. High resolution reflection seismic profiling over the tjellefonna fault in the møre-trøndelag fault complex, Norway

Abstract

The Møre-Trøndelag Fault Complex (MTFC) is one of the most prominent fault zones of Norway, both onshore and offshore. In spite of its importance, very little is known of the deeper structure of the individual fault segments comprising the fault complex. Most seismic lines have been recorded offshore or focused on deeper structures. This paper presents results from two reflection seismic profiles, located on each side of the Tingvollfjord, acquired over the Tjellefonna fault in the southeastern part of the MTFC. Possible kilometer scale vertical offsets, reflecting large scale northwest-dipping normal faulting, separating the high topography to the southeast from lower topography to the northwest have been proposed for the Tjellefonna fault or the Baeverdalen lineament. In this study, however, the Tjellefonna fault is interpreted to dip approximately 50-60° towards the southeast to depths of at least 1.3 km. Travel-time modeling of reflections associated with the fault was used to establish the geometry of the fault structure at depth, while detailed analysis of first P-wave arrivals in shot gathers, together with resistivity profiles, were used to define the near surface geometry of the fault zone. A continuation of the structure on the northeastern side of the Tingvollfjord is suggested by correlation of an in strike direction P-S converted reflection (generated by a fracture zone) seen on the reflection data from that side of the Tingvollfjord. The reflection seismic data correlate well with resistivity profiles and recently published near surface geophysical data. A highly reflective package forming a gentle antiform structure was also identified on both seismic profiles. This structure could be related to the folded amphibolite lenses seen on the surface or possibly by an important boundary within the gneissic basement rocks of the Western Gneiss Region. Abstract

We present reprocessed data from seven profiles from the Forsmark site, the location where Sweden will store its spent nuclear fuel. The reprocessed data show improved images of the crust below 1 km since lower frequencies were preserved in the new processing sequences. These sections together with data from a longer more regional profile to the south of the area indicate that there may be a previously unidentified larger southwesterly dipping deformation or fracture zone in the southern part of the Forsmark area, well away from the planned repository.

Abstract

The Cascades-2 sounding rocket was launched on 20 March 2009 at 11: 04: 00 UT from the Poker Flat Research Range in Alaska, and flew across a series of poleward boundary intensifications (PBIs). The rocket initially crosses a diffuse arc, then crosses the equatorward extent of one PBI (a streamer), and finally crosses the initiation of a separate PBI before entering the polar cap. Each of the crossings have fundamentally different in situ electron energy and pitch angle structure, and different ground optics images of visible aurora. It is found that the diffuse arc has a quasi-static acceleration mechanism, and the intensification at the poleward boundary has an Alfvenic acceleration mechanism. The streamer shows characteristics of both types of acceleration. PFISR data provide ionospheric context for the rocket observations. Three THEMIS satellites in close conjunction with the rocket foot point show earthward flows and slight dipolarizations in the magnetotail associated with the in situ observations of PBI activity. An important goal of the Cascades-2 study is to bring together the different observational communities (rocket, ground cameras, ground radar, satellite) with the same case study. 47. High resolution reflection seismic imaging of the Ullared Deformation Zone, southern Sweden

Abstract

The Ullared Deformation Zone (UDZ) is one of a few structures worldwide known to contain decompressed eclogite facies rocks of Precambrian age. Given the unique nature of the Ullared eclogites, a 15 km long reflection seismic profile was acquired across the UDZ in April 2007. The principal objective of the profile was to provide geometrical information on the deformation zone at depth. The profile was acquired along a crooked line which gave us the opportunity to extract strike and dip information of reflections from the data-set using non-standard seismic techniques, including a cross-dip correction. This cross-dip correction method proved to be very useful for constraining the geometry of the reflectors at depth. The most recent (from 1997) published geological interpretation of the UDZ area was based on aeromagnetic data and the UDZ was interpreted as a shear zone, or possibly a shear zone system. The seismic data and a detailed aeromagnetic analysis support the interpretation of the UDZ as a shear zone system. Based on differences in reflectivity and geometry, the UDZ can be separated into four different units. The north-easternmost unit is the only one where eclogites have been found so far. Structures of the eclogite bearing unit are interpreted to dip approximately 20-30° towards northeast at depth. © 2011 Elsevier B.V.

48. Reflection seismic investigations in the Dannemora area, central Sweden: Insights into the geometry of polyphase deformation zones and magnetite-skarn deposits

Abstract

The Bergslagen region is one of the most ore prospective districts in Sweden. Presented here are results from two nearly 25 km long reflection seismic profiles crossing this region in the Dannemora mining area. The interpretations are constrained by seismic wave velocity measurements on a series of rock samples, cross-dip analysis, prestack time migration, and swath 3-D imaging, as well as by other available geophysical and geological observations. A series of major fault zones is imaged by the seismic data, as is a large mafic intrusion. However, the most prominent feature is a package of east-dipping reflectors found east of the Dannemora area that extend down to at least 3 km depth. This package is associated with a polyphase, ductile-brittle deformation zone with the latest ductile movement showing east-side-up or reverse kinematics. Its total vertical displacement is estimated to be in the order of 2.5 km. Also clearly imaged in the seismic data is a steeply dipping reflector near the Dannemora mine that extends down to a depth of at least 2.2 km. The geological nature of this reflector is not known, but it could represent either a fluid-bearing fault zone or a deep-seated iron deposit, making it an important target for further detailed geophysical and geological investigations.
