Tedera ( Bituminaria bituminosa vars. albomarginata and crassiuscula) is a perennial pasture species with agronomic characters ideally suited to Mediterranean climates. Tedera seed has a period of after-ripening or primary dormancy typically lasting three months, which delays assessment and breeding of elite hybrid varieties. Temperature, chemical and mechanical methods were investigated in conjunction with in vitro culture to circumvent this dormancy period across a range of parental and hybrid genotypes. Temperature treatment of T5 (Tedera accession 5) and T48 (Tedera accession 48) alone was not sufficient to break dormancy (24.0% and 14.7% germination); however, when combined with soaking in gibberellic acid ( GA3) and mechanical scarification resulted in 79.7% and 84.3% germination respectively. In an effort to further improve this result for valuable hybrid genotypes, we combined mechanical scarification with removal of seed coat after imbibition and in vitro culture on B5 medium until radicle emergence. This resulted in breaking dormancy from 96% to 100% of parent seeds and 100% of hybrid seeds. Hardening the germinated F1 or F2 seedlings 4 d after first transfer to in vitro culture resulted in 100% survival of plants to soil. Embryogenic cell cultures of Picea mariana (black spruce) and the species complex Picea glauca-engelmannii (interior spruce) were maintained either as suspensions in liquid medium in 250 or 500-ml-capacity shake-flasks, 7-liter-capacity airlift or mechanically stirred bioreactors, or on agar-solidified medium. Cultures from each of the maintenance conditions were subsequently transferred to agar-solidified LP medium containing 40 µM (±) -abscisic acid for maturation into cotyledonary stage embryos. For both species, the highest maturation frequency resulted from cultures grown in the airlift bioreactor. With black spruce cells grown in the airlift bioreactor containing LP medium with 60 m M sucrose, a maximum of 7.1 g·liter dry weight and 2892 embryos·ml were obtained after 15 days. For interior spruce cells, a maximum dry weight of 5.9 g·liter and 2698 embryos·ml were obtained after 21 to 30 days. During culture over 2 wk, ammonia was almost completely utilized by both species, wherease nitrate was depleted to 40% of the initial concentration. Sucrose was rapidly hydrolyzed to glucose and fructose by both species. Black spruce cultures preferentially metabolized glucose, whereas interior spruce preferentially metabolized fructose. Improved growth of interior spruce cells in mechanically stirred bioreactors occurred when cultured in LP medium with 60 m M fructose as the sole carbon source. [ABSTRACT FROM AUTHOR]

Summary: To induce androgenesis in field pea, grass pea and the model legume species Medicago truncatula, isolated microspores of various genotypes of these three species were submitted to a range of abiotic stresses prior to and during their initial culture, in order to stimulate them to divide and form embryos. Some stress agents had a positive effect on androgenesis from the treated microspores. Submission of flower buds to a cold period prior to anther excision or microspore isolation, modifying the osmotic pressure of the medium during initial culture and electroporation of isolated microspores were the three major individual stress agents to have an impact on the efficiency of androgenetic proliferation and subsequent differentiation from the microspores of pea, grass pea and M. truncatula genotypes. A combination of osmotic and electric shocks significantly improved responses from isolated microspores and yielded microcalluses and then calluses, but only few underwent morphogenesis. Further work is under way to improve responses and extend them to other genotypes. The results reported here are, to the best of our knowledge, the first successful results from isolated microspores of these species. [Copyright &y& Elsevier]

Unlabelled: Legumes are recalcitrant to androgenesis and induction protocols were only recently developed for pea (Pisum sativum L.) and chickpea (Cicer arietinum L.), albeit with low regeneration frequencies. Androgenesis is thought to be mediated through abscisic acid (ABA) but other phytohormones, such as auxins, cytokinins, and gibberellins, have also been implicated. In view of improving induction protocols, the hormone content of pea, chickpea, and lentil anthers was measured after exposure to cold, centrifugation, electroporation, sonication, osmotic shock, or various combinations thereof using an analytical mass spectrometer. Indole-3-acetic acid (IAA) had a key function during the induction process. In pea, high concentrations of IAA-asparagine (IAA-Asp), a putative IAA metabolite, accumulated during the application of the different stresses. In chickpea, the IAA-Asp concentration increased 30-fold compared to pea but only during the osmotic shock treatment and likely as a result of the presence of exogenous IAA in the medium. In contrast, no treatment in lentil (Lens culinaris) invoked such an increase in IAA-Asp content. Of the various cytokinins monitored, only cis zeatin riboside increased after centrifugation and electroporation in pea and possibly chickpea. No bioactive gibberellins were detected in any species investigated, indicating that this hormone group is likely not linked to androgenesis in legumes. In contrast to the other stresses, osmotic shock treatment caused a reduction in the levels of all hormones analyzed, with the exception of IAA-Asp in chickpea. A short period of low hormone content might be a necessary transition phase for androgenesis induction of legumes., Key Message: Five androgenesis-inducing stress treatments changed content of ABA, auxin and cytokinin in anthers of three legumes. Osmotic shock treatment differed because it reduced hormone content to very low levels.

This is the first report on the production of double-haploid chickpea embryos and regenerated plants through anther culture using Canadian cultivar CDC Xena (kabuli) and Australian cultivar Sonali (desi). Maximum anther induction rates were 69% for Sonali and 63% for CDC Xena. Under optimal conditions, embryo formation occurred within 15-20 days of culture initiation with 2.3 embryos produced per anther for CDC Xena and 2.0 embryos per anther for Sonali. For anther induction, the following stress treatments were used: (1) flower clusters were treated at 4 degrees C for 4 days, (2) anthers were subjected to electric shock treatment of three exponentially decaying pulses of 50-400 V with 25 microF capacitance and 25 Omega resistance, (3) anthers were centrifuged at 168-1,509g for 2-15 min, and finally (4) anthers were cultured for 4 days in high-osmotic pressure (563 mmol) liquid medium. Anthers were then transferred to a solid embryo development medium and, 15-20 days later, embryo development was observed concomitant with a small amount of callus growth of 0.1-3 mm. Anther-derived embryos were regenerated on plant regeneration medium. Electroporation treatment of anthers enhanced root formation, which is often a major hurdle in legume regeneration protocols. Cytological studies using DAPI staining showed a wide range of ploidy levels from haploid to tetraploid in 10-30-day-old calli. Flow cytometric analysis of calli, embryos and regenerated plants showed haploid profiles and/or spontaneous doubling of the chromosomes during early regeneration stages.

Interior spruce (Picea glauca engelmannii complex) and black spruce (Picea mariana Mill.) cotyledonary somatic embryos were encapsulated in sodium alginate. Somatic embryo viability was retained, but germination occurred at a reduced frequency compared with the equivalent zygotic embryos. The addition of 0.5% (w/v) activated charcoal to the alginate capsule significantly enhanced root development and germination for somatic embryos but not for zygotic embryos. The possibility of developing an artiflcal endosperm was also investigated, by addition of Litvay (Litvay et al. 1981) nutrients with or without 90 mM sucrose to the alginate-charcoal capsule. This treatment significantly enhanced root development for all embryo categories with the exception of black spruce somatic embryos. Encapsulated and non-encapsulated somatic embryos survived one month cold storage at 4 °C without reduction in germination frequency.

For optimization of the transformation procedure with Pisum sativum L. stern explant callus was used to test the effect of disarmed Agrobacterium tumefaciens strains, cocultivation procedures (preconditioning of explants; use of Nicotiana tabacum L. nurse cultures), duration of cocultivation (2, 3 or 4 days), and agents for selection (kanamycin or hygromycin). The succinamopine strain EHA101(pBI1042) produced the highest percentage of transformed calli (77%) when used in conjunction with tobacco nurse culture during four days of cocultivation. Using this strain, kanamycin (76%) and hygromycin (77%) were equally effective selective agents, but for strain LBA4404(pBI1042) percentage of transformed calli was higher for hygromycin (63%) than for kanamycin (17%). The procedures and strains shown to be optimal for transformation of pea callus will now be complemented by a pea regeneration system.
