In article, there are presented digital tools development results for geo-information support to seaports activity within Industry 4.0 period, when while climate change and covid-19 pandemic. In study, there are used methods of data bases constructing, web-technologies and virtual reality tools. Also, there are used Foresight technologies, theory of decision making under uncertainties and risk management. As digital tools, there is proposed to use digital online platforms, which integrate heterogeneous hardware and software resources with the use of web-technologies in distributed networks and wide application of cloud services. In study, there are used data bases and tools of geo-information digital online platform EOS, including its Land Viewer product. As study result, there is demonstrated usage of geo-information support system for Russian seaport Ust-Luga. The research results presented in this article has significant scientific novelty and can be useful for private investors, public environmental organizations of the civil sector and state environmental control bodies.

Recently, there is digital transformation of university education, including new concepts for obtaining and presenting learning materials. In article, there is considered digital learning technologies for geo-information management within Industry 4.0 while modern global economic crisis in last years. In research, there are used Foresight technologies, theory of decision making under uncertainties and risk management. Also, there are used methods of data bases constructing, web-technologies and virtual reality tools. As base technology, authors propose to use digital educational platforms, which integrate heterogeneous hardware and software resources using web technologies in distributed networks and a wide use of cloud services. Authors propose to use Google Classroom as essential digital educational platform. As study result, there are proposed enlarged groups of didactic works in geo-information management, oriented on practical purposes and adapted to Covid-19 pandemic conditions. There are considered the issues of digital content creation within university education, essentially in practical training. Presented in article results of study have a significant scientific novelty and can be used in educational and training purposes, including the preparation of Master's programs in geo-information management.

The paper presents the technologies of geoinformation support within hydraulic structures construction and exploitation at the stage of engineering-geological, engineering-ecological and engineering-hydrometeorological surveys for environmental economics during the implementation of major projects in the field of sustainable development of cities and municipalities. The impact of dredging on the natural environment is considered, and a conclusion about the need to study the rate of sedimentation at different stages of exploration, design, construction, and operation of underwater engineering facilities are made. It is shown how the construction of the Marine Facade of St. Petersburg for receiving tourist ferries changed the need for dredging volumes and how this affected the total cost of work. The port of Bronka in Neva Bay is used as an example of sedimentation study during the period of 2005-2007. The developed technologies should help in managing natural risks in the implementation of major projects in the field of construction and operation of hydraulic structures within the framework of the environmental economics in a changing climate.

Abstract Background In spite of a great number of monomeric fluorescent proteins developed in the recent years, the reported fluorescent protein-based FRET pairs are still characterized by a number of disadvantageous features, complicating their use as reporters in cell biology and for high-throughput cell-based screenings. Results Here we screened some of the recently developed monomeric protein pairs to find the optimal combination, which would provide high dynamic range FRET changes, along with high pH- and photo-stability, fast maturation and bright fluorescence, and reliable detection in any fluorescent imaging system. Among generated FRET pairs, we have selected TagGFP-TagRFP, combining all the mentioned desirable characteristics. On the basis of this highly efficient FRET pair, we have generated a bright, high contrast, pH- and photo-stable apoptosis reporter, named CaspeR3 (Caspase 3 Reporter). Conclusion The combined advantages suggest that the TagGFP-TagRFP is one of the most efficient green/red couples available to date for FRET/FLIM analyses to monitor interaction of proteins of interest in living cells and to generate FRET-based sensors for various applications. CaspeR3 provides reliable detection of apoptosis, and should become a popular tool both for cell biology studies and high throughput screening assays.

Abstract Background Genetically encoded sensors developed on the basis of green fluorescent protein (GFP)-like proteins are becoming more and more popular instruments for monitoring cellular analytes and enzyme activities in living cells and transgenic organisms. In particular, a number of Ca2+ sensors have been developed, either based on FRET (Fluorescence Resonance Energy Transfer) changes between two GFP-mutants or on the change in fluorescence intensity of a single circularly permuted fluorescent protein (cpFP). Results Here we report significant progress on the development of the latter type of Ca2+ sensors. Derived from the knowledge of previously reported cpFP-based sensors, we generated a set of cpFP-based indicators with different spectral properties and fluorescent responses to changes in Ca2+ concentration. Two variants, named Case12 and Case16, were characterized by particular high brightness and superior dynamic range, up to 12-fold and 16.5-fold increase in green fluorescence between Ca2+-free and Ca2+-saturated forms. We demonstrated the high potential of these sensors on various examples, including monitoring of Ca2+ response to a prolonged glutamate treatment in cortical neurons. Conclusion We believe that expanded dynamic range, high brightness and relatively high pH-stability should make Case12 and Case16 popular research tools both in scientific studies and high throughput screening assays.

Abstract Background Freshwater planarians are widely used as models for investigation of pattern formation and studies on genetic variation in populations. Despite extensive information on the biology and genetics of planaria, the occurrence and distribution of viruses in these animals remains an unexplored area of research. Results Using a combination of Suppression Subtractive Hybridization (SSH) and Mirror Orientation Selection (MOS), we compared the genomes of two strains of freshwater planarian, Girardia tigrina. The novel extrachromosomal DNA-containing virus-like element denoted PEVE (Planarian Extrachromosomal Virus-like Element) was identified in one planarian strain. The PEVE genome (about 7.5 kb) consists of two unique regions (Ul and Us) flanked by inverted repeats. Sequence analyses reveal that PEVE comprises two helicase-like sequences in the genome, of which the first is a homolog of a circoviral replication initiator protein (Rep), and the second is similar to the papillomavirus E1 helicase domain. PEVE genome exists in at least two variant forms with different arrangements of single-stranded and double-stranded DNA stretches that correspond to the Us and Ul regions. Using PCR analysis and whole-mount in situ hybridization, we characterized PEVE distribution and expression in the planarian body. Conclusions PEVE is the first viral element identified in free-living flatworms. This element differs from all known viruses and viral elements, and comprises two potential helicases that are homologous to proteins from distant viral phyla. PEVE is unevenly distributed in the worm body, and is detected in specific parenchyma cells.

The paper presents the results of experiments on measuring cross-sections for the neutron transfer channels 9Be(3He, α)8Begs,3.03 in the reaction of the 3He (30 MeV) ions with the 9Be target. To describe the angular distributions, we use the Distorted Wave Born Approximation (DWBA) applying the FRESCO code. The results of the theoretical analysis are in agreement with the experimental data. In addition, we perform calculations based on the solution of the time-dependent Schrödinger equation (TDSE) for the weakly bound neutron of the 9Be nucleus. The TDSE approach allows us to determine the dynamics of the neutron transfer process and calculate the probabilities for the transfer and removal of the neutron of the 9Be nucleus in the 3He + 9Be reaction.
