Abstract

The paper presents the results of experiments on measuring cross-sections for the neutron transfer channels 9Be(3He, α)8Begs,3.03 in the reaction of the 3He (30 MeV) ions with the 9Be target. To describe the angular distributions, we use the Distorted Wave Born Approximation (DWBA) applying the FRESCO code. The results of the theoretical analysis are in agreement with the experimental data. In addition, we perform calculations based on the solution of the time-dependent Schrödinger equation (TDSE) for the weakly bound neutron of the 9Be nucleus. The TDSE approach allows us to determine the dynamics of the neutron transfer process and calculate the probabilities for the transfer and removal of the neutron of the 9Be nucleus in the 3He + 9Be reaction. Abstract

In article, there are presented digital tools development results for geo-information support to seaports activity within Industry 4.0 period, when while climate change and covid-19 pandemic. In study, there are used methods of data bases constructing, web-technologies and virtual reality tools. Also, there are used Foresight technologies, theory of decision making under uncertainties and risk management. As digital tools, there is proposed to use digital online platforms, which integrate heterogeneous hardware and software resources with the use of web-technologies in distributed networks and wide application of cloud services. In study, there are used data bases and tools of geo-information digital online platform EOS, including its Land Viewer product. As study result, there is demonstrated usage of geo-information support system for Russian seaport Ust-Luga. The research results presented in this article has significant scientific novelty and can be useful for private investors, public environmental organizations of the civil sector and state environmental control bodies.

Abstract

Recently, there is digital transformation of university education, including new concepts for obtaining and presenting learning materials. In article, there is considered digital learning technologies for geo-information management within Industry 4.0 while modern global economic crisis in last years. In research, there are used Foresight technologies, theory of decision making under uncertainties and risk management. Also, there are used methods of data bases constructing, web-technologies and virtual reality tools. As base technology, authors propose to use digital educational platforms, which integrate heterogeneous hardware and software resources using web technologies in distributed networks and a wide use of cloud services. Authors propose to use Google Classroom as essential digital educational platform. As study result, there are proposed enlarged groups of didactic works in geo-information management, oriented on practical purposes and adapted to Covid-19 pandemic conditions. There are considered the issues of digital content creation within university education, essentially in practical training. Presented in article results of study have a significant scientific novelty and can be used in educational and training purposes, including the preparation of Master's programs in geo-information management.

Cytotoxic humanized anti-TRBV9 monoclonal antibody. We performed first-in-human targeted depletion of TRBV-restricted T cells in an HLA–B*27-positive patient with AS. TCR repertoires Preparation of unique molecular identifier (UMI)-labeled TCRβ cDNA libraries was performed using HUMAN TCR RNA MULTIPLEX kit (MiLaboratories) according to the manufacturer's manuals. Sequencing was performed on Illumina MiSeq and NextSeq 550, using 150+150 nt paired-end mode. MIGEC v1.2.9 was used for UMI-based read grouping and error correction. MiXCR v3.0.13 was used for extraction of TCRβ CDR3 repertoires. VDJtools v1.2.1 was used for downstream post-analysis. The repertoires include only productive sequences with more than one read per UMI.

Abstract

Cytotoxic humanized anti-TRBV9 monoclonal antibody. A single intravenous (i.v.) dose of the antibody administered to rhesus macaques (Macaca mulatta), resulted in prominent depletion of peripheral blood TRBV9+ T cells in a dose-dependent manner. Animals After quarantine, 12 Macaca mulatta males aged 5–8 years were selected for this experiment. For TCR repertoire profiling peripheral blood samples were collected before and 3, 40, 150, and 300 days after a single administration of BCD-180. We divided the animals into two groups (n = 4 for each group) that received 1 mg or 10 mg BCD-180 i.v. per animal, along with a control group that included four animals and received human intravenous IgG immunoglobulins (Microgen). Animals were kept in accordance with the guidelines for accommodation and care of laboratory animals, with species-specific provisions for nonhuman primates. The local ethics committee approved experiments for these studies. TCR repertoires Sequencing was performed on Illumina HiSeq1500. MIGEC v1.2.9 was used for UMI-based read grouping and error correction. MiXCR v3.0.13 was used for extraction of TCRβ CDR3 repertoires. VDJtools v1.2.1 was used for downstream post-analysis. Abstract

The paper presents the technologies of geoinformation support within hydraulic structures construction and exploitation at the stage of engineering-geological, engineering-ecological and engineering-hydrometeorological surveys for environmental economics during the implementation of major projects in the field of sustainable development of cities and municipalities. The impact of dredging on the natural environment is considered, and a conclusion about the need to study the rate of sedimentation at different stages of exploration, design, construction, and operation of underwater engineering facilities are made. It is shown how the construction of the Marine Facade of St. Petersburg for receiving tourist ferries changed the need for dredging volumes and how this affected the total cost of work. The port of Bronka in Neva Bay is used as an example of sedimentation study during the period of 2005-2007. The developed technologies should help in managing natural risks in the implementation of major projects in the field of construction and operation of hydraulic structures within the framework of the environmental economics in a changing climate.

Abstract

Autoimmunity is intrinsically driven by memory T and B cell clones inappropriately targeted at self-antigens. Selective depletion or suppression of self-reactive T cells remains a holy grail of autoimmune therapy, but disease-associated T cell receptors (TCRs) and cognate antigenic epitopes remained elusive. A TRBV9-containing CD8+TCR motif was recently associated with the pathogenesis of ankylosing spondylitis, psoriatic arthritis and acute anterior uveitis, and cognate HLA-B*27-presented epitopes were identified. Following successful testing in nonhuman primate models, here we report human TRBV9+T cell elimination in ankylosing spondylitis. The patient achieved remission within 3 months and ceased anti-TNF therapy after 5 years of continuous use. Complete remission has now persisted for 4 years, with three doses of anti-TRBV9 administered per year. We also observed a profound improvement in spinal mobility metrics and the Bath Ankylosing Spondylitis Metrology Index (BASMI). This represents a possibly curative therapy of an autoimmune disease via selective depletion of a TRBV-defined group of T cells. The anti-TRBV9 therapy could potentially be applicable to other HLA-B*27-associated spondyloarthropathies. Such targeted elimination of the underlying cause of the disease without systemic immunosuppression could offer a new generation of safe and efficient therapies for autoimmunity.
