Abstract Blastocystis is a species complex that exhibits extensive genetic diversity, evidenced by its classification into several genetically distinct subtypes (ST). Although several studies have shown the relationships between a specific subtype and gut microbiota, there is no study to show the effect of the ubiquitous Blastocystis ST1 on the gut microbiota and host health. Here, we show that Blastocystis ST1 colonization increased the proportion of beneficial bacteria Alloprevotella and Akkermansia, and induced Th2 and Treg cell responses in normal healthy mice. ST1-colonized mice showed decreases in the severity of DSS-induced colitis when compared to non-colonized mice. Furthermore, mice transplanted with ST1-altered gut microbiota were refractory to dextran sulfate sodium (DSS)-induced colitis via induction of Treg cells and elevated short-chain fat acid (SCFA) production. Our results suggest that colonization with Blastocystis ST1, one of the most common subtypes in humans, exerts beneficial effects on host health through modulating the gut microbiota and adaptive immune responses.

Deletion of the gene for Themis affects T cell selection in the thymus, which would be expected to affect the TCR repertoire. We found an increased proportion of cells expressing Vα3.2 (TRAV9N-3) in the peripheral CD8+ T cell population in mice with germline Themis deficiency. Analysis of the TCRα repertoire indicated it was generally reduced in diversity in the absence of Themis, whereas the diversity of sequences using the TRAV9N-3 V-region element was increased. In wild type mice, Vα3.2+ cells showed higher CD5, CD6 and CD44 expression than non-Vα3-expressing cells, and this was more marked in cells from Themis-deficient mice. This suggested a virtual memory phenotype, as well as a stronger response to self-pMHC. The Vα3.2+ cells responded more strongly to IL-15, as well as showing bystander effector capability in a Listeria infection. Thus, the unusually large population of Vα3.2+ CD8+ T cells found in the periphery of Themis-deficient mice reflects not only altered thymic selection, but also allowed identification of a subset of bystander-competent cells that are also present in wild-type mice.

The human gut microbiota is a diverse and complex ecosystem that is involved in beneficial physiological functions as well as disease pathogenesis. Blastocystis is a common protistan parasite and is increasingly recognized as an important component of the gut microbiota. The correlations between Blastocystis and other communities of intestinal microbiota have been investigated, and, to a lesser extent, the role of this parasite in maintaining the host immunological homeostasis. Despite recent studies suggesting that Blastocystis decreases the abundance of beneficial bacteria, most reports indicate that Blastocystis is a common component of the healthy gut microbiome. This review covers recent finding on the potential interactions between Blastocystis and the gut microbiota communities and its roles in regulating host immune responses.

Abstract In the gut, various subsets of intraepithelial T cells (IELs) respond to self or non-self-antigens derived from the body, diet, commensal and pathogenic microbiota. Dominant subset of IELs in the small intestine are TCRαβCD8αα+ cells, which are derived from immature thymocytes that express self-reactive TCRs. Although most of TCRαβCD8αα+ IELs are thymus-derived, their repertoire adapts to microbial flora. Here, using high throughput TCR sequencing we examined how clonal diversity of TCRαβCD8αα+ IELs changes upon exposure to commensal-derived antigens. We found that fraction of CD8αα+ IELs and CD4+ T cells express identical αβTCRs and this overlap raised parallel to a surge in the diversity of microbial flora. We also found that an opportunistic pathogen (Staphylococcus aureus) isolated from mouse small intestine specifically activated CD8αα+ IELs and CD4+ derived T cell hybridomas suggesting that some of TCRαβCD8αα+ clones with microbial specificities have extrathymic origin. We also report that CD8ααCD4+ IELs and Foxp3CD4+ T cells from the small intestine shared many αβTCRs, regardless whether the later subset was isolated from Foxp3CNS1 sufficient or Foxp3CNS1 deficient mice that lacks peripherally-derived Tregs. Overall, our results imply that repertoire of TCRαβCD8αα+ in small intestine expends in situ in response to changes in microbial flora.

Helios transcription factor and semaphorin receptor Nrp-1 were originally described as constitutively expressed at high levels on CD4+Foxp3+ T regulatory cells of intrathymic origin (tTregs). On the other hand, CD4+Foxp3+ Tregs generated in the periphery (pTregs) or induced ex vivo (iTregs) were reported to express low levels of Helios and Nrp-1. Soon afterwards the reliability of Nrp-1 and Helios as markers discriminating between tTregs and pTregs was questioned and until now no consensus has been reached. Here, we used several genetically modified mouse strains that favor pTregs or tTregs formation and analyzed the TCR repertoire of these cells. We found that Tregs with variable levels of Nrp-1 and Helios were abundant in mice with compromised ability to support natural differentiation of tTregs or pTregs. We also report that TCR repertoires of Treg clones expressing high or low levels of Nrp-1 or Helios are similar and more alike repertoire of CD4+Foxp3+ than repertoire of CD4+Foxp3- thymocytes. These results show that high vs. low expression of Nrp-1 or Helios does not unequivocally identify Treg clones of thymic or peripheral origin.

Internal audit is performed in a diverse legal and cultural environment, for organizations that differ in their goals, size, complexity and structure. Internal auditing is performed by people both inside and outside the organization. Although the aforementioned differences may affect the practice of auditing in different environments, the application of the International Standards for the Professional Practice of Internal Auditing is an essential condition for the internal auditor and internal audit to fulfill their duties. Internal control is an immanent feature of the management process. In the functional sense, it is performed by each employee, in the institutional sense by a separate internal cell. Hence, the control system consists of: internal cells in the organizational structure, internal regulations, procedures and other control mechanisms in the enterprise/unit. Internal control operates on an ongoing basis and can respond immediately to any irregularities. Financial control is a special type of internal control - it deals with finances. Internal audit comes into contact with internal control during one of the many activities it undertakes, namely the examination and evaluation of the effectiveness of the existing internal control system.

To increase the anti-wear (AW) and anti-scuffing possibilities of commercially available lithium grease, this paper proposed enriching the original composition with functionalised carbon nanotubes (CNTs) at a concentration of 0.1% (w/w). The CNTs were modified by decorating them with nanoparticles of two metals with established tribological potential: copper and nickel. The AW and extreme-pressure properties were determined using the customised ISO-20623 test on a four-ball apparatus. The AW properties were determined using the standardised parameter MWSD (mean wear scar diameter) and the anti-scuffing properties using the last non-seizing load. The greases enriched with nanoadditives showed better AW properties compared to the reference grease at higher loads (1–1.2 kN). Particularly favourable results were observed for grease with the addition of Cu-decorated CNTs, for which the MWSD values were more than 50% lower than the reference. Optical microscopy, SEM and TEM microscopy with EDS analysis, and Raman spectroscopy were used to identify the wear mechanisms and characterise the role of nanoadditives in the lubrication process.

Abstract New public management introduces a managerial approach to managing the public sector and administration from the perspective of human resources management. This approach assumes the adaptation of efficiency mechanisms and management methods and techniques used in the private sector to the conditions of managing public organizations. The model assumes effective implementation of public cost accounting, an approach based on achieving results, decentralization of management, wider consideration of public-private partnerships and extensive use of market mechanisms. This management model is intended to ensure the economy, efficiency and effectiveness of the organization, including the privatization of some public services. Streszczenie Nowe zarządzanie publiczne wprowadza podejście menedżerskie do zarządzania sektorem publicznym i administracją z perspektywy zarządzania zasobami ludzkim. Takie podejście zakłada adaptację mechanizmów efektywnościowych oraz metod i technik zarządzania stosowanych w sektorze prywatnym do warunków zarządzania organizacjami publicznymi. Model zakłada efektywne wdrożenie rachunku kosztów publicznych, podejście oparte o osiąganie wyników, decentralizację zarządzania, szersze uwzględnienie partnerstwa publiczo-prywatnego oraz szerokie wykorzystanie mechanizmów rynkowych. Ten model zarządzania ma zapewnić gospodarność, efektywność i skuteczność organizacji, w tym prywatyzację części usług publicznych.

Objectives In a market economy, allocation decisions are more or less visibly related to the monetary savings of entities. The tendency of entities to save depends on both the interest rate on deposits and inflation, as well as the tax rate on income from capital (monetary savings). Also, the willingness of business entities to invest depends on the income from invested capital. Tax policy, its nature, implemented solutions, instruments in the form of reliefs and exemptions, and, above all, stability and predictability are an important element of effectively influencing taxpayers' behavior. Material and methods Induction was used as the main research method. It involves drawing general conclusions or establishing regularities based on the analysis of empirically identified phenomena and processes. Moreover, the work uses two general research methods, i.e. analytical and synthetic methods, characterized by a specific approach to the study of reality. Results To ensure economic growth and development, it is necessary to develop favorable tax solutions. Taxes should not affect the taxpayer's assets to an extent that prevents their further development. Therefore, it is important to balance the amount of tax collected from the company. Conclusions A simple consequence of the fiscal function of income taxes is a direct impact on the allocation of resources in the economy, because when the tax is paid, there is a final flow of income between the taxpayer and the state. The fiscal function of income taxes is always related to the allocation of resources, as it reduces the incomes of households and businesses, which limits their ability to invest, consume and save.

Abstract The stabilisation function of the state's finances encompasses measures aimed, inter alia, at achieving and maintaining a relatively high rate of economic growth, while limiting negative phenomena, i.e. high unemployment and inflation rates, mitigating business cycle fluctuations, stabilising the money market and making the most effective use of tangible factors of production. The role of the state in the economy and its influence on the behaviour of groups of economic units has undergone dynamic changes over the centuries, both with regard to the forms and scope of influence and the degree of interference of public authorities in the market mechanism Streszczenie Stabilizacyjna funkcja finansów pastwa obejmuje działania mające między innymi powodować osiągnięcie i utrzymanie relatywnie wysokiego tempa wzrostu gospodarczego, przy jednoczesnym ograniczaniu zjawisk negatywnych, tj. wysokiej stopy bezrobocia i inflacji, łagodzeniu wahań cyklu koniunkturalnego, stabilizację rynku pieniężnego oraz możliwie najbardziej efektywne wykorzystanie rzeczowych czynników produkcji. Rola państwa w gospodarce i jego wpływ na zachowanie się grup jednostek gospodarujących ulegała dynamicznym zmianom na przestrzeni wieków, zarówno odniesieniu do form i zakresu oddziaływania, jak i stopnia ingerencji władz publicznych w mechanizm rynkowy.

Objectives The purpose of the article is a theoretical analysis of the process of introducing a new product to the market, taking into account economic and social aspects. Material and methods Induction was used as the main research method. It involves drawing general conclusions or establishing regularities on the basis of analysis of empirically established phenomena and processes. It is a type of inference based on details about the general properties of a phenomenon or object. In addition, the paper uses two general research methods, i.e. analytical and synthetic methods, characterized by a particular approach to the study of reality. Results First: the process of implementing new products/services should correlate with the company's development strategy Second: the development and introduction of new products/services as part of the company's activities must be in line with the development strategy and mission. Third: each in the process of planning and marketing of new products / services - distinguish eight stages (milestones of the process). Fourth: The consumer communication strategy should be carefully designed, written out, and then implemented step by step. Conclusions First: the various organizational cells/accounting, marketing, legal departments, as well as development and implementation departments/ must know the essence of the product/service to be introduced for sale. Second: business units must have knowledge of: sales strategy, P&L, forecasts of A&P budgets, forecast of product profitability and implementation schedule. Third: when preparing a recommendation for planning and budgeting sales of new products/services, the individual organizational units of the form should know specific parameters. Fourth: the various organizational units should have knowledge of the prepared sales strategy.

The paper presents studies related to positive psychology that have been conducted in Poland. The article is based on a review of texts available in the databases of Google Scholar and EBSCO published by Polish researchers in the years 2001–2021. The selection criterion was the presence of some concepts in the title and/or abstract of articles that are constructs of positive psychology. Thirty intentionally selected articles were analyzed in terms of research objectives, variables, and measurement tools they described. In this way, a picture of research interests and methods of research on human and organization from the perspective of positive psychology was obtained. It seems that the main subjects of interest of Polish researchers were problems related to positive human qualities, life satisfaction, and mental well-being with a growing interest in positive society. The most common variable to study was life satisfaction, and the most frequently used measuring tool was the SWLS. It was concluded that there is a need for scientific exchange and integration within Polish researchers and the international community of positive psychologists.

The fascinating tribological phenomenon of carbon nanotubes (CNTs) observed at the nanoscale was confirmed in our numerous macroscale experiments. We designed and employed CNT-containing nanolubricants strictly for polymer lubrication. In this paper, we present the experiment characterising how the CNT structure determines its lubricity on various types of polymers. There is a complex correlation between the microscopic and spectral properties of CNTs and the tribological parameters of the resulting lubricants. This confirms indirectly that the nature of the tribological mechanisms driven by the variety of CNT–polymer interactions might be far more complex than ever described before. We propose plasmonic interactions as an extension for existing models describing the tribological roles of nanomaterials. In the absence of quantitative microscopic calculations of tribological parameters, phenomenological strategies must be employed. One of the most powerful emerging numerical methods is machine learning (ML). Here, we propose to use this technique, in combination with molecular and supramolecular recognition, to understand the morphology and macro-assembly processing strategies for the targeted design of superlubricants.

The article presents a new method of selecting operating parameters of ve-hicles which allows to prolong their useful life. The theoretical and practical aspects of the new approach were discussed. The key methods of optimizing the use of vehicles and their transport routes were analysed. The proposed method of selecting operating parameters of transport vehicles was based on elements of the dynamic programming model using heuristic algorithms. A model and then an algorithm and a computer program for selecting operating parameters for vehicles were developed.

Modernist architecture is one of the most significant movements which serve as grounds for contemporary creations. At the beginning of the 20th century, the impact of new structural and building technologies allowed designers to find new ways of architectural expression. It is rooted in thought-out composition and simplicity which serve as a background for the exposition of structure. The geometrical interplay of a building’s elements highlights the impressively thin structures and raw surfaces of novel building materials. Nowadays, in selected regions, the architecture of the Modernist Period is neglected or loosely refurbished. As an effect of this phenomenon, buildings are demolished or deprived of aesthetical values. Thus, this article aims at showing an overview of sectional research on good practice. We base the study of on-site design implementations of several modernist architecture refurbishments and focus on technology and design assumptions with optimised thermal modernisation. The paper reveals a set of examples for the refurbishment of modernist architecture, with calculations of heat energy coefficients of the initial and design phase. The presented thermal modernisations aim at adjusting buildings to new requirements concerning energy without any loss of initial architectural expression.

The article presents a computer algorithm to calculate the estimated operating cost analysis rail bus. This computer application system compares the cost of employment locomotive and wagon, the cost of using locomotives and cost of using rail bus. An intensive growth of passenger railway traffic increased a demand for modern computer systems to management means of transportation. Described computer application operates on the basis of selected operating parameters of rail buses.

Introduction. The progressive mechanisation of work in every area of life and the rapid development of means of transport have a major impact on the steadily increasing number of injuries. Spinal injuries are a major medical problem due to the complex structure of the spine, and particularly the neural elements located in the spine. Aim. The analysis of causes, the number and type of injuries of the spine and spinal cord trauma performed on the basis of the documentation from the Emergency Department (ED) of Lower Silesia Specialized Hospitalname Tadeusz Marciniak in Wroclaw and documentation from the Clinic and the University Clinical Hospital (UCH) in Wroclaw. Material and Methods. A retrospective analysis was performed on the documentation of 530 patients from the ED at Lower Silesia Specialized Hospital name Tadeusz Marciniak in Wroclaw who suffered a back injury and/or spinal cord injury and 45 patients from the Clinic and the Department of Neurosurgery at UCH in Wroclaw who were treated for a spine injury and spinal cord injury. Results. The analysis of the study material shows that: — more than a half of the ED patients (58.87%) and 44% of those hospitalized in the Department of Neurosurgery suffered a spinal injury during a traffic collision; — more men were patients in the Department of Neurosurgery (60%), whereas among the ED patients women were slightly more often hospitalized because of a spine injury (52.64%); — the vast majority of respondents with spinal injury were brought to hospital by emergency teams. Conclusions. 1. The predominant reason for spinal injuries and spinal cord injuries being motor vehicle accidents. 2. Mainly young people aged 20–35 years suffer spine and spinal cord injuries. 3. Spine injury most often refers to the cervical part of the spine. (JNNN 2014;3(4):153–156)

Ambient marketing as a new language of marketing communication and the original use in urban space Ambient marketing is one of the tools that belong to the so-called guerrilla marketing. It is a reaction to the rejection of classical types of marketing with their forms of persuasion. It aims at creating a new form of communication and a dialogue that often takes place in the urban areas. It achieves these goals by using the natural scenery of the city and integrating it within the creation of original, unique, and interactive objects and new communication situations.

The authors analyze a pre-election poster of the Slovak parliamentary party, the Slovak National Party (SNS). The poster has been selected for analysis as it is strikingly similar to the poster of the extreme right-wing party, Jobbik, which operates in Hungary. An attempt at ascertaining an instance of plagiarism has been made by first looking for plagiarized elements and then for creative elements of the poster. The analysis has been conducted in the communicative framework and is based on the principle of intertextuality.
