Background: Type 1 diabetes mellitus is a prevalent autoimmune disease worldwide. The knowledge of female particularities in metabolic dysfunction is of fundamental importance, leading to better choices for human therapy candidates. The aim of this study is to investigate the glucose flux peculiarities of female rats submitted to two classic experimental diabetes protocols. Methods: Female Wistar rats, 60 days old, were used to evaluate biochemical and hormonal serum parameters, in addition to skeletal muscle and liver energy stocks and 14C-glucose and 14C-alanine flux. Two different protocols, multiple (25 mg/kg dose) and single (65 mg/kg dose) intraperitoneal streptozotocin, were compared considering the alterations presented 48 h and 30 days after the drug administration. Results: The results showed few indicators of muscle and liver metabolic imbalance. High-single streptozotocin dose promoted 97% and 41% lower glycogen levels in liver and muscle respectively. Multiple-low streptozotocin dose promoted 63% lower lipid synthesis in liver. After 30 days, diabetic animals presented hyperglycaemia in both protocols, 589.5 (529.3/642.3) mg/dL to high-single dose and 374.2 (339.3/530.6) mg/dL to multiple-low dose. However, they did not present lower insulin levels, alterations on muscle glucose uptake, nor higher hepatic gluconeogenesis. Conclusion: In conclusion, this study demonstrates that females, at least Wistar rats, are less responsive to classic diabetes protocols established in literature, so mechanisms of experimental diabetes for females need more investigation. After which, therapeutic candidates should be evaluated in such a way sex bias does not present itself as a factor that hinders reproducibility in human studies.

Background and aim: Metabolic disturbances are known for their increasing epidemiological importance. Ilex paraguariensis presents a potential option for mitigating lipid metabolism imbalance. However, most of the literature to date has not considered sex bias. This study aimed to evaluate the effect of Ilex paraguariensis on the metabolism of different adipose tissue depots in males and females. Experimental procedure: After ovariectomy, female Wistar rats received daily treatment with the extract (1 g/kg) for forty-five days. Biochemical serum parameters and tissue metabolism were evaluated. Oxidation, lipogenesis and lipolysis were evaluated in brown, white visceral, retroperitoneal and gonadal adipose tissues. Results and conclusion: The results showed that treatment with the extract led to a reduced weight gain in ovariectomised females in comparison to control. The triglyceride concentration was decreased in males. Glucose oxidation and lipid synthesis in visceral and retroperitoneal adipose tissues were restored in ovariectomised females after treatment. The response to epinephrine decreased in visceral adipose tissue of control males; however, lipolysis in females did not respond to ovariectomy or treatment. These findings highlight the enormous potential effects of I. paraguariensis on lipid metabolism, modulating lipogenic pathways in females and lipolytic pathways in males. Furthermore, the sex approach applied in this study contributes to more effective screening of the effects of I. paraguariensis bioactive substances.

The influence of yerba mate extract (YME) and two concentrated fractions of methylxanthines–polyphenols (MXPL) and saponins (SFL) from yerba mate on the lipid metabolism of Wistar rats fed with a high-fat diet were examined. A Box–Behnken design was employed for the optimization of yerba mate extraction. Subsequently, one-step separation using solid phase fractionation was applied. HPLC–PDA and UPLC/Q-TOF-MS were used for quantification and identification of the main compounds. YME induced reductions on intra-abdominal fat weight gain and LDL plasma level. MXPL decreased plasma levels of cholesterol, LDL and triacylglycerols. In addition, this fraction increased lipogenesis of the muscle, hepatic glycogen synthesis and lipolysis in adipose tissue. SFL enhanced lipogenesis in adipose tissue and also fecal fat excretion comparing to both controls. Moreover, all mate preparations could entirely revert the hepatic lipogenesis induced by the high-fat diet. Thus, the yerba mate extract and fractions exhibited a potential anti-obesity effect on lipid metabolism of the Wistar rat and deserve further investigation.

FUNDAMENTO: A ressuscitação de parada cardíaca pode apresentar disfunção miocárdica determinada pelo tempo da isquemia, e a inibição da enzima conversora de angiotensina (ECA) pode reduzir a disfunção cardíaca durante a reperfusão. OBJETIVO: Investigar os efeitos da angiotensina-I e diferentes períodos de isquemia na recuperação funcional em corações de ratos isolados. MÉTODOS: Os corações isolados de ratos Wistar (n = 45; 250-300 g) foram submetidos a diferentes períodos de isquemia global (20, 25 ou 30 min) e reperfundidos (30 min) com o tampão Krebs-Henseleit, ou com a adição de 400 nmol/L de angiotensina-I, ou com 400 nmol/L de angiotensina-I + 100 µmol/L de captopril durante o período de reperfusão. RESULTADOS: A derivada positiva máxima de pressão (+dP/dt max) e o produto frequência-pressão foram reduzidos nos corações expostos à isquemia de 25 min (~ 73%) e à isquemia de 30 min (~ 80%) vs. isquemia de 20 min. A pressão diastólica final do ventrículo esquerdo (PDFVE) e a pressão de perfusão (PP) foram aumentadas nos corações expostos à isquemia de 25 min (5,5 e 1,08 vezes, respectivamente) e à isquemia de 30 min (6 e 1,10 vezes, respectivamente) vs. isquemia de 20 min. A angiotensina-I ocasionou uma diminuição no +dP/dt max e no produto frequência-pressão (~ 85-94%) em todos os períodos de isquemia e um aumento na PDFVE e na PP (6,9 e 1,25 vezes, respectivamente) apenas na isquemia de 20 min. O captopril foi capaz de reverter parcial ou completamente os efeitos da angiotensina-I na recuperação funcional nas isquemias de 20 e 25 min CONCLUSÃO: Os dados sugerem que a angiotensina-II participa direta ou indiretamente no dano pós-isquêmico e que a capacidade de um inibidor da ECA atenuar esse dano depende do tempo de isquemia.

In this paper, two methods for assessing the degree of melanization of pupal exuviae from the butterfly Heliconius erato phyllis , Fabricius 1775 (Lepidoptera, Nymphalidae, Heliconiini) are compared. In the first method, which was qualitative, the exuviae were classified by scoring the degree of melanization, whereas in the second method, which was quantitative, the exuviae were classified by optical density followed by analysis with appropriate software. The heritability (h 2 ) of the degree of melanization was estimated by regression and analysis of variance. The estimates of h 2 were similar with both methods, indicating that the qualitative method could be particularly suitable for field work. The low estimates obtained for heritability may have resulted from the small sample size ( n = 7-18 broods, including the parents) or from the allocation-priority hypothesis in which pupal color would be a lower priority trait compared to morphological traits and adequate larval development.

INTRODUÇÃO: Vários estudos têm apontado Ilex paraguariensis (erva-mate) como coadjuvante no manejo da obesidade. O objetivo do estudo foi avaliar ingestão alimentar, peso corporal, volume da diurese, quantidade de gordura abdominal, triglicérides e colesterol total plasmáticos de ratos Wistar machos tratados com extrato aquoso de Ilex paraguariensis. MÉTODOS: Formaram-se dois grupos de seis animais cada, um controle e outro tratado. O tratado recebia extrato de Ilex paraguariensis e o controle, água para hidratação. Todos receberam ração padrão. Para o preparo do extrato da erva, misturava-se 1 L de água aquecida à 80 ºC e 70 g de erva-mate. Em 15 minutos, essa mistura era coada. Após 8 semanas, os animais foram colocados em gaiolas metabólicas para avaliar a ingestão alimentar e hídrica e o volume de diurese e fezes. Nesse período, foi aferido o peso, foram coletadas amostras de sangue para quantificar triglicérides e colesterol e a gordura abdominal foi dissecada após a morte dos animais. Na análise estatística, foi utilizado o teste t de Student para o tratamento de todos os resultados, e P < 0,05 foi considerado significativo. RESULTADOS: Ingestão alimentar e hídrica, diurese, fezes e peso dos animais não apresentaram qualquer diferença significativa, assim como os níveis de triglicérides. A quantidade de gordura abdominal, assim como os níveis de colesterol, foi significativamente menor nos animais tratados. CONCLUSÃO: O extrato da erva-mate parece ter influência sobre o metabolismo dos lipídios sem interferir no peso corporal nem na ingestão alimentar e hídrica.
