Bacterial strains isolated from cankers of wild cherry trees (Prunus avium) in France were characterized using numerical taxonomy of biochemical tests, DNA-DNA hybridization, repeat sequence primed-PCR (rep-PCR) based on REP, ERIC and BOX sequences, heteroduplex mobility assay (HMA) of internal transcribed spacer (ITS) as well as pathogenicity on wild cherry trees and other species of Prunus. They were compared to reference strains of Pseudomonas syringae pathovars isolated from wild and sweet cherry and various host plants. Wild cherry strains were closely related to P syringae (sensu lato) in LOPAT group la (+ - - - +). Wild cherry strains were pathogenic to wild cherry trees and produced symptoms similar to those observed in orchards. They were pathogenic also, but at a lesser extent, to sweet cherry trees (cv. Napoleon). The wild cherry strains were collected from five different areas in France and appeared to constitute a very homogeneous group. They showed an homogenous profile of a biochemical and physiological characteristics. They were closely related by DNA-DNA hybridization and belonged to genomospecies 3 'tomato'. Rep-PCR showed that wild cherry strains constitute a tight group distinct from P.s. pv. morsprunorum races 1 and 2 and from other P syringae pathovars. HMA profiles indicated that the ITS of all wild cherry strains were identical but different from P.s. pv. persicae strains since the two heteroduplex bands with reduced mobility were generated by hybridization with the P.s. pv. persicae pathotype strain CFBP 1573. The 8 genomospecies of Gardan et al. (1999) have not been converted into formal species as they cannot be differentiated by biochemical tests. Therefore, the pathovar system within P syringae was currently used. P syringae pv. avii is proposed for this bacterium causing a wild cherry bacterial canker and strain CFBP 3846 (NCPPB 4290, ICMP 14479) is designated as the pathotype.

Le chancre bactérien est une maladie des agrumes dont l'incidence économique est globalement importante. Bien qu'ayant fait l'objet d'un grand nombre de publications scientifiques au cours des 40 dernières années, cette maladie demeure lune des contraintes de la filière agrumicole et il a été souligné que des recherches visant à préciser l'épidémiologie de cette maladie en zones tropicales étaient nécessaires. La contribution du Cirad à l'île de la Réunion se situe à ce niveau. Les principaux résultats acquis au cours de la dernière décennie ont concemé: - la caractérisation de l'agent pathogène, Xanthomonas axonopodis pv. citri (Xac), associé à cette maladie dans l'archipel des Mascareignes; - l'analyse des caractéristiques biologiques et épidémiologiques de la bactérie dans les conditions réunionnaises, ce qui a permis de préciser les périodes à risque en fonction du stade phénologique de l'hôte et des facteurs environnementaux, d'évaluer globalement l'importance biologique des populations associées à différents modes de survie de Xac, et de préciser l'importance de là qualité sanitaire des pépinières dans une stratégie de lutte Intégrée contre le chancre bactérien; - une analyse plus fine de l'épidémiologie de Xac en pépinières (exsudation de l'agent pathogène à partir de lésions foliaires, modélisation de l'évolution spatio-temporelle de la maladie dans des pépinières expérimentales en fonction d'itinéraires culturaux et des paramètres environnementaux); - la mise au point d'outils de diagnostic moléculaire basés sur là PCR; - une évaluation de la sensibilité à Xac d'une gamme de cultivars et hybrides d'agrumes (principalement orangers et mandariniers). Ces données et outils sont actuellement intégrés dans un schéma visant à moderniser les pépinières agrumicoles à la Réunion.

A cytochemical investigation was carried out on the development of an aggressive strain of #Xanthomonas campestris$ pv. #manihotis$, responsible for the cassava bacterial blight, to gain better insights into molecular and cellular mechanisms involved in leaf cell wall degradation by this pathogen. The use of anti-pectin monoclonal antibodies revealed that the plant middle lamellae were damaged during the infection process, from the epiphytic stage on the leaf surface to invasion of vascular bundles. In parallel, application of a beta-1,4-exoglucanase-gold probe to healthy and infected tissues indicated that primary and secondary cell walls were also altered. Quantitation of gold labeling confirmed that pectin was more severely degraded than cellulose. Accumulation of pectin-like compounds was also detected in occluded infected vessels. Bacterial-surrounding sheaths, which were routinely seen during pathogenesis early after leaf inoculation, had a dense or loosened fibrillar appearance and were differentiated from the pathogen cell wall. Close association occurred between extracellular fibrils and leaf cell walls, both at early and advanced stages of wall degradation. Bacterial extracellular sheaths were often seen deep in host cell walls, sometimes enclosing residual plant cell wall fragments. Our cytochemical data demonstrated that cell wall degradation of cassava by #Xanthomonas campestris$ pv. #manihotis$ plays an important role in host tissue colonization. It is also suggested that bacterial extracellular sheaths are involved in plant cell surface degradation. (Résumé d'auteur)

Pseudomonas syringae pv persicae, causal agent of a bacterial dieback has destroyed more than 1 million peach in the Rhône Valley since it occurred for the first time in 1966. There is no resistant variety and all the cultivated varieties are distributed in 3 classes of susceptibility according to the results of a 3-yr field trial. Carrying out inoculation tests on in vitro plants has been considered. Experiments were performed with either a hybrid almond x peach line or with 3 peach varieties corresponding to each previously defined susceptibility class. Two bacterial clones differing in aggressiveness were used for inoculation. The inoculation of rapidly growing almond x peach in vitro plants did not lead to any significant differentiation of the 2 bacterial clones. When treated by exposure either to negative temperature (-4 °C) for 1 hr or to low temperature (6 °C) for the duration of the experiment in addition with an inverted photoperiod (6/18) the in vitro plants reacted according to the aggressiveness of the inoculated clones. Moreover, the 3 peach varieties demonstrated a differential susceptibility to the bacterial clones similar to that observed in field. The test which has been developed on in vitro plants appeared to be useful in differentiating varieties with high susceptibility from those with low susceptibility but not accurate enough for varieties with intermediate susceptibility. It could be used as a preliminary test, either to discard the most susceptible varieties before introduction into orchard or before the field trial. It could also be performed to determine the aggressiveness of strains.; Pseudomonas syringae pv persicae, bactérie pathogène du pêcher, a détruit plus d’un million d’arbres dans la Vallée du Rhône depuis son apparition (1966). Aucune variété n’est résistante à cette bactérie, mais un gradient de sensibilité variétale existe et les variétés se répartissent en trois classes de sensibilité. Trois années d’expérimentation au champ sont au minimum nécessaires pour déterminer le comportement de chacune des nouvelles obtentions variétales vis-à-vis de ce Pseudomonas. L’utilisation du vitroplant, pour cribler les variétés vis-à-vis de P s pv persicae, a donc été envisagée. Le vitroplant en croissance réagit de la même manière face à des clones bactériens d’agressivités différentes. Sur un clone d’amandier x pêcher, hybride interspécifique, sensible à cette bactérie, un traitement thermique des vitroplants a permis d’obtenir des réponses différenciées après inoculation de 2 clones bactériens, l’un agressif et l’autre non agressif. Il s’agit soit d’une exposition d’une heure à -4 °C, soit d’un séjour des vitroplants à 6 °C avec une photopériode inversée (6/18 heures). Sur 3 variétés de pêcher, chacune appartenant à l’une des 3 classes de sensibilité à Ps pv persicae, le même type de traitement thermique a été appliqué après inoculation des 2 clones bactériens. Un gradient de sensibilité, conforme à celui constaté en verger, a été obtenu sur vitroplants entre les 3 variétés testées. Ce test permet de discriminer les variétés et porte-greffes très sensibles des variétés et porte-greffes peu sensibles. En revanche il est plus difficile de situer les variétés et porte-greffes de sensibilité intermédiaire. Ce test permettrait de repérer très tôt les variétés qu’il ne faut pas planter dans la région concernée par la maladie. Le vitroplant pourrait être également utilisé pour mesurer l’agressivité des souches.

Pseudomonas syringae, l’une des 50 espèces bactériennes phytopathogènes, est caractérisée par une grande variabilité génétique, physiologique et biologique qui s’exprime au travers des 52 pathovars qui la composent. L’espèce Pseudomonas syringae, décrite sur près de 400 espèces végétales (le seul pathovar syringae a été signalé sur 177 hôtes), possède 3 propriétés importantes : un pouvoir pathogène, un pouvoir glaçogène et une aptitude à la vie épiphyte. Dans une première partie, les principales caractéristiques de l’espèce, hormis les caractères génétiques non significativement développés ici, sont présentées. Dans la seconde partie, les 3 propriétés remarquables sont analysées en détail. L’aptitude épiphyte de Pseudomonas syringae s’exprime par une capacité à coloniser la surface des organes aériens des plantes, à s’y multiplier de façon importante. Elle conduit à l’existence de niveaux de population élevés au printemps, et en automne aussi pour les plantes pérennes; ces populations épiphytes constituent un réservoir d’inoculum et sont essentielles pour l’initiation de l’infection. La distribution de ces populations épiphytes, en général abondantes, n’est pas homogène ni entre plantes, ni entre organes d’une même plante ; leur répartition à la surface de la feuille qui constitue leur support essentiel est aussi très hétérogène. La nature de l’interaction entre l’hôte et les bactéries épiphytes n’est pas connue ; l’existence d’un lien entre spécificité parasitaire et la spécificité épiphyte au sein de l’espèce syringae est suspectée. Le pouvoir glaçogène de l’espèce syringae s’exprimant par une capacité à induire une rupture précoce de la surfusion de l’eau ne concerne qu’une fraction de pathovars, essentiellement le pathovar syringae. On a montré qu’il était impliqué dans la gélivité des plantes au printemps (vigne, arbres fruitiers, tomate, pomme de terre...) et qu’il est l’un des facteurs favorisant les premières phases de l’infection bactérienne. Le pouvoir pathogène de Pseudomonas syringae montre une spécificité d’hôte liée au pathovar considéré, sauf pour le pathovar syringae dont la gamme d’hôtes est actuellement contestée par plusieurs auteurs. Le développement de l’infection est conditionné par l’existence de voies de pénétration naturelles (elles sont limitées) ou artificielles (blessures) offertes à l’inoculum ; les conditions climatiques pendant les phases initiales de l’infection et durant l’incubation apparaissent capables de moduler l’intensité de l’infection. La connaissance de l’ensemble de ces données permet une esquisse générale de cycle biologique à partir duquel des stratégies de lutte peuvent être élaborées ; différents exemples sont alors présentés en appui. Diverses perspectives de recherches sur cette espèce bactérienne sont discutées., Pseudomonas syringae, one of the 50 phytopathogenic bacterial species, is the most frequently occurring bacterium under temperate climatic conditions. P syringae has been isolated from = 400 different plant hosts (177 hosts for pv syringae) and is characterized by high genetic, physiological and biological variability expressed through 52 pathovars. Three important properties are associated with this species: pathogenicity, ice nucleation activity and ability for epiphytic survival. The general characteristics of P syringae, apart from the genetic aspects, which have not been examined here, are presented in the first part of this review. In the second part, the 3 remarkable properties of the species have been analyzed. Epiphytic survival, expressed by a capacity to colonize aerial parts of plants or by a significant epiphytic multiplication characterizes most pathovars. High levels of epiphytic populations can be recovered in the spring and also in autumn for perennial plants. These epiphytic populations constitute an inoculum source and are essential to the development of infection. The generally abundant distribution of epiphytic populations is not homogeneous, either between or within plants; their distribution on the surface of the leaf which constitutes their main support is also very heterogeneous. The nature of the interaction between epiphytic bacteria and their host is not yet known, but a relation between host specificity for pathogenicity and host specificity for epiphytic capability within P syringae has been hypothesized. The ice nucleation activity of P syringae, expressed as the ability to induce an early nucleation of water, concerns only some pathovars, mainly the pathovar syringae. It has been shown to be involved in the ice nucleation of plants and frost damage in spring (eg grapevine, tomato, potato) and to be one of the favorable factors in the first stages of some bacterial infections. The pathogenicity of Pseudomonas syringae is dependent on the pathovar except for pv syringae which exhibits a rather wide host range. Some authors have contested this host range for the latter pathovar. The development of infection caused by Pseudomonas syringae is conditioned by the existence of natural (these are limited) or artificial (wounds) means of penetration into the plant tissues and by the inoculum level. Climatic conditions during the first stages of infection or during the incubation period appear to be involved in the modulation of disease intensity. These biological data are useful in understanding the life cycle of P syringae and in elaborating an efficient strategy for disease control; different examples are then presented for illustration. Finally, further developments of research on this phytopathogenic bacterium have been discussed.

Une méthodologie, basée sur l’utilisation de vitroplant de vigne, destinée à estimer la capacité de molécules chimiques à réduire les populations épiphytes de P s pv syringae, a été mise au point; les résultats des essais conduits soit sur des vitroplants axéniques, soit sur des vitroplants transférés en acclimatation et placés en conditions aseptiques ou non, et destinés à comparer l’efficacité de l’oxytétracycline, de la kasugamycine, de la fluméquine, de la bouillie bordelaise et du mancozèbe, concordent avec ceux obtenus sur des plants de vigne conduits en conteneurs. Dans ces essais, la kasugamycine a montré une bonne efficacité vis-à-vis de P s pv syringae; la fluméquine une efficacité moyenne; la bouillie bordelaise une efficacité limitée et variable selon les essais et le mancozèbe aucune efficacité. La technique de criblage de l’efficacité antibactérienne de molécules chimiques peut être développée sur plants en acclimatation en micro-serre, conduits en conditions subaseptiques; elle se situe entre le test réalisé en boîte de Petri (détermination de la concentration minimale inhibitrice) et l’essai de plein champ., Pseudomonas syringae pv syringae occurs as an epiphyte on the surface of grapevine, particularly on buds during spring. As it is also an ice nucleation active bacterium, it is considered as one of the factors causing spring frost damage to the grapevine. Some protection against frost injury could be achieved by reducing the level of P s pv syringae epiphytic populations. The chemicals available for control are limited to some copper formulations and antibiotics, both of which have some disadvantages. More efficient bactericides are required and consequently a great number of chemicals have to be tested. A method using grapevine in vitro plants has been developed to estimate the activity of molecules in reducing the epiphytic populations of P s pv syringae; the results of the experiments performed either on in vitro plants (fig 1) or on plants transferred to acclimatation (figs 2 and 3), and aimed at comparing the efficacy of oxytetracyclin, kasugamycin, flumequine, Bordeaux mixture and mancozebe agree with those obtained on plants grown in containers (fig 4). This method could first be extended to all the strains of the syringae pathovar whatever the host plant and then to the screening of chemicals active against any phytopathogenic bacterium of the genus Pseudomonas or Xanthomonas in so far as it has a significant epiphytic phase within its biological cycle.

National audience; An experiment on sensitivity to spring frost in vine buds was undertaken in the champagne vineyard (Vitis vinifera L Chardonnay) during the springs of 1987-1989 by means of differential thermal analysis (first exotherm) performed in freezing chambers, providing either convective cooling (on vine directly in the field) or radiative cooling on twigs. The sensitivity of buds was analysed in terms of bud phase, surface wetness and ice nuclei bacteria (Pseudomonas syringae) concentration. Figure 1 shows the evolution with phenological phase of mean freezing point of dry buds for 3 yr independently of ice nuclei concentration. Figure 2 reports this freezing point for dry vine buds which contain < 103 bacteria, ie quasi-free of ice nuclei. Figure 3 reports for stage 9 of Eichhhorn and Lorenz the freezing point of dry buds versus ice nuclei concentration. The evolution of the mean freezing point at different stages is plotted on figure 4 versus mean ice nuclei bacterial concentration for both dry and wetted vine buds. From this figure, one can see that frost sensitivity of buds is increased not only by phenology but also by wetness of bud surface. On the contrary, the influence of ice nuclei bacteria is not so obvious. It seems that this bacterial influence could be different depending on the origin of the bacteria. In figure 5 at stage 7, the freezing point of contaminated buds (x) is higher than that of buds which contain the same amount of bacteria (Δ). The only minor effect of natural ice nuclei bacteria concentration on buds was observed on the percentage of surviving buds for a given temperature : the concentration of bacteria observed on killed buds (formula, see attached document) is a little higher in figure 6 than those in surviving buds (+).; Une étude expérimentale de la gélivité des bourgeons de vigne in situ a été entreprise sur le vignoble champenois (Vitis vinifera L Chardonnay) de 1987 à 1989 en utilisant l’analyse thermique différentielle dans des chambres de refroidissement. L’analyse de la gélivité a consisté à étudier l’influence sur la température de rupture de surfusion de facteurs tels que le stade phénologique, le mouillage des organes végétaux et la teneur en bactéries dites glaçogènes. Au vu des résultats il apparaît que stade phénologique et mouillage sont des variables déterminantes, tandis que l’action des bactéries est beaucoup plus ténue et difficile à interpréter. Les informations recueillies doivent permettre à terme d’élaborer un modèle probabiliste du risque de rupture de surfusion dans le bourgeon.

International audience; Effectiveness of Firestop, a slow release formulation of flumequine, was evaluated under controlled conditions (greenhouse, controlled environment chamber) and in experimental orchards, against Erwinia amylovora (fire blight) in pear and apple, and Pseudomonas syringae pv persicae (bacterial dieback) in peach. Firestop (0.3 g ai |-1 for fire blight and 0.1 or 0.15 g ai |- 1 for bacterial dieback) was as efficient as Bordeaux mixture and less efficient than the conventional antibiotics, streptomycin or oxytetracycline. Firestop exhibits no phytotoxic effects, can be applied during the vegetative period and may be recommended in countries where classical antibiotics are not allowed for control of plant disease.; Firestop est une formulation de la fluméquine libérant lentement la matière active. Son efficacité est évaluée en serre et en verger dans le cas du feu bactérien des pommier et poirier (Erwinia amylovora) et du dépérissement bactérien du pêcher (Pseudomonas syringae pv persicae). A 0,3 g. |1 pour le feu bactérien et 0,1 ou 0,15 g. |1 pour le dépérissement bactérien, il s’est révélé tant en serre (tableaux I et II) qu’en champ (tableaux III et IV) aussi bon que la bouillie bordelaise, mais moins efficace que les antibiotiques streptomycine et oxytétracycline. Aucun effet phytotoxique n’étant induit par Firestop, il peut être recommandé, en particulier dans les pays où les antibiotiques ne sont pas autorisés.
