Abstract

We study the hydrodynamic properties of ultraclean interacting two-dimensional Dirac electrons with Keldysh quantum field theory. We study it from a weak-coupling and a strong-coupling perspective. We demonstrate that long-range Coulomb interactions play two independent roles: (i) they provide the inelastic and momentum-conserving scattering mechanism that leads to fast local equilibration; (ii) they facilitate the emergence of collective excitations, for instance plasmons, that contribute to transport properties on equal footing with electrons. Our approach is based on an effective field theory of the collective field coupled to electrons. Within a conserving approximation for the coupled system we derive a set of coupled quantum-kinetic equations. This builds the foundation of the derivation of the Boltzmann equations for the interacting system of electrons and plasmons. From this, we explicitly derive all the conservation laws and identify the extra contributions of energy density and pressure from the plasmons. We demonstrate that plasmons show up in thermo-electric transport properties as well as in quantities that enter the energy-momentum tensor, such as the viscosity. Abstract

In this paper we study thermo-electric transport in interacting two-dimensional Dirac-type systems using a phenomenological Boltzmann approach. We consider a setup that can accommodate electrons, holes, and collective modes. In the first part of the paper we consider the electron-hole hydrodynamics, a model that is popular in the context of graphene, and its transport properties. In a second part, we propose a novel type of hydrodynamics. In that setup, the `fluid' consists of electrons, holes, and plasmons. We study its transport properties, especially the thermo-electric behavior. The results of this part can also be adapted to the study of a fluid consisting of electrons and phonons. Abstract

Background Right‐sided heart failure (HF) due to severe tricuspid regurgitation (TR) is associated with reduced quality of life (QoL). Here, we analyzed the impact of TR on specific QoL dimensions and the effect of transcatheter tricuspid valve intervention (TTVI) on individual QoL items. Methods and Results In this study, we included 174 patients with HF (49% women; median age, 79 years; 97% New York Heart Association ≥3) with baseline QoL assessment undergoing TTVI by transcatheter edge‐to‐edge‐repair at our center between April 2016 and March 2022. QoL was assessed by the standardized Minnesota Living With HF Questionnaire. QoL change after TTVI and correlation to functional end points were analyzed. Abstract

Transcatheter tricuspid valve replacement (TTVR) is an increasingly used treatment technique for patients with severe tricuspid regurgitation (TR). Currently, available data from international registries and randomized controlled trials provide outcome data until a maximum follow-up of 2 years after the procedure. This case report presents 4-year follow-up data for an 84-year-old woman who underwent TTVR for torrential TR in 2019. The patient experienced durable TR reduction, symptomatic improvement, right ventricular reverse remodeling, and substantial improvement in liver and kidney function.

11. Long-Term Outcomes After Edge-to-Edge Repair of Secondary Mitral Regurgitation: 5-Year Results From the EuroSMR Registry.

Abstract

Mitral valve transcatheter edge-to-edge repair (M-TEER) reduces secondary mitral regurgitation (MR) in heart failure and impacts survival in selected patients as demonstrated in the COAPT (Cardiovascular Outcomes Assessment of the MitraClip Percutaneous Therapy for Heart Failure Patients with Functional Mitral Regurgitation) trial. However, long-term outcome data after M-TEER under real-world conditions are lacking. This study sought to assess long-term efficacy and survival after M-TEER in a large real-world registry. We analyzed patients with significant secondary MR undergoing M-TEER from the EuroSMR (European Registry of Transcatheter Repair for Secondary Mitral Regurgitation) registry. Long-term MR reduction, functional outcomes, survival rate, and predictors for all-cause mortality were assessed. In this study, 1,628 patients undergoing M-TEER (mean age 73.8 years, mean EuroSCORE II [European System for Cardiac Operative Risk Evaluation II] 6.9%, 86.6% NYHA functional class ≥III) with available long-term data were included. Five-year survival was 35.0%. Long-term MR reduction (MR grade ≤2+: baseline 4.1%, discharge 92.2%, 5-year follow-up 85.5%; P < 0.001) and functional improvement (NYHA ≤II: baseline 13.4%, 5-year follow-up 60.1%; P < 0.001) was observed. The degree of residual MR was associated with 5-year survival (residual MR grade ≤1+: 38.6%; 2+: 30.5%; ≥3+: 22.6%; P < 0.001). Independent predictors for 5-year all-cause mortality post–M-TEER included age, renal function, residual MR, NYHA functional class, left ventricular ejection-fraction, and COAPT trial eligibility (P < 0.01 for all). This extensive multicenter registry underscores the long-term efficacy of M-TEER in real-world clinical practice and identifies predictors for long-term survival. Abstract

Since transcatheter edge-to-edge repair (TEER) has become a valuable therapy in the treatment of both, mitral (MR) and tricuspid regurgitation (TR), the question of optimized patient selection has gained growing importance. After years of attributing rather little attention to the right ventricle (RV) and its function in the setting of valvular heart failure, this neglect has recently changed. The present review sought to summarize anatomy and function of the RV in a clinical context and aimed at presenting the current knowledge on how the RV influences outcomes after TEER for atrioventricular regurgitation. The anatomy of the RV is determined by its unique shape, which necessitates to use three-dimensional imaging methods for detailed and comprehensive characterization. Complex parameters such as RV to pulmonary artery coupling (RVPAc) have been developed to combine information of RV function and afterload which is primary determined by the pulmonary vasculature and LV filling pressure. Beyond that, TR, which is closely related to RV function also plays an important role in the setting of TEER. While mitral valve transcatheter edge-to-edge repair (M-TEER) leads to reduction of concomitant TR in some patients, the prognostic value of TR in the setting of M-TEER remains unclear. Overall, this review summarizes the current state of knowledge of the outstanding role of RV function and associated TR in the setting of TEER and outlines the unsolved questions associated with right-sided heart failure.

Abstract

Midkine (MK) is a 13-kDa heparin-binding cytokine and growth factor with anti-apoptotic, pro-angiogenic, pro-inflammatory and anti-infective functions, that enable it to partake in a series of physiological and pathophysiological processes. In the past, research revolving around MK has concentrated on its roles in reproduction and development, tissue protection and repair as well as inflammatory and malignant processes. In the recent few years, MK's implication in a wide scope of cardiovascular diseases has been rigorously investigated. Nonetheless, there is still no broadly accepted consensus on whether MK exerts generally detrimental or favorable effects in cardiovascular diseases. The truth probably resides somewhere in-between and depends on the underlying physiological or pathophysiological condition. It is therefore crucial to thoroughly examine and appraise MK's participation in cardiovascular diseases. In this review, we introduce the MK gene and protein, its multiple receptors and signaling pathways along with its expression in the vascular system and its most substantial functions in cardiovascular biology. Further, we recapitulate the current evidence of MK's expression in cardiovascular diseases, addressing the various sources and modes of MK expression. Moreover, we summarize the most significant implications of MK in cardiovascular diseases with particular emphasis on MK's advantageous and injurious functions, highlighting its ample diagnostic and therapeutic potential. Also, we focus on conflicting roles of MK in a number of cardiovascular diseases and try to provide some clarity and guidance to MK's multifaceted roles. In summary, we aim to pave the way for MK-based diagnostics and therapies that could present promising tools in the diagnosis and treatment of cardiovascular diseases.

Abstract

Pancreatic ductal adenocarcinoma (PDAC) presents a formidable clinical challenge due to its aggressiveness and resistance to treatment. Notably, PDAC exhibits a pronounced resistance to various therapeutic modalities, often attributed to a wide metabolic reprogramming involving also the Hexosamine Biosynthetic Pathway (HBP). Transcriptional and bioinformatics analyses, performed under inhibition of HBP in PDAC cell models, indicated that activation of ER stress and Unfolded Protein Response (UPR), due to HBP inhibition, activates an NRF2-dependent antioxidative response, characterized by activation of several genes involved in glutathione (GSH) metabolism. Indeed, inhibition of both HBP and the xCT protein, the cystine/glutamate antiporter involved in GSH synthesis, in PDAC cells significantly reduced cell proliferation, enhanced cell death, altered cellular redox status, heightened sensitivity to oxidative stress, increased dependence on glutamine metabolism, and induced ferroptosis. These findings underscore the potential of HBP inhibition to synergize with ERA, offering a promising therapeutic strategy for managing PDAC.

Abstract

De provincie Overijssel wil maatregelen nemen die bijdragen aan de natuurdoelen voor het beschermde natuurgebied De Wieden. In vier gebieden, 197 hectare in totaliteit worden hydrologische herstelmaatregelen genomen zoals verhoging van het waterpeil en maaiveldverlaging, waarna ze worden ontwikkeld tot natuurgebied. De vier gebieden liggen al in het aangewezen Natura 2000-gebied. Met deze maatregelen komt er meer ruimte voor moerasvogels en de otter. Ook wordt ingezet op de ontwikkeling van blauwgraslanden. Voor het besluit over het provinciaal inpassingsplan (PIP), de ontgrondingenvergunning, de watervergunning en het peilbesluit is een milieueffectrapport opgesteld. De provincie Overijssel heeft de Commissie voor de milieueffectrapportage gevraagd te adviseren over de kwaliteit en volledigheid van het opgestelde MER.

De Veluwe is voor de wespendief als Natura 2000-gebied aangewezen en de populatie staat hier onder druk door meerdere factoren. De provincie wil de omgevingsverordening en de provinciale beleidslijn Windenergie wijzigen. Hiervoor is een milieueffectrapport (MER) opgesteld. De provincie Gelderland heeft de Commissie voor de milieueffectrapportage gevraagd te adviseren over het MER. In dit advies spreekt de Commissie zich uit over de juistheid en de volledigheid van het MER.

We consider the quantum corrections to the conductivity of fermions interacting via a Chern-Simons gauge field, and concentrate on the Hartree-type contributions. The first-order Hartree approximation is only valid in the limit of weak coupling \lambda to the gauge field, and results in an antilocalizing conductivity correction, \sim \lambda^2 g \ln^2 T (g is the conductance). We consider the conductance of a normal-superconductor junction in bilayer graphene in the framework of the Dirac-Bogoliubov-De-Gennes equation. A remarkable suppression of the conductance at voltages just below the superconducting gap is found. This can be understood in terms of the spinor structures of the electron and hole excitations, in particular the reflected valence band hole being orthogonal to the incoming electron at normal incidence. Abstract

We investigate the amplitude of mesoscopic fluctuations of the differential conductance of a metallic wire at arbitrary bias voltage V. For non-interacting electrons, the variance increases with V. The asymptotic large-V behavior is \sim V/V_c (where eV_c=D/L^2 is the Thouless energy), in agreement with the earlier prediction by Larkin and Khmelnitskii. We find, however, that this asymptotics has a very small numerical prefactor and sets in at very large V/V_c only, which strongly complicates its experimental observation. This high-voltage behavior is preceded by a crossover regime, V/V_c \lesssim 30, where the conductance variance increases by a factor \sim 3 as compared to its value in the regime of universal conductance fluctuations (i.e., at V->0). We further analyze the effect of dephasing due to the electron-electron scattering on at high voltages. With the Coulomb interaction taken into account, the amplitude of conductance fluctuations becomes a non-monotonic function of V. Specifically, drops as 1/V for voltages V >> gV_c, where g is the dimensionless conductance. Abstract

We study the amplitude of mesoscopic Aharonov-Bohm oscillations in quasi-one-dimensional (Q1D) diffusive rings. We consider first the low-temperature limit of a fully coherent sample. The variance of oscillation harmonics is calculated as a function of the length of the leads attaching the ring to reservoirs. We further analyze the regime of relatively high temperatures, when the dephasing due to electron-electron interaction suppresses substantially the oscillations. We show that the dephasing length L_phi^AB governing the damping factor exp(-2pi R /L_phi^AB) of the oscillations is parametrically different from the common dephasing length for the Q1D geometry. Abstract

Development and homeostasis of blood vessels critically depend on the regulation of endothelial cell–cell junctions. VE-cadherin (VEcad)-based cell–cell junctions are connected to the actin cytoskeleton and regulated by actin-binding proteins. Coronin 1B (Coro1B) is an actin binding protein that controls actin networks at classical lamellipodia. The role of Coro1B in endothelial cells (ECs) is not fully understood and investigated in this study. Here, we demonstrate that Coro1B is a novel component and regulator of cell–cell junctions in ECs. Immunofluorescence studies show that Coro1B colocalizes with VEcad at cell–cell junctions in monolayers of ECs. Live-cell imaging reveals that Coro1B is recruited to, and operated at actin-driven membrane protrusions at cell–cell junctions. Coro1B is recruited to cell–cell junctions via a mechanism that requires the relaxation of the actomyosin cytoskeleton. By analyzing the Coro1B interactome, we identify integrin-linked kinase (ILK) as new Coro1B-associated protein. Coro1B colocalizes with α-parvin, an interactor of ILK, at the leading edge of lamellipodia protrusions. Functional experiments reveal that depletion of Coro1B causes defects in the actin cytoskeleton and cell–cell junctions. Finally, in matrigel tube network assays, depletion of Coro1B results in reduced network complexity, tube number and tube length. Together, our findings point toward a critical role for Coro1B in the dynamic remodeling of endothelial cell–cell junctions and the assembly of endothelial networks.

Abstract

Right ventricular dysfunction (RVD), as expressed by right ventricular to pulmonary artery coupling, has recently been identified as a strong outcome predictor in patients undergoing mitral valve edge-to-edge repair (M-TEER) for secondary mitral regurgitation (MR). The aim of this study was to define RVD in patients undergoing M-TEER for primary MR (PMR) and to evaluate its impact on procedural MR reduction, symptomatic development and 2-year all-cause mortality.This multicentre study included patients undergoing M-TEER for symptomatic PMR at nine international centres. The study cohort was divided into a derivation (DC) and validation cohort (VC) for calculation and validation of the best discriminatory value for RVD. A total of 648 PMR patients were included in the study. DC and VC were comparable regarding procedural success and outcomes at follow-up. Sensitivity analysis identified RVD as an independent predictor for 2-year mortality in the DC (hazard ratio [HR] 2.37, 95% confidence interval [CI] 1.47-3.81, p 0.001), which was confirmed in the VC (HR 2.06, 95% CI 1.36-3.13, p 0.001). Procedural success (MR ≤2+) and symptomatic improvement at follow-up (New York Heart Association [NYHA] class ≤II) were lower in PMR patients with RVD (MR ≤2+: 82% vs. 93%, p = 0.002; NYHA class ≤II: 57.3% vs. 66.5%, p = 0.09 for with vs. without RVD). In all PMR patients, the presence of RVD significantly impaired 2-year survival after M-TEER (HR 2.23, 95% CI 1.63-3.05, p 0.001).Mitral valve edge-to-edge repair is an effective treatment option for PMR patients. The presence of RVD is associated with less MR reduction, less symptomatic improvement and increased 2-year mortality. Accordingly, RVD might be included into pre-procedural prognostic considerations.

Abstract

Mitral regurgitation (MR) is associated with substantial morbidity and mortality. Within the past 15 years, mitral valve edge-to-edge repair (M-TEER) has developed from an experimental approach to a guideline-recommended, safe, and effective treatment option for patients with severe primary or secondary mitral regurgitation.This review covered relevant publications of M-TEER and summarizes the development of M-TEER devices within the last 15 years. It outlines anatomical challenges which drove the evolution of M-TEER devices, provides an overview about the current state of clinical application and research, and offers an outlook into the future of transcatheter mitral valve treatment.The development and refinement of new M-TEER device generations offer the possibility to treat a wide range of mitral valve anatomies. Choosing the best device for the individual anatomic properties of the patients and considering comorbidities is the key to maximized MR reduction, minimalized complication rates, and thus optimized postinterventional prognosis. Independent from prognostic implications, quality of life has become an important patient-centered outcome that can be improved by M-TEER in virtually all patients treated.

Abstract

Cancer genetics has to date focused on epithelial malignancies, identifying multiple histotype-specific pathways underlying cancer susceptibility. Sarcomas are rare malignancies predominantly derived from embryonic mesoderm. To identify pathways specific to mesenchymal cancers, we performed whole-genome germline sequencing on 1644 sporadic cases and 3205 matched healthy elderly controls. Using an extreme phenotype design, a combined rare-variant burden and ontologic analysis identified two sarcoma-specific pathways involved in mitotic and telomere functions. Variants in centrosome genes are linked to malignant peripheral nerve sheath and gastrointestinal stromal tumors, whereas heritable defects in the shelterin complex link susceptibility to sarcoma, melanoma, and thyroid cancers. These studies indicate a specific role for heritable defects in mitotic and telomere biology in risk of sarcomas.
