Brazil is the second largest ethanol producer in the world and largest using sugarcane feedstock. Bacteria contamination is one of the most important issues faced by ethanol producers that seek to increase production efficiency. Each step of production is a selection event due to the environmental and biological changes that occur. Therefore, we evaluated the influence of the selection arising from the ethanol production process on diversity and composition of bacteria. Our objectives were to test two hypotheses, (1) that species richness will decrease during the production process and (2) that lactic acid bacteria will become dominant with the advance of ethanol production. Bacterial community assemblage was accessed using 16S rRNA gene sequencing from 19 sequential samples. Temperature is of great importance in shaping microbial communities. Species richness increased between the decanter and must steps of the process. Low Simpson index values were recorded at the fermentation step, indicating a high dominance of Lactobacillus. Interactions between Lactobacillus and yeast may be impairing the efficiency of industrial ethanol production. Endogenous starter cultures are used in the production of several cheeses around the world, such as Parmigiano-Reggiano, in Italy, Époisses, in France, and Canastra, in Brazil. These microbial communities are responsible for many of the intrinsic characteristics of each of these cheeses. Bacteriophages are ubiquitous around the world, well known to be involved in the modulation of complex microbiological processes. However, little is known about phage–bacteria growth dynamics in cheese production systems, where phages are normally treated as problems, as the viral infections can negatively affect or even eliminate the starter culture during production. Furthermore, a recent metagenomic based meta-analysis has reported that cheeses contain a high abundance of phage-associated sequences. Here, we analyse the viral and bacterial metagenomes of Canastra cheese, a tradition artisanal cheese produced using an endogenous starter culture. We observe a very high phage diversity level, mostly composed of novel sequences. We detect several metagenomic assembled bacterial genomes at strain level resolution, and several putative phage-bacteria interactions, evidenced by the recovered viral and bacterial genomic signatures. We postulate that at least one bacterial strain detected could be endogenous to the Canastra region, in Brazil, and that its growth seems to be modulated by native phages present in this artisanal production system. This relationship is likely to influence the fermentation dynamics and ultimately the sensorial profile of these cheeses, with implication for all cheeses that employ similar production processes around the world.

Life on Earth is strictly dependent on liquid water. In polar terrestrial environments, water exists in solid state during almost the entire year. Polar microorganisms have not only to adjust their metabolism to survive at subzero temperatures, but also need to cope with extremely dry conditions. We investigated the presence of desiccation-adapted bacteria in Arctic permafrost and Antarctic surface soils and characterized their survivability to dryness. We selected desiccation tolerant cells by treating the soils with chloroform prior to cultivation, in order to mimic the stress of low water activity for long periods. From over 1000 colonies from different samples, 23 unique strains were selected and identified as members of phyla Firmicutes, Proteobacteria and Actinobacteria. About 60% of the strains survived after 50 days in anhydrobiosis. The competence to withstand desiccation varied between close related strains isolated from different locations, bringing the question if environmental conditions may play a role in the observed desiccation tolerance. Survivability was also affected by the solution in which the cells were suspended before drying; R2B medium being more protective than water. This is the first time that chloroform was used to select desiccation tolerant microorganisms from polar soils. The collection of polar microorganisms described herein opens the possibility of further experiments aiming to investigate the resistance mechanisms of polar anhydrobionts. Desiccation tolerance is fundamental to the survivability of microorganisms to the space environment and at the surface of thin-atmosphere planets like Mars. Therefore, the selected strains may open a road to better understand the limits of cold adapted life on Earth and beyond, and compare mechanisms of resistance with anhydrobionts from divergent extreme environments.

Here we investigated the diversity of bacterial communities from deep-sea surface sediments under influence of asphalt seeps at the Sao Paulo Plateau using next-generation sequencing (NGS) method. Sampling was performed at North São Paulo Plateau using the human occupied vehicle Shinkai 6500 and her support vessel Yokosuka. The microbial diversity was studied at two surficial sediment layers (0-1 and 1-4 cm) of five samples collected in cores in water depths ranging from 2,456-2,728 m. Bacterial communities were studied through sequencing of 16S rRNA gene on the Ion Torrent platform and clustered in operational taxonomic units. We observed high diversity of bacterial sediment communities as previously described by other studies. When we considered community composition, the most abundant classes were Alphaprotebacteria (27.7%), Acidimicrobiia (20%), Gammaproteobacteria (11.3%) and Deltaproteobacteria (6.6%). Most abundant OTUs at family level were from two uncultured bacteria from Actinomarinales (5.95%) and Kiloniellaceae (3.17%). The unexpected high abundance of Alphaproteobacteria and Acidimicrobiia in our deep-sea microbial communities may be related to the presence of asphalt seep at North São Paulo Plateau, since these bacterial classes contain bacteria that possess the capability of metabolizing hydrocarbon compounds.

The metacommunity concept studies the processes that structure communities on local and regional scales. This concept is useful to assess spatial variability. However, temporal patterns (e.g., ecological succession and colonization) are neglected in metacommunity studies, since such patterns require temporally extensive, and hard to execute studies. We used experimental habitats in temporary streams located within the Brazilian Cerrado to evaluate the importance of succession for the aquatic insect metacommunity. Five artificial habitats consisting of wrapped crushed rock were set transversally to the water flow in five streams. The habitats were sampled weekly to assess community composition, and replaced after sampling to identify new potential colonizers. We analyzed the accumulation of new colonizers after each week using a logistic model. We selected pairs of experimental habitats and estimated the Bray-Curtis dissimilarity index to assess the community composition trajectory during the experiment. We used the dissimilarity values in ANOVA tests, identifying the importance of time and space for the community. The number of new taxa stabilized in the third week, and we estimated a weekly increase of 1.61 new taxa in the community after stabilization. The overall pattern was a small change on community composition, but one stream had a higher weekly turnover. Our results showed a relevant influence of time in the initial communities of aquatic insects of temporary streams. However, we must observe the temporal pattern in a spatial context, once different streams have different successional history regarding number of taxa and community turnover. We highlight the importance of aerial dispersal and movement to seek oviposition sites as an important factor in determining colonization patterns.

The objective of this work is to perform a case study with data from a survey for two Brazilian SC podcasts (Dragões de Garagem and Fronteiras da Ciência) to evaluate the increase of science podcast media in Brazil, the involved potential, their advantages, shortcomings, and perspectives. We noted an increase of listeners over the years, probably due to the internet popularization and the massive increase of mobile phones. Scientific content is underexplored, despite the great interest of the public. Humorous and informal podcasts are the most appealing to the public and they usually listen to them on informal educational sites. The majority of the public is from the South and Southeast regions, they are young male adults with undergraduate or graduate degrees. SC podcasts, despite their potential to communicate science, still have shortcomings to overcome. Nevertheless, independent initiatives can solve this difficulty, making possible for the media to reach a varied audience, affecting different groups that would not have interest in a specific content before, or even the access itself to the scientific knowledge. Sedimentos de mar profundo são ambientes estáveis e homogêneos, apesar disso, eles apresentam uma grande variedade de habitats disponíveis, possibilitando uma alta diversidade microbiana. A distribuição espacial dos micro-organismos é influenciada por fatores locais e regionais. Os fatores locais são associados à estrutura do ambiente e os fatores regionais, a limitação na dispersão dos micro-organismos que compõem as comunidades e eventos históricos que eventualmente podem modificar o ambiente. Eventos como a liberação de hidrocarbonetos das camadas mais profundas do sedimento para superfície podem alterar os padrões de distribuição espacial das comunidades microbianas, devido o aumento na disponibilidade de carbono e consequentemente selecionando as espécies capazes de degradá-los. Esses eventos são denominados de exsudações de asfalto e foram encontradas na região de estudo. Considerando a falta de conhecimento e a importância dos micro-organismos em sedimentos de mar profundo do oceano Atlântico Sul, o objetivo desse estudo foi compreender os padrões de diversidade microbiana nessas regiões e também investigar como o óleo proveniente da exsudação de asfalto influência as comunidades de micro-organismos no seu entorno. Esse estudo foi realizado na região do Platô de São Paulo que foi dividido em duas regiões, norte e sul. A diversidade microbiana foi estudada em 14 amostras de sedimento de mar profundo, nove amostras na região norte e cinco na sul. A exsudação de asfalto foi encontrada na região norte, influenciando diretamente três das nove amostras. As comunidades foram estudadas através dos métodos de eletroforese em gel de gradiente desnaturante (DGGE), PCR quantitativa (qPCR) e sequenciamento de última geração (Ion Torrent). A distribuição espacial das comunidades foi analisada em diferentes escalas espaciais: verticalmente, variando com a profundidade do sedimento (≤ 4 cm), localmente, em cada uma das regiões amostradas (1-34 Km) e regionalmente, comparando as regiões norte e sul (> 250 Km). O perfil da comunidade obtido com a técnica de DGGE mostrou que as comunidades microbianas foram menos similares entre as regiões e apresentaram relação com a distância geográfica para arqueia e bactéria. Os valores de similaridade foi maior localmente do que regionalmente. A similaridade obtida nas camadas de profundidade analisadas foi alta e não houve relação com a distância geográfica. O número de células entre as camadas de profundidades foi diferente, com tendência de diminuição com o aumento da profundidade. As classes bacterianas mais abundantes foram Alphaproteobacteria (30%), Acidimicrobiia (18%), Gammaproteobacteria (16%), Deltaproteobacteria e Gemmatimonadetes (3%). A composição das comunidades influenciadas pela exsudação de asfalto não teve relação com a presença do óleo ou com as camadas de profundidade. A distância geográfica e a exsudação de asfalto foram importantes fatores para determinação da distribuição geográfica das comunidades microbianas em sedimento marinhos profundos do Platô de São Paulo. Apesar da ausência de relação entre o óleo proveniente da exsudação de asfalto e a composição das comunidades, a alta abundância de Alphaproteobacteria e a importância da distância dentro da região norte são indicativos do aumento da heterogeneidade causado pela exsudação de asfalto. Mais estudos procurando compreender a composição geoquímica dos sedimentos e do óleo são necessários para explicar como esses fatores influenciam a estruturação das comunidades microbianas estudadas. Deep-sea sediments are stable and homogeneous environments, however, they have a high variety of available habitats, allowing a high microbial diversity to occur. Microbial spatial distribution is determined by local and regional factors. Local factors are associated to environment structure and regional factors, to microbial dispersal limitation and historical events that may cause environmental changes. Historical events such as hydrocarbon emanation from sub-seafloor to seafloor may change the patterns of microbial spatial distribution, due to an increase of carbon, thus, selecting species capable to degrade them. These events are denominated as asphalt seep and they were found on the studied region. Considering the lack of knowledge and the importance of microorganisms on deep-sea sediments from South Atlantic ocean, this study aims to understand the patterns of microbial spatial distribution and how the oil from asphalt seep influence the microbial communities. This study was realized in São Paulo Plateau region. The plateau was divided in two regions, north and south. Microbial diversity was studied from 14 deep-sea sediment samples, nine samples from north region and five from south region. The asphalt seep was found in north region from São Paulo Plateau, directly affecting three of the nine samples. The communities were studied through denaturing gradient gel electrophoresis (DGGE), quantitative PCR (qPCR) and next generation sequencing (Ion Torrent). The spatial distribution of the communities was analyzed at different spatial scales: sediment depth (≤ 4 cm), local (1-34 Km) and regional (> 250 Km). Microbial communities were less similar between regions and showing relation with geographic distance to achaea and bacteria. Similarity values within regions were higher then between them, but the geographic distance was also important to both domains, despite samples being closer. Similarity values between sediment depths were high and have no relation with geographic distance. The cell number between sediment depths was different, with tendency to decrease with depth increase. The most abundant classes were Alphaproteobacteria (30%), Acidimicrobiia (18%), Gammaproteobacteria (16%), Deltaproteobacteria e Gemmatimonadetes (3%). The communities composition influenced by asphalt seep have no relation with oil presence and sediment depths. Geographic distance and asphalt seep were important factors to determine the spatial distribution of microbial communities in deep-sea sediments from São Paulo Plateau. Despite the absence of relation between oil from asphalt seep and communities composition, the high abundance of Alphaproteobacteria and the importance of distance within north region are indicative of heterogeneity increase caused by asphalt seep. More studies aiming to understand the geochemical composition from sediments and oil are necessary to explain how these factors influence the communities structure.

Extrasolar Earth and super-Earth planets orbiting within the habitable zone of M dwarf host stars may play a significant role in the discovery of habitable environments beyond Earth. Spectroscopic characterization of these exoplanets with respect to habitability requires the determination of habitability parameters with respect to remote sensing. The habitable zone of dwarf stars is located in close proximity to the host star, such that exoplanets orbiting within this zone will likely be tidally locked. On terrestrial planets with an icy shell, this may produce a liquid water ocean at the substellar point, one particular "Eyeball Earth" state. In this research proposal, HABEBEE: exploring the HABitability of Eyeball-Exo-Earths, we define the parameters necessary to achieve a stable icy Eyeball Earth capable of supporting life. Astronomical and geochemical research will define parameters needed to simulate potentially habitable environments on an icy Eyeball Earth planet. Biological requirements will be based on detailed studies of microbial communities within Earth analog environments. Using the interdisciplinary results of both the physical and biological teams, we will set up a simulation chamber to expose a cold- and UV-tolerant microbial community to the theoretically derived Eyeball Earth climate states, simulating the composition, atmosphere, physical parameters, and stellar irradiation. Combining the results of both studies will enable us to derive observable parameters as well as target decision guidance and feasibility analysis for upcoming astronomical platforms.
