Objetivou-se com o trabalho avaliar a presença de dormência física em sementes de Psidium cattleianum (araçá), obtidas de frutos de diferentes estádios de maturação. Os frutos foram coletados em 15 plantas matrizes no município de Lages-SC e classificados, de acordo com a tabela de cores de Munsell, em vermelho amarelado (5YR5/7), vermelho (10R5/6) e vermelho escuro (10R3/6). Após o beneficiamento, com auxílio de peneira e água corrente, as sementes dos frutos das três colorações foram avaliadas quanto: teor de água, massa seca, primeira contagem de germinação, germinação, plântulas anormais, sementes mortas, primeira contagem de emergência, emergência, dias para iniciar a emergência, índice de velocidade de emergência, curva de embebição em água e embebição em azul de metileno. Os teores de água estavam em 22,02; 23,53 e 23,08% para as sementes das colorações 1, 2 e 3, respectivamente. Não houve diferença significativa entre as sementes dos frutos das três colorações para todas as variáveis analisadas, exceto para o índice de velocidade de emergência, em que sementes de frutos da coloração 2 demonstraram-se mais vigorosas em relação às sementes das demais colorações. O processo de embebição em água foi caracterizado por uma curva trifásica, com início da protrusão radicular em 275, 323, 323 horas, para sementes de frutos de colorações 1, 2 e 3, respectivamente. Observou-se entrada do azul de metileno nas sementes já no primeiro dia, com absorção completa em 371 h (16 dias). Com base nos resultados obtidos, verifica-se que as sementes das três colorações do fruto já estavam na maturidade fisiológica, com resultados semelhantes em relação à massa seca. Conclui-se que sementes maduras de Psidium cattleianum Sabine não possuem dormência física, independente da coloração dos frutos estudados.

A principal forma de propagação de Schinus terebinthifolius é por sementes, contudo pouco se conhece sobre os testes para a avaliação da qualidade fisiológica das sementes dessa espécie. Os objetivos do presente trabalho foram avaliar a influência da temperatura no teste de germinação em sementes de S. terebinthifolius e verificar a eficiência do teste de condutividade elétrica. Sementes de três lotes foram submetidas a teste de germinação nas temperaturas de 20ºC e 25ºC e a testes de vigor: índice de velocidade de germinação, tempo médio de germinação, comprimento de plântula e condutividade elétrica, com leituras após 0, 2, 4, 6, 8, 10, 12, 24, 26 e 28 horas de embebição. Obtiveram-se plântulas mais vigorosas e resultados superiores para o índice de velocidade de germinação, tempo médio de germinação e comprimento de plântulas na temperatura de 25ºC. No teste de condutividade elétrica, após 10 horas de embebição, as leituras foram estabilizadas e permitiram a comparação entre lotes. A temperatura de 25ºC é mais adequada para a condução do teste de germinação de sementes de S. terebinthifolius, e o teste de condutividade elétrica não é recomendado para a análise da qualidade fisiológica das sementes de tal espécie.

SUMMARY: Sequoia sempervirens is a fast-growing, long-lived tree with pest-resistant wood. The objective of this study was to quantify the adventitious rooting ability of sequoia clones in two environments as well as determining rooting behavior during the evaluation period. Mini-cuttings of five clones grown in a clonal mini-garden were planted for rooting in two different environments: a mini-tunnel and an intermittently fogging greenhouse. Weekly assessments (from 30 to 93 days) of the variables were performed: survival, rooted mini-cuttings and number of roots emitted. Rooting speed varied in relation to the clones, occurring mainly between 50 and 60 days after planting, with the appearance of new roots between 70 and 80 days. The cultivation environment promoted distinct responses in the rooting process, being earlier in the greenhouse environment, although with a higher index in the mini-tunnel at the end of the evaluation. The process of differentiation of mini-cuttings occurs initially with the formation of calluses, around 15 days, followed by rooting (between 30 and 60 days) and the subsequent appearance of new roots. The rooting of sequoia mini-cuttings can be done in a mini-tunnel, with good indexes for all clones. It is recommended to keep mini-cuttings in this environment for 90 days for good root formation.

O armazenamento apropriado de sementes florestais consiste como um importante quesito para preservar a sua qualidade fisiológica. Desta forma, objetivou-se realizar a classificação fisiológica de sementes de Zanthoxylum rhoifolium de acordo com sua tolerância à dessecação e ao armazenamento e avaliar diferentes tratamentos para descongelamento. As sementes foram expostas à dessecação até 5% de teor de água e armazenadas em –20 ºC durante três meses. Em seguida, foram submetidas ao descongelamento em: B.O.D. a 25 ºC, estufa a 40 ºC, banho-maria a 50 ºC, e em temperatura ambiente de 18 a 25 ºC. A viabilidade das sementes foi avaliada pelo teste de tetrazólio, em todas as etapas. A viabilidade inicial das sementes foi de 74% e, após secagem, de 80%. As sementes da espécie Zanthoxylum rhoifolium são tolerantes à dessecação até 5% de umidade e ao armazenamento em -20º C, sendo classificadas como ortodoxas. Recomenda-se o descongelamento em B.O.D a 25 ºC. Recomenda-se o descongelamento em B.O.D a 25 ºC.

Gymnanthes klotzschiana is a tree native to the Atlantic Forest Biome. The objective of this study was to analyze the synchronization of five phenophases and their correlation with environmental factors during two years of survey in two distinct areas (Area 1: Mixed Alluvial Ombrophilous Forest; Area 2: Montane Semi-deciduous Seasonal Forest). Ten individuals were randomly selected in each area, with the following variables being studied: leaf fall and budding, flowering, and immature and ripe fruit production. The activity index and the intensity index were determined for each phenological event. Timing and interactions with environmental factors were tested using Spearman’s correlation. The photoperiod correlated with all phenophases and precipitation did not correlate with any. There was synchronization in the foliar fall and foliage in the two areas. The species can be characterized as semi-deciduous. Flowering and fruiting were annual, with the highest intensity of fruit production occurring in Area 1.

There has been an increase in the occurrence of fungal diseases in Ilex paraguariensis A. St.-Hill. (yerba mate) resulting from their domestication. Seed health tests may indicate pathogens prior to sowing, enabling their control. Our study sought to compare the efficacy of fungi detection methods on yerba mate seeds. Seeds from four lots were subjected to the following detection methods: BDA, V8 and filter paper; as well as the performance of asepsis with 1% sodium hypochlorite and 70% alcohol. The fungi found in the lots were: Penicillium spp., Cladosporium spp., Aureobasidium melanogenum, Aspergillus spp., Pestalotia spp., Nigrospora spp., Trichoderma spp., Fusarium graminearum, F. oxysporum and F. guttiforme. The asepsis in the seeds reduced the occurrence of contaminating organisms. There were differences between the evaluated lots regarding fungal incidence. In general, the BDA and V8 culture media showed better results in fungal detection.

The objective of this search was to quantify the genetic variability in progenies from grápia mother trees in natural populations, for initial growth characters of the seedlings. Seeds were collected from 13 mother trees, from four origins, located in the municipalities of Pareci Novo, São José do Sul and Aratiba in RS, and Seara in SC, as well as data from plants and place of origin. Biometrics were performed on 100 seeds lot from each mother tree, and an experiment was installed to evaluate progenies in a completely randomized design. It was evaluated the seedling emergence speed and the emergence percentage, the stem diameter and seedlings height, as well as the final survival percentage. A significative difference was found for all variables observed for seeds and seedlings of grápia mother trees. Seed length and width demonstrated high values for individual heritability in the narrow sense (above 0.6), being possible to use the seed size for an analysis of genetic variability between plants of the species, but not as an indicative variable of physiological quality and vigor. The correlation analysis indicated that the seedling emergence percentage is related to the characteristics of the mother plant plant (height and diameter). Thus, the use of seedlings for forest restoration areas is appropriate, as they have indications of genetic diversity.

The objective of this work was to evaluate the influence of types of drying on seeds' quality, as recalcitrant, Ocotea puberula and determine their degree of critical humidity and lethal degree. Seeds harvested in Brunópolis, SC, with an initial moisture content of 38%, were dried to 18%, with gradients of 2%; using the equation of target weight to ensure that gradient in the stove (35 oC) and in a desiccator with silica gel (25 oC). After drying, the seeds were evaluated for water content, percentage and germination speed index (GSI), tetrazolium and T50. It was observed that up to 32% water content did not change in seed quality, regardless of the type of drying and verified significant loss of germination after this value. We conclude that the type of drying does not affect the quality of the seed; however, because it is a recalcitrant seed, reducing the water content below 32% decreased germination, and its degree of critical humidity and seeds with 22% moisture content had no germination in this study is the degree of lethal humidity of this species. RESUMO Objetivou-se avaliar a influência de tipos de secagem na qualidade de sementes, consideradas recalcitrantes, de Ocotea puberula e determinar o seu grau de umidade crítico e grau letal. Sementes colhidas em Brunópolis, SC, com teor de água inicial de 38%, foram secas até 18%, com gradientes de 2%; utilizando a equação de peso alvo para garantir este gradiente, em estufa (35 oC) e em dessecador com sílica-gel (25 oC). Após cada secagem, as sementes foram avaliadas quanto ao teor de água, porcentagem e velocidade (IVG) de germinação, T50 e tetrazólio. Foi observado que até 32% de teor de água não houve alteração na qualidade das sementes, independente do tipo de secagem, sendo verificada significativa perda de germinação após esse valor. Conclui-se que o tipo de secagem não influencia na qualidade dessas sementes; porém, por tratar-se de uma semente recalcitrante, a redução do teor de água abaixo de 32% prejudica a germinação, sendo seu grau de umidade crítico e sementes com 22% de teor de água não tiveram germinação, sendo neste estudo o grau de umidade letal desta espécie.

Objetivou-se compreender os ciclos reprodutivo e vegetativo de Campomanesia guazumifolia (Cambess.) O. Berg., monitorando os eventos fenológicos e correlacionando com dados climáticos da região de estudo. Foram utilizados dois métodos de avaliação, o Índice de Atividade e o Índice de Intensidade (Índice de Fournier). A fenofase de floração ocorreu entre os meses de outubro a dezembro, e março a abril, com picos de intensidade de Fournier em novembro. A frutificação ocorreu de dezembro a abril, sendo que dezembro foi o mês de maior intensidade de frutos. A espécie apresentou período de maior queda foliar entre março e setembro, com pico de intensidade em agosto, e o brotamento com pico em setembro. Foi observada correlação positiva entre as variáveis climáticas de temperaturas máximas e mínimas mensais e a frutificação, e correlação negativa entre temperaturas máximas e médias mensais e a queda foliar.

Objetivou-se este estudo quantificar os componentes químicos e acompanhar a mobilização de reservas ao longo da germinação de sementes de Psidium cattleianum Sabine (Myrtaceae). Os frutos foram coletados de seis matrizes, localizadas em Lages, SC, e após o beneficiamento, foi determinada, para a semente, a composição química total, utilizando as porcentagens de proteínas, cinzas, lipídeos, carboidratos totais e umidade. Para os estudos relacionados à mobilização de reservas durante a germinação, inicialmente foi realizada a curva de embebição das sementes para a seleção dos pontos a serem utilizados (0, 6, 36, 120, 240 e 528 horas de embebição) com o objetivo de analisar as concentrações de proteína solúvel, açúcar solúvel e lipídeos totais. A composição química total das sementes de P. cattleianum é formada por carboidratos (55,78%), proteínas (13,30%) e lipídios (12,42%). O teor de água das sementes é de 17,49%, e 1,01% das sementes era composto por cinzas. As reservas de proteína solúvel, lipídeo e açúcar solúvel foram utilizadas durante a fase germinativa como fonte de energia e substrato para estruturas celulares. As sementes de P. cattleianum degradaram predominantemente lipídeos durante a germinação.

Informações sobre avaliação da qualidade de sementes dessas espécies não estão bem definidas, principalmente as relacionadas à adequação do teste de germinação. Objetivou-se testar substratos e temperaturas para realização do teste de germinação em sementes de Acca sellowiana (O. Berg) Burret. (goiaba-serrana), Campomanesia xanthocarpa O. Berg (guabiroba), Eugenia involucrata DC. (cereja-do-mato) e Eugenia pyriformis Camb. (uvaia). Sementes, de diferentes procedências, foram submetidas ao teste de germinação em substratos areia e rolo de papel tipo germitest, umedecidos com água destilada, e nas temperaturas 15°C, 25°C, 30°C, 35°C e 20-30°C, sob luz constante, em germinadores tipo BOD. O delineamento experimental utilizado foi o inteiramente casualizado com quatro repetições de 25 sementes/tratamento/lote/espécie, e as médias dos tratamentos foram comparadas pelo teste de Tukey (P < 0,01). O teste de germinação pode ser conduzido na temperatura de 25ºC e em substrato rolo de papel para Eugenia involucrata e Eugenia pyriformis. Ambos os substratos podem ser utilizados para o teste de germinação de Acca sellowiana, a 25ºC. A temperatura de 25ºC e alternância de 20-30ºC são indicadas para Campomanesia xanthocarpa, assim como, os substratos areia e rolo de papel.

Global warming represents one of the most important threats to biodiversity. Because the functional traits of plants influence the performance of individuals under different environmental conditions, we aimed to describe the functional traits of the most frequent tree species found in the Brazilian Mixed Subtropical Forest. We also aimed to test whether functional traits could be used to predict the impact of climate change on the future distribution of tree species. For selected species, certain functional traits were described, namely, wood density, leaf size, specific leaf area, leaf renovation regime, dispersal syndrome, maximum potential height, fruit type, presence of seed dormancy, and the desiccation tolerance of seeds. The geographical distributions of the selected species were modeled using the Maximum Entropy algorithm (MaxEnt) as a function of climatic variation. To predict the impact of climatic changes by 2070 relative to current climate, the best and worst scenarios in relation to the concentration of greenhouse gases (GHG) were used. While the best scenario assumes a strong reduction in GHG anthropogenic emissions, turning negative by 2070, the worst scenario assumes a continuous increase in GHG anthropogenic emissions over time. The functional traits were ordinated by Principal Components Analysis to identify the ecological strategies of each species. The relationship between functional traits and changes to the area of climatic suitability of each species was evaluated using regression trees. Functional traits predicted changes to suitable climatic areas in the worst-case scenario only. The suitable climatic area of more leathery leaf species was predicted to decline by 41.0% on average, whereas that of more membranous leaf species was predicted to decline by 14.0%. In particular, Araucaria angustifolia, which is considered the most representative species of the Mixed Subtropical Forest of Brazil, had the highest sensitivity to climate change. Consequently, our results suggest that this important forest formation will be strongly impacted by climate and will be under severe risk in the future.

Sequoia is a species that provides large volumes of wood, is resistant to decomposition and has seedling production by mini-cutting as a viable alternative. The aim of this work was to evaluate the rooting of mini-cuttings of different clones of Sequoia sempervirens and subsequent use of different concentrations of IBA in clones with low adventitious rooting. Mini-cuttings of different clones were used, from clonal mini-garden. In the experiment with the growth regulator, different concentrations of IBA (from 0 mg L -1 to 3000 mg L -1 ) were used. The material was placed to be rooted in a mini-tunnels and evaluate at 90 days. All experiments were installed in a completely randomized design, with a subsequent evaluation of survival (%), mini-cuttings with calluses (%), mini-cuttings rooted (%) and number of roots (only in the experiment with different concentrations of IBA). In the first experiment, the survival of mini-cuttings was high (close to 95%) and rooting showed great variation among the clones (22.8 to 100%). In the second experiment, using clones that previously presented rooting less than 50%, submitted to IBA application, high survival (88.1 to 99.4%) was observed. The clones presented different responses among themselves, but with improve in the rooting using growth regulator.

Sequoia is a fast-growing, long-living species, producing durable timber. The aim of this study was to test different methods for the vegetative rescue of Sequoia sempervirens trees over 40 years old, made at different periods of the year, and, later, testing the rooting of individualized cuttings in planned arrays. Twenty-four individual sequoias were rescued, applying girdling and semi-girdling at three different heights (-10, 0, and 30 cm). The first collection was made 90 days after application of the treatments, being repeated at 150, 240, and 360 days. The percentage of budding trees and the number of shoots per array were registered. In all collections, shoots produced cuttings, which were placed for rooting in mini-tunnels. Cutting survival (%), rooting (%), and number of roots were registered, per array, and per collection. The species vegetative rescue proved to be efficient for the production of shoots for stem cuttings, especially with girdling at 30 cm and semi-girdling at -10 cm. However, it is difficult to define the best method, mainly because of the genetic effect among stock plants. The rooting of cuttings, of recovered material, presented good results (average >65%), also with great differences among stock plants. The potential for rooting of cuttings varied according to different planting periods, with high rooting rates in all seasons, especially in summer. Sequoia sempervirens shows the potential for vegetative rescue and cloning by rooting of cuttings, and this may lead to new studies, with a view towards fixing clones. RESUMO Sequoia é uma espécie de rápido crescimento, longeva e de madeira durável. O objetivo do estudo foi testar diferentes métodos de resgate vegetativo em árvores adultas de Sequoia sempervirens, realizados em distintas épocas do ano e posterior enraizamento de estacas individualizadas por planta matriz. Resgataram-se 24 indivíduos de sequoia, aplicando-se anelamento e semianelamento em três diferentes alturas (-10, 0 e 30 cm). A primeira coleta foi efetuada 90 dias após aplicação dos tratamentos, sendo repetida aos 150, 240 e 360 dias. Contabilizou-se a porcentagem de árvores com brotações e o número de brotações por matriz. Em todas as coletas as brotações originaram estacas que foram colocadas para enraizar em estufim. No enraizamento das estacas analisou-se a sobrevivência (%), enraizamento (%) e número de raízes por matriz por coleta. O resgate vegetativo da espécie mostrou-se eficiente na produção de brotações para a estaquia, destacando-se o anelamento a 30 cm e o semianelamento a -10 cm. Entretanto, há dificuldade em definir qual o melhor método, principalmente em função do efeito genético entre matrizes. O enraizamento de estacas do material resgatado apresentou bons resultados (média >65%), também com grande diferença entre as matrizes. Diferentes épocas de estaquia alteraram o potencial de enraizamento, contudo, com altos índices de enraizamento em todas as estações do ano, principalmente no verão. Sequoia sempervirens apresenta potencial de resgate vegetativo e clonagem por estaquia, sendo possível conduzir novos trabalhos com vistas a fixação de clones.

RESUMO Acca sellowiana é uma espécie frutífera nativa da Região Sul do Brasil que está despertando grande interesse econômico devido ao alto potencial organoléptico de seus frutos. A principal forma de propagação da espécie é via sexuada. Objetivando determinar a qualidade sanitária das sementes, foram comparadas três formas de detecção de fungos, com ou sem a assepsia superficial, em sementes oriundas de quatro municípios do Estado do Rio Grande do Sul e de três municípios do Estado de Santa Catarina. Foram testados os meios de cultura Batata-Dextrose-Ágar (BDA), V8 (suco de tomate) e o método Blotter Test. Foram identificados os seguintes fungos: Aspergillus niger, Aspergillus flavus, Penicillium sp., Colletotrichum gloeosporioides, Fusarium sp., Curvularia sp., Alternaria sp., Trichoderma sp., Epicoccum sp. e Phomopsis sp. Os meios agarizados são mais sensíveis na detecção de fungos em sementes de goiaba serrana. A assepsia das sementes reduz a incidência de fungos infestantes. ABSTRACT Acca sllowiana (Berg) Burret is a native fruit species in southern Brazil, which is attracting great interest due to the high economic potential of its fruit organoleptic potential; the main form of propagation of the species is sexual way. Aiming to determine the sanitary quality of seeds, three forms of detection of fungi were compared with and without surface sterilization. The seeds were derived from four counties in the state of Rio Grande do Sul and three counties in the state of Santa Catarina. The culture media of Potato Dextrose Agar (PDA), V8 (tomato juice) and the method of “Blotter Test” were tested. The following fungi were identified: Aspergillus niger, Aspergillus flavus, Penicillium sp., Colletotrichum gloeosporioides, Fusarium sp., Curvularia sp., Alternaria sp. Trichoderma sp. Epicoccum spp. and Phomopsis sp. The agarized media are more sensitive in the detection of fungi in Acca sellowiana seeds. The sterilization of seeds reduces the incidence of weed fungi.

Objetivou-se, com este trabalho, verificar a possibilidade do uso de metodologia alternativa para o teste de tetrazólio em sementes de Araucaria angustifolia. Sementes de três lotes foram submetidas à imersão em água por 18 horas e tiveram seus tegumentos e tecido nutritivo removidos ou não. Os embriões foram imersos em diferentes concentrações – 1,0%, 0,5% e 0,1% de solução – a 25°C, por uma hora, além de ser utilizada metodologia já descrita para a espécie. Para comparação dos resultados, foi realizado teste de germinação. É possível utilizar a metodologia alternativa, por meio da imersão das sementes em água por 18 horas, seguida da remoção do tegumento e do megagametófito, e da imersão em solução de tetrazólio 0,1% ou 0,5% a 25°C por uma hora, para a avaliação da qualidade das sementes de A. angustifolia. The objective of this research was to verify the possible use of an alternative methodology for the tetrazolium test in A. angustifolia seeds. Seeds of three lots were submitted to immersion in water for 18 hours. The seeds had their coats and nutritive tissue removed and the embryos were soaked in 1.0%, 0.5% and 0.1% tetrazolium solution at 25 °C for one hour, applying the standard methodology described for the species. Germination tests were performed to verify the reliability of results through the tetrazolium test. Immersion in water for 18 hours followed by the removal of seed coat and nutritive tissue and immersion in tetrazolium solution at 0.1% or 0.5% at 25 °C for 1 hour was an efficient methodology to evaluate the seed viability of A. angustifolia.

Objetivou-se, neste estudo, verificar a influencia de diferentes metodos de secagem na qualidade de sementes de Acca sellowiana e realizar a classificacao fisiologica destas sementes quanto a tolerância a dessecacao e ao armazenamento. Para testar o comportamento das sementes em relacao a diferentes metodos de secagem, foram utilizados tres tratamentos - Controle, Secagem Lenta e Secagem Rapida. Nestes tratamentos, para ambas as secagens, o teor de agua foi de 20% de base umida. Para a classificacao fisiologica, sementes de A. sellowiana foram submetidas a secagem ate 12% e 5% de teor de agua, e ao armazenamento durante 90 dias, em refrigerador a 8 oC. Para sementes recem-colhidas e armazenadas por 30 e 90 dias, o tipo de secagem nao influenciou na qualidade de sementes de A. sellowiana. Em sementes armazenadas por 60 dias, o tratamento controle apresentou germinacao superior aos demais. Sementes de A. sellowiana podem ser fisiologicamente classificadas como intermediarias.

Objetivou-se verificar a eficiência do método de fracionamento em sementes de Eugenia involucrata na formação de plântulas, como forma de maximização de lotes para a produção de mudas. Sementes de E. involucrata foram submetidas aos tratamentos: a) Controle (sementes sem fracionamento), b) Sementes fracionadas transversalmente, com e sem hilo e c) Sementes fracionadas longitudinalmente, com e sem hilo, e, em seguida, à germinação em substrato rolo de papel tipo germitest, sob temperatura de 25 °C e luz branca constante, com quatro repetições de 25 sementes ou frações de sementes. Foram calculados a porcentagem de propágulos com capacidade de produzir plântulas normais (porcentagem de germinação) e a velocidade de germinação (índice de velocidade de germinação - IVG). As sementes sem fracionamento (controle) apresentaram maior porcentagem (72%) e velocidade de germinação (IVG = 0,64). A utilização de fracionamento não é eficiente para maximização de lotes de sementes de E. involucrata, pois a germinação foi diretamente afetada.

RESUMO Objetivou-se verificar a viabilidade de sementes e a integridade do DNA de Ocotea puberula durante armazenamento, com e sem o fruto. Sementes de cinco lotes foram armazenadas com e sem fruto, em câmara fria e seca (UR 40% ± 3% e temp. 10 °C ± 2 °C), pelos períodos de 0, 3, 6 e 9 meses. A cada período foram determinados o teor de água, germinação (%), tetrazólio (%) e integridade do DNA. O teste de germinação foi realizado em papel germitest, em germinador a 30 °C, sob luz constante. O teste de tetrazólio foi realizado com solução na concentração de 0,5%, pelo período de 1 hora. A integridade do DNA das sementes foi analisada em gel de agarose e a concentração de DNA mensurada com espectrômetro NanoDrop®. Foi verificado que somente sementes armazenadas sem o fruto mantiveram sua germinação por até três meses. Após esse período, a germinação das sementes foi reduzindo-se gradativamente, sendo nula aos 9 meses. Na análise da integridade do DNA de sementes foi possível visualizar sua degradação ao longo do armazenamento. Sementes de O. puberula, com ou sem fruto, armazenadas em sacos de papel kraft reduzem sua viabilidade e integridade de DNA após 3 meses de armazenamento em câmara fria e seca. ABSTRACT This study aimed to verify the DNA integrity and viability of Ocotea puberula seeds, with and without fruit, during storage. Seeds of five lots were stored, with and without fruit, in cold and dry chamber (40% ± 3% RH; 10 °C ± 2 °C) for 0, 3, 6, and 9 months. The following attributes were determined at each period: water content, germination potential (%), tetrazolium (%), and DNA integrity. Seed germination was conducted in germitest paper, in germinator at 30 °C under constant light. The tetrazolium test was performed in 0.5% solution for 1 hour. The DNA integrity of seeds was analyzed in agarose gel and DNA concentration was measured using a NanoDrop® spectrometer. The results showed that only seeds without fruit maintained germination for three months; after this period, the germination of seeds reduced gradually, becoming null after nine months. Seed degradation during storage was observed in the DNA integrity analysis. Ocotea puberula seeds, with or without fruit, stored in Kraft paper bags in cold and dry chamber had their viability and DNA integrity reduced after three months.

This research was aimed at studying effects of storage and accelerated aging on germination and profile of storage proteins in Handroanthus albus seeds. These were stored into a cold chamber (± 8 ºC; RH ± 40%) and after periods of 0, 3, 6, 9, and 12 months of storage, were subjected to accelerated aging for 0, 24, 48, 72, and 96 hours. Relationships between germination and proteins profile were assessed. Germination test was performed at 25 ºC, under constant light. For protein extraction, 125 mg of seeds were macerated in 2 mL of extraction buffer (1M Tris-HCl; pH 8.8) and applied to SDS-PAGE polyacrylamide gel at 80 V .15 h-1. Twelve month storage, combined with 72 hours accelerated aging have increased germination in approximately 65% when compared to non-aged seeds or to seeds with 24 h of accelerated aging. Besides beneficial effects, degradation and synthesis of different proteins were observed. It was concluded that germination of Handroanthus albus seeds, when not subjected to accelerated aging, is favored by storage in cold chamber during three to six months, or from nine to 12 months when subjected to accelerated aging process. Storage proteins may be associated to those increases, and hence further studies are needed. Objetivou-se estudar o efeito do armazenamento e do envelhecimento acelerado na germinação e no perfil das proteínas de reserva de sementes de Handroanthus albus. As sementes foram armazenadas em câmara fria e ao 0, 3, 6, 9 e 12 meses submeteu-se ao envelhecimento acelerado por 0, 24, 48, 72 e 96 h, e avaliou-se a germinação e o perfil proteico. O teste de germinação foi realizado sob luz constante a 25 ºC. Para a extração das proteínas, utilizaram-se 125 mg de sementes maceradas com 2 mL do tampão de extração (1M tris HCl pH 8,8) e aplicadas em gel SDS-PAGE (3% e 15%) a 80 V .15 h-1. O armazenamento por 12 meses associado a 72 h de envelhecimento acelerado aumentaram a germinação em aproximadamente 65% em relação às sementes não envelhecidas e envelhecidas por 24 h. Associado a este efeito benéfico, observou-se a degradação e síntese de diferentes proteínas. Conclui-se que a germinação de sementes de Handroanthus albus não envelhecidas artificialmente é favorecida pelo armazenamento em câmara fria por três ou seis meses, e por nove ou 12 meses, associado ao envelhecimento acelerado. As proteínas de reserva podem estar associadas a este acréscimo e necessitam ser mais bem estudadas.

Este trabalho objetivou avaliar a eficiência de tratamentos pré-germinativos na promoção da germinação e crescimento inicial de plântulas de palmiteiro. Os tratamentos avaliados foram: controle, imersão em solução de GA3, exposição ao etileno, imersão em água, imersão em H2SO4, escarificação mecânica, estratificação por 30 dias a 10°C, e escarificação seguida de estratificação. A imersão de sementes em solução de ácido giberélico (GA3; 2000 µL L-1 durante 24 h) ou sua exposição ao etileno (1000 µL L-1 durante 24 h) são eficientes para a promoção da emergência e crescimento inicial de plântulas de palmiteiro.

Objetivou-se neste trabalho descrever as características morfológicas de plântulas durante a germinação de Psidium cattleianum Sabine e Acca sellowiana (O. Berg) Burret. Para a descrição da germinação e das características morfológicas das plântulas, 100 sementes foram colocadas para germinar em substrato papel tipo germitest à temperatura de 25ºC. Foram analisados: tipo, forma e coloração da raiz; pilosidade, forma e coloração do hipocótilo e do epicótilo; consistência, textura, forma, nervação, coloração, tipo de bordo, ápice e base dos cotilédones e protófilos. A germinação de ambas é epígea fanerocotiledonar, formando plântula com sistema radicial axial, raiz principal pivotante longa, com paracotilédones e eofilos simples e opostos. Foram registradas glândulas e células secretoras em todos os órgãos, exceto na raiz, para as duas espécies. A semente de Psidium cattleianum inicia germinação aproximadamente 10 dias após a semeadura quando ocorre a abertura do opérculo e a protrusão radicular. Acca sellowiana, após 28 dias, apresenta hipocótilo longo, cilíndrico, ereto, de coloração marrom-avermelhada, glabro. Na ocorrência de total exposição dos paracotilédones, estes já apresentam coloração verde intensa, demonstrando o potencial fotossintetizante pela plântula em crescimento. A descrição do processo germinativo juntamente com a morfologia de plântulas constitui importante elemento de reconhecimento de espécies. This study describes the morphological characterization during seedling development of Psidium cattleianum Sabine e Acca sellowiana (O. Berg) Burret. To describe germination and seedling morphology, 100 seeds were allowed to germinate on germitest paper as substrate at a temperature of 25 °C. Were analyzed: type, shape and color of the root, hairiness, shape and coloration of hypocotyl and epicotyl, consistency, texture, shape, venation, color, type of board, apex and base of the cotyledons and protophilus. The germination is epigeous phanerocotyledonal for both species, forming seedlings with an axial root system, long pivoting main root with paracotyledons and simple eophylls, opposites. Glands and secreting cells were observed in all organs except in the root system for both species. The germination of the Psidium cattleianum seed begins approximately 10 days after sowing, when the operculum opens followed immediately by the root protrusion. Acca sellowiana shows a long, cylindrical, erect, brownish- red and glabrous hypocotyl after 28 days. When the paracotyledons are totally exposed, they immediately show an intense green color, demonstrating the photosynthetic potential of the seedling growth. The germination process description alongside the morphology of seedlings constitutes an important element for recognition of species.

A espécie Tabebuia serratifolia (ipê-amarelo) é utilizada para reflorestamento de áreas degradadas, além de possuir propriedades medicinais e madeireiras. Sua propagação é por sementes, as quais podem apresentar problemas de conservação, como alterações no seu potencial fisiológico ao longo do armazenamento. Para investigar a ocorrência dessas alterações fisiológicas e bioquímicas de sementes de ipê-amarelo, durante o armazenamento sob condições de câmara fria e ambiente, foram realizadas avaliações trimestrais da germinação, viabilidade, vigor, teor de água, de polifenóis e proteínas resistentes ao calor, em sementes armazenadas por doze meses. Para sementes armazenadas em câmara fria foram observados resultados superiores de germinação e vigor quando comparados com os obtidos na condição ambiente. Ocorreram decréscimos nos teores de polifenóis ao longo do armazenamento, já as proteínas resistentes ao calor mantiveram a mesma intensidade de bandas. Na condição ambiente, a germinação torna-se nula aos nove meses; enquanto que, em câmara fria, as sementes permanecem viáveis durante 12 meses de armazenamento. The species Tabebuia serratifolia (ipê amarelo) is used for reclamation of degraded areas, in addition it possess medicinal and timber properties. Its propagation is by seeds, which present problems of conservation, as changes in the percentage of germination throughout the storage. To investigate the possible changes in some physiological and biochemistry behaviors of "ipê-amarelo" seeds during the storage the germination, viability, vigor, protein content of resistant to heat and polyphenols were evaluated. The seeds were stored under environmental and cold chamber conditions for 12 months. Evaluations were made every 3 months. For seeds stored in cold chamber higher germination and vigor were observed as compared to the environmental condition. There were decreases in the levels of polyphenols during storage, but the heat shock proteins kept the same intensity of bands. In the environmental condition the germination becomes null on the 9th month, while in cold chamber the seeds remain viable during the 12 months of storage.
