Abstract. Climate change is increasing the frequency and intensity of wildfires globally, with significant impacts on society and the environment. However, our understanding of the global distribution of extreme fires remains skewed, primarily influenced by media coverage and regional research concentration. This inaugural State of Wildfires report systematically analyses fire activity worldwide, identifying extreme events from the March 2023–February 2024 fire season. We assess the causes, predictability, and attribution of these events to climate change and land use, and forecast future risks under different climate scenarios. During the 2023–24 fire season, 3.9 million km2 burned globally, slightly below the average of previous seasons, but fire carbon (C) emissions were 16 % above average, totaling 2.4 Pg C. This was driven by record emissions in Canadian boreal forests (over 9 times the average) and dampened by reduced activity in African savannahs. Notable events included record-breaking wildfire extent and emissions in Canada, the largest recorded wildfire in the European Union (Greece), drought-driven fires in western Amazonia and northern parts of South America, and deadly fires in Hawai’i (100 deaths) and Chile (131 deaths). Over 232,000 people were evacuated in Canada alone, highlighting the severity of human impact. Our analyses revealed that multiple drivers were needed to cause areas of extreme fire activity. In Canada and Greece a combination of high fire weather and an abundance of dry fuels increased the probability of fires by 4.5-fold and 1.9–4.1-fold, respectively, whereas fuel load and direct human suppression often modulated areas with anomalous burned area. The fire season in Canada was predictable three months in advance based on the fire weather index, whereas events in Greece and Amazonia had shorter predictability horizons. Formal attribution analyses indicated that the probability of extreme events has increased significantly due to anthropogenic climate change, with a 2.9–3.6-fold increase in likelihood of high fire weather in Canada and a 20.0–28.5-fold increase in Amazonia. By the end of the century, events of similar magnitude are projected to occur 2.22–9.58 times more frequently in Canada under high emission scenarios. Without mitigation, regions like Western Amazonia could see up to a 2.9-fold increase in extreme fire events. For the 2024–25 fire season, seasonal forecasts highlight moderate positive anomalies in fire weather for parts of western Canada and South America, but no clear signal for extreme anomalies is present in the forecast. This report represents our first annual effort to catalogue extreme wildfire events, explain their occurrence, and predict future risks. By consolidating state-of-the-art wildfire science and delivering key insights relevant to policymakers, disaster management services, firefighting agencies, and land managers, we aim to enhance society’s resilience to wildfires and promote advances in preparedness, mitigation, and adaptation.

Intimate partners' sexual desire for each other can fluctuate throughout the course of their relationship, and greater difference between partners' habitual level of sexual desire (i.e., sexual desire discrepancy [SDD]) is linked to lower sexual and relationship satisfaction (Mark, 2015). Although some couples view SDD as a natural and normal part of a sexual relationship, others experience significant distress and may seek therapy. Based on a prominent literature on attachment among couples (Birnbaum & Reis, 2019; Mark et al., 2018; Mikulincer & Shaver, 2007), the current study incorporated an actor-partner interdependence model to examine the dyadic associations between insecure attachment style (i.e., anxious, avoidant) and sexual desire among couples experiencing distressing or non-distressing levels of SDD. Couples (N = 202; 51% female, 48% male, 1% different gender; M = 28 years old, SD = 5.05) were recruited through social media (e.g., Instagram, Twitter) and completed a survey assessing sexual distress, adult attachment style, and sexual desire. Although anxious and avoidant attachment did not moderate an association between SDD and sexual distress, higher levels of avoidant attachment were associated with sexual distress for the individual, but not with their partner's distress. Findings suggest attachment may be essential for understanding individuals' experience of sexual distress when coping with discrepant levels of sexual desire in romantic relationships. The study of droplet break-up in microchannels can be used to improve designs for both biological or chemical microreactors and microfluidic devices e.g., heat exchangers and microchips. In this study, 3D computer simulations were carried out to investigate the detailed behavior of droplet breakup in a T-junction microchannel using the Volume of Fluid (VOF) and Level Set (LS) coupling methods (S-CLSVOF) implemented in OpenFoam. The evolution of droplet morphology and the rupture regimens were analyzed considering pressure results, velocity neck thickness, and tunnel width. The results show four rupture regimes for a range of capillary numbers and dimensionless initial droplet length of 0.005≤Ca≤0.055 and 1≤l0/w≤5. The stages of each rupture were also analyzed and compared with experimental and numerical results from literature. A detailed description of each step in the droplet breakup is also given, including analyses of the influence that the local pressure field and Laplace pressure have on the rupture processes. The 3D results reveal that the Laplace pressure decreases in the main channel due to continuous flux at the top and bottom of the 3D channel. This in turn, leads to a slight change in the rear curvature of the bubble. Laplace pressure is an important parameter that can be used to predict the opening tunnel point, large changes in curvature, and whether the droplet will breakup or not. Just before breakup, the 3D simulations captured a backflow near the neck, a region, characterized by pressure drops, and thus smaller breakup times.

The extension of cognition beyond the brain to the body and beyond the body to the environment is an area of debate in philosophy and the cognitive sciences. Yet, these debates largely overlook olfaction, a sensory modality used by most animals. Here, I use the philosopher's framework to explore the implications of embodiment for olfactory cognition. The philosopher's 4E framework comprises embodied cognition, emerging from a nervous system characterized by its interactions with its body. The necessity of action for perception adds enacted cognition. Cognition is further embedded in the sensory inputs of the individual and is extended beyond the individual to information stored in its physical and social environments. Further, embodiment must fulfill the criterion of mutual manipulability, where an agent's cognitive state is involved in continual, reciprocal influences with its environment. Cognition cannot be understood divorced from evolutionary history, however, and I propose adding evolved, as a fifth term to the 4E framework. We must, therefore, begin at the beginning, with chemosensation, a sensory modality that underlies purposive behavior, from bacteria to humans. The PROUST hypothesis (perceiving and reconstructing odor utility in space and time) describers how olfaction, this ancient scaffold and common denominator of animal cognition, fulfills the criteria of embodied cognition. Olfactory cognition, with its near universal taxonomic distribution as well as the near absence of conscious representation in humans, may offer us the best sensorimotor system for the study of embodiment.

Infidelity is reported at high rates despite strong societal prohibitions against it, leading to questions about whether outcomes support the motives driving infidelity. Little is known about whether motives behind infidelity correspond to perceived outcomes, including regret, but such information might help to explain the paradox of the high rates. Participants were recruited from a large prospective study on monogamy. Analyses were conducted on surveys from the 94 individuals who engaged in infidelity over the year. Using structured and open-ended measures, the authors examined how infidelity evolved, patterns among motives and outcomes, and regret. Infidelity typically began at work or online, lasted about one year, and involved sex as well as feelings of infatuation or love. Most (63.4%) reported not regretting their infidelity. Motives (anger, neglect, dissatisfaction, sex) were compared with outcomes (fulfilled needs, sexual satisfaction, distress) to assess concordance. Being motivated by feelings of neglect or relationship dissatisfaction was associated with needs fulfilled by infidelity; sex as a motive was associated with sexual satisfaction as an outcome. However, concordance in motives and outcomes was unrelated to regret. Sexual assault continues to plague college campuses despite years of targeted intervention. Recent prevention efforts have focused on the role of bystanders in reducing rates. However, limited work has explored actual intervention behaviors as opposed to intentions to intervene or reasons why individuals might choose not to help. College students (N = 1219; aged 19-62; 66% women) completed an online survey assessing experiences as a bystander witnessing a sexual assault, willingness to intervene, helping behavior, and associated beliefs and attitudes (rape myth attitudes, perceived peer norms, perceived self-efficacy). Fifteen percent witnessed a recent situation that they believed involved sexual assault, and most students (69%) intervened. Lower rape myth endorsement, greater self-efficacy regarding intervening, and greater perceived norms that peers would intervene on behalf of a peer predicted willingness to intervene. A personal history of sexual assault and knowing of another's assault increased the odds of perceiving an assault was underway. Knowing the victim increased the odds of intervening for all groups except men. Reports were compared among students who identified as men, women, heterosexuals, and sexual minorities. This research advances our understanding of how common it is to witness sexual assault, who is willing to intervene, and cognitive-affective barriers to intervention among college students. Specific recommendations for practitioners and educators are provided.

This study investigates the crucial role of Carbon Capture and Storage (CCS) technology in mitigating CO2 emissions from Poland’s power systems, which is essential not only for meeting climate targets but also for maintaining energy security in the country. Acknowledging natural gas as a transitional fuel, the focus is on evaluating the decarbonization potential of the natural gas combined cycle (NGCC) power plant. The NGCC with and without an amine-based carbon capture unit was modeled using IPSEpro (SimTech, version 7.0). It was found that the annual CO2 emission from 435.68 MWe (net) NGCC can be reduced from 1,365,501 tons (357.8 kgCO2/MWh) to 136,556 tons (42.9 kgCO2/MWh). On the other hand, the CCS reduced the net electric power of the NGCC from 435.68 MW to 363.47 MW and the net energy efficiency from 55.60% to 46.39%. Nonetheless, these results demonstrate the potential of using the amine-based CO2 capture technology in NGCC systems. This is especially important in the context of the decarbonization of the Polish power system.

A food-hoarding squirrel reshapes its physical environment through storing food. These changes have ramifications for future economic decisions that cascade into social and reproductive consequences. Food-hoarding strategies exist on a continuum from concentrated caches in a defended larder to scattered caches defended using memory and olfaction. These strategies emerge in response to specific physical environments. Because caches are pilfered, the hoarder must also respond to the competitive social environment. Here, we review recent studies, both from captivity and the field, on the socioeconomics and cognition of hoarding in tree squirrels and chipmunks. As ubiquitous inhabitants of an increasingly urbanized world, these studies illuminate the theoretical and applied research potential of the study of such decisions in squirrels.

To make maps from airborne odours requires dynamic respiratory patterns. I propose that this constraint explains the modulation of memory by nasal respiration in mammals, including murine rodents (e.g. laboratory mouse, laboratory rat) and humans. My prior theories of limbic system evolution offer a framework to understand why this occurs. The answer begins with the evolution of nasal respiration in Devonian lobe-finned fishes. This evolutionary innovation led to adaptive radiations in chemosensory systems, including the emergence of the vomeronasal system and a specialization of the main olfactory system for spatial orientation. As mammals continued to radiate into environments hostile to spatial olfaction (air, water), there was a loss of hippocampal structure and function in lineages that evolved sensory modalities adapted to these new environments. Hence the independent evolution of echolocation in bats and toothed whales was accompanied by a loss of hippocampal structure (whales) and an absence of hippocampal theta oscillations during navigation (bats). In conclusion, models of hippocampal function that are divorced from considerations of ecology and evolution fall short of explaining hippocampal diversity across mammals and even hippocampal function in humans. Arboreal animals often leap through complex canopies to travel and avoid predators. Their success at making split-second, potentially life-threatening decisions of biomechanical capability depends on their skillful use of acrobatic maneuvers and learning from past efforts. Here, we found that free-ranging fox squirrels (Sciurus niger) leaping across unfamiliar, simulated branches decided where to launch by balancing a trade-off between gap distance and branch-bending compliance. Squirrels quickly learned to modify impulse generation upon repeated leaps from unfamiliar, compliant beams. A repertoire of agile landing maneuvers enabled targeted leaping without falling. Unanticipated adaptive landing and leaping "parkour" behavior revealed an innovative solution for particularly challenging leaps. Squirrels deciding and learning how to launch and land demonstrates the synergistic roles of biomechanics and cognition in robust gap-crossing strategies.

Abstract Background Disruptions of SETBP1 (SET binding protein 1) on 18q12.3 by heterozygous gene deletion or loss-of-function variants cause SETBP1 disorder. Clinical features are frequently associated with moderate to severe intellectual disability, autistic traits and speech and motor delays. Despite the association of SETBP1 with neurodevelopmental disorders, little is known about its role in brain development. Methods Using CRISPR/Cas9 genome editing technology, we generated a SETBP1 deletion model in human embryonic stem cells (hESCs) and examined the effects of SETBP1-deficiency in neural progenitors (NPCs) and neurons derived from these stem cells using a battery of cellular assays, genome-wide transcriptomic profiling and drug-based phenotypic rescue. Results Neural induction occurred efficiently in all SETBP1 deletion models as indicated by uniform transition into neural rosettes. However, SETBP1-deficient NPCs exhibited an extended proliferative window and a decrease in neurogenesis coupled with a deficiency in their ability to acquire ventral forebrain fate. Genome-wide transcriptome profiling and protein biochemical analysis revealed enhanced activation of Wnt/β-catenin signaling in SETBP1 deleted cells. Crucially, treatment of the SETBP1-deficient NPCs with a small molecule Wnt inhibitor XAV939 restored hyper canonical β-catenin activity and restored both cortical and MGE neuronal differentiation. Limitations The current study is based on analysis of isogenic hESC lines with genome-edited SETBP1 deletion and further studies would benefit from the use of patient-derived iPSC lines that may harbor additional genetic risk that aggravate brain pathology of SETBP1 disorder. Conclusions We identified an important role for SETBP1 in controlling forebrain progenitor expansion and neurogenic differentiation. Our study establishes a novel regulatory link between SETBP1 and Wnt/β-catenin signaling during human cortical neurogenesis and provides mechanistic insights into structural abnormalities and potential therapeutic avenues for SETBP1 disorder.

In the evolving landscape of nephrology and kidney transplants, assessing renal functional reserve (RFR) in living kidney donors is essential for ensuring donor safety and successful transplantation. This study explores the use of the Intra-Parenchymal Renal Resistive Index Variation (IRRIV) test, a novel non-invasive method, to measure RFR in living donors. Our observational study included 11 participants undergoing living kidney donations, evaluated using the IRRIV-based Renal Stress Test (RST) before and 12 months post-nephrectomy. The study demonstrated significant changes in creatinine and eGFR CKD-EPI levels post-donation, with an average creatinine rise from 69 to 97 µmol/L and a reduction in eGFR from 104 to 66 mL/min/1.73 m2. These variations align with the expected halving of nephron mass post-nephrectomy and the consequent recruitment of RFR and hyperfiltration in the remaining nephrons. This pilot study suggests that the IRRIV-based RST is a practical, safe, and reproducible tool, potentially revolutionizing the assessment of RFR in living kidney donors, with implications for broader clinical practice in donor eligibility evaluation, even in borderline renal cases. Furthermore, it confirms the feasibility of RST in living kidney donors and allows us to assess the sample size in 48 donors for a future study.

Despite limited organ availability and post-transplant complications, kidney transplantation remains the optimal treatment for End-Stage Kidney Disease (ESKD). However, innovative dialysis technologies such as portable, wearable, and implantable bioartificial kidney systems are being developed with the aim of addressing these issues and improving patient care. An ideal implantable device could combine bioreactors and blood ultrafiltration to replicate key native cell functions for solute reabsorption, secretion, and endocrinologic activities. Today, the feasibility of an implantable bioreactor for renal cell therapy opens the challenge of developing a fully implantable bioartificial kidney based on silicon nanopore membranes to ensure immunological isolation, cell viability, and the possibility of maintaining a blood substrate for metabolic activities. Current technology is not sufficient to obtain an efficient artificial bioreactor to reach physiological blood purification, which requires a more complex system to produce an ultrafiltrate from the blood that can be processed by cells and eliminated as urine. The number of cells in the bioreactor, endocrine activity, immunological cell isolation, solute and fluid secretion/reabsorption, cell viability, blood and ultrafiltration flow control, and thrombogenicity are fundamental issues that require a new technology that today appears to be a challenge for the design of an implantable artificial kidney. This review aims to analyze the state of the art in this particular field of kidney replacement therapy to highlight the current limitations and possible future technology developments to create implanted and wearable organs capable of treating ESKD with artificial organs that can replicate all native kidneys functions.

Advances in stem cell technologies open up new avenues for modelling development and diseases. The success of these pursuits, however, relies on the use of cells most relevant to those targeted by the disease of interest, for example, midbrain dopaminergic neurons for Parkinson’s disease. In the present study, we report the generation of a human induced pluripotent stem cell (iPSC) line capable of purifying and tracing nascent midbrain dopaminergic progenitors and their differentiated progeny via the expression of a Blue Fluorescent Protein (BFP). This was achieved by CRISPR/Cas9-assisted knock-in of BFP and Cre into the safe harbour locus AAVS1 and an early midbrain dopaminergic lineage marker gene LMX1A, respectively. Immunocytochemical analysis and single-cell RNA sequencing of iPSC-derived neural cultures confirm developmental recapitulation of the human fetal midbrain and high-quality midbrain cells. By modelling Parkinson’s disease-related drug toxicity using 1-Methyl-4-phenylpyridinium (MPP+), we showed a preferential reduction of BFP+ cells, a finding demonstrated independently by cell death assays and single-cell transcriptomic analysis of MPP+ treated neural cultures. Together, these results highlight the importance of disease-relevant cell types in stem cell modelling.

Evidence suggests that there may be racial differences in risk factors associated with the development of Alzheimer’s disease and related dementia (ADRD). We used whole-genome sequencing analysis and identified a novel combination of three pathogenic variants in the heterozygous state (UNC93A: rs7739897 and WDR27: rs61740334; rs3800544) in a Peruvian family with a strong clinical history of ADRD. Notably, the combination of these variants was present in two generations of affected individuals but absent in healthy members of the family. In silico and in vitro studies have provided insights into the pathogenicity of these variants. These studies predict that the loss of function of the mutant UNC93A and WDR27 proteins induced dramatic changes in the global transcriptomic signature of brain cells, including neurons, astrocytes, and especially pericytes and vascular smooth muscle cells, indicating that the combination of these three variants may affect the neurovascular unit. In addition, known key molecular pathways associated with dementia spectrum disorders were enriched in brain cells with low levels of UNC93A and WDR27. Our findings have thus identified a genetic risk factor for familial dementia in a Peruvian family with an Amerindian ancestral background.
