Abstract

NMR is one of the most powerful techniques for the analysis of biological samples in the field of metabolomics. However, the high complexity of fluids, tissues, or other biological materials taken from living organisms is still a challenge for state‐of‐the‐art pulse sequences, thereby limiting the detection, the identification, and the quantification of metabolites. In this context, the resolution enhancement provided by broadband homonuclear decoupling methods, which allows for simplifying 1H multiplet patterns into singlets, has placed this so‐called pure shift technique as a promising approach to perform metabolic profiling with unparalleled level of detail. In recent years, the many advances achieved in the design of pure shift experiments has paved the way to the analysis of a wide range of biological samples with ultra‐high resolution. This review leads the reader from the early days of the main pure shift methods that have been successfully developed over the last decades to address complex samples, to the most recent and promising applications of pure shift NMR to the field of NMR‐based metabolomics. Abstract

International audience; This study explores the potential of profiling a microgram-scale soft tissue biopsy by NMR spectroscopy. The important elements of high resolution and high sensitivity for the spectral data are achieved through a unique probe, HR-μMAS, which allowed comprehensive profiling to be performed on microgram tissue for the first time under MAS conditions. Thorough spatially resolved metabolic maps were acquired across a coronal brain slice of rat C6 gliomas, which rendered the delineation of the tumor lesion. The results present a unique ex vivo NMR possibility to analyze tissue pathology that cannot be fully explored by the conventional approach, HR-MAS and in vivo MRS. Aside from the capability of analyzing a small localized region to track its specific metabolism, it could also offer the possibility to carry out longitudinal investigations on live animals due to the feasibility of minimally invasive tissue excision.

Abstract

International audience; Brain water and some critically important energy metabolites, such as lactate or glucose, are present in both intracellular and extracellular spaces (ICS/ECS) at significant levels. This ubiquitous nature makes diffusion MRI/MRS data sometimes difficult to interpret and model. While it is possible to glean information on the diffusion properties in ICS by measuring the diffusion of purely intracellular endogenous metabolites (such as NAA), the absence of endogenous markers specific to ECS hampers similar analyses in this compartment. In past experiments, exogenous probes have therefore been injected into the brain to assess their apparent diffusion coefficient (ADC) and thus estimate tortuosity in ECS. Here, we use a similar approach in mice by injecting sucrose, a well-known ECS marker, in either the lateral ventricles or directly in the prefrontal cortex. For the first time, we propose a thorough characterization of ECS diffusion properties encompassing (1) short-range restriction by looking at signal attenuation at high b values, (2) tortuosity and long-range restriction by measuring ADC time-dependence at long diffusion times and (3) microscopic anisotropy by performing double diffusion encoding (DDE) measurements. Overall, sucrose diffusion behavior is strikingly different from that of intracellular metabolites. Acquisitions at high b values not only reveal faster sucrose diffusion but also some sensitivity to restriction, suggesting that the diffusion in ECS is not fully Gaussian at high b. The time evolution of the ADC at long diffusion times shows that the tortuosity regime is not reached yet in the case of sucrose, while DDE experiments suggest that it is not trapped in elongated structures. No major difference in sucrose diffusion properties is reported between the two investigated routes of injection and brain regions. These original experimental insights should be useful to better interpret and model the diffusion signal of molecules that are distributed between ICS and ECS compartments.

Abstract

International audience; Analysis of microscopic specimens has emerged as a useful analytical application in metabolomics because of its capacity for characterizing a highly homogenous sample with a specific interest. The undeviating analysis helps to unfold the hidden activities in a bulk specimen and contributes to the understanding of the fundamental metabolisms in life. In NMR spectroscopy, micro($\mu$)-probe technology is well-established and-adopted to the microscopic level of biofluids. However, this is quite the contrary with specimens such as tissue, cell and organism. This is due to the substantial difficulty of developing a sufficient $\mu$-size magic-angle spinning (MAS) probe for sub-milligram specimens with the capability of high-quality data acquisition. It was not until 2012; a $\mu$MAS probe had emerged and shown promises to $\mu$g analysis; since, a continuous advancement has been made striving for the possibility of $\mu$MAS to be an effective NMR spectroscopic analysis. Herein, the mini-review highlights the progress of $\mu$MAS development-from an impossible scenario to an attainable solution-and describes a few demonstrative metabolic profiling studies. The review will also discuss the current challenges in $\mu$MAS NMR analysis and its potential to metabolomics.

Abstract

International audience; NMR has already proven to be a tremendous spectroscopic tool in metabonomics. However, due to its low detection sensitivity, NMR analyses can be challenging especially for mass-limited samples. To overcome this issue, here we introduce a 'new' reliable methodology for profiling $\mu$g/nL scale samples based on the Magic Angle Spinning (MAS) technique-High-Resolution $\mu$MAS (i.e. 1 mm HR-$\mu$MAS). A model study has been proposed: the localized metabolic profiling (i. e. mapping) of a garlic clove. We have been able to investigate 4 different regions (clove skin, flesh, core and inner epidermis), whereas only 3 regions were distinguished using the standard approach, HR-MAS with larger sample mass ($\sim$10 mg). The use of multivariate statistical analysis allowed an initial separation between the different regions by unsupervised PCA. Based on this PCA, a supervised discriminant analysis OPLS was carried out between the previously separated clusters to find the discriminant metabolites. The fact that these micro-detections have been able to offer such precise and accurate results on a model study, opens the way to new applications on heterogeneous biospecimens (such as tissues and cells), on the determination of NMR-based localized metabolic profiles to get the comprehensive fingerprint of their different anatomical regions.

Abstract

International audience; HR-MAS is a valuable NMR spectroscopic tool for metabolic profiling of biospecimens. It offers high resolution NMR spectra for identifying the metabolic compositions in heterogeneous samples such as biopsies and plant tissues. However, the HR-MAS experiments only provide the metabolic profile of an entire sample without any spatial distribution of the metabolites. Combining a pulsed-field gradient (PFG) for spatial encoding with HR-MAS for chemical shift acquisition, Fayon and his team have introduced the possibility of HR-MAS chemical shift imaging (CSI) experiment for acquiring spatial metabolic profiles of a whole organism (i.e. head, body and tail), but the spectral quality is insufficient for an in-depth analysis. Here, we explore the HR-MAS CSI experiments with superior spectral quality on various specimens including biopsy and food tissues, proving the capability of a comprehensive profiling: identification, quantification and multivariate data analysis.

Abstract

International audience; NMR has already proven to be a tremendqus spectroscopic tool in metabolomics. However, due to its low detection sensitivity, NMR analyses can be challenging especially for mass-limited samples. To overcome this issue, here we introduce a 'new' reliable methodology for profiling $\mu$g/nL scale samples based on the Magic Angle Spinning (MAS) technique-High-Resolution lIMAS (HR-$\mu$MAS). HR-$\mu$MAS offers an outstanding opportunity for the detection of metabolites in $\mu$g samples with good sensitivity and high spectral resolution, which coutd widen the existing NMR-based spectroscopy to metabolomic applications and pave the path to its utilization in numerous fields of study. ln comparison to the previous approach with the spinning $\mu$-coil (HR-MACS) [1], HR-$\mu$MAS renders improvements in probe practicality, sample-preparation, data repeatability and in the flexibility of NMR experiments. Moreover, we also show the possibility of performing metabolic quantification using HR-$\mu$ MAS-important analytical component in metabolomics. The excellent NMR spectroscopic capabilities of HR-$\mu$MAS shown here for NMR metabolic profiling on $\mu$g/nL samples have demonstrated that HR-$\mu$MAS could have an impact on future NMR-based metabolomics in biological applications.

24. Acid-free microwave-assisted hydrothermal extraction of pectin and porous cellulose from mango peel waste-towards a zero waste mango biorefinery : Towards a Zero Waste Mango Biorefinery

Abstract

Mango is the second most consumed tropical fruit after banana and the by-products of mango processing (peel, kernel and seed) roughly comprise 35-60% of the total fruit weight, thus representing a potentially high volume resource of exploitable biobased chemicals and materials. Herein, conversion and characterisation of waste mango peels from three different cultivars (Alphonso, Honey and Tommy Atkins) into pectin and porous cellulose using low temperature microwave-assisted acid-free hydrolytic conditions is reported. Microwave-assisted acid-free extraction yielded up to 11.63% (dry weight basis) of pectin which was characterised by IR, NMR (both solution and solid phase) and TGA which showed close similarity to commercial (acid extracted) pectin. The degree of esterification of pectin was determined by 13C NMR (75.6-86.2%) and titrimetry (79.3-87.7%) and the pectin showed excellent gelling ability. The molecular weight as determined by GPC was in the range 14130 (Honey)-25540 (Tommy Atkins). Porosity measurements on the depectinated residue, i.e., residual cellulosic matter showed mesoporous characteristics: average pore diameter, 9.3 nm (Alphonso)-10.5 nm (Honey), however with poor surface area 16.3 m2 g-1 (Honey)-26.0 m2 g-1. Interestingly, a second microwave hydrothermal treatment on these residues retained mesoporosity whilst significantly increasing surface area (88.8 m2 g-1 (Honey)-124.0 m2 g-1 (Alphonso)) and pore volume by approximately six-fold. This is the first detailed combined study of microwave-assisted extraction to yield pectin and mesoporous cellulose towards a potential zero waste mango biorefinery.

Abstract

Localized information on a specimen is considered indispensable for deciphering biological activity. Magnetic resonance spectroscopy is a notable method because of its versatility; however, one limitation is the spectral quality on a static sample. This study explores an amalgamated method with two magnetic resonance experiments: high-resolution magic-angle spinning (HR-MAS) for high-quality spectral acquisition from a spinning sample and chemical shift imaging (CSI) for spatial localization. The advantage of HR-MAS CSI is its amenity for simultaneously profiling the metabolome—with good spectral data—at different spatial regions in a single experiment. Herein, 1H HR-MAS CSI (including a T2-contrast CSI) was described and performed on various food tissues and an intact organism. Different data analyses such as multivariate and quantification were explored to identify the metabolic variants in different anatomical regions and in one case, to assist in a spatial allocation. Abstract

The thermal degradation of olive oil using conventional and microwave heating under the same experimental conditions were compared. A powerful identification and quantification technique based on 31P NMR has been developed to characterise the differences between the minor components including diacylglycerol and free fatty acids in the heated samples. The 31P NMR spectra of the degraded olive oils, which contain OH groups derivatised with a phosphorus reagent, showed that conventional heating is more detrimental to the oil than microwave technique. Conventional heating leads to a significant increase in the diacylglycerol and free fatty acid contents as well as in the number of degradation compounds, which damage the olive oil quality. However, the main process that takes place on using microwave heating is isomerisation between diacylglycerols, a change that could give a potential longer shelf life to the olive oil. Abstract: Commercially available partly acetylated glycerols (mono- and diacetin) are mixtures of glycerol, 1- and 2-acetylglycerol, 1,2- and 1,3-diacetylglycerol, and triacetin. Diacetin and monoacetin are by-products of the biodiesel and triacetin production using glycerol esterification with acetic acid or triglyceride interesterification with methyl acetate. Usually, primary analytical methods involve chromatography (HPLC or GC), spectroscopy (MS or NIR), and wet chemical techniques (potentiometric, iodometric titration) which are often time-consuming due to sample preparation, extended analysis time and/or complicated data analysis. Moreover, these methods require pure mono- and diacetin as standard, which are commercially unavailable. In this work, a complete 31P and 13C chemical shift data for glycerol, mono-, di- and triacetin (including isomers) allows for the identification and quantification of these components in the commercial mixtures. This experimental protocol allows for rapid analysis of mixtures that include these six components. Quantitative 31P NMR and 13C NMR results were validated to those obtained with other analytical methods, such as GC and HPLC-ELSD. 13C NMR is preferred due to allows to measure the content of triacetin, which has no free hydroxyl group, and no signal was detected by 31P NMR. Abstract

Brain water and some critically important energy metabolites, such as lactate or glucose, are present in both intracellular and extracellular spaces (ICS/ECS) at significant levels. This ubiquitous nature makes diffusion MRI/MRS data sometimes difficult to interpret and model. While it is possible to glean information on the diffusion properties in ICS by measuring the diffusion of purely intracellular endogenous metabolites (such as NAA), the absence of endogenous markers specific to ECS hampers similar analyses in this compartment. In past experiments, exogenous probes have therefore been injected into the brain to assess their apparent diffusion coefficient (ADC) and thus estimate tortuosity in ECS. Here, we use a similar approach in mice by injecting sucrose, a well-known ECS marker, in either the lateral ventricles or directly in the prefrontal cortex. For the first time, we propose a thorough characterization of ECS diffusion properties encompassing (1) short-range restriction by looking at signal attenuation at high b values, (2) tortuosity and long-range restriction by measuring ADC time-dependence at long diffusion times and (3) microscopic anisotropy by performing double diffusion encoding (DDE) measurements. Overall, sucrose diffusion behavior is strikingly different from that of intracellular metabolites. Acquisitions at high b values not only reveal faster sucrose diffusion but also some sensitivity to restriction, suggesting that the diffusion in ECS is not fully Gaussian at high b. The time evolution of the ADC at long diffusion times shows that the tortuosity regime is not reached yet in the case of sucrose, while DDE experiments suggest that it is not trapped in elongated structures. No major difference in sucrose diffusion properties is reported between the two investigated routes of injection and brain regions. These original experimental insights should be useful to better interpret and model the diffusion signal of molecules that are distributed between ICS and ECS compartments., (© 2021 The Authors. Abstract

The study of the metabolome within tissues, organisms, cells or biofluids can be carried out by several bioanalytical techniques. Among them, nuclear magnetic resonance (NMR) is one of the principal spectroscopic methods. This is due to a sample rotation technique, high-resolution magic angle spinning (HR-MAS), which targets the analysis of heterogeneous specimens with a bulk sample mass from 5 to 10 mg. Recently, a new approach, high-resolution micro-magic angle spinning (HR-μMAS), has been introduced. It opens, for the first time, the possibility of investigating microscopic specimens (<500 μg) with NMR spectroscopy, strengthening the concept of homogeneous sampling in a heterogeneous specimen. As in all bioanalytical approaches, a clean and reliable sample preparation strategy is a significant component in designing metabolomics (or -omics, in general) studies. The sample preparation for HR-μMAS is consequentially complicated by the μg-scale specimen and has yet to be addressed. This report details the strategies for three specimen types: biofluids, fluid matrices and tissues.
