Nociceptin/orphanin-FQ (N/OFQ) is a recently appreciated critical opioid peptide with key regulatory functions in several central behavioral processes including motivation, stress, feeding, and sleep. The functional relevance of N/OFQ action in the mammalian brain remains unclear due to a lack of high-resolution approaches to detect this neuropeptide with appropriate spatial and temporal resolution. Here we develop and characterize NOPLight, a genetically encoded sensor that sensitively reports changes in endogenous N/OFQ release. We characterized the affinity, pharmacological profile, spectral properties, kinetics, ligand selectivity, and potential interaction with intracellular signal transducers of NOPLight in vitro. Its functionality was established in acute brain slices by exogeneous N/OFQ application and chemogenetic induction of endogenous N/OFQ release from PNOC neurons. In vivo studies with fiber photometry enabled a direct recording of binding by N/OFQ receptor ligands, as well as the detection of natural or chemogenetically-evoked endogenous N/OFQ release within the paranigral ventral tegmental area (pnVTA). In summary, we show that NOPLight can be used to detect N/OFQ opioid peptide signal dynamics in tissue and freely-behaving animals.

Cancer is one of the most devastating diseases that the world is currently facing, accounting for 10 million deaths in 2020 (WHO). In the last two decades, advanced medical imaging has played an ever more important role in the early detection of the disease, as it increases the chances of survival and the potential for full recovery. To date, dynamic glucose-enhanced (DGE) MRI using glucose-based chemical exchange saturation transfer (glucoCEST) has demonstrated the sensitivity to detect both d-glucose and glucose analogs, such as 3-oxy-methyl-d-glucose (3OMG) uptake in tumors. As one of the recent international efforts aiming at pushing the boundaries of translation of the DGE MRI technique into clinical practice, a multidisciplinary team of eight partners came together to form the “glucoCEST Imaging of Neoplastic Tumors (GLINT)” consortium, funded by the Horizon 2020 European Commission. This paper summarizes the progress made to date both by these groups and others in increasing our knowledge of the underlying mechanisms related to this technique as well as translating it into clinical practice.

The integrity of myelinated axons relies on homeostatic support from oligodendrocytes (OLs), which is essential for brain function. However, the mechanisms by which OLs detect axonal spiking and rapidly control axon-OL metabolic coupling are largely unknown. Here, we combine optic nerve electrophysiology and two-photon imaging to study activity-dependent calcium (Ca2+) dynamics in OLs and metabolite fluxes in myelinated axons. Both high-frequency axonal firing and extracellular potassium (K+) elevations trigger a fast Ca2+response in OLs that is facilitated by barium-sensitive, inwardly rectifying K+channels. Using OL-specific Kir4.1 knockout mice (Kir4.1 cKO) we now demonstrate that, in addition to being crucial for K+clearance, oligodendroglial Kir4.1 regulates axonal energy metabolism and long-term axonal integrity. Before the manifestation of axonal damage, we observed reduced glucose transporter GLUT1 and monocarboxylate transporter MCT1 expression in myelin of young Kir4.1 cKO mice, suggesting early deficits in metabolite supply to axons. Strikingly, we found lower resting lactate levels and activity-induced lactate surges in optic nerve axons of young Kir4.1 cKO mice. Moreover, both axonal glucose uptake and consumption were hampered in the absence of oligodendroglial Kir4.1, uncovering a new role of OLs in regulating axonal glucose metabolism. Our findings reveal a novel model of axon-OL signaling and metabolic coupling in which OLs detect high-frequency axonal activity through K+signaling, which is critical in adjusting the axon-OL metabolic unit and in preserving long-term axonal health.

Background: Major retinal degenerative diseases, including age-related macular degeneration, diabetic retinopathy and retinal detachment, are associated with a local decrease in oxygen availability causing the formation of hypoxic areas affecting the photoreceptor (PR) cells. Here, we addressed the underlying pathological mechanisms of PR degeneration by focusing on energy metabolism during chronic activation of hypoxia-inducible factors (HIFs) in rod PR. Methods: We used two-photon laser scanning microscopy (TPLSM) of genetically encoded biosensors delivered by adeno-associated viruses (AAV) to determine lactate and glucose dynamics in PR and inner retinal cells. Retinal layer-specific proteomics, in situ enzymatic assays and immunofluorescence studies were used to analyse mitochondrial metabolism in rod PRs during chronic HIF activation. Results: PRs exhibited remarkably higher glycolytic flux through the hexokinases than neurons of the inner retina. Chronic HIF activation in rods did not cause overt change in glucose dynamics but an increase in lactate production nonetheless. Furthermore, dysregulation of the oxidative phosphorylation pathway (OXPHOS) and tricarboxylic acid (TCA) cycle in rods with an activated hypoxic response decelerated cellular anabolism causing shortening of rod photoreceptor outer segments (OS) before onset of cell degeneration. Interestingly, rods with deficient OXPHOS but an intact TCA cycle did not exhibit these early signs of anabolic dysregulation and showed a slower course of degeneration. Conclusion: Together, these data indicate an exceeding high glycolytic flux in rods and highlight the importance of mitochondrial metabolism and especially of the TCA cycle for PR survival in conditions of increased HIF activity.

Aromatically π-extended porphyrins possess exceptionally intense one-photon (1P) and sometimes two-photon (2P) absorption bands, presenting interest for construction of optical imaging probes and photodynamic agents. Here we investigated how breaking the molecular symmetry affects linear and 2PA properties of π-extended porphyrins. First, we developed the synthesis of porphyrins fused with two phthalimide fragments, termed syn-diarylphthalimidoporphyrins (DAPIP). Secondly, the photophysical properties of H(2), Zn, Pd and Pt DAPIP were measured and compared to those of fully-symmetric tetraarylphthalimidoporphyrins (TAPIP). The data were interpreted using DFT/TDDFT calculations and Sum-Over-States (SOS) formalism. Overall, the picture of 2PA in DAPIP was found to resemble that in centrosymmetric porphyrins, indicating that symmetry breaking, even as significant as by syn-phthalimido-fusion, induces a relatively small perturbation to the porphyrin electronic structure. Collectively, the compact size, versatile synthesis, high 1PA and 2PA cross-sections and bright luminescence makes DAPIP valuable chromophores for construction of imaging probes and other bio-applications.

The one-photon (1P) and two-photon (2P) absorption properties of three quadrupolar dyes, featuring thiophene as a donor and acceptors of varying strengths, are determined by a combination of experimental and computational methods employing the density functional theory (DFT). The emission shifts in different solvents are well reproduced by time-dependent DFT calculations with the linear response and state specific approaches in the framework of the polarizable continuum model. The calculations show that the energies of both 1P- and 2P-active states decrease with an increase of the strength of the acceptor. The 2P absorption cross-sections predicted by the response theory are accounted for by considering just one intermediate state (S1) in the sum-over-states formulation. For the chromophore featuring the stronger acceptor, the energetic positions of the 1P- and 2P-active states prevent the exploitation of the theoretically predicted very high 2P activity due to the competing 1P absorption into the S1 state.

Recent advances in laser technology have made three-photon (3P) microscopy a real possibility, raising interest in the phenomenon of 3P absorption (3PA). Understanding 3PA of organic chromophores is especially important in view of those imaging applications that rely on exogenous probes, whose optical properties can be manipulated and optimized. Here, we present measurements and theoretical analysis of the degenerate 3PA spectra of several phosphorescent metalloporphyrins, which are used in construction of biological oxygen probes. The effective 3PA cross-sections (σ((3))) of these porphyrins near 1700 nm, a new promising biological optical window, were found to be on the order of 1000 GM3 (1 GM3 = 10(−83) cm(6)s(2)), therefore being among the highest values reported to date for organic chromophores. To interpret our data we developed a qualitative four-states model specific for porphyrins and used it in conjunction with quantitative analysis based on the Time-Dependent Density Functional Theory (TDDFT)/a posteriori Tamm-Dancoff Approximation (ATDA)/Sum-Over-States (SOS) formalism. The analysis revealed that B (Soret) state plays a key role in the enhancement of 3PA of porphyrins in the Q band region, while the low-lying two-photon (2P)-allowed gerade states interfere negatively and diminish the 3PA strength. This study features the first systematic examination of 3PA properties of porphyrins, suggesting ways to improve their performance and optimize them for imaging and other biomedical applications.

Herein, we report the first calorimetric study of the protonation of planar and nonplanar free-base porphyrins: H2OETPP (strongly saddled by its substituents), H2T(tBu)P (strongly ruffled by its substituents), and the nominally planar porphyrins (npPs) H2OEP, H2TPP, H2T(nPe)P, and H2T(iPr)P. The observed enthalpies of protonation in solution (ΔHprotsoln) for formation of the dications in 1,1,2,2-tetrachloroethane with 2% trifluoroacetic acid are -45 ± 1 kcal mol-1 for the npPs, -52.0 kcal mol-1 for H2T(tBu)P, and -70.9 kcal mol-1 for H2OETPP. The corresponding enthalpies of protonation (ΔHDFT) obtained from DFT calculations (-27 ± 5, -42, and -63 kcal mol-1, respectively) reproduce this trend. The much more negative enthalpy of protonation seen for H2OETPP is consistent with this molecule being pre-deformed into the saddle structure favored by porphyrin dications. Except for OETPP, the calculated enthalpies of the first protonations (ΔH1) are significantly more positive than the enthalpies of the second protonations (ΔH2). In addition, the structural strain energies for the first protonations (ΔEst(1)) are also significantly more positive than ΔEst(2). According to the calculations, the monocations thus have higher proton affinities than the corresponding free-base porphyrins due to a structural strain effect, which is consistent with the generally elusive nature of the porphyrin monocation. The recent observations of monocations for free-base porphyrins with a high degree of saddling can be rationalized in terms of ΔH1 and ΔH2 being similar; so, the monocation is no longer an unstable intermediate.

Neurochemicals have a large impact on brain states and animal behavior but are notoriously hard to detect accurately in the living brain. Recently developed genetically encoded sensors obtained from engineering a circularly permuted green fluorescent protein into G-protein coupled receptors (GPCR) provided a vital boost to neuroscience, by innovating the way we monitor neural communication. These new probes are becoming widely successful due to their flexible combination with state of the art optogenetic tools and in vivo imaging techniques, mainly fiber photometry and 2-photon microscopy, to dissect dynamic changes in brain chemicals with unprecedented spatial and temporal resolution. Here, we highlight current approaches and challenges as well as novel insights in the process of GPCR sensor development, and discuss possible future directions of the field.

We report the use of microfluidics to functionalize suspended reduced graphene oxide flakes through the addition of aryl radical, generated in situ by reaction between aryl amines and isopentyl nitrite. Microfluidic enabled a tight control of temperature, reaction times and stoichiometric ratios, making it possible to tune the growth of oligomers on the surface of the flakes, which in turn affects the interactions of the functional material with the surrounding environment. The results suggest that shear stress phenomena within the reactor may play a role in the chemistry of graphene materials, by providing a constant driving force towards exfoliation of the layered structures. Scale up of the functionalization process is also reported, along with the grafting of dyes based on squaric acid cores. Photophysical characterization of the dye-modified flakes proves that the microfluidic approach is a viable method towards the development of new materials with tailored properties.

Room temperature phosphorescence is usually a prerogative of metal complexes. Molecules with a hexakis(phenylthio)benzene core constitute a rare example of all-organic chromophores with phosphorescence induced by environmental rigidification. Here we report covalent encapsulation of functionalized persulfurated benzene chromophores into silica nanoparticles as a method of rigidification for induction of phosphorescence. The developed nanoparticles display bright phosphorescence at ambient temperatures and possess high colloidal stability in water. The method permits incorporation of a large number of chromophores (ca. 40) per nanoparticle while preserving their emissivity. The luminescence of the nanoparticles is sensitive to quenching by molecular oxygen in the physiological oxygen range, potentially making them suitable as probes for phosphorescence lifetime imaging of oxygen in biological systems.

Silica nanoparticles (NPs) are versatile nanomaterials, which are safe with respect to biomedical applications, and therefore are highly investigated. The advantages of NPs include their ease of preparation, inexpensive starting materials and the possibility of functionalization or loading with various doping agents. However, the solubility of the doping agent(s) imposes constraints on the choice of the reaction system and hence limits the range of molecules that can be included in the interior of NPs. To overcome this problem, herein, we improved the current state of the art synthetic strategy based on Pluronic F127 by enabling the synthesis in the presence of large amounts of organic solvents. The new method enables the preparation of nanoparticles doped with large amounts of water-insoluble doping agents. To illustrate the applicability of the technology, we successfully incorporated a range of phosphorescent metalloporphyrins into the interior of NPs. The resulting phosphorescent nanoparticles may exhibit potential for biological oxygen sensing.

The synthesis, the structural characterization and the luminescence spectroscopy of new Ln(III) nitrate complexes (Ln = La, Eu, Gd, Tb and Lu) with the quinoline-based N, N’-bis(2-quinolylmethylidene)- 1,2-(R,R+S,S)-cyclohexanediamine, L1 and N, N’-bis(2-quinolylmethyl)- 1,2-(R,R+S,S)-cyclohexanediamine, L2 are presented. All the L1Ln(NO3)3 complexes and L2Ln(NO3)3 with Ln= La, Eu and Tb are isostructural. Metal ions such as La(III), Gd(III) and Lu(III) are able to promote intra-ligand (IL) fluorescence from L1 and L2. Gd(III) and Lu(III) in addition, are also able to stimulate IL phosphorescence at room temperature from the L2 ligand, by means of “paramagnetic effect” and/or “heavy-atom effect”. Both ligands are able to sensitize the typical f-f Eu(III) luminescence and only L2 the one of Tb(III) (green emission). The Eu(III) emission features for both complexes [L1Eu(NO3)3 and L2Eu(NO3)3] are in agreement with their structural properties. All the complexes under investigation reveal promising application in the field of luminescent devices and biomedicine. In particular, excitation at 360 nm, in the case of L1-based complexes, enable the use of glass optics and minimize interfering excitation by chromophores in biological media.

Silicon nanocrystals with an average diameter of 5nm and functionalized with 4,7-di(2-thienyl)-2,1,3-benzothiadiazole chromophores (TBT) and dodecyl chains exhibit near-infrared emission upon one-photon (1P) excitation at 515nm and two-photon (2P) excitation at 960nm. By using TBT chromophores as an antenna, we were able to enhance both 1P and 2P absorption cross-sections of the silicon nanocrystals to more efficiently excite their long-lived luminescence. These results chart a path to 2P-excitable imaging probes with long-lived oxygen-independent luminescence, a rare combination of properties that should allow for a substantial increase in imaging contrast.

We report on the synthesis and characterization of platinum nanoparticles (PtNps) inside the cavities of a PAMAM dendrimer decorated with [Ru(bpy)(3)](2+) units at the periphery. The phosphorescent ruthenium complexes are used as signaling units of the Pt2+ complexation in the dendritic architecture and as photosensitizer units in the photocatalytic production of H-2 from water. This is the first example of water photoreduction in which the catalyst and the sensitizer are anchored on a dendritic molecular scaffold. This study provides a new outlook in the design of new supramolecular systems and materials for developing artificial photosynthesis.

The synthesis of a new class of robust squaraine dyes, colloquially named 1,2-hemisquarimines (1,2-HSQiMs), through the microwave-assisted condensation of aniline derivatives with the 1,2-squaraine core is reported. In CH3CN, 1,2-HSQiMs show a broad absorption band with a high extinction coefficient and a maximum at around λ=530 nm, as well as an emission band centered at about λ=574 nm, that are pH dependent. Protonation of the imine nitrogen causes a redshift of both absorption and emission maxima, with a concomitant increase in the lifetime of the emitting excited state. Encapsulation of the chromophore into a cucurbit[7]uril host revealed fluorescence enhancement and increased photostability in water. The redox characteristics of 1,2-HSQiMs indicate that charge injection into TiO2 is possible; this opens up promising perspectives for their use as photosensitizers for solar energy conversion.

Three new iridium(III) corrole complexes, having symmetrically and asymmetrically substituted corrole frameworks and judiciously varied axial ligands are prepared and characterized by various spectroscopic techniques including the X-ray structures of two of them. The observed phosphorescence at ambient temperature appears at much longer wavelengths than the previously reported Ir(III) porphyrin/corrole derivatives. Efficiencies of these compounds in the generation of singlet oxygen are also studied for the first time.

Orexins (also called hypocretins) are hypothalamic neuropeptides that carry out essential functions in the central nervous system; however, little is known about their release and range of action in vivo owing to the limited resolution of current detection technologies. Here we developed a genetically encoded orexin sensor (OxLight1) based on the engineering of circularly permutated green fluorescent protein into the human type-2 orexin receptor. In mice OxLight1 detects optogenetically evoked release of endogenous orexins in vivo with high sensitivity. Photometry recordings of OxLight1 in mice show rapid orexin release associated with spontaneous running behavior, acute stress and sleep-to-wake transitions in different brain areas. Moreover, two-photon imaging of OxLight1 reveals orexin release in layer 2/3 of the mouse somatosensory cortex during emergence from anesthesia. Thus, OxLight1 enables sensitive and direct optical detection of orexin neuropeptides with high spatiotemporal resolution in living animals., Nature Methods, 19 (2), ISSN:1548-7105, ISSN:1548-7091
