Two binary aqueous mixtures containing small amphiphilic molecules TMAO (trimethylamine-N-oxide) and TBA (tert-butyl alcohol) are investigated. TMAO is osmolyte while TBA is a monohydrate alcohol and both possess bulky hydrophobic groups and polar heads namely, NO, in TMAO and OH in TBA.

7. Longitudinal genetic characterization of circulating tumor cells in metastatic breast cancer patients

Abstract

Background: Little is known about the evolution of genetic aberrations during metastatic cancer progression and in response to systemic treatment. Obtaining repeated tissue biopsies is often impractical. On the other hand, it has been shown that circulating tumor cells (CTCs) can be easily followed during disease course and genetic characterization at the single cell level is possible with high reliability Methods: Individual CTCs of a ER+ and HER2- de novo metastatic breast cancer patient treated with weekly paclitaxel/gemcitabine as first line therapy, were collected at three different time points (before start, after one and two cycles of treatment). Whole peripheral blood was enriched using the CellSearch® system and CTCs were sorted by DEPArrayTM device. The whole genome of single CTCs was amplified with Ampli1TM WGA kit and WGA quality was assessed by Ampli1TM QC Kit. Genome wide single cell copy number variation (CNV) profile was evaluated with Agilent SurePrint 180k array comparative genomic hybridization (aCGH). Results: CTCs count at CellSearch was 22, 75 and 15 at three time points respectively. A total of 25 single CTCs were collected and 23 (92%) showed high Genome Integrity Index (GII) as measured by Ampli1TM QC kit (GII = 3 or 4). For each time point multiple CTCs (3, 6 and 3 respectively) were selected for single cell aCGH analysis. The genomic profile was strikingly similar (1q gain, 12p, 13p, 16q and 17p losses) across individual cells of the same blood sample and throughout different time points evaluated. After 3 cycles of therapy a disease progression was documented by CAT-scan. Discussion: The observed high GII, low genetic heterogeneity and stable genome across different time points suggests the presence of an aggressive clone resistant to the treatment and cancer-associated genes analysis for sequence variants by NGS targeted sequencing is ongoing. Further patients with longitudinal follow-up will be enrolled in order to evaluate if the heterogeneity between aCGH profile at a given time point and longitudinal evolution of aCGH profiles can be associated with early treatment response. Results will be presented at the conference.

Abstract

Context: NF-kappa B is a family of transcription factors involved in cell proliferation, differentiation, and apoptosis. Objective: We have recently demonstrated that NF-kappa B is expressed in the growth plate and it mediates the growth-promoting effects of IGF-I on chondrogenesis and longitudinal bone growth. Humans with defects of the NF-kappa B pathway exhibit growth failure, which suggests a possible regulatory role for NF-kappa B in statural growth. We have previously reported a child with ectodermal dysplasia, immunodeficiency, and growth retardation, harboring a heterozygous mutation of I kappa B alpha, an essential component of the NF-kappa B pathway. Since he was found with low IGF-I and IGFBP-3, and elevated GH secretion, an IGF-I generation test was carried out: baseline IGF-I was low and only responded to a high dose of GH. Thus, the diagnosis of GH resistance was made. Results: To assess the underlying mechanisms of his GH resistance, we cultured the patient's skin fibroblasts with GH and/or IGF-I. While both GH and IGF-I induced cell proliferation and NF-kappa B activity in controls' fibroblasts, they had no effect on the patient's fibroblasts. In the fibroblasts of the patient's father (who displays mosaicism for the I kappa B alpha mutation), GH and IGF-I elicited an attenuated stimulatory effect. In addition, GH stimulated STAT5 phosphorylation and IGF-I mRNA expression in controls' and the father's fibroblasts, while IGF-I induced PI3K activity and mRNA and protein expression of TDAG51, a target gene for IGF-I. In contrast, none of these effects was elicited by GH or IGF-I in the patient's fibroblasts. Conclusion: Our findings suggest that this patient's I kappa B alpha mutation caused GH and IGF-I resistance which, in turn, contributed to his growth failure. 39. Molecular epidemiological survey on the vectors of <e1>Thelazia gulosa</e1>, <e1>Thelazia rhodesi</e1> and <e1>Thelazia skrjabini</e1> (Spirurida: Thelaziidae)

Abstract

A Polymerase Chain Reaction (PCR)- based assay developed for the specific identification of Thelazia gulosa, Thelazia rhodesi and Thelazia skrjabini (Nematoda, Spirurida), which cause bovine ocular thelaziosis, was evaluated for its usefulness in detecting the intermediate hosts and in estimating the infection prevalence of vectors in field conditions throughout 5 years (from 1997 to 2001). A total of 5190 flies were captured and identified as Musca larvipara, Musca osiris, Musca autumnalis, Musca tempestiva or Musca domestica. Genomic DNA was extracted from pools constituted by heads, thoraces, abdomens and wings of 10 flies of each species, and 2076 samples were subjected to a PCR assay to specifically detect the ribosomal ITS-1 sequence of bovine Thelazia. Amplicons were sequenced and subjected to digestion with CpoI restriction enzyme. M. autumnalis, M. larvipara, M. osiris and M. domestica species were shown to be PCR positive. T. gulosa was specifically detected by PCR in M. autumnalis, M. larvipara, M. osiris and M. domestica, whereas T. rhodesi is in M. autumnalis and M. larvipara. Of 27 positive samples, 23 were positive for T. gulosa and 4 for T. rhodesi, with a mean prevalence of 2·86% in the whole fly population collected. The highest mean prevalence values of infection were detected in M. autumnalis (4·46%) and M. larvipara (3·21%), and the former species was confirmed to be the vector of T. gulosa and T. rhodesi. This study is the first report of M. osiris as a vector of T. gulosa and M. larvipara as a vector of T. gulosa and T. rhodesi under natural conditions. The occurrence of Thelazia in fly populations in the Apulia region of Italy (in the 5 grazing seasons considered) indicates that cattle thelaziosis is enzootic in southern Italy. This molecular assay should be a useful epidemiological tool for assessing the role of different species of flies as intermediate hosts of thelaziae.

40. Polymorphic Behavior of Copolymers of Syndiotactic Polystyrene with m-Methylstyrene

Abstract

The influence of the presence of different amounts m-methylstyrene (m-MS), in the composition range 4−21 mol %, on the polymorphic behavior of syndiotactic polystyrene (s-PS) is presented. A comparison with the effect of the presence of p-methylstyrene is also presented. The behavior of copolymers subjected to solvent extraction or annealing procedures indicates that the presence of m-MS stabilizes the γ form of s-PS, which remains stable in a large temperature range, up to 210−220 °C. The helical mesoporous δ form of s-PS has not been observed in copolymers with m-MS. The presence of low contents of m-MS induces crystallization of the β form from polymer solution (by casting at high temperatures), whereas high concentrations of m-MS tend to stabilize the γ form. The copolymer samples crystallize from the melt in α and β forms depending on the concentration of m-MS, which stabilizes the β form for composition around 11 mol %, and induces crystallization in the α form for lower (4 mol %) and higher (21 mol %) concentration. Moreover, even when the memory of the α form is maintained in the melt, the presence of m-MS in a concentration around 11 mol % induces the crystallization of the β form, whereas, even though no memory of the α form is present in the melt, samples with 4 and 21 mol % of m-MS crystallize in the α form. The α form obtained in these copolymers, by melt-crystallization or annealing of the γ form, does not correspond to neither the disordered α‘, nor the ordered α‘ ‘ modifications observed in s-PS. The presence of m-MS comonomeric units induces structural variation in the crystal packing of the trans-planar chains, and a new crystalline form is obtained.

Abstract

We employed dynamic light scattering, radiowave dielectric spectroscopy and circular dichroism measurements to reveal the structural features of complexes formed in the interaction between DNA and cationic liposomes. Two distinct types of complexes form in these systems, as the charge ratio increases. They are: (i) DNA-coated liposomes, where the DNA binds electrostatically to the outside of the cationic lipid vesicle, (ii) multilamellar complexes followed by rupture and reorganization of the bilayer structure or clusters of DNA-coated vesicles with alternating sheets of DNA and lipid bilayers. The knowledge of the complex morphology occurring in lipid-DNA lipoplex formation could provide insight for the development of efficient artificial delivery systems in genetic engineering and gene therapy.

Abstract

The C6 methylene protons were selectively detected in 1H-NMR spectra of intact glioma cells incubated with [6-13C]-D-2-deoxyglucose ([6-13C]-2dG), a 13C-enriched glucose analog that is suitable for monitoring glucose utilization in brain tumors. Spectral editing via 1H–13C scalar coupling was performed with twin spin-echo double resonance (T-SEDOR), a pulse sequence which combines chemical specificity and high sensitivity, requires no solvent pre-saturation, and can easily be adapted to imaging protocols. This work demonstrates the suitability of the pulse sequence for monitoring [6-13C]-2dG uptake in living cells in vitro, in spite of line-broadening and the occurrence of other strong signals in the spectral region of interest (3.5–4.4 ppm). 46. Cluster Organization of Glycosphingolipid GD1a in Lipid Bilayer Membranes:  A Dielectric and Conductometric Study

Abstract

The dielectric and conductometric properties of mixed ganglioside/phospholipid vesicle aqueous suspensions were measured in the frequency range from 10 kHz to 10 MHz, where a pronounced dielectric dispersion due to surface bilayer polarization occurs. The concentration of the ganglioside employed, a disialoglycosphingolipid GD1a, was varied from 50 to 500 μg/mL (corresponding to a molar ratio [GD1a]/[DPPC] from 0.2 × 10-3 to 2 × 10-3) and the excess surface polarizability, attributed to ganglioside organization within the lipid bilayer, was evaluated. The dependence of the ganglioside concentration shows that a clusterization process occurs, giving rise to the formation of large in-phase aggregates whose coordination number, of the order of a few hundred dipoles, depends on temperature. The relevance, from a biological point of view, of a nonuniform distribution of GD1a in a lipid bilayer membrane is briefly discussed.
