Abstract

Surgery remains to be the mainstay of treatment for hepatocellular carcinoma (HCC). Nonetheless, its therapeutic efficacy is significantly impaired by postoperative recurrence, which occurs in more than half of cases as a result of intrahepatic metastasis or de novo tumorigenesis. For decades, most therapeutic strategies on inhibiting postoperative HCC recurrence have been focused on the residual tumor cells but satisfying therapeutic outcomes are barely observed in the clinic. In recent years, a better understanding of tumor biology allows us to shift our focus from tumor cells toward the postoperative tumor microenvironment (TME), which is gradually identified to play a pivotal role in tumor recurrence. In this review, we describe various surgical stress and surgical perturbation on postoperative TME. Besides, we discuss how such alternations in TME give rise to postoperative recurrence of HCC. Based on its clinical significance, we additionally highlight the potential of the postoperative TME as a target for postoperative adjuvant therapeutics. [ABSTRACT FROM AUTHOR]

20. Real-Time Transfer Case 4WD Control Development to Improve Vehicle Coasting Stability

Abstract

The real-time transfer case installed on the four-wheel drive vehicle with a longitudinally mounted engine, can distribute not only the engine drive torque, but also the engine drag torque between the front and rear axles, to improve vehicle traction and stability performance. When under vehicle coasting condition, the engine drag torque may cause vehicle rear wheel slippage on low friction adhesion road, which causes a vehicle to be unstable and spin in. Although the current engine Drag Torque Control technology has the ability to reduce rear wheel longitudinal slippage by engine torque control, but the control effect is poor, the wheel slip ratio is large and the slipping duration is long, the vehicle still has the tendency to spin in. This paper proposed a new transfer case coasting control strategy to avoid this problem. Through the pre-load control based on vehicle coasting characteristics and closed-loop feedback control based on rear axle slip state, the rapid dynamic distribution of engine drag torque between the front and rear axle is realized to avoid single axle slippage and improve vehicle coasting stability. Through test vehicle verification, the proposed control strategy could significantly improve vehicle coasting deceleration and stability performance.

41. Dynamic studies of guest molecules in Metal-Organic Frameworks using Solid-State NMR

Abstract

Metal-organic frameworks (MOFs) are a new class of porous materials that possess large three-dimensional voids in their structures. In this work, Solid-state NMR (SSNMR) is used to examine the dynamics of guest molecules at various temperatures in MOFs that possess different types of channels. Chapter 2 introduces the 13CO2 adsorption behavior in α-Mg formate studied by 13C SSNMR in conjunction with molecular dynamic simulation. 1H-13C cross polarization technique has successfully determined the adsorption sites of CO2 in this type of MOF to be hydrogen atoms. The dynamic analysis detects that at low temperature, all of the adsorbed CO2 undergo a combined motion consisting of a localized rotation upon a single hydrogen site and a non-localized two-sites hopping between two hydrogen sites, whereas at high temperature, a small portion of the adsorbed CO2 molecules obey only a localized rotation while the majority still follow the combined motion. Chapter 3 studies the ethylene adsorption behavior in α-Mg formate and CPO-27-M (M = Mg and Zn) by means of 2H SSNMR. Two types of ethylene are found in α-Mg formate following distinct motions. The majority of the adsorbed ethylene undergoes a combined motion of localized rotation and non-localized two-sites hopping, whereas the minority follows a localized rotation only. In CPO-27-M, all the adsorbed ethylene follows a combined motion of a localized rotation upon the unsaturated metal sites and a simultaneous non-localized hopping between six unsaturated metal sites. The affinity of ethylene towards CPO-27-Mg is found to be stronger than in CPO-27-Zn.

45. Sizable dynamics in small pores: CO2 location and motion in the α-Mg formate metal–organic framework.

Abstract

Metal–organic frameworks (MOFs) are promising materials for carbon dioxide (CO2) adsorption and storage; however, many details regarding CO2 dynamics and specific adsorption site locations within MOFs remain unknown, restricting the practical uses of MOFs for CO2 capture. The intriguing α-magnesium formate (α-Mg3(HCOO)6) MOF can adsorb CO2 and features a small pore size. Using an intertwined approach of 13C solid-state NMR (SSNMR) spectroscopy, 1H–13C cross-polarization SSNMR, and computational molecular dynamics (MD) simulations, new physical insights and a rich variety of information have been uncovered regarding CO2 adsorption in this MOF, including the surprising suggestion that CO2 motion is restricted at elevated temperatures. Guest CO2 molecules undergo a combined localized rotational wobbling and non-localized twofold jumping between adsorption sites. MD simulations and SSNMR experiments accurately locate the CO2 adsorption sites; the mechanism behind CO2 adsorption is the distant interaction between the hydrogen atom of the MOF formate linker and a guest CO2 oxygen atom, which are ca. 3.2 Å apart. Abstract

Record greenhouse gas emissions have spurred the search for clean energy sources such as hydrogen (H2) fuel cells. Metal–organic frameworks (MOFs) are promising H2 adsorption and storage media, but knowledge of H2 dynamics and adsorption strengths in these materials is lacking. Variable-temperature (VT) 2H solid-state NMR (SSNMR) experiments targeting 2H2 gas (i.e., D2) shed light on D2 adsorption and dynamics within six representative MOFs: UiO-66, M-MOF-74 (M = Zn, Mg, Ni), and α-M3(COOH)6 (M = Mg, Zn). D2 binding is relatively strong in Mg-MOF-74, Ni-MOF-74, α-Mg3(COOH)6, and α-Zn3(COOH)6, giving rise to broad 2H SSNMR powder patterns. In contrast, D2 adsorption is weaker in UiO-66 and Zn-MOF-74, as evidenced by the narrow 2H resonances that correspond to rapid reorientation of the D2 molecules. Employing 2H SSNMR experiments in this fashion holds great promise for the correlation of MOF structural features and functional groups/metal centers to H2 dynamics and host–guest interactions. Abstract

As regulators in gene expression, microRNAs take part in most biological processes including cell differentiation, apoptosis, cell cycle, and epithelial-to-mesenchymal transition (EMT). In order to evaluate their roles in EMT process, microRNA expression profile changes induced by EGF or TGF-β treatment on nasopharyngeal carcinoma cell HK-1 were analyzed, and miR-21, miR-148a, miR-505, and miR-1207-5p were found to be upregulated in growth factors-induced EMT process. miR-21 is already known as an oncogenic miRNA to promote metastasis, however, the exact functions of other three miRNAs in EMT are unclear. To our surprise, we found that miR-148a, miR-505, and miR-1207-5p can suppress EMT and metastasis phenotypes in HK-1 cells both in vitro and in vivo, which may relate to their inhibition on EMT and Wnt signaling molecules. MiRNAs confer robustness to biological processes by posttranscriptional repression of key transcriptional programs that are related to previous developmental stages or to alternative cell fates. Our findings indicate that miRNA feedback circuit is tuned to respond to growth factors-induced EMT, and we suggested a new negative feedback loop which may be an important element of the EMT process and confer biological robustness.
