Abstract

The initial rostrocaudal patterning of the neural tube leads to differential expression of Hox genes that contribute to the specification of motor neuron (MN) subtype identity. Although several 3' Hox mRNAs are expressed in progenitors in a noisy manner, these Hox proteins are not expressed in the progenitors and only become detectable in postmitotic MNs. MicroRNA biogenesis impairment leads to precocious expression and propagates the noise of Hoxa5 at the protein level, resulting in an imprecise Hoxa5-Hoxc8 boundary. Here we uncover, using in silico simulation, two feed-forward Hox-miRNA loops accounting for the precocious and noisy Hoxa5 expression, as well as an ill-defined boundary phenotype in Dicer mutants. Finally, we identify mir-27 as a major regulator coordinating the temporal delay and spatial boundary of Hox protein expression. Our results provide a novel trans Hox-miRNA circuit filtering transcription noise and controlling the timing of protein expression to confer robust individual MN identity.

Abstract

The X-linked disease Barth syndrome (BTHS) is caused by mutations in TAZ; TAZ is the main determinant of the final acyl chain composition of the mitochondrial-specific phospholipid, cardiolipin. To date, a detailed characterization of endogenous TAZ has only been performed in yeast. Further, why a given BTHS-associated missense mutation impairs TAZ function has only been determined in a yeast model of this human disease. Presently, the detailed characterization of yeast tafazzin harboring individual BTHS mutations at evolutionarily conserved residues has identified seven distinct loss-of-function mechanisms caused by patient-associated missense alleles. However, whether the biochemical consequences associated with individual mutations also occur in the context of human TAZ in a validated mammalian model has not been demonstrated. Here, utilizing newly established monoclonal antibodies capable of detecting endogenous TAZ, we demonstrate that mammalian TAZ, like its yeast counterpart, is localized to the mitochondrion where it adopts an extremely protease-resistant fold, associates non-integrally with intermembrane space-facing membranes and assembles in a range of complexes. Even though multiple isoforms are expressed at the mRNA level, only a single polypeptide that co-migrates with the human isoform lacking exon 5 is expressed in human skin fibroblasts, HEK293 cells, and murine heart and liver mitochondria. Finally, using a new genome-edited mammalian BTHS cell culture model, we demonstrate that the loss-of-function mechanisms for two BTHS alleles that represent two of the seven functional classes of BTHS mutation as originally defined in yeast, are the same when modeled in human TAZ.

8. Effect of Rare Earth Erbium on Microstructure and Mechanical Properties of A356 Aluminum Alloy

Abstract

For conventional A356 aluminum alloy, the addition of rare earth elements is an effective way to modify its microstructures and improve its mechanical properties. The effect of rare earth Er on microstructures and properties of as-cast A356 aluminum alloy was studied by differential scanning calorimetry (DSC), X-ray diffraction (XRD) and scanning electron microscopy (SEM). The results indicate that rare earth element Er is a good modifier for A356 aluminum alloy, which is able to improve obviously the microstructure of as-cast alloy. The addition of Er refines the primary α-Al phase, decreases the secondary dendrite arm spacing and the dendrite arm diameter, and modifies the eutectic Si in as-cast structure simultaneously. When the amount of Er reaches 0.4% (mass fraction, the same below), the refinement effect is the most significant, secondary dendrite arm spacing decreases from 53.6μm to 17.5μm and the morphology of eutectic Si transforms from thick lath-shaped to short rod-like or round granular. Compared with A356 alloy, the tensile strength and elongation of A356 aluminum alloy with the addition of 0.4%Er increase by 15.1% and 29.8% respectively.

12. The Relationship between Teachers' and Students' Preferences Regarding Adopting Various Classroom Activities in Adult, English as a Foreign Language (EFL) Classrooms in Taiwan

Abstract

Bilingual education has been a focus in Taiwan especially alter the educational reform in 2002 (Ministry of Education, 2003). With the educational reform, many teaching methods and approaches had been explored and implemented recently to enhance English as a Foreign Language (EFL) education. The traditional teaching approaches focused on the drills such as teaching students' subordinate language skills, or dividing students into different levels according to their mastery of language. Doing translation exercises, mimicking, and memorizing dialogue patterns were examples of the typical classroom activities which result in EFL students' lack of communicative competence. Learner-centered approaches had become the current trend and are gradually replacing the traditional EFL teaching approaches. Learner-centered education focused on maximizing learners' needs for meaningful communicative opportunities in students' second language (L2), promoting learners' active engagement in the classroom, and emphasizing learners' primary role in the learning processes. The main purpose of this study was to compare teachers' and students' preferences in various EFL instructional activities; examine how well teachers were aware of their students' learning preferences; and provide suggestions or alternative teaching ideas to promote classroom interaction and effectiveness. The subjects of the study were Taiwanese EFL college students. The teacher participants were English teachers employed or had been employed in Taiwanese colleges or universities. This research design utilized both qualitative and quantitative methods with more emphasis on quantitative method. The survey contained a six-point Likert Scale questionnaire and open-ended questions to collect EFL students and teachers' perceptions and attitude in implementing various classroom activities. Descriptive statistics were implemented for data analysis and to determine if there was significant difference or mismatch between the perceptions of two group participants.
