5. miR-133b Promotes Esophageal Squamous Cell Carcinoma Metastasis.

Abstract

Background: Evaluation of biological changes at the molecular level has important clinical implications for improving the survival rate of esophageal squamous cell carcinoma (ESCC). Therefore, we plan to analyze and elucidate the expression of microRNA-133b (miR-133b), M2 pyruvate kinase (PKM2), and signal transducer and activator of transcription 3 (STAT3) in ESCC and their associated clinicopathological significance. Methods: The 72 patients with ESCC were selected as the experimental study group. Normal adjacent tissues (NAT) were matched as the control group. In this study, in situ hybridization was used to detect the expression of miR-133b in ESCC, and tissue expressions of PKM2 and STAT3 were detected by immunohistochemistry, and literature review was conducted. Results: Studies had shown that the positive expression of miR-133b in NAT was significantly higher than that in ESCC (χ2 = 9.007, P =.003). PKM2 and STAT3 in ESCC had a significantly higher positive expression levels than those of NAT (χ2 = 56.523, P =.000; χ2 = 72.939, P =.000). From correlation analysis, there was a negative correlation between miR-133b and PKM2(r = −0.515, P <.001), a negative correlation between miR-133b and STAT3(r = −0.314, P =.007), and a positive correlation between PKM2 and STAT3(r = 0.771, P <.001). Conclusions: In ESCC, our study demonstrated that downregulation of miR-133b and upregulation of PKM2 and STAT3. We predict that miR-133b may inhibit the STAT3 pathway by downregulating PKM2. Abstract

Acetylation of extracellular proteins has been observed in many independent studies where particular attention has been given to the dynamic change of the microenvironmental protein post‐translational modifications. While extracellular proteins can be acetylated within the cells prior to their micro‐environmental distribution, their deacetylation in a tumor microenvironment remains elusive. Here it is described that multiple acetyl‐vWA domain‐carrying proteins including integrin β3 (ITGB3) and collagen 6A (COL6A) are deacetylated by Sirtuin family member SIRT2 in extracellular space. SIRT2 is secreted by macrophages following toll‐like receptor (TLR) family member TLR4 or TLR2 activation. TLR‐activated SIRT2 undergoes autophagosome translocation. TNF receptor associated factor 6 (TRAF6)‐mediated autophagy flux in response to TLR2/4 activation can then pump SIRT2 into the microenvironment to function as extracellular SIRT2 (eSIRT2). In the extracellular space, eSIRT2 deacetylates ITGB3 on aK416 involved in cell attachment and migration, leading to a promotion of cancer cell metastasis. In lung cancer patients, significantly increased serum eSIRT2 level correlates with dramatically decreased ITGB3‐K416 acetylation in cancer cells. Thus, the extracellular space is a subcellular organelle‐like arena where eSIRT2 promotes cancer cell metastasis via catalyzing extracellular protein deacetylation. [ABSTRACT FROM AUTHOR]

Abstract

Cubic perovskite-related ceramic CaCu3Ti4O12 has a very high dielectric constant larger than 10000 in the kilohertz frequency region. It is almost constant in the 100–600 K region. The ceramic compounds were prepared by conventional solid-state reaction at different sintering temperatures 1040 degrees C(C04), 1060 degrees C(C06), 1080 degrees C(C08), and 1100 degrees C(C10). Impedance spectroscopy on CaCusTi4O12 ceramics demonstrates that they are electrically heterogeneous and consist of semi-conducting grains with insulating grain boundaries. It is shown that C06 is the most suitable one for device applications. Surface observation and analysis of the grain-boundary-grained structure were carried out by using scanning electron microscopy, which confirms that the samples have clearly different crystal grain sizes and C06 has a larger grain size of about 20 μm. Compared to the dielectric relaxation spectroscopy, the mechanical relaxation spectroscopy on C06 was performed and a mechanical loss peak was found, which was attributed to the thermally activated behaviour being immanent in the grains.

Abstract

Thin films of Nd3+-substituted Bi3.15Nd0.85Ti3O12 (BNT) were fabricated on the (111)Pt/TiO2/SiO2/Si substrates by a metalorganic deposition (MOD) technique. These thin films are possessed of a single-phase bismuth-layered structure showing the preferred (001) and (117) orientation. The values of the remanent polarization Pr and coercive field Ec of the BNT thin film are 27 μC/cm2 and 157 kV/cm, respectively. The results of fatigue and retention tests revealed that the BNT thin film was not fatigued up to 1.44×1010 switching cycles and the retained charge was unchanged after 1×105 s. The leakage current behaviour of the BNT thin film was investigated at room temperature and their conduction mechanisms were also discussed. The I–V characteristics of the film show the ohmic behaviour for applied field lower than 40 kV/cm. Nonlinearity in the I–V behaviour was observed at an applied field above 40 kV/cm. In the high field region (E>95 kV/cm) the sample shows Schottky emission.

Abstract

Both Nd substituted and W doped bismuth titanate thin films were deposited by chemical solution deposition (CSD) technique on the (111)Pt/Ti/SiO2/(100)Si substrates. B-site doping with W6+, enhanced the remnant polarization and improved the fatigue property. Remnant polarization of Bi3.15Nd0.85Ti2.98W0.02O12 (BNTW) is 27 μ C/cm2, which is higher than that of Bi3.15Nd0.85Ti3O12 (BNT) deposited in the same condition. Good fatigue endurance was confirmed at 1 M Hz frequency up to 1010 switching cycles in both BNTW and BNT thin films. However, at the frequency of 50 kHz, the BNTW thin films show an improved fatigue property. W-doped effect is discussed.
