Amplification of the epidermal growth factor receptor gene (EGFR) represents one of the most commonly observed genetic lesions in glioblastoma (GBM); however, therapies targeting this signaling pathway have failed clinically. Here, using human tumors, primary patient-derived xenografts (PDX), and a murine model for GBM, we demonstrate that EGFR inhibition leads to increased invasion of tumor cells. Further, EGFR inhibitor-treated GBM demonstrates altered oxidative stress, with increased lipid peroxidation, and generation of toxic lipid peroxidation products. A tumor cell subpopulation with elevated aldehyde dehydrogenase (ALDH) levels was determined to comprise a significant proportion of the invasive cells observed in EGFR inhibitor-treated GBM. Our analysis of the ALDH1A1 protein in newly diagnosed GBM revealed detectable ALDH1A1 expression in 69% (35/51) of the cases, but in relatively low percentages of tumor cells. Analysis of paired human GBM before and after EGFR inhibitor therapy showed an increase in ALDH1A1 expression in EGFR-amplified tumors (P < 0.05, n = 13 tumor pairs), and in murine GBM ALDH1A1-high clones were more resistant to EGFR inhibition than ALDH1A1-low clones. Our data identify ALDH levels as a biomarker of GBM cells with high invasive potential, altered oxidative stress, and resistance to EGFR inhibition, and reveal a therapeutic target whose inhibition should limit GBM invasion.

Glioblastoma multiforme (GBMs) are recurrent lethal brain tumours. Recurrent GBMs often exhibit mesenchymal, stem-like phenotypes that could explain their resistance to therapy. Analyses revealed that recurrent GBMs have increased tension and express high levels of glycoproteins that increase the bulkiness of the glycocalyx. Studies showed that a bulky glycocalyx potentiates integrin mechanosignalling and tissue tension and promotes a mesenchymal, stem-like phenotype in GBMs. Gain- and loss-of-function studies implicated integrin mechanosignalling as an inducer of GBM growth, survival, invasion and treatment resistance, and a mesenchymal, stem-like phenotype. Mesenchymal-like GBMs were highly contractile and expressed elevated levels of glycoproteins that expanded their glycocalyx, and they were surrounded by a stiff extracellular matrix that potentiated integrin mechanosignalling. Our findings suggest that there is a dynamic and reciprocal link between integrin mechanosignalling and a bulky glycocalyx, implying a causal link towards a mesenchymal, stem-like phenotype in GBMs. Strategies to ameliorate GBM tissue tension offer a therapeutic approach to reduce mortality due to GBM.

12. Highly efficient ultraviolet and visible light emissions from upconversion transitions in Ho/Tm/Yb co-doped lutecia-stabilized zirconia single crystals excited by a 980 nm laser.

- Abstract
High-quality transparent cubic lutecia-stabilized zirconia (LSZ) and LSZ:Yb0.25Tm0.5Ho0.03 single crystals were fabricated by the OFZ (optical floating zone) method for the first time. For comparison, YSZ and YSZ:Yb0.25Tm0.5Ho0.03 single crystals were also fabricated. The transmittance of the LSZ single crystal (80.7%) is slightly lower than that of the YSZ single crystal (86.3%) in the visible light region, which is attributed to the LSZ single crystal with a higher density (6.604 g cm−3) in comparison with that of the YSZ single crystal (5.973 g cm−3). No absorption peaks were observed in the transmission and absorption spectra of LSZ and YSZ within 200–1100 nm, indicating that both crystals are excellent host crystals. The density of LSZ:Yb0.25Tm0.5Ho0.03 single crystals (6.596 g cm−3) is also higher than that of YSZ:Yb0.25Tm0.5Ho0.03 single crystals (6.005 g cm−3). The absorption spectra of LSZ:Yb0.25Tm0.5Ho0.03 and YSZ:Yb0.25Tm0.5Ho0.03 single crystals exhibited several absorption peaks at 363, 453, 678, and 785 nm and a strong broad absorption band within 850–1000 nm. The upconversion photoluminescence (UCPL) spectrum of the LSZ:Yb0.25Tm0.5Ho0.03 single crystal excited at 980 nm exhibited several emission peaks at 308, 373, 488, 540, 550, 655 and 794 nm; similar emission peaks were observed in the UCPL spectrum of the YSZ:Yb0.25Tm0.5Ho0.03 single crystal. The intensities of the emission peaks of the former are higher than those of the corresponding peaks of the latter, which is due to the former having a higher density and lower phonon energy in comparison with the latter. Power curves indicated that the 308 and 373 nm emissions are 4-photon upconversion processes, the 488 nm emission is a 3-photon process, and the 540 nm and 655 nm emissions are 2-photon processes. These results indicate that under the excitation at 980 nm, the LSZ:Yb0.25Tm0.5Ho0.03 single crystal has high efficiency over the YSZ:Yb0.25Tm0.5Ho0.03 single crystal in ultraviolet and visible light emissions. PurposeTo identify mediators of glioblastoma antiangiogenic therapy resistance and target these mediators in xenografts.Experimental designWe conducted microarray analysis comparing bevacizumab-resistant glioblastomas (BRG) with pretreatment tumors from the same patients. We established novel xenograft models of antiangiogenic therapy resistance to target candidate resistance mediator(s).ResultsBRG microarray analysis revealed upregulation versus pretreatment of receptor tyrosine kinase c-Met, which underwent further investigation because of its prior biologic plausibility as a bevacizumab resistance mediator. BRGs exhibited increased hypoxia versus pretreatment in a manner correlating with their c-Met upregulation, increased c-Met phosphorylation, and increased phosphorylation of c-Met-activated focal adhesion kinase and STAT3. We developed 2 novel xenograft models of antiangiogenic therapy resistance. In the first model, serial bevacizumab treatment of an initially responsive xenograft generated a xenograft with acquired bevacizumab resistance, which exhibited upregulated c-Met expression versus pretreatment. In the second model, a BRG-derived xenograft maintained refractoriness to the MRI tumor vasculature alterations and survival-promoting effects of bevacizumab. Growth of this BRG-derived xenograft was inhibited by a c-Met inhibitor. Transducing these xenograft cells with c-Met short hairpin RNA inhibited their invasion and survival in hypoxia, disrupted their mesenchymal morphology, and converted them from bevacizumab-resistant to bevacizumab-responsive. Engineering bevacizumab-responsive cells to express constitutively active c-Met caused these cells to form bevacizumab-resistant xenografts.ConclusionThese findings support the role of c-Met in survival in hypoxia and invasion, features associated with antiangiogenic therapy resistance, and growth and therapeutic resistance of xenografts resistant to antiangiogenic therapy. Therapeutically targeting c-Met could prevent or overcome antiangiogenic therapy resistance.

Inhibition of VEGF signaling leads to a proinvasive phenotype in mouse models of glioblastoma multiforme (GBM) and in a subset of GBM patients treated with bevacizumab. Here, we demonstrate that vascular endothelial growth factor (VEGF) directly and negatively regulates tumor cell invasion through enhanced recruitment of the protein tyrosine phosphatase 1B (PTP1B) to a MET/VEGFR2 heterocomplex, thereby suppressing HGF-dependent MET phosphorylation and tumor cell migration. Consequently, VEGF blockade restores and increases MET activity in GBM cells in a hypoxia-independent manner, while inducing a program reminiscent of epithelial-to-mesenchymal transition highlighted by a T-cadherin to N-cadherin switch and enhanced mesenchymal features. Inhibition of MET in GBM mouse models blocks mesenchymal transition and invasion provoked by VEGF ablation, resulting in substantial survival benefit.

34. Gentiopicroside Ameliorated Ductular Reaction and Inflammatory Response in DDC-induced Murine Cholangiopathies Model

- Abstract
Background: Cholangiopathies comprise a spectrum of diseases without curative treatments. Pharmacological treatments based on bile acid (BA) metabolism regulation represent promising therapeutic strategies for the treatment of cholangiopathies. Gentiopicroside (GPS), derived from the Chinese medicinal herb Gentianae Radix, exerts pharmacological effects on bile acid metabolism regulation and oxidative stress.Objective: The present study aims to investigate the effect of GPS on 3,5-diethoxycarbonyl-1,4dihydrocollidine (DDC)-induced cholangiopathy.Methods: Two independent animal experiments were designed to evaluate the comprehensive effect of GPS on chronic DDC diet-induced cholangiopathy, including bile duct obliteration, ductular reaction, BA metabolism reprogramming, liver fibrosis, oxidative stress and inflammatory responses.Results: In the first pharmacological experiment, three doses of GPS (5, 25 and 125 mg/kg) were injected intraperitoneally into mice fed a DDC diet for 14 days. DDC induced a typical ductular reaction, increased periductal fibrosis and mixed inflammatory cell infiltration in the portal areas. GPS treatment showed dose-dependent improvements in the ductular reaction, BA metabolism, fibrosis, oxidative stress and inflammatory response. In the second experiment, a high dose of GPS was injected intraperitoneally into control mice for 28 days, resulting in no obvious histologic changes and significant serologic abnormalities in liver function. However, GPS inhibited DDC-induced oxidative stress, serum and hepatic BA accumulation, proinflammatory cytokine production, and immunocyte infiltration. Specifically, the GPS-treated groups showed decreased infiltration of monocyte-derived macrophages and CD4+ and CD8+ T lymphocytes, as well as preserved Kupffer cells.Conclusion: GPS alleviated chronic DDC diet-induced cholangiopathy disorder by improving the ductular reaction, periductal fibrosis, oxidative stress and inflammatory response. Its dosage-dependent pharmacological effects indicated that GPS warrants its further evaluation in clinical trials for cholangiopathy.

One defining feature of domestic movies in 2016 is the outstanding performance made by art movies. In general, these art movies improved the overall quality of Chinese movies and the aesthetic appreciation of Chinese viewers. While highlighting the values of art and thought amid the commercial or entertainment movies, the art movies aroused public concern for domestic movies. Domestic art movies in 2016 are characterized by a close link between their art form and avant–garde style and their realist approach, affinity to the people and thought–provoking insight. Such characteristics enabled them to make new contributions to the development of realism. Some exhibited the producers’ relentless exploration and pursuit of innovation, vividly demonstrating the diversity of contemporary art movies. For example, some delivered an impressive modern interpretation of traditional Chinese aesthetic philosophy–"the extraordinary lies in the ordinary;"some had a novel camera expression and unique space–time composition; some established an in–depth connection between laughter and critical thinking by combining comic and tragic elements; some rendered innovative presentations by integrating modern style with ethnic culture; and some adopted ideographic means to enhance the significance of art movies. At present, however, Chinese art movies still lack a grand spiritual vision, with their themes to be further enriched and their concepts and expressions to be better integrated. Domestic art movies need to be endowed with more profound significance.
