Acinetobacter baumannii é um cocobacillus Gram-negativo que causa doenças graves em humanos em todo o mundo. A. baumannii é um agente patogénico oportunista e altamente perigoso, sendo considerado pela Organização Mundial de Saúde como uma ameaça de prioridade máxima para a saúde pública. Devido à sua grande capacidade de adquirir resistência aos medicamentos, juntamente com a sua tendência natural para se disseminar e sobreviver em ambientes hostis, tais como unidades de cuidados intensivos, A. baumannii é considerado um importante agente patogénico a estudar, impulsionando assim a vanguarda da investigação biomédica. O sucesso de A. baumannii como agente patogénico principal pode ser atribuído à presença de fatores de virulência, tais como a capacidade de formar biofilmes, aderência, colonizar e invadir células epiteliais humanas, o repertório de determinantes de resistência aos medicamentos e a sua maior capacidade de adquirir material genético de outros agentes patogénicos bacterianos. Neste estudo, pretendemos caracterizar epidemiologicamente uma coleção de isolados clínicos de A. baumannii, recolhidos durante um surto ocorrido num hospital da área metropolitana de saúde de Lisboa e correlacionar o perfil de resistência aos antibióticos com os clones encontrados e estudar a relação genética entre estas estirpes. Sessenta e oito isolados clínicos de A. baumannii foram incluídos no estudo. A diversidade genética foi estudada pela técnica de M13-PCR “fingerprinting” para a diversidade local e a tipagem de sequência multilocus (MLST), utilizando o esquema Oxford, que permitiu a identificação dos clones em circulação e uma comparação mundial. No final deste projeto, identificamos os clones de A. baumannii responsáveis por este surto inesperado e preocupante. Este estudo permitiu detetar a ocorrência de transmissão clonal de A. baumannii. A disseminação destes isolados em ambientes hospitalares é extremamente preocupante, principalmente devido à falta de opções terapêuticas, e precisa de ser cuidadosamente abordada com o objetivo de fornecer medidas adequadas para evitar a ocorrência de casos futuros e a sua disseminação para outros ambientes, em termos de comunidade. Acinetobacter baumannii is a Gram-negative coccobacillus that causes severe disease in humans worldwide. A. baumannii is an opportunistic and highly dangerous pathogen, being considered by the World Health Organization as a top priority threat for public health. Because of its great capacity to acquire drug resistance, together with its natural tendency to disseminate and survive in hostile environments, such as intensive care units, A. baumannii is an important pathogenic agent to study, thus driven the forefront of biomedical research. A. baumannii success as a major pathogen can be attributed to the presence of virulence factors, such as the ability to form biofilms, adherence, to colonize and invade human epithelial cells, the repertoire of drug resistance determinants and its increased capacity to acquire genetic material from other bacterial pathogenic agents. In this study, we aim to characterize epidemiologically a collection of clinical isolates of A. baumannii, collected during an outbreak that occurred in a hospital in the Lisbon metropolitan health care area, and to correlate the drug resistance profile with the clones found and study the genetic relatedness between these strains. Sixty-eight clinical isolates of A. baumannii were included in the study. The genetic diversity was studied by M13-PCR fingerprinting for local diversity and multilocus sequence typing (MLST), using the Oxford scheme, to allow the identification of the clones in circulation and a worldwide comparison. At the end of this project, we identify A. baumannii clones responsible for this unexpected and worrying outbreak. This study provides evidence of clonal transmission of A. baumannii. The spread of these isolates in hospital environments is extremely worrisome, mainly due to the lack of therapeutic options, and needs to be carefully addressed aiming to provide suitable measures to avoid the occurrence of future cases and its dissemination to other environments, as to the community. The increased average lifespan, together with low birth rates, are transforming the European Union's age pyramid. Currently, we are experiencing a transition towards a much older population structure. Given that the institutions that provide care to these population groups are limited by budgetary constraints, it is imperative to optimize several processes, among which route planning and staff scheduling stand out. This dissertation aims to develop a mathematical programming model to support the planning of routes and human resources for providers of Home Social Services. Beyond general Vehicle Routing Problems assumptions, the proposed model also considers the following features: i) working time regulations, ii) mandatory breaks, iii) users' autonomy, and iii) meals' distribution. The present model, implemented using GAMS software, focuses simultaneously on two objective functions: minimization of operating costs, and maximization of equity through the minimization of differences in teams' working times. Chebyshev's method was chosen to solve the developed multiobjective model. The model was built based on a Portuguese Private Institution of Social Solidarity. Through the application of the model, significant improvements are obtained when compared to the current planning of the partner institution, such as it is the case of an improved workload distribution between caregivers and routes that will result in lower costs for the institution. This model is fully enforceable to other institutions that provide services similar or equal to the institution used as a reference. O aumento da esperança média de vida, juntamente com baixas taxas de natalidade, estão a transformar a pirâmide etária da União Europeia. Atualmente, estamos a vivenciar uma transição direcionada para uma estrutura populacional muito mais envelhecida. Dado que as instituições que prestam cuidados a esta fração se encontram limitadas por restrições orçamentais, torna-se imperativo otimizar vários processos, dos quais se destacam planeamento de rotas e escalonamento de funcionárias. Esta dissertação visa introduzir um modelo de programação matemática com a finalidade de apoiar o planeamento de rotas e recursos humanos para prestadores de Serviços de Apoio Domiciliário. O modelo assenta, além dos pressupostos de um "Vehicle Routing Problem", nos seguintes: i) regulações de tempo de trabalho, ii) pausas obrigatórias, iii) autonomia dos utentes, e iv) distribuição de refeições. O modelo, desenvolvido através de software GAMS, foca-se em duas funções objetivo, simultaneamente: minimização dos custos operacionais, e maximização da equidade, através da minimização das diferenças nos tempos de trabalho das equipas. O método de Chebyshev foi o escolhido para desenvolver o modelo multiobjetivo. O modelo foi construído tendo por base uma Instituição Particular de Solidariedade Social em Portugal. Através da aplicação do modelo, obtêm-se melhorias significativas, quando comparado com o atual planeamento da instituição parceira, como é o caso de uma melhor distribuição da carga de trabalho entre as funcionárias e das rotas que resultam da redução dos custos operacionais da instituição. Este modelo é totalmente extensível a outras instituições que prestem serviços semelhantes ou iguais à instituição utilizada como referência.

As far as I know, this is the first volume dedicated to scientific collections in a conservation journal, something I have to thank to the previous director of Conservar Património, António João Cruz, and the current director, Elin Figueiredo. The fact that there are only four articles does not mean that the problem is uncommon or uninteresting, but only that we are in its infancy. I hope that these four articles will open up new perspectives of conservation research or at least awaken the curiosity of conservator-restorers, especially those currently in training. The mammalian gut is a heterogeneous environment inhabited by a large and diverse microbial community, including bacteria and their viral predators, bacteriophages (phages). Dynamic interactions between virulent phages and bacteria in the gut are still poorly understood, which is also an obstacle for the design of successful therapeutic interventions based on phages. Independent experiments have shown that virulent phages were found to have no major effects on their targeted bacteria in the gut, in spite of sustainable phage amplification. This suggests that there are unknown factors in the gut environment that modulate these interactions. Using comparative transcriptomics analysis of E. coli grown in vitro and in vivo (within the mammalian gut) we found that in the gut, bacteria downregulate the expression of genes related to phage receptors, which provides an explanation for the lack of selection of phage-resistant bacteria during in vivo experiments. We also found that the acquisition of a pathogenicity island commonly found in human E. coli isolates affects phage susceptibility possibility by downregulating a defense mechanism against invading DNA. Finally, we examined the repartition of phages and bacteria through mucosal and luminal gut sections and observed a heterogeneous spatial distribution of these two antagonist populations, supporting the hypothesis of source-sink dynamics. Altogether our data demonstrates that multiple factors encompassing, spatial distribution, bacterial physiology and defenses against foreign DNA modulate the interactions between bacteria and phages within the gut.; L'intestin des mammifères est peuplé de nombreux et divers microbes comprenant des bactéries et leurs prédateurs viraux, des bactériophages (phages). Les interactions entre les phages et les bactéries intestinales sont encore mal comprises. Des expériences indépendantes ont montré que les phages virulents n’avaient aucun effet majeur sur l’abondance des bactéries intestinales ciblées, en dépit de leur amplification durable. Cela suggère que des facteurs encore inconnus de l'environnement intestinal modulent ces interactions. À l’aide d’une analyse transcriptomique comparative de la bactérie Escherichia coli cultivée in vitro et in vivo (dans l’intestin de mammifères), nous avons constaté que, dans l’intestin, les bactéries réduisent l’expression de gènes liés aux récepteurs du phage. Ceci permet d’expliquer l’absence de sélection des bactéries mutées devenues résistantes aux phages lors d'expériences in vivo. D’autre part, nous avons montré que l'acquisition d'un îlot de pathogénicité, souvent associé aux souches intestinales humaines d'E. coli, affecte la susceptibilité aux phages en régulant négativement un mécanisme de défense contre l'ADN étranger. Enfin, nous avons examiné la répartition des phages et des bactéries dans les parties mucosales et luminales de l’intestin et avons observé une distribution spatiale hétérogène de ces deux populations antagonistes, corroborant l'hypothèse d'une dynamique « source-sink ». Globalement, nos données démontrent que de multiples facteurs incluant la distribution spatiale, la physiologie bactérienne et les défenses contre l’ADN étranger modulent les interactions entre bactéries et phages dans l’intestin des mammifères.

Vital Brazil Mineiro da Campanha (1865-1950) é conhecido pela sua descoberta que até hoje salva milhares de vidas: a especificidade dos soros antiofídicos. Este artigo integra-se nas comemorações dos 150 anos de Vital Brazil, com o objetivo de apresentar e discutir suas contribuições para a medicina tropical brasileira e internacional, relativamente ao estudo dos envenenamentos por animais peçonhentos e à descoberta da soroterapia antiofídica específica. Pretende abordar as biografias de Otto Wucherer (1820-1873) e Vital Brazil e o início das pesquisas sobre Ofidismo no Brasil, com similaridades em suas trajetórias, nomeadamente ao estudo do ofidismo e seu tratamento. A discussão da especificidade dos soros travada entre Vital Brazil e Albert Calmette (1863-1933) que se prolongou por vários anos (1902 a 1914), encerrada por Maurice Arthus (1862-1945), com ganho de causa para Vital Brazil, que em janeiro de 1916 pode comprovar sua tese na prática. L’histoire des cabinets royaux de physique portugais est mal connue. Les vastes collections d’instruments scientifiques qui les composaient, organisées au cours des xviiie et xixe siècles, ainsi que d’importantes bibliothèques et collections d’histoire naturelle destinées à l’éducation des jeunes Infants et au divertissement de la cour, ont presque intégralement disparu. Dans de nombreux cas, leur mémoire n’a survécu qu’à travers un petit nombre d’inventaires, de récits d’aristocrates de l’époque et de quelques voyageurs étrangers de passage à Lisbonne.Les collections du cabinet royal de physique, organisées en forme systématique et enrichies par le mathématicien vénitien Michel Franzini (c. 1740-1810) durant les trois dernières décennies du xviiie siècle, ont été progressivement démontées et éparpillées, au cours du xixe siècle, avec l’émergence des institutions universitaires, polytechniques et militaires et, à partir de 1910, avec l’avènement de la République et la nationalisation des biens royaux. Celles-ci avaient déjà subi les conséquences de leur transfert au Brésil (1807-1821) et de l’occupation et de la confiscation françaises (1807-1810).Une étude récente menée au sein des collections des musées techniques et universitaires, des institutions d’enseignement, ainsi que des anciennes résidences royales, a permis de faire ressurgir de nombreux instruments ayant appartenu aux collections royales. Ces objets d’origine diverse (Angleterre, Allemagne, Pays-Bas, France, Italie, Portugal) et de nature hétéroclite (physique, chimie, astronomie, mathématique, etc.) contribuent à mieux faire connaître et comprendre les cabinets de physique des monarques portugais. Cet article présente, sous une forme succincte, les résultats de cette recherche, en discutant plusieurs aspects de la formation et de la dispersion, de la provenance et de la « muséalisation » des cabinets. Little is known about the royal cabinets of physics of Portugal between the 17th and 19th centuries. Set up at different points in time, in tandem cabinets of natural history and libraries, the cabinets served to educate young princes of the Portuguese monarchy. In addition to their education uses, they also served for entertainment of the court. Although most of the instruments were scattered or lost during 19th century, there is a significant documentary record left behind about their constitution and content, most notably invoices, inventories and travel diaries of foreign visitors. This article provides a survey of recent research on the composition, the uses, and subsequent dissolution of the physics cabinets.The collections of the Royal Cabinet of Physics, were systematized and enriched by the Venetian mathematician working for the Portuguese court Michael Franzini (?-1810) between 1770s and 1790s. Throughout the 19th century, the collections were gradually dispersed with the advent of the Republic, the nationalization of royal property and the subsequent emergence of universities, polytechnics and military from 1910. The collections have already suffered the consequences a transfer to Brazil (1807-1821) and French occupation and confiscation of these objects (1807-1810).A recent study about collections in museums, universities and schools, as well as former royal residences, can help restitute many instruments that belonged to the Portuguese royal collections. These objects of diverse origin (England, Germany, Netherlands, France, Italy, Portugal) and heterogeneous nature (physics, chemistry, astronomy, mathematics, etc.) contribute to a better understanding of the Portuguese monarchs’ relation to science and technology in early modern period.

Relatório de Estágio do Mestrado em História da Arte, Património e Turismo Cultural - extinto apresentado à Faculdade de Letras No âmbito do estágio curricular do Mestrado de História da Arte, Património e Turismo Cultural, pela Faculdade de Letras da Universidade de Coimbra, realizado entre Setembro de 2015 e Março de 2016, no Mosteiro de Santa Claraa-Velha, tutelado pela Direção Regional da Cultura do Centro, foi proposto um estudo sobre o funcionamento do Serviço Educativo e o impacto da divulgação em instituições culturais. A escolha deste tema assentou no facto de as minhas funções dentro da instituição, se concentrassem maioritariamente no Serviço Educativo da entidade de acolhimento. Neste relatório, começo por fazer uma breve contextualização histórica do monumento e espaço museológico, e uma análise sobre o modo de funcionamento, das suas valências e gestão levada a cabo pela DRCC. Posteriormente elabora-se uma reflexão sobre o papel do Serviço Educativo e a importância de um Departamento de divulgação em instituições culturais. Esta reflexão vai servir de ponte para uma exposição detalhada da situação do Serviço Educativo do Mosteiro de Santa Clara-a-Velha, e as mudanças sofridas desde da sua abertura até à atualidade. O relatório conclui com a memória descritiva das atividades levadas a cabo ao longo dos seis meses, e da proposta de actividade programada por mim e pela outra estagiária, Beatriz Barroca. The idiopathic vestibular syndrome (IVS) is characterized by a lesion of the vestibular portion of the vestibulocochlear nerve (VIII). There is no known etiology and it usually leads to vestibular clinical signs with an acute onset and severe presentation, which enter in remission in three to four weeks, without any instituted medication. This study intends to analyse and compare the results of the neurological examinations and the respective ancillary diagnostic exams with the disease progression and prognosis of each animal suspected with idiopathic vestibular disorder, followed at Clínica de Referência Veterinária (RRV). In general, the evolution of the described clinical cases was according with the literature. The proposed causes for IVS are early Otitis interna/media, abnormal production, circulation and absorption of the endolymphatic fluid, neuritis of the vestibular portion of the vestibulocochlear nerve, Cuterebra larvae aberrant migration (not present in Portugal) and herpesvirus-1 infection of vestibular labyrinth and ganglion in dogs., Orientação: David Orlando Alves Ferreira
